We present a representation of nitrogen and phosphorus cycling in the vegetation demography model the Functionally Assembled Terrestrial Ecosystem Simulator (FATES), within the Energy Exascale Earth System (E3SM) land model. This representation is modular, and designed to allow testing of multiple hypothetical approaches for carbon-nutrient coupling in plants. The model tracks nutrient uptake, losses via turnover from both live plants and mortality into soil decomposition, and allocation during tissue growth for a large number of size- and functional-type-resolved plant cohorts within a time-since-disturbance-resolved ecosystem. Root uptake is governed by fine root biomass, and plants vary in their fine root carbon allocation in order to balance carbon and nutrient limitations to growth. We test the sensitivity of the model to a wide range of parameter variations and structural representations, and in the context of observations at Barro Colorado Island, Panama. A key model prediction is that plants in the high-light-availability canopy positions allocate more carbon to fine roots than plants in low-light understory environments, given the widely different carbon versus nutrient constraints of these two niches within a given ecosystem. This model provides a basis for exploring carbon-nutrient coupling with vegetation demography within Earth System Models (ESMs).

While climate change is impacting all parts of the globe, boreal forests are experiencing disproportionally higher rates of temperature increase, thus drastically impacting one of the largest biomes in the world. Changes in high-latitude forests have strong implications to the regional water and carbon cycling, and shifts in canopy cover (i.e., abundance and shifts between evergreen and deciduous species) will alter albedo, ecosystem productivity, and surface and canopy water fluxes. For example, high latitude warming may increase nutrient availability via a deepening of the soil active layer. Warming also induces permafrost degradation and loss, leading to strong interactions that alter the hydrology, and soil biological and physical processes. These climate-related interactions will affect plant competitive interactions, survival, and ultimately community distribution and carbon storage. In this study we explore how vegetation dynamics will be affected by changes in plant and microbial N competition, differences in nutrient demand due to shifts in PFTs (e.g., faster resource acquisition in deciduous plants), and ultimately carbon allocation. To be able to accurately predict and model these complex ecological processes we are using a new demographic vegetation model (FATES; Functionally-Assembled Terrestrial Ecosystem Simulator) that is coupled to ELMv1, the land surface model in the global Earth System Model - E3SM. We present here the newly implemented representation of nutrient competition, acquisition, and extensible approach of nutrient and carbon allocation within plants in the ELM-FATES model. This work has successfully coupled the interactions of nutrients between soil biogeochemistry (BGC) in ELM and plant productivity and carbon in FATES, with improved model hypothesis testing for plant’s nutrient storage capacity. With the inclusion of nutrient cycling in the previously ‘carbon-only’ ELM-FATES where the largest competition was light driven, the productivity and biomass storage was significantly reduced for the simulated boreal forests. After conducting an uncertainty quantification experiment (i.e., using large ensembles to generate surrogate models) in order to test the model parameter sensitivity, we found that model parameters related to carbon storage and leaf economics had the largest sensitivity on plant processes. We then applied a Bayesian inference approach using neural networks to calibrate the model parameters against observational datasets, and greatly improved model predicts to match field inventory data. These newly represented ecological-based processes have helped to improve the representation of these vulnerable forests in an Earth System Model.

