1. Machine Learning to Improve the Sensing of Biomolecules by Conical Track-Etched Nanopore

Abstract

Single nanopore is a powerful platform to detect, discriminate and identify biomacromolecules. Among the different devices, the conical nanopores obtained by the track-etched technique on a polymer film are stable and easy to functionalize. However, these advantages are hampered by their high aspect ratio that avoids the discrimination of similar samples. Using machine learning, we demonstrate an improved resolution so that it can identify short single- and double-stranded DNA (10- and 40-mers). We have characterized each current blockade event by the relative intensity, dwell time, surface area and both the right and left slope. We show an overlap of the relative current blockade amplitudes and dwell time distributions that prevents their identification. We define the different parameters that characterize the events as features and the type of DNA sample as the target. By applying support-vector machines to discriminate each sample, we show accuracy between 50% and 72% by using two features that distinctly classify the data points. Finally, we achieved an increased accuracy (up to 82%) when five features were implemented.

Abstract

Protein adsorption at the liquid−solid interface is an old but not totally solved topic. One challenge is to find an easy way to characterize the protein behavior on nanoparticles and make a correlation with its intrinsic properties. This work aims to investigate protein adsorption on gold nanoparticles and the colloidal properties. The protein panel was chosen from different structural categories (mainly-α, mainly-β or mix-αβ). The result shows that the colloidal stability with salt addition does not depend on the structural category. Conversely, using the single nanopore technique, we show that the mainly-α proteins form a smaller corona than the mainly-β proteins. We assign these observations to the lower internal energy of α-helices, making them more prone to form a homogeneous corona layer.

Abstract

The modification of the inner nanopore wall by polymers is currently used to change the specific properties of the nanosystem. Among them, the polyethylene glycol (PEG) is the most used to prevent the fouling and ensure the wettability. However, its properties depend mainly on the chain structure that is very difficult to estimate inside this confined space. Combining experimental and simulation approaches, we provide an insight to the consequence of the PEG presence inside the nanopore on the nanopore properties. We show, in particular, that the cation type in the electrolyte, together with the type of electrolyte (water or urea), is at the origin of the ion transport modification in the nanopore.

Abstract

Several neurodegenerative diseases have been linked to proteins or peptides that are prone to aggregate in different brain regions. Aggregation of amyloid-β (Aβ) peptides is recognized as the main cause of Alzheimer's disease (AD) progression, leading to the formation of toxic Aβ oligomers and amyloid fibrils. The molecular mechanism of Aβ aggregation is complex and still not fully understood. Nanopore technology provides a new way to obtain kinetic and morphological aspects of Aβ aggregation at a single-molecule scale without labeling by detecting the electrochemical signal of the peptides when they pass through the hole. Here, we investigate the influence of nanoscale geometry (conical and bullet-like shape) of a track-etched nanopore pore and the effect of molecular crowding (polyethylene glycol-functionalized pores) on Aβ fibril sensing and analysis. Various Aβ fibril samples that differed by their length were produced by sonication of fibrils obtained in the presence of epigallocatechin gallate. The conical nanopore functionalized with polyethylene glycol (PEG) 5 kDa is suitable for discrimination of the fibril size from relative current blockade. The bullet-like-shaped nanopore enhances the amplitude of the current and increases the dwell time, allowing us to well discern the fibrils. Finally, the nanopore crowded with PEG 20 kDa enhances the relative current blockade and increases the dwell time; however, the discrimination is not improved compared to the "bullet-shaped" nanopore.

Abstract

Solid-state nanopores are an emerging technology used as a high-throughput, label-free analytical method for the characterization of protein aggregation in an aqueous solution. In this work, we used Levodopamine to coat a silicon nitride nanopore surface that was fabricated through a dielectric breakdown in order to reduce the unspecific adsorption. The coating of inner nanopore wall by investigation of the translocation of heparin. The functionalized nanopore was used to investigate the aggregation of amyloid-β and α-synuclein, two biomarkers of degenerative diseases. In the first application, we demonstrate that the α-synuclein WT is more prone to form dimers than the variant A53T. In the second one, we show for the Aβ(42)-E22Δ (Osaka mutant) that the addition of Aβ(42)-WT monomers increases the polymorphism of oligomers, while the incubation with Aβ(42)-WT fibrils generates larger aggregates.

Abstract

International audience; Protein detection and identification at the single-molecule level are major challenges in many biotechnological fields. Solid-state nanopores have raised attention as label-free biosensors with high sensitivity. Here, we use solid-state nanopore sensing to discriminate two closely related proteins, α-thrombin and γ-thrombin. We show that aptamer functionalization improves protein discrimination thanks to a significant difference in the relative current blockade amplitude. To enhance discrimination, we postprocessed the signals using machine learning and training algorithms and we were able to reach an accuracy of 98.8% using seven features and ensemble methods.

