The explosive hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) is a contaminant at many military sites. RDX bioremediation as a clean-up approach has been gaining popularity because of cost benefits compared to other methods. RDX biodegradation has primarily been linked to six functional genes (diaA, nfsI, pnrB, xenA, xenB, xplA). However, current methods for gene quantification have the risk of false negative results because of low theoretical primer coverage. To address this, the current study designed new primer sets using the EcoFunPrimer tool based on sequences collected by the Functional Gene Pipeline and Repository and these were verified based on residues and motifs. The primers were also designed to be compatible with the SmartChip Real-Time PCR system, a massively parallel singleplex PCR platform (high throughput qPCR), that enables quantitative gene analysis using 5,184 simultaneous reactions on a single chip with low volumes of reagents. This allows multiple genes and/or multiple primer sets for a single gene to be used with multiple samples. Following primer design, the six genes were quantified in RDX-contaminated groundwater (before and after biostimulation), RDX-contaminated sediment, and uncontaminated samples. The final 49 newly designed primer sets improved upon the theoretical coverage of published primer sets, and this corresponded to more detections in the environmental samples. All genes, except diaA, were detected in the environmental samples, with xenA and xenB being the most predominant. In the sediment samples, nfsI was the only gene detected. The new approach provides a more comprehensive tool for understanding RDX biodegradation potential at contaminated sites.

The objectives of this experiment were to determine (1) the effect of a Ralgro implant administered at 30 to 90 d of age on suckling-phase growth rate and weaning weight and (2) the effect of a Revalor-G implant at weaning on postweaning performance. A total of 194 suckling steer calves weighing 111 ± 25.6 kg at branding from 3 locations were used. Within location, steer calves were stratified by cow age and then randomly assigned to 2 experimental treatments: implanted with Ralgro (IMPL; n = 97) or no implant (NIMP; n = 97). At weaning, IMPL steers were reimplanted with Revalor-G. The steers were weighed at the time of branding, at weaning, after the preconditioning period, and after the stocker phase. Implanted steers gained 9 kg more (P 0.5) in the preconditioning period; however, a 35.7% increase (P = 0.01) in IMPL ADG was recorded in the other 2 locations. The highly anisotropic and qualitatively different nature of the normal state in- and out-of-plane charge dynamics in high- T c cuprates cannot be accommodated within the conventional Boltzmann transport theory. The variation of in-plane and out-of-plane resistivities with temperature and hole content are anomalous and cannot be explained by Fermi-liquid theory. In this study, we have proposed a simple phenomenological model for the dc resistivity of cuprates by incorporating two firmly established generic features of all hole doped cuprate superconductors—(i) the pseudogap in the quasiparticle energy spectrum and (ii) the T -linear resistivity at high temperatures. This T -linear behavior over an extended temperature range can be attributed to a quantum criticality, affecting the electronic phase diagram of cuprates. Experimental in-plane and out-of-plane resistivities ( ρ p ( T ) and ρ c ( T ), respectively) of double-layer Y(Ca)123 have been analyzed using the proposed model. This phenomenological model describes the temperature and the hole content dependent resistivity over a wide range of temperature and hole content, p . The characteristic PG energy scale, e g ( p ), extracted from the analysis of the resistivity data, agrees quite well with those found in variety of other experiments. Various other extracted parameters from the analysis of ρ p ( T ) and ρ c ( T ) data showed systematic trends with changing hole concentration. We have discussed important features found from the analysis in detail in this paper.

The out-of-plane ( c -axis) resistivity, ρ c ( T ), of high- T c cuprates have been modeled in this study. The non-Fermi liquid like temperature dependence of ρ c ( T ) has been described by considering (i) the full impact of the pseudogap (PG) in the electronic density of states (EDOS) and (ii) the presence of a quantum critical point (QCP) beneath the superconducting dome at slightly overdoped region. This simple phenomenological model describes the experimental ρ c ( T ) data over a wide range of hole content (from the underdoped to slightly overdoped regions) remarkably well. The PG energy scale, e g (dominated by the anti-nodal parts of the Brillouin zone) extracted from the analysis of ρ c ( T ) data was found to decrease almost linearly with increasing hole concentration, p , in the CuO 2 planes. We have also discussed about the possible origin of more conventional behavior of ρ c ( T ) observed in the deeply overdoped side of the T – p phase diagram in this paper.

