Maintaining adequate planter downforce is critical for achieving timely and uniform crop emergence across the field. On-farm research studies were conducted in different regions across South Georgia, USA to investigate the effect of planter downforce in varying soil textures. Six fields were planted in cotton with four different growers from 2017 to 2019. Soil electrical conductivity (EC) was mapped in each field to characterize areas of varying soil textures into low, medium and high soil EC zones. For each field, the downforce treatments consisted of a grower-preferred downforce along with a relatively lower and higher downforce dependent on prevalent field conditions and the downforce system available on the grower’s planter. Results varied from field to field depending on soil texture variability or the type of downforce system. In three out of six fields, emergence was delayed and reduced in areas of high soil EC (heavy textured soils) whereas soil EC or downforce did not affect crop emergence in two fields. In one field, increasing planter downforce from the grower preferred value of 555 N to 1110 N improved crop emergence in heavy textured soils. In some cases, it was noted that even the highest downforce (1110 N) was not sufficient to place seeds at the desired seeding depth in the heavy textured soils. More on-farm studies are required to thoroughly understand planter downforce response in varying field conditions that exist during planting. Future studies should consider better quantification of in-field soil variability and changes in planter downforce during field operation.

Aims: Determine if the use of novel chlorophyll a fluorescence parameters could be utilized to predict yield loss of cotton exposed to sublethal rates of 2,4-dichlorophenoxyacetic acid (2,4-D) at various growth stages. Study Design: All trials were arranged in a randomized complete block design with four replications. Treatment means were subjected to analysis of variance and linear regression was utilized to determine relationship between chlorophyll a parameters and yield. Place and Duration of Study: University of Georgia Gibbs Farm in Tifton, GA, USA and the Sunbelt Agricultural Exposition in Moultrie, GA, USA during the 2013 growing season. Methodology: Two sublethal rates of 2,4-D were applied to cotton at six distinct growth stages. The rates consisted of 2 g and 40 g ae ha-1 equivalent to 1/421 and 1/21 of the full rate (0.532 kg ae ha-1), respectively. The sublethal rates were applied to cotton at six growth stages, including the four leaf, nine leaf, first bloom, two, four and six weeks after first bloom growth stages. A fluorometer was used to obtain the fluorescence parameters Fv/Fm, ΦEO and PIABS from the uppermost fully expanded leaves at various intervals after 2,4-D exposure. Results: Despite yield losses ranging from 20 – 90% of the non-treated control, no consistent patterns resulted from utilizing fluorescence transients to detect 2,4-D injury and overall instances of significant difference were minimal and typically not biologically relevant. In many cases, treatments exposed to 2,4-D that exhibited yield loss showed evidence of greater photosynthetic efficiency than the non-treated control. In the majority of instances, many of fluorescence parameters measured fell within ranges observed in previous studies in cotton produced under typical or non-stressed conditions. Conclusion: While it has been proven as a valuable tool in other plant screening endeavors, chlorophyll a fluorescence were not able to detect the effects of sub-lethal rates of 2,4-D on cotton, even in instances that resulted in severe yield loss.

Novel yield component traits and fiber quality indices have been recently developed by breeders to screen for desirable cotton (Gossypium hirsutum L.) lines, but assessing these components in response to water-induced yield variability has received little attention. We investigated the hypothesis that differential sensitivities of whole-crop and intra-boll yield components to drought will largely explain water-induced yield and fiber quality variation in cotton. To test this hypothesis, two cotton cultivars were grown in the field under five contrasting irrigation regimes during the 2013 and 2014 growing seasons near Camilla, GA. Measurements included predawn leaf water potential (ΨPD) throughout the growing season and extensive yield component and fiber quality assessments at the end of the season. Water-induced yield variability was primarily associated with ΨPD at the flowering and boll development phase of crop growth. Boll density (bolls ha−1) was the dominant driver of drought-induced yield loss, but reduced boll mass and seed number per boll also contributed somewhat to yield loss. By comparison, increased drought severity decreased fiber density but increased individual fiber mass, producing a peak in total fiber weight per seed at a −0.7 MPa ΨPD irrigation threshold and increasing lint percentage in stressed treatments. Thus, the negative impacts of drought on overall boll mass and seed number per boll are partially offset by increased dry matter partitioning toward fiber growth. Fiber length declined with increased drought severity, whereas fiber micronaire increased in the most severely stressed treatments. This indicates that increasing drought severity during flowering and boll development decreases the number of individual fibers per seed and their final length, but their thickness is increased. The end result is a decline in the overall fiber quality index (Q-score) under drought stress.

