Abstract: Understanding the interaction between the fusion plasma and the plasma-facing materials (PFMs) is crucial for achieving the optimal performance, safety, and lifetime of the fusion devices. Relevant materials have been intensely studied to determine fuel retention and the composition of the co-deposited layers on the PFMs by depth profile analysis using Laser-Induced Breakdown Spectroscopy (LIBS) and Calibration-Free (CF)-LIBS. However, the comparison between the layers containing a mixture of different (seeding) gases using LIBS, has not been studied systematically. Consequently, the aim of this work is the LIBS depth profile analysis of Be-based samples containing the fuel (D) and one of the seeding gases (N, Ne, He) with variable concentrations to study the potential impact on fuel retention in the PFMs. The LIBS measurements were performed using Nd:YAG laser (1064 nm) under Ar atmosphere (at 2 and 100 mbar). In LIBS the ablation rate was evaluated, spectral lines of all relevant elements except N and Ne were detected and compared with the SIMS depth profiles. The reasons for the non-detectability of Ne are discussed in detail.

Abstract: Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

Abstract: Within the 9th European Framework programme, since 2021 EUROfusion is operating five tokamaks under the auspices of a single Task Force called ‘Tokamak Exploitation’. The goal is to benefit from the complementary capabilities of each machine in a coordinated way and help in developing a scientific output scalable to future largre machines. The programme of this Task Force ensures that ASDEX Upgrade, MAST-U, TCV, WEST and JET (since 2022) work together to achieve the objectives of Missions 1 and 2 of the EUROfusion Roadmap: i) demonstrate plasma scenarios that increase the success margin of ITER and satisfy the requirements of DEMO and, ii) demonstrate an integrated approach that can handle the large power leaving ITER and DEMO plasmas. The Tokamak Exploitation task force has therefore organized experiments on these two missions with the goal to strengthen the physics and operational basis for the ITER baseline scenario and for exploiting the recent plasma exhaust enhancements in all four devices (PEX: Plasma EXhaust) for exploring the solution for handling heat and particle exhaust in ITER and develop the conceptual solutions for DEMO. The ITER Baseline scenario has been developed in a similar way in ASDEX Upgrade, TCV and JET. Key risks for ITER such as disruptions and run-aways have been also investigated in TCV, ASDEX Upgrade and JET. Experiments have explored successfully different divertor configurations (standard, super-X, snowflakes) in MAST-U and TCV and studied tungsten melting in WEST and ASDEX Upgrade. The input from the smaller devices to JET has also been proven successful to set-up novel control schemes on disruption avoidance and detachment.

Abstract: In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

Abstract: Building on prior analysis of ASDEX Upgrade (AUG) experiments (Henderson et al 2023 Nucl. Fusion 63 086024), this study compares simple analytical formula predictions for divertor detachment onset and reattachment timescales in JET experiments. Detachment onset primarily scales with divertor neutral pressure, impurity concentration, power directed to the targets, machine size, and integral perpendicular power decay length. JET experiments, focusing on seeding mixtures of Ne and Ar, align with the detachment onset predictions. Radiation efficiencies among the impurities show good agreement with the model predictions, contrasting with AUG observations which suggested higher efficiency for Ar and lower efficiency for Ne. The time taken to re-ionise the neutral volume in front of the outer target in fully detached divertor conditions was measured following both abrupt increases in injected neutral beam power and, separately, cutting of the impurity gas flow. Re-ionisation of the neutrals occurs within approximately 1 s on JET, which aligns with the simple model prediction derived from AUG data. While the AUG results are not new, their comparison with the JET results enhances understanding, reinforcing confidence in using simple models to predict future reactor scenarios.

Abstract: The experimental and theoretical analysis were focused on experiments conducted to assess the effect of plasma isotopes, protium (H), deuterium (D), and tritium (T) on ion cyclotron resonance heating (ICRH) related plasma wall interactions. Comparison of L-mode discharges with N = 1 ^3 He and N = 1 H minority ICRH heating scenarios were done for different isotopes. For the selected pulses, the behaviour of high-Z, mid-Z and low-Z intrinsic impurity and radiated power behaviour was investigated based on data from VUV, visible spectroscopy, and bolometry diagnostic at Joint European Torus. It was found that for N = 1 ^3 He scenario during radiofrequency antennas operation, core W, Ni content, Be source and the radiated power are higher for π /2 in comparison to dipole antenna phasing. Lowest core Ni, W content and radiated power is clearly observed for H plasmas in comparison to D and T, where for this ICRH scenario behaviour was similar. However, lower Be photon flux is observed for T in comparison to D plasmas. Be sputtering by He particles is responsible for such an effect. Additionally, several computer simulations were conducted using the COREDIV code. The difference in the electron temperature was due to the difference in the isotope masses. Increased temperature in the central plasma in the case of T plasmas leads to higher radiation in the central plasma in comparison to H plasmas. As a result, the power across separatrix is lower and the temperature on the divertor plate decreases with the increase of the isotope mass. At these temperatures on the divertor plate, W is not sputtered by the main plasma ions H, D and T and by He. For the N = 1 H ICRH scenario clear difference between D and T plasma was observed with higher metallic impurity content for T plasma in comparison to D. Impurity content in the plasmas is found to be sensitive to the power balance between the antenna straps. Its minimum is observed for the maximum of P _cen / P _tot .

7. First SOLPS-ITER predictions of the impact of cross-field drifts on divertor and scrape-off layer conditions in double-null configuration of STEP

Abstract: Introduction of cross-field drifts in SOLPS-ITER simulations of connected double-null plasmas in STEP with the ion $\nabla B \times B$ drift towards the upper divertors was found to enhance the detachment of the inner divertors with decreased target densities and ion and heat fluxes, while simultaneously complicating the access to detachment in the outer lower divertor by increasing the target temperature and heat loads to levels above the engineering limits. The $\nabla B \times B$ drift was observed to significantly affect the radial heat transport between the core and the scrape-off layer (SOL), altering the poloidal temperature and pressure profiles and, consequently, the poloidal conductive and convective heat transport in the SOL. As a result, up-down asymmetries of 52:48 and 58:42 biased towards the outer lower and upper inner divertors, respectively, were observed to arise in the unmitigated power entering the divertor regions, breaking the up-down symmetry seen in simulations without the drift terms and contradicting with earlier experimental observations on the low-field side. Moreover, the upstream electron density was found to decrease noticeably in the core and separatrix regions following the activation of the drifts due to an increased share of the neutrals arriving from D _2 injections near the upper and lower X-points ionizing already in the private flux regions.

Abstract: JET, the world’s largest operating tokamak with unique Be/W wall and tritium handling capability, completed a Deuterium-Tritium (D-T) campaign in 2021 (Maggi et al 29th Fusion Energy Conf. ) following a decade of preparatory experiments, dedicated enhancements, technical rehearsals and training (Horton et al 2016 Fusion Eng. Des. 109–111 925). Operation with tritium raises significant technical, safety and scientific challenges not encountered in standard protium or deuterium operation. This contribution describes the tritium operational requirements, pulses and technical preparations, new operating procedures, lessons learned and details on the achieved operational availability and performance. The preparation and execution of the recent JET tritium experiments benefitted from the previous experience in 1991 (Preliminary Tritium Experiment), 1997 (DTE1 campaign) and 2003 (Trace Tritium Campaigns) and consisted of the following five phases: technical rehearsals and scenario preparation, tritium commissioning, 100% tritium campaign, D-T campaign (DTE2), tritium clean-up. Following the clean-up JET resumed normal operation and is currently undertaking a further D-T campaign (DTE3).

