Abstract Cisplatin is an antineoplastic medicine used in the treatment for various types of cancer. Among its side effects is ototoxicity, which may result in a bilateral and irreversible hearing loss. The ototoxic effect in the pediatric population has a bigger impact as it compromises language acquisition. The discovery of drugs with otoprotective effects and the optimal way to administer them have become significant challenges in minimizing the impact of cisplatin regarding auditory function. The objective was to understand otoprotective drugs and their relevance in the preventive treatment to cisplatin-induced ototoxicity in childhood. An integrative review was conducted by consulting databases including PubMed, Bireme, MedLine, LILACS, SciELO, and ClinicalTrials.gov. The search strategy was performed by crossing descriptors (DeCS and MeSH) and free terms. Studies published in English, Spanish, and Portuguese were selected, with no publication year restrictions. Subsequently, articles were selected according to inclusion and exclusion criteria. A total of 736 articles were found in PubMed, 431 in Bireme, 425 in MedLine, 6 in LILACS, 0 in SciELO, and 4 in ClinicalTrials.gov. After document analysis, 12 articles were selected for full analysis. Evidence was found for 8 substances with potential otoprotective effects when used with cisplatin, which tend to minimize the impact of cisplatin regarding auditory function. The substances found were: Amifostine, Dexamethasone, Genistein, Ginkgo Biloba, Lycopene, N-acetylcysteine, Polydatin also Sodium Thiosulfate. In general, these drugs are applied before, during, or after cisplatin infusion, depending on the chosen drug, via intravenous, oral, or transtympanic injections, acting as antioxidant therapy. The biochemical effects of these substances are relevant to their potential otoprotective properties, including the inactivation of oxygen free radicals and electrophilic platinum species. The use of these substances can reduce ototoxicity, decreasing cisplatin-induced hearing loss and improving the confort of life, especially for children.

From 1984 until 1994 there were 7 cases of newborn with esophageal atresia without fistulae, five of which were treated through primary anastomosis and two of these presented exceptional long gaps. This article is an analysis of these 7 newborns, their post-operative complications and the therapeutics used. Two deaths occurred due to associated serious cardiac pathologies, which made delayed primary anastomosis impossible, and one sudden death four months after the esophageal anastomosis. Three of these cases present a good stature-weight progression and one remains slightly under the 5 percentile. Considering our results, the delayed primary anastomosis in the esophageal atresias with long gap, in spite of the post-operative complications, shall continue to be the author's choice.

The use of gold for artefact productions underwent substantial developments during Iron Age in western Europe, with the diversification of alloy compositions and the introduction of new technologies and aesthetics. This work presents a detailed study of a Late Iron Age earring from the western Iberian Peninsula and a discussion taking into consideration the alloys and technologies used at the time. The earring has a complex structure, composed of block- and strip-twisted wires and granulation. Characterization by multifocus optical microscopy, portable X-ray fluorescence spectrometry (pXRF), micro-XRF and scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS) were performed to obtain information on the chemical composition and manufacturing techniques. Results show that the earring was manufactured with wires and granules made of a silver-rich alloy, covered with a gold-rich layer 45 μm thick on average, which acted as an external coating making the entire earring resemble gold. This is a rare example of an Iron Age gilded earring, and its detailed study provides further information on the manufacturing solutions adopted by the Iron Age goldsmiths.

In order to evaluate the influence of different nitrogen rates on the hormetic effect of glyphosate on common bean crop, proposed to study the combination of nitrogen fertilization in succession to low glyphosate doses. The study was conducted under field conditions at the experimental farm of the State University of São Paulo-UNESP, Ilha Solteira campus, located in the city of Selvíria-MS, Brazil. The experimental design was a randomized block with split plots arranged in four blocks. The plots were composed of four glyphosate doses (0, 7.2, 14.4 and 21.6 g of acid equivalent (AE) ha-1) and the subplots were composed of five nitrogen doses (0, 15, 30, 45 and 60 kg ha-1), with urea as the source. Bean growth was stimulated with a maximum point in the 11 g AE ha-1 sub-dose of glyphosate, however, only in the absence of nitrogen under cover. Leaf N content was higher at the estimated 12 and 11 g AE ha-1 glyphosate doses with 30 and 45 kg ha-1 nitrogen. Hormetic effect on yield was obtained with increments of up to 500 kg ha-1 or 21% at the estimated 10 g AE ha-1 low glyphsate dose. Cooking time was affected by low glyphosate doses, depending on the amount of nitrogen. It was possible to prove the influence of nitrogen on glyphosate hormesis in common bean for dry matter mass, leaf N content, grain yield and cooking time.