9. Real-Time Fast Amyloid Seeding and Translocation of α-Synuclein with a Nanopipette

Abstract

The detection to α-synuclein (αS) assemblies as a biomarker of synucleinopathies is an important challenge for further development of an early diagnosis tool. Here, we present proof of concept real-time fast amyloid seeding and translocation (RT-FAST) based on a nanopipette that combines in one unique system a reaction vessel to accelerate the seed amplification and nanopore sensor for single-molecule αS assembly detection. RT-FAST allows the detection of the presence αS seeds WT and A53T variant in a given sample in only 90 min by adding a low quantity (35 μL at 100 nM) of recombinant αS for amplification. It also shows cross-seeding aggregation by adding mixing seeds A53T with WT monomers. Finally, we establish the dependence between the capture rate of aggregates by the nanopore sensor and the initial seed concentration from 200 pM to 2 pM, which promises further development toward a quantitative analysis of the initial seed concentration.

Abstract

International audience; In two decades, the solid state and polymer nanopores became attractive method for the protein sensing with high specificity and sensitivity. They also allow the characterization of conformational changes, unfolding, assembly and aggregation as well the following of enzymatic reaction. This review aims to provide an overview of the protein sensing regarding the technique of detection: the resistive pulse and ionic diodes. For each strategy, we report the most significant achievement regarding the detection of peptides and protein as well as the conformational change, protein-protein assembly and aggregation process. We discuss the limitations and the recent strategies to improve the nanopore resolution and accuracy. A focus is done about concomitant problematic such as protein adsorption and nanopore lifetime.

Abstract

International audience; The Aβ(1-42) aggregation is a key event in the physiopathology of Alzheimer's disease (AD). Exogenous factors such as environmental pollutants, and more particularly pesticides, can corrupt Aβ(1-42) assembly and could influence the occurrence and pathophysiology of AD. However, pesticide involvement in the early stages of Aβ(1-42) aggregation is still unknown. Here, we employed conical track-etched nanopore in order to analyse the Aβ(1-42) fibril formation in the presence of pyrimethanil, a widely used fungicide belonging to the anilinopyrimidine class. Our results evidenced a pro-aggregating effect of pyrimethanil on Aβ(1-42). Aβ(1-42) assemblies were successfully detected using conical nanopore coated with PEG. Using an analytical model, the large current blockades observed (>0.7) were assigned to species with size close to the sensing pore. The long dwell times (hundreds ms scale) were interpreted by the possible interactions amyloid/PEG using molecular dynamic simulation. Such interaction could leave until splitting phenomena of the dimer structure. Our work also evidences that the pyrimethanil induce an aggregation of Aβ(1-42) mechanism in two steps including the reorganization prior the elongation phase.

Abstract

International audience; The heparin contamination by oversulfated chondroitin (OSCS) was at the origin of one major sanitary problem of last decade. Here we propose a novel strategy to detect OSCS from heparin solution based on conical nanopore functionalized with poly-L-lysine deposition to ensure its re-usability. This sensor is an excellent to detect low heparin concentration (from 25 ng/ml to 3 mu g/ml) using the modification of ionic current rectification. It also allows following the kinetic of heparin degradation by heparinase with a good correlation with results obtained by classical methods. The sensor is sensitive to the inhibition of heparinase by OSCS until a concentration of 200 pg/l representing 0.01% in weight in a heparin. This resolution is one order of magnitude lower than the one obtained by chromatography. For the first time, it was reached without fluorescence labeling

Abstract

Expérience réalisée au GANIL; International audience; The salinity gradient between brine and fresh water is an abundantsource of power which can be harvested by two major membrane methods:pressure-retarded osmosis and reversed electrodialysis. Nowadays, the lattertechnology is close to real application, but it still suffers from low power yield. Lowmembrane selectivity and complex membrane fabrication are the main limitingfactors. To improve that, we design a couple of ion-selective membranes based onthe track-etched polymer nanopore functionalized by highly charged hydrogels.Two nanopore geometries are compared (cylindrical and conical shape) togenerate osmotic energy with gel functions and more importantly can be scaledup. Experiments from the single nanopore and multipore membrane to stackedmembranes show complete characterization from ionic transportation to energygeneration and a clear relationship from the single pore to stacked membranes. Inthe actual experiment conditions, a power density of 0.37 W m−2 at pH 7 wasachieved. By improving ionic tracks and reducing intermembrane distances, it canbe a good candidate for industrial applications.

