Plum production has good potential for commercial growers and homeowners in Florida and other mild winter areas throughout the Gulf coast. However, most seasonal plums on the market are from California and will not perform well enough in Florida to produce fruit. From 1998 to 2001, plum cultivars have been released from the University of Florida's breeding program that adapt to our mild winters and high disease pressures. Because they have cropped reliably and produced high quality fruit, these cultivars are recommended for trial in Florida. The names of all University of Florida plum cultivars begin with the prefix 'Gulf.' These cultivars are Japanese type plums (Prunus salicina Lindl.) and have resistance to plum leaf scald (Xylella fastidiosa) and bacterial spot (Xanthomonas campestris). Fruit size is satisfactory (about 1 1/2 to 2 in. diameter) and fruit quality is good. They ripen in early to late May or about two weeks before California plums. The flexible acquisition of substrates from nutrient pools is critical for microbes to prevail in competitive environments. To acquire glucose from diverse glycoside and disaccharide substrates, many free-living and symbiotic bacteria have developed, alongside hydrolysis, a non-hydrolytic pathway comprised of four biochemical steps and conferred from a single glycoside utilization gene locus (GUL). Mechanistically, this pathway integrates within the framework of oxidation and reduction at the glucosyl/glucose C3, the eliminative cleavage of the glycosidic bond and the addition of water in two consecutive lyase-catalyzed reactions. Here, based on study of enzymes from the phytopathogen Agrobacterium tumefaciens, we reveal a conserved Mn 2+ metallocenter active site in both lyases and identify the structural requirements for specific catalysis to elimination of 3-keto-glucosides and water addition to the resulting 2-hydroxy-3-keto-glycal product, yielding 3-keto-glucose. Extending our search of GUL-encoded putative lyases to the human gut commensal Bacteroides thetaiotaomicron, we discover a Ca 2+ metallocenter active site in a putative glycoside hydrolase-like protein and demonstrate its catalytic function in the eliminative cleavage of 3-keto-glucosides of opposite (α) anomeric configuration as preferred by the A. tumefaciens enzyme (β). Structural and biochemical comparisons reveal the molecular-mechanistic origin of 3-keto-glucoside lyase stereo-complementarity. Our findings identify a basic set of GUL-encoded lyases for glucoside metabolism and assign physiological significance to GUL genetic diversity in the bacterial domain of life., (© 2024 The Authors. Objectives: Internationally, corticosteroids are still the mainstay treatment for patients with idiopathic sudden sensorineural hearing loss (ISSHL). This is a retrospective monocentric study investing the impact of adding N-acetylcysteine (NAC) to prednisolone treatment on patients with ISSHL at a tertiary university otorhinolaryngology department., Methods: 793 patients (median age 60 years; 50.9% women) with a new diagnosis of ISSHL from 2009 to 2015 were included in the study. 663 patients received NAC administration in addition to standard tapered prednisolone treatment. Univariate and multivariable analysis were performed to identify independent factors regarding negative prognosis of hearing recovery., Results: Mean initial ISSHL and hearing gain after treatment in 10-tone pure tone audiometry (PTA) were 54.8 ± 34.5 dB and 15.2 ± 21.2 dB, respectively. In univariate analysis, treatment with prednisolone and NAC was associated with a positive prognosis of hearing recovery in the Japan classification in 10-tone PTA. In multivariable analysis on Japan classification in 10-tone PTA including all significant factors from univariate analysis, negative prognosis of hearing recovery were age > median (odds ratio [OR] 1.648; 95% confidence interval [CI] 1.139-2.385; p = 0.008), diseased opposite ear (OR 3.049; CI 2.157-4.310; p < 0.001), pantonal ISSHL (OR 1.891; CI 1.309-2.732; p = 0.001) and prednisolone alone without NAC treatment (OR 1.862; CI 1.200-2.887; p = 0.005)., Conclusions: Prednisolone treatment combined with NAC resulted in better hearing outcomes in patients with ISSHL than treatment without NAC., (© 2023. Biological degradation of natural product glycosides involves, alongside hydrolysis, β-elimination for glycosidic bond cleavage. Here, we discover an O-glycoside β-eliminase (OGE) from Agrobacterium tumefaciens that converts the C3-oxidized O-β-D-glucoside of phloretin (a plant-derived flavonoid) into the aglycone and the 2-hydroxy-3-keto-glycal elimination product. While unrelated in sequence, OGE is structurally homologous to, and shows effectively the same Mn 2+ active site as, the C-glycoside deglycosylating enzyme (CGE) from a human intestinal bacterium implicated in β-elimination of 3-keto C-β-D-glucosides. We show that CGE catalyzes β-elimination of 3-keto O- and C-β-D-glucosides while OGE is specific for the O-glycoside substrate. Substrate comparisons and mutagenesis for CGE uncover positioning of aglycone for protonic assistance by the enzyme as critically important for C-glycoside cleavage. Collectively, our study suggests convergent evolution of active site for β-elimination of 3-keto O-β-D-glucosides. Background: Adequate staffing is crucial for high quality patient care and nurses' wellbeing. Nurses' professional assessment on adequacy of staffing is the gold standard in measuring staffing adequacy. However, available measurement instruments lack reliability and validity., Objectives: To develop and psychometrically test an instrument to measure nurses' perceived adequacy of staffing (PAS) of general hospital wards., Design: A multicenter cross-sectional psychometric instrument development and validation study using item response theory., Settings: Ten general nursing wards in three teaching hospitals in the Netherlands., Participants: A sample of 881 participants, including third and fourth year nursing students and nurse/care assistants., Methods: The 13-item self-reported questionnaire was developed based on a previous Delphi study and two focus groups. We interviewed five nurses to evaluate the content validity of the instrument. The field test for psychometric evaluation was conducted on ten general wards of three teaching hospitals in the Netherlands. Structural validity of the item bank was examined by fitting a graded response model (GRM) and inspecting GRM fit. Measurement invariance was assessed by evaluating differential item functioning (DIF) of education level and work experience. We examined convergent validity by testing the hypothesis that there was at least a moderate correlation between the PAS instrument and a single item measuring adequacy of staffing. Internal consistency and reliability across the scale were also examined., Results: A total of 881 measurements were included in the analysis. The data fitted the GRM adequately and item fit statistics were good. DIF was detected for work experience for the protocols item, but the impact on total scores was negligible. The hypothesis was confirmed and the item bank reliably measured two standard deviations around the mean., Conclusions: The Nurse Perceived Adequacy of Staffing Scale (NPASS) for nurses of general hospital wards in the Netherlands has sufficient reliability and validity and is ready for use in nurse staffing research and practice., Competing Interests: None., (© 2023 Published by Elsevier Ltd.)