Abstract: This paper reports the first experiment carried out in deuterium–tritium addressing the integration of a radiative divertor for heat-load control with good confinement. Neon seeding was carried out for the first time in a D–T plasma as part of the second D–T campaign of JET with its Be/W wall environment. The technical difficulties linked to the re-ionisation heat load are reported in T and D–T. This paper compares the impact of neon seeding on D–T plasmas and their D counterpart on the divertor detachment, localisation of the radiation, scrape-off profiles, pedestal structure, edge localised modes and global confinement.

Abstract: The roles of the molecularly assisted ionization (MAI), recombination (MAR) and dissociation (MAD) reaction chains with respect to the purely atomic ionization and recombination processes were studied experimentally during detachment in low-confinement mode (L-mode) plasmas in JET with the help of experimentally inferred divertor plasma and neutral conditions, extracted previously from filtered camera observations of deuterium Balmer emission, and the reaction coefficients provided by the ADAS, AMJUEL and H2VIBR atomic and molecular databases. The direct contribution of MAI and MAR in the outer divertor particle balance was found to be inferior to the electron–atom ionization (EAI) and electron–ion recombination (EIR). Near the outer strike point, a strong atom source due to the D2+-driven MAD was, however, observed to correlate with the onset of detachment at outer strike point temperatures of Te,osp=0.9–2.0 eV via increased plasma-neutral interactions before the increasing dominance of EIR at Te,osp< 0.9 eV, followed by increasing degree of detachment. The analysis was supported by predictions from EDGE2D-EIRENE simulations which were in qualitative agreement with the experimental observations.

Abstract: Measurements of the ion currents to and plasma conditions at the low-field side (LFS) divertor target plate in low-confinement mode plasmas in the JET ITER-like wall materials configuration show that the core plasma density required to detach the LFS divertor plasma is independent of the hydrogenic species protium, deuterium and tritium, and a 40 %/60 % deuterium–tritium mixture. This observation applies to a divertor plasma configuration with the LFS strike line connected to the horizontal part of the LFS divertor chosen because of its superior diagnostic coverage. The finding is independent of the operational status of the JET cryogenic pump. The electron temperature (Te) at the LFS strike line was markedly reduced from 25 eV to 5 eV over a narrow range of increasing core plasma density, and observed to be between 2 eV and 3 eV at the onset of detachment. The electron density (ne) peaks across the LFS plasma when Te at the target plate is 1 eV, and spatially moves to the X-point for higher core densities. The density limit was found approximately 20 % higher in protium than in tritium and deuterium–tritium plasmas.

12. Validation of SOLPS-ITER simulations with kinetic, fluid, and hybrid neutral models for JET-ILW low-confinement mode plasmas

Abstract: For JET L-mode plasmas in low-recycling conditions (electron temperature at the outer strike point, Te,ot≳30eV), SOLPS-ITER simulations agree within the error bars for the experimental profiles at the low-field side (LFS) divertor target. The peak Balmer-α (Dα) emission in the LFS divertor agrees within the error bars of the KS3 filterscope diagnostic, but is approximately 30% lower than the peak value of the KT1 spectrometer. Simulations have been performed with fluid, kinetic, and hybrid models for the neutrals. The large fluid-kinetic discrepancies of more than a factor 2 are successfully corrected by using a hybrid fluid-kinetic approach, for which kinetic atoms are transferred to the fluid population when the local Knudsen number of the atom becomes smaller than a user-defined transition Knudsen number Knt. The hybrid-kinetic discrepancies are limited to a few % for Knt≤100. When increasing the upstream density to high-recycling conditions, at the onset of detachment (Te,ot≈5eV), the simulations predict more than a factor 2 lower peak ion saturation current to the LFS divertor than the experiments. Also the Dα emission is underpredicted with approximately a factor 2. For these high-recycling conditions, the fluid-kinetic discrepancies are limited to maximum 50%, which are again corrected by using the hybrid approach.

13. EDGE2D-EIRENE and ERO2.0 predictions of nitrogen molecular break-up and transport in the divertor of JET low-confinement mode plasmas

Abstract: EDGE2D-EIRENE simulations of nitrogen-seeded partially detached JET l-mode plasmas show that the divertor N I to N V radiation distributions are highly sensitive to the upstream electron density, and less sensitive to changes in the assumed cross-field particle diffusivity for nitrogen ions. The EDGE2D-EIRENE simulations reproduce the peak intensities of N I to N V as measured by vertically viewing divertor filterscopes to within 50 % for a narrow range of the upstream electron density within the experimental uncertainties, while the predicted profiles are narrower than the measured ones. Including nitrogen atoms only in the ERO2.0 simulations implies lower N III and N IV peak intensities by one and two thirds, respectively, compared to EDGE2D-EIRENE. If nitrogen is assumed to recycle exclusively as molecules instead of atoms, ERO2.0 predicts that the N III and N IV intensities in the divertor increase by up to a factor of two and that the time-averaged, volume-integrated number of N5+ to N7+ ions in the plasma also increases by approximately a factor of 2.

Abstract: Simulations of JET ITER-like wall high-confinement mode plasmas, including type-I edge-localised modes (ELMs), using JINTRAC for the background plasmas and ERO2.0 for tungsten erosion and transport, predict virtually perfect screening of the primary W erosion sources at the divertor targets during both the ELM and inter-ELM phases. The largest source of W influx to the main plasma is predicted to be the outer vertical divertor due to sputtering by energetic fuel (D, T) atoms from charge-exchange reactions. ERO2.0 predictions accurately reproduce the measured W I emission in the low-field side divertor, but underpredict the W II emission by a factor of 10. Potential reasons for the W II discrepancy include uncertainties in the atomic data, assumptions on the sheath properties and the sputtering angle distribution, and the impact of metastable states.

Abstract: The dimensionless isotope mass scaling experiment between pure Deuterium and pure Tritium plasmas with matched $\rho^*$ , $\nu^*$ , β _n , q and $T_e/T_i$ has been achieved in JET L-mode with dominant electron heating (NBI+ohmic) conditions. 28% higher scaled energy confinement time $B_t\tau_{E,\mathrm{th}}/A$ is found in favour of the Tritium plasma. This can be cast in the form of the dimensionless energy confinement scaling law as $\Omega_i \tau_{E,\mathrm{th}} \sim A^{0.48 \pm 0.16}$ . This significant isotope mass scaling is consequently seen in the scaled one-fluid heat diffusion coefficient $A \chi_{\mathrm{eff}}/B_t$ which is around 50% lower in the Tritium plasma throughout the whole plasma radius. The isotope mass dependence in the particle transport channel is negligible, supported also by the perturbative particle transport analysis with gas puff modulation. The comparison of the edge particle fuelling or ionisation profiles from the EDGE2D-EIRENE simulations show that the absolute density differences that are necessary for the dimensionless match in the confined plasma dominate over any isotope mass dependencies of particle fuelling and ionization profiles at the plasma edge. Local GENE simulation results indicate a mild anti-gyroBohm effect at $\rho_\mathrm{tor}$ = 0.6 and thereby a small isotope mass dependence in favour of Tritium on heat transport and a negligible effect on particle transport. A significant fraction of the isotope scaling and reduced heat transport observed in the Tritium plasma is not captured in the GENE and ASTRA-TGLF-SAT2 simulations by simply changing the isotope mass for the same input profiles.

