Cancer resistance to chemotherapy and radiation therapies presents significant challenges, necessitating the exploration of alternative approaches. Targeting specific proteins at the molecular level, particularly their active sites, holds promise in addressing this issue. We investigated the potential of 4′-methoxy-2-nitrochalcone (MNC) as an MCL-1 inhibitor, examining its chemical and structural characteristics to elucidate its biological activity and guide the selection of potential candidates. We conducted a docking study, followed by synthesis, structural characterization, theoretical calculations, and in vitro experiments to comprehensively evaluate MNC. The docking results revealed MNC’s excellent binding within the active site of MCL-1. At 50 µM, MNC demonstrated 99% inhibition of HCT116 cell proliferation, with an IC50 value of 15.18 µM after 24 h. Treatment with MNC at 30.36 and 15.18 µM resulted in reduced cell density. Notably, MNC exhibited marked cytotoxicity at concentrations of 15.58 µM and 7.79 µM, inducing high frequencies of plasma membrane rupture and apoptosis, respectively. Our findings highlight the significant biological potential of MNC as an MCL-1 inhibitor. Furthermore, we propose exploring chalcones with hydrogen bond acceptor substituents as promising candidates for studying inhibitors targeting this protein. In conclusion, our study addresses the challenge of cancer resistance by investigating MNC as an MCL-1 inhibitor. Through detailed characterization and experimental validation, we establish the efficacof MNC in inhibiting cell proliferation and inducing cytotoxic effects. These results underscore the potential of MNC as a valuable therapeutic agent and suggest the use of chalcones with hydrogen bond acceptor substituents as a basis for developing novel MCL-1 inhibitors.

O isolamento social ocasionado pela pandemia do COVID-19 levou a uma corrida dos sistemas de ensino na busca de estratégias para garantia da continuidade do ensino mesmo que distante do espaço escolar. Devido a isso, este trabalho pautou-se em um relato de experiência que envolveu o uso da Tecnologia Digital de Informação e Comunicação (TDIC) na disciplina de Ciências com os alunos da escola Municipal Apulcro Mota, na tentativa de contornar as diferentes limitações já presentes no setor educacional público, potencializadas pela pandemia. Como TDIC, foi utilizado principlamente o software Stellarium Astronomy devido a sua gratuidade, facíl acesso e apresentar situações reais. De modo complementar os alunos foram incentivados a construção de um simulador de fases da Lua, visando trabalhar os contéudos da disciplina de Ciências de forma atrativa para reduzir a evasão e falta de motivação dos alunos e atingir os objetivos propostos da disciplina. Notou-se que quando a TDIC’s utilizada pelo docente consegue motivar os alunos, estes ficam atentos as aulas e se interessam mais quando o conteúdo é trabalhado de tal forma que possibilite a contextualização com as suas vivências para além do espaço escolar.

An 18-month-old Springer Spaniel was presented for investigation of ascites, exercise intolerance and suspected cor triatriatum dexter. Division of the right atrium into two separate chambers by an anomalous perforated membrane consistent with cor triatriatum dexter, was confirmed during echocardiography. However, a routine agitated saline contrast (bubble) study yielded results that could not be explained by the congenital heart defect alone. Computed tomography angiography was performed and revealed a dilated ventral internal vertebral venous plexus and a short, dilated vessel in the mid-lumbar region redirecting blood flow away from the caudal vena cava and into the azygos vein. Balloon dilatation of the orifice within the cor triatriatum dexter membrane successfully reduced pressure within the caudal chamber of the right atrium, and enhanced return via the caudal vena cava with subsequent resolution of clinical signs. This case report highlights the use of cross-sectional imaging in dogs with cor triatriatum dexter and unexpected contrast study results, as a means by which concurrent vascular anomalies may be identified.

