The beauty production cross section for deep inelastic scattering events with at least one hard jet in the Breit frame together with a muon has been measured, for photon virtualities Q^2 > 2 GeV^2, with the ZEUS detector at HERA using integrated luminosity of 72 pb^-1. The total visible cross section is sigma_b-bbar (ep -> e jet mu X) = 40.9 +- 5.7 (stat.) +6.0 -4.4 (syst.) pb. The next-to-leading order QCD prediction lies about 2.5 standard deviations below the data. The differential cross sections are in general consistent with the NLO QCD predictions; however at low values of Q^2, Bjorken x, and muon transverse momentum, and high values of jet transverse energy and muon pseudorapidity, the prediction is about two standard deviations below the data., 18 pages, 4 figures

A resonance search has been made in the K0s p and K0s pbar invariant-mass spectrum measured with the ZEUS detector at HERA using an integrated luminosity of 121 pb-1. The search was performed in the central rapidity region of inclusive deep inelastic scattering at an ep centre-of-mass energy of 300-318 GeV for exchanged photon virtuality, Q2, above 1 GeV2. Recent results from fixed-target experiments give evidence for a narrow baryon resonance decaying to K+ n and K0s p, interpreted as a pentaquark. The results presented here support the existence of such state, with a mass of 1521.5+/-1.5(stat.)^{+2.8}_{-1.7}(syst.) MeV and a Gaussian width consistent with the experimental resolution of 2 MeV. The signal is visible at high Q2 and, for Q2>20 GeV2, contains 221+/-48 events. The probability of a similar signal anywhere in the range 1500-1560 MeV arising from fluctuations of the background is below 6x10^{-5}., 4 eps figures, 15 pages, 1 table, accepted by Physics Letters B

The human cytochrome P4503A forms show expression patterns subject to developmental influence. CYP3A7 and CYP3A4 are generally classified as the major fetal and adult liver forms, respectively. However, characterization of CYP3A4, -3A5, and -3A7 developmental expression has historically been confounded by the lack of CYP3A isoform-specific antibodies or marker enzyme activities. Therefore, the objective of this study was to characterize the developmental expression of hepatic CYP3A forms from early gestation to 18 years of age using up to 212 fetal and pediatric liver samples. Based on immunoquantitation, CYP3A5 protein expression was found to be highly variable, generally independent of age, and more frequently observed for African-American individuals. For differentiation of CYP3A4 and -3A7 levels, dehydroepiandrosterone metabolite patterns for expressed CYP3A forms were characterized and used for simultaneous quantitation of protein levels within liver microsome samples. The major metabolite formed by CYP3A4, 7beta-hydroxy-dehydroepiandrosterone, was identified based on cochromatography and mass spectra matching with the authentic standard. Kinetic analysis showed a 34-fold greater intrinsic clearance of 7beta-hydroxy-dehydroepiandrosterone by CYP3A4 versus -3A7, whereas CYP3A7 showed the highest 16alpha-hydroxy-dehydroepiandrosterone intrinsic clearance. Metabolite profiles for the expressed enzymes were fit to a multiple response model and CYP3A4 and -3A7 levels in fetal and pediatric liver microsome samples were calculated. Fetal liver microsomes showed extremely high CYP3A7 levels (311-158 pmol/mg protein) and significant expression through 6 months postnatal age. Low CYP3A4 expression was noted for fetal liver (< or =10 pmol/mg), with mean levels increasing with postnatal age.

This tutorial reviews the recent computational advances in two-dimensional (2D) correlation spectroscopy, presents the theory, and provides examples applying 2D correlation analysis. Two-dimensional correlation analysis is a method for visualizing the relationships among the variables in multivariate data and their temporal behavior by applying the complex cross-correlation function. This function measures correlations that occur at the same rate or frequency with respect to the data acquisition time. The complex cross-correlation function yields real and imaginary components that contain information about the phase behavior of the variables. The real component provides information about mutually dependent in-phase variations. Variations that occur out-of-phase (with time lags or leads) are given by the imaginary component. Two-dimensional correlation analysis is a general analysis method that can be used for the treatment of data from a variety of applications including image, distribution, environmental, and kinetic analysis.