Abstract: After the second Deuterium–Tritium Campaign (DTE2) in the JET tokamak with the ITER-Like Wall (ILW) and full tritium campaigns that preceded and followed after the DTE2, a sequence of fuel recovery methods was applied to promote tritium removal from wall components. The sequence started with several days of baking of the main chamber walls at 240 °C and at 320 °C. Subsequently, baking was superimposed with Ion-Cyclotron Wall Conditioning (ICWC) and Glow Discharge Conditioning (GDC) cleaning cycles in deuterium. Diverted plasma operation in deuterium with different strike point configurations, including a Raised Inner Strike Point (RISP) configuration, and with different plasma heating—Ion Cyclotron Resonance Frequency (ICRF) and Neutral Beam Injection (NBI)—concluded the cleaning sequence. Tritium content in plasma and in the pumped gas was monitored throughout the experiment. The applied fuel recovery methods allowed reducing the residual tritium content in deuterium NBI-heated plasmas to about 0.1% as deduced from neutron rate measurements. This value is well below the requirement of 1% set by the maximum 14 MeV fusion neutron budget allocated in the ensuing deuterium plasma campaign. The quantified tritium removal over the course of the experiment was $\left( {13.4 \pm 0.7} \right) \times {10^{22}}$ atoms or $\left( {0.67 \pm 0.03} \right)$ g with ∼58% attributed to baking, ∼12.5% to ICWC, ∼26% to GDC, and ∼3.5% to first low power RISP plasmas. The experimentally estimated amount of removed tritium is in good agreement with long-term tritium accounting by the JET tritium reprocessing plant, in which the unaccounted amount was reduced by $0.71$ g after the cleaning experiment.

Abstract: While current tokamak experiments are beginning to use real-time feedback control systems to manage the plasma exhaust, future tokamaks still require validation of theoretical models used to predict the threshold impurity concentration required to sufficiently reduce the power and particle fluxes to the divertor. This work exploits new spectroscopic measurements of the divertor nitrogen concentration, cN, in partially detached N2-seeded H-mode plasmas on ASDEX Upgrade (AUG) and JET with the ITER-Like Wall (JET-ILW) to test the parameter dependencies of the power flowing to the outer divertor, Pdiv,outer, and the separatrix electron density, ne,sep. A least-squares regression of the AUG measurements demonstrates that the threshold cN required for detachment scales as cN∝Pdiv,outer1.19±0.32ne,sep-2.77±0.36. This scaling of ne,sep is also consistent with the measurements from JET which, at constant Pdiv,outer, show cN∝ne,sep-2.43±0.27. The dependencies of Pdiv,outer and ne,sep is demonstrated over at least a factor of two change in both parameters and indicates a stronger dependence on ne,sep in comparison to the Lengyel model, which could be due to the assumption in this model that the heat flux channel width is independent of density. This first assessment of detachment scaling with impurity seeding highlights the need for further analysis of the systematic uncertainties of the measurement and more consistent scenarios from more tokamaks to investigate the machine size scaling.

Abstract: A remote LIBS system is being considered as an analysis tool in ITER for monitoring the erosion and fuel retention in the first wall. This necessitates further investigation of the performance of LIBS for thick co-deposited layers consisting of ITER-relevant materials. The main goal of this work is determining the fuel content of the samples and D depth profile by LIBS and to compare the results with those obtained by other methods. The studied samples were Be-based mixed coatings on W substrates, containing D and in some cases C, O, or both. These impurities are relevant not only for ITER, but also for other fusion devices, such as JET-ILW. The laser ablation was performed at 10 mbar Ar pressure using a 5 ns pulse Nd:YAG laser at 1064 nm. Suitable Be and Ar spectral lines were employed for the evaluation of the electron temperature of the plasma using multi-elemental Saha-Boltzmann (MESB) plots. The electron density was obtained from the Stark broadening of the Dα and Hα spectral lines following deconvolution. The average elemental content of each coating was then obtained by calibration-free LIBS (CF-LIBS) and are in agreement with other techniques (TOF–ERDA, IBA).

Abstract: SOLPS 5.0 simulations assuming convection-dominated radial ion transport show qualitative and quantitative agreement with measurements of detached high-field side (HFS) divertor conditions for unseeded low-power L-mode plasmas in ASDEX Upgrade, while simultaneously maintaining a reasonable match to the density and temperature measurements at the low-field side (LFS) and HFS midplanes within the scatter of the data. The decreased diffusive transport from the high-density region in the HFS divertor volume across the separatrix into the core enables spatial extension of the high-density front to above the X-point in agreement with spectroscopic measurements. The suppressed ion fuelling from this region into the core plasma allows increasing the neutral D2 fuelling to experimental levels, leading to agreement with the measured sub-divertor neutral fluxes within 30%. Detachment of the HFS divertor is observed as a significant decrease of the target ion flux and as reproduction of the characteristic roll-over behaviour of the integrated target ion current at increasing upstream density.

20. Estimation of 2D distributions of electron density and temperature in the JET divertor from tomographic reconstructions of deuterium Balmer line emission

Abstract: Estimates for 2D distributions of electron density, ne, and temperature, Te, have been obtained using reflection-corrected tomographic reconstructions of deuterium Balmer line emission in the JET divertor. The solutions are obtained by a two-dimensional Monte Carlo optimization model, based on intensity ratios of the Balmer Dα, Dγ and Dε emission and the ADAS photon emission coefficients with additional constraints provided by line-integrated spectroscopic measurements of ne and Te. Synthetic benchmarks utilizing EDGE2D-EIRENE simulation outputs yield reproduction of reference ne and Te distributions within 25% with statistical error estimates of 5–15%. Experimental analysis of an L-mode ne ramp shows extension and movement of the outer divertor high-ne region with ne,div up to 1.5 × 1020 m−3 from the outer strike point to above the X-point and decrease of the divertor Te to the range of 0.5–1.0 eV within 25% agreement with the line-integrated measurements. Comparison between the local estimates and their corresponding line integrals suggests underestimation of ne and Te by up to 75% and 50%, respectively, due to the geometric effects of line integration.

Abstract: High speed visible imaging allows to visualize the 2-dimensional dynamics of fast phenomena in the boundary layer of fusion plasmas. Here we describe the two high speed visible cameras currently operating in the JET tokamak, enabling the simultaneous observation of a large fraction of the JET tokamak vacuum vessel with the appropriate configuration for many different plasma phenomena. As an example we discuss recent observations on the spatial and temporal dynamics of visible emission at the outer wall during Edge Localized Modes (ELMs) and the penetration of Shattered Injected Pellet (SPI) into the plasma.

