Tibial torsion assessment is crucial for understanding deformities and malalignments that can lead to joint pathologies in dogs. Different methods such as radiography, computed tomography (CT), and three-dimensional (3D) volume-rendering techniques have been employed to measure tibial torsion. This study compared the accuracy and reliability of tibial torsion angle (TTa) measurements obtained using radiography and ultrasound tilting techniques against those obtained using the 3D volume-rendering method in small-to-medium-sized non-chondrodystrophic dogs. Seven dogs with 11 hind limbs were included in this study. Descriptive statistics revealed mean TTa values for radiography (1.6° ± 5.14°), ultrasound (2.92° ± 3.98°), CT (4.57° ± 3.44°), and 3D volume-rendering method (5.29° ± 3.30°). Intraclass correlation coefficient (ICC) analysis indicated excellent intra- and interobserver agreement between the radiography and ultrasound methods. Correlation analysis showed positive correlations between all the methods. These findings demonstrate that radiography and the ultrasound tilting technique are reliable alternatives for measuring TTa. Although slightly lower ICC values were observed than those of the 3D volume-rendering technique, the radiography and ultrasound methods still exhibited good to excellent reliability, suggesting that these alternative methods could be effective diagnostic tools for assessing TTa in clinical settings with high accuracy and reliability.

Abstract Background Transcranial direct current stimulation (tDCS) is a promising treatment for major depressive disorder (MDD). This study evaluated its antidepressant and cognitive effects as a safe, effective, home-based therapy for MDD. Methods This double-blind, sham-controlled, randomized trial divided participants into low-intensity (1 mA, n = 47), high-intensity (2 mA, n = 49), and sham (n = 45) groups, receiving 42 daily tDCS sessions, including weekends and holidays, targeting the dorsolateral prefrontal cortex for 30 minutes. Assessments were conducted at baseline and weeks 2, 4, and 6. The primary outcome was cognitive improvement assessed by changes in total accuracy on the 2-back test from baseline to week 6. Secondary outcomes included changes in depressive symptoms (HAM-D), anxiety (HAM-A), and quality of life (QLES). Adverse events were monitored. This trial was registered with ClinicalTrials.gov (NCT04709952). Results In the tDCS study, of 141 participants (102 [72.3%] women; mean age 35.7 years, standard deviation 12.7), 95 completed the trial. Mean changes in the total accuracy scores from baseline to week 6 were compared across the three groups using an F-test. Linear mixed-effects models examined the interaction of group and time. Results showed no significant differences among groups in cognitive or depressive outcomes at week 6. Active groups experienced more mild adverse events compared to sham but had similar rates of severe adverse events and dropout. Conclusions Home-based tDCS for MDD demonstrated no evidence of effectiveness but was safe and well-tolerated. Further research is needed to address the technical limitations, evaluate broader cognitive functions, and extend durations to evaluate its therapeutic potential.

Jejunal haemorrhage syndrome (JHS) is a sporadic and fatal enterotoxaemic disease in dairy cows associated with acute development and poor prognosis despite treatment. A 5-year-old Holstein cow with no reported pregnancy, three calving numbers, and 303 days in milk presented with hypothermia, discomfort, and inappetence. Anaemia, dehydration, faeces with blood clots, and absence of rumen and bowel movements were observed. We identified the presence of neutrophilia, hyperglycaemia, hypoproteinaemia, azotaemia, hyperlactatemia, hypocalcaemia, hypermagnesemia, hypokalaemia, and hypochloraemia through blood analyses. Necropsy and histopathologic examination revealed a dilated bluish-purple jejunum, blood clots within the jejunum, neutrophil infiltration into the submucosa of the jejunum, and vascular necrosis. Retrospective examination revealed extraordinary patterns of rumination time, activity, rumen mobility, and rumen temperature using biosensors and decreased milk yield. The abnormalities in the affected cow were detected before recognition by farm workers. To the best of our knowledge, this is the first report to examine data from biosensors in a cow with JHS. Our findings suggest that using biometric data may help understand the development of JHS.

