1. The Effects of Hydroalcoholic Extract of Matricaria Recutita on the Hormonal Pituitary-Testis Axis and Testis Tissue Changes of Mature Male Rats

Abstract

Background & Objectives: Matricaria recutita is one of the most ancient and well- known medicinal plants, and its role in the treatment of a wide range of diseases has been studied . The purpose of this study was to investigate the effect of Matricaria recutita on spermatogenesis and the pituitary-gonadal axis in male adult rats. Materials & Methods: In this experimental study, the animals were divided into two groups: the control group, which received 1 ml of distilled water orally, and the experimental group, which received 100 mg/kg of Matricaria recutita extract via gavage feeding once daily for an 8-week period. After the treatment period, several fertility indices such as the weight of the reproductive organs, sperm count, sperm motility and vitality, and testis histological changes as well as blood serum levels of testosterone, estrogen, FSH, and LH were measured. Results: Our statistical analysis showed a significant increase in the weight of the reproductive organs, sperm count, and blood testosterone in the group which received 100 mg/kg of Matricaria recutita hydroalcholic extract . Conclusion: The results of this study demonstrated that hydroalcholic extract of Matricaria recutita could increase the function of the hormonal pituitary-testis axis and spermatogenesis in male rats.

2. Study on the immunopathologic effects of 4(3H)-quinazolinone-2-ethyl-2-phenyl ethyl (QEPE) in newborn Balb/C mice

Abstract

Quinazolinones are heterocyclic and water insoluble compounds with various pharmacological and biological characteristics (antibacterial, antiswelling, antifungal, parkinson and etc.). They are used for treatment HIV and cancer. This study investigated the effects of 4(3H) quinazlonones-2-ethyl-2-phenyl ethyl (QEPE) as a new quinazolinons compounds on the spleen and immunocompetent cells of newborn Balb/C mice. Pregnant mice were divided into 3 groups (n=10) of control, sham and experimental, received distilled water, methyl cellulose %0.05 (the solvent) and 100 mg/kg body weight (ip) of QEPE (most effective dose), respectively, on days 8 to 15 of gestation. Examinations indicated an increase in weight of spleen in experimental group. Pathological studies showed increase in capsule thickness and number of macrophage cells of experimental group. Statistical analysis showed significant differences in morphological studies between experimental, sham and control groups. The statistical data on capsule thickness and number of macrophage cells indicated significant differences between groups. Detailed observations showed an increase in the volume of monocyte, neutrophile and eosinophile, in response to QEPE, but the volume of lymphocyte and basophile were similar in experimental, control and sham groups. The damages caused by this dose of QEPE could have been the reason for the increase in the number of immonucompetent and macrophage cells. Some studies showed damages to the organs such as livers and hearts would lead to the increase in the thickness of spleen capsule, consequently, increase in its weight and leading to splenomegaly. So, QEPE can not be an appropriate candidate for drug development. QEPE in lower doses might be an appropriate candidate for increase of immune system resistance.

4. cAMP-dependent protein kinase inhibits α7 nicotinic receptor activity in layer 1 cortical interneurons through activation of D1/D5 dopamine receptors

Abstract

Key points Protein kinases can modify the function of many proteins including ion channels. However, the role of protein kinase A in modifying nicotinic receptors in the CNS has never been investigated. We showed through whole-cell recordings of layer 1 prefrontal cortical interneurons that α7 nicotinic responses are negatively modulated by protein kinase A. Furthermore, we show that stimulation of dopamine receptors can similarly attenuate α7 nicotinic responses through the activation of protein kinase A. These results suggest how the interaction of the cholinergic and dopaminergic systems may influence neuronal excitability in the brain. Abstract Phosphorylation of ion channels, including nicotinic acetylcholine receptors (nAChRs), by protein kinases plays a key role in the modification of synaptic transmission and neuronal excitability. α7 nAChRs are the second most prevalent nAChR subtype in the CNS following α4β2. Serine 365 in the M3–M4 cytoplasmic loop of the α7 nAChR is a phosphorylation site for protein kinase A (PKA). D1/D5 dopamine receptors signal through the adenylate cyclase–PKA pathway and play a key role in working memory and attention in the prefrontal cortex. Thus, we examined whether the dopaminergic system, mediated through PKA, functionally interacts with the α7-dependent cholinergic neurotransmission. In layer 1 interneurons of mouse prefrontal cortex, α7 nicotinic currents were decreased upon stimulation with 8-Br-cAMP, a PKA activator. In HEK 293T cells, dominant negative PKA abolished 8-Br-cAMP's effect of diminishing α7 nicotinic currents, while a constitutively active PKA catalytic subunit decreased α7 currents. In brain slices, the PKA inhibitor KT-5720 nullified 8-Br-cAMP's effect of attenuating α7 nicotinic responses, while applying a PKA catalytic subunit in the pipette solution decreased α7 currents. 8-Br-cAMP stimulation reduced surface expression of α7 nAChRs, but there was no change in single-channel conductance. The D1/D5 dopamine receptor agonist SKF 83822 similarly attenuated α7 nicotinic currents from layer 1 interneurons and this attenuation of nicotinic current was prevented by KT-5720. These results demonstrate that dopamine receptor-mediated activation of PKA negatively modulates nicotinic neurotransmission in prefrontal cortical interneurons, which may be a contributing mechanism of dopamine modulation of cognitive behaviours such as attention or working memory.

Abstract

Understanding how dopaminergic (DA) neurons of the substantia nigra pars compacta (SNc) govern movements requires a detailed knowledge of how different neurotransmitter systems modulate DA neuronal excitability. We report a heterogeneity of electrophysiological properties between medial and lateral SNc neurons modulated by cholinergic neurotransmission. Lateral DA neurons received mainly excitatory (nicotinic or glutamatergic) mediated cholinergic neurotransmission. Medial DA neurons received predominantly GABAergic currents mediated by presynaptic nicotinic receptors or biphasic GABAergic and nicotinic neurotransmission conveyed by GABA and ACh corelease, which inhibited DA neurons. To examine whether cholinergic signaling in the SNc controls mouse behavior, we used optogenetics in awake behaving mice and found that activation of cholinergic terminals in the medial SNc decreased locomotion, whereas activation in the lateral SNc increased locomotion. Our findings provide novel insights on how cholinergic inputs to subregions of the SNc regulate the excitability of DA neurons differentially, resulting in different patterns of motor behavior.

Abstract

Salviaspecies which are medicinal herbs have been used to improve male reproductive functions in traditional medicine. They also, have been used to treat several diseases. In this present study, effects of Salvia hypoleucaon male rat reproductive function were investigated by sperm analysis and assessment of CREM expression at mRNA and protein levels.