22. Measurement of N+ flows in the high-field side scrape-off layer of ASDEX upgrade with different degrees of inner divertor detachment

Abstract: Toroidal and poloidal flows of injected N+ ions were measured in the high-field side (HFS) scrape-off layer (SOL) of ASDEX Upgrade by Doppler spectroscopy with different degrees of HFS divertor detachment. In high-recycling conditions, the results suggest reversed parallel N+ flow away from the inner divertor in the near SOL close to the separatrix, while the flow is towards the inner divertor throughout the SOL in detached conditions. The measured poloidal N+ flows were directed away from the HFS divertor in the near SOL for all density cases. Divertor plasma oscillations, characterized by momentary peaking of the HFS target ion flux and decrease of the HFS SOL density, were observed slightly before the roll-over of the ion saturation current to the HFS target and lead to an increase in the N+ flow towards the HFS divertor. SOLPS and ERO simulations of the experiment predict entrainment below 50% between the velocities of N+ and D+ ions, suggesting that N+ ions are quantitatively a limited proxy for measuring D+ flows. 23. Loading of deuterium and helium by Pilot-PSI plasma and their detection by in-situ LIBS

Abstract: Laser-induced breakdown spectroscopy (LIBS) is a promising method for quantifying the fuel content of the plasma-facing components of ITER both in between plasma discharges (in-situ) and after maintenance operations. The aim of the present study is to test the applicability of in-situ LIBS for monitoring deuterium (D) and helium (He) content of W samples exposed to fusion relevant plasma fluxes in the linear plasma device Pilot-PSI.The D loading was performed during 1000s of plasma exposure at low (200-300 °C) surface temperatures. Despite of low intensity and noisy LIBS spectra, H and D lines, at 656.1 and 656.3nm, respectively, could be fitted with Lorentzian contours and reliably resolved at 1.2 mbar background pressure of argon.In the case of He loading, the samples were also exposed to plasma during 1000s while the surface temperature reached values up to 720 °C at the center. Already at 10–2 mbar residual pressure of the device, the He I line at 587.6nm was visible for the first 2–3 laser shots.We demonstrated that in-situ LIBS is a reliable method for detection of He and D retention in ITER-relevant materials. Nevertheless, for measuring relative and absolute concentrations of D and He in the ITER-relevant samples, further studies are needed. 24. ERO and PIC simulations of gross and net erosion of tungsten in the outer strike-point region of ASDEX Upgrade

Abstract: We have modelled net and gross erosion of W in low-density l-mode plasmas in the low-field side strike point region of ASDEX Upgrade by ERO and Particle-in-Cell (PIC) simulations. The observed net-erosion peak at the strike point was mainly due to the light impurities present in the plasma while the noticeable net-deposition regions surrounding the erosion maximum could be attributed to the strong E ×B drift and the magnetic field bringing eroded particles from a distance of several meters towards the private flux region. Our results also imply that the role of cross-field diffusion is very small in the studied plasmas. The simulations indicate net/gross erosion ratio of 0.2–0.6, which is in line with the literature data and what was determined spectroscopically. The deviations from the estimates extracted from post-exposure ion-beam-analysis data (∼0.6–0.7) are most likely due to the measured re-deposition patterns showing the outcomes of multiple erosion-deposition cycles. 25. Determination of deuterium depth profiles in fusion-relevant wall materials by nanosecond LIBS

Abstract: The ablation and fuel-retention characteristics of aluminum–tungsten (AlW) and beryllium–tungsten (BeW) samples have been determined using Laser Induced Breakdown Spectroscopy (LIBS) and compared to results obtained using Secondary Ion Mass Spectrometry (SIMS). The measurements have been made both at vacuum (of the order of 10−4Pa) and at 50Pa of argon to especially enhance the intensities of the spectral lines of H and D. For reliable evaluation of the ablation rate of the samples the electron density ne and temperature Te of the LIBS plasma have been determined with the help of selected of spectral lines of Be, Al, and W. The electron density ne has been obtained from Stark broadening lines of Al I (308.7nm and 394.4nm) and Be I (457.3nm) and Te from the Saha–Boltzmann plot using W I and W II spectral lines having a higher value of the energy of upper states in order to prevent the influence of self-absorption on the results. The results indicate similar ablation characteristics between AlW (AlWD) and BeW (BeWD) samples but the inclusion of deuterium in the coating increases the ablation rate by a factor of 10 for both sample types. Concerning fuel retention more than one order of magnitude less D is retained in the AlWD sample than in BeWD. In the presence of background argon, the H and D lines were stronger and more easily distinguishable. This is a positive sign considering the real application in ITER where LIBS measurements are foreseen to be done during maintenance breaks. However the higher pressure gave a better signal, it is still far from the measurement conditions planned for ITER which need to be tested separately. Keywords: LIBS, Beryllium, Aluminum

27. A neural network algorithm for cloud fraction estimation using NASA-Aura OMI VIS radiance measurements

Abstract: The discrimination of cloudy from cloud-free pixels is required in almost any estimate of a parameter retrieved from satellite data in the ultraviolet (UV), visible (VIS) or infrared (IR) parts of the electromagnetic spectrum. In this paper we report on the development of a neural network (NN) algorithm to estimate cloud fractions using radiances measured at the top of the atmosphere with the NASA-Aura Ozone Monitoring Instrument (OMI). We present and discuss the results obtained from the application of two different types of neural networks, i.e., extreme learning machine (ELM) and back propagation (BP). The NNs were trained with an OMI data sets existing of six orbits, tested with three other orbits and validated with another two orbits. The results were evaluated by comparison with cloud fractions available from the MODerate Resolution Imaging Spectrometer (MODIS) flying on Aqua in the same constellation as OMI, i.e., with minimal time difference between the OMI and MODIS observations. The results from the ELM and BP NNs are compared. They both deliver cloud fraction estimates in a fast and automated way, and they both performs generally well in the validation. However, over highly reflective surfaces, such as desert, or in the presence of dust layers in the atmosphere, the cloud fractions are not well predicted by the neural network. Over ocean the two NNs work equally well, but over land ELM performs better.

Abstract: Introduction: There are several potential causes of QRS-axis deviation in the ECG, but there is limited data on the prognostic significance of QRS-axis deviation in ACS patients. Subjects and methods: We evaluated the long-term prognostic significance of acute phase frontal plane QRS-axis deviation and its shift during hospital stay in ACS patients. A total of 1026 patients who met the inclusion criteria were divided into three categories: normal (n = 823), left (n = 166) and right/extreme axis (n = 37). Results: The median survival time was 9.0 years (95% CI 7.9—10.0) in the normal, 3.6 years (95% CI 2.4—4.7) in the left and 1.3 years (95% CI 0.2—2.4) in the right/extreme axis category. Both short and long-term all-cause mortality was lowest in the normal axis category and highest in the right/extreme axis category. Compared to normal axis, both admission phase QRS-axis deviation groups were independently associated with a higher risk of all-cause mortality. When including left ventricular hypertrophy in the ECG, only the right/extreme axis retained its statistical significance (aHR 1.76; 95% CI 1.16—2.66, p = 0.007). Axis shift to another axis category had no effect on mortality. Conclusion: In ACS patients, acute phase QRS-axis deviation was associated with higher risk of all-cause mortality. Among the axis deviation groups, right/extreme QRS-axis deviation was the strongest predictor of mortality in the multivariable analysis. Further studies are required to investigate to what extent this association is caused by pre-existing or by ACS-induced axis deviations. Abstract: Introduction: Atrial fibrillation (AF) is a frequent finding in acute coronary syndrome (ACS), but there is conflicting scientific evidence regarding its long-term impact on patient outcome. The aim of this study was to survey and compare the ≥10-year mortality of ACS patients with sinus rhythm (SR) and AF. Methods: Patients were divided into 2 groups based on rhythm in their 12-lead ECGs: (1) SR (n = 788) at hospital admission and discharge (including sinus bradycardia, physiological sinus arrhythmia, and sinus tachycardia) and (2) AF/atrial flutter (n = 245) at both hospital admission and discharge, or SR and AF combination. Patients who failed to match the inclusion criteria were excluded from the final analysis. The main outcome surveyed was long-term all-cause mortality between AF and SR groups during the whole follow-up time. Results: Consecutive ACS patients (n = 1,188, median age 73 years, male/female 58/42%) were included and followed up for ≥10 years. AF patients were older (median age 77 vs. 71 years, p < 0.001) and more often female than SR patients. AF patients more often presented with non-ST-elevation myocardial infarction (69.8 vs. 50.4%, p < 0.001), had a higher rate of diabetes (31.0 vs. 22.8%, p = 0.009), and were more often using warfarin (32.2 vs. 5.1%, p < 0.001) or diuretic medication (55.1 vs. 25.8%, p < 0.001) on admission than patients with SR. The use of warfarin at discharge was also more frequent in the AF group (55.5 vs. 14.8%, p < 0.001). The rates of all-cause and cardiovascular mortality were higher in the AF group (80.9 vs. 50.3%, p < 0.001, and 73.8 vs. 69.6%, p = 0.285, respectively). In multivariable analysis, AF was independently associated with higher mortality when compared to SR (adjusted HR 1.662; 95% CI: 1.387–1.992, p < 0.001). Conclusion: AF/atrial flutter at admission and/or discharge independently predicted poorer long-term outcome in ACS patients, with 66% higher mortality within the ≥10-year follow-up time when compared to patients with SR.

