Microsatellite loci are commonly used markers in population genetic studies. In this study, we present 40 novel and polymorphic microsatellite loci elaborated for the ichneumonid parasitoid Latibulus argiolus (Rossi, 1790). Reaction condition optimisation procedures allowed 14 of these loci to be co-amplified in two PCRs and loaded in two multiplex panels onto a genetic analyser. The assay was tested on 197 individuals of L. argiolus originating from ten natural populations obtained from the host nests of paper wasps. The validated loci were polymorphic with high allele numbers ranging from eight to 27 (average 17.6 alleles per locus). Both observed and expected heterozygosity values were high, ranging between 0.75 and 0.92 for HO (mean 0.83) and from 0.70 to 0.90 for HE (mean 0.85). The optimized assay showed low genotyping error rate and negligible null allele frequency. The designed multiplex panels could be successfully applied in relatedness analyses and genetic variability studies of L. argiolus populations, which would be particularly interesting considering the coevolutionary context of this species with its social host.

The discovery of two undisturbed caves in Jamaica with 14C and 210Pb dating indicating that the oldest layers of guano were ca. 200 years old in the first cave (81 cm long core), and as much as 4300 years old in the second cave (129 cm long core) provides exciting possibilities to examine past ecological communities. We analyzed genetic and pollen profiles in these ancient bat guano deposits and revealed DNA sequences most similar to arthropods, mainly insects such as caddisflies (Trichoptera), butterflies (Lepidoptera) and mayflies (Ephemeroptera), suggesting a potential source for past dietary data. Palynological analysis failed to recover well-preserved pollen grains and spores older than ca. 200 years, however in layers preserving pollen, angiosperms were the most abundant plants observed, with a regular occurrence of the potato (Solanaceae) and pepper families (Piperaceae), which are frequently bat-dispersed. In general, changes in frequency of particular plant taxa appear to reflect changes of vegetation and land use in the cave vicinity; however, some changes could be linked to hurricanes, leading to forest canopy damage and promoting the growth of light-demanding species such as guarumo (Cecropia). Higher amounts of mangrove (Rhizophora) pollen have also been recorded in the periods of heavy hurricane activities. Our data highlight the value of bat guano deposits in caves as a resource for the analysis of past ecological systems and stress the conservation values of these deposits.

Background Colourless microalgae of the Prototheca genus are the only known plants that have consistently been implicated in a range of clinically relevant opportunistic infections in both animals and humans. The Prototheca algae are emerging pathogens, whose incidence has increased importantly over the past two decades. Prototheca wickerhamii is a major human pathogen, responsible for at least 115 cases worldwide. Although the algae are receiving more attention nowadays, there is still a substantial knowledge gap regarding their biology, and pathogenicity in particular. Here we report, for the first time, the complete nuclear genome, organelle genomes, and transcriptome of the P. wickerhamii type strain ATCC 16529. Results The assembled genome size was of 16.7 Mbp, making it the smallest and most compact genome sequenced so far among the protothecans. Key features of the genome included a high overall GC content (64.5%), a high number (6081) and proportion (45.9%) of protein-coding genes, and a low repetitive sequence content (2.2%). The vast majority (90.6%) of the predicted genes were confirmed with the corresponding transcripts upon RNA-sequencing analysis. Most (93.2%) of the genes had their putative function assigned when searched against the InterProScan database. A fourth (23.3%) of the genes were annotated with an enzymatic activity possibly associated with the adaptation to the human host environment. The P. wickerhamii genome encoded a wide array of possible virulence factors, including those already identified in two model opportunistic fungal pathogens, i.e. Candida albicans and Trichophyton rubrum, and thought to be involved in invasion of the host or elicitation of the adaptive stress response. Approximately 6% of the P. wickerhamii genes matched a Pathogen-Host Interaction Database entry and had a previously experimentally proven role in the disease development. Furthermore, genes coding for proteins (e.g. ATPase, malate dehydrogenase) hitherto considered as potential virulence factors of Prototheca spp. were demonstrated in the P. wickerhamii genome. Conclusions Overall, this study is the first to describe the genetic make-up of P. wickerhamii and discovers proteins possibly involved in the development of protothecosis.

