IntroductionSystemic lupus erythematosus (SLE) is an autoimmune disease in which circulating immune complexes can cause different types of glomerulonephritis, according to immune deposits and to the type of glomerular cell injury. Proliferative lesions represent the most severe form of lupus nephritis (LN) and often lead to kidney failure and death. Mucosal-associated invariant T (MAIT) cells are a subset of innate-like T cells that recognize microbial-derived ligands from the riboflavin synthesis pathway. Although abundant in peripheral blood, MAIT cells are enriched in mucosal and inflamed tissues. While previous studies have reported concordant results concerning lower MAIT cell frequencies in the blood of SLE patients, no information is known about MAIT cell function and LN severity and outcome.MethodsIn the current study, we analyzed the baseline phenotype and function of peripheral blood MAIT cells by flow cytometry in 26 patients with LN and in a control group of 16 healthy individuals.ResultsWe observe that MAIT cell frequencies are markedly reduced in blood of LN patients. MAIT cells from patients have an altered phenotype in terms of migration, proliferation and differentiation markers, notably in most severe forms of LN. Frequencies of PMA/ionomycin stimulated MAIT cells secreting effector molecules, such as proinflammatory IL-17 and cytotoxic protein granzyme B, are higher in LN patients. Patients undergoing a complete renal remission after immunosuppressive therapy had higher MAIT cell frequency, lower expression of proliferation marker Ki-67 and granzyme B (GzB) at inclusion. Remarkably, GzB production defines a predictive model for complete remission.DiscussionWe report here that blood MAIT cells display proinflammatory and cytotoxic function in severe lupus nephritis which may play a pathogenesis role, but without association with systemic lupus activity. Finally, low cytotoxic profile of MAIT cells may represent a promising prognostic factor of lupus nephritis remission one year after induction therapy.

O presente artigo em desenvolvimento traz à apreciação o recorte de uma tese de doutoramento já defendida na área de Letras, com enfoque nos Estudos Literários. Trata-se da exposição de alguns dos principais achados acerca da imersão na plataforma virtual de autopublicação literária Wattpad, uma comunidade que abarca trabalhos autorais de caráter essencialmente eletrônico. Para tanto, elegemos os feitos de Anna Todd, wattpader estadunidense responsável por assinar os livros que integram a saga After, sucesso no ciberespaço e para além dos limites virtuais, uma vez ter passado também a protagonizar as páginas dos livros impressos, as graphic novels e as telas do cinema. Partindo dessa premissa, objetivou-se elucidar o contexto de manufatura de suas obras, assim como as especificidades que demarcam o seu processo criativo de elaboração textual. Diante disso, alguns importantes aportes de natureza teórica foram selecionados a fim de trilharmos o caminho aqui pretendido, tal como é o caso de Franco Moretti, Anne Jamison e Henry Jenkins, dentre outros nomes que contribuem aos diálogos entre Literatura e Cultura Digital. Em linhas gerais, verificamos que o distanciamento em relação ao objeto de análise selecionado se faz por vezes necessário à aproximação de uma compreensão significativa a respeito das práticas literárias eletrônicas em tempos contemporâneos e digitais.

Various host compounds have been investigated to produce an alternative attractant for monitoring and controlling pest insects. Several studies evidenced that volatile molecules released by coffee and mangoes attracted both male and female Ceratitis capitata (Wiedemann), a significant pest in fruit crops. In this study, we tested the effectiveness of the commercial glycolic extract of coffee (Coffea arabica L.) and a commercial mango essence (Mangifera indica L.) to attract C. capitata. We identified the main volatile compounds (VOCs) emitted from such products. The experiments were conducted in field cages and showed that the glycolic extract of coffee attracts both sexes of C. capitata, while the mango essence attracts only males. After seven days, the pH of the coffee extract and mango essence did not change. These results indicate that attractants based on commercial fruit extracts and essences can be an option for integrated pest management of the Mediterranean fruit fly in orchards.