Two experiments were designed to investigate the effects of feeding monensin and/or slow release urea with a fibrolytic feed enzyme (Optimase; Alltech, Inc., Nicholasville, KY) on performance, milk production, calf growth performance, and blood metabolites in beef cows. Spring-calving cows and heifers were used in a completely randomized design in Exp. 1 (N = 84; 534 ± 68 kg initial BW) and Exp. 2 (N = 107; 508 ± 72 kg initial BW). Exp. 1 supplements were formulated to meet cow protein requirements and fed daily and included 1) cottonseed meal with no monensin (control); or 2) monensin added to control to supply 200 mg per head per d (MON). In Exp. 2, experimental supplements included 1) cottonseed meal/wheat middlings (CS) fed at a rate to provide adequate DIP and CP according to , 2) the CS plus soybean hulls and 61 g per cow per d Optimase (OPT), 3) the CS plus monensin to supply 200 mg per cow per d (MON2), and 4) OPT plus MON2 (Combo). Cows were fed in last trimester through early lactation in Exp. 1 and during 2nd trimester in Exp. 2. Data were analyzed using the Mixed procedure in SAS with animal as the experimental unit. In Exp. 1, treatment did not affect cow BW or BCS change (P0.19). Calf birth BW was not affected by dam treatment (P = 0.24); however, calves from dams consuming MON weighed more (P0.04) at d 45 and at trial end. Calves also had greater (P = 0.04) ADG from birth to trial end. Milk production did not significantly differ among treatments (P0.41). In Exp. 2, mean cow BW and BCS were similar (P0.35) among treatments on d 90. However, from d 0 to 54, cows assigned to the OPT supplement gained less BCS (P = 0.02) compared with cows assigned to the CS supplement. Cumulative BCS gain was greater (P0.01) for CS-fed cows than for cows fed the OPT and MON2 supplements, although it was not significantly different for cows fed the Combo supplement. These studies indicate that the influence of monensin on cow BW and BCS change is inconsistent. The potential for monensin supplementation to positively impact calf performance during early lactation seems to be clearer. Replacing a portion of oilseed N in the supplement with Optimase may marginally reduce cow performance. Further research is needed to determine both the effects of monensin and the implications of combining monensin with Optimase on forage intake and cow performance at various stages of production.

Sulfotransferase 1A1 (SULT1A1) (thermostable phenol sulfotransferase, TS PST1, P-PST) is important in the metabolism of thyroid hormones. SULT1A1 isolated from human platelets displays wide individual variations not only in the levels of activity, but also in thermal stability. The activity of the allelic variant or allozyme SULT1A1*1, which possesses an arginine at amino acid position 213 (Arg213) has been shown to be more thermostable than the activity of the SULT1A1*2 allozyme which possesses a histidine at this position (His213) when using p-nitrophenol as the substrate. We isolated a SULT1A1*1 cDNA from a human liver cDNA library and expressed both SULT1A1*1 and SULT1A1*2 in eukaryotic cells. The allozymes were assayed using iodothyronines as the substrates and their biochemical properties were compared. SULT1A1*1 activity was more thermostable and more sensitive to NaCl than was SULT1A1*2 activity when assayed with 3,5,3'-triiodothyronine (T(3)). Sensitivities to 2,6-dichloro-4-nitrophenol (DCNP) and apparent K(m) values for SULT1A1*1 and for SULT1A1*2 with iodothyronines were similar. Based on K(m) values, the preferences of these SULT1A1 allozymes for iodothyronine substrates were the same (3,3'-diiodothyronine (3,3'-T(2))>3', 5',3-triiodothyronine (rT(3))>T(3)>thyroxine (T(4))>>3,5-diiodothyronine (3,5-T(2))). SULT1A1*1 activity was significantly higher than the SULT1A1*2 activity with T(3) as the substrate. Potential differences in thyroid hormone sulfation between individuals with predominant SULT1A1*1 versus SULT1A1*2 allozymes are most likely due to differences in catalytic activity rather than substrate specificity.

We have compared changes in haemodynamic variables before and during transurethral resection of the prostate in 22 patients under general or spinal anaesthesia. In the general anaesthetic group there was a significant decrease in cardiac output (mean 32% (SEM 5%)) and mean arterial pressure (14% (3%)) after induction of anaesthesia and a significant decrease (27% (3%)) in heart rate before the start of resection. Otherwise, variables remained close to baseline values. In the subarachnoid block group, mean arterial pressure decreased after induction (16% (2%)), but overall variables remained unchanged. We conclude that with both these anaesthetic techniques the greatest changes in haemodynamic variables occurred shortly after induction, and that these changes were greater during general than spinal anaesthesia. The resection period was not associated with significant haemodynamic changes. (Br. J. Anaesth. 1994; 72: 267–271)

More detailed sequence standards that keep up with revolutionary sequencing technologies will aid the research community in evaluating data.