Abstract

International audience; This study aims to evaluate the effect of ionic liquid intercalation in smectite on fluorescent dye adsorption. In the first step, we investigated the influence of interlayer cation of Montmorillonite (Li+, Na+, K+, Rb+, and Cs+) on the intercalation of 1-butyl-3-methylimidazolium chloride. The adsorption data was well described by the pseudo-second-order kinetics and Langmuir adsorption isotherm. The material structure was investigated by Fourier transform infrared (FTIR) and wide-angle X-ray diffraction (XRD) showing that the ionic liquid intercalation is occurred. Then, the organo-clays were used to the adsorption of a fluorescein. In presence of ionic liquid the dye adsorption is a little bit improved. The interaction between the dye and the ionic liquid are highlighted based on the fluorescence lifetime. This interesting result confirms that the dye is not released compared to the pristine Montmorillonite.

Abstract

The modification of the inner nanopore wall by polymers is currently used to change the specific properties of the nanosystem. Among them, the polyethylene glycol (PEG) is the most used to prevent the fouling and ensure the wettability. However, its properties depend mainly on the chain structure that is very difficult to estimate inside this confined space. Combining experimental and simulation approaches, we provide an insight to the consequence of the PEG presence inside the nanopore on the nanopore properties. We show, in particular, that the cation type in the electrolyte, together with the type of electrolyte (water or urea), is at the origin of the ion transport modification in the nanopore.

Abstract

International audience; Single nanopore is a powerful platform to detect, discriminate and identify biomacromolecules. Among the different devices, the conical nanopores obtained by the track-etched technique on a polymer film are stable and easy to functionalize. However, these advantages are hampered by their high aspect ratio that avoids the discrimination of similar samples. Using machine learning, we demonstrate an improved resolution so that it can identify short single-and double-stranded DNA (10-and 40-mers). We have characterized each current blockade event by the relative intensity, dwell time, surface area and both the right and left slope. We show an overlap of the relative current blockade amplitudes and dwell time distributions that prevents their identification. We define the different parameters that characterize the events as features and the type of DNA sample as the target. By applying support-vector machines to discriminate each sample, we show accuracy between 50% and 72% by using two features that distinctly classify the data points. Finally, we achieved an increased accuracy (up to 82%) when five features were implemented.

Abstract

International audience; Biomimetic and bio‐inspired membranes draw great attention and generate extensive efforts to solve environmental, health, and energy issues. In this field, track‐etched membranes present the advantage to be tuned from a single pore to a large‐scale multipore membrane. On one hand, from the large‐scale membrane to a single nanopore, the research becomes more fundamental, meticulous, and quantitative to describe concrete processes inside confined spaces. This allows understanding the role of the surface phenomena and nanoscale effects. On the other hand, working from a single pore to a multipore membrane allows perfect control of the membrane properties. This ensures a promising ability for upscale from the lab findings to applications. In this review, a global insight on the track‐etched nanopore/membrane is taken through presentation of fabrication and functionalization methods, transport properties, and the related applications. By functionalization, track‐etched nanopores can be used to understand ion transport under confined spaces with high aspect ratio, to analyze single molecules, to design stimuli‐responsive membranes, and to generate energy from salinity gradient and light

Abstract

International audience; Protein aggregation is involved in many diseases, including Parkinson's and Alzheimer's. The latter is characterized by intraneuronal deposition of amyloid aggregates composed of the tau protein. Although large and insoluble aggregates are typically found in affected brains, intermediate soluble oligomers are thought to represent crucial species for toxicity and spreading. Nanopore sensors constitute an emerging technology that allows the detection of the size and populations of molecular assembly present in a sample. Here, we employed conical nanopores to obtain the particle distributions during tau aggregation. We identified three distinct populations, monomers, oligomers, and fibrils, which we could quantify along the aggregation process. By comparing tau wild type with a mutant carrying the disease-associated P301L mutation, we showed that the latter mutation promotes the formation of oligomers. We furthermore highlighted that the P301L mutation promotes fibril breakage. This work demonstrates that conical nanopore is a powerful tool to measure and quantify transient protein aggregate intermediates.

Abstract

This research reports the transport behaviors of long flexible polymers (hyaluronic acid) through long conical track-etched nanochannels with and without grafted enzymes. The impacts of the channel diameter and the polymer regimes in solution (dilute and semi-dilute) have been investigated. Without enzymes, the experimental results can be well explained by the analytical models of the scaling law of de Gennes. Then, the corresponding enzymes (hyaluronidase) were grafted inside the channel. When enzymes are located at the base side, polymers get degraded at the entrance and the degraded products are detected. When enzymes are grafted at the tip side, the extension of translocation duration due to the binding of substrate–enzyme is observed. This is for the first time that the enzymatic degradation reactions are characterized in situ at the single molecule level by nanopore technology.

