1. Cytoprotective Small Compound M109S Attenuated Retinal Ganglion Cell Degeneration Induced by Optic Nerve Crush in Mice

Abstract

BAX plays an essential role in retinal ganglion cell (RGC) death induced by optic nerve injury. Recently, we developed M109S, an orally bioactive and cytoprotective small compound (CPSC) that inhibits BAX-mediated cell death. We examined whether M109S can protect RGC from optic nerve crush (ONC)-induced apoptosis. M109S was administered starting 5 h after ONC for 7 days. M109S was orally administered in two groups (5 mg/kg twice a day or 7.5 mg/kg once a day). The retina was stained with anti-BRN3A and cleaved Caspase-3 (active Caspase-3) that are the markers of RGC and apoptotic cells, respectively. ONC decreased the number of BRN3A-positive RGC and increased the number of active Caspase-3-expressing apoptotic cells. In ONC-treated retina, there were cells that were double stained with anti-BRN3A and ant-cleaved Caspase-3, indicating that apoptosis in BRN3A-positive RGCs occurred. M109S inhibited the decrease of BRN3A-positive cells whereas it inhibited the increase of active Caspase-3-positive cells in the retina of ONC-treated mice, suggesting that M109S inhibited apoptosis in RGCs. M109S did not induce detectable histological damage to the lungs or kidneys in mice, suggesting that M109S did not show toxicities in the lung or kidneys when the therapeutic dose was used. The present study suggests that M109S is effective in rescuing damaged RGCs. Since M109S is an orally bioactive small compound, M109S may become the basis for a portable patient-friendly medicine that can be used to prevent blindness by rescuing damaged optic nerve cells from death.

Abstract

Abstract Mounting evidence implicates dysfunctional GABAAR-mediated neurotransmission as one of the underlying causes of learning and memory deficits observed in the Ts65Dn mouse model of Down syndrome (DS). The specific origin and nature of such dysfunction is still under investigation, which is an issue with practical consequences to preclinical and clinical research, as well as to the care of individuals with DS and anxiety disorder or those experiencing seizures in emergency room settings. Here, we investigated the effects of GABAAR positive allosteric modulation (PAM) by diazepam on brain activity, synaptic plasticity, and behavior in Ts65Dn mice. We found Ts65Dn mice to be less sensitive to diazepam, as assessed by electroencephalography, long-term potentiation, and elevated plus-maze. Still, diazepam pre-treatment displayed typical effectiveness in reducing susceptibility and severity to picrotoxin-induced seizures in Ts65Dn mice. These findings fill an important gap in the understanding of GABAergic function in a key model of DS.

Abstract

Down syndrome (DS), which results from the complete or partial trisomy of chromosome 21 (trisomy-21), is the most common genetically defined cause of intellectual disability. Trisomy-21 also produces, or is associated with, many neurodevelopmental phenotypes and neurological comorbidities, including delays and deficits in fine and gross motor development. The Ts65Dn mouse is the most studied animal model for DS and displays the largest known subset of DS-like phenotypes. To date, however, only a small number of developmental phenotypes have been quantitatively defined in these animals. Here, we used a commercially available high-speed, video-based system to record and analyze the gait of Ts65Dn and euploid control mice. Longitudinal treadmill recordings were performed from p17 to p35. One of the main findings was the detection of genotype- and sex-dependent developmental delays in the emergence of consistent, progressive-intensity gait in Ts65Dn mice when compared to control mice. Gait dynamic analysis showed wider normalized front and hind stances in Ts65Dn mice compared to control mice, which may reflect deficits in dynamic postural balance. Ts65Dn mice also displayed statistically significant differences in the variability in several normalized gait measures, which were indicative of deficits in precise motor control in generating gait.

