The bamboo worm Omphisa fuscidentalis is a kind of popular edible insect in Indochina Peninsula, however, the genus affiliation of this insect is controversial. Previous taxonomic studies from China and Thailand using limited morphology and single mitochondrial gene evidence indicated that, those samples distributed in different areas from Asia could belong to different species in different genera. To understand its taxonomic status and phylogenetic relationship with related taxa, we first sequenced and characterised the complete mitochondrial genome (mitogenome) of the moth O. fuscidentalis’ mitogenome is 15,347 bp in size, and containing 13 protein-coding genes (PCGs), 2 rRNAs, 22 tRNAs, and a control region. Further, 23 of the 37 genes are encoded by the J-strand while 14 are encoded by the N-strand. Most of PCGs are initiated by ATN codons, but 3 of the 13 PCGs harboured the incomplete termination codon by ATT. In the gene order of O. fuscidentalis, the ancestor of Lepidoptera sequence trnI-trnQ-trnM morphed into trnM-trnI-trnQ, as observed in other Pyraloidea species. A phylogenetic tree was then constructed based on the nucleotide sequences of mitochondrial 13 PCGs from 39 Pyraloidea species and two Tortricoidea species (outgroups) using Bayesian inference and maximum likelihood, which determined that O. fuscidentalis belongs to Crambidae. Accordingly, the bamboo worm does not belong to the genus Chilo in the subfamily Crambinae, or to the subfamily Spilomelinae. This study provides fundamental data to clarify the taxonomic status and better understand the biology of this edible insect.

Radiographic joint space width (JSW) has been a standard for measuring knee osteoarthritis (OA) structural change. Limitations in the responsiveness of this approach might be overcome by instead measuring 3D JSW on weight-bearing CT (WBCT). This study compared the responsiveness of 3D JSW measurements using WBCT with the responsiveness of radiographic 2D JSW.Standing, fixed-flexion knee radiographs (XR) and WBCT were acquired ancillary to the 144- and 168-month Multicenter Osteoarthritis Study visits. Tibiofemoral JSW was measured on both XR and WBCT. Responsiveness to change was defined by the standardized response mean (SRM) for change in JSW (1) at predetermined mediolateral locations (JSWx) on both modalities and (2) in the following subregions measured on WBCT images: central medial and lateral femur (CMF/CLF) and tibia (CMT/CLT), and anterior and posterior tibia (AMT/ALT, PMT/MLT).Baseline and 24-month follow-up JSWx measurements were completed for 265 participants (58.1% women). Responsiveness of 3D JSWx for medial tibiofemoral compartment on coronal WBCT (SRM range: -0.18, -0.24) exceeded that for 2D JSWx (-0.10, -0.16). Responsiveness of 3D JSW subregional mean (-0.06, -0.36) and maximal (-1.14, -1.75) CMF and CMT and maximal CLF/CLT 3D JSW changes were statistically significantly greater in comparison with respective medial and lateral 2D JSWx (P ≤ 0.002).Subregional 3D JSW on WBCT is substantially more responsive to 24-month changes in tibiofemoral joint structure compared to radiographic measurements. Use of subregional 3D JSW on WBCT could enable improved detection of OA structural progression over a 24-month duration in comparison with measurements made on XR.