Climatic extreme events are expected to occur more frequently and potentially be stronger in the future, increasing the likelihood of unprecedented climate extremes (UCEs), or record-breaking events such as prolonged droughts, to occur. To prepare for UCEs and their impacts, we need to develop a better understanding of terrestrial ecosystem responses to events such as extreme drought. We know that intense, extreme droughts can substantially affect ecosystem stability and carbon cycling through increased plant mortality and delaying ecosystem recovery. Our ability to predict such effects is limited due to the lack of experiments focusing on climatic excursions beyond the range of historical experience.We explore the response of forest ecosystems to UCEs using two dynamic vegetation demographic models (VDMs), ED2 and LPJ-GUESS, in which the abundances of different plant functional types, as well as tree size- and age-class structure, are emergent properties of resource competition. We investigate the hypothesis that ecosystem responses to UCEs (e.g., unprecedented droughts) cannot be extrapolated from ecosystem responses to milder extremes, as a result of non-linear ecosystem responses (e.g. due to plant plasticity, functional diversity, and trait combinations). We evaluate each model’s mechanisms and state variables prior, during, and after a continuum of drought intensities ultimately reaching very extreme drought scenarios (i.e., 0% to 100% reduction in precipitation for drought durations of 1-year, 2-year, and 4-year scenarios) at two dry forested sites: Palo Verde, Costa Rica (i.e. tropical) and EucFACE, Australia (i.e. temperate). Both models produce nonlinear responses to these UCEs. Due to differences in model structure and process representation, the model’s sensitivity of biomass loss diverged based on either duration or intensity of droughts, as well as different model responses at each site. Biomass losses in ED2 are sensitive to drought duration, while in LPJ-GUESS they are mainly driven by drought intensity. Elevated atmospheric CO2 concentrations alone did not buffer the ecosystems from carbon losses during UCEs in the majority of our simulations. Our findings highlight discrepancies in process formulations and uncertainties in models, notably related to availability in plant carbohydrate storage and the diversity of plant hydraulic schemes. This shows that different hypotheses of plant responses to UCEs exist in two similar models, reflecting knowledge gaps, which should be tested with gap-informed field experiments. This iterative modeling-experiment framework would help improve predictions of terrestrial ecosystem responses and climate feedbacks.

Arctic shrub expansion has been widely reported in recent decades, with large impacts on carbon budgets, albedo, and warming rates in high latitudes. However, predicting shrub expansion across regions remains challenging because the underlying controls remain unclear. Observational studies and models typically use relationships between observed shrub presence and current environmental suitability (climate and topography) to predict shrub expansion, but such approaches omit potentially important biotic-abiotic interactions and non-stationary relationships. Here, we use long-term high-resolution satellite imagery across Alaska and western Canada to show that observed shrub expansion has not been controlled by environmental suitability during 1984-2014, but rather can only be explained by accounting for seed dispersal and fire. These findings provide the impetus for better observations of recruitment and for incorporating currently underrepresented processes of seed dispersal and fire in land models to project shrub expansion and future climate feedbacks. Integrating these dynamic processes with projected fire extent and climate, we estimate that shrubs will expand into 25% of the non-shrub tundra by 2100, in contrast to 39% predicted using a relationship with increasing suitability alone. Thus, using environmental suitability alone likely overestimates and misrepresents the spatial pattern of shrub expansion and its associated carbon sink.

Hence, we simulated an old-growth tropical forest using six versions of four terrestrial models differing in scale of vegetation structure and representation of biogeochemical (BGC) cycling, all driven with CO2 forcing from the preindustrial period to 2100. The models were benchmarked against tree inventory and eddy covariance data from a Brazilian site for present-day predictions. All models predicted positive vegetation growth that outpaced mortality, leading to continual increases in present-day biomass accumulation. Notably, the two vegetation demographic models (VDMs) (ED2 and ELM-FATES) always predicted positive stem diameter growth in all size classes. The field data, however, indicated that a quarter of canopy trees didn't grow over the 15-year period, and while high interannual variation existed, biomass change was near neutral. With a doubling of CO2, three of the four models predicted an appreciable biomass sink (0.77 to 1.24 Mg ha−1 year−1). ELMv1-ECA, the only model used here that includes phosphorus constraints, predicted the lowest biomass sink relative to initial biomass stocks (+21%), lower than the other BGC model, CLM5 (+48%). Models projections differed primarily through variations in nutrient constraints, then carbon allocation, initial biomass, and density-dependent mortality. The VDM's performance was similar or better than the BGC models run in carbon-only mode, suggesting that nutrient competition in VDMs will improve predictions. We demonstrate that VDMs are comparable to nondemographic (i.e., “big-leaf”) models but also include finer scale demography and competition that can be evaluated against field observations.