The paper presents a review of the need for the planning and monitoring of construction work, a description of the characteristics of site construction work, particularly the degree of task repetition required for project execution, a review of two planning models comprising the critical path model, which is based on the concept that a project consists of a series of discrete activities, and the linear scheduling-flow line model, which is based on the concept that a project consists of a series of repetitive activities. The practices of contractors when confronted with a construction scheduling problem and the use of the techniques derived from the models are examined. Six successful case studies are included, including two renovation projects and four commercial office buildings, two based on the critical path method, two based on the flow line method, and two that use both models in appropriate sections. The paper concludes with the general observations that: Few contractors use planning techniques to their full potential; critical path methods are best suited to nonrepetitive projects; flow line is best suited to repetitive projects; and as most contracts have dual characteristics, no single system can meet all a contractor’s requirements for planning and monitoring construction work.

Virulence-factor activity relationship (VFAR) is a concept that was developed as a way to relate the architectural and biochemical components of a microorganism to its potential to cause human disease. Development of these relationships requires specialised bioinformatics databases that do not exist at present. A pilot-scale VFAR database was designed for three different waterborne organisms: Escherichia coli, Norovirus and Cryptosporidium, to evaluate VFAR relationships. For the web-based database, each organism has separate pages containing virulence genes, occurrence genes, primer sets and probes, taxonomy, outbreaks, and serotype/species/genogroup/genotype. As the database continues to grow, it will be possible to relate the occurrence and prevalence of certain genes in various microorganisms to outbreak data and, subsequently, to establish the utility of using a combination of specific genes as markers of virulence and in establishing virulence-factor activity relationships (VFARs). The database and the VFARs established will be of use to the regulatory community as a way to assist with prioritising those organisms, which need to be regulated.

During a 16-month period, 10 goats with listeriosis were identified in 2 herds that shared 3 bucks, including 1 that died of listeriosis. Using DNA fingerprinting, we determined that a single genetically unique Listeria monocytogenes strain had infected all goats from which isolates were available. All isolates were unable to metabolize rhamnose (rhamnose-negative), whereas as a species, L monocytogenes is considered to have a rhamnose-positive phenotype. Therefore, these isolates would have been characterized as a species other than L monocytogenes if any of a variety of commercial bacterial identification kits had been used for speciation. Silage was not fed to either herd, and L monocytogenes was not isolated from vaginal or rectal swab specimens obtained from healthy goats or from samples of feed. Because the 3 bucks were the only common elements between the 2 herds, our results suggest a venereal route of transmission for listeriosis.

Exposure of swine to Trichinella spiralis was evaluated using a combination of 3 consecutive enzyme-linked immunosorbent assays (ELISAs) based on larval T. spiralis excretory-secretory antigen as screening test and western blot analysis as confirmatory test. Ninety-three of 32,693 domestic swine sera collected in Georgia over a 5-yr period contained antibodies specific to T. spiralis (prevalence of exposure = 0.28%). The highest prevalence (0.52%) of exposure to T. spiralis was in samples from stockyards and salebarns. Prevalence of exposure in samples from cull sows from 1 slaughter house was 0.38% compared with 0.17% in samples obtained from farms. Pepsin-HCl digestion of diaphragms from 49 swine from 6 seropositive farms revealed 0.01 larvae/g in 4 swine from 3 farms. Determination of T. spiralis infection status of farms appears to be accurately determined with this combination of exploratory ELISAs and confirmatory western blot analysis.

Derivation of biological information of a macromolecule isolate based on sequence similarity is playing a significant role in numerous areas of biological research. However, it is often the case that a researcher obtains more macromolecule isolates than can be sequenced practically, due either to the high cost of sequencing or lack of specialized equipment and personnel. To overcome this difficulty, we study the problem of obtaining biological information (such as sequence information) about a macromolecule isolate using only (i) the fragmentation pattern of that isolate obtained from digestion with enzymes and (ii) a database D of sequences. We investigate a three phase approach to solving this problem. In the first phase, we obtain a restriction pattern of the isolate while analytically deriving the corresponding restriction maps of the sequences in the database. In the second phase, we identify a set S [symbol: see text] D of sequences which have restriction maps that are most similar to the unknown isolate's restriction pattern. This task is complicated by the fact that we have only approximate fragment lengths for the unknown isolate and that we do not know the actual ordering of the unknown isolate's fragments. Despite these difficulties, we derive experimental results which indicate maximum matching techniques are effective in identifying the correct set most of the time. In the third phase, we use the set S to infer biological information (such as sequence information or hierarchical classification information) about the unknown isolate. We demonstrate experimentally that the closeness of the sequences in the set S to each other can be used to infer the relatedness of the unknown isolate to the sequences of the set S. Furthermore, the confidence of this inferred information is strongly correlated to the minimum pairwise relatedness of any two elements in S.

