Abstract

AbstractIn this experimental study, we delve into the performance of an active mixer that utilizes liquid metal droplets. Our mixer configuration follows an innovative Y-type design, where two liquid metal-based pumping mechanisms operate within the minichannel inlet branches. By applying alternating current voltage to both sides of the liquid metal, we induce a surface tension gradient, propelling the fluid from the reservoirs toward the minichannel. Notably, the two entering liquids are distinctly colored, allowing us to quantify mixing using a Mixing Index (MI) definition and image processing techniques. Our investigation centers on four key variables governing mixer performance: applied voltage, liquid metal droplet diameter, input angle between the branches, and minichannel width. Given the experimental nature of our research, we employ a central composite design method within the framework of design of experiments (DoE). From our obtained experimental results, we establish correlations between MI and the other variables. Our examination of the four mentioned parameters revealed that liquid metal droplet diameter exerts the most significant influence on the mixing index. Following this, in descending order of impact, we find applied voltage, input angle, and channel width. In the optimal dimensions of our mixer, we achieve a remarkable maximum MI of 0.867. 12. The Effect of Error Correction Feedback on the Collocation Competence of Iranian EFL Learners

Abstract

Collocations are one of the most important elements in language proficiency but the effect of error correction feedback of collocations has not been thoroughly examined. Some researchers report the usefulness and importance of error correction (Hyland, 1990; Bartram & Walton, 1991; Ferris, 1999; Chandler, 2003), while others showed that error correction was of little or no use (Leki, 1990; Truscott, 1996). However, teachers use error correction feedback of collocations most of the time in teaching even though it has not been proven whether there was effect on the improvement of collocation competence of EFL students or not. The researcher tried to investigate the effect of error correction feedback on various categories of lexical collocations (V+N, Adj.+N, N+N), and grammatical collocations (Adj.+Prep.,V+Prep., N) in this study based on Benson, Benson and Ilson's (1986) collocations model. The results revealed that error correction feedback was significant and had a positive effect on the collocation competence at advanced and intermediate levels, but not at the elementary level.

Abstract

Background and Objectives: Host genetic changes like single nucleotide polymorphisms (SNPs) are one of the main factors influencing susceptibility to viral infectious diseases. This study aimed to investigate the association between the host SNP of Toll-Like Receptor3 (TLR3) and Toll-Like Receptor7 (TLR7) genes involved in the immune system and susceptibility to COVID-19 in a sample of the Iranian population. Materials and Methods: This retrospective case-control study evaluated 244 hospitalized COVID-19 patients as the case group and 156 suspected COVID-19 patients with mild signs as the control group. The genomic DNA of patients was genotyped for TLR7 (rs179008 and rs179009) and TLR3 (rs3775291 and rs3775296) SNPs using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: A significant association between rs179008 SNP in the TLR7 gene and the susceptibility of COVID-19 was found between case and control groups. The AT genotype (Heterozygous) of TLR7 rs179008 A>T polymorphism showed a significant association with a 2.261-fold increased odds of COVID-19 (P=0.003; adjusted OR: 2.261; 99% CI: 1.117-4.575). In addition, a significant association between TC genotype of TLR7 rs179009 T>C polymorphism and increased odds of COVID-19 (P< 0.0001; adjusted OR: 6.818; 99% CI: 3.149-14.134) were determined. The polymorphism frequency of TLR3 rs3775291 and rs3775296 genotypes were not significantly different between the case and control groups (P> 0.004167). Conclusion: SNPs in TLR7 rs179008 and rs179009 genotypes are considered host genetic factors that could be influenced individual susceptibility to COVID-19. The SNPs in TLR3 (rs3775296 and rs3775291) showed no significant association with COVID-19 in Iranian population. [ABSTRACT FROM AUTHOR]

18. Targeted hydrolysis of native potato protein:A novel workflow for obtaining hydrolysates with improved interfacial properties

