ABSTRACT Pericytes are located around blood vessels, in close contact with endothelial cells. We discovered that pericytes dampen pro-inflammatory endothelial cell responses. Endothelial cells co-cultured with pericytes had markedly reduced expression of adhesion molecules (PECAM-1 and ICAM-1) and proinflammatory cytokines (CCL-2 and IL-6) in response to bacterial stimuli (Brucella ovis, Listeria monocytogenes, or Escherichia coli lipopolysaccharide). Pericyte-depleted mice intraperitoneally inoculated with either B. ovis, a stealthy pathogen that does not trigger detectable inflammation, or Listeria monocytogenes, developed peritonitis. Further, during Citrobacter rodentium infection, pericyte-depleted mice developed severe intestinal inflammation, which was not evident in control mice. The anti-inflammatory effect of pericytes required connexin 43, as either chemical inhibition or silencing of connexin 43 abrogated pericyte-mediated suppression of endothelial inflammatory responses. Our results define a mechanism by which pericytes modulate inflammation during infection, which shifts our understanding of pericyte biology: from a structural cell to a pro-active player in modulating inflammation.IMPORTANCEA previously unknown mechanism by which pericytes modulate inflammation was discovered. The absence of pericytes or blocking interaction between pericytes and endothelium through connexin 43 results in stronger inflammation, which shifts our understanding of pericyte biology, from a structural cell to a player in controlling inflammation.

Category: Midfoot/Forefoot; Diabetes; Other Introduction/Purpose: Non-traumatic lower extremity amputations (LEA), especially those performed in dysvascular and diabetic patients, are known to have poor long-term prognosis. Perioperative mortality has been reported at between 4 and 10%, and the 1 and 5 year mortality rates range between 22-33% and 39-69%, respectively. While poor outcomes in these patients have been described, there is no consensus as to the predictors of mortality. The purpose of the study is to determine the percentage of patients who had a complication following transmetatarsal amputation (TMA) and identify associated risk factors for complications and mortality. Methods: We queried our institution’s administrative database to identify 247 TMA procedures performed in 229 patients between January, 2002 and December, 2016. Electronic health records were reviewed to document complications defined as reoperation, amputation and mortality. Mortality was also verified using the National Death Index. Additionally, we recorded risk factors including diabetes, A1c level, end stage renal disease (ESRD), cardiovascular disease (CVD), peripheral vascular disease (PVD), history of revascularization, contralateral amputation, and neuropathy. The majority of the study patients were males (157, 69%) and the average age was 57 years (range 24-91). The median BMI was 28 (range 16-58) and 29% of the study patients were obese with a BMI ≥ 30. Fishers Exact tests were used to compare categorical variables. Kruskal-Wallis and Independent T-tests were used to compare numeric data. All data were analyzed using SAS/STAT software version 9.4 (Carey, NC) and a 0.05 level of significance was defined apriori. Results: The conversion rate to below (BKA) or above knee amputation (AKA) was 26% (64 of 247). Males (p=.0274), diabetics (p=.0139), patients in ESRD (p=.019), and patients with a history of CVD (p=.0247) or perioperative revascularization (p=.022) were more likely to undergo further amputation following an index TMA. BMI was significantly higher in patients requiring BKA/AKA (p=.0305). There were no significant differences in age (p=.2723) or A1c levels (p=.4219). The overall mortality rate was 35% (84 of 229). Gall-forming insects are sophisticated sedentary herbivores that present high level of specifcity with host plant, but their performance can be affected by biotic and abiotic factors. In this study we have tested two predictions: a) plants that have a greater number of conspecifc neighbors have greater richness and abundance of gall-forming insects, and b) interspecifc competition is a force capable of shaping the organization of gall-forming insect communities in super-host plants. We used the Copaifera oblongifolia (Fabaceae)/galling insects’ system to test these predictions. Fieldwork was carried out in areas of Cerrado (Brazilian Savanna) in northern Minas Gerais, Brazil. To test the frst hypothesis, we evaluated with generalized linear mixed models, the effects of the number of conspecifc neighbors on the richness and abundance of galls associated with 67 C. oblongifolia individual plants belonging to two populations. To test the second hypothesis, we used null models to evaluate whether a plant of C. oblongifolia colonized by a species of gall is preferred or avoided by another species of gall. A total of 2901 gall-forming insects belonging to 15 species were collected from the host plant C. oblongifolia. We observed negative relationships between the number of conspecifc neighbors and the abundance and richness of gall-forming insects associated C. oblongifolia. Thus, our data did not support the resource concentration hypothesis. Instead, we used the resource dilution theory to explain the negative relation between resource concentration and frequency of attack by galling insects. Our results also showed that the co-occurrence pattern of gall-forming insects in the host plant did not differ from those expected by chance. Therefore, the structure of the gall-forming insect community associated to single C. oblongifolia plants cannot be attributed to deterministic factors such as interspecifc competition. https://doi.org/10.25260/EA.19.29.1.0.751