Forest disturbance and regrowth are key processes in forest dynamics but detailed information of these processes is difficult to obtain in remote forests as the Amazon. We used chronosequences of Landsat satellite imagery to determine the sensitivity of surface reflectance from all spectral bands to windthrow, clearcutting, and burning and their successional pathways of forest regrowth in the Central Amazon. We also assess whether the forest demography model Functionally Assembled Terrestrial Ecosystem Simulator (FATES) implemented in the Energy Exascale Earth System Model (E3SM) Land Model (ELM), ELM-FATES, accurately represents the changes for windthrow and clearcut. The results show that all spectral bands from Landsat satellite were sensitive to the disturbances but after 3 to 6 years only the Near Infrared (NIR) band had significant changes associated with the successional pathways of forest regrowth for all the disturbances considered. In general, the NIR decreased immediately after disturbance, increased to maximum values with the establishment of pioneers and early-successional tree species, and then decreased slowly and almost linearly to pre-disturbance conditions with the dynamics of forest succession. Statistical methods predict that NIR will return to pre-disturbance values in about 39 years (consistent with observational data of biomass regrowth following windthrows), and 36 and 56 years for clearcut and burning. The NIR captured the observed successional pathways of forest regrowth after clearcut and burning that diverge through time. ELM-FATES predicted higher peaks of initial forest responses (e.g., biomass, stem density) after clearcuts than after windthrows, similar to the changes in NIR. However, ELM-FATES predicted a faster recovery of forest structure and canopy-coverage back to pre-disturbance conditions for windthrows compared to clearcuts. The similarity of ELM-FATES predictions of regrowth patterns after windthrow and clearcut to those of the NIR results suggest that the dynamics of forest regrowth for these disturbances are represented with appropriate fidelity within ELM-FATES and useful as a benchmarking tool.

Tropical forests absorb large amounts of atmospheric CO2 through photosynthesis but elevated temperatures suppress this absorption and promote monoterpene emissions. Using 13 CO2 labeling, here we show that monoterpene emissions from tropical leaves derive from recent photosynthesis and demonstrate distinct temperature optima for five groups (Groups 1-5), potentially corresponding to different enzymatic temperature-dependent reaction mechanisms within β-ocimene synthases. As diurnal and seasonal leaf temperatures increased during the Amazonian 2015 El Nino event, leaf and landscape monoterpene emissions showed strong linear enrichments of β-ocimenes (+4.4% °C-1 ) at the expense of other monoterpene isomers. The observed inverse temperature response of α-pinene (-0.8% °C-1 ), typically assumed to be the dominant monoterpene with moderate reactivity, was not accurately simulated by current global emission models. Given that β-ocimenes are highly reactive with respect to both atmospheric and biological oxidants, the results suggest that highly reactive β-ocimenes may play important roles in the thermotolerance of photosynthesis by functioning as effective antioxidants within plants and as efficient atmospheric precursors of secondary organic aerosols. Thus, monoterpene composition may represent a new sensitive 'thermometer' of leaf oxidative stress and atmospheric reactivity, and therefore a new tool in future studies of warming impacts on tropical biosphere-atmosphere carbon-cycle feedbacks.