The cervical spine is a common site of injury in the vertebral column, with severe injuries often associated with damage to the spinal cord. Several studies have been performed to better understand the mechanisms of such situations and develop ways to treat or even prevent them. Among the most advantageous and most widely used methods are computational models, as they offer unique features such as providing information on strains and stresses that would otherwise be difficult to obtain. Therefore, the main objective of this work is to help better understand the mechanics of the neck by creating a new finite element model of the human cervical spine that accurately represents most of its components. The initial geometry of the cervical spine was obtained using the computer tomography scans of a 46-year-old female. The complete model was then sectioned, and a functional spinal unit consisting of the C6–C7 segment was simulated to initiate the validation process. The reduced model was validated against experimental data obtained from in vitro tests that evaluated the range of motion of various cervical segments in terms of flexion–extension, axial rotation, and lateral bending.

Abstract Hymenaea martiana is a species popularly known in Northeastern Brazil as “jatobá” and used in folk medicine to treat pain and inflammation. The aim of this work was to evaluate the antinociceptive and anti-inflammatory activity of H. martiana. In the present study, we carried out an investigation about the effects of the crude ethanolic extract (Hm-EtOH) and the ethyl acetate fraction (Hm-AcOEt) in models of nociception and inflammation in mice. Chemical (acetic acid-induced writhing and formalin) and thermal stimuli (hot plate) were used for the evaluation of antinociceptive activity, while for the anti-inflammatory profile paw edema induced by carrageenan was used, along with leukocyte migration to the peritoneal cavity. The presence of the flavonoid astilbin in the samples was characterized through HPLC-DAD-MS analysis. Hm-EtOH and Hm-AcOEt (100, 200 and 400 mg.kg-1, i.p.) significantly reduced the number of abdominal contortions and decreased the paw licking time in the formalin test. In the hot plate, the extract increased the latency time of animals. Hm-EtOH and Hm-AcOEt inhibited significantly the increase in the edema after the administration of carrageenan. Hm-EtOH and Hm-AcOEt inhibited leukocyte migration in the peritonitis test. HPLC-DAD-MS analysis of Hm-EtOH and Hm-AcOEt revealed the presence of the flavonoid astilbin in the samples. According to the results of this study, both Hm-EtOH and Hm-AcOEt have antinociceptive and anti-inflammatory activities, which could be related with the presence of flavonoid in the extracts. The results reinforce the popular use of this plant.

European silver ,Potosí silver ,argentiferous copper ,PIXE ,EDXRF ,Archaeology ,CC1-960 - Abstract
The circulation trading routes and the characterization of the silver metal used in the European continent in the 15⁻17th centuries are historical issues that are still open. This study aimed to bring an insight into the silver processed within a chronological framework in the Portuguese territory, relating the analytical data with the known historical information. This investigation developed on 230 high silver coins from two important Portuguese coin collections was based mainly on surface particle-induced X-ray emission (PIXE) analysis, complemented with a few energy-dispersive X-ray fluorescence (EDXRF) analyses. The silver processed in different timelines was discriminated based on the variation of the impurity contents, namely Au and Bi. European silver with high Au and Hg and low Pb and Bi contents supplied the 15th century chronologies, being replaced at the dawn of the 16th century by a new metal entering the Portuguese capital. This new metal, with low Au and high Bi contents, was probably derived from European argentiferous copper ores. By the end of the 1500s, the Philippine chronologies reveal the newly discovered Potosí silver, identified for the first time based on PIXE minor and trace element surface contents, distinguishable from the European silver in use until 1578 in the Portuguese territory, by Au contents