Background: In 2021, the World Health Organization (WHO) set sodium benchmarks for packaged foods to guide countries in setting feasible and effective sodium reformulation programs. We modeled the dietary and health impact of full compliance with the WHO’s sodium benchmarks in Australia and compared it to the potential impact of Australia’s 2020 sodium reformulation targets. Methods: We used nationally representative data on food and sodium intake, sodium levels in packaged foods, and food sales volume to estimate sodium intake pre- and post-implementation of the WHO and Australia’s sodium benchmarks for 24 age-sex groups. Using comparative risk assessment models, we then estimated the potential deaths, incidence, and disability-adjusted life years averted from cardiovascular disease, chronic kidney disease, and stomach cancer based on the reductions in sodium intake. Results: Compliance with the WHO’s sodium benchmarks for packaged foods in Australia could lower mean adult sodium intake by 404 mg/day, corresponding to a 12% reduction. This could prevent about 1770 deaths/year (95% uncertainty interval 1168–2587), corresponding to 3% of all cardiovascular disease, chronic kidney disease, and stomach cancer deaths in Australia, and prevent some 6900 (4603–9513) new cases, and 25 700 (17 655–35 796) disability-adjusted life years/year. Compared with Australian targets, the WHO benchmarks will avert around 3 and a half times more deaths each year (1770 versus 510). Conclusions: Substantially greater health impact could be achieved if the Australian government strengthened its current sodium reformulation program by adopting WHO’s more stringent and comprehensive sodium benchmarks.

Purpose: Difficulty understanding speech in noise is a common communication problem. Clinical tests of speech in noise differ considerably from real-world listening and offer patients limited intrinsic motivation to perform well. In order to design a test that captures motivational aspects of real-world communication, this study investigated effects of gamification, or the inclusion of game elements, on a laboratory spatial release from masking test. Method: Fifty-four younger adults with normal hearing completed a traditional laboratory and a gamified test of spatial release from masking in counterbalanced order. Masker level adapted based on performance, with the traditional test ending after 10 reversals and the gamified test ending when participants solved a visual puzzle. Target-to-masker ratio thresholds (TMRs) with colocated maskers, separated maskers, and estimates of spatial release were calculated after the 10th reversal for both tests and from the last six reversals of the adaptive track from the gamified test. Results: Thresholds calculated from the 10th reversal indicated no significant differences between the traditional and gamified tests. A learning effect was observed with spatially separated maskers, such that TMRs were better for the second test than the first, regardless of test order. Thresholds calculated from the last six reversals of the gamified test indicated better TMRs in the separated condition compared to the traditional test. Conclusions: Adding gamified elements to a traditional test of spatial release from masking did not negatively affect test validity or estimates of spatial release. Participants were willing to continue playing the gamified test for an average of 30.2 reversals of the adaptive track. For some listeners, performance in the separated condition continued to improve after the 10th reversal, leading to better TMRs and greater spatial release from masking at the end of the gamified test compared to the traditional test. Differential diagnosis between medulloblastoma (MB), ependymoma (EP) and astrocytoma (PA) is important due to differing medical treatment strategies and predicted survival. The aim of this study was to investigate non-invasive MRI-based radiomic analysis of whole tumors to classify the histologic tumor types of pediatric posterior fossa brain tumor and improve the accuracy of discrimination, using a random forest classifier.MRI images of 99 patients, with 59 MBs, 13 EPs and 27 PAs histologically confirmed by surgery and pathology before treatment, were included in this retrospective study. Registration was performed between the three sequences, and high- throughput features were extracted from manually segmented tumors on MR images of each case. The forest-based feature selection method was adopted to select the top ten significant features. Finally, the results were compared and analyzed according to the classification.The top ten contributions according to the classifier of wavelet features all came from the ADC sequence. The random forest classifier achieved 100% accuracy on the training data and validated the best accuracy (0.938): sensitivity=1.000, 0.948 and 0.808, specificity=0.952, 0.926 and 1.000 for EP, MB and PA, respectively.A random forest classifier based on the ADC sequence of the whole tumor provides more quantitative information than TIWI and T2WI in differentiating pediatric posterior fossa brain tumors. In particular, the histogram percentile value showed great superiority, which added diagnostic value in pediatric neuro-oncology.