The progressive lung destruction in cystic fibrosis (CF) is tightly associated with chronic bacterial infection and neutrophil-dominated airway inflammation. CF pulmonary disease is complicated by episodes of acute exacerbations, contributing to irreversible lung damage. We hypothesized that circulating subsets of neutrophils from clinically stable adults with CF present some phenotypic specificities that could amplify their activation during an infectious episode. The aim of the present study was to examine the different neutrophil subsets in whole blood and in the low density neutrophils (LDN) that co-purify with peripheral blood mononuclear cells (PBMC) in clinically stable adults with CF and in CF adults during pulmonary exacerbations compared to healthy donors. Blood samples were obtained from 22 adults with CF (16 in stable state and 6 during pulmonary exacerbations) and from 20 healthy donors. Flow cytometry analysis of 13 different markers related to lineage (CD45, CD15), maturity (CD16, CD10, and CD33), activation (CD62L, CD11b, CD66b, and CD114), metabolism (GLUT-1, LOX1) and immunosuppression (PD1, PD-L1) was carried out within whole blood and within the LDN fraction. Unsupervised analysis of flow cytometry data was performed using visual t-distributed stochastic neighbor embedding (vi-tSNE). A significant increase in the CD11b expression in neutrophils from CF patients during exacerbations was observed compared to neutrophils from stable CF patients or to healthy donors, indicative of a circulating activation state due to an infectious status. The percentage of LDN was not increased in stable CF patients but increased during exacerbations. Analysis of neutrophil subsets using the double CD16/CD62L labeling revealed a significant increase in the CD16high/CD62Llow subset in all CF patients compared to healthy donors. In contrast, an increase in the CD16low/CD62Lhigh subset was observed only in CF patients during exacerbations. Unsupervised analysis identified a PD-L1high/CD114high population that was present in stable CF patients and as well as in CF patients during exacerbations.

Os lançamentos literários impressos da autoria de influenciadores digitais adolescentes, entre blogueiros e youtubers, seguramente contribuiu ao aquecimento do nicho de consumo dedicado ao público jovem, em grande parcela órfão de publicações cativas às temáticas presentes em seus cotidianos, assinadas por pares – ídolos da Web. Um levantamento de obras impressas produzidas por administradores de blogs e canais do YouTube, entre janeiro de 2008 e dezembro de 2016, trouxera-nos à tona relevantes achados no que concerne à divergência entre dados catalográficos e os gêneros de fato contemplados nos livros analisados. Essa ocorrência nos conduz a refletir a respeito das dificuldades de classificação dos conteúdos abordados pelos jovens autores no mundo do papel, processo esse mediado pelo mercado editorial. Logo, pretendemos apresentar considerações gerais sobre tal problemática, lançando luzes e perspectivas à produção literária brasileira contemporânea cujo público-alvo é constituído por sujeitos ainda em processo de formação enquanto leitores, amantes dos seres e estares dessa Literatura. Utilizamos como base o supracitado mapeamento, ademais, aportes teóricos pertinentes ao campo temático, convictos de que as manifestações ciberespaciais têm transfigurado a arte literária, agora, mais do que nunca, plural e conectada às próprias possibilidades. Há muito se vêm desenvolvendo pesquisas na área da educação acerca da formação de professores e dos estágios curriculares, como uma das formas de garantir a melhoria dos cursos de licenciatura. Muitos desses estudos buscam compreender como ocorre o processo de identificação do docente em formação com o ofício de ensinar. Nessa perspectiva, este trabalho teve como objetivo analisar a relação entre os estágios supervisionados e o processo de construção da identidade docente do professor de língua portuguesa. Desenvolvemos uma pesquisa qualitativa e documental, na qual analisamos o discurso dos estagiários manifestado em seus relatórios escritos de vivências no campo do estágio. O corpus foi composto pelos trabalhos de 25 graduandos, concluintes do Estágio Curricular Supervisionado em Português 4, do curso de licenciatura em Letras-Português da Universidade Federal de Pernambuco, componente que consiste na regência de turma de ensino médio. Realizamos a análise dos dados com base em categorias teóricas atinentes à identidade e profissionalização docente (CORACINI, 2000; NAGAKOME, 2012; AGUIAR, 2016), à formação docente (SILVESTRE e PLACCO, 2011) e ao estágio (COSTA e LIMA, 2012; PIMENTA e LIMA, 2006). Constatamos que os estagiários reconhecem o estágio como importante etapa formativa do curso, espaço de reflexão acerca da prática pedagógica e processo de construção e reafirmação da identidade e da profissionalidade docente. Ademais, foram identificados diferentes sentimentos e impactos – tanto positivos quanto negativos – causados pela vivência do exercício profissional dos formandos, que também apontaram o que ainda pode ser feito para garantir uma formação mais consistente.