The ZEUS inclusive differential cross-section data from HERA, for charged and neutral current processes taken with e+ and e- beams, together with differential cross-section data on inclusive jet production in e+ p scattering and dijet production in \gamma p scattering, have been used in a new NLO QCD analysis to extract the parton distribution functions of the proton. The input of jet data constrains the gluon and allows an accurate extraction of \alpha_s(M_Z) at NLO; \alpha_s(M_Z) = 0.1183 \pm 0.0028(exp.) \pm 0.0008(model) An additional uncertainty from the choice of scales is estimated as \pm 0.005. A search for lepton-flavor-violating interactions $e p \to \mu X $ and $e p\to \tau X $ has been performed with the ZEUS detector using the entire HERA I data sample, corresponding to an integrated luminosity of 130 pb^{-1}. The data were taken at center-of-mass energies, $\sqrt{s}$, of 300 and 318 GeV. No evidence of lepton-flavor violation was found, and constraints were derived on leptoquarks (LQs) that could mediate such interactions. For LQ masses below $\sqrt{s}$, limits were set on $\lambda_{eq_1} \sqrt{\beta_{\ell q}}$, where $\lambda_{eq_1}$ is the coupling of the LQ to an electron and a first-generation quark $q_1$, and $\beta_{\ell q}$ is the branching ratio of the LQ to the final-state lepton $\ell$ ($\mu$ or $\tau$) and a quark $q$. For LQ masses much larger than $\sqrt{s}$, limits were set on the four-fermion interaction term $\lambda_{e q_\alpha} \lambda_{\ell q_\beta} / M_{\mathrm{LQ}}^2$ for LQs that couple to an electron and a quark $q_\alpha$ and to a lepton $\ell$ and a quark $q_\beta$, where $\alpha$ and $\beta$ are quark generation indices. Some of the limits are also applicable to lepton-flavor-violating processes mediated by squarks in $R$-Parity-violating supersymmetric models. Multijet production rates in neutral current deep inelastic scattering have been measured in the range of exchanged boson virtualities 10 < Q2 < 5000 GeV2. The data were taken at the ep collider HERA with centre-of-mass energy sqrt(s) = 318 GeV using the ZEUS detector and correspond to an integrated luminosity of 82.2 pb-1. Jets were identified in the Breit frame using the k_T cluster algorithm in the longitudinally invariant inclusive mode. Measurements of differential dijet and trijet cross sections are presented as functions of jet transverse energy E_{T,B}{jet}, pseudorapidity eta_{LAB}{jet} and Q2 with E_{T,B}{jet} > 5 GeV and -1 < eta_{LAB}{jet} < 2.5. Next-to-leading-order QCD calculations describe the data well. The value of the strong coupling constant alpha_s(M_Z), determined from the ratio of the trijet to dijet cross sections, is alpha_s(M_Z) = 0.1179 pm 0.0013(stat.) {+0.0028}_{-0.0046}(exp.) {+0.0064}_{-0.0046}(th.), 22 pages, 5 figures

The production of D^*+, D^0, D^+, D_s^+ and Lambda_c^+ charm hadrons and their antiparticles in ep scattering at HERA was measured with the ZEUS detector using an integrated luminosity of 79 pb^-1. The measurement has been performed in the photoproduction regime with the exchanged-photon virtuality Q^2 < 1 GeV^2 and for photon-proton centre-of-mass energies in the range 130 < W < 300 GeV. The charm hadrons were reconstructed in the range of transverse momentum p_T(D, Lambda_c) > 3.8 GeV and pseudorapidity |eta(D, Lambda_c)| < 1.6. The production cross sections were used to determine the ratio of neutral and charged D-meson production rates, R_u/d, the strangeness-suppression factor, gamma_s, and the fraction of charged D mesons produced in a vector state, P_v^d. The measured R_u/d and gamma_s values agree with those obtained in deep inelastic scattering and in e^+e^- annihilations. The measured P_v^d value is smaller than, but consistent with, the previous measurements. The fractions of c quarks hadronising as a particular charm hadron, f(c -> D, Lambda_c), were derived in the given kinematic range. The measured open-charm fragmentation fractions are consistent with previous results, although the measured f(c -> D^*+) is smaller and f(c -> Lambda_c^+) is larger than those obtained in e^+e^- annihilations. The applicability of two-dimensional (2D) correlation spectroscopy to kinetic data is demonstrated. The photodecomposition of Mo(CO)6 was used as a model kinetic system. The effects of the steady-state and time-average methods for static spectrum removal were evaluated. The time-average method provided 2D-correlation spectra that were more easily interpreted. The influence of noise was evaluated with the synthetic data. The methods of static spectral removal were affected differently by the presence of noise and the time-average method was detrimentally affected by noise. A low-pass Fourier filtering method was devised to remove noise in the 2D-correlation spectra. Kinetic simulations were demonstrated as useful tools for interpretation of noisy data.