Of the Prototheca genus, Prototheca wickerhamii has the highest clinical significance in humans. However, neither nuclear nor organellar genomes of this species were sequenced until now. The hitherto determined and analyzed mitochondrial and plastid genomes of the alleged P. wickerhamii species belong in fact to another species, recently named Prototheca xanthoriae. This study provides a first insight into the organellar genomes of a true P. wickerhamii (type strain ATCC 16529). The P. wickerhamii mitochondrion had a 53.8-kb genome, which was considerably larger than that of Prototheca ciferrii (formerly Prototheca zopfii gen. 1) and Prototheca bovis (formerly Prototheca zopfii gen. 2), yet similarly functional, with the differences in size attributable to a higher number of introns and the presence of extra unique putative genes. The 48-kb plastid genome of P. wickerhamii, compared to autotrophic Trebouxiophyceae, was highly reduced due to the elimination of the photosynthesis-related genes. The gene content of the plastid genome of P. wickerhamii was, however, very similar to other colourless Prototheca species. Plastid genome-based phylogeny reinforced the polyphyly of the genus Prototheca, with Helicosporidium and Auxenochlorella branching within clades of Prototheca species. Phylogenetic reconstruction also confirmed the close relationship of P. wickerhamii and P. xanthoriae, which is reflected in the synteny of their organellar genomes. Interestingly, the entire set of atp genes was lost in P. wickerhamii plastid genome, while being preserved in P. xanthoriae.

Bats are currently killed in large numbers at wind turbines worldwide, but the ultimate reason why this happens remains poorly understood. One hypothesis is that bats visit wind turbines to feed on insects exposed at the turbine towers. We used single molecule next generation DNA sequencing to identify stomach contents of 18 bats of four species (Pipistrellus pygmaeus, Nyctalus noctula, Eptesicus nilssonii and Vespertilio murinus) found dead under wind turbines in southern Sweden. Stomach contents were diverse but included typically diurnal flies, e.g. blow-flies (Calliphoridae), flesh-flies (Sarcophagidae) and houseflies (Muscidae) and also several flightless taxa. Such prey items were eaten by all bat species and at all wind turbine localities and it seems possible that they had been captured at or near the surface of the turbines at night. Using sticky traps, we documented an abundance of swarming (diurnal) ants (Myrmica spp.) and sometimes blow-flies and houseflies at the nacelle house. Near the base of the tower the catches were more diverse and corresponded better with the taxa found in the bat stomachs, including various diurnal flies. To evaluate if flies and other insects resting on the surface of a wind turbine are available to bats, we ensonified a house fly (Musca) on a smooth (plastic) surface with synthetic ultrasonic pulses of the frequencies used by the bat species that we had sampled. The experiment revealed potentially useful echoes, provided the attack angle was low and the frequency high (50–75 kHz). Hence resting flies and other arthropods can probably be detected by echolocating bats on the surface of a wind turbine. Our findings are consistent with published observations of the behavior of bats at wind turbines and may actually explain the function of some of these behaviors.

Bursaphelenchus piceae sp. n. is described from the bark of Norway spruce, Picea abies. Adults and propagative juveniles of the nematode are found in larval galleries of the six-toothed spruce bark beetle, Pityogenes chalcographus, while its dauer juveniles aggregate in Malpighian tubules of older larvae, pupae and adult beetles. The new species is characterised by the body length of 719 (530-945) μm in female and 632 (474-808) μm in male, its moderately slender body (a = 41.4 (35.4-50.2) and 42.6 (35.1-47.2) in female and male, respectively), and small spicules (16.0 (14.0-18.3) μm along arc and 13.7 (12.2-15.5) μm along chord). The extended anterior vulval lip in female, lateral fields with four incisures, long post-uterine sac extending for 67-91% of vulva-anus distance, and number (7) and arrangement of male caudal papillae suggest that B. piceae sp. n. is closely related to the xylophilus-group yet it differs by the relatively small, claw-like spicules with no cucullus at the tip. The close relation of B. piceae sp. n. to the xylophilus-group has been confirmed by DNA sequencing and phylogenetic analysis. Morphologically B. piceae sp. n. most closely resembles B. tokyoensis and B. fagi but can be separated from both by the unique shape of the spicules. The taxonomic separation of the new species is also confirmed by the unique molecular profile of the ITS region (ITS-RFLP). The presence of dauer juveniles of B. piceae sp. n. in Malpighian tubules of adult P. chalcographus may lead to extensive damage to this organ as shown by local expansion of the tubule basement membrane and degradation of its cellular epithelium. Detailed phylogenetic analysis revealed that B. piceae sp. n. together with five other bark beetle-associated Bursaphelenchus species, namely B. trypophloei, B. masseyi, B. tiliae, B. tokyoensis and B. fagi, constitutes a small phylogenetic clade that is most closely located to, but separate from, the xylophilus-group.

