Abstract

ZusammenfassungTraditionell werden pharmazeutische Zubereitungen der Weidenmistel häufig bei der Therapie von Blasen- und Nierenzellkarzinomen eingesetzt. Dennoch existieren bislang nur wenige wissenschaftliche Untersuchungen zu Wirkmechanismen von Weidenmistelpräparaten. Daher wurde mithilfe verschiedener Testverfahren die Hemmung der Viabilität von Tumorzellen durch den Weidenmistelextrakt Iscucin® Salicis bestimmt und die beteiligten Mechanismen untersucht. Des Weiteren wurde die Hemmung der Zellmigration analysiert. Ergebnisse Der Extrakt hemmte Zellwachstum und -migration in vitro, wobei die Sensitivität der einzelnen Tumorzelllinien variierte. Vor allem sprachen neuroepitheliale Tumorzellen und Bronchialkarzinomzellen gut an. Zudem wurden pro-apoptotische Marker aktiviert. Klinische Untersuchungen stehen aus, um zu klären, bei welchen Tumoren das größte Potenzial für eine Behandlung mit Iscucin® Salicis besteht.

Abstract

Zusammenfassung Hintergrund und Ziel Mistelpräparate, insbesondere aus der Apfelbaummistel (Viscum album ssp. album, Mali), werden häufig von Mammakarzinom-Patientinnen als komplementäre Therapiemaßnahme nach Resektion, Chemo- oder Radiotherapien angewendet. In klinischen Studien wurde bereits der Nutzen von Mistelpräparaten nachgewiesen. Da die dahinter liegenden Wirkmechanismen bisher nicht vollständig aufgeklärt sind, sollten die potenziell antitumoralen Effekte eingehender untersucht werden. Material und Methoden Es wurden In-vitro-Untersuchungen zu den Effekten eines wässrigen Extrakts aus Viscum album ssp. album, Mali (Iscucin® Mali) auf die Zellviabilität, auf verschiedene Phasen der Angiogenese (Zellproliferation und Kapillarausbildung) sowie auf die Zellmigration humaner Tumorzellen durchgeführt. Ergebnisse Zwei verschiedene Assays gaben Hinweise auf antiproliferative Effekte mit einer halbmaximalen Hemmung bei Konzentrationen Diskussion und Schlussfolgerung Ein Panel aus verschiedenen Untersuchungsmethoden zeigte in Abhängigkeit von der Zelllinie antitumorale Effekte von Iscucin® Mali, vor allem im Bereich der Kapillarausbildung und der Zellmigration. Dies lässt vermuten, dass sowohl das Tumorwachstum als auch die Metastasierung verzögert werden können. Weitere Studien sind erforderlich, um die Relevanz der gewonnenen Daten im klinischen Bereich zu untermauern.

Abstract

Ethnopharmacological relevance Mistletoe has been used since ancient times in Europe mostly for medicinal purposes. Since 1917, mistletoe preparations have been applied in cancer therapy and today are the most frequently used complementary medicine in tumor treatment. The main cytotoxic constituents of Viscum album are lectins and viscotoxins. Aim of the study The aim of this in vitro study was to investigate the antiproliferative potential of Viscum album preparations from different host trees and to assess the impact of mistletoe lectin 1 (ML-1) and viscotoxin A (VT-A) in comparison to a structurally similar lectin and thionin. Materials and methods By means of widely accepted 2D Alamar Blue Assay, based on population counting of living cells using a fluorescent cell viability dye, the potential impact to inhibit tumor cell of the mistletoe preparations (Iscucin®) and their single compounds (ML-1 and VT-A) on the cell growth of six human cancer cell lines were evaluated. Also the mixture of ML-1 and VT-A corresponding to the contents in the specific mistletoe preparations were monitored. Ricin and purothionin were used as reference lectin and reference thionin, respectively. Results The lung carcinoma cell line HCC827 was very sensitive to the Iscucin® preparations. Very strong antiproliferative effects were found with Iscucin® Salicis and Tiliae and a strong with Iscucin® Crataegi, Mali and Populi. The IC50 concentrations of the Iscucin® preparations correlated with their respective ML-1 contents, but the ML-1 levels were much lower than the IC50 concentration of isolated ML-1 (1 ng/ml – 56 ng/ml). ML-1 was much more effective than ricin. Iscucin® preparations, ML-1 and ricin showed antiproliferative activity on human tumor cells. VT-A and purothionin had no effect on cell viability in the concentration ranges tested. Conclusion The complete mistletoe extract is more potent to inhibit tumor cell proliferation than isolated ML-1 at an equivalent concentration level. Phenolic compounds found in all Iscucin® preparations might contribute to uphold the cytotoxic activity of ML-1 by antioxidative action. However, further studies are necessary to evaluate the role of VT-A and possible synergistic actions to the antiproliferative effect of aqueous mistletoe extracts.