Abstract

In this work, we investigated the impact of different polyelectrolytes (polyacrylic acid, polystyrene sulfonate, dextran sulfate, and chondroitin sulfate) on lysozyme properties when they form colloidal complexes. To this aim, we characterized (i) the size and stability of the different polyelectrolyte-lysozyme complexes upon addition of NaCl (different concentrations) by diffusion light scattering, and (ii) the structure and accessibility of lysozyme active site in such complexes by fluorescence quenching and time resolved fluorescence analysis. We then used these results to explain the antibacterial activity variations among colloidal complexes and compared with free lysozyme. Our findings show that colloidal complexes that are more prone to swelling (i.e., lysozyme complexed with polystyrene sulfonate) are less stable upon salt addition. In these colloids, the enzymatic site is also more accessible. However, the antibacterial activity does not depend on the swelling properties because no large structural modification of the active site occurs.

Abstract

International audience; The understanding of the interactions between nanomaterials, biomolecules, and polyphenols is fundamental in food chemistry, toxicology, and new emerging fields, such as nanomedicine. Here, we investigated the effect of the resveratrol, a principal actor in drug-delivery application on the interaction between bovine serum albumin (BSA), employed as a vector for the delivery of polyphenol drugs, and gold nanoparticle (gNP), the most promising tool in theranostic applications. Through a combination of experimental techniques, which includes an initial evaluation by dynamic light scattering and surface plasmon resonance spectroscopy, we were able to evaluate the evolution of the gold nanoparticle aggregation with increasing ionic strength and the consequences of the BSA and resveratrol addition. To investigate the mechanisms of the interactions, we pursued at the single-molecule level using solid-state nanopore and fluorescence correlation spectroscopy. Our results show that without resveratrol, the BSA is adsorbed on the gNP in water or saline solution. In the presence of resveratrol, the BSA is normally absorbed on gNP in water, but the salt addition leads to its desorption. The resveratrol clearly plays a fundamental role, changing the protein behavior and making the BSA adsorption a reversible process in the presence of salt.

We studied their influence on the ionic diode properties We have considered different couples of polyelectrolytes: poly-l-lysine/poly(acrylic acid) and poly(ethyleneimine)/poly(acrylic acid) as well as the influence of cross-linking. The results show that the nanopores decorated with poly(ethyleneimine)/poly(acrylic acid) exhibit an interesting behavior. Indeed, at pH 3, the nanopore is open only at the low salt concentration, while at pH 7, it is already open. The nanopores functionalized with poly-l-lysine/poly(acrylic acid) do not show an inversion of ionic transport properties with the pH as expected. After cross-linked to prevent large conformational changes, the ionic diode properties are dependent on the pH.

Abstract

International audience; Hybrid pigments based on carminic acid (CA) stabilized by clay minerals have been prepared. Three different pathways were used: CA@saponite; CA@Al-pillared saponite; and hydrogels of saponite-CA or montmorillonite-CA covered by polyorganosilane (POS). Pigments were characterized by a set of different techniques including X-Ray diffraction (XRD), transmission electron microscopy (TEM), infrared spectroscopy (FTIR), solid state nuclear magnetic resonance (NMR) and time-resolved fluorescence (TRF) spectroscopies. The color parameters, i.e., L*a*b*, were measured. In addition, the photostability of the samples under visible light was evaluated. The results showed that CA@PilSap presented a greater stability compared to CA@Saponite. The pigments obtained from hydrogels presented the greatest light stability due to the coverage with POS that acts as a protective layer.

Abstract

The fundamental understanding of the transport mechanisms of objects across a single nanopore is one key point to develop Coulter counters at the nanoscale for macromolecule or nanoparticle detection. In this area, nanoparticles have been less investigated than biomacromolecules such as DNA or proteins due to their self-aggregation in the presence of salts. In this work, the transport of modified latex nanoparticles across solid-state nanopores was investigated. To prevent their aggregation, their surface was modified with a low molecular weight single strand DNA coating. Then the coated nanoparticles were successfully detected across a single pore material in 200 mM NaCl buffer. The experimental capture rate was compared to that of the predictive model. It reveals that the nanoparticle entrance inside the nanopore is mainly governed by diffusion and required a weak energy. For relative current blockades, the predictive model should take into account both the nanopore shape and the additional charge due to ssDNA coating.