Abstract

Introduction: Parents/legal guardians are medical decision-makers for their minor children. Lack of parental capacity to appreciate the implications of the diagnosis and consequences of refusing recommended treatment may impede pediatric patients from receiving adequate medical care. Child and adolescent psychiatrists (CAPs) need to appreciate the ethical considerations relevant to overriding parental medical decision-making when faced with concerns for medical neglect.Methods: Two de-identified cases illustrate the challenges inherent in clinical and ethical decision-making reflected in concerns for parental capacity for medical decision-making. Key ethical principles are reviewed.Case 1: Treatment of an adolescent with an eating disorder ethically complex due to the legal guardian's inability to adhere with treatment recommendations leading to the patient's recurrent abrupt weight loss.Case 2: Questions of parental decisional capacity amid treatment of an adolescent with schizoaffective disorder raised due to parental mistrust of diagnosis, disagreement with treatment recommendations, and lack of appreciation of the medical severity of the situation with repeated discharges against medical advice and medication nonadherence.Discussion: Decisions to question parental capacity for medical decision-making when risk of imminent harm is low but concern for medical neglect exists are controversial. Systematic review of cases concerning for medical neglect benefits from the assessment of parental decisional capacity, review of ethical standards and principles.Conclusion: Recognition of the importance of parental decision-making capacity as relates to parental autonomy and medical neglect and understanding key ethical principles will enhance the CAP's capacity in medical decision-making when stakes are high and absolute recommendations are lacking.

Abstract

We investigate the mass structure of a strong gravitational lens galaxy at $z=0.350$, taking advantage of the milli-arcsecond (mas) angular resolution of very long baseline interferometric (VLBI) observations. In the first analysis of its kind at this resolution, we jointly infer the lens model parameters and pixellated radio source surface brightness. We consider several lens models of increasing complexity, starting from an elliptical power-law density profile. We extend this model to include angular multipole structures, a separate stellar mass component, additional nearby field galaxies, and/or a generic external potential. We compare these models using their relative Bayesian log-evidence (Bayes factor). We find strong evidence for angular structure in the lens; our best model is comprised of a power-law profile plus multipole perturbations and external potential, with a Bayes factor of $+14984$ relative to the elliptical power-law model. It is noteworthy that the elliptical power-law mass distribution is a remarkably good fit on its own, with additional model complexity correcting the deflection angles only at the $\sim5$ mas level. We also consider the effects of added complexity in the lens model on time-delay cosmography and flux-ratio analyses. We find that an overly simplistic power-law ellipsoid lens model can bias the measurement of $H_0$ by $\sim3$ per cent and mimic flux ratio anomalies of $\sim8$ per cent. Our results demonstrate the power of high-resolution VLBI observations to provide strong constraints on the inner density profiles of lens galaxies., 24 pages, 15 figures, 3 tables. Abstract

Poly(acrylic acid) (PAA) bulk gels and threads, typically derived via free-radical polymerization, are of interest as anionic polyelectrolyte mimics of cellular cytosol and as models for early protocells. The thread dimensions have been limited by the diameters of readily-available glass or plastic capillaries, and threads with diameters of less than 50 µm have been difficult to achieve. Here, we report a useful approach for achieving crosslinked, partially neutralized PAA, namely poly(acrylate), gel threads with diameters of a few microns when dry. This technique utilizes coaxial electrospinning to effectively produce capillaries (shells) of polystyrene loaded with a gel-forming precursor mixture composed of 3 M acrylic acid, methylene-bisacrylamide, potassium persulfate and 2.2 M NaOH in the core, followed by thermally-induced polymerization and then the removal of the polystyrene shell. Relatively long (up to 5 mm), continuous PAA threads with thicknesses of 5–15 µm are readily obtained, along with a multitude of PAA gel particles, which result from the occasional break-up of the fluid core prior to gel formation during the electrospinning process. The threads and beads are of the sizes of interest to model ancient protocells, certain functional aspects of excitable cells, such as myocytes and neurons, and various membraneless organelles.