A 19-month-old Korean native cow died the following day after consuming new silage. Grossly, the liver showed enlargement, redness, and haemorrhages in all the lobes. Additionally, many of the bur-shaped fruits of the cocklebur (Xanthium strumarium) were found in the gastric contents. The histological evaluation confirmed centrilobular hepatic necrosis. Additionally, carboxyatractyloside (CATR), a material fatal to animals found in the cocklebur, was detected in the gastric contents using high-performance liquid chromatography-quadrupole-time of flight mass spectrometry (HPLC-Q-TOF-MS). Based on the pathological findings and analytical confirmation, CATR intoxication was diagnosed. Therefore, careful feeding and elimination of the cocklebur is essential for minimising economic loss.

Cryptosporidium parvum is an enteric protozoan, which causes severe diarrhoea in a wide range of vertebrate hosts including ruminants and humans. C. parvum infections are responsible for immense economic losses to the livestock industry; furthermore, the zoonotic spread of the infection may lead to mortality in immunosuppressed humans. In the present study, we report two cases of severe cryptosporidiosis in goat kids in the Republic of Korea. Both cases were afflicted with severe diarrhoea upon presentation; the histopathological examinations revealed atrophied and fused intestinal villi and numerous circular basophilic organisms in the jejunum and ileum. Both cases were diagnosed with cryptosporidiosis based on the results of the histopathological analysis, amplification of the C. parvum gp60 gene, modified Ziehl-Neelsen staining, and C. parvum antigen ELISA. According to the phylogenetic analysis using the C. parvum gp60 gene for the genetic subtypes, the C. parvum isolates were identified as subtype IIdA15G1 with zoonotic potential. This is the first pathological report of caprine cryptosporidiosis induced by C. parvum subtype IId in the Republic of Korea. Considering the clinical manifestations associated with the pathological lesions and the zoonotic significance of these findings, the continuous monitoring and prevention of C. parvum infections in goats are essential for minimising the economic losses in ruminant farms and in maintaining public health safety standards.

While a large body of work in sentence comprehension has explored how different types of linguistic information are used to guide syntactic parsing, less is known about the effect of discourse structure. This study investigates this question, focusing on the main and subordinate discourse contrast manifested in the distinction between restrictive relative clauses (RRCs) and appositive relative clauses (ARCs) in American English. In three self-paced reading experiments, we examined whether both RRCs and ARCs interfere with the matrix clause content and give rise to the agreement attraction effect. While the standard attraction effect was consistently observed in the baseline RRC structures, the effect varied in the ARC structures. These results collectively suggest that discourse structure indeed constrains syntactic dependency resolution. Most importantly, we argue that what is at stake is not the static discourse structure properties at the global sentence level. Instead, attention should be given to the incremental update of the discourse structure in terms of which "discourse questions" are active at any given moment of a discourse. The current findings have implications for understanding the way discourse structure, specifically the active state of discourse questions, constrains memory retrieval.

Friendships are important for the mental well-being of emerging adults. Socially prescribed perfectionism, where individuals feel pressured to be perfect by others, can be destructive, leading to conflict with others, depressive symptoms, and problematic drinking. However, its impact on friendships is not well-explored. This study examined 174 emerging adult friendship dyads using a 4-wave, 4-month dyadic design. Data were analyzed using longitudinal actor-partner interdependence models. Using a novel friend-specific measure of socially prescribed perfectionism, we found that an individual's perceived expectation to be perfect from a friend was positively associated with increased conflict between friends, as well as with higher levels of depressive symptoms and problematic drinking in the individual. Findings lend credence to longstanding theoretical accounts and case histories suggesting socially prescribed perfectionism leads to harmful individual and relational outcomes and extends them to the specific context of friendships.

