Abstract

This thesis aimed to investigate vibrational spectroscopies for the identification of biochemical markers of leukaemias and lymphomas. In a preliminary study using the blood proteins albumin, fibrinogen and globulin, Drop Coating Deposition Raman Spectroscopy was explored and extended for use with Fourier Transform infrared spectroscopy for leukaemia blood sample analysis. Due to low sample volumes and minimal preparation required it was identified as a potential alternative to blood centrifugation to obtain the buffy coat for analysis. These studies identified that it was capable of detecting low levels of protein from small, highly concentrated droplets. Thus this method, alongside cytospin centrifugation, was used for the spectroscopic analysis of different blood fractions. Due to the low number of lymphoma samples obtained, only a feasibility study is outlined in this thesis. Samples were collected from leukaemia patients and healthy volunteers. Infrared and Raman spectra were measured of whole blood and buffy coat samples cytospun onto slides and whole blood and plasma pipetted by drop coating deposition. Multivariate statistical analysis was employed to extract key spectral differences between the pathologies and develop classification models for diagnosing chronic lymphoblastic leukaemia from previously treated and untreated patient groups. Principal component analysis followed by linear discriminant analysis was employed to identify the largest variances in the data and leave one sample out cross validation evaluated the performance of the spectral models measured on different blood components in diagnosing leukaemia. The buffy coat infrared model correctly classified 59% of the spectra, and blood droplet Raman 62%. The treated and untreated groups were then combined, which improved classification to 83% for buffy coat infrared and 71% for blood droplet Raman. These findings highlight the potential of drop coating deposition spectroscopy of whole blood for leukaemia diagnosis, although further work is required to achieve a clinically validated method.

Abstract

Introduction: Within Australia, the introduction of the National Disability Insurance Scheme has led to a growth in paediatric occupational therapists working in community settings. This growth has increased the demand for support from more senior paediatric occupational therapists to novice clinicians. Mentoring has long been valued by occupational therapists as a means to provide this support. Despite its apparent benefit, there is limited research on the contribution of mentoring as distinct from supervision, and its impact on mentees' skills and confidence in providing care. This study examined the contribution of mentorship to the development of professional capability in paediatric occupational therapy practice from the perspective of mentors and mentees. Methods: Interpretive description methodology was used. In‐depth interviews were conducted with nine mentors and eight mentees from three Australian states. Data were analysed inductively and thematically. Findings: All participants reflected on the challenges presented by the complexity of practice, requiring a knowledge base that mentees perceived they did not possess. The essential nature of non‐judgemental, emotional support allowed mentees to feel safe to discuss their concerns when they were often overwhelmed by practice. Mentorship was viewed as capacity building, building competence by scaffolding clinical reasoning and supporting theory to practice translation while developing resilience to cope with complexity. Conclusion: Study participants reported that successful mentorship assisted novice practitioners to integrate knowledge and skills required for complex clinical and professional reasoning. The emotional support provided through the relationship supported novice therapists to build their confidence and resilience while promoting professional identity and socialisation into the profession. The study raised questions related to how the profession best supports novice paediatric therapists in the current employment contexts, and the need to review how bodies of knowledge relevant to paediatrics are brought together to be used by both novice clinicians and senior therapists, who support them, for translation to effective practice. Abstract

We collected price and promotional data from seven supermarkets operating in San Luis Obispo, CA, for one year, from 2017 to 2018. Using the data, we created a series of variables to measure prices and promotional activity. These variables were subjected to an exploratory regression analysis. The research uncovered a number of findings that help explain price variation in supermarkets and motivate future research. Average prices, price variation, promotional frequency, and promotional depth are all interrelated in important, and in some cases unexplored, ways.

46. Does brand familiarity have any impact on repairing trust between a company and customer in the time of crisis?

Abstract

Extant research has shown trust to be damaged between a company and customer if a crisis or violation occurs. Prior research has focused on examining what the best course of action would be for a company to take in a crisis, and how best to repair the trust afterwards. However, there has been scant investigation into whether repairing that trust is directly linked to how familiar a customer is with the brand. This paper presents a review of the existing literature on trust, trust repair and brand familiarity, and conducts two research studies to examine the implications of repairing trust after a crisis or violation for a familiar brand and an unfamiliar brand. 256 questionnaires were collected and analysed using SPSS, with questions centred on how a crisis affects a customer’s perception of trust, loyalty and forgiveness based on their familiarity with a brand under three conditions – denial, admittance and prior knowledge of the crisis. results show that brand familiarity does impact upon repairing trust between a company and customer, and their ability to forgive and remain loyal to the familiar brand, as respondents are shown to be less forgiving and are thus unlikely to trust them post crisis. By admitting and apologising for the crisis, a familiar brand is more likely to be able to regain trust. Respondents place great emphasis on the role of the media, with most respondents likely to adopt the media portrayal of the crisis. The paper concludes with future research directions suggested.