Inflammation, immune cells and the host microbiota are intimately linked in the pathophysiology of obesity and diabetes. Here the authors show mucosal-associated invariant T cells fuel inflammation in the tissues and serve a function in promoting metabolic breakdown, polarising macrophage populations and inducing dysbiosis of the intestinal microbiota.

O presente artigo versa sobre os impactos de um atraso na inclusão das questões que tangem a saúde mental infanto-juvenil à agenda de políticas públicas no Brasil. Nota-se que nas últimas décadas houve uma elevação da importância do conhecimento a respeito das Políticas Públicas no país, de seu funcionamento e suas finalidades. Em um primeiro momento, este artigo busca fazer um apanhado geral do que são essas ações de políticas públicas, principalmente no que tange as questões de saúde mental do público infantil e juvenil. Faz-se um levantamento acerca da Reforma Psiquiátrica Brasileira, como se deu e a forma como ainda vem se modificando, evoluindo e retrocedendo, com o passar do tempo, focando-se especificamente na Reforma Psiquiátrica no contexto da infância e adolescência, visto que essa teve um acréscimo muito tardio à agenda do SUS provocando consequências bem presentes nos dias atuais. A partir disto, apresenta-se dados que indicam a escassez de serviços especializados de cuidado à saúde mental para essa faixa etária, contando como estratégia de enfrentamento para essa defasagem a rede de apoio matricial feita pela Estratégia de Saúde da Família com o CAPSi, expondo seus impasses e peculiaridades.

Cystic fibrosis (CF) is a rare genetic multisystemic disease, the manifestations of which are due to mutations in the gene encoding the CF transmembrane conductance regulator (CFTR) protein and can lead to respiratory insufficiency and premature death. CFTR modulators, which were developed in the past decade, partially restore CFTR protein function. Their clinical efficacy has been demonstrated in phase 3 clinical trials, particularly in terms of lung function and pulmonary exacerbations, nutritional status, and quality of life in people with gating mutations (ivacaftor), homozygous for the F508del mutation (lumacaftor/ivacaftor and tezacaftor/ivacaftor), and in those with at least one F508del mutation (elexacaftor/tezacaftor/ivacaftor). However, many questions remain regarding their long-term safety and effectiveness, particularly in patients with advanced lung disease, liver disease, renal insufficiency, or problematic bacterial colonization. The impact of CFTR modulators on other important outcomes such as concurrent treatments, lung transplantation, chest imaging, or pregnancies also warrants further investigation. The French CF Reference Network includes 47 CF centers that contribute patient data to the comprehensive French CF Registry and have conducted nationwide real-world studies on CFTR modulators. This review seeks to summarize the results of these real-world studies and examine their findings against those of randomized control trials.

Natural killer (NK) cells are innate lymphocytes that play an important role in immunosurveillance, acting alongside other immune cells in the response against various types of malignant tumors and the prevention of metastasis. Since their discovery in the 1970s, they have been thoroughly studied for their capacity to kill neoplastic cells without the need for previous sensitization, executing rapid and robust cytotoxic activity, but also helper functions. In agreement with this, NK cells are being exploited in many ways to treat cancer. The broad arsenal of NK-based therapies includes adoptive transfer of in vitro expanded and activated cells, genetically engineered cells to contain chimeric antigen receptors (CAR-NKs), in vivo stimulation of NK cells (by cytokine therapy, checkpoint blockade therapies, etc.), and tumor-specific antibody-guided NK cells, among others. In this article, we review pivotal aspects of NK cells’ biology and their contribution to immune responses against tumors, as well as providing a wide perspective on the many antineoplastic strategies using NK cells. Finally, we also discuss those approaches that have the potential to control glioblastoma—a disease that, currently, causes inevitable death, usually in a short time after diagnosis.

