Abstract Our legs act as our primary contact with the surrounding environment, generating external forces that enable agile motion. To be agile, the nervous system has to control both the magnitude of the force that the feet apply to the ground and the point of application of this force. The purpose of this study was to characterize the performance of the healthy human neuromechanical system in controlling the force-magnitude and position of an externally applied force. To accomplish this, we built an apparatus that immobilized participants but allowed them to exert variable but controlled external forces with a single leg onto a ground embedded force plate. We provided real-time visual feedback of either the leg force-magnitude or force-position that participants were exerting against the force platform and instructed participants to best match their real-time signal to prescribed target step functions. We tested target step functions of a range of sizes and quantified the responsiveness and accuracy of the control. For the control of force-magnitude and for intermediate step sizes of 0.45 bodyweights, we found a bandwidth of 1.8 ± 0.5 Hz, a steady-state error of 2.6 ± 0.9%, and a steady-state variability of 2.7 ± 0.9%. We found similar control performance in terms of responsiveness and accuracy across step sizes and between force-magnitude and position control. Increases in responsiveness correlated with reductions in other measures of control performance, such as a greater magnitude of overshooting. We modelled the observed control performance and found that a second-order model was a good predictor of external leg force control. We discuss how benchmarking force control performance in young healthy humans aids in understanding differences in agility between humans, between humans and other animals, and between humans and engineered systems.

Traditional approaches used in the pharmaceutical industry are not precise or versatile enough for customized medicine formulation and manufacture. Here the authors produce a method to form coatings, with accurate dosages, as well as a means of closely controlling dissolution kinetics.

Mangroves are found globally throughout tropical and sub-tropical inter-tidal coastlines. These highly biodiverse and carbon-dense ecosystems have multi-faceted value, providing critical goods and services to millions living in coastal communities and making significant contributions to global climate change mitigation through carbon sequestration and storage. Despite their many values, mangrove loss continues to be widespread in many regions due primarily to anthropogenic activities. Accessible, intuitive tools that enable coastal managers to map and monitor mangrove cover are needed to stem this loss. Remotely sensed data have a proven record for successfully mapping and monitoring mangroves, but conventional methods are limited by imagery availability, computing resources and accessibility. In addition, the variable tidal levels in mangroves presents a unique mapping challenge, particularly over geographically large extents. Here we present a new tool—the Google Earth Engine Mangrove Mapping Methodology (GEEMMM)—an intuitive, accessible and replicable approach which caters to a wide audience of non-specialist coastal managers and decision makers. The GEEMMM was developed based on a thorough review and incorporation of relevant mangrove remote sensing literature and harnesses the power of cloud computing including a simplified image-based tidal calibration approach. We demonstrate the tool for all of coastal Myanmar (Burma)—a global mangrove loss hotspot—including an assessment of multi-date mapping and dynamics outputs and a comparison of GEEMMM results to existing studies. Results—including both quantitative and qualitative accuracy assessments and comparisons to existing studies—indicate that the GEEMMM provides an accessible approach to map and monitor mangrove ecosystems anywhere within their global distribution.

The black rot pathogen Xanthomonas campestris utilizes molecules of the diffusible signal factor (DSF) family as signals to regulate diverse processes contributing to virulence. DSF signal synthesis and transduction requires proteins encoded by the rpf gene cluster. RpfF catalyzes DSF synthesis, whereas the RpfCG two-component system links the perception of DSF to alteration in the level of the second messenger cyclic di-GMP. As this nucleotide can exert a regulatory influence at the post-transcriptional and post-translational levels, we have used comparative proteomics to identify Rpf-regulated processes in X. campestris that may not be revealed by transcriptomics. The abundance of a number of proteins was altered in rpfF, rpfC, or rpfG mutants compared with the wild type. These proteins belonged to several functional categories, including biosynthesis and intermediary metabolism, regulation, oxidative stress or antibiotic resistance, and DNA replication. For many of these proteins, the alteration in abundance was not associated with alteration in transcript level. A directed mutational analysis allowed us to describe a number of new virulence factors among these proteins, including elongation factor P and a putative outer membrane protein, which are both widely conserved in bacteria.