9. Soluble prion protein and its N-terminal fragment prevent impairment of synaptic plasticity by Aβ oligomers: Implications for novel therapeutic strategy in Alzheimer's disease

Abstract

The pathogenic process in Alzheimer's disease (AD) appears to be closely linked to the neurotoxic action of amyloid-β (Aβ) oligomers. Recent studies have shown that these oligomers bind with high affinity to the membrane-anchored cellular prion protein (PrPC). It has also been proposed that this binding might mediate some of the toxic effects of the oligomers. Here, we show that the soluble (membrane anchor-free) recombinant human prion protein (rPrP) and its N-terminal fragment N1 block Aβ oligomers-induced inhibition of long-term potentiation (LTP) in hippocampal slices, an important surrogate marker of cognitive deficit associated with AD. rPrP and N1 are also strikingly potent inhibitors of Aβ cytotoxicity in primary hippocampal neurons. Furthermore, experiments using hippocampal slices and neurons from wild-type and PrPC null mice (as well as rat neurons in which PrPC expression was greatly reduced by gene silencing) indicate that, in contrast to the impairment of synaptic plasticity by Aβ oligomers, the cytotoxic effects of these oligomers, and the inhibition of these effects by rPrP and N1, are independent of the presence of endogenous PrPC. This suggests fundamentally different mechanisms by which soluble rPrP and its fragments inhibit these two toxic responses to Aβ. Overall, these findings provide strong support to recent suggestions that PrP-based compounds may offer new avenues for pharmacological intervention in AD.

Abstract

While the direct detection of the dark-matter particle remains very challenging, the nature of dark matter could be possibly constrained by comparing the observed abundance and properties of small-scale sub-galactic mass structures with predictions from the phenomenological dark-matter models, such as cold, warm or hot dark matter. Galaxy-galaxy strong gravitational lensing provides a unique opportunity to search for tiny surface-brightness anomalies in the extended lensed images (i.e. Einstein rings or gravitational arcs), induced by possible small-scale mass structures in the foreground lens galaxy. In this paper, the first in a series, we introduce and test a methodology to measure the power spectrum of such surface-brightness anomalies from high-resolution Hubble Space Telescope (HST) imaging. In particular, we focus on the observational aspects of this statistical approach, such as the most suitable observational strategy and sample selection, the choice of modelling techniques and the noise correction. We test the feasibility of the power-spectrum measurement by applying it to a sample of galaxy-galaxy strong gravitational lens systems from the Sloan Lens ACS Survey, with the most extended, bright, high-signal-to-noise-ratio lensed images, observed in the rest frame ultraviolet. Abstract

We present DECORAS, a deep learning based approach to detect both point and extended sources from Very Long Baseline Interferometry (VLBI) observations. Our approach is based on an encoder-decoder neural network architecture that uses a low number of convolutional layers to provide a scalable solution for source detection. In addition, DECORAS performs source characterization in terms of the position, effective radius and peak brightness of the detected sources. We have trained and tested the network with images that are based on realistic Very Long Baseline Array (VLBA) observations at 20 cm. Also, these images have not gone through any prior de-convolution step and are directly related to the visibility data via a Fourier transform. We find that the source catalog generated by DECORAS has a better overall completeness and purity, when compared to a traditional source detection algorithm. DECORAS is complete at the 7.5$\sigma$ level, and has an almost factor of two improvement in reliability at 5.5$\sigma$. We find that DECORAS can recover the position of the detected sources to within 0.61 $\pm$ 0.69 mas, and the effective radius and peak surface brightness are recovered to within 20 per cent for 98 and 94 per cent of the sources, respectively. 12. Red quasars blow out molecular gas from galaxies during the peak of cosmic star formation

Abstract

Recent studies have suggested that red quasars are a phase in quasar evolution when feedback from black hole accretion evacuates obscuring gas from the nucleus of the host galaxy. Here, we report a direct link between dust-reddening and molecular outflows in quasars at $z\sim2.5$. By examining the dynamics of warm molecular gas in the inner region of galaxies, we detect outflows with velocities 500--1000 km s$^{-1}$ and infer timescales of $\approx0.1$ Myr that are due to ongoing quasar energy output. We observe outflows only in systems where quasar radiation pressure on dust in the vicinity of the black hole is sufficiently large to expel their obscuring gas column densities. This result is in agreement with theoretical models that predict radiative feedback regulates gas in the nuclear regions of galaxies and is a major driving mechanism of galactic-scale outflows of cold gas. Our findings suggest that radiative quasar feedback ejects star-forming gas from within nascent stellar bulges at velocities comparable to those seen on larger scales, and that molecules survive in outflows even from the most luminous quasars., Comment: Accepted by MNRAS. 18 figures and 3 tables