O Bioma Caatinga está presente na região Nordeste brasileira, possuindo diversas espécies de plantas endêmicas como a Opuntia fícus-indical, Prosopis juliflora, Calotropis procera e Bromelia laciniosa, sendo a base alternativa da alimentação animal na Caatinga nos períodos de seca, onde a raça Boer vem assumindo importante papel no agronegócio na região Semiárida. Portanto, o objetivo da revisão foi analisar as espécies de plantas cultivadas na Caatinga para alimentação de caprino nos períodos de seca. Pelo fato da escassez da chuva nesse bioma se prolongar por vários meses durante o ano, houve a necessidade de adaptar as plantas que são ricas em proteínas brutas, altas concentrações de nutrientes e amido, que serve para fornecer alimentos dos caprinos, proporcionando aumento do peso e a produção de leite. Dessa forma, tem contribuído para adaptação da raça na Caatinga independente das condições ambientais e garante um excelente manejo alimentar alternativo em todas as épocas do ano.

Os campos cirúrgicos constituem uma das principais barreiras contra a incursão de microrganismos no sítio cirúrgico, além de proteção contra possíveis doenças aos profissionais de saúde. Dessa forma, o objetivo desse estudo é destacar as vantagens do campo cirúrgico descartável frente ao campo reprocessado, visto que em todos os procedimentos cirúrgicos utilizam-se esses materiais. Realizou-se o levantamento de dados em um hospital de pequeno porte da cidade de Ariquemes - RO, com intuito de conhecer o custo final do reprocessamento do campo de tecido, a fim de auxiliar na tomada de decisão, através da comparação do custo frente ao campo descartável. Diante de todo o levantamento, o valor final do pacote cirúrgico reprocessável do hospital avaliado é de R$40,08 (quarenta reais e oito centavos) e o custo real do descartável é de R$135,24 (cento e trinta e cinco reais e vinte e quatro centavos). Ambos apresentaram vantagens e desvantagens: o reprocessado é de baixo custo, mas devido aos inúmeros processos de lavagem e esterilização, reduz a capacidade das barreiras microbianas, já o descartável reduz os desperdícios ocorridos nos procedimentos cirúrgicos, mas aumenta o resíduo infectante. O resultado da pesquisa subsidiará discussões sobre a substituição de campos de tecido reutilizáveis por campos descartáveis, favorecendo argumentações relativas às vantagens e desvantagens dessa possibilidade considerando os recursos humanos, materiais, estruturais e financeiros.

ABSTRACT Big defensins, ancestors of β-defensins, are composed of a β-defensin-like C-terminal domain and a globular hydrophobic ancestral N-terminal domain. This unique structure is found in a limited number of phylogenetically distant species, including mollusks, ancestral chelicerates, and early-branching cephalochordates, mostly living in marine environments. One puzzling evolutionary issue concerns the advantage for these species of having maintained a hydrophobic domain lost during evolution toward β-defensins. Using native ligation chemistry, we produced the oyster Crassostrea gigas BigDef1 (Cg-BigDef1) and its separate domains. Cg-BigDef1 showed salt-stable and broad-range bactericidal activity, including against multidrug-resistant human clinical isolates of Staphylococcus aureus. We found that the ancestral N-terminal domain confers salt-stable antimicrobial activity to the β-defensin-like domain, which is otherwise inactive. Moreover, upon contact with bacteria, the N-terminal domain drives Cg-BigDef1 assembly into nanonets that entrap and kill bacteria. We speculate that the hydrophobic N-terminal domain of big defensins has been retained in marine phyla to confer salt-stable interactions with bacterial membranes in environments where electrostatic interactions are impaired. Those remarkable properties open the way to future drug developments when physiological salt concentrations inhibit the antimicrobial activity of vertebrate β-defensins. IMPORTANCE β-Defensins are host defense peptides controlling infections in species ranging from humans to invertebrates. However, the antimicrobial activity of most human β-defensins is impaired at physiological salt concentrations. We explored the properties of big defensins, the β-defensin ancestors, which have been conserved in a number of marine organisms, mainly mollusks. By focusing on a big defensin from oyster (Cg-BigDef1), we showed that the N-terminal domain lost during evolution toward β-defensins confers bactericidal activity to Cg-BigDef1, even at high salt concentrations. Cg-BigDef1 killed multidrug-resistant human clinical isolates of Staphylococcus aureus. Moreover, the ancestral N-terminal domain drove the assembly of the big defensin into nanonets in which bacteria are entrapped and killed. This discovery may explain why the ancestral N-terminal domain has been maintained in diverse marine phyla and creates a new path of discovery to design β-defensin derivatives active at physiological and high salt concentrations.