The phytopathogenic bacterium Xanthomonas axonopodis pv. citri is responsible for the canker disease affecting citrus plants throughout the world. Here, we have evaluated the role of bacterial attachment and biofilm formation in leaf colonization during canker development on lemon leaves. Crystal violet staining and confocal laser scanning microscopy analysis of X. axonopodis pv. citri strains expressing the green fluorescent protein were used to evaluate attachment and biofilm formation on abiotic and biotic (leaf) surfaces. Wild-type X. axonopodis pv. citri attached to and formed a complex, structured biofilm on glass in minimal medium containing glucose. Similar attachment and structured biofilm formation also were seen on lemon leaves. An X. axonopodis pv. citri gumB mutant strain, defective in production of the extracellular polysaccharide xanthan, did not form a structured biofilm on either abiotic or biotic surfaces. In addition, the X. axonopodis pv. citri gumB showed reduced growth and survival on leaf surfaces and reduced disease symptoms. These findings suggest an important role for formation of biofilms in the epiphytic survival of X. axonopodis pv. citri prior to development of canker disease.

Cyclic di-GMP is an almost ubiquitous second messenger in bacteria that was first described as an allosteric activator of cellulose synthase but is now known to regulate a range of functions, including virulence in human and animal pathogens. Two protein domains, GGDEF and EAL, are implicated in the synthesis and degradation, respectively, of cyclic di-GMP. These domains are widely distributed in bacteria, including plant pathogens. The majority of proteins with GGDEF and EAL domains contain additional signal input domains, suggesting that their activities are responsive to environmental cues. Recent studies have demonstrated that a third domain, HD-GYP, is also active in cyclic di-GMP degradation. In the plant pathogen Xanthomonas campestris pv. campestris, a two-component signal transduction system comprising the HD-GYP domain regulatory protein RpfG and cognate sensor RpfC positively controls virulence. The signals recognized by RpfC may include the cell-cell signal DSF, which also acts to regulate virulence in X. campestris pv. campestris. Here, we review these recent advances in our understanding of cyclic di-GMP signaling with particular reference to one or more roles in the bacterial pathogenesis of plants.

The gum gene cluster of Xanthomonas campestris pv. campestris comprises 12 genes whose products are involved in the biosynthesis of the extracellular polysaccharide xanthan. These genes are expressed primarily as an operon from a promoter upstream of the first gene, gumB. Although the regulation of xanthan synthesis in vitro has been well studied, nothing is known of its regulation in planta. A reporter plasmid was constructed in which the promoter region of the gum operon was fused to gusA. In liquid cultures, the expression of the gumgusA reporter was correlated closely with the production of xanthan, although a low basal level of β-glucuronidase activity was seen in the absence of added carbon sources when xanthan production was very low. The expression of the gumgusA fusion also was subject to positive regulation by rpfF, which is responsible for the synthesis of the diffusible signal factor (DSF). The expression of the gumgusA fusion in bacteria recovered from inoculated turnip leaves was maximal at the later phases of growth and was subject to regulation by rpfF. These results provide indirect support for the operation of the DSF regulatory system in bacteria in planta.

Inoculation of pepper leaves, Capsicum annuum cv. Early Calwonder ECW 10R, with strains of Xanthomonas campestris led to an accumulation of the phenolic conjugates feruloyltyramine (FT) and p-coumaroyltyramine (CT) 24 h postinoculation in nonhost- and gene-for-gene-determined incompatible interactions with X. campestris pv. campestris and X. campestris pv. vesicatoria, respectively. In contrast, neither compound was detected in compatible interactions with X. campestris pv. vesicatoria. The accumulation of FT and CT was preceded by an increase in the extractable activity of tyrosine decarboxylase as well as increases in the transcription of genes encoding phenylalanine ammonia-lyase and tyramine hydroxycinnamoyl transferase. No such changes were detected in compatible interactions. Very rapid accumulation of FT and CT occurred (4 h postinoculation) in pepper in response to a X. campestris pv. campestris mutant carrying a deletion of the hrp gene cluster. In contrast, hrp mutants of X. campestris pv. vesicatoria failed to elicit the production of FT and CT. These observations suggest the existence of hrp gene-dependent and -independent activation mechanisms of a defense response involving hydroxycinnamoyltyramines.

