1. Genetically conditioned interaction among microRNA‐155, alpha‐klotho, and intra‐renal RAS in male rats: Link to CKD progression.

Abstract

Incident chronic kidney disease (CKD) varies in populations with hypertension of similar severity. Proteinuria promotes CKD progression in part due to activation of plasminogen to plasmin in the podocytes, resulting in oxidative stress‐mediated injury. Additional mechanisms include deficiency of renal alpha‐klotho, that inhibits Wnt/beta‐catenin, an up regulator of intra‐renal renin angiotensin system (RAS) genes. Alpha‐klotho deficiency therefore results in upregulation of the intra‐renal RAS via Wnt/beta‐catenin. In hypertensive, Dahl salt sensitive (DS) and spontaneously hypertensive rats (SHR), we investigated renal and vascular injury, miR‐155, AT1R, alpha‐klotho, and TNF‐α. Hypertensive high salt DS (DS‐HS), but not SHR developed proteinuria, plasminuria, and glomerulosclerosis. Compared to DS low salt (DS‐LS), in hypertensive DS‐HS alpha‐klotho decreased 5‐fold in serum and 2.6‐fold in kidney, whereas serum mir‐155 decreased 3.3‐fold and AT1R increased 52% in kidney and 77% in aorta. AT1R, alpha‐klotho, and miR‐155 remained unchanged in prehypertensive and hypertensive SHR. TNF‐α increased by 3‐fold in serum and urine of DS‐HS rats. These studies unveiled in salt sensitive DS‐HS, but not in SHR, a genetically conditioned dysfunction of the intermolecular network integrated by alpha‐klotho, RAS, miR‐155, and TNF‐α that is at the helm of their end‐organ susceptibility while plasminuria may participate as a second hit. Abstract

Onco-nephrology is an emerging field in medicine. Patients with cancer may suffer from kidney diseases because of the cancer itself and cancer-related therapy. It is critical for nephrologists to be knowledgeable of cancer biology and therapy in order to be fully integrated in the multidisciplinary team and optimally manage patients with cancer and kidney diseases. In a recent international meeting, the key issues in this challenging clinical interface were addressed, including many unresolved basic science questions, such as the high tumor incidence in kidney transplant recipients. To this end, 70 highly qualified faculty members were gathered from all over the world to discuss these issues in 8 plenary sessions, including 5 keynote lectures. In addition, 48 young nephrologists and oncologists were invited to present their original observations that were highlighted in 2 large poster sessions.

15. COX-2-derived prostaglandins as mediators of the deleterious effects of nicotine in chronic kidney disease.

Abstract

Tobacco smoking has been identified as a risk factor in the progression of chronic kidney disease (CKD). In previous studies, we showed that nicotine induces cyclooxygenase (COX)-2 expression in vivo and in vitro and that the administration of nicotine in vivo worsens the severity of renal injury in a model of subtotal renal ablation. In the present study, we tested the role of COX-2-derived prostaglandins on the deleterious effects of nicotine in CKD. Sham and 5/6 nephrectomy (5/6Nx) rats received tap water or nicotine (100 μg/mL) in the drinking water for 12 wk. Additional groups also systemically received the COX-2 inhibitor NS-398 (1.5 mg·kg-1·day-1 via osmotic minipump). The administration of nicotine worsened renal injury and proteinuria in 5/6Nx rats and increased proteinuria in sham rats. 5/6Nx rats had increased cortical production of the prostaglandins PGE2, PGI2, PGD2, and PGF2α and of thromboxane A2. In these rats, nicotine reduced the production of all prostaglandins examined except thromboxane A2. Treatment with the COX-2 inhibitor NS-398 resulted in complete inhibition of all prostaglandins studied and ameliorated renal injury and proteinuria in 5/6Nx rats on nicotine but not in 5/6 Nx rats on tap water. Nicotine also reduced the expression of megalin in all groups examined, and this was partially prevented by COX-2 inhibition. In the present study, we showed that in CKD, nicotine worsens renal injury at least in part by producing an imbalance in the production of prostaglandins. This imbalance in the production of prostaglandins likely plays a role in the deleterious effects of smoking on the progression of CKD. [ABSTRACT FROM AUTHOR]

