Objective: A set of microsatellite markers with high polymorphism from Tsaiya duck were used for the genetic monitoring and genetic structure analysis of Brown and White Tsaiya duck populations in Taiwan.Methods: The synthetic short tandem repeated probes were used to isolate new microsatellite markers from the genomic DNA of Tsaiya ducks. Eight populations, a total of 566 samples, sourced from Ilan Branch, Livestock Research Institute were genotyped through novel and known markers. The population genetic variables were calculated using optional programs in order to describe and monitor the genetic variability and the genetic structures of these Tsaiya duck populations.Results: In total 24 primer pairs, including 17 novel microsatellite loci from this study and seven previously known loci, were constructed for the detection of genetic variations in duck populations. The average values for the allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.29, 5.370, 0.591, 0.746, and 0.708, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting Brown Tsaiya duck cluster and a spreading White Tsaiya duck cluster. The Brown Tsaiya ducks and the White Tsaiya ducks with Pekin ducks were just split to six clusters and three clusters when K was set equal to 6 and 3 in the Bayesian cluster analysis. The individual phylogenetic tree revealed eight taxa, and each individual was assigned to its own population.Conclusion: According to our study, the 24 novel microsatellite markers exhibited a high capacity to analyze relationships of inter- and intra-population in those populations with a relatively limited degree of genetic diversity. We suggest that duck farms in Taiwan could use the new (novel) microsatellite set to monitor the genetic characteristics and structures of their Tsaiya duck populations at various intervals in order to ensure quality breeding and conservation strategies.

International audience; This study aimed to isolate, culture, and characterize duck primordial germ cells (PGCs) and to compare these cells with chicken PGCs. We first cultured Muscovy duck (Cairina moschata) circulating PGCs and gonadal PGCs (gPGCs) in the modified serum-containing medium used to amplify chicken PGCs. gPGCs were found to proliferate better in serum-free chemically defined medium than in serum-containing medium. Thereafter, gPGCs were similarly isolated from 2 other duck breeds, the Pekin duck (Anas platyrhynchos) and the hybrid mule duck (C. moschata x A. platyrhynchos), and amplified for a limited period of time in the chemically defined culture condition, but sufficiently to be characterized and transplanted. Cultured gPGCs of all 3 duck breeds were characterized by Periodic acid-Schiff staining, immunocytochemical staining, and expression analysis of germline-specific and pluripotency genes. Cultured duck gPGCs colonized the gonads after being genetically labeled and injected into recipient embryos. Taken together, these results demonstrate that duck PGCs retain their germline characteristics after being isolated, expanded in vitro, and genetically modified. Further studies are required to establish the optimal conditions for long-term culture of duck PGCs, which may involve supplementing the culture medium with other growth factors or compounds.

The objective of this study is trying to solve water pollution problems related to duck house wastewater by developing a novel duck house wastewater treatment technology. A pilot-scale sequencing batch reactor (SBR) system using different hydraulic retention times (HRTs) for treating duck house wastewater was developed and applied in this study. Experimental results showed that removal efficiency of chemical oxygen demand in untreated duck house wastewater was 98.4, 98.4, 87.8, and 72.5% for the different HRTs of 5, 3, 1, and 0.5 d, respectively. In addition, removal efficiency of biochemical oxygen demand in untreated duck house wastewater was 99.6, 99.3, 90.4, and 58.0%, respectively. The pilot-scale SBR system was effective and deemed capable to be applied to treat duck house wastewater. It is feasible to apply an automatic SBR system on site based on the previous case study of the farm-scale automatic SBR systems for piggery wastewater treatment.