Seed mass and oil content of the quiescent cotton seed are positively associated with seedling vigor. In contrast, seed size has been negatively associated with lint yield due to selection for cultivars with greater lint percent. The current study addressed the hypothesis that planting seed mass and total oil + protein calorie content of the quiescent cotton seed would be strongly predictive of seedling vigor across most field conditions and that the impact of seed traits on yield would be dependent upon yield environment. When considered in each yield environment, seedling vigor was positively associated with seed size and the total oil + protein calorie content per seed in four out five environments tested. Regression analysis of cultivar mean oil + protein kcal content per seed versus seedling vigor across all environments indicated a strong, positive relationship between the two parameters (r2 = 0.65). Although lint percent was positively correlated with lint yield in every environment, planting seed mass and calorie content were not correlated with lint yield in four of the five environments tested or when cultivar means for lint yield and seed characteristics were averaged across all environments. Thus, it is concluded that individual planting seed mass and total energy content for oil + protein are strong predictors of early seedling vigor. Furthermore, selecting commercially available cultivars with characteristics indicative of seedling vigor does not appear to limit lint yield in most environments tested.

Temperature and drought are major abiotic limitations to crop productivity worldwide. While abiotic stress physiology research has focused primarily on fully expanded leaves, no studies have investigated photosynthetic tolerance to concurrent drought and high temperature during leaf ontogeny. To address this, Gossypium hirsutum plants were exposed to five irrigation treatments, and two different leaf stages were sampled on three dates during an abnormally dry summer. Early in the growing season, ontogenic PSII heat tolerance differences were observed. Photosystem II was more thermotolerant in young leaves than mature leaves. Later in the growing season, no decline in young leaf net photosynthesis (PN) was observed as leaf temperature increased from 31 to 37°C, as average midday leaf water potential (ΨMD) declined from -1.25 to -2.03MPa. In contrast, mature leaf PN declined 66% under the same conditions. Stomatal conductance (gs) accounted for 84-98% of variability in leaf temperature, and gs was strongly associated with ΨMD in mature leaves but not in young leaves. We conclude that young leaves are more photosynthetically tolerant to heat and drought than mature leaves. Elucidating the mechanisms causing these ontogenic differences will likely help mitigate the negative impacts of abiotic stress in the future.

Much effort has been expended to improve irrigation efficiency and drought tolerance of agronomic crops; however, a clear understanding of the physiological mechanisms that interact to decrease source strength and drive yield loss has not been attained. To elucidate the underlying mechanisms contributing to inhibition of net carbon assimilation under drought stress, three cultivars of Gossypium hirsutum were grown in the field under contrasting irrigation regimes during the 2012 and 2013 growing season near Camilla, Georgia, USA. Physiological measurements were conducted on three sample dates during each growing season (providing a broad range of plant water status) and included, predawn and midday leaf water potential (ΨPD and ΨMD), gross and net photosynthesis, dark respiration, photorespiration, and chlorophyll a fluorescence. End-of-season lint yield was also determined. ΨPD ranged from -0.31 to -0.95MPa, and ΨMD ranged from -1.02 to -2.67MPa, depending upon irrigation regime and sample date. G. hirsutum responded to water deficit by decreasing stomatal conductance, increasing photorespiration, and increasing the ratio of dark respiration to gross photosynthesis, thereby limiting PN and decreasing lint yield (lint yield declines observed during the 2012 growing season only). Conversely, even extreme water deficit, causing a 54% decline in PN, did not negatively affect actual quantum yield, maximum quantum yield, or photosynthetic electron transport. It is concluded that PN is primarily limited in drought-stressed G. hirsutum by decreased stomatal conductance, along with increases in respiratory and photorespiratory carbon losses, not inhibition or down-regulation of electron transport through photosystem II. It is further concluded that ΨPD is a reliable indicator of drought stress and the need for irrigation in field-grown cotton.

