Brazil is the center of diversity and dispersion of species of the genus Passiflora. Two species of passion fruit Passiflora tenuifila BRS VT (passiflora garlic) and another of Passiflora setacea BRS PC were used in this study. The plants were grown on the Embrapa Cerrados experimental farm. The fruits were washed, minimally cut and their constituents separated. Both species differed in the proportion of wet mass between husk, seed, and pulp. As expected, both species had a higher proportion of husks and seeds and a lower amount of pulp. A semi-purification procedure for the proteins present in the seeds and husks was described and the molecular aspects were analyzed by polyacrylamide gel electrophoresis (SDS-PAGE). The quantification of soluble proteins was performed using the Bradford method. The analysis of the soluble protein extracted from the seeds and husks showed that these values are up to 70 times higher for the seeds of P. tenuifila and 28.5 times for P. setacea, in the husks. It was verified that both species present a similar protein profile, observed by the intense and diverse bands found in the polyacrylamide gel, mainly in the range of 32 to 19 kDa. This work opens up an unexplored field of tracking bioactive proteins and/or peptides including forms of nanostructure systems that protect other bioactive molecules. The objective of this work was to present an analytical procedure to semi-purify seed proteins of two Passiflora species and use electrophoresis as an analysis tool for further screening of their protein profiles and selection of a molecular marker to differentiate them. Coffee, besides being one of the most consumed stimulating beverages in the world, has important bioactive activities, which have been attracting increasing attention from researchers. However, the standard process of roasting causes changes in its chemical composition. In the present study, extracts obtained from green and roasted beans (light, medium and dark) of Coffea arabica Linnaeus were submitted to high-power ultrasonic extraction and atomization by spray drying. Colorimetric analysis was used to classify the roasting levels of the dried extract samples. The effects of the roasting process on the bioactivity of the dried extracts were verified through the following assays: caffeine, chlorogenic acid and caffeic acid, by HPLC-PDA; total phenolics by Folin–Ciocalteu; antioxidant activity by DPPH, FRAP, ABTS and ORAC; antiproliferative activity, using the MTT assay; and cell cycle and apoptosis by flow cytometry in metastatic prostate cancer cell lines from bone (PC-3) and brain (DU-145). The results showed that the lowest levels of caffeine, chlorogenic and caffeic acids were observed in dark roasted coffee. In comparison to medium and dark extracts in PC-3 cells, the green and light coffee extracts had higher antioxidant activities and promoted cytotoxicity followed by cell cycle arrest in phase S and apoptosis induction. Thus, the roasting level of the coffee extracts may be related to the potential chemoprotective effects of Coffea arabica L. in prostate cancer cells.

Coffee roasting needs precise control and innovative techniques that are economically viable to monitor and improve its consistency. In this study, mass spectrometry was used as a tool to screen chemical markers that appear on the surface of coffee beans (whole bean) along the roasting process. A non-target and non-volatile approach was used with an ambient technique (EASI) coupled to a single quadrupole mass analyzer to monitor roasting chemical changes in the coffee bean. Green (raw), soft, medium, dark and very dark roasted coffee beans showed a decrease in ions in the range of m/z 500–600, whereas an increase in abundance in the m/z 800–900 range was clearly observed in the most roasted coffees. A multivariate approach through PCA separated the different roasts in 70% of the variance using PC1 and PC2. The major ions in the range of m/z 500–600 were characterized by ESI-MS and also HPLC-fluorescence as the N-alkanoyltryptamides, surface constituents of coffee wax layer which are almost fully degraded in darker roasts. The ions in the range of m/z 800–900 were characterized as di-and triacylglicerols and its increase during the roasting process was systematically observed. For these classes of chemical markers of the roasting process, ESI-MS showed also the sodium and potassium adducts with good relative abundances.

Coffee is a popular drink consumed all over the world. Besides its long-recognized stimulant effect, it has important nutritional and health effects. However, the type of bean processing modifies the composition of brewed coffee and possibly its bioactivity. In this study, extracts obtained from green and roasted beans of Coffea canephora (Coffea canephora var. robusta) were submitted to spray- or freeze-drying and were tested for antiproliferative activity, using MTT assay, and their influence on the cell cycle and apoptosis by flow cytometry analysis. Moreover, colors and nutrient contents were measured to identify the changes due to the roasting process. The results obtained showed that extracts from green and light roasted beans exhibited strong bioactive capacity. Coffee extracts promoted a decrease in cell viability, modulated cell cycle and induced apoptosis in human prostate carcinoma cell line (DU-145). The level of roasting reduced this property, but the type of drying did not in all cases.