We present a minimal extension of the left-right symmetric model based on the gauge group $SU(3)_{c} \times SU(2)_{L} \times SU(2)_{R} \times U(1)_{B-L} \times U(1)_{X}$, in which a vector-like fermion pair ($\zeta_L$ and $\zeta_R$) charged under the $U(1)_{B-L} \times U(1)_X$ symmetry is introduced. Associated with the symmetry breaking of the gauge group $SU(2)_{R} \times U(1)_{B-L} \times U(1)_{X}$ down to the Standard Model (SM) hypercharge $U(1)_Y$, Majorana masses for $\zeta_{L, R}$ are generated and the lightest mass eigenstate plays a role of the dark matter (DM) in our universe by its communication with the SM particles through a new neutral gauge boson "$X$". We consider various phenomenological constraints of this DM scenario, such as the observed DM relic density, the LHC Run-2 constraints from the search for a narrow resonance, and the perturbativity of the gauge couplings below the Planck scale. Combining all constraints, we identify the allowed parameter region which turns out to be very narrow. In this contribution, we re-assess some aspects of axionic electrodynamics by coupling non-linear electromagnetic effects to axion physics. We present a number of motivations to justify the coupling of the axion to the photon in terms of a general non-linear extension of the electromagnetic sector. Our emphasis in the paper relies on the investigation of the constitutive permittivity and permeability tensors, for which the axion contributes by introducing dependence on the frequency and wave vector of the propagating radiation. Also, we point out how the axion mass and the axion-photon-photon coupling constant contribute to a dispersive behavior of the electromagnetic waves, in contrast to what happens in the case of non-linear extensions, when effective refractive indices appear which depend only on the direction of the propagation with respect to the external fields. The axion changes this picture by yielding refractive indices with dependence on the wavelength. We apply our results to the special case of the Born-Infeld Electrodynamics and we show that it becomes birefringent whenever the axion is coupled. The paper is supplemented by an Appendix, where we approach the recent discussion on the controversy in defining the Poynting vector of axionic electrodynamics. This case report details a rare case of squamous cell carcinoma present in the renal pelvis of a fused pelvic kidney, a congenital complete renal fusion anomaly. A 52-year-old man with no previous urological history presented with intermittent haematuria and weight loss. He was found to have a large complex mass (14.3 cm×4.5 cm×15.5 cm) in the right moiety of a pelvic fused kidney on CT and subsequently underwent an open hemi-nephrectomy. He recovered well following surgery, however, unfortunately developed rapid recurrence, and died from disease progression after 6 months. This report demonstrates the use of 3D anatomical reconstruction to help pre-operative planning of complex surgical cases. We also include histological analysis of the tumour in detail. This case report emphasises that squamous cell carcinoma can occur due to precipitating factors such as renal calculi or infection, which are more prevalent in fusion anomalies of the kidney. Although a rare malignancy, this tumour type is associated with an aggressive behaviour and poor prognosis.

Antimalarial drugs with novel modes of action and wide therapeutic potential are needed to pave the way for malaria eradication. Violacein is a natural compound known for its biological activity against cancer cells and several pathogens, including the malaria parasite, Plasmodium falciparum (Pf). Herein, using chemical genomic profiling (CGP), we found that violacein affects protein homeostasis. Mechanistically, violacein binds Pf chaperones, PfHsp90 and PfHsp70-1, compromising the latter's ATPase and chaperone activities. Additionally, violacein-treated parasites exhibited increased protein unfolding and proteasomal degradation. The uncoupling of the parasite stress response reflects the multistage growth inhibitory effect promoted by violacein. Despite evidence of proteotoxic stress, violacein did not inhibit global protein synthesis via UPR activation - a process that is highly dependent on chaperones, in agreement with the notion of a violacein-induced proteostasis collapse. Our data highlight the importance of a functioning chaperone-proteasome system for parasite development and differentiation. Thus, a violacein-like small molecule might provide a good scaffold for development of a novel probe for examining the molecular chaperone network and/or antiplasmodial drug design. publishersversion published