Osteoporosis is a disease characterized by low bone mass, most commonly caused by an increase in bone resorption that is not matched by sufficient bone formation. The most common complications of postmenopausal osteoporosis are bone-related defects and fractures. Fracture healing is a multifactorial bone regeneration process, influenced by both biological and mechanical factors related to age, osteoporosis and stability of the osteosynthesis. During the treatment of bone defects in osteoporotic conditions, imbalanced bone remodeling is the leading cause for implant failure. To overcome these problems, ethyl-2,5-dihydroxybenzoate (E-2,5-DHB), a drug that promotes bone formation and inhibits bone resorption, was used. E-2,5-DHB-incorporating titanium (Ti) implants using poly(lactic-co-glycolic acid) (PLGA) coating for local delivery of E-2,5-DHB were developed and the effects on bone healing of femoral defects were evaluated in an osteoporotic model. The release of E-2,5-DHB resulted in decreased bone resorption and increased bone formation around the implant. Thus, it was confirmed that, in the osteoporotic model, bone healing was increased and implant fixation was enhanced. These results suggested that E-2,5-DHB-coated Ti implants have great potential as an ultimate local drug delivery system for bone tissue scaffolds.

Focusing effect of laser-driven positron jets by self-generated target sheath fields has been observed for the first time experimentally and the results are supported by the computational studies. In the experiment, OMEGA EP short-pulse (0.7 ps, 500 J) irradiates mm-size gold targets with a concave back surface and reference flat-surface targets. Both targets exhibited positrons with quasi-monoenergetic energy peaks while targets with concave curvature also showed increased number of positrons at the detector. The data is consistent with hybrid-PIC simulations confirming that the time-varying electric fields driven by electrons escaping from the target significantly change the trajectories of positrons. These simulations show a small radius of curvature on the rear side increases the relative focusing effect and the positrons to electrons ratio in the escaping plasma. For the smallest radius of curvature, positron jets that are up to 10 times denser can be achieved.

We report experimental results on relativistic electron beam (REB) transport in a set of cold and shock-heated carbon samples using the high-intensity kilojoule-class OMEGA EP laser. The REB energy distribution and transport were diagnosed using an electron spectrometer and x-ray fluorescence measurements from a Cu tracer buried at the rear side of the samples. The measured rear REB density shows brighter and narrower signals when the targets were shock-heated. Hybrid PIC simulations using advanced resistivity models in the target warm-dense-matter (WDM) conditions confirm this observation. We show that the resistivity response of the media, which governs the self-generated resistive fields, is of paramount importance to understand and correctly predict the REB transport.

We derive exact expressions for effective elastodynamic properties of two-phase composites in the long-wavelength (quasistatic) regime via homogenized constitutive relations that are local in space. This is accomplished by extending the ‘strong-contrast’ expansion formalism that was previously applied to the static problem. These strong-contrast expansions explicitly incorporate complete microstructural information of the composite via an infinite set of n -point correlation functions. Utilizing the rapid-convergence properties of these series expansions (even for extreme contrast ratios), we extract accurate approximations that depend on the microstructure via the spectral density, which is easy to compute or measure for any composite. We also investigate the predictive power of modifications of such approximation formulas postulated elsewhere (Kim and Torquato 2020 Proc. Natl Acad. Sci. 117 8764) to extend their applicability beyond the quasistatic regime . The accuracy of these nonlocal microstructure-dependent approximations is validated by comparison to full-waveform simulation results for certain models of dispersions. We apply our formulas to a variety of models of nonhyperuniform and hyperuniform disordered composites. We demonstrate that hyperuniform systems are less lossy than their nonhyperuniform counterparts in the quasistatic regime, and stealthy hyperuniform media can be perfectly transparent for a wide range of wavenumbers. Finally, we discuss how to utilize our approximations for engineering composites with prescribed elastic wave characteristics.