Abstract: An overview of recent results obtained at the tokamak ASDEX Upgrade (AUG) is given. A work flow for predictive profile modelling of AUG discharges was established which is able to reproduce experimental H-mode plasma profiles based on engineering parameters only. In the plasma center, theoretical predictions on plasma current redistribution by a dynamo effect were confirmed experimentally. For core transport, the stabilizing effect of fast ion distributions on turbulent transport is shown to be important to explain the core isotope effect and improves the description of hollow low-Z impurity profiles. The L–H power threshold of hydrogen plasmas is not affected by small helium admixtures and it increases continuously from the deuterium to the hydrogen level when the hydrogen concentration is raised from 0 to 100%. One focus of recent campaigns was the search for a fusion relevant integrated plasma scenario without large edge localised modes (ELMs). Results from six different ELM-free confinement regimes are compared with respect to reactor relevance: ELM suppression by magnetic perturbation coils could be attributed to toroidally asymmetric turbulent fluctuations in the vicinity of the separatrix. Stable improved confinement mode plasma phases with a detached inner divertor were obtained using a feedback control of the plasma β. The enhanced Dα H-mode regime was extended to higher heating power by feedback controlled radiative cooling with argon. The quasi-coherent exhaust regime was developed into an integrated scenario at high heating power and energy confinement, with a detached divertor and without large ELMs. Small ELMs close to the separatrix lead to peeling-ballooning stability and quasi continuous power exhaust. Helium beam density fluctuation measurements confirm that transport close to the separatrix is important to achieve the different ELM-free regimes. Based on separatrix plasma parameters and interchange-drift-Alfvén turbulence, an analytic model was derived that reproduces the experimentally found important operational boundaries of the density limit and between L- and H-mode confinement. Feedback control for the X-point radiator (XPR) position was established as an important element for divertor detachment control. Stable and detached ELM-free phases with H-mode confinement quality were obtained when the XPR was moved 10 cm above the X-point. Investigations of the plasma in the future flexible snow-flake divertor of AUG by means of first SOLPS-ITER simulations with drifts activated predict beneficial detachment properties and the activation of an additional strike point by the drifts. Abstract: A sequence of fuel recovery methods was tested in JET, equipped with the ITER-like beryllium main chamber wall and tungsten divertor, to reduce the plasma deuterium concentration to less than 1% in preparation for operation with tritium. This was also a key activity with regard to refining the clean-up strategy to be implemented at the end of the 2nd DT campaign in JET (DTE2) and to assess the tools that are envisaged to mitigate the tritium inventory build-up in ITER. The sequence began with 4 days of main chamber baking at 320 °C, followed by a further 4 days in which Ion Cyclotron Wall Conditioning (ICWC) and Glow Discharge Conditioning (GDC) were applied with hydrogen fuelling, still at 320 °C, followed by more ICWC while the vessel cooled gradually from 320 °C to 225 °C on the 4th day. While baking alone is very efficient at recovering fuel from the main chamber, the ICWC and GDC sessions at 320 °C still removed slightly higher amounts of fuel than found previously in isotopic changeover experiments at 200 °C in JET. Finally, GDC and ICWC are found to have similar removal efficiency per unit of discharge energy. The baking week with ICWC and GDC was followed by plasma discharges to remove deposited fuel from the divertor. Raising the inner divertor strike point up to the uppermost accessible point allowed local heating of the surfaces to at least 800 °C for the duration of this discharge configuration (typically 18 s), according to infra-red thermography measurements. In laboratory thermal desorption measurements, maintaining this temperature level for several minutes depletes thick co-deposit samples of fuel. The fuel removal by 14 diverted plasma discharges is analysed, of which 9, for 160 s in total, with raised inner strike point. The initial D content in these discharges started at the low value of 3%–5%, due to the preceding baking and conditioning sequence, and reduced further to 1%, depending on the applied configuration, thus meeting the experimental target.

Abstract: Laser-induced breakdown spectroscopy (LIBS) is one of the most promising methods for quantitative in-situ determination of fuel retention in plasma-facing components (PFCs) of magnetically confined fusion devices like ITER and JET. In this article, the current state of understanding in LIBS development for fusion applications will be presented, based on a complete review of existing results and complemented with newly obtained data. The work has been performed as part of a research programme, set up in the EUROfusion Consortium, to address the main requirements for ITER: (a) quantification of fuel from relevant surfaces with high sensitivity, (b) the technical demonstration to perform LIBS with a remote handling system and (c) accurate detection of fuel at ambient pressures relevant for ITER. For the first goal, the elemental composition of ITER-like deposits and proxies to them, including deuterium (D) or helium (He) containing W–Be, W, W–Al and Be–O–C coatings, was successfully determined with a typical depth resolution ranging from 50 up to 250 nm per laser pulse. Deuterium was used as a substitute for tritium (T) and in the LIBS experiments deuterium surface densities below 1016 D/cm2 could be measured with an accuracy of ∼30%, confirming the required high sensitivity for fuel-retention investigations. The performance of different LIBS configurations was explored, comprising LIBS systems based on single pulse (pulse durations: ps–ns) and double pulse lasers with different pulse durations. For the second goal, a remote handling application was demonstrated inside the Frascati-Tokamak-Upgrade (FTU), where a compact, remotely controlled LIBS system was mounted on a multipurpose deployer providing an in-vessel retention monitor system. During a shutdown phase, LIBS was performed at atmospheric pressure, for measuring the composition and fuel content of different area of the stainless-steel FTU first wall, and the titanium zirconium molybdenum alloy tiles of the toroidal limiter. These achievements underline the capability of a LIBS-based retention monitor, which complies with the requirements for JET and ITER operating in DT with a beryllium wall and a tungsten divertor. Concerning the capabilities of LIBS at pressure conditions relevant for ITER, quantitative determination of the composition of PFC materials at ambient pressures up to 100 mbar of N2, the D content could be determined with an accuracy of 25%, while for atmospheric pressure conditions, an accuracy of about 50% was found when using single-pulse lasers. To improve the LIBS performance in atmospheric pressure conditions, a novel approach is proposed for quantitative determination of the retained T and the D/T ratio. This scenario is based on measuring the LIBS plume emission at two different time delays after each laser pulse. On virtue of application of a double pulse LIBS system, for LIBS application at N2 atmospheric pressure the distinguishability of the spectra from H isotopes could be significantly improved, but further systematic research is required.