During the 2018 growing season, cotton plants in several fields in Georgia were observed with symptoms that included leaf curling, reddening and drooping of leaves, subsequent distortion of leaf growth above the nodes where reddened leaves were first observed, and shortening of upper internodes and their discoloration to deep green. These symptoms were visible in the upper portion of the plants, resembling “cotton blue” disease caused by cotton leafroll dwarf virus (CLRDV) (family Luteoviridae, genus Polerovirus). The symptoms were observed sporadically with less than 10% disease incidence within affected fields. CLRDV is a phloem-limited virus with single-stranded, positive-sense RNA genome, transmitted by aphids (Aphis gossypii) in a persistent, circulative, nonpropagative manner (Distefano et al. 2010; Sharman et al. 2015). A total of 93 symptomatic and asymptomatic leaves and petioles were collected from several different commercial cotton fields in the southern part of the state. Total RNA was extracted using the modified cetyltrimethylammonium bromide method (Sharman et al. 2015). Complementary DNA (cDNA) was synthesized from 2.5 µg of total RNA using Superscript III reverse transcription (Invitrogen, U.S.A.) and specific reverse primers targeting different open reading frames (ORFs) of the virus genome. The cDNA was used for polymerase chain reaction (PCR) with primers CLRDV3675F and Pol3982R targeting ORF 3 of the virus (310-bp product, Sharman et al. 2015) and primers 17 and 18 targeting ORF 5 (1,065-bp product, Distefano et al. 2010). A new set of primers, SB11F (5′-AGGTTTTCTGGTAGCAGTACCAATATCAACGTTA-3′) and SB11R (5′-TATCTTGCATTGTGGATTTCCCTCATAA-3′), was developed to amplify the 803-bp fragment spanning complete ORF 3 and ORF 4, encoding virus coat protein and movement protein genes. Using the three different sets of primers, products of predicted sizes were amplified from the symptomatic tissues, but not from asymptomatic tissues collected from fields and negative controls including asymptomatic leaves from plants grown in an insect-free controlled growth chamber and DNase/RNase free water (Invitrogen). Reverse transcription PCR products were gel purified, cloned into TOPO-TA cloning vector (Invitrogen), and sequenced using Sanger sequencing (GeneScript, NJ). The consensus sequence of three clones for ORF 3 and 4 from this study (MK290759 and MK290760) when analyzed using NCBI-BLASTn showed 97% sequence identity with the Brazilian and Argentinean isolates (KF906261.1 and KF359947.1, respectively) and many other isolates from South American and Asian countries. ORF 3 and 4 sequences from Georgia shared 98 to 99% identity among themselves. The partial sequences for ORF 5 (MK513938 and MK513939) shared 95% identity with the isolate from Argentina (KF359946.1 and KF359947.1). The presence of the virus was detected in all the symptomatic samples collected from 14 counties in Georgia, covering the entire cotton growing region of the state. To the best of our knowledge, this is the first report of CLRDV in Georgia, U.S.A. CLRDV infection of cotton has been reported in other cotton-growing regions in Africa, Asia, South America, and, more recently, in Alabama, U.S.A. (Avelar et al. 2018; Correa et al. 2005; Distefano et al. 2010; Mukherjee et al. 2012). With its rapid spread, this virus could pose an imminent threat and cause severe economic losses to the cotton industry. Further research is needed to elucidate epidemiology, symptomatology, vector transmission, and crop losses owing to CLRDV in the United States.