Current climate change scenarios indicate warmer temperatures and the potential for more extreme droughts in the tropics, such that a mechanistic understanding of the water cycle from individual trees to landscapes is needed to adequately predict future changes in forest structure and function. In this study, we contrasted physiological responses of tropical trees during a normal dry season with the extreme dry season due to the 2015–2016 El Niño-Southern Oscillation (ENSO) event. We quantified high resolution temporal dynamics of sap velocity (Vs), stomatal conductance (gs) and leaf water potential (ΨL) of multiple canopy trees, and their correlations with leaf temperature (Tleaf) and environmental conditions [direct solar radiation, air temperature (Tair) and vapor pressure deficit (VPD)]. The experiment leveraged canopy access towers to measure adjacent trees at the ZF2 and Tapajós tropical forest research (near the cities of Manaus and Santarém). The temporal difference between the peak of gs (late morning) and the peak of VPD (early afternoon) is one of the major regulators of sap velocity hysteresis patterns. Sap velocity displayed species-specific diurnal hysteresis patterns reflected by changes in Tleaf. In the morning, Tleaf and sap velocity displayed a sigmoidal relationship. In the afternoon, stomatal conductance declined as Tleaf approached a daily peak, allowing ΨL to begin recovery, while sap velocity declined with an exponential relationship with Tleaf. In Manaus, hysteresis indices of the variables Tleaf-Tair and ΨL-Tleaf were calculated for different species and a significant difference (p < 0.01, α = 0.05) was observed when the 2015 dry season (ENSO period) was compared with the 2017 dry season (“control scenario”). In some days during the 2015 ENSO event, Tleaf approached 40°C for all studied species and the differences between Tleaf and Tair reached as high at 8°C (average difference: 1.65 ± 1.07°C). Generally, Tleaf was higher than Tair during the middle morning to early afternoon, and lower than Tair during the early morning, late afternoon and night. Uncertainties surrounding vegetation response to increased disturbance rates associated with climate change remains a major global change issue for Amazon forests. Additionally, turnover rates computed as the average of mortality and recruitment rates in the Western Amazon basin are doubled when compared to the Central Amazon, and notable gradients currently exist in specific wood density and aboveground biomass (AGB) between these two regions. This study investigates the extent to which the variation in disturbance regimes contributes to these regional gradients. To address these issues, we evaluated disturbance-recovery processes under two scenarios of increased disturbance rates in a complex Central Amazon forest using first ZELIG-TROP, a dynamic vegetation gap model which we calibrated using long-term inventory data, and second using the Community Land Model (CLM), a global land surface model that is part of the Community Earth System Model (CESM). Upon doubling the mortality rate in the Central Amazon to mirror the natural disturbance regime in the Western Amazon of ∼2% mortality, at steady-state, AGB significantly decreased by 41.9% and there was no significant difference between the modeled AGB of 104 Mg C ha−1 and empirical AGB from the western Amazon datasets of 107 Mg C ha−1. We confirm that increases in natural disturbance rates in the Central Amazon will result in terrestrial carbon loss associated with higher turnover. However, different processes were responsible for the reductions in AGB between the models and empirical datasets. We observed that with increased turnover, the subsequent decrease in wood density drives the reduction in AGB in empirical datasets. However, decrease in stand basal area was the driver of the drop in AGB in ZELIG-TROP, and decreased leaf area index (LAI) was the driver in CLM. Further comparisons found that stem density, specific wood density, and basal area growth rates differed between the two Amazonian regions. This suggests that: (1) the variability between regions cannot be entirely explained by the variability in disturbance regime, but rather potentially sensitive to intrinsic environmental factors; or (2) the models are not accurately simulating all forest characteristics in response to increased disturbances. Last, to help quantify the impacts of increased disturbances on climate and the earth system, we evaluated the fidelity of tree mortality and disturbance in a global land surface model: CLM. For a 100% increase in annual mortality rate, both ZELIG-TROP and CLM were in close agreement with each other and predicted a net carbon loss of 41.9 and 49.9%, respectively, with an insignificant effect on aboveground net primary productivity (ANPP). Likewise, a 20% increase in mortality every 50 years (i.e. periodic disturbance treatment) resulted in a reciprocal biomass loss of 18.3 and 18.7% in ZELIG-TROP and CLM, respectively.

We describe an implementation of the Ecosystem Demography (ED) concept in the Community Land Model. The structure of CLM(ED) and the physiological and structural modifications applied to the CLM are presented. A major motivation of this development is to allow the prediction of biome boundaries directly from plant physiological traits via their competitive interactions. Here we investigate the performance of the model for an example biome boundary in eastern North America. We explore the sensitivity of the predicted biome boundaries and ecosystem properties to the variation of leaf properties using the parameter space defined by the GLOPNET global leaf trait database. Furthermore, we investigate the impact of four sequential alterations to the structural assumptions in the model governing the relative carbon economy of deciduous and evergreen plants. The default assumption is that the costs and benefits of deciduous vs. evergreen leaf strategies, in terms of carbon assimilation and expenditure, can reproduce the geographical structure of biome boundaries and ecosystem functioning. We find some support for this assumption, but only under particular combinations of model traits and structural assumptions. Many questions remain regarding the preferred methods for deployment of plant trait information in land surface models. In some cases, plant traits might best be closely linked to each other, but we also find support for direct linkages to environmental conditions. We advocate intensified study of the costs and benefits of plant life history strategies in different environments and the increased use of parametric and structural ensembles in the development and analysis of complex vegetation models.

