1. Structural characterization of the Aspergillus niger citrate transporter CexA uncovers the role of key residues S75, R192 and Q196

Abstract: The Aspergillus niger CexA transporter belongs to the DHA1 (Drug-H+ antiporter) family. CexA homologs are exclusively found in eukaryotic genomes, and CexA is the sole citrate exporter to have been functionally characterized in this family so far. In the present work, we expressed CexA in Saccharomyces cerevisiae, demonstrating its ability to bind isocitric acid, and import citrate at pH 5.5 with low affinity. Citrate uptake was independent of the proton motive force and compatible with a facilitated diffusion mechanism. To unravel the structural features of this transporter, we then targeted 21 CexA residues for site-directed mutagenesis. Residues were identified by a combination of amino acid residue conservation among the DHA1 family, 3D structure prediction, and substrate molecular docking analysis. S. cerevisiae cells expressing this library of CexA mutant alleles were evaluated for their capacity to grow on carboxylic acid-containing media and transport of radiolabeled citrate. We also determined protein subcellular localization by GFP tagging, with seven amino acid substitutions affecting CexA protein expression at the plasma membrane. The substitutions P200A, Y307A, S315A, and R461A displayed loss-of-function phenotypes. The majority of the substitutions affected citrate binding and translocation. The S75 residue had no impact on citrate export but affected its import, as the substitution for alanine increased the affinity of the transporter for citrate. Conversely, expression of CexA mutant alleles in the Yarrowia lipolytica cex1Δ strain revealed the involvement of R192 and Q196 residues in citrate export. Globally, we uncovered a set of relevant amino acid residues involved in CexA expression, export capacity and import affinity.

2. Crustal Imaging of Portugal Mainland Using Magnetotelluric Data

Abstract: Abstract The comprehensive mapping of resistivities in depth and its interpretation is of great importance to unravel the electrical properties of deep‐seated rocks, providing significant insights into the structure of the crust. The first 3D resistivity model for Portugal mainland is here reported using data from 31 broadband MT soundings spaced 50 × 50 km apart. The model shows large crustal volumes with contrasting resistivity values. The central and northern regions are characterized by a high resistivity crustal domain spreading in‐depth (103–105 Ω.m) that correlates well with voluminous granitoid bodies and high‐grade metamorphic rocks. To the west and the south, roughly corresponding to the Portuguese Western Shore and the South Portuguese Zone (SPZ) respectively, the crustal resistivity tends to decrease unevenly to values between 1 and 103 Ω.m. Interconnected graphite in structural discontinuities, possibly forming a regional mid‐crustal décollement at ≈13–15 km depth, are highlighted by multiple low resistivity crustal domains (1–100 Ω.m). Similarly, the model reveals a very large E‐W low resistivity crustal domain in SPZ possibly representing a deep‐seated major décollement. The present study aims to contribute to a better understanding of the physical properties characterizing the concealed crust in Portugal mainland, providing also indirect information on its composition and structure.

3. Life stress and Bipolar Disorder: regarding a clinical case

Abstract: Introduction Research on life stress in bipolar disorder largely fails to account for the possibility of a dynamic relationship between psychosocial stress and episode initiation. The kindling hypothesis states that over the course of recurrent affective disorders, there is a weakening temporal relationship between major life stress and episode initiation that could reflect either a progressive sensitization or progressive autonomy to life stress. Objectives To explore the concept of the Kindling model applied to bipolar disorder and to present a clinical case of a bipolar patient whose latter mood episodes were caused by adverse life events. Methods We performed a non-systematic literature review using the most relevant papers found on the database PubMed with the keywords “kindling effect”, “allostatic load”, “bipolar disorder” and “prevention”. Description of the clinical case report. Results The phenomenon of kindling was first discovered by Goddard in 1967 who described it in epilepsy. Later, Post applied it to the bipolar disorder, arguing that the initial episodes of both unipolar and bipolar affective disorders are often precipitated by psychosocial stressors, but after multiple recurrences, not only do precipitated episodes continue to occur, but so do spontaneous ones as well. We present the case report of a 62 years old woman, divorced, diagnosed with type 1 bipolar disorder since she was 20 years old. She always have had poor adherence to her medication and follow-up with Psychiatry consultation, with a non-containing sociofamily environment that does not promote clinical stability. Over the time, her admissions on the Psychiatry ward were more frequent and precipitated by adverse life events, mainly caused by the deteriorated relationship with her children. Conclusions The kindling model clarifies aspects of the longitudinal course of episode development, recurrence, and progression to spontaneity, as well as further conceptual and theoretical rationales for intervention in order to prevent illness progression. 4. Intensity and distribution in the space-time of the rain erosivity in Goias and Federal District states