Entomopathogenic nematodes (EPNs) from Heterorhabditidae and Steinernematidae families are extensively used to control insect pests. In Brazil, however, relatively few studies have identified and characterized these entomopathogens. The objective of this study was to identify and characterize an EPN isolate obtained from soil samples collected in the state of Paraná, Brazil. An isolate (UEL 08) of Heterorhabditis was detected in a soil sample collected from a pasture area cultivated with Brachiaria grass in Londrina, state of Paraná, Brazil (23°34ʹ311ʹʹS, 050°58ʹ298ʹʹW), using the insect-baiting technique with Galleria mellonella larvae as hosts. The nematode was identified through morphometric studies and molecular analyses based on amplification of the rDNA ITS region. Although we identified certain morphometric differences compared with the original description, the molecular data indicated that the ITS sequence obtained for the UEL 08 isolate is identical to the reference sequence of H. amazonensis (DQ665222) and presented 100% similarity. Thus, the findings of our morphological and molecular studies confirmed that the isolated nematode is H. amazonensis, which is the first time this species has been registered in Paraná. Study of the biological characteristics of H. amazonensis (UEL 08) revealed that the isolate has two distinct life cycles – one short (216 h) and the other long (288 h) – and produces two generations in both cycles. We observed that H. amazonensis (UEL 8) was pathogenic and virulent to the three evaluated hosts, although with different virulence against these hosts. The larvae of G. mellonella and Alphitobius diaperinus were more susceptible than adult Dichelops (Diacereus) melacanthus, with 100%, 85%, and 46% mortality, respectively. Furthermore, an in vivo production assay revealed a mean daily yield of 3.4 × 103 infective juveniles/g host larvae. Resumo Nematoides entomopatogênicos (NEP) das famílias Heterorhabditidae e Steinernematidae são amplamente utilizados no controle de insetos-pragas. No Brasil, os estudos relacionados a caracterização e identificação destes entomopatógenos são recentes e escassos. Nesse sentido, o objetivo deste estudo foi isolar NEP de amostras de solos coletadas em diferentes áreas no estado do Paraná, Brasil. Um isolado Heterorhabditis (UEL 08) detectado em amostra de solo em área de pastagem cultivada com braquiária, localizada em Londrina, Paraná, Brasil (23º34´311´´S, 050º58´298´´W), utilizando o método de “inseto-isca” com lagartas de Galleria mellonella. Para a identificação foram realizados estudos de morfometria e identificação molecular a partir da amplificação da região ITS. Algumas diferenças foram encontradas em termos de morfometria em comparação com a descrição original, entretanto, os dados moleculares demonstraram que a sequência obtida para o isolado UEL 08 é idêntica à sequência de referência de H. amazonensis (DQ665222), com a qual apresentou 100% de similaridade. Os estudos das características biológicas de H. amazonensis (UEL 08) revelaram que o isolado tem dois ciclos de vida distintos, um curto (216 h) e outro longo (288 h), sendo que ocorrem duas gerações em ambos os ciclos. O isolado UEL 08 H. amazonensis foi patogénico e virulento sobre os três hospedeiros avaliados. Notadamente, as larvas de G. mellonella e Alphitobius diaperinus foram consideradas mais susceptíveis do que os adultos do percevejo Dichelops (Diacereus) melacanthus, com percentagens de mortalidade de 100%, 85% e 46% de mortalidade, respectivamente. O ensaio de produção in vivo revelou um rendimento médio diário de 3,4 × 103 juvenis infectantes/g de larva hospedeira.