An automated method for monitoring a polyethylene recycling process has been developed. The products of catalyzed polyethylene cracking are alkanes, monounsaturated alkenes (monoene) and dienes, which were analyzed by gas chromatog-raphy-mass spectrometry. Three expert systems, a knowledge based expert system, a univariate rule building expert system and a fuzzy multivariate expert system, were developed and evaluated for their ability to correctly classify products of polyethylene cracking by their degree of unsaturation. Two test sets were used in the evaluation. One contained spectra from a mass spectral library. The second was a collected set of hydrocarbon standards. On both test sets, the rule building expert systems performed better than the knowledge based expert system. The chemometric detectors were effective for mathematically resolving chromatograms by chemical functionality.

The flowering pot plant, clerodendrum (Clerodendrum speciosum), exhibits typical symptoms of chilling injury following transient exposure (48 h or more) to temperatures of 2°C. Symptoms include a decline in a leaf-water content and an increase in electrolyte leakage. Plants grown for 3 days or longer at day temperatures of 17–22°C displayed a much slower growth rate and were hardened to chilling for 48 h at 2°C, relative to control plants grown at 32°C. Similar results were obtained in plants treated with triazoles through a single foliar spray application at least 10 days prior to chilling. Both treatments inhibited the chilling-induced degradation of membrane proteins and phospholipids. It is proposed that at least some elements of the mechanisms by which both of these hardening techniques act, are analogous.

We perform a set of experiments on photoheating in a water droplet containing gold nanoparticles (NPs). Using photocalorimetric methods, we determine efficiency of light-to-heat conversion (eta) which turns out to be remarkably close to 1, (0.97eta1.03). Detailed studies reveal a complex character of heat transfer in an optically stimulated droplet. The main mechanism of equilibration is due to convectional flow. Theoretical modeling is performed to describe thermal effects at both nano- and millimeter scales. Theory shows that the collective photoheating is the main mechanism. For a large concentration of NPs and small laser intensity, an averaged temperature increase (at the millimeter scale) is significant (approximately 7 degrees C), whereas on the nanometer scale the temperature increase at the surface of a single NP is small (approximately 0.02 degrees C). In the opposite regime, that is, a small NP concentration and intense laser irradiation, we find an opposite picture: a temperature increase at the millimeter scale is small (0.1 degrees C) but a local, nanoscale temperature has strong local spikes at the surfaces of NPs (approximately 3 degrees C). These studies are crucial for the understanding of photothermal effects in NPs and for their potential and current applications in nano- and biotechnologies.

The thermo-optical properties of gold nanoparticles suspended in liquid water droplets have been explored. A water droplet containing suspended gold nanoparticles was irradiated with a laser beam and the efficiency of light-to-heat conversion (η) was determined. Heat generated by the optically stimulated nanoparticles was found to dissipate through several mechanisms. Convection currents were observed visually inside the droplet by the addition of silica beads (˜3-10 micrometers). This was determined to be the dominant mechanism of stirring within the droplet.