The presence of acrylamide in foods such as coffee can pose risks to human health. Coffee consumption and production in Latin America have increased in recent years. According to the FAO, world coffee consumption is divided into five groups of countries according to average yearly per capita consumption: (i) between 8.0 kg and 12 kg; (ii) between 4.1 kg and 8.0 kg; (iii) between 2.4 kg and 4.5 kg; (iv) between 0.8 kg and 2.4 kg; and (v) less than 0.8 kg. In this work, the countries of Latin America are divided into three groups according to annual per capita coffee consumption: group 1: between 4.1 kg and 8.0 kg (Brazil, Argentina, Colombia and Costa Rica); group 2: between 2.4 kg and 4.5 kg (Guyana, French Guyana, Honduras, Suriname, Uruguay and Venezuela); and group 3: between 0.8 kg and 2.4 kg (Bolivia, Chile, Cuba, El Salvador, Ecuador, Guatemala, Haiti, Jamaica, Mexico, Nicaragua, Panama, Paraguay, Peru, Puerto Rico and Dominican Republic). There are great differences in the consumption of acrylamide-forming foods among populations from different countries. The literature shows a range from 0.5 to 4.21 µg of acrylamide per 300 mL of coffee and the World Health Organization recommends a maximum daily limit for acrylamide intake of 1 µg/kg bw. Thus, coffee is a beverage that contributes greatly to acrylamide intake in almost all population groups. From the calculated data on acrylamide intake in Latin America, it was possible to estimate the daily intake in the region’s countries. According to the maximum limit set by the WHO of 1 µg acrylamide per day per bw, for people in the countries of group 1, coffee makes a large contribution to the intake of acrylamide. Thus, there should be focus on reducing the levels of acrylamide in foods, since they are the predominant sources of this substance for some populations.

Coffee plays an important economic and social role, worldwide. The global acceptance of coffee as a drink and commodity can be demonstrated not only in the number of consumers but even in water quality studies, where caffeine has been considered an anthropogenic chemical marker and its concentration has been associated with population density (Buerge et al., 2003). Moreover, nowadays coffee is no longer seen just as a stimulant, due to the caffeine content, but is also considered as a beverage with important antioxidant properties due the bioactive compounds it contains (Liu and Kitts, 2011).

Vitamin C is an essential nutrient for human physiology. In Brazil, vitamin C supplements are expensive and most of the population obtains vitamin C through its consumption of fruits and vegetables. Therefore, the vitamin C assay in food is crucial in post-harvest studies to conserve and minimize losses of this highly sensitive nutrient. This study proposes a method for analyzing vitamin C by High Performance Liquid Chromatography using a hydrogen type ion exchange column, and demonstrates that it is more efficient than the traditional methods of reverse phase column (C18) for complex matrixes and low levels of this analyte. Chromatograms were baseline resolved and peak purity evaluation showed spectral homogeneity by photo diode array detector. This method was also tested using the best extraction solution to stabilize vitamin C, demonstrating that 0.05 M of superpure sulfuric acid (also the mobile phase) was the most efficient solution for this purpose.

In the recent years the way of thinking about human health necessarily passes by human food. Recent discoveries are not only concerned about valuable biomolecules but also contaminants. Thus, the screening of substances in animal and vegetable matrices by analytical techniques is focused on the presence and absence of target substance. In both cases, the majority of these substances are present as traces or in very low levels. Contaminants could be naturally present in the food, inserted on it or even developed on it as a consequence of food processing or cooking. Pesticides, mycotoxins, dioxins, acrylamide, Sudan red, melamine and now 4(5)-methylimidazole can be, at present, be listed as some of the world big problems related to food contaminants and adulterants. With the development of liquid chromatography coupled to mass spectrometry (LC-MS-MS), in the last few decades, analysis of some food contaminants in trace levels trace become less laborious, more accurate and precise. The multiple approach of those techniques make possible to obtain many results in one single run. On the other hand, European Union (2002/657/EC) established regulations for analytical methods regarding mass spectrometry as detection tool, showing the importance of this technique in food quality control. The EU criteria uses identification points (IPs) that could be achieved basically with four product ions (including molecular ion) or reduced with the use of high resolution equipments. This kind of mass spectrometers made the IPs criteria more accessible, as the exact mass information is a differential tool. In view of this the aim of this review is to present the actual scenario for mass spectrometry analysis in a complex vegetable food matrix such as roasted coffee, with emphasis on needs and challenges regarding the LC-MS technique in order to meet and contribute to food safety standards in this complex matrix.