Abstract: The ability of rainfall to promote soil disaggregation and transport depends on its intensity and distribution over time, allowing proper soil and water management practices in order to minimize their effects in and outside food production fields. The objective of this study was to verify the intensity of the spatial and temporal distribution of rainfall erosivity in Goias and Federal District states using historical series of rainfall data available at the National Institute of Meteorology (Inmet) from 1986 to 2017. Rainfall erosivity (EI) for Goias and Federal District were estimated using the modified Fournier index. Annual average for factor R were obtained by the sum of the EI in the 12 months of the year. Data were placed to descriptive analysis and classification according to their erosive capacity. The annual mean of the factor R for the study region was 8,834±5.2 MJ mm ha-1 h-1 ano-1, being classified as a strong erosive potential. However, considering the territory, its intensity in Goiás and DF varied from very strong to moderate throughout the year, indicating the need to adopt a conservationist practice that protects the soil from the kinetic energy of rainfall. It is concluded, therefore, that the intensity and distribution of the rainfall erosivity fluctuate both between the municipalities of Goiás (from 4.514±2.5 to 11.215±1.8 MJ mm ha-1 h-1 year-1), and throughout the year (from 180±1.5 to 1.519±4.4 MJ mm ha-1 h-1 month-1).

Abstract: Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). 6. Beneficial Effects of Renin-Angiotensin System Blockers on Testicular Morphology

Abstract: Background: The renin-angiotensin system (RAS) is a set of peptides, enzymes, and receptors specially involved in the control of extracellular fluid volume and blood pressure (BP); however, some of its components have already been identified in the testis, such as angiotensinogen, angiotensin converting enzyme, and renin. Objectives: The aim of this study was to evaluate whether renin-angiotensin system blockers have effects on the testicular morphology of animals fed a high energy density (HED) diet. Materials and Methods: Male C57BL/6 mice were fed initially a standard chow (SC) or a HED diet. After 8 weeks, HED animals were randomized into 4 groups, each group receiving one of the following treatments for the next 6 weeks: HED-A: aliskiren (50 mg/kg/d); HED-E: enalapril (30 mg/kg/d); HED-L: losartan (10 mg/kg/d); and untreated HED group. The BP was measured biweekly. At the end of treatment, all animals were killed and the testes were processed for morphometric and stereological parameters including density of seminiferous tubules per area, density of length and the total length of the seminiferous tubules, height of the epithelium, and diameter of the seminiferous tubules. Samples were tested for their homoscedasticity and the differences between the groups were tested by one-way analysis of variance (ANOVA), followed by the Holm-Sidak post-test. In all cases, the significance level adopted was P ≤ .05. Results: Compared to SC, HED groups presented an increase in BP, normalized by all RAS blockers. However, the HED diet caused testis alterations that were not affected by aliskiren or losartan. Only enalapril maleate was capable of reversing such alterations. Conclusions: Further studies are still needed to answer why only enalapril was able to reveal the morphological changes caused by the high energy diet; so enalapril could be suggested as the drug of choice for patients with previous reproductive dysfunction.