Culture supernatants of Xanthomonas campestris pv. campestris contain an enzymic activity capable of degrading gp120, a proline-rich glycoprotein associated with the extracellular matrix of the vascular bundles in petioles of turnip (Brassica campestris). This activity did not reside in any of the three previously characterized proteases of X. campestris pv. campestris that were identified by their action against the model substrate β-casein. The novel enzyme was purified by ion-exchange and size-exclusion high-performance liquid chromatography (HPLC). The enzyme, which has no activity against β-casein, is active against some plant glycoproteins of the hydroxyproline-rich class such as extensin from potato and tomato and gpS-3, a glycoprotein induced in B. campestris petioles by wounding. Other hydroxyproline-rich glycoproteins, such as the solanaceous lectins, were not substrates however. Studies of the products released upon degradation of tomato extensin suggested that the degradative mechanism was proteolysis. Inhibitor studies suggested that the enzyme was a zinc-requiring metalloprotease. Extracellular matrix glycoproteins of the proline-rich and hydroxy-proline-rich classes have been implicated in plant resistance to microbial attack, hence their degradation by X. campestris pv. campestris may have considerable significance for black rot pathogenesis.

A panel of monoclonal antibodies that recognize plant extracellular matrix glycoproteins previously implicated in plant-microbe interactions was used to study the effects of pathogen inoculation and wounding on glycoproteins in petioles of Brassica campestris. The panel of monoclonals comprised two sets: JIM11, JIM12, and JIM20 recognize epitopes carried on hydroxyproline-rich glycoproteins (HRGPs) (M. Smallwood, A. Beven, N. Donovan, S. J. Neill, J. Peart, K. Roberts, and J. P. Knox, Plant J. 5:237–246, 1994); MAC204 and MAC265 recognize glycoproteins of the Rhizobium infection thread (K. A. VandenBosch, D. J. Bradley, S. Perotto, G. W. Butcher, and N. J. Brewin, EMBO J. 8:335–342, 1989). Wounding or inoculation of petioles with avirulent strains of pathovars of Xanthomonas campestris induced the synthesis of two new groups of antigens: gp160 ran as a smear on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with apparent molecular mass from 120 to 200 kDa and was recognized by JIM20 and MAC204; gpS remained in the stacking gel on SDS-PAGE and was recognized by JIM11, JIM20, and MAC204. The response to virulent strains of pathovars of X. campestris was either less pronounced or absent. gpS comprised several components that were resolved by cation-exchange chromatography. Some of these components were characterized as extensin-like HRGPs. The level of induction of the gpS group of antigens by virulent strains was not altered by mutation of a number of genes required for basic pathogenicity or by heat-killing the bacteria.

In previous work, the pigB transcriptional unit of Xanthomonas campestris was implicated in the generation of a diffusible factor (DF) that regulates xanthomonadin and extracellular polysaccharide (EPS) production. Similarly, the rpfF and rpfB genes were implicated in the generation of a diffusible factor (DSF) that regulates the production of extracellular enzymes and EPS. In this study, extracel-lular cross-feeding studies with rpfF and pigB mutant strains provided evidence for the existence of two separate intercellular signaling systems with overlapping roles in the regulation of EPS production.

Pre-treatment of leaves of pepper (Capsicum annuum) with lipopolysaccharide (LPS) preparations from enteric bacteria and Xanthomonas campestris could prevent the hypersensitive response caused by an avirulent X. campestris strain. By use of a range of deep-rough mutants, the minimal structure in Salmonella LPS responsible for the elicitation of this effect was determined to be lipid A attached to a disaccharide of 2-keto-3-deoxyoctulosonate; lipid A alone and the free core oligosaccharide from a Salmonella Ra mutant were not effective. For Xanthomonas, the core oligosaccharide alone had activity although lipid A was not effective. The results suggest that pepper cells can recognize different structures within bacterial LPS to trigger alterations in plant response to avirulent pathogens.

Une enquête épidémiologique sur la trypanosomose bovine a été réalisée en Gambie de novembre 1987 à octobre 1989. Elle a concerné la race trypanotolérante Ndama en zone de faible et forte pression glossinienne et visait la connaissance des paramètres de production de cette race en élevage traditionnel villageois et l'étude des facteurs les influençant. Naissances et mortalités, changements de poids des animaux, expulsion d'oeufs de strongles, fréquences des infections annuelles et mensuelles de la trypanosomose sont présentés ainsi que les variations de l'hématocrite. Les auteurs concluent que la production bovine de race Ndama en zone infestée par la trypanosomose est rentable mais conditionnée par la disponibilité en nourriture et la gestion du troupeau au sens large (contrôle des feux, sélection, traitement anthelminthique régulier, complémentation).