Background: The incidence of facial skin cancer increases worldwide, resulting in more surgical resections and reconstructions. Reconstructive surgery aims to approach a normal facial anatomy to optimize the quality of life. Objective automated assessment of the esthetic outcome of facial reconstructions could provide feedback for the improvement of surgical techniques and preoperative patient expectation management. Objective: This systematic literature review aimed to assess whether modern technologies can create automated objective measurements of surgical and non-surgical facial interventions outcomes using 3D surface imaging technology. Methods: A systematic literature search was conducted in Embase, Medline (Ovid), Web of Science, and Cochrane on May 19, 2021. All English literature was collected on surgical and non-surgical invasive facial interventions in which 3D surface imaging technology was used for objective automated assessment of outcomes. Results: Fourteen articles were included in the systematic review. 3D surface imaging technology and automated assessment techniques were found for skin malignancy, cleft lip repair, rhinoplasty, orthognathic surgery, and injectables. Ten 3D surface imaging technology hardware systems and 12 software systems were described. Four studies compared 3D surface imaging techniques to conventional methods. Ten studies used 3D surface imaging techniques for the evaluation of the surgical outcome, without comparison to 2D photography, validated scores, or a panel. Two studies validated the hardware system. Conclusion: This systematic literature review shows that 3D surface imaging technology has the potential for automated objective assessment of facial intervention outcomes. Future studies are necessary to study and validate these tools for standard clinical use in patients with facial interventions.

To evaluate changes in macular blood flow in patients with various stages of diabetic retinopathy (DR) by optical coherence tomography angiography (OCTA).The data of 94 patients (122 eyes) with type 2 diabetes mellitus admitted from January 2018 to May 2021 were recorded, including 31 patients (42 eyes) with no DR (NDR group), 33 patients (40 eyes) with mild non-proliferative DR (mild NPDR group) and 30 patients (40 eyes) with moderate NPDR (moderate NPDR group). Forty healthy patients (40 eyes) who were examined during the same period were selected as control group. The blood flow densities at the superficial capillary plexus (SCP), deep capillary plexus (DCP) and 300μm-wide region near the foveal avascular zone (FAZ) (FD300), FAZ area, FAZ perimeter and acircularity index (AI) were compared. Correlations of blood flow densities at the SCP, DCP and FD300, FAZ area, FAZ perimeter and AI with the severity of DR were investigated by multivariate regression analysis. A receiver operating characteristic (ROC) curve was plotted to analyze the predictive efficacy of the OCTA indices for early DR.Blood flow densities at the SCP, DCP and FD300 significantly decreased, while FAZ perimeter and AI increased in the NDR and DR groups compared with those of the control group (P0.05). Blood flow densities at the SCP, DCP and FD300 of the DR group were lower than those of the NDR group, but the FAZ perimeter and AI of the former were larger (P0.05). Blood flow densities at the SCP, DCP and FD300 in the macular zone of DR patients declined with increasing DR stage, while FAZ perimeter and AI increased (P0.05), without collinearity among the variables. The area under the curve of FD300 for predicting early DR was highest (0.804), with a sensitivity and specificity of 84.13% and 67.19%, respectively. AI with a cut-off value of 1.05 showed the optimal specificity (87.84%) and sensitivity (73.62%) for predicting early DR.The flow density at FD300 declines, while FAZ perimeter and AI increase in the macular zone of patients with early DR. These three OCTA indicators show close associations with DR stage, and can function as predictors of early DR.

Remote testing of auditory function can be transformative to both basic research and hearing healthcare; however, historically, many obstacles have limited remote collection of reliable and valid auditory psychometric data. Here, we report performance on a battery of auditory processing tests using a remotely administered system, Portable Automatic Rapid Testing. We compare a previously reported dataset collected in a laboratory setting with the same measures using uncalibrated, participant-owned devices in remote settings (experiment 1, n = 40) remote with and without calibrated hardware (experiment 2, n = 36) and laboratory with and without calibrated hardware (experiment 3, n = 58). Results were well-matched across datasets and had similar reliability, but overall performance was slightly worse than published norms. Analyses of potential nuisance factors such as environmental noise, distraction, or lack of calibration failed to provide reliable evidence that these factors contributed to the observed variance in performance. These data indicate feasibility of remote testing of suprathreshold auditory processing using participants' own devices. Although the current investigation was limited to young participants without hearing difficulties, its outcomes demonstrate the potential for large-scale, remote hearing testing of more hearing-diverse populations both to advance basic science and to establish the clinical viability of auditory remote testing.