Abstract: Two visible cameras with the same wide-angle view have been used to study the gross beryllium (Be) erosion in JET. An absolutely calibrated spectroscopy system employed in the past for the same object was used to quantitatively infer the camera photon fluxes and also to validate the results. To extract the effective sputtering yield, a simplified method is applied from the quotient of the measured radiances of the D α and the Be II line at 527 nm. The obtained results are in reasonably good agreement with those already published. The main benefit of using cameras is that the global view of the plasma–wall interaction surfaces of the whole vessel is monitored, and the different sources and their wetted areas can be identified and quantified. In addition, the measurement of the particle flux distributions can be used for the validation of three-dimensional erosion-transport codes.

Abstract: Background: \ud Spontaneous coronary artery dissection (SCAD) is an increasingly recognized cause of acute coronary syndromes (ACS) afflicting predominantly younger to middle-aged women. Observational studies have reported a high prevalence of extracoronary vascular anomalies, especially fibromuscular dysplasia (FMD) and a low prevalence of coincidental cases of atherosclerosis. PHACTR1/EDN1 is a genetic risk locus for several vascular diseases, including FMD and coronary artery disease, with the putative causal noncoding variant at the rs9349379 locus acting as a potential enhancer for the endothelin-1 (EDN1) gene.\ud \ud Objectives: \ud This study sought to test the association between the rs9349379 genotype and SCAD.\ud \ud Methods: \ud Results from case control studies from France, United Kingdom, United States, and Australia were analyzed to test the association with SCAD risk, including age at first event, pregnancy-associated SCAD (P-SCAD), and recurrent SCAD.\ud \ud Results: \ud The previously reported risk allele for FMD (rs9349379-A) was associated with a higher risk of SCAD in all studies. In a meta-analysis of 1,055 SCAD patients and 7,190 controls, the odds ratio (OR) was 1.67 (95% confidence interval [CI]: 1.50 to 1.86) per copy of rs9349379-A. In a subset of 491 SCAD patients, the OR estimate was found to be higher for the association with SCAD in patients without FMD (OR: 1.89; 95% CI: 1.53 to 2.33) than in SCAD cases with FMD (OR: 1.60; 95% CI: 1.28 to 1.99). There was no effect of genotype on age at first event, P-SCAD, or recurrence.\ud \ud Conclusions: \ud The first genetic risk factor for SCAD was identified in the largest study conducted to date for this condition. This genetic link may contribute to the clinical overlap between SCAD and FMD.

Abstract: Heavy alcohol use is one of the top causes of disease and death in the world. The brain is a key organ affected by heavy alcohol use. Here, our aim was to measure changes caused by heavy alcohol use in the human brain metabolic profile. We analyzed human postmortem frontal cortex and cerebrospinal fluid (CSF) samples from males with a history of heavy alcohol use (n = 74) and controls (n = 74) of the Tampere Sudden Death Series cohort. We used a nontargeted liquid chromatography mass spectrometry-based metabolomics method. We observed differences between the study groups in the metabolite levels of both frontal cortex and CSF samples, for example, in amino acids and derivatives, and acylcarnitines. There were more significant alterations in the metabolites of frontal cortex than in CSF. In the frontal cortex, significant alterations were seen in the levels of neurotransmitters (e.g., decreased levels of GABA and acetylcholine), acylcarnitines (e.g., increased levels of acylcarnitine 4:0), and in some metabolites associated with alcohol metabolizing enzymes (e.g., increased levels of 2-piperidone). Some of these changes were also significant in the CSF samples (e.g., elevated 2-piperidone levels). Overall, these results show the metabolites associated with neurotransmitters, energy metabolism and alcohol metabolism, were altered in human postmortem frontal cortex and CSF samples of persons with a history of heavy alcohol use. publishedVersion

Abstract: Dementia is among the most frequent causes of institutionalization. To serve the purpose of preventive strategies, there are no follow-up studies that have evaluated the actual impact of post-stroke dementia on institutionalization. We therefore compared the institutionalization rate and length of stay in an institutional care facility of patients with post-stroke dementia with stroke patients without dementia.We included 410 consecutive patients aged 55 to 85 years with ischemic stroke who were admitted to Helsinki University Hospital (The SAM cohort). Hospitalization and nursing home admissions were reviewed from national registries. Dementia was diagnosed using the Diagnostic and Statistical Manual of Mental Disorders 3rd edition (DSM-III) criteria using extensive clinical assessments performed 3 months post-stroke. The cohort had a follow-up 21 years later.Compared to patients without dementia, post-stroke dementia was associated with shorter survival time (6.60 vs 10.10 years, p 0.001), shorter time spent not institutionalized (5.40 vs 9.37 years, p 0.001), but not with time spent permanently institutionalized (0.73 vs 1.10 years, p = 0.08). Post-stroke dementia was associated with higher rates and earlier permanent institutionalization compared to absence of post-stroke dementia (HR 1.53, 95% CI 1.07-2.18) in a Cox regression model adjusting for age, status of living alone at baseline, modified Rankin Scale at baseline, history of atrial fibrillation, and cardiac failure.Post-stroke dementia is associated with earlier permanent institutionalization. Due to significantly shorter survival, the time spent in nursing homes was not significantly longer in patients with post-stroke dementia compared with patients without post-stroke dementia.

Abstract: Background A positive T wave in lead aVR (aVRT+) is an independent prognostic predictor of cardiovascular mortality in the general population as well as in cardiovascular disease. Subjects and methods We evaluated the prognostic impact of aVRT+ in an ECG recorded as close to hospital discharge as possible in acute coronary syndrome patients (n = 527). We divided the patients into three categories based on the findings in the admission ECG: ST elevation, global ischemia and other ST/T changes. Results In the whole study population, and in all the three ECG subgroups, the 10-year all-cause mortality rate was higher in the aVRT+ group than in the aVRT− group. In Cox regression analysis, the age and gender adjusted hazard ratio (HR) for aVRT+ to predict all-cause mortality in the whole study population was 1.43 (95% confidence interval [CI] 1.12–1.83; p = 0.004). To predict cardiovascular mortality, the age and gender adjusted HR for aVRT+ was 1.54 (95% CI 1.14–2.07; p = 0.005) in the whole study population and 2.07 (95% CI 1.07–4.03; p = 0.032) in the category with other ST/T changes. Conclusion In ACS patients with or without ST elevation, but with ischemic ST/T changes in their presenting ECG, a positive or isoelectric T wave in lead aVR in an ECG recorded in the subacute in-hospital stage is associated with all-cause and cardiovascular mortality during long-term follow-up. Clinicians should pay attention to this simple ECG finding at hospital discharge.