13. Pharmacological Modulation of Three Modalities of CA1 Hippocampal Long-Term Potentiation in the Ts65Dn Mouse Model of Down Syndrome

Abstract

The Ts65Dn mouse is the most studied animal model of Down syndrome. Past research has shown a significant reduction in CA1 hippocampal long-term potentiation (LTP) induced by theta-burst stimulation (TBS), but not in LTP induced by high-frequency stimulation (HFS), in slices from Ts65Dn mice compared with euploid mouse-derived slices. Additionally, therapeutically relevant doses of the drug memantine were shown to rescue learning and memory deficits in Ts65Dn mice. Here, we observed that 1 μM memantine had no detectable effect on HFS-induced LTP in either Ts65Dn- or control-derived slices, but it rescued TBS-induced LTP in Ts65Dn-derived slices to control euploid levels. Then, we assessed LTP induced by four HFS (4xHFS) and found that this form of LTP was significantly depressed in Ts65Dn slices when compared with LTP in euploid control slices. Memantine, however, did not rescue this phenotype. Because 4xHFS-induced LTP had not yet been characterized in Ts65Dn mice, we also investigated the effects of picrotoxin, amyloid beta oligomers, and soluble recombinant human prion protein (rPrP) on this form of LTP. Whereas ≥10 μM picrotoxin increased LTP to control levels, it also caused seizure-like oscillations. Neither amyloid beta oligomers nor rPrP had any effect on 4xHFS-induced LTP in Ts65Dn-derived slices.

Abstract

After primary infection, varicella-zoster virus (VZV) establishes latency in neurons of the dorsal root and trigeminal ganglia. Many questions concerning the mechanism of VZV pathogenesis remain unanswered, due in part to the strict host tropism and inconsistent availability of human tissue obtained from autopsies and abortions. The recent development of induced pluripotent stem (iPS) cells provides great potential for the study of many diseases. We previously generated human iPS cells from skin fibroblasts by introducing four reprogramming genes with non-integrating adenovirus. In this study, we developed a novel protocol to generate sensory neurons from iPS cells. Human iPS cells were exposed to small molecule inhibitors for 10 days, which efficiently converted pluripotent cells into neural progenitor cells (NPCs). The NPCs were then exposed for two weeks to growth factors required for their conversion to sensory neurons. The iPS cell-derived sensory neurons were characterized by immunocytochemistry, flow cytometry, RT-qPCR, and electrophysiology. After differentiation, approximately 80% of the total cell population expressed the neuron-specific protein, βIII-tubulin. Importantly, 15% of the total cell population co-expressed the markers Brn3a and peripherin, indicating that these cells are sensory neurons. These sensory neurons could be infected by both VZV and herpes simplex virus (HSV), a related alphaherpesvirus. Since limited neuronal populations are capable of supporting the entire VZV and HSV life cycles, our iPS-derived sensory neuron model may prove useful for studying alphaherpesvirus latency and reactivation.