Elexacaftor-tezacaftor-ivacaftor induces rapid clinical improvement in patients with cystic fibrosis (CF) and advanced pulmonary disease, often leading to suspend the indication for lung transplantation. Yet no long-term data is available in lung transplant candidates.Lung transplant candidates (defined as being waitlisted for lung transplantation or considered for listing within 3 months) who have initiated elexacaftor-tezacaftor-ivacaftor were identified in the French cohort of patients with CF and advanced pulmonary disease. Patients were prospectively followed to evaluate treatment safety and effectiveness from initiation to July 20th, 2021.Among the 331 patients with advanced CF pulmonary disease who initiated elexacaftor-tezacaftor-ivacaftor, 65 were lung transplant candidates (17 listed for transplantation, 48 considered for listing within 3 months). Median [IQR] follow-up time was 363 [329; 377] days. At the end of the follow-up period, two patients were transplanted five and 11 days following treatment initiation, two were listed for transplantation, and 61 no longer met transplantation criteria. Improvement in percent predicted forced expiratory volume in 1 s (ppFEVIn lung transplant candidates eligible for elexacaftor-tezacaftor-ivacaftor, the rapid improvement following initiation of treatment persisted over one year with a reduction in treatment burden and lung transplantation could be safely deferred in most patients.

Childhood IgA nephropathy (IgAN) includes a wide spectrum of clinical presentations, from isolated hematuria to acute nephritis with rapid loss of kidney function. However, implication of such factors, notably soluble CD89, in childhood IgAN pathogenesis remains unclear. Here, we studied these biomarkers in a cohort of 67 patients with childhood IgAN and 42 pediatric controls. While Gd-IgA1 was only moderately increased in patient plasma, levels of circulating IgA complexes (soluble CD89-IgA and IgG-IgA) and free soluble CD89 were markedly increased in childhood IgAN. Soluble CD89-IgA1 complexes and free soluble CD89 correlated with proteinuria, as well as histological markers of disease activity: mesangial, endocapillary hypercellularity and cellular crescents. Soluble CD89 was found in patient's urine but not in urine from pediatric controls. Mesangial deposits of soluble CD89 were detected in biopsies from patients with childhood IgAN. Serum chromatographic fractions containing covalently linked soluble CD89-IgA1 complexes or free soluble CD89 from patients induced mesangial cell proliferation in vitro in a soluble CD89-dependent manner. Recombinant soluble CD89 induced mesangial cell proliferation in vitro which was inhibited by free soluble recombinant CD71 (also a mesangial IgA receptor) or mTOR blockers. Interestingly, injection of recombinant soluble CD89 induced marked glomerular proliferation and proteinuria in mice expressing human IgA1. Thus, free and IgA1-complexed soluble CD89 are key players in mesangial proliferation. Hence, our findings suggest that soluble CD89 plays an essential role in childhood IgAN pathogenesis making it a potential biomarker and therapeutic target.