7. Influence of a 6-month physical training program on serum and urinary concentrations of trace metals in middle distance elite runners

Abstract: Background The aim of this survey was to determine the effects of an aerobic physical training program of six months duration on the serum and urinary concentrations of essential trace elements among middle distance runners and untrained, non-sportsmen participants. Methods 24 well-trained, middle-distance (1500 and 5000 m), aerobic male runners (AG) were recruited at the beginning of their training season and 26 untrained males formed the control group (CG). All participants were from the same region of Spain, and all of them had been living in this area for at least two years. Serum and urine of samples of Cobalt (Co), Copper (Cu), Manganese (Mn), Molybdenum (Mo), Selenium (Se), Vanadium (V) and Zinc (Zn) were obtained at the beginning of the training season, and six months later, from all participants. All samples were analyzed with inductively coupled plasma mass spectrometry (ICP-MS). Results Two-way ANOVA showed significant differences relative to group effect in serum concentrations of Co, Cu, Mn, Mo, Se and Zn. Attending to time effect, there were differences in Mn (p = 0.003) and Zn (p = 0.001). The group x time interaction revealed differences only in the case of Mn (p = 0.04). In urine, significant differences between group were obtained in Co, Cu, Mn, Se and V. Time effect showed changes in Co, Cy, Mo and Se. Finally, the group and time interaction revealed significant differences in urinary Cu (p = 0.001), Mn (p = 0.01) and Se (p = 0.001). Conclusions A six-month aerobic training program for well-trained athletes induced modifications in the body values of several minerals, a fact which may reflect adaptive responses to physical exercise. The obtained data could be interesting for physicians or coaches in order to consider specific modifications in sportsmen’s diets as well as to determine specific nutritional supplementation strategies.

Abstract: Asthma often remains uncontrolled, despite the fact that the pharmacological treatment has undergone large changes. We studied changes in the treatment of asthma over a 20-year period and identified factors associated with the regular use of inhaled corticosteroid (ICS) treatment. Changes in the use of medication were determined in 4617 randomly selected subjects, while changes in adults with persistent asthma were analysed in 369 participants. The study compares data from three surveys in 24 centres in 11 countries. The use of ICSs increased from 1.7% to 5.9% in the general population and the regular use of ICSs increased from 19% to 34% among persistent asthmatic subjects. The proportion of asthmatic subjects reporting asthma attacks in the last 12 months decreased, while the proportion that had seen a doctor in the last 12 months remained unchanged (42%). Subjects with asthma who had experienced attacks or had seen a doctor were more likely to use ICSs on a regular basis. Although ICS use has increased, only one-third of subjects with persistent asthma take ICSs on a regular basis. Less than half had seen a doctor during the last year. This indicates that underuse of ICSs and lack of regular healthcare contacts remains a problem in the management of asthma.

Abstract: Although Amazonian oils present great potential for various applications, they have not been extensively explored for commercial use. In this study, the effects of enzymatic interesterification of buriti oil in relation to its triacylglycerol composition, regiospecific distribution of fatty acids, and minority compounds were evaluated. The results indicated that the lipase used in the reaction showed higher specificity for oleic acid and the sn-1 and sn-3 positions of triacylglycerol, generating more unsaturated structured lipids. There were increases of 11% and 12.5% in unsaturated-unsaturated-unsaturated triacylglycerol types and reductions of 12.1% and 16.2% in saturated-unsaturated-unsaturated triacylglycerol types after 6 and 24 hours of reaction, respectively. At 24 h of reaction, the structured lipid formed was totally unsaturated at the three triacylglycerol positions. In addition, as the reaction conditions were mild, the carotenoids and phenolic compounds were maintained in the structured lipids. The results indicate that the enzymatic interesterification can be an alternative to produce structured lipids with new functionalities, and diversify the application of this oil from the Amazon.