Abstract

Peptides and protein hydrolysates are promising alternatives to substitute chemical additives as functional food ingredients. In this study, we present a novel workflow for producing a potato protein hydrolysate with improved emulsifying and foaming properties using quantitative proteomics and bioinformatic prediction to facilitate targeted hydrolysis design. Based on previous studies, we selected 15 potent emulsifier peptides derived from abundant potato proteins as targets. Through in silico analysis, we determined that from a range of industrial proteases (Neutrase (Neut), Alcalase (Alc), Flavourzyme (Flav) and Trypsin (Tryp)), Tryp was found more likely to release peptides resembling the targets. After applying all proteases individually, hydrolysates were assayed for in vitro emulsifying and foaming properties. No direct correlation between degree of hydrolysis and interfacial properties was found. Tryp (E/S = 3%) produced a hydrolysate (DH = 5.4%) with high aqueous solubility and the highest (P < 0.05) emulsifying and foaming abilities, validating the hypothesis. Using LC-MS/MS, we identified >10,000 peptides in each hydrolysate. Peptide mapping revealed that random overlapping with known peptide emulsifiers is not sufficient to quantitatively describe hydrolysate functionality. However, validated release of targeted peptides by 3% Tryp appears to increase surface activity of the hydrolysate. Our data also suggest that terminal hydrophobic anchor domains may be important for high interfacial partitioning and activity. While modest yields and residual unhydrolyzed protein indicate room for process improvement, this work shows that bioinformatics-guided and data-driven targeted hydrolysis is a promising, interdisciplinary approach to facilitate process design for production of functional hydrolysates from alternative protein sources.

Abstract

Peptide emulsifiers derived from plant proteins are gathering growing interest as green and sustainable replacements for chemical additives in food. Potato (Solanum tuberosum) is one of the most important crops for both human consumption and industrial processing. Globally, the annual production of potato starch exceeds 3,000,000 MT with more than 200,000 MT of potato protein isolated as a side-stream, providing an enormous source of raw protein. The direct isolation of food-grade protein is in many cases regarded as cost-ineffective. Nevertheless, potato proteins may be a valuable source of functional peptides.Previously, we demonstrated amphiphilicity-based bioinformatic prediction of peptide emulsifiers embedded in potato proteins1,2. Amongst the predicted peptides, especially one (γ1), derived from the storage protein patatin, showed exceptional emulsifying activity in vitro. Although patatin is the most abundant protein in potatoes, it is not a single protein, but a family of highly homologous isoforms. Using bottom-up proteomics in combination with multiple sequence alignment and in silico digestion, we identified several γ1 variants. The variants consist of both full-length isoforms with single amino acid substitutions and tryptic variants/truncations from different patatin isoforms. The emulsifying activity of the γ1 variants, physical stability of the emulsions during storage, and interfacial properties were investigated3. Furthermore, the interfacial conformation of the peptides was investigated by SRCD and supplemented by NMR and benchtop CD for selected peptides in micellar model systems. Based on these results, we are able to i) evaluate the full potential of using γ1 variants as peptide emulsifiers in food; ii) provide novel insight on the structure/function relationship of amphipathic, α-helical peptide emulsifiers; iii) combine in vitro functional validation with in silico proteolysis to design a scalable and targeted enzymatic hydrolysis, resulting in a hydrolysate with improved emulsifying properties; and iv) apply the hydrolysates as stabilizers for encapsulation of fish oil for foods.

27. Texture and viscoelastic characteristics of silver carp (Hypophthalmichthys molitrix) surimi affected by combination of washing regimes and hydrogen peroxide

Abstract

This study aimed to apply H2O2 at different concentrations in combination with mince:water (M:W) ratios and different washing cycles (WC) in order to produce surimi gel from silver carp without compromising its quality characteristics. Color, texture, microstructure and rheological properties of surimi gels were investigated. Water holding capacity, texture profile and gel strength, showed a greater dependency on number of washing cycles than the M:W ratios and percentage of H2O2 (p<0.05), i.e., higher washing cycles, firmer surimi gel. Accordingly, T2 (one WC, 2% H2O2, 1:3), T10 (two WC, 1% H2O2, 1:2) and T16 (three WC, 1% H2O2, 1:2) treatments resulted the most cohesive and resilient surimi compared to the rest (p<0.05), confirmed by SEM images. However, all treated fish mince samples with H2O2, resulted in a surimi gel with lower texture quality compared to the control surimi prepared by conventional washing process without H2O2 (p<0.05). A temperature sweep test was conducted based on the LVR stress and frequency values and the aforementioned surimi gels exhibited an obvious valley shape pattern at temperature range of 48-62°C. In the creep-recovery test, the Burgers model satisfactorily described the internal structure of the surimi gel samples as the lowest deformation belonged to the control samples followed by T2. However, after 300s strain, neither of surimi gels were fully recovered their original shape. Altogether, further studies are needed to clarify the effects of H2O2 in reduction of washing cycles, without significantly affecting the textural and rheological properties of resultant surimi gel.