The number of solvents capable of dissolving myelin and proteolipid protein (PLP) and of being used as a mobile phase for the separation of myelin proteins by reversed-phase high-performance liquid Chromatography (RP-HPLC) is very limited. In a thorough study, we found that aqueous tetrahydrofuran (THF) fulfilled such a requirement. The maximal amount of protein extracted corresponded to a THF/water ratio of 4:1 v/v and a polarity index of 5.16. This mixture dissolved a purified PLP preparation completely, 60% of proteins from fresh myelin, and 20% of white matter total proteins. Sodium dodecyl sulphate-poly-acrylamide gel electrophoresis (SDS-PAGE) of those extracts, followed by densitometric analysis, showed that the amount and type of proteins dissolved depended on the polarity (i.e., the content of water) of the solvent mixture used. This selective effect was greater for basic protein in myelin preparations. Crude extracts highly enriched in basic protein can be prepared. In addition, the solvent system THF/ water proved to be very useful as a mobile phase in RP-HPLC for separating myelin proteins. Using a C3 column and a linear gradient from 30% to 100% THF in water, both containing 0.1% trifluoroacetic acid (TFA), we separated completely the three main protein fractions of central nervous system (CNS) myelin in a short period of time. The high solubility power of THF/water mixtures prolonged greatly the life of the column.

Sub-project 6 of the European Integrated Project APROSYS, implemented within the 6th Framework Programme, has developed an integrated safety system for side impact protection. For this, two different innovative technologies have been applied for the first time as a technology showcase in a car: shape memory alloy based actuators and a pre-crash sensing subsystem using radar and stereo-vision observing the side of a vehicle. Several technical challenges had to be met during development work. The sensing subsystem detects an imminent collision and decides about the probability of a collision. It activates the actuator before the crash occurs in order to reduce intrusion into the passenger compartment. This article describes the sensing subsystem, the actuator subsystem and finally the testing programme that was executed to evaluate the complete system.

The EU funded integrated project APROSYS aims to increase the safety of road users. Among others, subproject 6 (SP6) "intelligent safety systems" develops, realises and verifies novel side pre-crash systems. Previously we presented an application analysis, the resulting system definition and the selection of sensors of a suitable side sensing system. The sensing system is based on a stereo video rig and a short-range radar subsystem. Now, a prototype system has been built up and first data were taken. These were analysed, a data fusion module was implemented and the data analysis algorithms were optimised with respect to the specific requirements of the side pre-crash application.

As a first result, we adapted the fields of view of the sensors to the specific needs of side pre-crash sensing. As a second result of simulation we identified false alarm discrimination as a major challenge of side sensing.

The relationship of rotatable bond count (N(rot)) and polar surface area (PSA) with oral bioavailability in rats was examined for 434 Pharmacia compounds and compared with an earlier report from Veber et al. (J. Med. Chem. 2002, 45, 2615). N(rot) and PSA were calculated with QikProp or Cerius2. The resulting correlations depended on the calculation method and the therapeutic class within the data superset. These results underscore that such generalizations must be used with caution.

Cross sections for e(+)p neutral current deep inelastic scattering have been measured at a center-of-mass energy of roots=318 GeV with the ZEUS detector at HERA using an integrated luminosity of 63.2 pb(-1). The double-differential cross section, d(2)sigma/dxdQ(2), is presented for 200 GeV2 lt Q(2) lt 30 000 GeV2 and for 0.005 lt x lt 0.65. The single- differential cross sections dsigma/dQ(2), dsigma/dx and dsigma/dy are presented for Q(2) gt 200 GeV2. The effect of Z- boson-exchange is seen in dsigma/dx measured for Q(2) gt 10 000 GeV2. The data presented here were combined with ZEUS e(+)p neutral current data taken at roots=300 GeV and the structure function F-2(em) was extracted. All results agree well with the predictions of the Standard Model.

The photoproduction of bottom quarks in events with two jets and a muon has been measured with the ZEUS detector at HERA using an integrated luminosity of 110 ${\mathrm{pb}}^{\mathrm{\ensuremath{-}}1}$. The fraction of jets containing b quarks was extracted from the transverse momentum distribution of the muon relative to the closest jet. Differential cross sections for bottom production as a function of the transverse momentum and pseudorapidity of the muon, of the associated jet and of ${x}_{\ensuremath{\gamma}}^{\mathrm{jets}},$ the fraction of the photon's momentum participating in the hard process, are compared with MC models and QCD predictions made at next-to-leading order. The latter give a good description of the data.