Abstract: Buriti oil is an example of an Amazonian palm oil of economic importance. The local population uses this oil for the prevention and treatment of different diseases; however, there are few studies in the literature that evaluate its properties. In this study, detailed chemical and antioxidant properties of Buriti oil were determined. The predominant fatty acid was oleic acid (65.6%) and the main triacylglycerol classes were tri-unsaturated (50.0%) and di-unsaturated-mono-saturated (39.3%) triacylglycerols. The positional distribution of the classes of fatty acids on the triacylglycerol backbone indicated a saturated and unsaturated fatty acid relationship similar in the three-triacylglycerol positions. All tocopherol isomers were present, with a total content of 2364.1 mg·kg−1. α-tocopherol constitutes 48% of the total tocopherol content, followed by γ- tocopherol (45%). Total phenolic (107.0 mg gallic acid equivalent·g−1 oil) and β-carotene (781.6 mg·kg−1) were particularly high in this oil. The highest antioxidant activity against the free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) was obtained at an oil concentration of 50 mg·mL−1 (73.15%). The antioxidant activity evaluated by the Oxygen Radical Absorbance Capacity (ORAC) was 95.3 μmol Trolox equivalent·g−1 oil. These results serve to present Buriti oil as an Amazonian resource for cosmetic, food and pharmaceuticals purposes.

14. Preliminary validation of a questionnaire to measure basic psychological needs in Physical Education

Abstract: The self-determination theory is a psychological approach to motivation that focuses on causes and consequences of human behavior regulation. According several authors, this theoretical framework could provide important information about the student’s motivational process to physical education class, however, in Portugal does not exists any instrument to measure the basic psychological needs in this domain. So, the main propose of this study is the preliminary adaptation to physical education contexts of Basic Psychological Needs Exercise Scale (Portuguese version: BPNESp), and determine their initial psychometrics properties through an exploratory factor analysis. This propose was accomplished with a sample of 150 students (n=150) from de 2nd and 3rd CEB, aged from 11 to 16 years (M = 13.39, SD = 1.44) with different levels of sports practice. Results revealed a factorial structure just like the original model (12 items grouped in 3 factors, with 4 items hitch factor) and presents acceptable values of validity and reliability. Those findings allow us to conclude, that questionnaire can be used in future investigations to measure the basic psychological needs in physical education.

15. A mechanistic view of mitochondrial death decision pores

Abstract: Mitochondria increase their outer and inner membrane permeability to solutes, protons and metabolites in response to a variety of extrinsic and intrinsic signaling events. The maintenance of cellular and intraorganelle ionic homeostasis, particularly for Ca2+, can determine cell survival or death. Mitochondrial death decision is centered on two processes: inner membrane permeabilization, such as that promoted by the mitochondrial permeability transition pore, formed across inner membranes when Ca2+ reaches a critical threshold, and mitochondrial outer membrane permeabilization, in which the pro-apoptotic proteins BID, BAX, and BAK play active roles. Membrane permeabilization leads to the release of apoptogenic proteins: cytochrome c, apoptosis-inducing factor, Smac/Diablo, HtrA2/Omi, and endonuclease G. Cytochrome c initiates the proteolytic activation of caspases, which in turn cleave hundreds of proteins to produce the morphological and biochemical changes of apoptosis. Voltage-dependent anion channel, cyclophilin D, adenine nucleotide translocase, and the pro-apoptotic proteins BID, BAX, and BAK may be part of the molecular composition of membrane pores leading to mitochondrial permeabilization, but this remains a central question to be resolved. Other transporting pores and channels, including the ceramide channel, the mitochondrial apoptosis-induced channel, as well as a non-specific outer membrane rupture may also be potential release pathways for these apoptogenic factors. In this review, we discuss the mechanistic models by which reactive oxygen species and caspases, via structural and conformational changes of membrane lipids and proteins, promote conditions for inner/outer membrane permeabilization, which may be followed by either opening of pores or a rupture of the outer mitochondrial membrane.