A 2-week-old emu chick (Dromaius novaehollandiae) of approximately 200 g body weight was presented for necropsy with a history of weakness, diarrhea, pallor of the head, and acute death. Hemorrhagic enteritis with mild hepatomegaly was noted on gross examination. Microscopic examination revealed necrohemorrhagic enteritis with intralesional intranuclear basophilic viral inclusion bodies in intestinal epithelial cells; splenic lymphoid necrosis and fibrin exudation; hepatocellular vacuolar change; and multiple clusters of small gram-negative bacilli in the liver, spleen, yolk sac, and intestine. Transmission electron microscopy of negatively stained fecal specimens and thin sections of small intestine revealed clusters of viral particles consistent with adenovirus and rotavirus. Attempts at viral isolation from pooled tissue specimens were unsuccessful. Escherichia coli was isolated from specimens of liver and intestine and from an abdominal swab.

In 1 of 2 outbreaks of eastern equine encephalomyelitis virus (EEEV) infection in swine, 280 of 350 pigs died. Histopathologic findings in the brain were typical of EEEV infection. In addition, multifocal necrosis and inflammation were seen in the myocardium. Follow-up investigation in 1 of the herds revealed antibody response to EEEV in surviving pigs and in adult swine. Growth retardation was noticed in surviving pigs. Histologic examination revealed mild lesions in the brain of the survivors.

The value of using a combined infusion of morphine with a variable dose of ketamine for postoperative analgesia following upper abdominal surgery was assessed in a double-blind randomised study of 40 elderly patients. Four groups of 10 patients received an infusion of morphine at 1 mg.h-1, either alone, or combined with ketamine at a rate of 5, 10 or 20 mg.h-1. The addition of ketamine to a continuous infusion of morphine did not significantly improve either analgesia or postoperative lung function. Increasing the dose of ketamine resulted in an increased incidence of postoperative dreaming (p < 0.01).

Porcine small intestinal explants maintained in vitro were inoculated with Salmonella choleraesuis to study the characteristics of its invasion of enterocytes. The explants were fixed at selected intervals for up to 12 hours after inoculation and examined by conventional light microscopy, immunoperoxidase staining, and transmission electron microscopy. Although there was diffuse loss of villous enterocytes during the first hour of incubation, the villi were reepithelialized by the end of 2 hours of culture, and the mucosal epithelium remained intact and appeared to be viable through 12 hours of culture. Intraepithelial S choleraesuis were not detected before 6 hours after inoculation, but after 12 hours of incubation, bacteria were numerous within enterocytes. Ultrastructurally, penetration of the brush border by S choleraesuis resulted in focal loss of microvilli. Bacteria were endocytosed into membrane-bound vacuoles where most remained, but a few were free within the cytoplasm of enterocytes. Invasion of the explants closely resembled that described for live animal and cell culture models of Salmonella spp invasion.

Small intestinal explants from weaned pigs were cultured under a variety of conditions. Explants maintained villus-to-crypt ratio between 1:1 and 1.5:1 for 48 hours. The mucosal epithelium remained well preserved and retained good cellular morphologic features, as determined by light and electron microscopy. Between 48 and 72 hours, considerable mucosal degeneration was evident. Best results were obtained when the explants were cultured on a rocking platform placed in an atmosphere of 95% O2 and 5% CO2, using supplemented RPMI 1640 cell culture medium.

During amino acid starvation the synthesis of rRNA and tRNA is specifically inhibited (stringently controlled) in wild type Escherichia coli but not in relaxed strains carrying the relA mutation. We have found that the in vivo transcription of a hybrid rrnB rRNA operon, in which the normal promoter region has been replaced by the lambda PL promoter, is under stringent control even though this promoter lacks the "stringent discriminator" sequence postulated to be required for stringent control. Furthermore, we have found that transcription of the rrnB operon from a phage T7 promoter, as well as T7 genes in general, by phage T7 RNA polymerase is also subject to stringent control in vivo. These results are consistent with the idea that stringent control acts in a relatively nonspecific manner to inhibit some step(s) in transcription that are often rate-limiting for very active transcription. The relative simplicity of transcription by phage T7 RNA polymerase should offer a good system to study the molecular mechanisms of stringent control.

This paper outlines an investigation into the optimization of traditional formwork construction, using an interation technique. The work was carried out, in part, at the C.S.I.R.O. Division of Building Research, Highett, Victoria, and the computing was executed on the C.S.I.R.O. 3200 C.D.C. Computer system. The original work for this paper was carried out using imperial units. However, for presentation the results have been converted directly into metric S.I.

