Observations of glacier mass changes are key to understanding the response of glaciers to climate change and related impacts, such as regional runoff, ecosystem changes, and global sea level rise. Spaceborne optical and radar sensors make it possible to quantify glacier elevation changes, and thus multi-annual mass changes, on a regional and global scale. However, estimates from a growing number of studies show a wide range of results with differences often beyond uncertainty bounds. Here, we present the outcome of a community-based inter-comparison experiment using spaceborne optical stereo (ASTER) and synthetic aperture radar interferometry (TanDEM-X) data to estimate elevation changes for defined glaciers and target periods that pose different assessment challenges. Using provided or self-processed digital elevation models (DEMs) for five test sites, 12 research groups provided a total of 97 spaceborne elevation-change datasets using various processing approaches. Validation with airborne data showed that using an ensemble estimate is promising to reduce random errors from different instruments and processing methods but still requires a more comprehensive investigation and correction of systematic errors. We found that scene selection, DEM processing, and co-registration have the biggest impact on the results. Other processing steps, such as treating spatial data voids, differences in survey periods, or radar penetration, can still be important for individual cases. Future research should focus on testing different implementations of individual processing steps (e.g. co-registration) and addressing issues related to temporal corrections, radar penetration, glacier area changes, and density conversion. Finally, there is a clear need for our community to develop best practices, use open, reproducible software, and assess overall uncertainty to enhance inter-comparison and empower physical process insights across glacier elevation-change studies.

Climate models predict amplified warming at high elevations in low latitudes, making tropical glacierized regions some of the most vulnerable hydrological systems in the world. Observations reveal decreasing streamflow due to retreating glaciers in the Andes, which hold 99 % of all tropical glaciers. However, the timescales over which meltwater contributes to streamflow and the pathways it takes – surface and subsurface – remain uncertain, hindering our ability to predict how shrinking glaciers will impact water resources. Two major contributors to this uncertainty are the sparsity of hydrologic measurements in tropical glacierized watersheds and the complication of hydrograph separation where there is year-round glacier melt. We address these challenges using a multi-method approach that employs repeat hydrochemical mixing model analysis, hydroclimatic time series analysis, and integrated watershed modeling. Each of these approaches interrogates distinct timescale relationships among meltwater, groundwater, and stream discharge. Our results challenge the commonly held conceptual model that glaciers buffer discharge variability. Instead, in a subhumid watershed on Volcán Chimborazo, Ecuador, glacier melt drives nearly all the variability in discharge (Pearson correlation coefficient of 0.89 in simulations), with glaciers contributing a broad range of 20 %–60 % or wider of discharge, mostly (86 %) through surface runoff on hourly timescales, but also through infiltration that increases annual groundwater contributions by nearly 20 %. We further found that rainfall may enhance glacier melt contributions to discharge at timescales that complement glacier melt production, possibly explaining why minimum discharge occurred at the study site during warm but dry El Niño conditions, which typically heighten melt in the Andes. Our findings caution against extrapolations from isolated measurements: stream discharge and glacier melt contributions in tropical glacierized systems can change substantially at hourly to interannual timescales, due to climatic variability and surface to subsurface flow processes.

This mixed-methods case study examines data for over 1,500 student awards at two dual language elementary schools collected over the course of two years. Using descriptive statistics, this paper examines student representation by demographics in quarterly scholastic awards. Through document analysis of award descriptions, we analyze how students were selected by their teachers. Using Critical Race Theory (CRT) as a theoretical framework, findings show that across both schools, Latinx students were underrepresented in scholastic awards while White students were overrepresented. After examining and discussing student award data, this paper contextualizes the data within the literature on unconscious bias, inequity in DLBE programs, data-based SWPBIS systems, and explores implications for further research and school-based leadership.

In the frame of the European Project on Ocean Acidification (EPOCA), the response of an Arctic pelagic community (pCO2 was investigated. For this purpose 9 large-scale in situ mesocosms were deployed in Kongsfjorden, Svalbard (78°56.2´ N, 11°53.6´ E), in 2010. The present study investigates effects on the communities of particle-attached (PA; >3 μm) and free-living (FL; < 3 μm > 0.2 μm) bacteria by Automated Ribosomal Intergenic Spacer Analysis (ARISA) in 6 of the mesocosms, ranging from 185 to 1050 μatm initial pCO2, and the surrounding fjord. ARISA was able to resolve, on average, 27 bacterial band classes per sample and allowed for a detailed investigation of the explicit richness and diversity. Both, the PA and the FL bacterioplankton community exhibited a strong temporal development, which was driven mainly by temperature and phytoplankton development. In response to the breakdown of a picophytoplankton bloom, numbers of ARISA band classes in the PA community were reduced at low and medium CO2 (~ 185–685 μatm) by about 25%, while they were more or less stable at high CO2 (~ 820–1050 μatm). We hypothesise that enhanced viral lysis and enhanced availability of organic substrates at high CO2 resulted in a more diverse PA bacterial community in the post-bloom phase. Despite lower cell numbers and extracellular enzyme activities in the post-bloom phase, bacterial protein production was enhanced in high CO2 mesocosms, suggesting a positive effect of community richness on this function and on carbon cycling by bacteria.