Abstract

International audience; Large-scale climate patterns influenced temperature and weather patterns around the globe in 2011. In particular, a moderate-to-strong La Nina at the beginning of the year dissipated during boreal spring but reemerged during fall. The phenomenon contributed to historical droughts in East Africa, the southern United States, and northern Mexico, as well the wettest two-year period (2010-11) on record for Australia, particularly remarkable as this follows a decade-long dry period. Precipitation patterns in South America were also influenced by La Nina. Heavy rain in Rio de Janeiro in January triggered the country's worst floods and landslides in Brazil's history. The 2011 combined average temperature across global land and ocean surfaces was the coolest since 2008, but was also among the 15 warmest years on record and above the 1981-2010 average. The global sea surface temperature cooled by 0.1 degrees C from 2010 to 2011, associated with cooling influences of La Nina. Global integrals of upper ocean heat content for 2011 were higher than for all prior years, demonstrating the Earth's dominant role of the oceans in the Earth's energy budget. In the upper atmosphere, tropical stratospheric temperatures were anomalously warm, while polar temperatures were anomalously cold. This led to large springtime stratospheric ozone reductions in polar latitudes in both hemispheres. Ozone concentrations in the Arctic stratosphere during March were the lowest for that period since satellite records began in 1979. An extensive, deep, and persistent ozone hole over the Antarctic in September indicates that the recovery to pre-1980 conditions is proceeding very slowly. Atmospheric carbon dioxide concentrations increased by 2.10 ppm in 2011, and exceeded 390 ppm for the first time since instrumental records began. Other greenhouse gases also continued to rise in concentration and the combined effect now represents a 30% increase in radiative forcing over a 1990 baseline. Most ozone depleting substances continued to fall. The global net ocean carbon dioxide uptake for the 2010 transition period from El Nino to La Nina, the most recent period for which analyzed data are available, was estimated to be 1.30 Pg C yr(-1), almost 12% below the 29-year long-term average. Relative to the long-term trend, global sea level dropped noticeably in mid-2010 and reached a local minimum in 2011. The drop has been linked to the La Nina conditions that prevailed throughout much of 2010-11. Global sea level increased sharply during the second half of 2011. Global tropical cyclone activity during 2011 was well-below average, with a total of 74 storms compared with the 1981-2010 average of 89. Similar to 2010, the North Atlantic was the only basin that experienced above-normal activity. For the first year since the widespread introduction of the Dvorak intensity-estimation method in the 1980s, only three tropical cyclones reached Category 5 intensity level-all in the Northwest Pacific basin. The Arctic continued to warm at about twice the rate compared with lower latitudes. Below-normal summer snowfall, a decreasing trend in surface albedo, and above-average surface and upper air temperatures resulted in a continued pattern of extreme surface melting, and net snow and ice loss on the Greenland ice sheet. Warmer-than-normal temperatures over the Eurasian Arctic in spring resulted in a new record-low June snow cover extent and spring snow cover duration in this region. In the Canadian Arctic, the mass loss from glaciers and ice caps was the greatest since GRACE measurements began in 2002, continuing a negative trend that began in 1987. New record high temperatures occurred at 20 m below the land surface at all permafrost observatories on the North Slope of Alaska, where measurements began in the late 1970s. Arctic sea ice extent in September 2011 was the second-lowest on record, while the extent of old ice (four and five years) reached a new record minimum that was just 19% of normal. On the opposite pole, austral winter and spring temperatures were more than 3 degrees C above normal over much of the Antarctic continent. However, winter temperatures were below normal in the northern Antarctic Peninsula, which continued the downward trend there during the last 15 years. In summer, an all-time record high temperature of -12.3 degrees C was set at the South Pole station on 25 December, exceeding the previous record by more than a full degree. Antarctic sea ice extent anomalies increased steadily through much of the year, from briefly setting a record low in April, to well above average in December. The latter trend reflects the dispersive effects of low pressure on sea ice and the generally cool conditions around the Antarctic perimeter.