This work simulates the production of methyl crotonate from various industrial wastewaters. In the upstream process, wastewater is fermented into volatile fatty acids which are then converted into polyhydroxyalkanoates (PHA) by means of mixed microbial cultures. In the downstream, PHA undergoes a series of thermolysis and esterification reactions to produce methyl crotonate. The origin of the wastewater was found to have a great influence on the composition of the PHA with the effluent of a candy bar factory producing a high polyhydroxybutyrate/polyhydroxyvalerate ratio of 86/14 in favour of methyl crotonate production. It was observed that the use of intracellular polyhydroxybutyrate, instead of purified, significantly lowers the number of separation steps and yet reduces the methyl crotonate recovery by only 20 %. An operating pressure higher than 18 bar led to more transesterification of polyhydroxybutyrate, producing byproducts instead of methyl crotonate. Operational processes in the care sector are becoming increasingly digitalized due to rising workload. Insufficient involvement of caregivers in this digitalization process results in poorly integrated technological developments along with a lack of technology acceptance. Therefore, in order to lay a firm foundation for successful technology development, it is utmost essential to understand the intention of professionals to use technology in outpatient care. This paper develops a model that outlines the factors influencing the intention of caregivers to use digital technologies in outpatient care, based on a case study examining the introduction of a textile sensor mat. In the developed model, the care situation is investigated for the first time as a factor influencing the intention to use digital technologies, in addition to technology acceptance. Apart from deriving the factors of the model and inspecting the strength of their influences, the practical relevance for technology developers in other care contexts is also determined. Practical Relevance : Demographic change in the German population is leading to major challenges in various sectors. Specifically, in the outpatient care sector, which is already severely affected by a shortage of skilled workers, this change is particularly noticeable with an increase in patients and a high workload for caregivers. Therefore, digital technologies are increasingly being used to make the daily work of caregivers easier in terms of physical and psychological factors. Background: Poor inter-professional collaboration might negatively influence adequate planning of operative procedures. Interventions capable of improving inter-professional collaboration will positively impact professional practice and health care outcomes. Radboud University Medical Center (UMC) redesigned their operating room (OR) scheduling method by implementing cross-functional teams (CFTs). In this center, positive effects of CFTs were already demonstrated in a mono-center study. This study aims to confirm these effects by comparing the Radboud data with data from 6 other similar centers using a nationwide OR benchmark collaborative., Study Design: The effect of CFTs was measured by the performance indicator "raw utilization." The Kruskal-Wallis one-way ANOVA was applied to compare OR performance among all 7 centers. The Wilcoxon-Mann-Whitney test was used to determine differences in OR performance between Radboud UMC and the control group., Results: Operating room performance differed significantly among all 7 centers (p<0.0005). Radboud UMC demonstrated the highest median raw utilization of 94% vs 85% in the control group (p<0.0005). Box-and-whisker plots validated the reduced variation during the years, indicating an organizational learning effect. Therefore, not only a better performance than the control group, but also a gradual improvement of this performance during the years., Conclusions: This study shows that multidisciplinary collaboration in CFTs during the perioperative phase has a positive influence on OR scheduling and use of OR time. Other national databases considering mortality rates also support the idea that introducing CFTs is not only an important condition for improving OR performance, but also for improving quality of care., (Copyright © 2015 American College of Surgeons. Purpose: The purpose of this paper is to present the effect of the introduction of cross-functional team (CFT)-based organization, rather than, on planning and performance of OR teams., Design/methodology/approach: In total, two surgical departments of the Radboud University Nijmegen Medical Center (RUNMC) in the Netherlands were selected to illustrate the effect on performance. Data were available for a total of seven consecutive years from 2005 until 2012 and consisted of 4,046 OR days for surgical Department A and 1,154 OR days for surgical Department B on which, respectively 8,419 and 5,295 surgical cases were performed. The performance indicator "raw utilization" of the two surgical Departments was presented as box-and-whisker plots per year (2005-2011). The relationship between raw utilization (y) and years (x) was analyzed with linear regression analysis, to observe if performance changed over time., Findings: Based on the linear regression analysis, raw utilization of surgical Department A showed a statistically significant increase since 2006. The variation in raw utilization reduced from IQR 33 percent in 2005 to IQR 8 percent in 2011. Surgical Department B showed that raw utilization increased since 2005. The variation in raw utilization reduced from IQR 21 percent in 2005 to IQR 8 percent in 2011., Social Implications: Hospitals need to improve their productivity and efficiency in response to higher societal demands and rapidly escalating costs. The RUNMC increased their OR performance significantly by introduction of CFT-based organization in the operative process and abandoning the so called functional silos., Originality/value: The stepwise reduction of variation - a decrease of IQR during the years - indicates an organizational learning effect. This study demonstrates that introducing CFTs improve OR performance by working together as a team.