We describe here the successful coupling of real-time polymerase chain reaction (PCR) and high-resolution melting (HRM) analysis to rapidly identify 15 forensically important species of blowfly from the family Calliphoridae (Diptera), which occur in Poland. Two short regions (119 and 70 base pairs, respectively) of cytochrome oxidase gene subunit I with sufficient sequence diversity were selected. In the case of lacking taxa (e.g., reference species) these amplicons can be synthesized using sequences deposited in gene banks. The technique utilizes low template DNA concentration and is highly reproducible. The melting profile was not altered up to a 10,000-fold difference in DNA template concentration (ranging from 5 pg to 50 ng). The several HRM runs performed on different specimens from Poland belonging to the same species and on different days resulted in only minor variations in the amplification curves and in melting temperatures of the peaks. Intraspecific variation in a larger scale was tested using synthesized oligonucleotides from cosmopolitan Lucilia illustris originating from Poland, France, Great Britain, India, and USA. As HRM PCR analysis is sensitive to even single base changes, all geographic variants of this species were identified. This technique is also cost-effective and simple, and it may even be used by non-geneticists. A working protocol was ultimately constructed for the purpose of rapid and accurate species identification in most countries in Europe regardless of which stage or which part of a blowfly was collected.

Descriptions of the developmental stages of Sarcophaga (Liopygia) argyrostoma (R.-D.) are given. Scanning electron microscope images of most of its immature stages are presented for the first time. The sequence of mitochondrial cytochrome c oxidase subunit I (COI) gene fulfilling DNA barcoding standards is presented for the first time.

The abiotic conditions of the desert habitat fluctuate in a circadian rhythm of hot days and cold nights. Species living in desert habitats evolved many adaptations to increase their chances of survival. However, abiotic conditions in xerothermic habitats of a temperate climate are much different. Diurnal fluctuations are not as strong, but animals have to cope with seasonal changes and hibernate during the winter, which may potentially influence their adaptations to critical temperature conditions. We attempted to assess heat resistance adaptations using the example of a widely distributed xerothermic ant Formica cinerea. Using Real-Time PCR, we measured the expression of three heat shock protein genes (Hsp60, Hsp75, Hsp90) and assessed the adaptations of F. cinerea to enable foraging in risk prone conditions. The analysis of gene expression using the Generalized Linear Model surprisingly indicated that there was no significant effect of temperature when comparing workers from the control (23ºC) with workers foraging on the surface of hot sand (47-54ºC). As a next step we tried to estimate the threshold of a thermal resistance with the use of thermal chambers. Expression of all Hsps genes increase compare to the control group, expression of Hsp60 and Hsp90 continued up to 45ºC.

Based on published information about the glacial, postglacial, and recent distribution of the root vole, Microtus oeconomus, we hypothesized that a population inhabiting the pristine wetland in eastern Poland (Bialowieza Primeval Forest) might comprise a high diversity of haplotypes. The support for this hypothesis was provided by an analysis of partial cytb gene sequences from 149 voles sampled within a two-hectare plot during a nine-year study. In this population, we identified eight haplotypes (PLB1–PLB8), four of which were new to the root vole. These haplotypes were characterized by low nucleotide diversity (π = 0.0054, SE = 0.0019), the absence of transversional differences between sequences, and no changes in the encoded amino acid sequence: features suggesting a lack of immigration from the distant populations. The haplotype number and their frequency distribution in males and females did not differ significantly. An assessment of the persistence of matrilines in the local population throughout the study period revealed that the haplotype composition was relatively stable for only about 3 years. A more complete haplotype network for root voles in Europe was constructed by combining the newly identified haplotypes with the 45 previously described haplotypes. Two of the haplotypes detected in this study occupy key positions in this network: PLB5, as the closest link to the North European group, and PLB8, as an ancestor to many other Central European haplotypes.