Abstract: Genome-wide association studies have discovered hundreds of loci associated with complex brain disorders, but it remains unclear in which cell types these loci are active. Here we integrate genome-wide association study results with single-cell transcriptomic data from the entire mouse nervous system to systematically identify cell types underlying brain complex traits. We show that psychiatric disorders are predominantly associated with projecting excitatory and inhibitory neurons. Neurological diseases were associated with different cell types, which is consistent with other lines of evidence. Notably, Parkinson’s disease was genetically associated not only with cholinergic and monoaminergic neurons (which include dopaminergic neurons) but also with enteric neurons and oligodendrocytes. Using post-mortem brain transcriptomic data, we confirmed alterations in these cells, even at the earliest stages of disease progression. Our study provides an important framework for understanding the cellular basis of complex brain maladies, and reveals an unexpected role of oligodendrocytes in Parkinson’s disease. © 2020, The Author(s), under exclusive licence to Springer Nature America, Inc.

Abstract: Haptoglobin (Hp) is a plasma protein that binds free hemoglobin and protects tissues from oxidative damage. An Hp2 allele has been associated with an increased risk of cardiovascular complications. On the other hand, recent studies have suggested that Hp1 allele increases risk to develop severe cerebral small vessel disease. We aimed to replicate this finding in a first-ever stroke patient cohort. Hp was genotyped by PCR and gel electrophoresis in the Helsinki Stroke Aging Memory Study in patients with DNA and magnetic resonance imaging (MRI) available (SAM, n = 316). Lacunar infarcts and white matter lesions (WML) classified by Fazekas grading from brain MRI were associated with Hp genotypes. As population controls, we used participants of Cardiovascular diseases&mdash, a sub study of Health 2000 Survey (n = 1417). In the SAM cohort, 63.0% of Hp1-1 carriers (n = 46), 52.5% of Hp1-2 carriers (n = 141) and 51.2% of Hp2-2 carriers (n = 129) had severe WML (p = 0.372). There was no difference in severe WMLs between Hp1-1 vs. Hp1-2 and Hp2-2 carriers (p = 0.201). In addition, 68.9% of Hp1-1 carriers (n = 45), 58.5% of Hp1-2 carriers (n = 135), and 61.8% of Hp2-2 carriers (n = 126) had one or more lacunar lesions (p = 0.472). There was no difference in the number of patients with at least one lacunar infarct between Hp1-1 vs. Hp1-2 and Hp2-2 groups (p = 0.322). Neither was there any difference when diabetic patients (type I and II) were examined separately. Hp1 allele is not associated with an increased risk for cerebral small vessel disease in a well-characterized Finnish stroke patient cohort.

Abstract: Background Human saliva contains approximately 700 bacterial species. It has been reported that the salivary microbiome of a large family of closely related individuals consisting of multiple households is similar but the relatedness of salivary bacteria between generations of parents and their children has not yet been investigated. The objectives were to investigate the entirety of salivary bacterial DNA profiles and whether and how families share these profiles and also compare these communities between grandparents and their first daughter generations (F1) using 16S rRNA gene amplicon sequencing. Results The most abundant phyla in two separate families were Firmicutes, Bacteroidetes, Proteobacteria, Fusobacteria and Actinobacteria. Family ties explained 13% of the variance between individuals’ bacterial communities (R2 = 0.13; P = 0.001). Mothers shared more OTUs with adult children compared to fathers, but this linkage seemed to be weaker in the nuclear family with older adult children. We identified 29 differentially abundant genus level OTUs (FDR < 0.05) between families, which accounted for 31% of the total identified genus level OTUs. Conclusions Our results indicate that adult family members share bacterial communities and adult children were more similar to mothers than fathers. The observed similarity in oral microbiome between parent–child pairs seemed to weaken over time. We suggest that our analysis approach is suitable for relatedness study of multigenerational salivary bacteria microbiome.

Abstract: Upstream transcription factor 1 (USF1) regulates the transcription of many genes related to cell and organism survival processes such as stress and immune response, regulation of cellular senesce, and carcinogenesis. In this study, our aim was to investigate the effect of USF1 single nucleotide variations (SNVs) on longevity in the Vitality 90+ study, a population-based study of nonagenarians (90 ±1 years of age) living in the area of Tampere municipality, Finland. Altogether 509 voluntary nonagenarians (115 males, 394 females) were genotyped using the 5'-nuclease assay for rs2774279G A, rs2516839T C, and rs2073658C T SNVs. During the 4 years of follow-up, the total mortality rate was 64.2%. In the study, we found that the frequency of C-allele of rs2516839 among nonsurviving nonagenarians (52.5%) was higher than those who survived (41.2%; P = 0.0006, odds ratio = 1.575, 95% confidence interval [CI]: 1.215-2.041). Furthermore, carriage of this variation and its haplotypes had a significant gender by genotype interaction (P 0.05) on mortality. Kaplan-Meier log-rank test during 4-years of follow-up showed significantly higher mortality rate in the case of CC genotype carriage than other genotype carriages in nonagenarian women (P 0.0001). In addition, after adjusting for age in Cox regression analysis, cardiovascular disease, diabetes, infectious disease, dementia, and living place (nursing home or home), CC genotype of rs2516839T C was found to be associated with shorter life expectancy in nonagenarian women (hazard ratio = 2.27; 95% CI, 1.34-3.85 P = 0.002). In conclusion, rs2516839 variation and related haplotypes of the USF1 gene are strongly related to all-cause mortality in Finnish nonagenarians, especially among women.

Abstract: Objective Dental bacterial DNA and bacterial-driven inflammation markers have previously been detected in intracranial aneurysm tissue samples. This study aimed (i) to assess the possible presence of dental infectious foci, (ii) and the possible association between typical odontogenic bacteria and clinical dental findings in patients undergoing pre-operative dental examination before surgical treatment of saccular intracranial aneurysm. Ninety patients with an intracranial aneurysm were recruited to the study, and the patients’ teeth were routinely investigated. Clinical data and bacterial samples from the gingival pockets were collected from a subpopulation of 60 patients. Abstract: Background and Purpose— Haptoglobin (Hp) is an acute phase plasma protein protecting tissues from oxidative damage. It exists in 2 variant alleles ( hp1/hp2 ) giving rise to 3 protein isoforms with different biochemical properties and efficiency to limit oxidative stress. We previously found that hp2 variant is associated with stroke risk in the patients with carotid stenosis and the risk of ischemic cardiovascular events in a general population cohort. This study examined the hypothesis that Hp genotype is associated with general cardiovascular risk in patients with stroke. Methods— Hp was genotyped in SAM study (Helsinki Stroke Aging Memory, n=378). A total of 1426 individuals ascertained from a nationally representative cross-sectional health survey served as population controls. Results— Hp genotype frequencies were 15.6% ( hp1-1 ), 44.2% ( hp1-2 ), and 40.2% ( hp2-2 ) in patients with stroke. During a mean of 7.5-year follow-up after first-ever stroke, hp2 carriers had a substantially higher rate of cardiac deaths (24.5% versus 8.5%; P =0.006) and a trend toward more fatal strokes (23.5% versus 13.6%; P =0.122). The combined risk of ischemic cardiovascular deaths was 2.4-fold higher among hp2 carriers (95% confidence interval, 1.28–4.43) after adjustment for major cardiovascular risk factors. Conclusions— Hp2 allele is associated with premature ischemic cardiovascular deaths after first-ever ischemic stroke.