Abstract Selaginella convoluta (Arn.) Spring is a species popularly known as “jericó”, and used in folk medicine as analgesic and anti-inflammatory. This study aimed to investigate in mice the antinociceptive and anti-inflammatory activities of the hexane (Sc-Hex) and chloroform (Sc-CHCl3) fractions (100, 200 and 400 mg/kg) obtained by partition of crude ethanol extract from S. convoluta. The preliminary phytochemical analysis of the fractions was performed. Antinociceptive activity was evaluated by writhing, formalin and hot-plate tests. Anti-inflammatory activity was evaluated using carrageenan-induced pleurisy. The rota-rod test was used to evaluate motor coordination. Preliminary phytochemical screening showed that the Sc-Hex and the Sc-CHCl3 were positive for the presence of flavonoids, anthracene derivatives, quinones, triterpenes and steroids. Inhibition of writhing was observed for fractions tested. The Sc-Hex at all doses tested was effective in reducing the nociceptive behavior produced by formalin only in the second phase. However, the Sc-CHCl3 decreased the paw licking time in the first and second phases. In the hot plate no significant effect was observed for any fraction. In the rota-rod test, treated mice did not demonstrate any significant motor performance changes. In the carrageenan-induced pleurisy, Sc-CHCl3 (200 mg/kg) reduced cell migration to the pleural cavity. These results reveal the antinociceptive properties of S. convoluta , which support, in part, its traditional use, since the fractions did not presented significant activity in the inflammatory response profile. We further verify that this antinociceptive effect could be by activation of nociceptive peripheral pathway.

Wastewater treatment plants (WWTPs) are significant reservoirs of bacterial resistance. This work aims to identify the determinants of resistance produced by Gram-negative bacteria in the influent and effluent of two WWTPs in Portugal. A total of 96 wastewater samples were obtained between 2016 and 2019. The numbers of total aerobic and fecal contamination bacteria were evaluated, and genomic features were searched by polymerase chain reaction (PCR) and Next-Generation Sequencing (NGS). Enterobacteriaceae corresponded to 78.6% (n = 161) of the 205 isolates identified by 16sRNA. The most frequent isolates were Escherichia spp. (57.1%, n = 117), followed by Aeromonas spp. (16.1%, n = 33) and Klebsiella spp. (12.7%, n = 26). The remaining 29 isolates (14.1%) were distributed across 10 different genera. Among the 183 resistant genes detected, 54 isolates produced extended spectrum β-lactamases (ESBL), of which blaCTX-M-15 was predominant (37 isolates; 68.5%). A KPC-3 carbapenemase-producing K. oxytoca was identified (n = 1), with blaKPC-3 included in a transposon Tn4401 isoform b. A higher number of virulence genes (VG) (19 genes) was found in the E. coli 5301 (O25b-ST131-B2) isolate compared with a commensal E. coli 5281 (O25b-ST410-A) (six genes). Both shared five VG [Enterobactin; Aerobactin, CFA/1 (clade α); Type1 (clade γ1); Type IV]. In conclusion, this work highlights the role of relevant clinical bacteria in WWTPs, such as KPC-3-producing K. oxytoca, and, for the first time, a CTX-M-15-producing Ochromobactrum intermedium, a human opportunistic pathogen, and a SED-1-producing Citrobacter farmeri, an uncommon CTX-M-type extended-spectrum beta-lactamase.

Terrestrial plants, due to their sessile nature, are highly exposed to environmental pressure and therefore need to produce very effective molecules that enable them to survive all the threats. Myrica and Morella (Myricaceae) are taxonomically close genera, which include species of trees or shrubs with edible fruits that exhibit relevant uses in traditional medicine. For instance, in Chinese or Japanese folk medicine, they are used to treat diarrhea, digestive problems, headache, burns, and skin diseases. A wide array of compounds isolated from different parts of Myrica and/or Morella species possess several biological activities, like anticancer, antidiabetic, anti-obesity, and cardio-/neuro-/hepatoprotective activities, both in vitro and in vivo, with myricanol, myricitrin, quercitrin, and betulin being the most promising. There are still many other compounds isolated from both genera whose biological activities have not been evaluated, which represents an excellent opportunity to discover new applications for those compounds and valorize Morella/Myrica species.