Attention is biased toward features aligning with task goals and stimuli previously allocated attentional priority (selection history). The relationship between selection history and the strategic control of attention has scarcely been explored. In the present study, we utilized a modified version of the Adaptive Choice Visual Search (ACVS) task to determine whether the choice of visual search strategy varies with the strategies participants have elected to use in the past. Participants were tasked with searching through stimuli presented in two task-relevant colors on each trial to find one of two targets. The distribution of stimuli rendered in these two colors was manipulated between subjects, with one group receiving more imbalanced displays during learning. Participants who experienced the more imbalanced displays quickly learned the optimal visual search strategy of searching through the less abundant color, which maximized performance. Critically, these participants retained their tendency toward this learned strategy in a subsequent test phase in which displays were less imbalanced, in contrast to participants who completed the same test phase but had only experienced the less imbalanced displays. Our results demonstrate that, without explicit instruction, the choice of visual search strategy is to some degree dependent upon selection history.

Active learning approaches to biology teaching, including simulation-based activities, are known to enhance student learning, especially of higher-order skills; nonetheless, there are still many open questions about what features of an activity promote optimal learning. Here we designed three versions of a simulation-based tutorial called Understanding Experimental Design that asks students to design experiments and collect data to test their hypotheses. The three versions vary the experimental design task along the axes of feedback and constraint, where constraint measures how much choice students have in performing a task. Using a variety of assessments, we ask whether each of those features affects student learning of experimental design. We find that feedback has a direct positive effect on learning. We further find that small changes in constraint have only subtle and mostly indirect effects on learning. This work suggests that designers of tools for teaching higher-order skills should strive to include feedback to increase impact and may feel freer to vary the degree of constraint within a range to optimize for other features such as the ability to provide immediate feedback and time-on-task.

A theory of change is a prediction about desired outcomes and the actions that may prompt them. Research on educational change suggests that explicit theories of change are essential for effectively engaging in academic change work. While many centers for teaching and learning (CTL) express aims around organizational change, a recent study was not able to identify any theories of change made public in annual reports or websites. Therefore, we outline an adaptable process for other CTLs to discuss and make explicit their own theories of change. This process involves four steps, moving across scale of a CTL (large group, small group, and individual). We offer recommendations for other types of CTLs, such as "centers of one," to develop a theory of change and theorize about current and potential utility of a theory of change for a CTL's work.

NIMA-related kinase 1 (NEK1) is a serine/threonine and tyrosine kinase that is highly expressed in mammalian germ cells. Mutations in Nek1 induce anemia, polycystic kidney and infertility. In this study we evaluated the role of NEK1 in meiotic spindle formation in both male and female gametes. Our results show that the lack of NEK1 provokes an abnormal organization of the meiosis I spindle characterized by elongated and/or multipolar spindles, and abnormal chromosome congression. The aberrant spindle structure is concomitant with the disruption in localization and protein levels of myosin X (MYO10) and α-adducin (ADD1), both of which are implicated in the regulation of spindle formation during mitosis. Interaction of ADD1 with MYO10 is dependent on phosphorylation, whereby phosphorylation of ADD1 enables its binding to MYO10 on mitotic spindles. Reduction in ADD1 protein in NEK1 mutant mice is associated with hyperphosphorylation of ADD1, thereby preventing the interaction with MYO10 during meiotic spindle formation. Our results reveal a novel regulatory role for NEK1 in the regulation of spindle architecture and function during meiosis.

