Abstract

We describe changes in pulmonary function measures across time in Duchenne muscular dystrophy patients treated with glucocorticoids (GCs) 1 year compared to GC naïve patients in the Cooperative International Research Group Duchenne Natural History Study, a multicenter prospective cohort study. 397 participants underwent 2799 pulmonary function assessments over a period up to 10 years. Fifty-three GC naïve participants ( 1 month exposure) were compared to 322 subjects with 1 year cumulative GC treatment. Forced vital capacity (FVC), peak expiratory flow rate (PEFr), maximal inspiratory and expiratory pressures were performed and calculated as a percent predicted (%p). GC treatment slowed the rate of pulmonary decline as measured by FVC%p, in patients aged 7-9.9 years. GC treatment slowed 12 and 24-month progression of percent predicted spirometry to a greater degree in those with baseline FVC%p from 80-34%. GC treatment resulted in higher peak absolute FVC and PEFr values with later onset of decline. Progression to an absolute FVC 1 liter was delayed by GC treatment. Patients who reached a FVC below 1 L were 4.1 times more likely to die (p = 0.017). Long-term glucocorticoid treatment slows pulmonary disease progression in Duchenne dystrophy throughout the lifespan.

Abstract

Although Carpinus caroliniana (American hornbeam), Fagus grandifolia (American beech), and Gymnocladus dioicus (Kentucky coffeetree) have considerable landscape merit, they are not readily available in nurseries. Little research has been conducted on the cultural preferences of these species during nursery production. This study tested the effects of light quality and extended photoperiod on these species. Container-grown seedlings were subjected to three light treatments: 18-hour photoperiod using high pressure sodium lights (HPS), 18-hour photoperiod using incandescent lights (INC), and natural light and daylength (NAT). After 14 weeks, stem height of F. grandifolia seedlings was greatest for HPS light and least for NAT light. After 8 and 12 weeks, light source had no impact on stem heights of C. caroliniana. In 1997, stem height and caliper of G. dioicus were greatest using HPS light. In a repeat of the G. dioicus, experiment in 1998, HPS and INC light treatments resulted in greater stem height, caliper, and shoot and root dry weight compared to NAT treatment. The increase in height of seedlings under the HPS and INC treatments compared to the NAT treatments resulted from a greater number of seedlings that produced a second flush of growth under the HPS and INC treatments.

Abstract

The role of exchange and structural heterozygosity in radiation-induced non-disjunction and loss in the oocyte of Drosophila melanogaster has been investigated by irradiating 2 types of females, one heterozygous and the other homozygouus for a multiply inverted X-chromosome. The frequencies of exchange in tetrads in the oocytes of the two genotypes were estimated as ∽ 24% for the heterozygote and ∽ 90% for the homozygote. An examination of the first 7 daily broods from both kinds of females irradiated with 4000 R of X-rays revealed no significant difference in X-chromosome loss or non-disjunction between the 2 genotypes. The average non-disjunction frequencies for the entire period were 1.57 ± 0.19% for the homozygotes and 1.74 ± 0.24% for the heterozygotes. The corresponding frequencies of loss were 5.21 ± 0.37% and 7.19 ± 0.67%. The values obtained for stage 8–14 oocytes from homozygous and heterozygous females irradiated with 500 R were 0.22 ± 0.21% and 0.33% ± 0.18%, respectively, for non-disjunction, and 2.46 ± 0.33% and 2.57 ± 0.33%, respectively, for loss. The results indicate that neither structural heterozygosity of homologues nor exchange between homologues modifies the frequencies with which they non-disjoin or are lost following irradiation of the oocyte.
