Abstract

Cordyceps militaris specimens were continuously collected by Entomopathogenic Fungal Culture Collection (EFCC), Kangwon National University from different mountains, national parks and recreation parks of Korea from 1986 to 2002, mainly from late May to October of each year. Dry specimens of C. militaris along with their isolates have been preserved in EFCC. Fruiting of C. militaris was induced from single ascospore isolates as well as their combinations in brown rice medium. Fruiting experiments showed that combinations of single ascospore isolates produced fertile fruiting bodies, but single isolates could not produce any fruiting bodies. It was shown that two isolates of the opposite mating types were required to produce fertile stromata. However, combinations of the same mating type isolates produced no fruiting body, showing that C. militaris is a bipolar, heterothallic fungus.

Abstract

In order to breed a Cordyceps bassiana isolate that stably forms fruiting body in artificial cultivation, isolates derived from subculturing and single spores were tested through mating. From C. bassiana EFCC 783, three subcultured isolates EFCC 2830, EFCC 2831 and EFCC 2832 were obtained and fourteen single conidial isolates were obtained from these three subcultured isolates. Two different morphological types were found in the fourteen single conidial isolates. One type was able to form synnemata and another type was not able to form synnemata. Since switch of morphological type was not observed despite their continuous subculturing, cross was performed between the two types and the formation of fruiting body was examined. Ascospores were obtained from a selected fruiting body formed by hybrid of the cross. Self-cross and combinational cross of the ascospore-derived isolates generated hybrids that stably produce high quality fruiting body in artificial media.

Abstract

Inonotus obliquus (Pers.:Fr.) Pil. is a white rot fungus that belongs to the family Hymenochaetaceae of Basidiomycetes. Extracts and fractions of this fungus have been known to have biological activities, including antimutagenic, anticancer, antioxidative, and immunostimulating effects. Recently, there have been reports that the anti-inflammatory and antinociceptive properties of the methanol extract of I. obliquus may be due to the inhibition of inducible nitric oxide (NO) synthase (iNOS) and cyclooxygenase-2 (COX-2) expression via the down-regulation of nuclear factor kappaB (NF-kappaB) binding activity. However, the effects of I. obliquus on Akt and mitogen-activated protein kinase (MAPK) activation of inflammatory mediator production have not yet been elucidated. In the present study, a 70% ethanol extract of I. obliquus (IOE70) showed antioxidative effects. We also tested the ability of the I. obliquus extract to inhibit the inflammatory cascades in lipopolysaccharide (LPS)-induced RAW 264.7 macrophage cells. The NO inhibition of IOE70 was better than that of other ethanol extracts from I. obliquus. To investigate the mechanism by which IOE 70 inhibits NO production and iNOS and COX-2 expression, we examined the activations of IkappaBalpha, Akt, and c-Jun NH(2) -terminal kinase (JNK) in LPS-activated macrophages. IOE70 markedly inhibited the phosphorylation of IkappaBalpha, Akt, and MAPKs in dose-dependent manners in LPS-activated macrophages. Taken together, these experiments demonstrated that IOE70 inhibition of LPS-induced expression of iNOS and COX-2 protein is mediated by Akt and JNK. Based on our findings, the most likely mechanism that can account for this biological effect of IOE70 involves the inhibition of NF-kappaB through the phosphatidylinositol 3-kinase/Akt/IkappaB pathway and the inhibition of JNK activation. Thus, IOE70 might have useful clinical applications in the management of inflammatory diseases and may also be useful as a medicinal food.

Abstract

Higher Fungi were collected twice a month from May to September 2004 during field survey trips to Seoraksan and Odaesan National Parks. All the collected specimens were investigated for the morphological characters of carpophores and other features, and deposited in the herbarium of the Entomopathogenic Fungal Culture Collection (EFCC), Kangwon National University, Chuncheon. Among the identified specimens, three genera Rhodotus, Hotermannia and Sebacina and four species Rhodotus palmatus, Gomphus clavatus, Holtermannia corniformis and Sebacina incrustans were confirmed as new to Korea and reported here with descriptions.