Here, we review the state of knowledge regarding MTF tree mortality, create a conceptual framework with testable hypotheses regarding the drivers, mechanisms and interactions that may underlie increasing MTF mortality rates, and identify the next steps for improved understanding and reduced prediction. Increasing mortality rates are associated with rising temperature and vapor pressure deficit, liana abundance, drought, wind events, fire and, possibly, CO2 fertilization-induced increases in stand thinning or acceleration of trees reaching larger, more vulnerable heights. The majority of these mortality drivers may kill trees in part through carbon starvation and hydraulic failure. The relative importance of each driver is unknown. High species diversity may buffer MTFs against large-scale mortality events, but recent and expected trends in mortality drivers give reason for concern regarding increasing mortality within MTFs. Models of tropical tree mortality are advancing the representation of hydraulics, carbon and demography, but require more empirical knowledge regarding the most common drivers and their subsequent mechanisms. We outline critical datasets and model developments required to test hypotheses regarding the underlying causes of increasing MTF mortality rates, and improve prediction of future mortality under climate change.

Strong winds associated with severe convective storms are dominant natural drivers of tree mortality in the Amazon. Why forests vary with respect to their vulnerability to wind events and how the predicted increase in storm events might affect forest ecosystems within the Amazon are not well understood. We found that windthrows are common in the Amazon region extending from northwest (Peru, Colombia, Venezuela, and west Brazil) to central Brazil, with the highest occurrence of windthrows in the northwest Amazon. More frequent winds, produced by more frequent severe convective systems, in combination with well-known processes that limit the anchoring of trees in the soil, help to explain the higher vulnerability of the northwest Amazon forests to winds. Projected increases in the frequency and intensity of convective storms in the Amazon have the potential to increase wind-related tree mortality. A forest demographic model calibrated for the northwestern and the central Amazon showed that northwestern forests are more resilient to increased wind-related tree mortality than forests in the central Amazon. Our study emphasizes the importance of including wind-related tree mortality in model simulations for reliable predictions of the future of tropical forests and their effects on the Earth' system.

Disturbance-induced tree mortality is a key factor regulating the carbon balance of a forest, but tree mortality and its subsequent effects are poorly represented processes in terrestrial ecosystem models. It is thus unclear whether models can robustly simulate moderate (non-catastrophic) disturbances, which tend to increase biological and structural complexity and are increasingly common in aging US forests. We tested whether three forest ecosystem models – Biome-BGC (BioGeochemical Cycles), a classic big-leaf model, and the ZELIG and ED (Ecosystem Demography) gap-oriented models – could reproduce the resilience to moderate disturbance observed in an experimentally manipulated forest (the Forest Accelerated Succession Experiment in northern Michigan, USA, in which 38% of canopy dominants were stem girdled and compared to control plots). Each model was parameterized, spun up, and disturbed following similar protocols and run for 5 years post-disturbance. The models replicated observed declines in aboveground biomass well. Biome-BGC captured the timing and rebound of observed leaf area index (LAI), while ZELIG and ED correctly estimated the magnitude of LAI decline. None of the models fully captured the observed post-disturbance C fluxes, in particular gross primary production or net primary production (NPP). Biome-BGC NPP was correctly resilient but for the wrong reasons, and could not match the absolute observational values. ZELIG and ED, in contrast, exhibited large, unobserved drops in NPP and net ecosystem production. The biological mechanisms proposed to explain the observed rapid resilience of the C cycle are typically not incorporated by these or other models. It is thus an open question whether most ecosystem models will simulate correctly the gradual and less extensive tree mortality characteristic of moderate disturbances.