Abstract

Cassiopeia A is one of the best-studied supernova remnants. Its shocked ejecta emits brightly in radio and X-rays. Its unshocked ejecta can be studied through infrared emission, the radio-active decay of $^{44}$Ti, and low frequency free-free absorption due to cold gas internal to the shell. Free-free absorption is affected by the mass, geometry, temperature, and ionisation conditions in the absorbing gas. Observations at the lowest radio frequencies constrain a combination of these properties. We use LOFAR LBA observations at 30-77 MHz and L-band VLA observations to compare $u-v$-matched images with a common resolution of 17". We simultaneously fit, per pixel, for the emission measure and the ratio of the emission from the unabsorbed front of the shell versus the absorbed back of the shell. We explore the effects that low temperatures and a high degree of clumping can have on the derived physical properties, such as mass and density. We also compile published radio flux measurements, fit for the absorption processes that occur in the radio band, and consider how they affect the secular decline of the source. We find a mass in the unshocked ejecta of $M = 2.95 \pm {0.48} \,M_{\odot}$ for an assumed gas temperature of $T=100$ K. This estimate is reduced for colder gas temperatures and if the ejecta are clumped. We measure the reverse shock to have a radius of $114$" $\pm $6". We also find that a decrease in the amount of mass in the unshocked ejecta (as more and more material meets the reverse shock and heats up) cannot account for the observed low frequency behaviour of the secular decline rate. To reconcile our low frequency absorption measurements with models that predict little mass in the unshocked ejecta we need the ejecta to be very clumped, or the temperature in the cold gas to be low ($\sim10$ K). Abstract

Stringent observational constraints on the sub-galactic matter power spectrum would allow one to distinguish between the concordance $\Lambda$CDM and the various alternative dark-matter models that predict significantly different properties of mass structure in galactic haloes. Galaxy-galaxy strong gravitational lensing provides a unique opportunity to probe the sub-galactic mass structure in lens galaxies beyond the Local Group. Here, we demonstrate the first application of a novel methodology to observationally constrain the sub-galactic matter power spectrum in the inner regions of massive elliptical lens galaxies on 1-10 kpc scales from the power spectrum of surface-brightness anomalies in highly magnified galaxy-scale Einstein rings and gravitational arcs. The pilot application of our approach to Hubble Space Telescope (HST/WFC3/F390W) observations of the SLACS lens system SDSS J0252+0039 allows us to place the following observational constraints (at the 99 per cent confidence level) on the dimensionless convergence power spectrum $\Delta^{2}_{\delta\kappa}$ and the standard deviation in the aperture mass $\sigma_{\rm AM}$: $\Delta^{2}_{\delta\kappa}, Comment: 16 pages, 10 figures, updated and resubmitted to MNRAS together with an accompanying methodology paper in response to reviewer's comments

Abstract

International audience; Faraday rotation measurements using the current and next generation of low-frequency radio telescopes will provide a powerful probe of astronomical magnetic fields. However, achieving the full potential of these measurements requires accurate removal of the time-variable ionospheric Faraday rotation contribution. We present ionFR, a code that calculates the amount of ionospheric Faraday rotation for a specific epoch, geographic location, and line-of-sight. ionFR uses a number of publicly available, GPS-derived total electron content maps and the most recent release of the International Geomagnetic Reference Field. We describe applications of this code for the calibration of radio polarimetric observations, and demonstrate the high accuracy of its modeled ionospheric Faraday rotations using LOFAR pulsar observations. These show that we can accurately determine some of the highest-precision pulsar rotation measures ever achieved. Precision rotation measures can be used to monitor rotation measure variations – either intrinsic or due to the changing line-of-sight through the interstellar medium. This calibration is particularly important for nearby sources, where the ionosphere can contribute a significant fraction of the observed rotation measure. We also discuss planned improvements to ionFR, as well as the importance of ionospheric Faraday rotation calibration for the emerging generation of low-frequency radio telescopes, such as the SKA and its pathfinders.