Honey bee abdominal pigmentation is one of the most recognisable traits and it is often used by beekeepers as an indicator of M-lineage subspecies purity. However, this approach may negatively impact population diversity and is futile if there is no association between tergite colour patterns and the genetic background. To assess whether this trait can be used as a proxy for introgression proportions in M-lineage subspecies, we genotyped, with highly informative SNP assays, A. m. mellifera and A. m. iberiensis individuals displaying four different colour phenotypes. The SNP data detected highly introgressed bees exhibiting a black phenotype and, at the same time, pure or marginally introgressed bees with yellow banding patterns, in both subspecies. Despite these observations, contrary to A. m. iberiensis , in A. m. mellifera , introgression proportions revealed to be a significant predictor of abdominal pigmentation. Therefore, abdominal pigmentation could be used by A. m. mellifera conservationists to guide colony selection when genetic tools are unavailable. Cancer is one of the leading causes of death worldwide and requires intense and growing research investments from the public and private sectors. This is expected to lead to the development of new medicines. A determining factor in this process is the structural understanding of molecules with potential anticancer properties. Since the major compounds used in cancer therapies fail to encompass every spectrum of this disease, there is a clear need to research new molecules for this purpose. As it follows, we have studied the class of quinolinones that seem effective for such therapy. This paper describes the structural elucidation of a novel dihydroquinoline by single-crystal X-ray diffraction and spectroscopy characterization. Topology studies were carried through Hirshfeld surfaces analysis and molecular electrostatic potential map; electronic stability was evaluated from the calculated energy of frontier molecular orbitals. Additionally, in silico studies by molecular docking indicated that this dihydroquinoline could act as an anticancer agent due to their higher binding affinity with human aldehyde dehydrogenase 1A1 (ALDH 1A1). Tests in vitro were performed for VERO (normal human skin keratinocytes), B16F10 (mouse melanoma), and MDA-MB-231 (metastatic breast adenocarcinoma), and the results certified that compound as a potential anticancer agent. A Dihydroquinoline derivative was tested against three cancer cell lines and the results attest that compound as potential anticancer agent.

Chalcones exhibit a broad spectrum of biological activities, mainly due to α,β-unsaturated ketone, and are precursors of the biosynthesis of flavonoids present in plants. These compounds have been shown to be useful in the biological approach, proven by the broad spectrum of biological activities reported, and also in the technological approach, considering their potential as nonlinear optic (NLO) material. In this context, this work aimed to examine the crystallization and characterization of fluorinated chalcone (E)-1-(4-fluorophenyl)-3-(4-isopropylphenyl)prop-2-en-1-one (DFC). A comprehensive structural study of DFC was carried out to understand the process of stabilizing the crystalline lattice through X-ray diffraction, infrared spectroscopy, and molecular modeling studies. Finally, the electrical properties of DFC were calculated by using the supermolecule method (SM). DFC molecules are connected by means of CH···O and CH···F intermolecular contacts, forming dimers which play an important role in the stabilization of crystal packing. Molecular modeling studies indicated that this compound could act as an anti-tuberculosis ligand because of its high binding affinity with the M. tuberculosis enoyl-acyl carrier protein, InhA. On the other hand, theoretical calculations were performed to evaluate the NLO properties of DFC and indicated that it showed good potential. Schistosomiasis control in endemic areas depends on several factors, including mass drug delivery programs and interrupting the transmission of disease by controlling the intermediate host snails in the freshwater ecosystem using molluscicides. However, the use of the gold standard molluscicide, i.e., niclosamide, has been considered problematic due to its high cost, toxicity for aquatic organisms, and the emergence of niclosamide-resistant snail populations. In this work, we report the in silico driven identification of novel naphthoquinone compounds with high molluscicidal activity against Biomphalaria glabrata. For this purpose, we developed statistically robust and validated shape-based and machine learning models using B. glabrata bioassay compounds data. Using these models, we prioritized fourteen naphthoquinone compounds for further in vivo testing against adult, newly-hatched, and embryo of B. glabrata snails. Among them, compounds 3, 5, 6, 7, and 12 were the best candidates, presenting moderate potency against adult snails (LC50: 28.98–102.24 μM) and high potency (LC50: 14.52–0.45 μM) against newly-hatched snails and embryos. To summarize, the in silico approach explored here allowed us to discover five new molluscicidal candidates for prospective field studies.