A search for events containing isolated tau leptons and large missing transverse momentum, not originating from the tau decay, has been performed with the ZEUS detector at the electron-proton collider HERA, using 130 pb^-1 of integrated luminosity. A search was made for isolated tracks coming from hadronic tau decays. Observables based on the internal jet structure were exploited to discriminate between tau decays and quark- or gluon-induced jets. Three tau candidates were found, while 0.40 +0.12 -0.13 were expected from Standard Model processes, such as charged current deep inelastic scattering and single W-boson production. To search for heavy-particle decays, a more restrictive selection was applied to isolate tau leptons produced together with a hadronic final state with high transverse momentum. The subjet multiplicity has been measured in neutral current e+p interactions at Q**2 > 125 GeV**2 with the ZEUS detector at HERA using an integrated luminosity of 38.6 pb-1. Jets were identified in the laboratory frame using the longitudinally invariant K_T cluster algorithm. The number of jet-like substructures within jets, known as the subjet multiplicity, is defined as the number of clusters resolved in a jet by reapplying the jet algorithm at a smaller resolution scale y_cut. Measurements of the mean subjet multiplicity, < n_sbj >, for jets with transverse energies E_T,jet >15 GeV are presented. Next-to-leading-order perturbative QCD calculations describe the measurements well. The value of alpha_s(M_Z), determined from < n_sbj > at y_cut=10**-2 for jets with 25 < E_T,jet < 71 GeV, is alpha_s (M_Z) = 0.1187 +/- 0.0017 (stat.) +0.0024 / -0.0009 (syst.) +0.0093 / -0.0076 (th.)., Comment: 27 pages, 4 figures, 3 tables; submitted to Phys. A study of the $\overline{\ensuremath{\nu}}$-jet mass spectrum in ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}\overline{\ensuremath{\nu}}X$ events at a center-of-mass energy 300 GeV has been performed with the ZEUS detector at the HERA collider at DESY using an integrated luminosity of 47.7 ${\mathrm{pb}}^{\ensuremath{-}1}.$ The mass spectrum is in good agreement with that expected from standard model processes over the $\overline{\ensuremath{\nu}}$-jet mass range studied. No significant excess attributable to the decay of a narrow resonance is observed. By using both ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}{e}^{+}X$ and ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}\overline{\ensuremath{\nu}}X$ data, mass-dependent limits are set on the s-channel production of scalar and vector resonant states. Couplings to first-generation quarks are considered and limits are presented as a function of the ${e}^{+}q$ and $\overline{\ensuremath{\nu}}q$ branching ratios. These limits are used to constrain the production of leptoquarks and R-parity violating squarks.