Abstract

Context. The LOw Frequency ARray (LOFAR) radio telescope is a giant digital phased array interferometer with multiple antennas distributed in Europe. It provides discrete sets of Fourier components of the sky brightness. Recovering the original brightness distribution with aperture synthesis forms an inverse problem that can be solved by various deconvolution and minimization methods Aims. Recent papers have established a clear link between the discrete nature of radio interferometry measurement and the "compressed sensing" (CS) theory, which supports sparse reconstruction methods to form an image from the measured visibilities. Empowered by proximal theory, CS offers a sound framework for efficient global minimization and sparse data representation using fast algorithms. Combined with instrumental direction-dependent effects (DDE) in the scope of a real instrument, we developed and validated a new method based on this framework Methods. We implemented a sparse reconstruction method in the standard LOFAR imaging tool and compared the photometric and resolution performance of this new imager with that of CLEAN-based methods (CLEAN and MS-CLEAN) with simulated and real LOFAR data Results. We show that i) sparse reconstruction performs as well as CLEAN in recovering the flux of point sources; ii) performs much better on extended objects (the root mean square error is reduced by a factor of up to 10); and iii) provides a solution with an effective angular resolution 2-3 times better than the CLEAN images. Conclusions. Sparse recovery gives a correct photometry on high dynamic and wide-field images and improved realistic structures of extended sources (of simulated and real LOFAR datasets). This sparse reconstruction method is compatible with modern interferometric imagers that handle DDE corrections (A- and W-projections) required for current and future instruments such as LOFAR and SKA, Published in A&A, 19 pages, 9 figures

Abstract

We present the Multifrequency Snapshot Sky Survey (MSSS), the first northern-sky LOFAR imaging survey. In this introductory paper, we first describe in detail the motivation and design of the survey. Compared to previous radio surveys, MSSS is exceptional due to its intrinsic multifrequency nature providing information about the spectral properties of the detected sources over more than two octaves (from 30 to 160 MHz). The broadband frequency coverage, together with the fast survey speed generated by LOFAR's multibeaming capabilities, make MSSS the first survey of the sort anticipated to be carried out with the forthcoming Square Kilometre Array (SKA). Two of the sixteen frequency bands included in the survey were chosen to exactly overlap the frequency coverage of large-area Very Large Array (VLA) and Giant Metrewave Radio Telescope (GMRT) surveys at 74 MHz and 151 MHz respectively. The survey performance is illustrated within the "MSSS Verification Field" (MVF), a region of 100 square degrees centered at J2000 (RA,Dec)=(15h,69deg). The MSSS results from the MVF are compared with previous radio survey catalogs. We assess the flux and astrometric uncertainties in the catalog, as well as the completeness and reliability considering our source finding strategy. We determine the 90% completeness levels within the MVF to be 100 mJy at 135 MHz with 108" resolution, and 550 mJy at 50 MHz with 166" resolution. Images and catalogs for the full survey, expected to contain 150,000-200,000 sources, will be released to a public web server. We outline the plans for the ongoing production of the final survey products, and the ultimate public release of images and source catalogs., Comment: 23 pages, 19 figures. Abstract

We report the discovery of a new gravitational lens system from the CLASS survey. Radio observations with the VLA, the WSRT and MERLIN show that the radio source B0850+054 is comprised of two compact components with identical spectra, a separation of 0.7 arcsec and a flux density ratio of 6:1. VLBA observations at 5 GHz reveal structures that are consistent with the gravitational lens hypothesis. The brighter of the two images is resolved into a linear string of at least six sub-components whilst the weaker image is radially stretched towards the lens galaxy. UKIRT K-band imaging detects an 18.7 mag extended object, but the resolution of the observations is not sufficient to resolve the lensed images and the lens galaxy. Mass modelling has not been possible with the present data and the acquisition of high-resolution optical data is a priority for this system., Comment: 5 pages, 4 figures, accepted for publication in MNRAS

Abstract

Abell 2256 is one of the best known examples of a galaxy cluster hosting large-scale diffuse radio emission that is unrelated to individual galaxies. It contains both a giant radio halo and a relic, as well as a number of head-tail sources and smaller diffuse steep-spectrum radio sources. The origin of radio halos and relics is still being debated, but over the last years it has become clear that the presence of these radio sources is closely related to galaxy cluster merger events. Here we present the results from the first LOFAR Low band antenna (LBA) observations of Abell 2256 between 18 and 67 MHz. To our knowledge, the image presented in this paper at 63 MHz is the deepest ever obtained at frequencies below 100 MHz in general. Both the radio halo and the giant relic are detected in the image at 63 MHz, and the diffuse radio emission remains visible at frequencies as low as 20 MHz. The observations confirm the presence of a previously claimed ultra-steep spectrum source to the west of the cluster center with a spectral index of -2.3 \pm 0.4 between 63 and 153 MHz. The steep spectrum suggests that this source is an old part of a head-tail radio source in the cluster. For the radio relic we find an integrated spectral index of -0.81 \pm 0.03, after removing the flux contribution from the other sources. This is relatively flat which could indicate that the efficiency of particle acceleration at the shock substantially changed in the last \sim 0.1 Gyr due to an increase of the shock Mach number. In an alternative scenario, particles are re-accelerated by some mechanism in the downstream region of the shock, resulting in the relatively flat integrated radio spectrum. In the radio halo region we find indications of low-frequency spectral steepening which may suggest that relativistic particles are accelerated in a rather inhomogeneous turbulent region., Comment: 13 pages, 13 figures, accepted for publication in A\&A on April 12, 2012