We propose a model of Majorana fermions with quartic self-couplings. These Majorana fermions acquire masses via a type II seesaw mechanism in which the physical eigenstates are identified as a light Majorana fermion and another heavy Majorana fermion. On a physical basis, the quartic self-couplings involve axial currents of these Majorana fermions, and also the interaction of the axial current for the light particle with the heavy particle one. We introduce two auxiliaries gauge fields in this model, and we study the stability conditions of the correspondent effective potential of the model. The ground state of the effective potential introduces two 4-vectors as scales of vacuum expected values, and consequently, the dynamical Lorentz symmetry breaking (DLSB) emerges in the model. We use the expansion of the effective action to calculate the effective Lagrangian up to second order in the auxiliary fields as fluctuations around the ground state. This mechanism generates dynamics for the auxiliary gauge fields, mixed mass terms, longitudinal propagation, and Chern-Simons term through radiative corrections. After the diagonalization, the two gauge fields gain masses through an analogous type II seesaw mechanism in which a gauge boson has a light mass, and the other one acquires a heavy mass. In this scenario of Lorentz symmetry breaking, we obtain the correspondent dispersion relations for the Majorana fermions and the gauge boson fields. Posteriorly, we analyze the neutrino's oscillations in the presence of a DLSB parameter, in the transition $\nu_{e} \rightarrow \nu_{\mu}$. The properties of the modified Maxwell electrodynamics (ModMax) are investigated in presence of external and uniform electric and magnetic fields. We expand the non-linear theory around an electromagnetic background up to second order in the propagating fields to obtain the permittivity and permeability tensors, dispersion relations, group velocity and refractive indices as functions of external fields. The case with perpendicular background fields is contemplated. The phenomenon of birefringence is discussed and the fundamental role of the ModMax parameter becomes clear. We calculate the difference of the refractive indices in terms of this parameter and the external fields. Finally, we set up a scenario where the axion is present and compute the interaction energy for the coupled axion-ModMax electrodynamics if a magnetic background field is considered. This calculation is carried out in the framework of the gauge-invariant, but path-dependent variables formalism. Objectives: The objectives of this study were to determine whether conducting a clinical audit was achievable in a group of centres that perform interventional cardiac procedures and to report the success and complications rates in dogs diagnosed with patent ductus arteriosus (PDA). Methods: This was a multi-centre, European-wide, prospective study. Patient data were entered into a bespoke database prior to commencing interventional closure of PDA in all animals undergoing this procedure during the study period. The database was designed to gather clinical audit information, after completion of the procedure, such as discharge outcome, complication rate and medium-term outcome. Results: A total of 339 cases were included from five participating centres. The process of performing clinical audit was achieved in all centres. The successful discharge outcome was 95.9% with a complication rate of 4.1%. The procedure-related mortality was 0.6%. A total of 149 cases (43.9%) were either lost to follow-up or had not yet had a follow-up within the time period. Of the remaining 169 cases in which follow-up was available, 157 (92.9%) had a successful medium-term outcome Conclusions: This study demonstrates that the process of performing a clinical audit is achievable in veterinary clinical interventions across different centres. These results provide a benchmark for future comparison in our ongoing clinical audit and validate the process of clinical audit for other centres performing cardiac interventions. The use of clinical audit should be considered in other aspects of veterinary medicine.

