Bei vielen biomedizinischen Problemen in einer immer älter werdenden Gesellschaft können Biomaterialien einen wichtigen Beitrag dazu leisten, nicht mehr (voll) funktionsfähige Bestandteile des Körpers zu unterstützen oder zu ersetzen und damit die Lebensqualität des Patienten zu erhöhen. Aufgrund der Komplexität biologischer Materialien und Prozesse sowie zur Unterstützung von Heilungsprozessen ist eine stetige Weiterentwicklung synthetischer Biomaterialien erforderlich. Daher beschäftigt sich diese Arbeit mit der Entwicklung neuer nanoporöser Nanopartikel auf Basis von Organosilica und mit der Optimierung von diesen und anderen Biomaterialien in Form von Siliconkompositen mit Organosilica-Füllstoffen. Es werden sowohl reine nanoporöse Organosilica-Nanopartikel (NPOSNP) mit aromatischen Biphenylen-Brückeneinheiten als auch Kern-Schale-Nanopartikel (engl.: core-shell nanoparticles; CSNPs) mit Organosilica-Hülle synthetisiert und untersucht. Letztere bauen auf ausgiebig untersuchten nanoporösen SilicaNanopartikeln (NPSNP) im Kern auf und enthalten Phenylen-Brücken in der Schale. Die hergestellten Nanopartikel dienen als Wirkstofffreisetzungssysteme, zum einen in partikulärer Form, zum anderen innerhalb von Siliconkompositen, denen sie als Füllstoff beigemischt werden. In dieser Funktion zeigen die OrganosilicaNanopartikel einen weichmachenden Effekt bei geringen Füllstoffanteilen sowie eine bessere Verarbeitbarkeit aufgrund der durch die hydrophoben Gruppen verhältnismäßig schwachen Füllstoff-Polymer-Wechselwirkungen. Mit zunehmendem Füllstoffanteil ist eine Verstärkung des Materialverbundes festzustellen. Die Freisetzung des neuroprotektiven Wirkstoffes Rolipram ist sowohl aus den NPOSNPs als auch aus Siliconkompositen möglich, bei letzterem bis zu mindestens 100 Tagen. Ein spezielles Freisetzungssystem stellen die CSNPs dar. Durch den unterschiedlichen chemischen Charakter von Kern und Schale adsorbieren Gastmoleküle unterschiedlich stark in Abhängigkeit ihrer Polarität in verschiedenen Regionen des Partikels, was eine rasche Freisetzung aus der hydrophoben Schale und eine verzögerte Abgabe aus dem hydrophilen Kern zur Folge hat. Dies kann sowohl mittels Farbstoffen als auch mit verschiedenen Wirkstoffen belegt werden. Der letzte Teil der Arbeit beschäftigt sich mit der Stabilität der Nanopartikel unter Beladungs- und Freisetzungsbedingungen in physiologischem Medium mit Fokus auf den Physisorptionseigenschaften. Es kann eine rasche Degradation von Silica festgestellt werden, wohingegen die NPOSNPs kaum eine Veränderung innerhalb von 70 Tagen zeigen. Die einzigartigen Physisorptions- und Degradationseigenschaften der CSNPs spiegeln den Materialverbund von Silica und Organosilica wider., With regard to many biomedical problems of an aging society, biomaterials are able to make an important contribution to support or replace parts of the body that do not function anymore (completely) to improve a patients quality of life. Due to the complexity of biological materials and processes as well as to promote healing, it is necessary to enhance the properties of synthetic biomaterials. Therefore, this work deals with the development of new nanoporous nanoparticles based on organosilica and with the optimisation of these and other biomaterials as silicone composites with organosilica fillers. Pure nanoporous organosilica nanoparticles (NPOSNPs) with biphenylene bridging units as well as core-shell nanoparticles (CSNPs) with organosilica shell are synthesised and characterised. The latter are built upon intensively studied nanoporous silica nanoparticles (NPSNPs) as core with phenylene bridging units in the shell. The manufactured nanoparticles serve as drug delivery systems, as particles as well as within silicone composites, to which they are added as fillers. In this function, the organosilica nanoparticles show a plasticizing effect at low amounts of fillers as well as a better processing due to their hydrophobic groups, which weaken filler-polymerinteractions. With increasing filler content, a reinforcement of the material is observed. It is possible to release the neuroprotective agent rolipram from NPOSNPs and from silicone composites, in the latter case for up to at least 100 days. CSNPs present a special release system. Due to the different chemical character of core and shell, guest molecules adsorb in different parts of the particle depending on their polarity, which can result in a burst release from the hydrophobic shell and a delayed release from the hydrophilic core. This has been proven utilising dyes and different kinds of drugs. The last part deals with the stability of the nanoparticles under loading and release conditions in physiological media and focusses on the physisorption properties. A rapid degradation of silica is determined, whereas NPOSNPs showed hardly any alteration within 70 days. The unique physisorption- and degradation properties of CSNPs reflect the material’s composition from silica and organosilica.

In this work we present the generation of new core–shell network nanostructures of macroscopic dimensionality by a two-step process analogous to the seeded-growth method in colloidal nanoparticle modification. The nanoparticle-based core network is assembled first and in a separate second step it is coated with a continuous metal oxide shell by sol–gel methods. The interparticle contact of the nanoparticles comprising the core network is kept intact throughout the process. By analyzing the local elemental distribution, the shells can be shown to be homogeneous over the macroscopic network monolith. The shell network can be used to considerably reinforce the mechanical strength of the final core–shell network structure.