Three-jet production in the reaction ep --> eXp has been studied with the ZEUS detector at HERA using an integrated luminosity of 42.74 pb^-1. The data were measured in the kinematic region 5 < Q^2 < 100 GeV^2, 200 < W < 250 GeV and 23 < M_X < 40 GeV. The diffractive signal was selected by requiring a large rapidity gap in the outgoing proton direction. Jets were reconstructed in the centre-of-mass system of X using the exclusive k_T-algorithm. A sample of three-jet events in diffraction has been identified. Differential cross sections were measured as a function of the jet pseudorapidity and jet transverse momentum with respect to the virtual photon-Pomeron axis. The jets going in the Pomeron direction are broader than those going in the virtual-photon direction. This is consistent with models predicting that gluons are predominantly produced in the Pomeron direction and quarks in the virtual-photon direction., 14 pages, 5 figures and 7 tables. The first measurement of inclusive Ds+- photoproduction at HERA has been performed with the ZEUS detector for photon-proton centre-of-mass energies 130 < W < 280 GeV. The measured cross section for 3 < pt(Ds) < 12 GeV and |eta(Ds)|< 1.5 is sigma(ep -> Ds X) = 3.79 +- 0.59 (stat.) +0.26-0.46 (syst.) +- 0.94 (br.) nb, where the last error arises from the uncertainty in the Ds decay branching ratio. The measurements are compared with inclusive D*+- photoproduction cross sections in the same kinematic region and with QCD calculations. The Ds cross sections lie above a fixed-order next-to-leading order calculation and agree better with a tree-level O(alpha,alpha_s^3) calculation that was tuned to describe the ZEUS D* cross sections. The ratio of Ds+- to D*+- cross sections is 0.41 +- 0.07 (stat.) +0.03-0.05 (syst.) +- 0.10 (br.). From this ratio, the strangeness-suppression factor in charm photoproduction, within the LUND string fragmentation model, has been calculated to be gamma_s = 0.27 +- 0.05 +- 0.07 (br.). The immunofluorescence technique is one of the most useful methods for localizing antigens in several tissues, including the central nervous system. For immunohistochemical procedures, especially immunofluorescence methods, formaldehyde is commonly used as a fixative agent. But for some protocols, mainly in neurobiology, glutaraldehyde is necessary to recognize a number of small molecules (haptens) whose antisera have been raised using glutaraldehyde as the cross-linking agent. This is a severe limitation because glutaraldehyde gives rise to a strong autofluorescence on tissue that precludes the observation of specific immunofluorescence staining. In this paper we present a new method that allows the use of immunofluorescence techniques on glutaraldehyde-fixed tissues. The new method consists of a treatment of tissue sections with the Schiffs reagent (leucobasic fuchsin) followed by a reduction of the Schiff-dye with sodium borohydride. This reduced dye produces a quenching of glutaraldehyde-induced fluorescence on the tissue. The goal of the new method is to make possible the use of a great number of available glutaraldehyde-raised antisera for immunofluorescence techniques, a useful tool in both basic and clinical research.

The mercury-silver (Hg-Ag) argentaffin technique, known to stain specifically proteins in the lateral components of triads/diads in striated muscle cells, was applied to the central nervous system of adult rats. Following fixation in glutaraldehyde, axons in white and gray matter were selectively stained, but not perikarya or their proximal axon and dendrites. Neural tissues were postfixed 24 hr in 5% (w/v) mercuric acetate in 2% (v/v) acetic acid in distilled water, stained for 12-24 hr in darkness at 37-43 C with ammoniacal silver nitrate solution, freshly prepared by adding concentrated ammonia to 60% (w/v) silver nitrate solution until a small amount of silver oxide precipitate remained undissolved. Samples were then washed with freshly prepared 5% (w/v) sodium sulfite and distilled water. All steps were carried out using dark-colored glass flasks. Samples were dehydrated with ethanol and embedded in Paraplast or Poly Bed. Electron microscopy showed the silver-reducing protein inside the axons. Methylation abolished Hg-Ag axonal reactivity indicating that carboxyl groups were necessary for silver staining. Proteins with solubility properties characteristic of neurofilament proteins were involved in Hg-Ag staining. In the cerebellum the plexus of parallel fibers in the molecular layer were not stained, while basket cell axonal processes reacted intensely. The method appears to distinguish neuronal protein variants related to cytotypic differences in cytoskeletal neurofilaments.

Spermatocytes at meiotic metaphase I and anaphase I have a characteristic centromeric filament in a variety of vertebrate organisms. This centromeric filament was first demonstrated on mouse spermatocytes and its presence is now extended to spermatocytes from the human, rat, golden hamster, bull, and chicken. The visualization of this filament was possible through the use of a novel silver-staining technique, which allows a high contrast between the filament and the centromeric chromatin. In the species cited, the centromeric filament shares an intense staining, a short (0.2–0.6 μm) length, a curved and branched shape, and location inside the centromeric chromatin of seemingly every homologue of the complement. The similarity of staining reactivity and the observation of transitional structures during first meiotic prophase strongly suggest that the centromeric filament is a remnant of a lateral element of the synaptonemal complex, which stays specifically at both centromeric regions of each bivalent. This filament is not found at the second meiotic division or at the centromeres of mitotic chromosomes. It is assumed that this centromeric filament joins the two sister chromatids of each homologue at the centromere and thus ensures the proper coorientation of sister kinetochores at metaphase I. Further testable assumptions on the functions of this filament are presented.Key words: synaptonemal complex, meiosis, meiotic centromeres.