BACKGROUND AND AIMS: Identify novel metabolite associations with blood pressure (BP) salt-sensitivity and hypertension. METHODS AND RESULTS: The Genetic Epidemiology Network of Salt Sensitivity (GenSalt) Replication study includes 698 Chinese participants who underwent a 3-day baseline examination followed by a 7-day low-sodium feeding and 7-day high-sodium feeding. Latent mixture models identified three trajectories of blood pressure (BP) responses to the sodium interventions. We selected 50 most highly salt-sensitive and 50 most salt-resistant participants for untargeted metabolomics profiling. Multivariable adjusted mixed logistic regression models tested the associations of baseline metabolites with BP salt-sensitivity. Multivariable adjusted mixed linear regression models tested the associations of BP salt-sensitivity with metabolite changes during the sodium interventions. Identified metabolites were tested for associations with hypertension among 1,249 Bogalusa Heart Study (BHS) participants using multiple logistic regression. Fifteen salt-sensitivity metabolites were associated with hypertension in the BHS. Baseline values of serine, 2-methylbutyrylcarnitine and isoleucine directly associated with high salt-sensitivity. Among them, serine indirectly associated with hypertension while 2-methylbutyrylcarnitine and isoleucine directly associated with hypertension. Baseline salt-sensitivity status predicted changes in 14 metabolites when switching to low-sodium or high-sodium interventions. Among them, glutamate, 1-carboxyethylvaline, 2-methylbutyrylcarnitine, 3-methoxytyramine sulfate, glucose, alpha-ketoglutarate, hexanoylcarnitine, gamma-glutamylisoleucine, gamma-glutamylleucine, and gamma-glutamylphenylalanine directly associated with hypertension. Conversely, serine, histidine, threonate and 5-methyluridine indirectly associated with hypertension. Together, these metabolites explained an additional 7% of hypertension susceptibility when added to a model including traditional risk factors. CONCLUSIONS: Our findings contribute to the molecular characterization of BP response to sodium and provide novel biological insights into salt-sensitive hypertension.

The most accurate method to measure population salt intake is to collect the complete 24-h urinary sodium excretion (24-h UNa) but it is resource intensive and is rarely carried out frequently. We, therefore, assessed the use of spot urinary sodium concentration to monitor relative changes in population salt intake in between 24-h urine surveys.We used 24-h (n = 2020) and spot urine (n = 21 711) samples drawn from adult participants in separate, cross-sectional, nationally representative surveys in England, repeated between 2006 and 2014.As population average 24-h UNa fell from 2006 to 2014 (from 8.7 to 7.6 g/day, i.e. by 12%) with the ongoing salt reduction programme, spot sodium concentration fell by a similar extent (from 106.1 to 93.1 mmol/l, i.e. by 13%). The regression slopes of 24-h UNa and spot sodium concentration ran parallel (P value = 0.1009) in a linear regression modelling the difference in their year-on-year changes [by regressing the 24-h UNa or sodium concentration values on time, estimation method (24-h versus spot), and their interaction term]. In contrast, when 24-h UNa was estimated by applying the Kawasaki, Tanaka, or INTERSALT formulas to spot sodium concentrations, almost no change was detected from 2006 to 2014 (±1%) and their regression slopes were significantly different from that of the measured 24-h UNa (all P values0.0001).Spot urinary sodium concentration drawn from random and representative samples of the population accurately reflected relative changes in population average 24-h UNa, and can therefore, be used in between 24-h urine surveys to monitor population salt reduction programmes. Formulas commonly used to estimate 24-h UNa were unsuitable to do so.