Abstract

Protein adsorption at the liquid&ndash, solid interface is an old but not totally solved topic. One challenge is to find an easy way to characterize the protein behavior on nanoparticles and make a correlation with its intrinsic properties. This work aims to investigate protein adsorption on gold nanoparticles and the colloidal properties. The protein panel was chosen from different structural categories (mainly-&alpha, mainly-&beta, or mix-&alpha, &beta, ). The result shows that the colloidal stability with salt addition does not depend on the structural category. Conversely, using the single nanopore technique, we show that the mainly-&alpha, proteins form a smaller corona than the mainly-&beta, proteins. We assign these observations to the lower internal energy of &alpha, helices, making them more prone to form a homogeneous corona layer.

International audience; This review gives an overview of protein adsorption at solid/liquid interface. Compared to the other ones, we have focus on three main questions with the point of view of the protein. The first question is related to the kinetic and especially the using of Langmuir model to describe the protein adsorption. The second question is about the concept of hard and soft protein. In this part, we report the protein structural modification induced by adsorption regarding their intrinsic structure. This allows formulating of a new concept to classify the protein to predict their behavior at solid/liquid interface. The last question is related to the protein corona. We give an overview about the soft/hard corona and attempt to make correlation with the concept of hard/soft protein.

Abstract

International audience; Hybrid materials based on montmorillonite, a cationic polymer and carminic acid were prepared. The surface charge of montmorillonite was inversed thanks to the cationic polymer, polydiallyldimethylammonium chloride. Samples were characterized by a set of different techniques including Infrared (IR) and Nuclear Magnetic Resonance (NMR) spectroscopies to highlight the nature of the interactions between the organic and inorganic parts. The photo-stability of the samples was tested for different durations, and L*a*b* parameters were measured. It was possible to propose an approach for the degradation mechanism of the supported dye thanks to EPR (electron paramagnetic resonance) spectroscopy. The experimental data were in agreement with the theoretical periodic DFT calculations where a molecular scheme of the adsorption complex was proposed, and the importance of hydration on the stability of the adsorption complex highlighted.

Abstract

Hybrid materials based on one organic dye, the carminic acid, and montmorillonite have been prepared and characterized by a multi-technical approach. Aluminum and tin II cations were used in the experimental mixtures in order to have a chelating effect with the carminic acid. X-Ray diffraction patterns (XRD) did not highlight any intercalation of the organic molecule in the interlayer space, a result confirmed by transmission electron microscopy (TEM) micrographs. The fixation of the organic molecules on the phyllosilicates was then investigated by 13 C solid state nuclear magnetic resonance (NMR) and time resolved fluorescence spectroscopies. The spectra showed the role that the ketone and catechol functions of the dye played for its fixation and stressed on the decrease of its mobility when the lake pigment was formed. The role of the clay edges and/or surface was highlighted by this set of experimental data.

Abstract

Amphotericin B (AmB) is a well-known polyene which self-organizes into membrane cell in order to cause the cell death. Its specific action towards fungal cell is not fully understood but was proved to become from sterol composition. The mechanism was shown experimentally to require the formation of stable sterol/polyene couples which could then organize in a nanochannel. This would allow the leakage of ions responsible for the death of fungal cells, only. In this present study, we investigate the arrangement of AmB/sterols in biological membrane using molecular dynamic simulations in order to understand the role of the sterol structure on the antifungal action of the polyene. We show in particular that the nanochannels tend to close up when cell was composed with cholesterol (animal cell) due to strong interaction between amphotericin and sterol. On the other side, with ergosterol (fungal cell) the largest interactions between amphotericin and lipid membrane lead to the appearance of large hole that could favor the important leakage of ions and thus, the fungal cell death. This work appears as a good complement in the extensive studies linked to the understanding of the antifungal molecules in membrane cells.

Abstract

International audience; myloid fibrils are involved in several neuro-degenerative diseases. However, because of their polymorphism and low concentration, they are challenging to assess in real-time with conventional techniques. Here, we present a new approach for the characterization of the intermediates: protofibrils and "end-off" aggregates which are produced during the amyloid formation. To do so, we have fashioned conical track-etched nanopores that are functionalized to prevent the fouling. Using these nanopores, we have followed the kinetic of amyloid growth to discriminate the different intermediates protofibrils and "end-off. Then, the nanopore was used to characterize the effect of promoter and inhibitor of the fibrillation process. Finally, we have followed in real-time the degradation of amyloid with peptase. Compare with the SiN nanopore, the track-etched one features exceptionally high success rate via functionalization and detection in "one-pot". Our results demonstrate the potential for a conical nanopore to be used as a routine technique for the characterization of the amyloid growth and/or degradation.