The volatile gas isoprene is emitted in teragrams per annum quantities from the terrestrial biosphere and exerts a large effect on atmospheric chemistry. Isoprene is made primarily from recently fixed photosynthate; however, alternate carbon sources play an important role, particularly when photosynthate is limiting. We examined the relative contribution of these alternate carbon sources under changes in light and temperature, the two environmental conditions that have the strongest influence over isoprene emission. Using a novel real-time analytical approach that allowed us to examine dynamic changes in carbon sources, we observed that relative contributions do not change as a function of light intensity. We found that the classical uncoupling of isoprene emission from net photosynthesis at elevated leaf temperatures is associated with an increased contribution of alternate carbon. We also observed a rapid compensatory response where alternate carbon sources compensated for transient decreases in recently fixed carbon during thermal ramping, thereby maintaining overall increases in isoprene production rates at high temperatures. Photorespiration is known to contribute to the decline in net photosynthesis at high leaf temperatures. A reduction in the temperature at which the contribution of alternate carbon sources increased was observed under photorespiratory conditions, while photosynthetic conditions increased this temperature. Feeding [2-13C]glycine (a photorespiratory intermediate) stimulated emissions of [13C1–5]isoprene and 13CO2, supporting the possibility that photorespiration can provide an alternate source of carbon for isoprene synthesis. Our observations have important implications for establishing improved mechanistic predictions of isoprene emissions and primary carbon metabolism, particularly under the predicted increases in future global temperatures. © 2014 American Society of Plant Biologists. Treatment for hemorrhagic shock secondary to a spontaneous hemoperitoneum includes restoration of IV volume and surgical control of hemorrhage. This study was designed to determine if limited fluid volume resuscitation (LFVR) with hypertonic saline (HS) and hyperoncotic fluids (hydroxyethylstarch [HES]) results in more rapid cardiovascular stabilization in dogs with spontaneous hemoperitoneum versus conventional resuscitation (CR) with large volume resuscitation. Eighteen client-owned dogs presenting in hemorrhagic shock with a spontaneous hemoperitoneum were enrolled. Dogs were randomized to be fluid resuscitated with up to 90 mL/kg of an isotonic crystalloid (CR group) or up to 8 mL/kg of 7.2% Na chloride (i.e., HS) combined with up to 10 mL/kg of 6% HES. Measurements of vital signs, lactate, packed cell volume (PCV), total solids (TS), and blood pressure were made at standard time points. The primary end point was time to stabilization of hemodynamic parameters (measured in min). Dogs in the LFVR group achieved hemodynamic stabilization significantly faster (20 min; range, 10–25 min) than those in the CR group (35 min; range, 15–50 min; P = .027). Future studies are warranted to further investigate potential benefits associated with LFVR in dogs with spontaneous hemoperitoneum.

In addition to natural storm incongruity, climate change can alter storm formation, duration, frequency, and intensity. This model-based investigation assessed the impacts of multiple storms of different intensities and occurrence frequencies on the long-term dynamics of subtropical dry forests in Puerto Rico. Using the previously validated individual-based gap model ZELIG-TROP, we developed a new hurricane damage routine and parameterized it with site- and species-specific hurricane effects. A baseline case with the reconstructed historical hurricane regime represented the control condition. Ten treatment cases, reflecting plausible shifts in hurricane regimes, manipulated both hurricane return time (i.e. frequency) and hurricane intensity. The treatment-related change in carbon storage and fluxes were reported as changes in aboveground forest biomass (AGB), net primary productivity (NPP), and in the aboveground carbon partitioning components, or annual carbon accumulation (ACA). Increasing the frequency of hurricanes decreased aboveground biomass by between 5% and 39%, and increased NPP between 32% and 50%. Decadal-scale biomass fluctuations were damped relative to the control. In contrast, increasing hurricane intensity did not create a large shift in the long-term average forest structure, NPP, or ACA from that of historical hurricane regimes, but produced large fluctuations in biomass. Decreasing both the hurricane intensity and frequency by 50% produced the highest values of biomass and NPP. For the control scenario and with increased hurricane intensity, ACA was negative, which indicated that the aboveground forest components acted as a carbon source. However, with an increase in the frequency of storms or decreased storms, the total ACA was positive due to shifts in leaf production, annual litterfall, and coarse woody debris inputs, indicating a carbon sink into the forest over the long-term. The carbon loss from each hurricane event, in all scenarios, always recovered over sufficient time. Our results suggest that subtropical dry forests will remain resilient to hurricane disturbance. However carbon stocks will decrease if future climates increase hurricane frequency by 50% or more.