Tigecycline is a broad spectrum antibiotic having activity against multiresistant isolates. In vitro susceptibility testing is difficult to perform with the use of traditional microbiological techniques. The aim of this study was to evaluate the disk diffusion test with three different Mueller-Hinton agar brands, and the Vitek 2 automated system in comparison with the standard broth microdilution method against 200 gram-negative isolates (Escherichia coli, Klebsiella pneumoniae, Enterobacter cloacae, Serratia marcescens and Acinetobacter baumannii). Among Enterobacteriaceae, the Becton Dickinson agar had the lowest rate of minor (32.5%) and major errors (3.8%). No very major errors were found. For A. baumanni, the rate of minor and major errors was lower. A high rate of agreement (94%) was found between the broth microdilution method and the Vitek 2 system. Our results show that there are important differences between agars used for the disk diffusion test, and that Vitek 2 is a valid tool for susceptibility testing in clinical laboratories.La tigeciclina es un antibiótico de amplio espectro con actividad frente a bacterias multirresistentes. Existen dificultades en la determinación de la actividad in vitro a través de las técnicas microbiológicas convencionales. El objetivo del estudio fue evaluar tres marcas diferentes de medio agar Mueller-Hinton para utilizar en el método de difusión con disco y el método automatizado Vitek 2, y compararlos con la prueba tradicional de microdilución manual (Paneles Trek) frente a 200 aislamientos de microorganismos gram negativos (Escherichia coli, Klebsiella pneumoniae, Enterobacter cloacae, Serratia marcescens y Acinetobacter baumannii). Para el grupo de las enterobacterias, el medio con mejor desempeño fue el producido por Becton Dickinson, que tuvo 32,5% de errores menores y 3,8% de errores mayores. No se presentaron errores mayores con ningún medio. Se encontró una alta concordancia (94%) entre el método de microdilución manual y el Vitek 2. Para A. baumannii, el medio con mejor desempeño fue el Mueller-Hinton elaborado por Becton Dickinson, con 12,5% de errores menores y 2,5% de errores mayores. Los resultados sugieren que el método Vitek 2 es una herramienta válida en la determinación de la sensibilidad a la tigeciclina y que existen diferencias muy grandes en la prueba de difusión con disco según la marca comercial de medio utilizado.

Isolated from the marine bacteria Serinicoccus sp., seriniquinone (SQ1) has been characterized by its selective activity in melanoma cell lines marked by its modulation of human dermcidin and induction of autophagy and apoptosis. While an active lead, the lack of solubility of SQ1 in both organic and aqueous media has complicated its preclinical evaluation. In response, our team turned its effort to explore analogues with the goal of returning synthetically accessible materials with comparable selectivity and activity. The analogue SQ2 showed improved solubility and reached a 30-40-fold greater selectivity for melanoma cells. Here, we report a detailed comparison of the activity of SQ1 and SQ2 in SK-MEL-28 and SK-MEL-147 cell lines, carrying the top melanoma-associated mutations, BRAFV600E and NRASQ61R, respectively. These studies provide a definitive report on the activity, viability, clonogenicity, dermcidin expression, autophagy, and apoptosis induction following exposure to SQ1 or SQ2. Overall, these studies showed that SQ1 and SQ2 demonstrated comparable activity and modulation of dermcidin expression. These studies are further supported through the evaluation of a panel of basal expression of key-genes related to autophagy and apoptosis, providing further insight into the role of these mutations. To explore this rather as a survival or death mechanism, autophagy inhibition sensibilized BRAF mutants to SQ1 and SQ2, whereas the opposite happened to NRAS mutants. These data suggest that the seriniquinones remain active, independently of the melanoma mutation, and suggest the future combination of their application with inhibitors of autophagy to treat BRAF-mutated tumors.

Chinese hamster ovary (CHO) cells are widely used for producing biopharmaceuticals, and engineering gene expression in CHO is key to improving drug quality and affordability. However, engineering gene expression or activating silent genes requires accurate annotation of the underlying regulatory elements and transcription start sites (TSSs). Unfortunately, most TSSs in the published Chinese hamster genome sequence were computationally predicted and are frequently inaccurate. Here, we use nascent transcription start site sequencing methods to revise TSS annotations for 15 308 Chinese hamster genes and 3034 non-coding RNAs based on experimental data from CHO-K1 cells and 10 hamster tissues. We further capture tens of thousands of putative transcribed enhancer regions with this method. Our revised TSSs improves upon the RefSeq annotation by revealing core sequence features of gene regulation such as the TATA box and the Initiator and, as exemplified by targeting the glycosyltransferase gene Mgat3, facilitate activating silent genes by CRISPRa. Together, we envision our revised annotation and data will provide a rich resource for the CHO community, improve genome engineering efforts and aid comparative and evolutionary studies.

Clusters of similar or dissimilar objects are encountered in many fields. Frequently used approaches treat each cluster's central object as latent. Yet, often objects of one or more types cluster around objects of another type. Such arrangements are common in biomedical images of cells, in which nearby cell types likely interact. Quantifying spatial relationships may elucidate biological mechanisms. Parent‐offspring statistical frameworks can be usefully applied even when central objects ("parents") differ from peripheral ones ("offspring"). We propose the novel multivariate cluster point process (MCPP) to quantify multi‐object (e.g., multi‐cellular) arrangements. Unlike commonly used approaches, the MCPP exploits locations of the central parent object in clusters. It accounts for possibly multilayered, multivariate clustering. The model formulation requires specification of which object types function as cluster centers and which reside peripherally. If such information is unknown, the relative roles of object types may be explored by comparing fit of different models via the deviance information criterion (DIC). In simulated data, we compared a series of models' DIC; the MCPP correctly identified simulated relationships. It also produced more accurate and precise parameter estimates than the classical univariate Neyman–Scott process model. We also used the MCPP to quantify proposed configurations and explore new ones in human dental plaque biofilm image data. MCPP models quantified simultaneous clustering of Streptococcus and Porphyromonas around Corynebacterium and of Pasteurellaceae around Streptococcus and successfully captured hypothesized structures for all taxa. Further exploration suggested the presence of clustering between Fusobacterium and Leptotrichia, a previously unreported relationship. [ABSTRACT FROM AUTHOR]