To enhance fuel safety and to achieve a power uprating, a new concept PWR (Pressurized Water Reactor) fuel, which is called a dual cooled fuel rod, has been studied since 2007. Although there are some challenging problems about each mechanical component, fuel rod supporting structures are especially dealt within this paper. In the dual-cooled annular fuel rod, an inner flow passage as well as an outer one is used not only to enhance the fuel safety but to achieve a power uprating through the decreasing fuel center temperature and increasing the heat transfer area between fuel and coolant. As a result, the diameter of dual-cooled fuel rods becomes 1.6 times bigger than a conventional solid fuel rod and the gap between the annular fuel and the spacer grid’s straps is narrowed. This is a current key issue of fuel rod supports. To accommodate the narrowed gap, spacer grids, such as the cantilever type, hemi-sphere type, circular insertion type, etc., were suggested. For some of these configurations, patents have been applied. The grid springs in a spacer grid play the role of holding the fuel rods in an appropriate position and preventing fuel rods from dropping during normal reactor operation. In the case of the dual cooled fuel rod, the total mass is increased. So, the stiffness of a grid spring has to be increased more than that of a conventional grid spring. However, if the stiffnesses of springs are increased too much, fuel rods can bend due to the prohibition of their axial slip. So, it is necessary to design an appropriate stiffness. In this paper, the minimum spring force to prevent dual-cooled fuel rods from dropping during normal reactor operation is calculated. The spring characteristics of a cantilever type and a hemi-sphere type are predicted. A finite element analysis is carried out by using the commercial code ABAQUS. The analysis results are verified by experiments. Finally, it is checked whether the property of the suggested springs satisfies the minimum required spring force. Based on the obtained results, a kind of spacer grid candidate for dual cooled fuel rods, i.e. a spacer grid with hybrid supports is suggested.

Somitogenesis is regulated by a molecular oscillator that drives dynamic gene expression within the pre-somitic mesoderm. Previous mathematical models of the somitogenesis clock that invoke the mechanism of delayed negative feedback predict that its oscillation period depends on the sum of delays inherent to negative-feedback loops and inhibitor half-lives. We develop a mathematical model that explores the possibility that positive feedback also plays a role in determining the period of clock oscillations. The model predicts that increasing the half-life of the positive regulator, Notch intracellular domain (NICD), can lead to elevated NICD levels and an increase in the oscillation period. To test this hypothesis, we investigate a phenotype induced by various small molecule inhibitors in which the clock is slowed. We observe elevated levels and a prolonged half-life of NICD. Reducing NICD production rescues these effects. These data provide the first indication that tight control of the turnover of positive as well as negative regulators of the clock determines its periodicity.

The identification of the H3K4 trimethylase, PRDM9, as the gene responsible for recombination hotspot localization has provided considerable insight into the mechanisms by which recombination is initiated in mammals. However, uniquely amongst mammals, canids appear to lack a functional version of PRDM9 and may therefore provide a model for understanding recombination that occurs in the absence of PRDM9, and thus how PRDM9 functions to shape the recombination landscape. We have constructed a fine-scale genetic map from patterns of linkage disequilibrium assessed using high-throughput sequence data from 51 free-ranging dogs, Canis lupus familiaris. While broad-scale properties of recombination appear similar to other mammalian species, our fine-scale estimates indicate that canine highly elevated recombination rates are observed in the vicinity of CpG rich regions including gene promoter regions, but show little association with H3K4 trimethylation marks identified in spermatocytes. By comparison to genomic data from the Andean fox, Lycalopex culpaeus, we show that biased gene conversion is a plausible mechanism by which the high CpG content of the dog genome could have occurred.

The RAD9-RAD1-HUS1 (9-1-1) complex is a heterotrimeric PCNA-like clamp that responds to DNA damage in somatic cells by promoting DNA repair as well as ATR-dependent DNA damage checkpoint signaling. In yeast, worms, and flies, the 9-1-1 complex is also required for meiotic checkpoint function and efficient completion of meiotic recombination; however, since Rad9, Rad1, and Hus1 are essential genes in mammals, little is known about their functions in mammalian germ cells. In this study, we assessed the meiotic functions of 9-1-1 by analyzing mice with germ cell-specific deletion of Hus1 as well as by examining the localization of RAD9 and RAD1 on meiotic chromosomes during prophase I. Hus1 loss in testicular germ cells resulted in meiotic defects, germ cell depletion, and severely compromised fertility. Hus1-deficient primary spermatocytes exhibited persistent autosomal γH2AX and RAD51 staining indicative of unrepaired meiotic DSBs, synapsis defects, an extended XY body domain often encompassing partial or whole autosomes, and an increase in structural chromosome abnormalities such as end-to-end X chromosome-autosome fusions and ruptures in the synaptonemal complex. Most of these aberrations persisted in diplotene-stage spermatocytes. Consistent with a role for the 9-1-1 complex in meiotic DSB repair, RAD9 localized to punctate, RAD51-containing foci on meiotic chromosomes in a Hus1-dependent manner. Interestingly, RAD1 had a broader distribution that only partially overlapped with RAD9, and localization of both RAD1 and the ATR activator TOPBP1 to the XY body and to unsynapsed autosomes was intact in Hus1 conditional knockouts. We conclude that mammalian HUS1 acts as a component of the canonical 9-1-1 complex during meiotic prophase I to promote DSB repair and further propose that RAD1 and TOPBP1 respond to unsynapsed chromatin through an alternative mechanism that does not require RAD9 or HUS1.