Abstract: Psychotic symptoms in Parkinson’s disease (PD) are frequent, disabling, and an important prognostic factor. Thus, screening instruments for detecting psychosis in PD are needed. For this purpose, we applied the Parkinson’s Psychosis Questionnaire (PPQ), a short structured questionnaire, which requires no specific training, along with the Brief Psychiatric Rating Scale, expanded version (BPRS-E), for rating general psychopathology, including psychotic symptoms. We evaluated, in a cross-sectional study, a Portuguese sample of 36 early-stage PD patients (mean age of 73 years; mean duration of illness of 3.2 years). The PPQ total score correlated with the BPRS-E total score (0.359; 𝑃=0.032) and with the BPRS-E-positive symptoms score (0.469; 𝑃=0.004). The prevalence of psychosis (41.7%) was higher than expected. Sampling bias and detection of minor psychotic phenomena may have contributed to this result. These findings suggest that the PPQ should be further evaluated as a feasible assessment for psychotic symptoms in PD.

These models are methodically constructed based on medical imaging data acquired through collaboration with hospitals and subsequently verified through empirical experimentation. Studies have shown that the main factor that influences the vulnerability to bridging vein rupture in older adults is the degenerative processes of nervous tissue. The most visible structural damage was observed in the outflow cuff segment. This phenomenon can be primarily attributed to specific geometric parameters associated with this anatomical region. The presented research emphasises the importance of computational models in understanding the pathomechanics of brain structures. As a result of the analyses, it was proven that the neurodegenerative processes of the brain that occur with age are crucial in understanding the higher incidence of subdural haematomas in elderly people. Abstract: Abstract: Infectious diseases have always been a concern for human health, responsible for numerous pandemics throughout history. Even with the advancement of medicine, new infectious diseases have been discovered over the years, requiring constant effort in medical research to avoid future problems. Like the emergence of new diseases, the increase in resistance of certain bacterial strains also becomes a concern, carried out through the misuse of antibiotics, generating the adaptation of certain microorganisms. Worldwide, the resistance developed by several bacterial strains is growing exponentially, creating awareness and developing novel strategies to control their evolution a mandatory research topic. Methicillin-resistant Staphylococcus aureus (MRSA) is an example of a bacterial strain that causes serious and mortal infections. The fact is that this bacterial strain started to develop resistance against commonly used antibiotics, first to penicillin and against methicillin. Thus, the treatment against infections caused by MRSA is limited and difficult due to its capacity to develop defense mechanisms against the antibiotic's action. Given the urgency to find new alternatives, the scientific community has been developing interesting research regarding the exploitation of natural resources to discover bioactive molecules that are able to inhibit/kill MRSA. In this sense, several natural matrices, namely plants, have shown great potential against MRSA, due to the presence of phenolic compounds, molecules with high antimicrobial capacity due to their chemical structure and arrangement.

Abstract: Inadequate infection control, wound care, and oral hygiene protocols in nursing homes provide challenges to residents’ quality of life. Based on the outcomes from a focus group meeting and a literature search, this narrative review evaluates the current and potential roles of antiseptics within nursing home infection management procedures. We examine contemporary strategies and concerns within the management of meticillin-resistant Staphylococcus aureus (MRSA; including decolonization regimes), chronic wound care, and oral hygiene, and review the available data for the use of antiseptics, with a focus on povidone-iodine. Compared with chlorhexidine, polyhexanide, and silver, povidone-iodine has a broader spectrum of antimicrobial activity, with rapid and potent activity against MRSA and other microbes found in chronic wounds, including biofilms. As no reports of bacterial resistance or cross-resistance following exposure to povidone-iodine exist, it may be preferable for MRSA decolonization compared with mupirocin and chlorhexidine, which can cause resistant MRSA strains. Povidone-iodine oral products have greater efficacy against oral pathogens compared with other antiseptics such as chlorhexidine mouthwash, highlighting the clinical benefit of povidone-iodine in oral care. Additionally, povidone-iodine-based products, including mouthwash, have demonstrated rapid in vitro virucidal activity against SARS-CoV-2 and may help reduce its transmission if incorporated into nursing home coronavirus 2019 control protocols. Importantly, povidone-iodine activity is not adversely affected by organic material, such as that found in chronic wounds and the oral cavity. Povidone-iodine is a promising antiseptic agent for the management of infections in the nursing home setting, including MRSA decolonization procedures, chronic wound management, and oral care.