Despite exhaustive literature describing drought stress effects on photosynthesis in Gossypium hirsutum, the sensitivity of photosynthetic electron flow to water deficit is heavily debated. To address this, G. hirsutum plants were grown at a field site near Camilla, GA under contrasting irrigation regimes, and pre-dawn water potential (ΨPD), stomatal conductance (gs), net photosynthesis (PN), actual quantum yield of photosystem II (ΦPSII) and electron transport rate (ETR) were measured at multiple times during the 2012 growing season. ΨPD values ranged from −0.3 to −1.1 MPa. Stomatal conductance exhibited a strong (r2 = 0.697), sigmoidal response to ΨPD, where gs was ≤0.1 mol m−2 s−1 at ΨPD values ≤ −0.86 MPa. Neither ΦPSII (r2 = 0.015) nor ETR (r2 = 0.010) was affected by ΨPD, despite exceptionally low ΨPD values (−1.1 MPa) causing a 71.7 % decline in PN relative to values predicted for well-watered G. hirsutum leaves at ΨPD = −0.3 MPa. Further, PN was strongly influenced by gs, whereas ETR and ΦPSII were not. We conclude that photosynthetic electron flow through photosystem II is insensitive to water deficit in field-grown G. hirsutum.

A standardized experiment was conducted during 2009 and 2010 at 20 location-years across U.S. cotton (Gossypium hirsutum L.)-producing states to compare the N use requirement of contemporary cotton cultivars based on their planting seed size. Treatments consisted of three cotton varieties with planting seed of different numbers of seed per kg and N rates of 0, 45, 90, and 134 kg ha–¹. Soil at each trial location was sampled and tested for nitrate presence. High levels of soil nitrate (>91 N-NO₃– kg ha–¹) were found in Arizona and western Texas, and soil nitrate in the range of 45 to 73 kg N-NO₃– ha–¹ was found at locations in the central United States. Cotton lint yield responded to applied N at 11 of 20 locations. Considering only sites that responded to applied N, highest lint yields were achieved with 112 to 224 kg ha–¹of applied plus pre-plant residual soil NO₃—translating to an optimal N requirement of 23 kg ha–¹ per 218 kg bale of lint produced. Among the varieties tested those with medium-sized seed produced higher yields in response to N than did larger and smaller seeded varieties. Varieties with larger seed had longer and stronger fibers, higher fiber length uniformity than small seeded varieties and decreased micronaire. Seed protein and oil increased and decreased slightly in response to increasing amounts of soil nitrate plus applied N, respectively.

Glyphosate-resistant (GR) biotypes of Palmer amaranth are now commonly found across the southern United States. Experiments were conducted to characterize physiological differences between a GR biotype and a glyphosate-susceptible (GS) biotype from North Carolina. The GR biotype had an 18-fold level of resistance based upon rates necessary to reduce shoot fresh weight 50%. Shikimate accumulated in both biotypes following glyphosate application, but greater concentrations were found in GS plants. Absorption and translocation of14C-glyphosate were studied in both biotypes with and without an overspray with commercial glyphosate potassium salt (840 g ae ha−1) immediately prior to14C-glyphosate application. Greater absorption was noted 6 h after treatment (HAT) in GS compared with GR plants, but no differences were observed at 12 to 72 HAT. Oversprayed plants absorbed 33 and 61% more14C by 48 and 72 HAT, respectively, than plants not oversprayed.14C distribution (above treated leaf, below treated leaf, roots) was similar in both biotypes. Together, these results suggest that resistance in this biotype is not due to an altered target enzyme or translocation but may be in part due to the rate of glyphosate absorption. These results also are consistent with resistance being due to increased gene copy number for the target enzyme.