We have developed a new, simple, and reproducible cytochemical method to specifically stain DNA at the electron microscopic level: the NAMA-Ur. It is based on the extraction of RNA and phosphate groups from phosphoproteins by a weak alkali hydrolysis (NA) which does not affect DNA, followed by blockage of the amino and carboxyl groups by methylation and acetylation (MA). Finally, sections are stained by uranyl (Ur), which can bind only to DNA. The efficiency of the pre-treatment (NA and MA) was demonstrated by X-ray microanalysis at the transmission electron microscopic level. The NAMA-Ur method has been successfully performed en bloc and on Lowicryl sections in mammalian and plant cells. A specific contrast is observed in the DNA-containing structures after this method, whose sensitivity allows visualization of electron-dense chromatin fibers of 10-12 nm composed of 3-nm DNA fibrils. This staining method has been combined with anti-DNA antibodies, providing complementary information to detect DNA in situ. SUMMARY A new technique for the visualization of DNA-containing structures in electron microscopy is described. Samples of glutaraldehyde-fixed bone marrow from rats were subjected to alkaline hydrolysis to remove RNA and the phosphate of phospho-proteins, followed by a combined blockage of protein carboxyl and amino groups through methylation-acetylation. After uranyl acetate staining of epoxy-embedded ultrathin sections, chromatin from all cell types showed a highly selective and intense electron opacity. Staining methods for DNA were also positive in semithin sections. This simple procedure could be very useful in ultrastructural cytochemistry of DNA and chromatin.

43. Selective extraction, solubilization, and reversed-phase high-performance liquid chromatography separation of the main proteins from myelin using tetrahydrofuran/water mixtures

The number of solvents capable of dissolving myelin and proteolipid protein (PLP) and of being used as a mobile phase for the separation of myelin proteins by reversed-phase high-performance liquid chromatography (RP-HPLC) is very limited. In a thorough study, we found that aqueous tetrahydrofuran (THF) fulfilled such a requirement. The maximal amount of protein extracted corresponded to a THF/water ratio of 4:1 v/v and a polarity index of 5.16. This mixture dissolved a purified PLP preparation completely, 60% of proteins from fresh myelin, and 20% of white matter total proteins. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of those extracts, followed by densitometric analysis, showed that the amount and type of proteins dissolved depended on the polarity (i.e., the content of water) of the solvent mixture used. This selective effect was greater for basic protein in myelin preparations. Crude extracts highly enriched in basic protein can be prepared. In addition, the solvent system THF/water proved to be very useful as a mobile phase in RP-HPLC for separating myelin proteins. Using a C3 column and a linear gradient from 30% to 100% THF in water, both containing 0.1% trifluoroacetic acid (TFA), we separated completely the three main protein fractions of central nervous system (CNS) myelin in a short period of time. The high solubility power of THF/water mixtures prolonged greatly the life of the column.

This study reports the persistence of axis-like structures in the centromeric region of both homologues during the metaphase-I and anaphase-I stages of meiotic division of mouse spermatocytes. A novel type of silver ‘argentaffin’ technique (NH4−Ag) is employed. This technique includes the treatment of glutaraldehyde-fixed tissues with dilute ammonium hydroxide followed by a reduction of aldehyde groups with sodium borohydride. Staining is accomplished with ammoniacal silver nitrate in darkness followed by sulfite washing. The lateral elements of synaptonemal complexes and the single chromosomal axes of diplotene spermatocytes show a prominent reactivity with this technique. The pattern of very small grains over condensed chromatin is uniform and gives only a light opacity to the electron beam. The presence of an axis-like structure is seen in every centromeric end of meiotic chromosomes at metaphase I and anaphase I. The chromatin (heterochromatin) that surrounds the centromeric filament and some material distributed in irregular linear arrays along some of the homologues also showed a higher electron opacity than the bulk of deoxyribonucleoprotein. While the former is related to C+ heterochromatin, the latter could represent dispersed material of diplotene axes. It is suggested that the disposal of axial material is differentially delayed at the centromeric regions. The present evidence supports the hypothesis that axial fragments or lateral-element segments persisting at these regions contribute to the cohesiveness of centromeres of sister chromatids during normal disjunction.