Cardiology and Cardiovascular Medicine - Abstract
Background Acute total occlusion of the unprotected left main coronary artery (ATOLMCA) is a dramatic entity with very high mortality. Owing to its infrequency, there is limited and inconsistent data regarding this population. Purpose To describe the clinical presentation, short- and long-term outcomes of patients with ATOLMCA. Methods This retrospective multicentric cohort study included all patients presenting with acute ( Results In the period of the study, 11,036 emergent coronary angiographies were performed in the participating centers, 59 of which were ATOLMCA (0.5%). Mean age of patients at the time of the event was 61.2 (±12.2) years. Seventy-three percent were male. At presentation, 72.9% of patients were in cardiogenic shock, and aborted cardiac arrest occurred in 27.1%. Right dominance was present in all patients except one, who had a balanced dominance. Primary percutaneous coronary intervention (PCI) was performed in 89.8% of the patients, with angiographic success being achieved in 55.6% of the procedures. Overall, the in-hospital mortality rate was 57.6%. Mortality was significantly higher in patients without angiographic criteria for PCI success (87.5 vs 36.7%, p Conclusion Patients with ATOLMCA have a dismal prognosis. Most patients present with cardiogenic shock, and a significant number develop cardiac arrest during the acute phase. Despite medical care, in-hospital mortality is high. Patients with left dominance may not even reach the hospital. Among survivors, long term outcomes are reasonable. Further studies are needed in order to improve the management and outcomes of patients with ATOLMCA. Funding Acknowledgement Type of funding sources: None.

Cardiology and Cardiovascular Medicine - Abstract
Background Percutaneous valve commissurotomy (PMC) is a viable alternative to mitral valve (MV) surgery in the treatment of patients with clinically significant mitral stenosis (MS). About 40% of patients treated with PMC will require at least one reintervention (either PMC or MVS) along time. Purpose To evaluate the long-term results of PMC in patients with rheumatic MS. Methods We retrospectively analysed all consecutive patients between 1991 and 2008 with clinically significant rheumatic MS undergoing PMC. Clinical and echocardiographic data were collected at baseline and during early and long-term follow-up. MACE was a composite of adverse events defined as all-cause mortality, MV re-intervention or cardiovascular hospitalization. Results A total of 124 patients were enrolled: 108 (87%) were female, with a mean age at the time of PMC of 46±11 years. At baseline, 34% patients were in NYHA class ≥ III and 81% had a Wilkins score ≤8; all patients had preserved biventricular systolic function and 83% presented pulmonary hypertension. Regarding associated valve disease, 46 patients had mild tricuspid regurgitation (TR), 19 mild aortic regurgitation (AR), 14 moderate IT and 5 moderated AR. Most of the procedures were successful (91%) and without complications (94%), with median improvement in MV area of 0.9 cm2 (IQR 0.5) and median reductions in mean transmitral gradient (MTG) of 6 mmHg (IQR 6) and in pulmonary artery systolic pressure (PASP) of 8 mmHg (IQR 10) early after PMC. During the mean follow-up of 20±6 years, 52 (42%) of patients had MV re-intervention (86% surgery and 14% re-PMC), 37 (30%) were hospitalized and 30 (24%) died. Concerning time-to-event analysis, approximately 80% of patients kept MACE-free after 10 years; after 30 years, more than 20% continued MACE-uneventful, approximately 50% were alive and about 45% were free from re-intervention. Considering patients submitted to surgical re-intervention, 9 underwent MV valvuloplasty and the others MV replacement with mechanical (32) or biological prothesis (11). At the same procedure, 23 patients were submitted to tricuspid annuloplasty, 9 to other valve replacement and one to coronary artery bypass graft. Using Cox regression, we found that the presence of moderate disease of other valves at PMC time was associated with a 2.3-fold greater rate of re-intervention compared to patients with none or mid disease of other valves (HRcrude 2.3; 95% IC 1.221–4.331, p=0.017). After adjusting for the success of the PMC and for mitral regurgitation after PMC, the observed effect remained significant (HRadjusted = 2.7; 95% CI 1.417–5.233, p=0.003). Conclusion PMC was safe and effective in clinically significant rheumatic MS. Most of the patients were free from adverse events after 10 years and half were alive after 30 years. Still, about 40% required re-intervention, with moderate disease of other valves as its independent predictor. Funding Acknowledgement Type of funding sources: None.