OBJECTIVE: To characterize hemostatic profiles in dogs with acute pancreatitis. DESIGN: Prospective and observational study. SETTING: Tertiary referral centers. ANIMALS: Fifteen client‐owned dogs with acute pancreatitis enrolled between December 1, 2011 and June 1, 2012. MATERIALS AND METHODS: Blood samples were collected on admission for measurement of platelet count, PCV, thromboelastography (TEG), antithrombin, prothrombin time, activated partial thromboplastin time, D‐dimer, von Willebrand factor, and fibrinogen values, which were compared to reference intervals derived from healthy dogs. The Wilcoxon rank‐sum test was used to test for differences in continuous variables between study subjects and reference intervals. MEASUREMENTS AND MAIN RESULTS: Dogs with acute pancreatitis were globally hypercoagulable using TEG when compared with reference intervals. Stable pluripotent feeder-free propagation of human embryonic stem cells (hESCs) prior to their therapeutic applications remains a major challenge. Matrigel™ (BD Singapore) is a murine sarcoma-derived extracellular matrix (ECM) widely used as a cell-free support combined with conditioned or chemically defined media; however, inherent xenogenic and immunological threats invalidate it for clinical applications. Using human fibrogenic cells to generate ECM is promising but currently suffers from inefficient and time-consuming deposition in vitro. We recently showed that macromolecular crowding (MMC) accelerated ECM deposition substantially in vitro. In the current study, we used dextran sulfate 500 kDa as a macromolecular crowder to induce WI-38 fetal human lung fibroblasts at 0.5% serum condition to deposit human ECM in three days. After decellularization, the generated ECMs allowed stable propagation of H9 hESCs over 20 passages in chemically-defined medium (mTEsR1) with an overall improved outcome compared to Matrigel in terms of population doubling while retaining teratoma formation and differentiation capacity. Of significance, only ECMs generated by MMC allowed the successful propagation of hESCs. ECMs were highly complex and in contrast to Matrigel, contained no vitronectin but did contain collagen XII, ig-h3 and novel for hESC-supporting human matrices, substantial amounts of transglutaminase 2. Genome-wide analysis of promoter DNA methylation states revealed high overall similarity between human ECM- and Matrigel-cultured hESCs; however, distinct differences were observed with 49 genes associated with a variety of cellular functions. Thus, human ECMs deposited by MMC by selected fibroblast lines are a suitable human microenvironment for stable hESC propagation and clinically translational settings. Through modifications to the ZELIG simulation model, including the development of species- and site-specific parameters and internal modifications, the capability to model and predict forest change within the 4500-ha Guanica State Forest in Puerto Rico can now be accomplished. Published datasets and additional data from the U.S. Forest Service Forest Inventory Anal- ysis were used to parameterize the new gap model, ZELIG-TROP. We used data from permanent plots