To help educators deliver their physiology laboratory courses remotely, we developed an inexpensive, customizable hardware kit along with freely available teaching resources. We based the course design on four principles that should allow students to conduct insightful experiments on different physiological systems. First, the experimental setup should not be constrained to laboratory environments. Second, students should be able to take this course without prior coding and electronics experience. Third, the hardware kit should be relatively inexpensive, and all other resources should be freely available. Fourth, all resources should be customizable for educators. The hardware kit consists of commercially available electronic components, with a microcontroller as its hub (Arduino friendly). All measurement systems can be assembled without soldering. The hardware kit is cost-effective (approximately the cost of a textbook) and can be customized depending upon instructional needs. All software is freely available, and we share all necessary codes in open-access online repositories for simple use and customizability. All lab manuals and additional video tutorials are also freely available online and customizable. In our particular course, we have weekly asynchronous physiology lectures and one synchronous laboratory session, where students can get help with their equipment. In this article, we only focus on the novel and open-source laboratory part of the course. The laboratory includes four units [data acquisition, ECG, electromyography (EMG), activity classification] and one final project. It is our intent that these resources will allow other educators to rapidly implement their own remote physiology laboratories or to extend our work into other pedagogical applications of wearable technology.

La mayoría de las bacterias fitopatógenas viven de forma epifítica sobre las plantas antes de la colonización y sobreviven en la superficie del tejido del hospedante, mediante la formación de biopelículas. La capacidad para producir exopolisacáridos es crítica para la formación de las mismas en diversas bacterias. En este trabajo, se estudió la formación de biopelículas de X. citri subsp. citri, bacteria responsable de la cancrosis de los cítricos, y su relación con la supervivencia epifítica y el desarrollo del cancro sobre hojas de plantas de limonero. Mediante tinción con cristal violeta y análisis de microscopía confocal de barrido láser, se observó que X. citri subsp. citri forma estructuras de biopelículas sobre hojas de limonero. En contraste, una cepa mutante defectiva en la biosíntesis del exopolisacárido xantano, denominada X. citri subsp. citri gumB, fue incapaz de formar dichas estructuras y su supervivencia epifítica y capacidad de desarrollar la enfermedad en la planta se vieron comprometidas. Estos resultados sugieren que la formación de biopelículas tiene una importancia clave en la fase epifítica de X. citri subsp. citri antes y durante el desarrollo del cancro.

Low‐grade serous ovarian carcinoma (LGSC) is a rare and lethal subtype of ovarian cancer. LGSC is pathologically, biologically, and clinically distinct from the more common high‐grade serous ovarian carcinoma (HGSC). LGSC arises from serous borderline ovarian tumours (SBTs). The mechanism of transformation for SBTs to LGSC remains poorly understood. To better understand the biology of LGSC, we performed whole proteome profiling of formalin‐fixed, paraffin‐embedded tissue blocks of LGSC (n = 11), HGSC (n = 19), and SBTs (n = 26). We identified that the whole proteome is able to distinguish between histotypes of the ovarian epithelial tumours. Proteins associated with the tumour microenvironment were differentially expressed between LGSC and SBTs. Fibroblast activation protein (FAP), a protein expressed in cancer‐associated fibroblasts, is the most differentially abundant protein in LGSC compared with SBT. Multiplex immunohistochemistry (IHC) for immune markers (CD20, CD79a, CD3, CD8, and CD68) was performed to determine the presence of B cells, T cells, and macrophages. The LGSC FAP+ stroma was associated with greater abundance of Tregs and M2 macrophages, features not present in SBTs. Our proteomics cohort reveals that there are changes in the tumour microenvironment in LGSC compared with its putative precursor lesion, SBT. These changes suggest that the tumour microenvironment provides a supportive environment for LGSC tumourigenesis and progression. Thus, targeting the tumour microenvironment of LGSC may be a viable therapeutic strategy. © 2024 The Pathological Society of Great Britain and Ireland. [ABSTRACT FROM AUTHOR]

The use of interferometry for heat transfer studies enables a complete field of view to be quantitatively examined at one time without disturbing the temperature distribution being measured. The Mach-Zehnder interferometer described has an elliptical field, with a 15cm minor axis and is capable of accommodating test chambers 102cm in length using the full field of view. The sensitivity of the instrument for various air pressures and temperature distributions within the test cell has been evaluated. The use of the instrument for identifying isotherms in the neighbourhood of a heated slab supported on skid bars is described. A Moiré technique which eliminates the effects of imperfections in the interferometer plates has also been used.