Abstract

International audience; Wine chemistry inspires and challenges with its complexity and intriguing composition. In this context, the composites based on the use of a model protein, a polyphenol of interest and montmorillonite in a model hydroalcoholic solution have been studied. A set of experimental characterization techniques highlighted the interactions between the organic and the inorganic parts in the composite. The amount of the organic part was determined by ultraviolet-visible (UV–VIS) and thermal analysis. X-ray diffraction (XRD) and transmission electronic microscopy (TEM) informed about the stacking/exfoliation of the layers in the composites. Vibrational and nuclear magnetic resonance spectroscopies methods stressed on the formation of a complex between the protein and the polyphenol before adsorption on the clay mineral. The mobility/rigidity of the organic parts were determined by fluorescence time resolved spectroscopy. Changes in the secondary structure of the protein occured upon complexation with polyphenol on clay mineral due to strong interactions. Although not representating faithfully enological conditions, these results highlight the range and nature of mechanisms possibly involved in wine fining.

Abstract

Solid-state nanopore technology for nanoparticle sensing is considered for the development of analytical tools to characterise their size, shape or zeta potential. In this field, it is crucial to understand how the nanopore inner surface influences the dynamic of nanoparticle translocation. Here, three single nanopores directly drilled in metal alloys (titanium nitride, titanium-tantalum and tantalum) are considered. The translocation of polystyrene nanoparticles coated with ssDNA is investigated by the resistive pulse method at different concentrations and voltages. The results show that the nanoparticle energy barrier for entrance into the pore decreases for nanopores that exhibits a higher surface energy and hydrophilicity, while the dwell time is found to depend on the nanopore surface state. Overall, this study demonstrates that the control of nanopore surface state must be taken into account for the resistive pulse experiments for nanoparticle detection.

Abstract

International audience; The antibiotic intercalation inside the layered double hydroxide (LDH) layers was usually considered for water decontamination but rarely for drug delivery. Here, tetracycline (TCH) and oxytetracycline (OXY) were immobilized in Zn2Al-Cl LDH following two methods: co-precipitation and anionic exchange. The interfacial concentration of antibiotic varies from 0.04 to 0.5 depending the method of immobilization. The antibiotics are not intercalated in the interlayer space allowing their release in 10 Hours. The antibacterial activity against both E. coli and S. epidermidis revealed that the loaded antibiotics are still active but less efficient compared to the free ones. After exposition to UV light or to high temperature storage (30, 60 and 120 degrees C), their antibacterial activity significantly decreases due to their degradation especially when antibiotic is loaded on material by co-precipitation. These results are promise to reduce antibiotic contamination in waters.

Abstract

To mimic the selectivity and properties of biological channels and pumps, adaptive systems can be designed using artificial membranes. One big advantage of those membranes is that they are able to respond to several stimuli at the same time. Here, it is proposed to tailor conical nanopores by grafting polyethyleneglycol (PEG)-spiropyrans in order to combine both light- and pH-responsive properties. The study of the ionic transport reveals that after UV irradiation the nanopore is open while after visible irradiation, it is closed. This gating property is due to the self-assembly of PEG-spiropyran. The reversibility of the assembly inside the nanopore strongly depends on the solvent. Indeed, water blocks the photoswitchable molecule in its zwitterionic form, while ethanol favors the conformational switching. Under UV irradiation, the pH can control the nanopore selectivity. It is demonstrated that at acidic pH, the nanopore is selective to anion while at neutral pH the cation transport is favored.

Abstract

We investigated ionic transport behavior in the case of uncharged conical nanopores. To do so, we designed conical nanopores using atomic layer deposition of Al2O3/ZnO nanolaminates and then coated these with trimethylsilane. The experimental results are supported by molecular dynamics simulations. The ionic transport reveals an unexpected behavior: (i) a current rectification and (ii) a constant conductance at low salt concentration which are usually reported for charged conical nanopore. To explain these results, we have considered different assumptions: (i) a default of functionalization, (ii) the adsorption anion and (iii) the slippage. The first one was refuted by the study of the poly-l-lysine transport through the nanopore. To verify the second assumption, we investigate the effect of pH on the current rectification and the molecular dynamics simulations. Finally our study demonstrates that the unexpected ionic transport is provided to a predominant effect of slippage due to the water organization at the solid/liquid interface.

Abstract

International audience; Hybrid pigments have attracted great interest due to their stability and physicochemical properties that can be used in paintings, artworks and in the field of cultural heritage. The study of new materials includes the search for new pigments by immobilization of the dyes in resistant substrates such clays minerals to get new colour fonts and materials with high stability. Organic-inorganic based lake pigments, at different pH, have been prepared. Ti- and Al-pillared montmorillonite were synthesized and loaded with carminic acid (CA) and alizarin (Aliz) organic dyes. The pillaring process was investigated by X-ray diffraction, textural analysis, transmission electronic microscopy and solid state nuclear magnetic resonance of Al-27. The interactions between the organic guest and the inorganic host were highlighted by infrared, C-13 and Al-27 solid state magnetic nuclear resonance and time resolved fluorescence spectroscopies. The spectra support complex formation between the chromophores and the inorganic matrix. The colours of the pigments prepared were pH dependent. Lake pigments based on Al-Pillared Mt were more stable under light than Ti-based hybrid, even in oil painting formulations.