Glyphosate resistance in Palmer amaranth was first confirmed in North Carolina in 2005. A survey that year indicated 17 and 18% of 290 populations sampled were resistant to glyphosate and thifensulfuron, respectively. During the fall of 2010, 274 predetermined sites in North Carolina were surveyed to determine distribution of Palmer amaranth and to determine if and where resistance to fomesafen, glufosinate, glyphosate, and thifensulfuron occurred. Palmer amaranth was present at 134 sites. When mortality for each biotype was compared to a known susceptible biotype for each herbicide within a rate, 93 and 36% of biotypes were controlled less by glyphosate (840 g ae ha−1) and thifensulfuron (70 g ai ha−1), respectively. This approach may have underestimated resistance for segregating populations due to lack of homogeneity of the herbicide resistance trait and its contribution to error variance. When mortality and visible control were combined, 98% and 97% of the populations were resistant to glyphosate and the ALS inhibitor thifensulfuron, respectively, and 95% of the populations expressed multiple resistance to both herbicides. This study confirms that Palmer amaranth is commonly found across the major row crop production regions of North Carolina and that resistance to glyphosate and ALS-inhibiting herbicides is nearly universal. No resistance to fomesafen or glufosinate was observed.

Inheritance of glyphosate resistance in a Palmer amaranth biotype from North Carolina was studied. Glyphosate rates for 50% survival of glyphosate-resistant (GR) and glyphosate-susceptible (GS) biotypes were 1288 and 58 g ha−1, respectively. These values for F1 progenies obtained from reciprocal crosses (GR×GSandGS×GRwere 794 and 501 g ha−1, respectively. Dose response of F1 progenies indicated that resistance was not fully dominant over susceptibility. Lack of significant differences between dose responses for reciprocal F1 families suggested that genetic control of glyphosate resistance was governed by nuclear genome. Analysis of F1 backcross (BC1F1) families showed that 10 and 8 BC1F1 families out of 15 fitted monogenic inheritance at 2000 and 3000 g ha−1glyphosate, respectively. These results indicate that inheritance of glyphosate resistance in this biotype is incompletely dominant, nuclear inherited, and might not be consistent with a single gene mechanism of inheritance. Relative 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) copy number varied from 22 to 63 across 10 individuals from resistant biotype. This suggested that variableEPSPScopy number in the parents might be influential in determining if inheritance of glyphosate resistance is monogenic or polygenic in this biotype.

Palmer amaranth (Amaranthus palmeriS. Wats.) has become difficult to control in row crops due to selection for biotypes that are no longer controlled by acetolactate synthase inhibiting herbicides and/or glyphosate. Early season interference in soybean [Glycine max(L.) Merr.] for 40 days after emergence by three glyphosate-resistant (GR) and three glyphosate-susceptible (GS) Palmer amaranth biotypes from Georgia and North Carolina was compared in the greenhouse. A field experiment over 2 years compared season-long interference of these biotypes in soybean. The six Palmer amaranth biotypes reduced soybean height similarly in the greenhouse but did not affect soybean height in the field. Reduction in soybean fresh weight and dry weight in the greenhouse; and soybean yield in the field varied by Palmer amaranth biotypes. Soybean yield was reduced 21% by Palmer amaranth at the established field density of 0.37 plant m−2. When Palmer amaranth biotypes were grouped by response to glyphosate, the GS group reduced fresh weight, dry weight, and yield of soybean more than the GR group. The results indicate a possible small competitive disadvantage associated with glyphosate resistance, but observed differences among biotypes might also be associated with characteristics within and among biotypes other than glyphosate resistance.

An alternative design for a particulate sampler will be presented. Two major distinctions separate this design from previous particulate samplers. First, a single body would be used rather than separate compartments for each sample. This has been proposed in order to reduce weight. The second distinction would be the abandoning of a motor in favor of a spring in order to provide the motive force of the device. It is hoped that this will again reduce weight and reduce complications in control. These changes from the standard design present other obstacles which must, in turn, be overcome. This paper is meant to evaluate the relative advantages and disadvantages of this design when compared to other, more standard, designs.