Cytochemical electron microscope methods have been useful for the localization of individual molecules or groups in situ. Techniques selective for DNA have been very thoroughly investigated, and have lead to the different Schiff-like techniques. The Feulgen-like method, with the osmium-ammine complex according to Cogliati and Gautier (1973) and developed by Moyone (1980) has been the only method to reveal chromatin fine structure, because of the specificity and high resolution it offers. Its use has enabled visualization of nucleosomal and non-nucleosomal (12 and 3nm) chromatin fibres in both animal and plant cells (Derenzini et al. 1982; Risueno et al., 1982). The main problem of this technique is the synthesis of the stain, as it is not commercially available. Recently, the localization of the DNA in the nucleus has been address by immunocytochemical procedures (Scheer et al., 1987). The use of monoclonal antibodies has made available a highly sensitive procedure to detect double and single-stranded DNA. However, this approach requires antibody and the development of tissue processing to preserve antigenicity.

‘Argentaffinity’, as opposed to ‘argyrophilia’, denotes the reduction of silver salts to metallic silver by tissue or cell component per se, without addition of any extraneous reducing substance and in the dark. Aldehydes, which are known to reduce diammine silver, are not needed for the argentaffin reaction to take place; thus, staining after ethanol or Carnoy fixation was entirely attributable to an intrinsic property of the cell components (Tandler, 1954). A novel type of ‘argentaffin’ histochemical staining reaction based on the reduction of diammine silver nitrate by mercuric acetate postfixed tissues has been recently applied to striated muscle cells (Tandler and Pellegrino de Iraldi, 1989; Tandler et al., 1989).

A selective silver staining of proteins within the lateral components of triads and diads in fixed striated muscle cells and in isolated sarcoplasmic reticulum vesicles using a mercuric acetate postflxation has been demonstrated by us.In the present work we modify the technique in order to study the mechanism of the reaction . First, the mercuric postfixation step was omitted and replaced by a treatment with diluted ammonia, a procedure thought to wash out the excess of the reducing aldehyde fixative. Second, a treatment with mercuric acetate was employed on the ammonia treated muscles. The rat gastrocnemius muscle was fixed by immersion (12-24 h) in 3% glutaraldehyde, washed briefly with water and placed (12-24 h) into an ammonia:distilled water (1:4 v/v) solution. After washing with water, 1% acetic acid and water, muscles were treated overnight with 50% (v/v) ammoniacal silver nitrate at 37°C in darkness, washed with three changes of 5% (w/v) sodium sulphite and water, embedded in Poly B, sectioned and examined unstained with a Siemens Elmiskop I electron microscope.

This paper deals with the developing of a new simple and repetitive technique to preferentially stain DNA-containing structures at the E.M. level that can be used “en bloc” and on sections. DNA staining techniques are scarce, being the osmium ammine one the only cytochemical method known until now that specifically stains DNA but not the proteins that are complexed with it, having been used to study chromatin ultrastructure. The osmium ammine synthesis is a complex process and, although an improvement has been lately reported, the stain is not comercially available.Glutaraldehyde-fixedAllium cepa L. root meristematic cells,Capsicum annuum L. & Scilla peruviana L. pollen grains were treated with 0.5N NaOH in 4% formaldehyde overnight, rinsed in water and 1% acetic acid, methanol dehydrated, and inmersed in methanol-acetic anhydride (5:1) at 25ºC overnight. Then, the samples were Epon embedded. To perform the technique on sections, some samples were glutaraldehyde or formaldehyde fixed, methanol dehydrated, Lowicryl embedded at -20ºC and ultrathin sections were treated in the same way as in the “en bloc” procedure.