Abstract

The pandemic of severe acute respiratory syndrome coronavirus 2 raised the attention towards bacterial coinfection and its role in coronavirus disease 2019 (COVID-19) disease. This study aims to systematically review and identify the pooled prevalence of bacterial coinfection in the related articles. A comprehensive search was conducted in international databases, including MEDLINE, Scopus, Web of Science, and Embase, to identify the articles on the prevalence of bacterial coinfections in COIVD-19 patients from 1 December 2019 until 30 December 2020. All observational epidemiological studies that evaluated the prevalence of bacterial coinfections in patients with COVID-19 were included without any restriction. Forty-two studies including a total sample size of 54,695 were included in the analysis. The pooled estimate for the prevalence of bacterial coinfections was 20.97% (95% CI: 15.95-26.46), and the pooled prevalence of bacterial coinfections was 5.20% (95% CI: 2.39-8.91) for respiratory subtype and 4.79% (95% CI: 0.11-14.61) for the gastrointestinal subtype. The pooled prevalence for Eastern Mediterranean Regional Office and South-East Asia Regional Office was 100% (95% CI: 82.35-100.00) and 2.61% (95% CI: 1.74-3.62). This rate of coinfection poses a great danger towards patients, especially those in critical condition. Although there are multiple complications and adverse effects related to extensive use of antibiotics to treat patients with COVID-19, it seems there is no other option except applying them, and it needs to be done carefully.

Abstract

Background and Objectives: Diabetes mellitus is the most common endocrine disorder worldwide. Cardiometabolic risk factors like truncal obesity and unhealthy life style including unhealthy diet contribute to development of type 2 diabetes (T2D) and its complications and attributable deaths. Therefore, modification of the patient's diet and lifestyle is a core of T2D treatment. ?-Oryzanol (ORZ) is a phytochemical found in rice bran oil. An increasing body of evidence has demonstrated health benefits of ORZ including decreasing oxidative stress and insulin resistance. This study will be conducted to examine possible effects of ORZ-fortified canola oil, as compared with plain canola and sunflower oils, on certain cardiometabolic indicators. Materials and Methods: A total of 90 adult subjects aged 20-65 y with confirmed diagnosis of T2D will randomly be allocated in one to the three groups to receive: (i) ORZ-fortified canola oil (14 mg ORZ/14g); (ii) plain canola oil (without ORZ); and (iii) sunflower oil. All oils will have equal amounts of vitamins D (4.5 µg =180 IU) and A (240 µg =800 IU) per serving, i.e. 14 g. The subjects will be instructed to use only the given oils for cooking during 12 weeks intervention period. Dietary, anthropometric and laboratory evaluations will be done for all participants before and after the intervention. Conclusions: The findings of this study will shed a light to our current knowledge of health aspects of cooking oils and also will input information to our future dietary recommendations to T2D patients and, probably, the general population. Abstract

Protein hydrolysates show great promise as bioactive food and feed ingredients and for valorization of side-streams from e.g., the fish processing industry. We present a novel approach for hydrolysate characterization that utilizes proteomics data for calculation of weighted mean peptide properties (length, molecular weight, and charge) and peptide-level abundance estimation. Using a novel bioinformatic approach for subsequent prediction of biofunctional properties of identified peptides, we are able to provide an unprecedented, in-depth characterization. The study further characterizes bulk emulsifying, foaming, and in vitro antioxidative properties of enzymatic hydrolysates derived from cod frame by application of Alcalase and Neutrase, individually and sequentially, as well as the influence of heat pre-treatment. All hydrolysates displayed comparable or higher emulsifying activity and stability than sodium caseinate. Heat-treatment significantly increased stability but showed a negative effect on the activity and degree of hydrolysis. Lower degrees of hydrolysis resulted in significantly higher chelating activity, while the opposite was observed for radical scavenging activity. Combining peptide abundance with bioinformatic prediction, we identified several peptides that are likely linked to the observed differences in bulk emulsifying properties. The study highlights the prospects of applying proteomics and bioinformatics for hydrolysate characterization and in food protein science.