Paracoccidioidomycosis (PCM) is a systemic mycosis, endemic in Latin America, caused by fungi of the genus Paracoccidioides. The treatment of PCM is complex, requiring a long treatment period, which often results in serious side effects. The aim of this study was to screen for inhibitors of a specific target of the fungus that is absent in humans. Methylcitrate synthase (MCS) is a unique enzyme of microorganisms and is responsible for the synthesis of methylcitrate at the beginning of the propionate degradation pathway. This pathway is essential for several microorganisms, since the accumulation of propionyl-CoA can impair virulence and prevent the development of the pathogen. We performed the modeling and molecular dynamics of the structure of Paracoccidioides lutzii MCS (PlMCS) and performed a virtual screening on 89,415 compounds against the active site of the enzyme. The compounds were selected according to the affinity and efficiency criteria of in vitro tests. Six compounds were able to inhibit the enzymatic activity of recombinant PlMCS but only the compound ZINC08964784 showed fungistatic and fungicidal activity against Paracoccidioides spp. cells. The analysis of the interaction profile of this compound with PlMCS showed its effectiveness in terms of specificity and stability when compared to the substrate (propionyl-CoA) of the enzyme. In addition, this compound did not show cytotoxicity in mammalian cells, with an excellent selectivity index. The stinkbug Euschistus heros is an important pest of Brazilian agricultural production with increasing importance in the Neotropical region. High reproductive potential and increased risk of resistance to insecticides are determining factors influencing its pest status. This work evaluated the effects of pyriproxyfen on the reproduction and morphological development of the reproductive system of E. heros. Pyriproxyfen was utilized at sublethal concentrations (LC30) in fourth-instar nymphs (N4). Sublethal pyriproxyfen resulted in a reduction in fertility and fecundity of 2 and 12% when only the males were treated, 47 and 53% when only the females were treated, and 32 and 46% when females and males were treated. Basal oocytes were larger and more previtellogenic in newly emerged pyriproxyfen-treated females. The nuclear area of testicular accessory cells shrunk after 10 and 15 days of age in treated insects. Similarly, testicular shape was altered, changing from an ellipsoid to an obpyriform pattern, characterized by hypertrophy of the basal region over time. The apparent area of the organ grew until ten days of age, then shrunk at 15 days of age. In summary, pyriproxyfen treatments disrupted the normal development of N4 of E. heros and diminished the reproductive potential of female and male adults.

The Neotropical brown stink bug, Euschistus heros, is the most important sucking pest of soybean crops, quantitatively and qualitatively affecting grain and seed. This study aimed to determine mouthpart deformities, feeding inhibition, and E. heros nymphs’ mortality when in contact with insect growth disruptors (IGDs). The effects of two insecticides of the chitin biosynthesis inhibitor group (CBIs), lufenuron and diflubenzuron, and the juvenile hormone analog pyriproxyfen were tested in the laboratory. CBI application promoted mouthpart deformities, feeding inhibition, alteration of metamorphosis, and mortality of E. heros nymphs. Although pyriproxyfen did not cause mouthpart deformation, it caused certain mortality for nymphs, possibly due to physiological disorders. Lufenuron caused feed inhibition and mortality in less time when compared with diflubenzuron and pyriproxyfen. It was also more toxic to 4th instar nymphs. The insecticides, when used in higher concentrations, increased nymph’s mortality in less time. Nymphs with deformed mouthparts were unable to feed and died. All IGDs in the tested concentrations caused 100% mortality and prevented adult formation.

Chemically induced toxicity is the leading cause of recent extinction of honey bees. In this regard, we developed an innovative artificial intelligence-based web app (BeeToxAI) for assessing the acute toxicity of chemicals to Apis mellifera. Initially, we developed and externally validated QSAR models for classification (external set accuracy ∼91%) through the combination of Random Forest and molecular fingerprints to predict the potential for chemicals to cause acute contact toxicity and acute oral toxicity to honey bees. Then, we developed and externally validated regression QSAR models ( R 2 = 0.75) using Feedforward Neural Networks (FNNs). Afterward, the best models were implemented in the publicly available BeeToxAI web app ( http://beetoxai.labmol.com.br/ ) . The outputs of BeeToxAI are: toxicity predictions with estimated confidence, applicability domain estimation, and color-coded maps of relative structure fragment contributions to toxicity. As an additional assessment of BeeToxAI performance, we collected an external set of pesticides with known bee toxicity that were not included in our modeling dataset. BeeToxAI classification models were able to predict four out of five pesticides correctly. The acute contact toxicity model correctly predicted all of the eight pesticides. Here we demonstrate that BeeToxAI can be used as a rapid new approach methodology for predicting acute toxicity of chemicals in honey bees.