Summary 1. We simulated two key components of severe hurricane disturbance, canopy openness and detritus deposition, to determine the independent and interactive effects of these components on woody plant recruitment and forest structure. 2. We increased canopy openness by trimming branches and added or subtracted canopy detritus in a factorial design. Plant responses were measured during the 4-year study, which followed at least 1 year of pre-manipulation monitoring. 3. The physical conditions of canopy openness and detritus deposition in our experiment resembled the responses to Hurricane Hugo, a severe category 4 hurricane that struck this forest in 1989. 4. Canopy detritus deposition killed existing woody seedlings and provided a mechanical barrier that suppressed seedling recruitment. The increase in understorey light caused by canopy trimming stimulated germination from the seed bank and increased seedling recruitment and density of pioneer species several hundred-fold when hurricane debris was absent. Many significant interactions between trimming and detritus deposition were evident from the manner in which seedling density, recruitment and mortality changed over time, and subsequently influenced the composition of woody stems (individuals ‡ 1 cm d.b.h.). 5. When the canopy was trimmed, stem densities increased > 2-fold and rates of recruitment into the stem size class increased > 25-fold. Trimming had no significant effect on stem mortality. The two dominant species that flourished following canopy trimming were the pioneer species Cecropia schreberiana and Psychotria berteriana. Deposition of canopy detritus had little effect on stems, although basal area increased slightly when detritus was added. There were no evident effects of the interactions between canopy trimming and detritus deposition on stems. 6. Synthesis. The separate and interactive effects of canopy openness and detritus deposition result in variable short-term trajectories of forest recovery. However, the short interval of increased canopy openness due to hurricane impacts and its influence on the recruitment of pioneer trees is the dominant factor that drives short-term recovery and may alter long-term structure and composition of the forest.

The dioecious, tropical palm Mauritia flexuosa has high ecological and economic value, but currendy some wild populations are harvested excessively, which is likely to increase. In this study, we investigated the population dynamics of this important palm, the effects of harvesting, and suggested sustainable harvesting regimes. Data were collected from populations in the Ecuadorian Amazon that were assumed to be stable. We used a matrix population model to calculate the density-independent asymptotic population growth rate (A, = 1.046) and to evaluate harvesting scenarios. Elasticity analysis showed that survival (particularly in the second and fifth size class) contributes more to the population growth rate, than growth and fecundity. To simulate a stable population at carrying capacity, density dependence was incorporated and applied to the seedling survival and growth parameters in the transition matrix. Harvesting scenarios were simulated with the density-dependent population models to predict sustainable harvesting regimes for the dioecious palm. We simulated the removal of only female palms and showed how both sexes are affected with harvest intensities between 15 and 75 percent and harvest intervals of 1-15 yr. By assuming a minimum female threshold, we demonstrated a continuum of sustainable harvesting schedules for various intensities and frequencies for 100 yr of harvest. Furthermore, by setting the population model's ? = 1.00, we found that a harvest of 22.5 percent on a 20 yr frequency for the M. flexuosa population in Ecuador is consistent with a sustainable, viable population over time.

Objective: To review the physiology of lactate production and metabolism, the causes of lactic acidosis, and the current applications of lactate monitoring in humans and animals. Data sources: Human and veterinary studies. Summary: Lactate production is the result of anaerobic metabolism. Tissue hypoxia due to hypoperfusion is the most common cause of lactic acidosis. Studies in critically ill humans have shown that serial lactate monitoring can be used to assess the severity of illness and response to therapy. Several veterinary studies have also shown lactate as a useful tool to assess severity of illness. Conclusions: Lactate measurement in critically ill veterinary patients is practical and can provide information to assess severity of illness. Further veterinary studies are needed to establish the value of serial lactate measurements for prognostic and therapeutic purposes. Information regarding lactate measurement in cats is limited, and further studies are warranted.

Objective: To describe the clinical manifestations and successful outcome following an inadvertent overdose of ketamine to a cat. Case summary: A 4-year-old neutered male domestic shorthair cat was evaluated for a urethral obstruction. Because of an inadvertent miscalculation of ketamine, 20 times the intended dose was administered intravenously, which resulted in cardiopulmonary arrest. Cardiopulmonary-cerebral resuscitation was successful, and short-term mechanical ventilation, fluids and intensive monitoring were utilized to achieve full recovery and subsequent discharge of the animal. New or unique information provided: Ketamine is a common anesthetic agent used in cats that is considered to have a wide therapeutic index and minimal cardiopulmonary depressant effects at recommended doses. Successful management of inadvertent ketamine overdose has been reported in children, but not in cats. Prompt CPCR and short-term mechanical ventilation may be necessary to treat a significant ketamine overdose. In cats, yohimbine may act as a partial antagonist of ketamine.
