Age-related diseases, such as osteoarthritis, Alzheimer’s disease, diabetes, and cardiovascular disease, are often associated with chronic unresolved inflammation. Neutrophils play central roles in this process by releasing tissue-degenerative proteases, such as cathepsin G, as well as pro-inflammatory leukotrienes produced by the 5-lipoxygenase (5-LO) pathway. Boswellic acids (BAs) are pentacyclic triterpene acids contained in the gum resin of the anti-inflammatory remedy frankincense that target cathepsin G and 5-LO in neutrophils, and might thus represent suitable leads for intervention with age-associated diseases that have a chronic inflammatory component. Here, we investigated whether, in addition to BAs, other triterpene acids from frankincense interfere with 5-LO and cathepsin G. We provide a comprehensive analysis of 17 natural tetra- or pentacyclic triterpene acids for suppression of 5-LO product synthesis in human neutrophils. These triterpene acids were also investigated for their direct interference with 5-LO and cathepsin G in cell-free assays. Furthermore, our studies were expanded to 10 semi-synthetic BA derivatives. Our data reveal that besides BAs, several tetra- and pentacyclic triterpene acids are effective or even superior inhibitors of 5-LO product formation in human neutrophils, and in parallel, inhibit cathepsin G. Their beneficial target profile may qualify triterpene acids as anti-inflammatory natural products and pharmacological leads for intervention with diseases related to aging.

Das Harz vieler Weihrauch-Arten (Boswellia spp.) enthält neben geruchsaktiven Verbindungen einen beträchtlichen Anteil organischer Säuren. Unter diesen nehmen die Boswelliasäuren eine besondere Stellung ein, denn sie kommen ausschließlich in dieser Gattung vor und zeigen interessante pharmakologische Wirkungen. Schwerpunkte der Arbeit waren die Speziesidentifizierung bei Weihrauchharzen sowie die Identifizierung und Quantifizierung eines Fremdharzes in einer Handelsprobe. Für diese Fragen ist die Säurefraktion das beste Untersuchungsobjekt, weswegen für deren Isolierung aus dem Harzextrakt eine Methode mittels Ionenaustauscher entwickelt wurde. Die Säurefraktion lässt sich dann leicht und in großem Maßstab in eine einzige definierte Boswelliasäure oder ein Derivat umwandeln; für diesen Ansatz wird der Begriff der „fokussierenden Synthese“ eingeführt. Im Zuge der Arbeit wurden außerdem zehn Triterpene aus dem Weihrauchharz in hoher Reinheit isoliert und in ihrer Struktur aufgeklärt., The resin from many frankincense species (Boswellia spp.) contains - apart from the fragrant compounds - a considerable amount of organic acids. Among these the boswellic acids are unique in that they are found only in this genus and show interesting pharmacological effects. Important goals in these studies were the species identification of frankincense resins and the identification and quantification of an adulterant resin in a commercial frankincense sample. As the whole mixture of organic acids is the best starting material for addressing these problems a method was developped for isolating the organic acids with an ion exchanger. The resulting mixture of resin acids can then be easily converted into a single boswellic acid (or derivative) in large amounts; the term „focussing synthesis“ is introduced for this approach. A total of ten triterpenoids were isolated or synthesized and their structure elucidated in the course of this work.

Die Reverse Transkription ist ein Schlüsselschritt im Vermehrungszyklus von HIV-1, der durch neue Non-Nucleosidische Reverse Transkriptase-Inhibitoren blockiert werden kann. Die anti-HIV-Aktivität der Driman-Sesquiterpene Mniopetal A-F sowie der strukturell verwandten Pilz-Metaboliten Kuehneromycin A/B und Panudial ist belegt, die der nahe verwandten Marasmone ist zu vermuten. Im Rahmen der Arbeit wurden die Metaboliten Anhydromarasmon, Kuehneromycin B und Panudial über eine variable und effiziente Synthesestrategie dargestellt. Im Falle der Totalsynthese des Anhydromarasmons war der Schlüsselschritt eine milde Übergangsmetall-induzierte Doppelbindungs-Isomerisierung mit Vaskas Komplex. Zwei weitere Zielmoleküle stellten Kuehneromycin B und Panudial dar, welche durch pH-abhängige Hydrolyse und in-situ-Decarboxylierung aus Kuehneromycin A in hohen Ausbeuten zugänglich wurden., AIDS is currently one of the most important global challenges in the field of infectious diseases. Reverse Transcription (RT) is one of the key steps during the replication cycle of HIV-1 and can be inhibited by novel Non-Nucleosidic Reverse Transcriptase inhibitors (NNRTIs). The anti-HIV-activity of the drimane sesquiterpenes Mniopetal A-F has been established and the inhibitory effect of the closely related Marasmones has been postulated. The fungal metabolites Anhydromarasmone, Kuehneromycin B and Panudial were synthesized using a variable and efficient strategy. In case of the total synthesis of Anhydromarasmone the key step was a mild transition metal-catalyzed double bond isomerization using Vaska's complex. The other two targets, Kuehneromycin B and Panudial, were synthesized from Kuehneromycin A in high yield via pH-dependent hydrolysis and in-situ-decarboxylation.