Due to the existence of new methodologies that have reduced the production costs of microbiological chitosan, this paper puts forward the use of agro-industrial residues in order to produce microbiological chitosan and to apply chitosan as an innovative resource for removing acid orange 7 (AO7) from wastewaters. The best culture conditions were selected by a full 24 factorial design, and the removal of the dye was optimized by a 23 central composite rotational design. The results showed that corn steep liquor (CSL) is an agro-industrial residue that can be advantageously used to produce microbiological chitosan with yields up to 7.8 g/kg of substrate. FT-IR spectra of the product showed typical peak distributions like those of standard chitosan which confirmed the extracted product was chitosan-like. The efficiency of removing low concentrations of AO7 by using microbiological chitosan in distilled water (up to 89.96%) and tap water (up to 80.60%) was significantly higher than the efficiency of the control (chitosan obtained from crustaceans), suggesting that this biopolymer is a better economic alternative for discoloring wastewater where a low concentration of the dye is considered toxic. The high percentage recovery of AO7 from the microbiological chitosan particles used favors this biopolymer as a possible bleaching agent which may be reusable.

Este trabalho teve como objetivos caracterizar e comparar a estrutura e a dinâmica populacional de Microphrys bicornutus Latreille, 1825 no fital Halimeda opuntia (Halimedaceae) coletado nas formações recifais de Picãozinho (submetida à visitação turística) e São Gonçalo (área controle), na costa de João Pessoa (Nordeste do Brasil), sob influência de fatores ambientais e do número de visitantes. Nas duas áreas de estudo as populações analisadas estiveram compostas por fêmeas e machos maduros e imaturos com significativa predominância de machos e de animais imaturos, freqüência de tamanho e períodos reprodutivos similares. O tamanho máximo dos exemplares, a freqüência de distribuição de tamanho e a razão sexual diferiram dos resultados obtidos para a espécie em outras latitudes e habitats. Dados de razão sexual evidenciam que independentemente do estágio de maturação, os machos apresentam predominância significativa (RS>1,0), e que a proporção de fêmeas diminui com o amadurecimento sexual. Sem sofrer influência da biomassa da alga, e da salinidade e temperatura da água, variações populacionais significativas foram associadas ao aumento de juvenis durante períodos chuvosos. A baixa densidade populacional e a maior desproporção da relação macho: fêmea em subárea de Picãozinho com maior fluxo de pessoas sugerem que estas variações podem ter sido induzidas pelo pisoteio das algas.The aim of this study was to characterize and compare the population structure and dynamic of Microphrys bicornutus Latreille, 1825 on the phytal Halimeda opuntia (Halimedaceae) collected on the reef areas of Picãozinho (visited by tourists) and São Gonçalo (control area), in João Pessoa coast (Northeastern of Brazil), under the influence of environmental conditions and visitant numbers. In both study areas the populations analyzed were composed similarly of mature and immature males and females, with predominance of males and immature animals, and similar size frequency and reproductive period. Sex-ratio, size frequency distribution, and size of the biggest specimens differed from those estimated in others latitudes and habitats for this species. Sex ratio estimation has made evident that independently of maturation stage, males tended to predominate significantly and the proportion of females to decrease with sexual maturation. Without exhibiting influence of the alga biomass, and of the salinity and temperature of the water, significant population variations were associated to the increase of juvenile during rainy periods. The low population density and the largest disproportion of the relationship male: female in Picãozinho subarea with larger flow of people, are suggestive that the trampling of the alga might have induced this population variation.

Teaching biology in high school is challenging, with numerous topics to cover and potential pitfalls to avoid. Traditional teaching methods, like using textbook pictures, can be ineffective, especially when teaching complex concepts like evolution, genetics, and kingdoms of classification. To address this, educators are turning to simulations to improve the teaching and learning process. Simulations have been used in various fields for decades, allowing scientists to model and communicate complex ideas effectively. Not surprisingly, science students also benefit from using simulations to enhance their understanding and knowledge integration. A study in Bangkok, Thailand, investigated the effectiveness of using simulations to teach selected topics in biology to Grade 12 students. The research compared a traditional lecture-based approach with a simulation-based method using an interactive computer simulation called PLIX. Results showed that students in the simulation-based group had significantly higher posttest scores than those in the traditional teaching group. This suggests that incorporating simulations into biology lessons can enhance students' understanding of complex scientific concepts. Educators are encouraged to integrate simulation-based teaching methods into their curricula to improve student learning and engagement in science education. [ABSTRACT FROM AUTHOR]