A synthetic strain of ducks (Anas platyrhynchos) was developed by introducing genes for long duration of fertility to be used as mother of mule ducklings and a seven-generation selection experiment was conducted to increase the number of fertile eggs after a single artificial insemination (AI) with pooled Muscovy semen. Reciprocal crossbreeding between Brown Tsaiya LRI-2 (with long duration of fertility) and Pekin L-201 (with white plumage mule ducklings) ducks produced the G0. Then G1 were intercrossed to produce G2 and so on for the following generations. Each female duck was inseminated 3 times, at 26, 29, and 32 weeks of age. The eggs were collected for 14 days from day 2 after AI. Individual data regarding the number of incubated eggs (Ie), the number of fertile eggs at candling at day 7 of incubation (F), the total number of dead embryos (M), the maximum duration of fertility (Dm) and the number of hatched mule ducklings (H) with plumage colour were recorded. The selection criterion was the breeding values of the best linear unbiased prediction animal model for F. The results show high percentage of exhibited heterosis in G2 for traits to improve (19.1% for F and 12.9% for H); F with a value of 5.92 (vs 3.74 in the Pekin L-201) was improved in the G2. Heritabilities were found to be low for Ie (h (2) = 0.07±0.03) and M (h (2) = 0.07±0.01), moderately low for Dm (h (2) = 0.13±0.02), of medium values for H (h (2) = 0.20±0.03) and F (h (2) = 0.23±0.03). High and favourable genetic correlations existed between F and Dm (rg = 0.93), between F and H (rg = 0.97) and between Dm and H (rg = 0.90). The selection experiment showed a positive trend for phenotypic values of F (6.38 fertile eggs in G10 of synthetic strain vs 5.59 eggs in G4, and 3.74 eggs in Pekin L-201), with correlated response for increasing H (5.73 ducklings in G10 vs 4.86 in G4, and 3.09 ducklings in Pekin L-201) and maximum duration of the fertile period without increasing the embryo mortality rate. The average predicted genetic response for F was 40% of genetic standard deviation per generation of selection. The mule ducklings' feather colour also was improved. It was concluded that this study provided results for a better understanding of the genetics of the duration of fertility traits in the common female duck bred for mule and that the selection of a synthetic strain was effective method of improvement.

The long-term effects of Great Lakes' contaminants on male reproductive performance resulting from lactational and dietary exposure were investigated. Female C57BL/6 J mice (F-0) were mated with DBA/2 J male mice to produce B6D2-F1 offspring. Dams (F-0) were fed one of three treatment diets during lactation. The three treatment diets were: (i) diet C, containing lab chow and fish oil; (ii) diet I, containing 60% Iowa carp and 40% lab chow; and (iii) diet G, containing 60% Great Lakes' carp and 40% lab chow. Diet C served as a lab-chow control treatment, while diet I served as a fish-diet control treatment. Diet G contained 2500 µg of polychlorinated biphenyls (PCBs) and 300 µg of 1,1-dichloro-2,2-bis-[p-chlorophenyl]ethylene (4,4′-DDE) per kilogram of feed. The offspring (F-1) were continued on their respective maternal treatment diets from weaning until termination. Greater neonatal mortality and lesser body weights were observed in the F-1 male mice on diet G. Lesser fecundity was observed in one-year-old F-1 male mice on diet G following pairing with non-treated female mice. The sperm concentration of F-1 male mice on diet G was less than 30% of that of mice on diets C and I. The in vitro fertilizing ability of the sperm decreased to 10% of that of mice on the two control diets. Sperm motility, velocity, linearity and amplitude of lateral head (ALH) displacement were also decreased in 15-month-old F-1 male mice on diet G.

The objectives of this study were to understand the possible mechanism of duck sperm toxicity induced by arsenic exposure in vivo, and to investigate the roles of the antioxidant L-ascorbic acid in ameliorating the arsenic-induced sperm impairment. To test the acute toxicity, the percentages of mortality of mature drakes treated with different concentrations of trivalent sodium arsenite, As (III), and pentavalent sodium arsenate, As (V) were measured. The LD50 value of As (III) for mature drakes was 4.89±1.49 ppm. Although As (V) didn't cause any deaths even at a concentration of 40 ppm, the chronic toxicity of As (V) on sperm quality was shown by a decreased fertilization rate. When the concentrations of As (V) were above 0.4 ppm, fertilization rates were lower than those of 0.04 ppm and control. Drakes treated with 40 ppm of As (V) had the highest malondialdehyde (MDA) level in the testis tissue, 3.100±0.218 µmole/g testis. This showed that 40 ppm of As (V) significantly induced lipid peroxidation in testis tissue. For the 1.2 ppm As (III) treatment, several significant effects were observed: (1) sperm motility was decreased most dramatically by 52.0±9.1% after three days of incubation; (2) fertilization rate of artificially inseminated semen was the lowest, 26.4±15.4; (3) the MDA concentration in testis tissue, 7.846±0.246 µmole/g testis, was significantly higher than the others (p