Hydrocotyle umbellata L. is a medicinal herb for the treatment of some health problems including hypertension, according to traditional medicine. Even so, its vascular effects and the pharmacological action mechanisms have not been analyzed.This experiment aimed to analyze the effects of hydroalcoholic extract of Hydrocotyle umbellata L. (HEHU) on isolated vessels and verify the interaction of hibalactone (chemical marker) against CaVascular reactivity experiments were performed using rat aortas with (EHEHU (0-560 μg/mL) induced relaxation of the pre-contracted arteries in a concentration-dependent manner. Objetivo: Verificar a associação entre a ocorrência de discrepâncias medicamentosas na admissão hospitalar e o tempo de internação em pacientes cirúrgicos em um hospital de ensino. Método: Trata-se de estudo de coorte retrospectivo, realizado com todos os pacientes internos na clínica cirúrgica no período compreendido entre os anos de 2014 a 2018. As discrepâncias foram avaliadas comparando os medicamentos da primeira prescrição médica em ambiente hospitalar e os medicamentos constantes na melhor história possível da medicação, document este constituinte do prontuário farmacêutico que detalha o uso dos medicamentos em domicílio na semana que antecede a internação. Essas informações foram obtidas através de entrevista estruturada com o paciente, aplicada por farmacêutico clínico nas primeiras 24h de entrada do paciente no hospital. Background Endoscopic remission (ER) is currently endorsed as one of the main treatment targets in Crohn’s Disease (CD). In a previous study, we have shown that transmural remission (TR) is associated with better clinical outcomes up to 1-year. It is unknown if these results still hold over a longer follow-up Methods This was a multicenter study, including 333 CD patients with magnetic resonance enterography (MRE) and colonoscopy evaluation performed within a 6-month interval and at least 5-years of follow-up. Patients were classified as having TR (inactive MRE and colonoscopy), ER (active MRE and inactive colonoscopy), and no remission (NR) (active colonoscopy). The need for surgery, hospitalization, steroids, and biologics was evaluated at 5-years of follow-up. Results Patients with TR presented lower rates of surgery (1.9% vs 17.9% vs 23.7%, P Conclusion TR was associated with improved clinical outcomes over 5-years of follow-up. Going beyond ER appears to provide significant clinical benefits in the short and long-term.

Only ~1% of all drug candidates against Neglected Tropical Diseases (NTDs) have reached clinical trials in the last decades, underscoring the need for new, safe and effective treatments. In such context, drug repositioning, which allows finding novel indications for approved drugs whose pharmacokinetic and safety profiles are already known, emerging as a promising strategy for tackling NTDs. Chemogenomics is a direct descendent of the typical drug discovery process that involves the systematic screening of chemical compounds against drug targets in high-throughput screening (HTS) efforts, for the identification of lead compounds. However, different to the one-drug-one-target paradigm, chemogenomics attempts to identify all potential ligands for all possible targets and diseases. In this review, we summarize current methodological development efforts in drug repositioning that use state-of-the-art computational ligand- and structure-based chemogenomics approaches. Furthermore, we highlighted the recent progress in computational drug repositioning for some NTDs, based on curation and modeling of genomic, biological, and chemical data. Additionally, we also present in-house and other successful examples and suggest possible solutions to existing pitfalls.

Aim: Computer-aided drug design approaches were applied to identify chalcones with antiplasmodial activity. Methodology: The virtual screening was performed as follows: structural standardization of in-house database of chalcones; identification of potential Plasmodium falciparum protein targets for the chalcones; homology modeling of the predicted P. falciparum targets; molecular docking studies; and in vitro experimental validation. Results: Using these models, we prioritized 16 chalcones with potential antiplasmodial activity, for further experimental evaluation. Among them, LabMol-86 and LabMol-87 showed potent in vitro antiplasmodial activity against P. falciparum, while LabMol-63 and LabMol-73 were potent inhibitors of Plasmodium berghei progression into mosquito stages. Conclusion: Our results encourage the exploration of chalcones in hit-to-lead optimization studies for tackling malaria.

