39. 2-phenylthio-3,3,3-trifluoropropene, its sulfoxide or sulfone in Diels-Alder cycloadditions

Abstract

The Diels-Alder reaction of 2-phenylthio-3,3,3-trifluoropropene 1 and its derivatives sulfoxide 2 and sulfone 3, respectively was carried out with cyclopentadiene, 2,3-dimethylbutadiene, butadiene and isoprene to give the [4+2] cycloadducts in good to excellent yields. The particular reactivity of 2-phenylsulfinyl-3,3,3-trifluoropropene 2 is revealed as an alpha,beta-isomerisation via cycloaddition, sulfenic acid elimination, readdition followed by retro Diels-Alder. Abstract

The trifluoromethylated enamines 1 and 2 condense with monochloromethyleneiminium chlorides, N,N-dimethylacetamide dimethylacetal and chloroiminium chlorides prepared from lactams and gaseous phosgene to give the new 2-aza-1,3-dienes 5a-c, 6, 13a-f, 19a-c and 20a-c which then cyclize with sodium hydride to pyridin-4-ones 22 and 23a,b,e,f.

Abstract

The trifluoromethylated enamines 4 and 5 obtained from 3-trifluoroacetyl-pyrrolidinone 1 and ammonia or methylamine, are good precursors for the synthesis of new 4-azapentamethine cyanines 8-10. These 1,5-bis-electrophiles condense with ammonia or phenylhydrazine to give 5H-pyrrolo[2,3-d] pyrimidines 18, 19, pyrimidinone 20 and pyrrolo-[3,2-f]-1,2,4-triazepines 21, 22. The structure and configuration of all the heterocycles are confirmed by their NMR and UV/vis data and by comparison with analogous compounds.

3-Trifluoroacetyl- and other 3-acyl-1-methyl-lactams (1-5) undergo the Robinson annelation with alpha,beta-unsaturated ketones to produce spirocyclic cyclohexenones (6), their precursors hydroxycyclohexanones (7) or 3-acyl-3-oxobutyl-lactams (8) depending on the substitution.

The hydroxylation of Baylis-Hillman adducts 1 is shown to proceed with high levels of diastereocontrol. A mechanistic rationale to explain this selectivity is discussed.

Abstract

The title reactions lead to unique diastereoisomers of pyrrolizidines 1a [dimethyl 2,3-dehydro-1-methyl-3-cyanopyrrolizidine-1,2-dicarboxylate] or 1b [dimethyl 2,3-dehydro-1-(ethoxycarbonylmethyl)1-3-phenyl-pyrrolizidine-1,2-dicarboxylate] with relative stereochemistry (1a: trans; 1b: cis) established by X-ray diffraction analysis. The heterocycle adopts a different conformation in 1a and 1b.

Abstract

The synthesis of new vinyldiazomethanes from ethyl 2-diazo-4,4,4-trifluoroacetoacetate and Wittig or Horner-Emmons-Wadsworth (HEW) reagents is described. Vinyldiazomethanes which have a double bond bearing a carbonyl group undergo reductive cyclization in the presence of triphenylphosphine providing a new route to the pyridazine chemistry.

Abstract

Heterocyclisations of 1,4-bis-nucleophiles with the tetrahydroazepin-2-one 1 and gamma-butyrolactone 2 gave imidazolidine 9, 12, oxazolidines 3, 10, 13, 15 and thiazolidine 4 as a mixture of two diastereoisomers in various proportions. The structure and the relative configurations of these heterocycles were established by X-ray diffraction analysis and by comparison of F-19- and C-13-NMR data.

Abstract

The syntheses of alpha- and beta-trifluoromethyl epoxysulfones 1 and 2 are described. Compound 1 reacts with nucleophiles and bis-nucleophiles to furnish trifluoromethyl ketones and triRuoromethyl heterocycles in good yield, while its isomer 2 leads to the opposite thiazole regioisomers with thioamides.