The aroma volatile compounds in tomatoes are strongly linked to taste and flavor. Commercial tomato varieties, however, generally lack flavor. Tomato breeding programs have therefore started to focus on increasing the flavor-enhancing volatiles, which can be more easily altered than the sugar content. Analytical efforts that identify key flavor volatiles are therefore an important part of tomato improvement programs. Based on this real-world goal, an upper-level undergraduate analytical laboratory exercise has been developed that utilizes solid-phase microextraction (SPME) and gas chromatography-mass spectrometry to characterize tomato volatiles. Students adsorb volatile organic compounds (VOCs) and a spiked internal standard onto SPME fibers, and these volatiles are thermally desorbed in a gas chromatograph--mass spectrometer (GC-MS) for identification and quantification. VOCs from hybrid heirloom tomatoes (open-pollinated, older cultivars not bred for industrial agriculture) and store-bought tomatoes are compared and normalized to the internal standard, and students determine which volatiles contribute to positive (and negative) taste perceptions. The laboratory exercise has been employed successfully in our Instrumental Analysis course over three years. Using previously obtained data, the exercise was also modified for remote (Zoom) laboratory learning. The experimental protocol is robust with few technical issues, and students produce replicable data. Because students can relate easily to the subject matter of the research, engagement is high. A large, complex data set is produced in this exercise, and students learn to draw conclusions from this data set that mimic real-world, authentic research.

The mammalian ortholog of yeast Slx4, BTBD12, is an ATM substrate that functions as a scaffold for various DNA repair activities. Mutations of human BTBD12 have been reported in a new sub-type of Fanconi anemia patients. Recent studies have implicated the fly and worm orthologs, MUS312 and HIM-18, in the regulation of meiotic crossovers arising from double-strand break (DSB) initiating events and also in genome stability prior to meiosis. Using a Btbd12 mutant mouse, we analyzed the role of BTBD12 in mammalian gametogenesis. BTBD12 localizes to pre-meiotic spermatogonia and to meiotic spermatocytes in wildtype males. Btbd12 mutant mice have less than 15% normal spermatozoa and are subfertile. Loss of BTBD12 during embryogenesis results in impaired primordial germ cell proliferation and increased apoptosis, which reduces the spermatogonial pool in the early postnatal testis. During prophase I, DSBs initiate normally in Btbd12 mutant animals. However, DSB repair is delayed or impeded, resulting in persistent γH2AX and RAD51, and the choice of repair pathway may be altered, resulting in elevated MLH1/MLH3 focus numbers at pachynema. The result is an increase in apoptosis through prophase I and beyond. Unlike yeast Slx4, therefore, BTBD12 appears to function in meiotic prophase I, possibly during the recombination events that lead to the production of crossovers. In line with its expected regulation by ATM kinase, BTBD12 protein is reduced in the testis of Atm(-/-) males, and Btbd12 mutant mice exhibit increased genomic instability in the form of elevated blood cell micronucleus formation similar to that seen in Atm(-/-) males. Taken together, these data indicate that BTBD12 functions throughout gametogenesis to maintain genome stability, possibly by co-ordinating repair processes and/or by linking DNA repair events to the cell cycle via ATM.