21. Agricultural land classification with soil conservation purposes in plots of horticultural use, Alto Motatán sub-basin, Mérida-Venezuela

Se determinó los Índices de Productividad y Riesgos de Erosión con el propósito de clasificar tierras agrícolas con fines de conservación en áreas montañosas tropicales. El área de estudio está ubicada en la Finca Gavilán 2, perteneciente al Comité de Riego Cruz Chiquita, localizado en la subcuenca Alto Motatán, parroquia La Venta, municipio Miranda, del estado Mérida. Se describieron 18 perfiles de suelo que representan 4,89 ha, obteniendo las características físicas y químicas que permitieron determinar las variables requeridas para calcular los índices antes indicados. Los resultados obtenidos muestran que los suelos tienen una productividad variable desde baja a muy alta, predominando una baja productividad (51,12%). Los factores más limitantes para la productividad de los suelos son los altos porcentajes de fragmento grueso y la poca profundidad efectiva del suelo. Todas las parcelas presentan un moderado riesgo de erosión, siendo la pendiente el principal factor que potencia la erosión del suelo. Finalmente, se obtuvo que el 63,80% de las parcelas clasificaron como tierras agrícolas en reserva (R), por presentar una productividad baja a moderada y moderados riesgos de erosión. El 36,20% restante clasifican como tierras en condición subcrítica (S), por tener productividad alta a muy alta y moderados riesgos de erosión. Se recomienda la utilización de prácticas conservacionistas para preservar la capa arable de estos suelos y mejorar los rendimientos de los cultivos. In order to classify agricultural lands with conservation purposes in mountain tropical areas, the Productivity Index and Erosion Risk Index were determined. The study area is located at the "Gavilán 2" farm, which belongs to the Cruz Chiquita Irrigation Committee, located at the sub-basin "Alto Motatán", "La Venta" parrish, Miranda municipality, Mérida State. 18 profiles of soil that represent 4.89 ha, were described, obtaining the physical and chemical characteristics that enabled to determine the required variables to calculate the indexes mentioned above. The obtained results shows that soils have a variable productivity from low to high, predominating a low productivity (51.12%). The most limiting factors for productivity in soils are the high percentages of coarse fragment and the low effective of soil depth. All the plots shows a moderate erosion risk, being the slope the main factor that increases the soil erosion. Finally, it was obtained that 63.80% of the plots classified as agricultural lands in reserve, since they present a low to moderate productivity and moderate erosion risk. The 36.20% remaining classify as lands in sub-critical condition, having a high to higher productivity and moderate erosion risk. The utilization of conservation practices to preserve the topsoil of these soils and to improve the yields of cultivations, are recommended.

Laser-Induced Breakdown Spectroscopy is a technique where a pulsed laser beam focused on a sample, produce ablation of small quantities of material and formation of plasma. Spectroscopy analysis of the radiation emitted by this plasma, allows detection of the elements in the sample and determination its concentration. LIBS is an efficient analytic technique for the traces analisys of samples in any aggregation states: solid, liquids or gaseous. It is a rapid, local and not destructive method; it allows working at large. The Atomic and Molecular Spectroscopy Laboratory (LEAM) works in this develops applications of this technique to spectroscopic analisys of materials of technological interest. Abstract

Results of Mössbauer spectroscopy for the chalcopyrite-type of alloys CuFe(S1−zSez)2 in the range of composition 0≤z≤0.45 are presented. Room temperature spectra show two contributions: one is a magnetic spectrum (six lines) with a value of the hyperfine field near to that of chalcopyrite (z=0, in which an antiferromagnetic order occurs belowT=823 K), the other having only one line. The relative area of the single-line contribution increases asz increases. Forz=0.2, we also made Mössbauer measurements as a function of temperature. The ratio of the two contributions evolves according to a Boltzmann law, in which the single line corresponds to the excited state at 81 K above the antiferromagnetic ground state.