SINCE the introduction of aircraft carriers to the Fleet of the U. S. Navy their role has continually increased in scope as well as in importance. This has been due chiefly to advances made in aircraft and associated weapons and systems. These advances have had a direct effect on the electric power requirements of the ship service and emergency power system. Fig. 1 shows how ship service generating plants have continually grown, the CV4 in 1930 to the CVA(N) at the present time. The generating capacity of the CVA(N) is ten times greater than that of the CV4. Although this increased power demand has been necessitated by a number of factors such as higher ship's speeds, larger hulls, and more electronic gear, this discussion will cover only certain special power and lighting systems which are considered to be broad enough in scope to be treated individually.

IN all power systems the continuity of power service is dependent upon the characteristics of the devices that are employed in the distribution systems and the proper co-ordination of these devices. It is desirable to have these devices so arranged as to isolate any fault with a minimum disruption of power in the remainder of the power systems. The methods of accomplishing this on navy ships and the limitations of available protective devices were discussed in a previous paper.1 Shipboard power plants have continued to become larger, thus resulting in higher available fault currents. This paper describes these larger systems and discusses the shipboard protective systems and devices employed.

An epizootic of equine infectious anemia (EIA) involved 35 horses on a farm in south Georgia. During a 126-day period, 21 of these horses became seropositive for EIA. After the initial diagnosis in July, the horses were tested every 7 to 10 days. At least one additional horse was found to be seropositive on each testing day. As soon as they were determined to be seropositive, the horses were removed from the herd and sent to slaughter. The removal of the seropositive horses, however, did not stop the epizootic. We believe the initial infection was from a 7-year-old stallion that recently had been purchased or from 1 of 2 mares that were seropositive for EIA on the first test. None of the horses had been tested for EIA at the time of purchase or within 60 days before the epizootic.

Phialemonium obovatum was found to be the cause of osteomyelitis in a German shepherd dog. Histologic examination of the biopsied material from the left tibia revealed septate, irregularly branched hyphae, swollen cells, and ovate-to-spherical cells divided by a transverse septum. The majority of the fungal elements were hyaline, but a few had lightly pigmented cell walls that had a greenish yellow tint. The presence of melanin in the cell walls of the hyphae and especially in their septa was verified by the use of the Fontana-Masson silver stain. P. obovatum formed moist, off-white-to-ochraceous, spreading colonies with a characteristic green pigment on their reverse side. The pigment was more prominent in cultures grown at 37 degrees C than in those grown at 25 degrees C. The isolate also grew at 40 degrees C. The dog isolate produced characteristic adelophialides without conspicuous collarettes and also basal septa from the creeping vegetative hyphae growing on the surface of the medium. The numerous obovate phialoconidia were smooth and one-celled.

It has been shown that the transcription of rRNA in Escherichia coli is feedback-regulated by its own transcription products through a negative feedback loop which appears to require the assembly of rRNA into complete ribosomes. In order to examine whether the feedback loop involves the ribosomes' main function, translation, we have constructed a strain in which the chromosomal copy of infB, encoding IF2, was placed under lac promoter/operator control, and the effects of limitation of translation initiation factor IF2 on the regulation were examined. By varying the concentration of a lac operon inducer, isopropyl thiogalactoside (IPTG), it was possible to vary the cellular concentration of IF2. Under the growth conditions used, decreasing the concentration of IF2 about twofold affected the growth rate only slightly, but further deprivation of IF2 resulted in a significant decrease in growth rate, an increase in RNA content and a large accumulation of non-translating ribosomes. These accumulated ribosomes were apparently unable to cause feedback regulation of rRNA synthesis in the absence of sufficient IF2. When a higher concentration of IPTG was added to these IF2-deficient cells, a rapid increase in the IF2 level and a significant decrease in the rate of RNA accumulation were observed before the new steady-state growth was attained. These results indicate that IF2 apparently is necessary for feedback regulation of stable RNA and imply that ribosomes must enter translation for feedback regulation to occur.

Of 119 consecutive patients with adenocarcinoma of the prostate, 54 patients had clinical Stage C disease and are the subject of this study. Of these 54 patients, 52 underwent transurethral prostatectomy and bilateral orchiectomy as their initial treatment; estrogens were not prescribed. The mean age of the patients with Stage C disease was seventy-three years, the average survival 6.4 years, and the five and ten-year survivals were 66 and 20 per cent, respectively. Stage C patients treated in this series did as well as those treated with radiation or radical extirpation.