While there is a wealth of knowledge about core object recognition—our ability to recognize clear, high-contrast object images, how the brain accomplishes object recognition tasks under increased uncertainty remains poorly understood. We investigated the spatiotemporal neural dynamics underlying object recognition under increased uncertainty by combining MEG and 7 Tesla fMRI in humans during a threshold-level object recognition task. We observed an early, parallel rise of recognition-related signals across ventral visual and frontoparietal regions that preceded the emergence of category-related information. Recognition-related signals in ventral visual regions were best explained by a two-state representational format whereby brain activity bifurcated for recognized and unrecognized images. By contrast, recognition-related signals in frontoparietal regions exhibited a reduced representational space for recognized images, yet with sharper category information. These results provide a spatiotemporally resolved view of neural activity supporting object recognition under uncertainty, revealing a pattern distinct from that underlying core object recognition.

X-ray bursts are among the brightest stellar objects frequently observed in the sky by space-based telescopes. A type-I X-ray burst is understood as a violent thermonuclear explosion on the surface of a neutron star, accreting matter from a companion star in a binary system. The bursts are powered by a nuclear reaction sequence known as the rapid proton capture process (rp process), which involves hundreds of exotic neutron-deficient nuclides. At so-called waiting-point nuclides, the process stalls until a slower β+ decay enables a bypass. One of the handful of rp process waiting-point nuclides is 64Ge, which plays a decisive role in matter flow and therefore the produced X-ray flux. Here we report precision measurements of the masses of 63Ge, 64,65As and 66,67Se—the relevant nuclear masses around the waiting-point 64Ge—and use them as inputs for X-ray burst model calculations. We obtain the X-ray burst light curve to constrain the neutron-star compactness, and suggest that the distance to the X-ray burster GS 1826–24 needs to be increased by about 6.5% to match astronomical observations. The nucleosynthesis results affect the thermal structure of accreting neutron stars, which will subsequently modify the calculations of associated observables.

We report on the modulation of interlayer exchange coupling (IEC) between the ferromagnet (FM) Fe and insulating antiferromagnet (AFM) CoO and its impact on the Gilbert damping by a time-resolved magneto-optical Kerr effect technique. By inserting a wedge spin conductor Ag layer at the interface of Fe/CoO, it is revealed that both uniform spin precession frequency and Gilbert damping constant of Fe film decrease with increasing Ag thickness up to ∼2 nm, and above that with eliminated IEC, these two parameters reach the intrinsic values of the Fe film. The precession frequency and damping also show similar variation tendency with temperature for attenuated IEC. These results prove that the exchange coupling at the FM–AFM interface plays an essential role for the spin angular momentum transfer to the insulating AFM layer, and the spin pumping is insignificant even for the very thin spin conductor layer. Our findings may have general insights into the role of the FM/AFM interface in the magnetization dynamics and spin angular momentum transfer for future spintronic applications.

Antiretroviral therapy has effectively suppressed HIV infection and replication and prolonged the lifespan of HIV-infected individuals. In the meantime, various complications including type 2 diabetes associated with long-term antiviral therapy have shown steady increases. Metformin has been the front-line anti-hyperglycemic drug of choice and the most widely prescribed medication for the treatment of type 2 diabetes. However, little is known about the effects of Metformin on HIV infection and replication. In this study, we showed that Metformin treatment enhanced HIV gene expression and transcription in HIV-transfected 293T and HIV-infected Jurkat and human PBMC. Moreover, we demonstrated that Metformin treatment resulted in increased CREB expression and phosphorylation, and TBP expression. Furthermore, we showed that Metformin treatment increased the recruitment of phosphorylated CREB and TBP to the HIV LTR promoter. Lastly, we showed that inhibition of CREB activation significantly abrogated Metformin-enhanced HIV gene expression. Taken together, these results demonstrated that Metformin treatment increased HIV transcription, gene expression, and production though increased CREB phosphorylation and recruitment to the HIV LTR promoter. These findings may help design the clinical management plan and HIV cure strategy of using metformin to treat type 2 diabetes, a comorbidity with an increasing prevalence, in people living with HIV.