38. Electric field gradient sign determination in low magnetic fields perpendicular to the gamma rays in57Fe Mössbauer spectroscopy

Abstract

Abstract: The determination, by Mssbauer57Fe spectroscopy, of the sign and the asymmetry parameter of the electric field gradient (EFG), in a non-magnetic powder sample, needs the use of an external magnetic field. Usually a “parallel-magnetic-field-to-the-gamma-rays” configuration is employed. In the present work, theoretically generated spectra in both configurations, parallel an perpendicular, are compared. The result is that the spectra in the perpendicular configuration show always a larger asymmetry for the same fields intensitics. In consequence, the sign determination of the EFG, is possible with magnetic fields significantly smaller in the perpendicular case than in the parallel one. Furthermore, the practical consequences are important, because EFG studies with conventional electromagnets, allowing only fields lower than 25 kGauss are feasible. As an example. Mssbauer experimental results obtained at room temperature for natural pyrite (FeS2) in perpendicular fields of 15 and 20 kGauss, created by a conventional electromagnet, are shown here.

Abstract

Onco-nephrology is an emerging field in medicine. Patients with cancer may suffer from kidney diseases because of the cancer itself and cancer-related therapy. It is critical for nephrologists to be knowledgeable of cancer biology and therapy in order to be fully integrated in the multidisciplinary team and optimally manage patients with cancer and kidney diseases. In a recent international meeting, the key issues in this challenging clinical interface were addressed, including many unresolved basic science questions, such as the high tumor incidence in kidney transplant recipients. To this end, 70 highly qualified faculty members were gathered from all over the world to discuss these issues in 8 plenary sessions, including 5 keynote lectures. In addition, 48 young nephrologists and oncologists were invited to present their original observations that were highlighted in 2 large poster sessions.

Abstract

Background: Allergic bronchopulmonary aspergillosis is a rare entity, caused by a hypersensitivity reaction of the immune system to Aspergillus fumigatus, characterized by the presence of pulmonary infiltrates, bronchiectasis and increased eosinophils in peripheral blood., Case Report: 42-year-old male with diabetes mellitus, with poor adherence to treatment, went to the emergency room due to rapidly progressive dyspnea and severe hypoxemia, pneumonia and bilateral pleural effusion were evident, laboratory studies reported, HbA1c 9.4%, eosinophils 3650 cells/mL, serum IgE for Aspergillus fumigatus 0.84 IU/mL. Treatment was started with supplemental oxygen, prednisone and itraconazole, to which the patient showed clinical and radiological improvement., Conclusions: Invasive mycoses have a mortality of 21 to 80%, frequently in immunocompromised patients, they are usually not described in patients with diabetes mellitus; The prevalence of fungal colonization among diabetic patients has been described as 37% compared to 4.7% in non-diabetic patients.