Abstract

A newly engineered implementation of a collimated SX700‐style beam line for soft x‐rays is described. This facility is operational at the Advanced Light Source and delivers high brightness undulator beams to a scanning zone plate microscope and to an array of end stations for x‐ray spectroscopic studies of wet surfaces. Switching between branches is motorized, servo‐steering systems maintain throughput and the monochromator works together with the elliptical undulator for a fully automated facility.

Abstract

The design and initial commissioning of the first IR beamline at the ALS has been described previously. We report the final commissioning and first results of the mid-IR spectromicroscopy beamline 1.4.3. In addition, several improvements and two new branchlines are presented. Beamline 1.4.2 is connected to the front end under vacuum and consists of a Bruker Rapid- and Step-Scan vacuum FTIR bench. The modulated light is then coupled into a UHV surface science chamber for grazing incidence reflection studies. Several more external ports are available from the Bruker bench. Beamline 1.4.1 receives light from a separate port on the beamline 1.4 front end and connects to an optical table for photoluminescence and other experiments using photons with energies up to 6 eV.

Abstract

A simple semi-continuous piston corer for stratigraphic sampling up to 15 m of organic lake sediments has been developed. The design is flexible so that components can be assembled to suit the depth of core required. For cores up to 6 m, the apparatus is easily deployed from a small boat with the minimum of difficulty. With increasingly long cores, the operational complexity increases. For the designed maximum core depth, two workers on the surface and one SCUBA diver are required to operate the corer. A small stable inflatable boat or raft is required as a platform.

Abstract

Concentrations of zinc and copper in a series of large oligotrophic lakes on Vancouver Island, Canada, have been increasing since mining operations began near the south end of Buttle lake in 1966. Declines in species diversity of both phytoplankton and periphyton have occurred since the beginning of mining activity with the disappearance of metal sensitive forms such asAsterionella andRhizosolenia. Cladoceran and calanoid copepod numbers and species diversity increase as the metal concentrations decline downstream from the mine. Bioassays carried out withDaphnia pulex show increasing toxicity downstream. Metal concentrations in Zooplankton and metallothionein concentrations in rainbow trout liver are correlated to metal concentrations in the water. The water was not acutely toxic to rainbow trout in laboratory simulations of Buttle Lake conditions or duringin situ exposures (1 month). Specific sublethal deleterious effects have not yet been determined.

Abstract

Twenty-three softwater lakes in British Columbia, Canada, were sampled before fall overturn to assess the in situ conditions associated with internal phosphorus loading. Hypolimnetic sediment and water results were correlated with physical and morphological characteristics of each lake. The concentration of soluble reactive phosphorus (SRP) measured 1 m above the sediment–water interface was strongly correlated with the oxidation reduction potential (ORP) measured at the same depth. Above 0 mV, soluble reactive phosphorus was below 3 μgL−1 at 1 m above the sediment–water interface. Below 0 mV, soluble reactive phosphorus concentrations increased exponentially with decreasing oxidation reduction potential. A model using sediment loss on ignition (LOI) and spring overturn phosphorus (SOP) can predict the hypolimnetic oxidation reduction potential. Lakes with areal water loading values < 10 m yr−1 typically had higher loss on ignition and spring overturn phosphorus concentrations. High loss on ignit...