As fronteiras brasileiras vêm sendo observadas com mais atenção por pesquisadores das ciências sociais e, especialmente, pelas políticas de segurança pública brasileiras. Esse interesse é recente e inédito, ainda sabe-se pouco sobre os cerca de dezessete mil quilômetros que separam e aproximam o Brasil dos seus países vizinhos. Entretanto, muitos conceitos vêm sendo discutidos no intuito de tornar a temática das fronteiras menos “romântica” e mais acadêmica, ao menos em tese. Como por exemplo o de Cidades-gêmeas, de Zona de Fronteira etc. Nesse contexto, a região sul do país, a que faz fronteira com a República Oriental do Uruguai e com a Argentina, torna-se um espaço frutífero para análises empíricas, tendo em vista que é a que possui mais municípios conurbados, cento e noventa e sete só no estado do Rio Grande do Sul. Dessa forma, o presente estudo visa compreender um pouco das dinâmicas urbanas dessas cidades-gêmeas, analisando alguns processos de socialização como é o caso dos Palestinos, especialmente na fronteira de Sant’Ana do Livramento (BR) e Rivera (ROU), utilizando-se dos conceitos de Pórtico e de Cidade para pensar esses espaços que não são nem brasileiros nem uruguaios.

In this study, we investigated the influence of soil management systems on the C retention capacity of soil with an emphasis on the silt and clay fractions of two subtropical soils with different mineralogy and climate. A percepção ambiental dos usuários quanto a importância da existência e manutenção dos parques ambientais municipais ainda é insuficiente. O objetivo dessa pesquisa foi identificação do perfil dos frequentadores do Parque Ambiental Municipal Adhemar Monteiro, em Paragominas (PA). O método utilizado foi o quantitativo com investigação básica. A obtenção de dados primários ocorreu com a aplicação de 338 formulários semiestruturados com sete questões objetivas e três subjetivas, durante 30 dias do mês de janeiro de 2019. Quanto aos secundários, eles foram obtidos em bases armazenadoras de informações científicas como a Coordenação de Aperfeiçoamento de Pessoal do Ensino Superior, Scientific Eletronic Library Online, Google Scholar, WebScience e periódicos especializados. A análise dos dados obtidos indicou que os usuários do parque são adultos (144 = 42,6%), a maioria é residente em Paragominas a quase 20 anos (112 = 33,1%) com isso, evidenciaram o surgimento e a evolução dessa Unidade de Conservação; atualmente, a manutenção do mesmo é considerada boa (165 = 48,8%). Por isso, utilizam esse espaço verde para lazer (208 = 61,5%). Todavia, foram efetuadas sugestões de melhorias como incrementar a diversidade de animais (133 = 39,3%), e segurança (9 = 2,7%). Logo, a percepção ambiental sobre a importância do parque à comunidade está atrelada a melhorias que podem ser efetuadas nesse local.

O acumulo de residuos solidos, especialmente aqueles oriundos dos centros urbanos torna-se um dos grandes problemas de poluicao do meio ambiente na atualidade, e que e sustentado na triade consumo, recurso e residuo. o objetivo desta pesquisa e analisar quantiqualitativamente a relacao entre o crescimento populacional, o de residuos solidos e a disposicao final desses residuos em um recorte temporal compreendido entre 2008 e 2017, na regiao norte. O metodo aplicado foi o dedutivo. A pesquisa tambem foi exploratoria. Em relacao a abordagem, esta pesquisa teve caracteristica quantiqualitativa. A coleta dos dados secundarios foi efetuada a partir de acesso a plataforma de dados livres do Panorama dos Residuos Solidos no Brasil publicado pela Associacao Brasileira de Empresas de Limpeza Publica e Residuos Especiais – ABRELPE. A geracao de residuos solidos de 2008 a 2017 aumentou 75,28% e o crescimento populacional foi 8,51%. Entre os anos de 2008 e 2012 a producao de residuos solidos apresentou maiores percentuais (54,21%), enquanto que o crescimento populacional foi de 6,14%. Em relacao a disposicao final dos residuos solidos houve um aumento d 2008 a 2017 de 5,4% dos residuos encaminhados a aterros sanitarios e 1,4% dos encaminhados a lixoes. Houve ainda, uma diminuicao dos residuos dispostos em aterros controlados de 6,8% no mesmo periodo. Dessa forma e confirmada a forte correlacao entre o crescimento populacional e o de residuos solidos.