Abstract: The ablation and fuel-retention characteristics of aluminum–tungsten (AlW) and beryllium–tungsten (BeW) samples have been determined using Laser Induced Breakdown Spectroscopy (LIBS) and compared to results obtained using Secondary Ion Mass Spectrometry (SIMS). The measurements have been made both at vacuum (of the order of 10−4Pa) and at 50Pa of argon to especially enhance the intensities of the spectral lines of H and D. For reliable evaluation of the ablation rate of the samples the electron density ne and temperature Te of the LIBS plasma have been determined with the help of selected of spectral lines of Be, Al, and W. The electron density ne has been obtained from Stark broadening lines of Al I (308.7nm and 394.4nm) and Be I (457.3nm) and Te from the Saha–Boltzmann plot using W I and W II spectral lines having a higher value of the energy of upper states in order to prevent the influence of self-absorption on the results. The results indicate similar ablation characteristics between AlW (AlWD) and BeW (BeWD) samples but the inclusion of deuterium in the coating increases the ablation rate by a factor of 10 for both sample types. Concerning fuel retention more than one order of magnitude less D is retained in the AlWD sample than in BeWD. In the presence of background argon, the H and D lines were stronger and more easily distinguishable. This is a positive sign considering the real application in ITER where LIBS measurements are foreseen to be done during maintenance breaks. However the higher pressure gave a better signal, it is still far from the measurement conditions planned for ITER which need to be tested separately. Keywords: LIBS, Beryllium, Aluminum

Abstract: Background: Precise cause of death (CoD) ascertainment is crucial in any cancer screening trial to avoid bias from misclassification due to excessive recording of diagnosed cancer as a CoD in death certificates instead of non-cancer disease that actually caused death. We estimated whether there was bias in CoD determination between screening (SA) and control arms (CA) in a population-based prostate cancer (PCa) screening trial. Methods: Our trial is the largest component of the European Randomized Study of Screening for Prostate Cancer with more than 80,000 men. Randomly selected deaths in men with PCa (N = 442/2568 cases, 17.2%) were reviewed by an independent CoD committee. Median follow-up was 16.8 years in both arms. Results: Overdiagnosis of PCa was present in the SA as the risk ratio for PCa incidence was 1.19 (95% confidence interval (CI) 1.14-1.24). The hazard ratio (HR) for PCa mortality was 0.94 (95% CI 0.82-1.08) in favor of the SA. Agreement with official CoD registry was 94.6% (k = 0.88) in the SA and 95.4% (k = 0.91) in the CA. Altogether 14 PCa deaths were estimated as false-positive in both arms and exclusion of these resulted in HR 0.92 (95% CI 0.80-1.06). Conclusions: A small differential misclassification bias in ascertainment of CoD was present, most likely due to attribution bias (overdiagnosis in the SA). Maximum precision in CoD ascertainment can only be achieved with independent review of all deaths in the diseased population. Abstract: Background and aims Focal adhesions (FA) play an important role in the tissue remodeling and in the maintenance of tissue integrity and homeostasis. Talin and vinculin proteins are among the major constituents of FAs contributing to cellular well-being and intercellular communication. Methods Microarray analysis (MA) and qRT-PCR low-density array were implemented to analyze talin-1, talin-2, meta-vinculin and vinculin gene expression in circulating blood and arterial plaque. Results All analyzed genes were significantly and consistently downregulated in plaques (carotid, abdominal aortic and femoral regions) compared to left internal thoracic artery (LITA) control. The use of LITA samples as controls for arterial plaque samples was validated using immunohistochemistry by comparing LITA samples with healthy arterial samples from a cadaver. Even though the differences in expression levels between stable and unstable plaques were not statistically significant, we observed further negative tendency in the expression in unstable atherosclerotic plaques. The confocal tissue imaging revealed gradient of talin-1 expression in plaque with reduction close to the vessel lumen. Similar gradient was observed for talin-2 expression in LITA controls but was not detected in plaques. This suggests that impaired tissue mechanostability affects the tissue remodeling and healing capabilities leading to development of unstable plaques. Conclusions The central role of talin and vinculin in cell adhesions suggests that the disintegration of the tissue in atherosclerosis could be partially driven by downregulation of these genes, leading to loosening of cell-ECM interactions and remodeling of the tissue.

49. Manner of death and demographic effects on microbial community composition in organs of the human cadaver

Abstract: The microbiome serves important functions in human health, and postmortem, the microbial signatures of colonized organ tissue could be useful in helping to predict the manner of death in cases where this information is not known. We surveyed the microbiota (16S rRNA V4 amplicon sequencing) of 265 organ tissue samples including liver, blood, brain, heart, prostate, spleen and uterus from cadavers in Italy, Finland and the United States with confirmed manners of death comprising either accidental death, natural death, homicide, and suicide. Geographic locality (i.e. nationality) had a strong effect on observed microbial composition. Differing PERMANOVA results between unweighted and weighted UniFrac (nearly inverse results) suggest that specific bacteria may be associated with ethnicity and age, but that these differences are negligible when taking into account the relative abundance of bacterial taxa; weighted UniFrac measures suggest that although taxonomic composition may not vary significantly between different manners of death, PMI, or BMI categories, the relative abundance of specific taxa vary significantly. Various tissues exhibit differential associations with bacteria, and prostate and uterus were substantially different compared to other organs. For example, in Italian cadavers, the bacteria MLE1-12 permeated nearly all tissues, except the prostate and uterus. We identified specific bacterial ASVs as biomarkers of either natural or accidental death and suicide, but not for homicide. While the manner of death may have an impact on microbial associations, further investigation under more controlled conditions will be needed to validate whether these associations are predictive in forensic determinations.ImportanceThe utilization of microbial data in the context of forensic investigations holds great promise for the field of forensic science. Identification of taxa that are associated with postmortem interval (PMI), specific manners of death (MOD), or other traits such as age, sex, ethnicity, and nationality may allow investigators to refine the circumstantial details surrounding the death of an individual. In this study we find nationality (geographic location of cadaver) to be a dominant predictor of cadaver microbiome composition. We also identify a number of cadaver-specific traits to be associated with microbial alpha- and beta diversity, as well as bacterial taxa that are differentially associated with these traits.

50. Mild Chronic Traumatic Encephalopathy Neuropathology in People With No Known Participation in Contact Sports or History of Repetitive Neurotrauma

Abstract: It has been asserted that chronic traumatic encephalopathy (CTE) pathology is only present in former athletes and others who have been exposed to repetitive concussions, subconcussive blows, or both. We hypothesized that CTE pathology would be present in men who had no known history of repetitive neurotrauma. Comprehensive medical record reviews and health surveys completed by a family member were available for the 8 men in this case series, none of whom had known exposure to repetitive neurotrauma but 2 of whom had a history of traumatic brain injury (TBI). Postmortem tissue was immunostained for hyperphosphorylated tau (p-tau) to assess for CTE pathology, Braak stage, and aging-related p-tau. The neuropathologist was blind to age, personal history, and clinical history. Six of the 8 cases (75%) showed p-tau in neurons, astrocytes, and cell processes around small blood vessels in an irregular pattern at the depths of the cortical sulci. The changes were focal and limited in terms of overall extent, and some of the cases had a clearer pattern of pathology and some could be considered equivocal. Two of the 8 cases had a history of TBI and one of them showed CTE pathology. Five of the 6 cases with no known history of neurotrauma appeared to meet consensus criteria for CTE. This study adds to the emerging literature indicating that CTE pathology is present in people not known to have experienced multiple concussions or subconcussive blows to the head.