Cardiology and Cardiovascular Medicine - Abstract
Background Percutaneous valve commissurotomy (PMC) is an established treatment in patients with significative mitral stenosis (MS). Although rheumatic MS incidence has decreased in the last century, it remains a prevalent pathology worldwide. The Wilkins score (WS) is a reference in echocardiographic assessment of MS; a score ≤8 is considered a predictor of treatment success and score between 9 and 11 is a “grey zone” (WGZ) in which doubts persists regarding PMC success. Purpose To evaluate the early and long-term results of PMC in patients with rheumatic MS and to compare long-term events between patients with WS ≤8 and patients in WGZ. Methods We retrospectively analysed all patients between 1991 and 2008 with significative rheumatic MS undergoing PMC. Data were collected at baseline and during long-term follow-up. M ACE was defined as a composite of all-cause mortality, mitral valve re-intervention or cardiovascular hospitalization. Results In our cohort, 124 patients were included. Most were female (87%), mean age at the time of repair was 46±11 year-old and mean follow-up was 20±6 years. Before the procedure, 81% had WS ≤8 and 19% were in WGZ. Both groups had similar baseline characteristics, namely age at first intervention, NYHA class and follow-up time. All patients had preserved biventricular systolic function, 83% presented PH, mean transvalvular gradient (TVG) and mitral valve area (MVA) were 12.8 mmHg and 1.0 cm2, respectively. Most of the procedures were successful (91%) and without complications (94%). Mean MVA improvement was similar in both groups [0.9 cm2 in WS ≤8 and 0.8 cm2 in WGZ, t(102)=0.173, p=0.863]; there was also no significative difference in TVG and PASP reduction after PMC. During long-term follow-up, re-intervention and mortality occurred in 40% and 23% in WS ≤8 and in 50% and 29% in WGZ, respectively, and none of these differences was statistically significant (p=0.389 and p=0.544, respectively). Concerning time-to-event analysis, approximately 80% of patients kept uneventful and >90% alive after 10 years in both groups and no significant difference in M ACE events and all-cause mortality between WS ≤8 and WGZ was observed (Log Rank, p=0,419 and p=0.950, respectively). Conclusion PMC was safe and effective in clinically significant rheumatic MS in both WS ≤8 and WS 9–11, with similar MVA improvement. After 10 years, approximately 80% of patients were MACE-free and >90% alive in both groups. There was no difference in all-cause mortality and in a composite of all-cause death, mitral valve re-intervention or cardiovascular hospitalization concerning WS groups. Cardiology and Cardiovascular Medicine - Abstract
Background Percutaneous valve commissurotomy (PMC) is a viable alternative to mitral valve surgery in the treatment of patients with clinically significant mitral stenosis (MS). Although rheumatic MS incidence has decreased in developed countries, it remains a prevalent healthcare problem in Cardiology clinics Purpose To evaluate the early and long-term results of PMC in patients with rheumatic MS and to compare long-term events between patients with and without pulmonary hypertension (PH). Methods We retrospectively analysed all consecutive patients between 1991 and 2008 with clinically significant rheumatic MS undergoing PMC. Clinical and echocardiographic data were collected at baseline and during long-term follow-up. MACE was a composite of adverse events defined as all-cause mortality, mitral valve re-intervention or hospitalization for a cardiovascular cause. Results A total of 124 patients were enrolled: 87% were female, with a mean age at the time of repair of 46±11 year-old and a mean follow-up of 20±6 years. Before the procedure, 34% were in NYHA class ≥ III and 81% had a Wilkins score ≤8; all patients had preserved biventricular systolic function, 83% presented PH, mean transvalvular gradient (TVG) and mitral valve area (MVA) were 12.8 mmHg and 1.0 cm2, respectively. Most of the procedures were successful (91%) and without complications (94%), with a mean MVA improvement of 0.9 cm2 and reduction of 8.5 mmHg in TVG and 9.7 mmHg in pulmonary artery systolic pressure (PASP) after PMC. During long-term follow-up, 42% of patients were submitted to re-intervention (most of them surgically) and 24% died. In patients non-submitted to re-intervention, TVG and PASP remained similar with early post-procedure evaluation (p=0.109 and p=0.777, respectively), while MVA reduced over time, yet still statistically superior to baseline MVA (1.6 cm2 vs 1.0 cm2, p Conclusion PMC was safe and effective in clinically significant rheumatic MS. After a long-term follow-up patients maintained the reduction in TVG and PASP and a smaller but significative improvement in MVA. Most of the patients were free from adverse events after 10 years and half were alive after 30 years. There was no difference in all-cause mortality and in a composite of all-cause death, mitral valve re-intervention or cardiovascular hospitalization concerning PH presence. Osteochondral (OC) defects combine damage to cartilage and subchondral bone, posing a significant challenge to their repair due to the dissimilar characteristics and regenerative capabilities between the two tissues. Here, we propose novel OC bilayer composites, drawing inspiration from corresponding biological tissues and using a combination of simple and reproducible techniques. Cartilage-like materials based on poly(vinyl alcohol) (PVA) hydrogels were produced with nanofiber reinforcements acquired from high-performance fibers (Kevlar® and Zylon®), while bone-like materials were obtained by adding magnesium-substituted calcium phosphate ceramics to PVA. All composites were sterilized by gamma irradiation to rule out the possibility of undesirable effects resulting from the process, and then fully characterized. The results indicated that nanofibers and bioceramics incorporated into the PVA networks form promising structures with multiple interesting properties. The composites resembling cartilage and bone showed high biomimicry with natural tissues, being able to reconcile exceptional mechanics with the requirements of adequate porosity, liquid content, and biological behavior. The developed materials reveal a high potential for use in OC tissue repair applications.