Abstract

Background The therapeutic use of Helleborus niger L. is manifold due to its specific phytochemical composition. Two compound groups, the ranunculin derivates including protoanemonin and the steroidal saponins, are also associated with toxicity (genotoxicity, disintegration of membrane structures). Therefore, in vitro investigations were performed on safety aspects of a Helleborus niger aqueous fermented extract (HNE). In addition its therapeutic potential against various cancer cell lines was assessed to gain insight into the respective mechanisms of action. Methods To evaluate the safe use of HNE, Ames and hemolytic tests were carried out. Two angiogenesis assays in 2D and 3D design were conducted to assess the anti-angiogenetic potential, for which human umbilical vein endothelial cells (HUVEC) were chosen. A panel of tumor cell lines was used in 2D and 3D proliferation assays as well in the migration- and invasion-assay. All investigations were performed with HNE compared to reference substances. The 2D proliferation assay was additionally performed with isolated compounds of HNE (characteristic steroidal saponins). Results HNE did not exhibit any genotoxic potential. Concentrations up to 10 μl/ml were classified as non-hemolytic. HNE exerted anti-angiogenetic effects in HUVEC and anti-proliferative effects in five cancer cell lines, whereas hellebosaponin A and D as well macranthosid I did not show comparable effects neither singly nor in combination. Due to the inherent instability of protoanemonin in isolated form, parallel investigations with protoanemonin could not be performed. HNE (600–1000 μg/ml) inhibited the migration of certain cancer cells by > 80% such as Caki-2, DLD-1 and SK-N-SH. Conclusion HNE exhibit neither genotoxic nor hemolytic potential. The present investigations verify the anti-angiogenetic effects on HUVEC, the anti-proliferative effects and migration-inhibiting properties on tumor cells. The lower effect of the relevant steroidal saponins compared to the whole extract underlines the fact that the latter is more effective than a blend of isolated pharmacologically active components. Abstract

Objectives While Taraxacum officinale (dandelion) extracts showed antitumor effects on adult cancer cells, effects on pediatric tumor cells as a single agent or in combination with mistletoe extracts are hitherto unknown. Material and methods The anti-proliferative effects of an aqueous fermented Taraxacum officinale extract (Taraxacum) on a pediatric cancer cell line panel were assessed by cell viability assays (MTT). In two neuroblastoma cell lines, SH-SY5Y and Kelly, the effects on cell cycle distribution (PI staining), mitochondrial integrity (MitoTracker staining), invasion (Boyden chamber assay) and migration (Scratch-assay) as well as the synergistic effects of the co-treatment of Taraxacum and mistletoe preparations (Iscucin® Tiliae or Iscucin® Pini) were investigated. Results All tested cancer cell lines were more susceptible to Taraxacum than the normal human fibroblast cell line, NHDF-C. In neuroblastoma cell lines Taraxacum caused apoptosis and loss of mitochondrial integrity as well as an inhibition of invasion and migration. The simultaneous therapy of Taraxacum and the mistletoe extracts revealed synergistic effects. Conclusion This preclinical data support the use of Taraxacum as a potential adjuvant application in pediatric oncology.

Abstract

Das Waldbingelkraut (Mercurialis perennis L.) - eine Pflanze aus der Familie der Wolfsmilchgewachse (Euphorbiaceae) - war bereits in vorchristlicher Zeit und im Mittelalter als probates Mittel zur Behandlung von eitrigen Wunden, Ekzemen und Geschwuren bekannt. Auch als starkes Abfuhrmittel und bei Menstruationsbeschwerden wurde Mercurialis verwendet. Hydroalkoholische Auszuge des Krautes und wassrig-fermentierte Extrakte finden heute insbesondere in der Komplementarmedizin Anwendung. Indikationen solcher Mercurialis-Zubereitungen sind z.B. schlecht heilende Wunden, Entzundungen, Verbrennungen, Hamorrhoiden sowie trockene Bindehautentzundung des Auges (Konjunktivitis). Die Inhaltsstoffe von M. perennis und deren Pharmakologie galten bislang als wenig erforscht. Neue phytochemische Untersuchungen zeigen, dass die Pflanze Alkaloide, atherisches Ol, n-Alkylresorcine sowie Depside und Flavonoide enthalt. In einem In-vitro-Entzundungsmodell an isolierten menschlichen Monozyten wurde nun erstmals die immunmodulatorische Wirkung der fermentierten Mercurialis-Urtinktur nachgewiesen - ein Befund, der mit einem beschleunigten Wundheilungsprozess nach Anwendung von Mercurialis-Urtinkturpraparaten im Einklang steht. Der Beitrag halt eine Ruckschau auf altere Literatur zur Pharmakognosie von M. perennis und stellt Daten aus der aktuellen Forschung vor.