Using magnetic measurements structural irregularities in electrodeposited nanotubes are inferred. The correlation between morphology and magnetic properties is very useful for studying materials. Through the use of the magnetic switching field distribution, the association with the surface irregularities imperfections of the magnetic nanotubes was performed. The distribution of magnetic switching field presents well behaved in smooth-surface nanotubes and, for nanotubes with irregularities, the distribution of switching fields has a noisy behavior, added to that smoothed signal. This noise is due to the superficial irregularities on the tubes surface because the magnetic barriers occurring in imperfect structures. Pinning centers due to irregularities are the principal aspect to observe in this situation. Magnetic measurements were associated directly to the morphology obtained by scanning electron microscopy.

As part of a drug discovery program aimed at the identification of anti- Trypanosoma cruzi metabolites from Brazilian flora, four acetogenins (1-4) were isolated from the seeds of Porcelia macrocarpa and were identified by NMR spectroscopy and HRESIMS. The new compounds 1 and 2 displayed activity against the trypomastigote (IC50 = 0.4 and 3.6 μM) and amastigote (IC50 = 23.0 and 27.7 μM) forms. The structurally related known compound 3 showed less potency to the amastigotes, with an IC50 value of 58 μM, while the known compound 4 was inactive. To evaluate the potential mechanisms for parasite death, parameters were evaluated by fluorometric assays: (i) plasma membrane permeability, (ii) plasma membrane electric potential (ΔΨp), (iii) reactive oxygen species production, and (iv) mitochondrial membrane potential (ΔΨm). The results obtained indicated that compounds 1 and 2 depolarize plasma membranes, affecting ΔΨp and ΔΨm and contributing to the observed cellular damage and disturbing the bioenergetic system. In silico studies of pharmacokinetics and toxicity (ADMET) properties predicted that all compounds were nonmutagenic, noncarcinogenic, nongenotoxic, and weak hERG blockers. Additionally, none of the isolated acetogenins 1-4 were predicted as pan-assay interference compounds.

Os produtos químicos perigosos, especialmente os agrotóxicos e produtos químicos industriais, vem sendo considerados responsáveis por uma queda dramática no número das abelhas. Consequentemente, a avaliação de risco ecológico de novos produtos químicos é vital e necessária. Uma vez que os ensaios experimentais em abelhas são caros, demorados e apresentam um problema ético, existe uma necessidade muito urgente de desenvolver métodos alternativos para avaliar a ecotoxicidade em abelhas. Na presente revisão, nós apresentamos os principais avanços tecnológicos utilizados para identificar e filtrar com segurança, compostos potencialmente tóxicos para abelhas. Em seguida, nós destacamos os principais esforços e armadilhas na integração, preparo e padronização de dados necessários para construção de modelos computacionais e sugerimos roteiros que possam contribuir para otimizar os resultados da pesquisa e conduzir a construção de modelos computacionais mais eficientes e preditivos.

Schistosomiasis is a parasitic disease caused by trematode worms of the genus Schistosoma and affects over 200 million people worldwide. The control and treatment of this neglected tropical disease is based on a single drug, praziquantel, which raises concerns about the development of drug resistance. This, and the lack of efficacy of praziquantel against juvenile worms, highlights the urgency for new antischistosomal therapies. In this review we focus on innovative approaches to the identification of antischistosomal drug candidates, including the use of automated assays, fragment-based screening, computer-aided and artificial intelligence-based computational methods. We highlight the current developments that may contribute to optimizing research outputs and lead to more effective drugs for this highly prevalent disease, in a more cost-effective drug discovery endeavor.