Abstract

Background: In nursing education, contact with real scenarios implies the design of favorable experiences to develop prioritization, reasoning, critical thinking, and management skills that support future practice. In the context of the teaching-learning process, simulation emerges as a support strategy, but its use and management require the knowledge and appropriation of teachers. Clinical simulation during education promotes growth in technical skills and aptitudes such as critical thinking, emotional management, organization, delegation, and teamwork. The culmination positively impacts the student, reflecting on their confidence, security, and adaptability to unexpected or unknown situations and risks., Objective: The aim of this scoping review is to determine the socioemotional skills described during the teaching-learning process mediated by medium- and high-fidelity clinical simulation in nursing students., Methods: The main concepts and limits of the research area will be determined according to the 5 phases of a scoping review proposed by Arksey and O'Malley. Research articles and postgraduate theses published between 2010 and 2023 in English and Spanish will be considered. Dissertation-type documents, book chapters, editorials, abstracts, and articles focused on clinical simulation among nursing professionals will be excluded. The articles will be retrieved from databases available at the Universidad Cooperativa de Colombia, along with CINAHL, Scielo, and PubMed. The search strategy will be based on the Population-Concept-Context framework. Article selection will be carried out by 2 independent evaluators who will review titles and abstracts in stage 1 and the full text in stage 2. A database of retrieved articles will be built with the variables of interest. A qualitative thematic analysis will be conducted by 5 independent reviewers to provide an overview of the literature, focusing on identifying similarities and contrasts between studies and contributions related to the aspects of social skills described in nursing students., Results: The investigation has not yet started. The findings aim to focus on variables within the academic environment that, when correlated with the clinical simulation experience, may determine student learning. The working hypothesis is that students who experience greater satisfaction or possess better communication skills also demonstrate superior performance during high-fidelity simulation activities. The most relevant results will be contrasted considering the stated objective and knowledge gaps. Key aspects will also be compared with other reviews addressing related topics such as communication, self-efficacy, and self-confidence. Skills described by other authors that were not considered in the initial literature review will also be mentioned., Conclusions: Educational institutions are responsible for including learning experiences in controlled environments such as medium- and high-fidelity simulation to ensure the acquisition of technical capabilities and additional socioemotional skills. Abstract

Metronidazole (MTZ) is the most common drug used against Trichomonas vaginalis ( T. vaginalis ) infections; however, treatment failures and high rates of recurrence of trichomoniasis have been reported, suggesting the presence of resistance in T. vaginalis to MTZ. Therefore, research into new therapeutic options against T. vaginalis infections has become increasingly urgent. This study investigated the trichomonacidal activity of a series of five imidazole carbamate compounds (AGR-1, AGR-2, AGR-3, AGR-4, and AGR-5) through in vitro susceptibility assays to determine the IC 50 value of each compound. All five compounds demonstrated potent trichomonacidal activity, with IC 50 values in the nanomolar range and AGR-2 being the most potent (IC 50 400 nM). To gain insight into molecular events related to AGR-induced cell death in T. vaginalis , we analyzed the expression profiles of some metabolic genes in the trophozoites exposed to AGR compounds and MTZ. It was found that both AGR and MTZ compounds reduced the expression of the glycolytic genes ( CK , PFK , TPI , and ENOL ) and genes involved in metabolism ( G6PD , TKT , TALDO , NADHOX , ACT , and TUB ), suggesting that disturbing these key metabolic genes alters the survival of the T. vaginalis parasite and that they probably share a similar mechanism of action. Additionally, the compounds showed low cytotoxicity in the Caco-2 and HT29 cell lines, and the results of the ADMET analysis indicated that these compounds have pharmacokinetic properties similar to those of MTZ. The findings offer significant insights that can serve as a basis for future in vivo studies of the compounds as a potential new treatment against T. vaginalis .

Abstract

The parasite Trichomonas vaginalis is the etiologic agent of trichomoniasis, the most common non-viral sexually transmitted disease worldwide. This infection often remains asymptomatic and is related to several health complications. The traditional treatment for trichomoniasis is the use of drugs of the 5-nitroimidazole family, such as metronidazole; however, scientific reports indicate an increasing number of drug-resistant strains. Benzimidazole derivatives could offer an alternative in the search for new anti-trichomonas drugs. In this sense, two attractive candidates are the compounds O 2 N-BZM7 and O 2 N-BZM9 (1 H -benzimidazole derivatives), since, through in vitro tests, they have shown a higher trichomonacide activity. In this study, we determined the effect on the expression level of metabolic genes in T. vaginalis . The results show that genes involved in redox balance ( NADHOX , G6PD::6PGL ) are overexpressed, as well as the gene that participates in the first reaction of glycolysis ( CK ); on the other hand, structural genes such as ACT and TUB are decreased in expression in trophozoites treated with the compound O 2 N-BZM9 , which would probably affect its morphology, motility and virulence. These results align with the trichomonacidal activity of the compounds, with benzimidazole O 2 N-BZM9 being the most potent, with an IC 50 value of 4.8 μM. These results are promising for potential future therapeutic applications.