Recent studies demonstrated that freshwater clam (Corbicula fluminea) has lipid-lowering and hepatoprotective activities, but its effect on immune responses has not yet been addressed. Here we showed that ethanol extracts of C. fluminea (ECF) reduced nitrite oxide, interleukin-1beta, interleukin-6, and tumor necrosis factor-alpha in lipopolysaccharide-activated macrophages. Further, ECF was fractionated into n-hexane, ethyl acetate, ethanol, and water soluble fractions. Of these, the ethyl acetate soluble fraction (EACF) had the highest capacity to inhibit pro-inflammatory mediators expression. The underlying mechanisms for the anti-inflammatory activity of EACF were demonstrated as down-regulation of ERK1/2, JNK1/2, and p38 phosphorylation and NF-kappaB activity. Using gas chromatography-mass spectrometric analysis EACF was found to be composed mainly of fatty acids and steroids. Our results provide evidence that freshwater clam has anti-inflammatory activity, and support the possibility for the development of freshwater clam as a health supplement or adjuvant therapeutic agent for either preventing or treating inflammation related diseases.

Composting is fast becoming the primary disposal option for leaves and grass in the U.S. due to proposed bans on the land filling and incineration of these wastes. To date, few systematic field scale studies have been done to compare the effects of turning regime, pile size and feed stock mix ratios on the rate of compost production or the properties of finished compost. In this study, leaf, grass and brush mixtures of 4:1:1, 4:2:1, 4:3:1, as well as 4:2:1 and 4:3:1 with staged grass addition, were formed into windrows and piles and composted for 136 days. Using a windrow turner, one series of windrows (#1) was turned seven times every four weeks, while the second series of windrows (#2) was turned once every four weeks. The piles were turned using a front-end loader once every four weeks. Results showed that windrow temperatures rose to 55–62° C in 6 days and remained at this temperature through day 60 and then slowly declined to around 45° C by day 100. Pile temperatures rose to higher levels (68–74° C) and remained above 60° C for 100 days. Windrow oxygen concentrations ranged between 1 and 4% while the oxygen concentrations in the piles fluctuated greatly between 2 and 18%. Organic matter loss was fastest in windrows series #1 and slowest in the piles. The pH of all the treatments rose from neutral to slightly alkaline (pH>8). The bulk density increased by more than 400% for windrows in series #1 but by only 150%, and 100%, respectively, for windrows in series #2 and the piles. The results indicate that turning frequency has a significant effect on bulk density, a slight effect on the rate of organic matter loss, and an insignificant effect on temperature and oxygen concentration. The mix ratio of leaves to grass, on the other hand, had no significant effect on most measured parameters. The composts from all the treatments were stable after 60 days of compo sting as indicated by oxygen uptake rates of less than 0.1 mg O2/g organic matter/hr. None of the composts inhibited seed germination or root elongation as measured by the Cress seed test.

Because of proposed bans on the landfilling and incineration of leaves, grass and brush, large-scale composting is fast becoming the primary disposal option for yard trimmings in many states. Few systematic studies have been done to compare the effects of turning regime, feedstock mix ratio, or windrow vs. pile configuration on composting and the characteristics of finished compost. In this study, various ratios of leaves, grass and brush were mixed and composted in two series of windrows; and one set of static piles. One windrow series (#1) was turned seven times every four weeks, while the other windrow series (#2), and the piles, were turned once every four weeks.