Abstract

Until now, only two Cordyceps species out of more than 500 reported have been shown as having Beauveria as their anamorphs. The first of these, C. brongniartii, was grown in vitro from Coleopteran larvae infected with B. brongniartii in Japan [1]. Nearly two decades later, it was recently reported as wild, growing on Coleopteran larva in Japan [2]. The second species, C. bassiana, was described as growing from a Lepidopteran cadaver in the wild, from China [3, 4]. Recently, C. bassiana has been successfully grown in vitro, with the aim of developing it for commercial use [5-7]. Cordyceps have been a subject of curiosity for hundreds of years. They have been prized for their hidden medicinal values, especially in the Eastern Asian society. A more recent, worldwide interest in these fungi as a medicinal mushrooms has instigated the development of much research. The challenge to develop it commercially is in understanding their growth habit in culture conditions. The growth of C. militaris, the cosmopolitan species, was the first to be attempted in vitro [8, 9]. Ever since then, many researchers have tried to grow its stromata under different nutritional and environmental conditions. Besides the biotic factors, the bipolar heterothallic type of mating system has been shown as the principal genetic factor for fertile stromata production of C. militaris [10]. The mysteries of Cordyceps are slowly being deciphered. A wide range of variation in colony texture, pigmentation and growth rate has been observed in C. militaris [11]. Also, it has been shown that colony pigmentation has no correlation with mating type in C. militaris [12]. C. bassiana isolates produce synnemata very profusely from both single isolates and their combinations, although perithecial stromata are also produced, usually from crossings of mating-compatible isolates [5]. C. bassiana isolates either produce yellow pigmentation or do not produce any pigmentation and remain white. It was also observed that two isolates, when inoculated on opposite sides of agar plates, either avoided each other at the meeting line or overlapped each other and induced thick mycelial growth. Hence, the aim of this study is to understand relationship between fruiting body formation and colony pigmentation and vegetative compatibility in C. bassiana.

Abstract

A specimen of Beauveria bassiana was collected from Yang-yang of Gangwon province, Korea in October 2006. Conidial isolates were prepared from the specimen by the dilution method and inoculated in brown rice medium for fruiting body production. After nearly two months incubation for perithecial stromata developed from single isolates as well as from their combinations. They were determined as Cordyceps bassiana by observing the stromatal characters and their conidial structures. This is the first report of the development of C. bassiana from B. bassiana cultures.

Abstract

Recently, Cordyceps species (Clavicipitaceae, Hypocreales, Ascomycota) have been searched for bio-active compounds by biochemists and pharmacologists to cure different diseases. Cordyceps species are specially known as medicinal mushrooms in East Asian countries, including Korea. Among Cordyceps species, C. militaris is one of the vigorously studied species for the pharmacological values such as immunomodulatory, immunoenhancing and anti-tumor activities (Liu et al., 1997; Yoo et al., 2004; Zhao et al., 2000) and antioxidant, anti-inflammatory, antiangiogenic and antinociceptive effects (Won and Park, 2005). Bio-active compounds such as cordycepin and exopolysaccharides have been extracted from mycelium of Cordyceps militaris in broth culture. For higher production of cordycepin and exopolysaccharides, different nutritional factors have been studied in broth culture of C. militaris (Kim and Yun, 2005; Mao and Zhong, 2006). The limitation for the further production of bio-active compounds from fruiting bodies of Cordyceps species is due to the extreme difficulty in their culture and in vitro production of fruiting bodies. Apart from the submerged culture of mycelium, research on fruiting body production of C. militaris on various media is going on. Recently, in vitro stromata production of C. militaris was successfully established in brown rice medium (Choi et al., 1999; Sung, 1996; Sung et al., 1999, 2002). Other researchers have successively grown C. militaris fruiting bodies on alternative insect pupae (Harada et al., 1995; Sato and Shimazu, 2002). But, unstable fruiting body production poses a serious problem for large scale extraction of bio-active compounds from stromata of C. militaris (Shrestha et al., 2004b; Sung, 1996). More recently, bipolar heterothallism was shown during fruiting body production of C. militaris by inoculating single spore strains in pair-wise combinations in rice pupae medium (Shrestha, 2003; Shrestha et al., 2004a, 2005a, b). Relationship between colony pigmentation and mating type was also investigated by crossing strains with different pigmentation types (Shrestha et al., 2005a). In previous studies, strains of few specimens of C. militaris were used for fruiting body production. In this study, isolates derived from a large number of specimens were used to select potential strains for higher stromata production. Similarly, inter-strain crossing was also made using strains derived from different isolates for fruiting body production and compared with those of intra-strain crossings. Both intra-strain crossings and inter-strain crossings among strains could produce large number of stromata in brown rice medium.

Abstract

Effects of various preservation periods and subcultures on fruiting body formation of Cordyceps militaris were investigated using EFCC C-10995 single ascospore strains. Fruiting body formation by original strains was profuse when preserved at 4℃ for 5~6 months. Fruiting from subcultures was stable till second to sixth subcultures, after which it decreased sharply. The more the colony color of subcultures changed, the less the fruiting bodies formed. Liquid inoculum preparation of single ascospore strains in the same or separate broths did not affect fruiting body formation. Similarly, two strains C-10995-3 and C-10995-6 in different numbers during liquid inoculum preparation produced similar fruiting bodies.