Detectors and Experimental Techniques - Abstract
The CMS Detector will be upgraded for the High-Luminosity LHC to include a MIP Timing Detector (MTD). The MTD will consist of barrel and endcap timing layers, BTL and ETL, respectively, providing precision timing of charged particles. The BTL sensors are based on LYSO Ce scintillating crystals coupled to SiPMs that are read out by TOFHIR2 ASICs in the front-end system. A resolution of 30 ps for MIP signals is expected at the beginning of HL-LHC operation degrading to 60 ps at the end of operation due to the SiPMs radiation damage. Relative to the first version of the front-end ASIC, TOFHIR2X implements improved circuitry for mitigation of the SiPM dark current noise as well as a new current mode discriminator. We present an overview of the TOFHIR2 requirements and design, simulation results and the first measurements with TOFHIR2X silicon samples coupled to LYSO/SiPM prototype sensors.

Gastroenterology ,General Medicine - Abstract
Background Intravenous steroids (IV-S) remain the cornerstone of treatment of Acute Severe Ulcerative Colitis (ASUC). However, one-third of these patients are refractory to this treatment. Several indices have been developed to predict the failure of IV-S for ASUC, using clinical data at day 3 of treatment. However, indices able to predict IV-S failure based on admission data are lacking. Methods A retrospective analysis of all consecutive hospital admissions due to ASUC, in a Portuguese tertiary centre, between February 2007 and April 2021 was performed. Only patients receiving IV-S for at least 3 consecutive days were included in this study. IV-S non-response was defined as the need for a second-line therapy or surgery. The following clinical data was collected from each patient: age, gender, disease duration, Montreal disease extension, previous ASUC, previous and on admission medical therapies (oral mesalazine, rectal mesalazine, oral steroids, immunosuppressants and monoclonal antibodies), fever on admission and weeks of symptoms’ worsening before admission. The following parameters on admission were also registered: C-reactive protein, albumin level, hemoglobin, leucocytes count, platelets count, Mayo endoscopic subscore and presence of transverse colonic dilatation on abdominal X-ray ≥5.5 cm. Univariate analysis was used to identify predictive factors of IV-S failure. Results 68 patients (64.7% female), with a mean age of 40.4 (±16.2) years, were included in this study. 9 patients (13.2%) did not respond to IV-S therapy. On univariate analysis, only transverse colonic distension on X-ray was more frequent in IV-S non-responders (33.3% vs 6.8%, p=0.04). Conclusion Clinical data on admission, including previous medication, analytical parameters and endoscopic activity were not predictive of IV-S refractoriness in this population. However, abdominal X-ray showed to be useful to predict the need for a rescue therapy. These findings highlight, in one hand, the need of larger studies to identify on admission predictors of IV-S refractoriness and, in other hand, the importance of abdominal X-ray, in patients admitted due to ASUC.