Abstract

In nature, ion channels are highly selective pores and act as gate to ensure selective ion transport, allowing ions to cross the membrane. By mimicking them, single solid state nanopore devices emerge as a new, powerful class of molecule sensors that allow for the label-free detection of biomolecules (DNA, RNA, and proteins), non-biological polymers, as well as small molecules. In this review, we exhaustively describe the fabrication and functionalization techniques to design highly robust and selective solid state nanopores. First we outline the different materials and methods to design nanopores, we explain the ionic conduction in nanopores, and finally we summarize some techniques to modify and functionalize the surface in order to obtain biomimetic nanopores, responding to different external stimuli.

Abstract

The direct capture of energy from salinity gradient is limited by low energetic efficiency. Here we propose to investigate an original membrane for capture of osmotic energy based conical track-etched nanopore. In order to improve the surface charge at pH 7, the nanopore was functionalized with chitosan / poly(acrylic acid) self-assembly. We observed for single nanopore a large osmotic current around 0.6 nA for a concentration gradient c,Tip/c,Base = 1000. This corresponds to energy around 25 pW per nanopore. Then, we demonstrate that this result can be upscale for a multi-pore membrane in the case of a low density of nanopores.

Abstract

We report on an indirect optical method for the determination of glucose via the detection of hydrogen peroxide (H2O2) that is generated during the glucose oxidase (GOx) catalyzed oxidation of glucose. It is based on the finding that the ultraviolet (~374 nm) and visible (~525 nm) photoluminescence of pristine zinc oxide (ZnO) nanoparticles strongly depends on the concentration of H2O2 in water solution. Photoluminescence is quenched by up to 90 % at a 100 mM level of H2O2. The sensor constructed by immobilizing GOx on ZnO nanoparticles enabled glucose to be continuously monitored in the 10 mM to 130 mM concentration range, and the limit of detection is 10 mM. This enzymatic sensing scheme is supposed to be applicable to monitoring glucose in the food, beverage and fermentation industries. It has a wide scope in that it may be extended to numerous other substrate or enzyme activity assays based on the formation of H2O2, and of assays based on the consumption of H2O2 by peroxidases.

Abstract

International audience; In the present study, the adsorption capacities of two intercalated smectites, CTAB-saponite and CTAB-montmorillonite with a cationic surfactant, were investigated with three fluorescent dyes namely Rhodamine 640 perchlorate rhodamine (Rho), sulforhodamine B (SR) and Kiton red 620 (KR). The adsorption isotherms fit well with the non-linear Langmuir isotherm model and the maximum adsorption capacities of all the composites are determined. The photophysical properties such as anisotropy and fluorescence lifetime of all the fluorescent dyes over the clay materials are determined. The set of experimental data based on X-Ray diffraction (XRD), transmission electron microscopy (TEM), Thermal analysis (TG-DTA) and fluorescence measurements allow highlighting the presence or the absence of interactions between the dyes and the modified clay minerals.

Abstract

International audience; A simple synthetic route based on nanosphere lithography has been developed in order to design largescale nanoarray for specific control of protein anchoring. This technique based on two-dimensional (2D) colloidal crystals composed of polystyrene spheres allows the easy and inexpensive fabrication of large arrays (up to several centimeters) by reducing the cost. A silicon wafer coated with a thin adhesion layer of chromium (15 nm) and a layer of Gold (50 nm) is used as a substrate. PS spheres aredeposited on the gold surface using the floating-transferring technique. The PS spheres were then functionalized with PEG-biotin and the defects by self-assembly monolayer (SAMs) PEG to prevent unspecific adsorption. Using epifluorescence microscopy, we show that after immersion of sample ontarget protein (avidin and anti-avidin) solution, the latter are specifically located on polystyrene spheres. Thus, these results are meaningful for exploration of devices based on large-scale nanoarray of PS spheres and can be used for detection of target proteins or simply to pattern a surface with specific proteins.

Abstract

Layered double hydroxide (LDH) nanohybrid intercalated biomolecules, including oligonecluotides, genes and peptides/proteins, have attracted particular attention since they exhibit improved safety and effectiveness as successful delivery biosystems. The current study specifically investigated the adsorption of nisin peptide and precisely the control of the release of the payload. Adsorption occurred from peptide solution in contact with zinc-aluminum LDH at room temperature, looking out over the influence of the Zn2+/Al3+ ratio, the anion exchange capacity, the nature of the intercalated anion, the host matrix, and the host morphology. Higher adsorption was obtained, around 80% of the loaded nisin and successful intercalation was verified by X-ray diffraction. The in-vitro release tests of the nisin from the biohybrid formulation was held over 25days in PBS medium (0.01M, pH7,4) and showed that no burst release phenomenon occurred at the beginning step, in addition, a sustained-time release of nisin was obtained compared with the free nisin. Therefore, these preliminary results are encouraging for the development of bioprotectors based on nisin intercalated LDH and being implemented in the food and medical industries.

Abstract

International audience; Lysozyme from hen egg white is composed by a unique linear chain of 129 amino acids. It is known to inhibit Gram positive bacteria and to form amyloid fibrils at low pH, under 75 °C. This work investigates the effect of the fibrillation and/or adsorption onto a layered double hydroxide material on the antibacterial properties of lysozyme. The kinetics of adsorption follows a behavior of pseudo second order model. The X-ray diffraction and the Fourier transform infrared spectroscopy highlight that adsorption occurs only on the external surface of the material. Interestingly, the amyloid fibrils of lysozyme retain their antibacterial properties when they are adsorbed on the layered double hydroxide; even if their activity is lowered, the active site of the enzyme is not fully denatured and is still accessible. This is confirmed by the study of the tryptophan using time-resolved fluorescence spectroscopy.

Abstract

Biological channels have served as inspiration to design stimuli-response artificial nanopores. Here we propose an original approach to design a pH-gate nanopore based on polyethylenimine and chondroitin-4-sulfate (ChS) layer-by-layer self-assembly. This approach is interesting because it is rapid and permits monitoring in real time of functionalization. The study of ionic transport through these single nanopores reveals a selectivity on anions and pH-gate properties at low salt concentration. It is open at pH below 4 or 5 depending on salt concentration. These properties are explained by the modification of both charge and conformation of ChS as well as swelling of the polyelectrolyte complex.

Abstract

The importance of DNA structures in medicine has motivated researchers to develop sequencing methods with low costs and high speeds. The potential use of nanofluidics for the analysis of the translocation of polynucleotides through a nanopore is a very interesting solution. Herein, we demonstrate both by experiments and by explicit solvent molecular dynamics simulations that the translocation of polyadenylic acid and polycytidylic acid can significantly affect the ionic flux behavior. In particular, the nanochannel enhances the velocity difference between polynucleotides during the translocation process. Our results revealed the important role of the strand structures during their translocation, as already observed in biochannels. The folding/unfolding process leads to stronger interactions with the nanopore wall that decrease the velocity of the strands. This behavior could lead to the detection of different polynucleotides in a mixture.

Abstract

The development of nanometric Coulter counters for nanoparticle detection is an attractive and promising field of research. In this work, we have studied the influence of the nanopore surface state on charged polymer nanoparticle translocations. To make this, the translocation of carboxylate modified polystyrene microspheres (diameter 40, 70 and 100 nm) has been investigated through two kinds of high aspect ratio nanopores (negative and uncharged). The latter were tailored by a single track-etched and atomic layer deposition technique. It was shown that the mobility and the energy barrier are strongly dependent on nanopore surface charge. Typically if the latter exhibits negative surface charge, the microsphere mobility increases and the global energy barrier of entrance inside the nanopore decreases with its diameter, converse to the uncharged nanopore.

Abstract

The confinement and the understanding of ion transport through ionic channels when they are confined inside solid-state nanopores smaller than 10 nm remains a challenge. Here we report on the fabrication of biomimetic nanopores with high length (5 μm)/diameter (smaller than 10 nm) ratio obtained using both a track-etched technique and atomic layer deposition on flexible membranes. These membranes present uniform hydrophobic nanopores with a low roughness inside the pores. Gramicidin A is then confined inside nanopores (diameter 10.6, 5.7, and ∼2 nm) leading to the NaCl ionic transport mechanism through a hybrid nanopore similar to the biological ones especially for small diameter (5.7 and ∼2 nm).

Abstract

We report the production of a robust hybrid polyene/artificial solid-state membrane. The production of this membrane is based on the confinement of Amphotericin B inside the cylindrical nanopores of a polymeric membrane. The experiments supported by molecular dynamics simulations reveal new filtration properties never seen in biological membranes. Indeed this biomimetic membrane exhibits higher permeability to anions than to monovalent (Na+ and K+) and divalent (Ca2+) cations. This work opens a new route of separation in the domain of nanobiofiltration, especially for tunable nanodevices based on differential ion conduction, using a fundamental understanding of the confinement mechanism.
