Abstract This data descriptor presents a curated dataset for pathogen detection and identification (Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans) directly from whole-blood samples. The dataset was created using differential cell lysis combined with rapid extraction, digestion, and mass spectrometry-based proteomics. Our method offers a rapid diagnostic alternative to traditional culture, enabling timely disease management, such as sepsis. Highlighting our dataset’s uniqueness, it features a three-tier structure: Spectral Libraries of Pathogens for identifying peptide peaks for putative biomarkers; Spiked pathogen in blood MS data for biomarker panel optimization through varied concentration samples; and Parallel Reaction Monitoring (PRM) data from sepsis patients for validating our biomarker panel, achieving 83.3% sensitivity within seven hours without microbial enrichment culture. This dataset serves as a comprehensive reference for bioinformatic tool development and biomarker panel proposals, advancing microbial detection, antimicrobial resistance, and epidemiological studies.

ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others.

Introdução: As Doenças Cardiovasculares (DCV) lideram o número de óbitos entre homens e mulheres. Contudo, em mulheres os prognósticos são piores, sugerindo haver fisiopatologia diferente para doenças e risco cardiovascular entre os sexos, ainda assim, mulheres são subdiagnosticadas e sub representadas em estudos sobre DCV. Objetivo: Avaliar o risco cardiovascular em mulheres em idade adulta em uma cidade da Zona da Mata de Pernambuco. Materiais e Métodos: Estudo transversal, realizado com mulheres adultas no qual foram investigadas variáveis socioeconômicas, antropométricas e de predição do risco cardiovascular. As mulheres foram subdivididas em grupos: eutrofia, com sobrepeso e obesidade conforme o índice de massa corporal. A coleta de dados ocorreu entre abril e dezembro de 2022. As análises estatísticas foram realizadas, pelo programa SPSS, sendo realizados os testes: Anova one-way, Kruskal - Wallis e correlação de Pearson, para dados paramétricos, não paramétricos e correlação, respectivamente. Resultados: Foram avaliadas 54 mulheres das quais 25,9% foram classificadas como eutróficas, 31,5% com sobrepeso e 42,6% com obesidade. O perfil socioeconômico foi semelhante entre os grupos, contudo, o grupo com obesidade apresentou índices elevados de: pressão arterial sistólica e diastólica; percentual de gordura corporal e maior risco cardiovascular. Também foram encontradas correlações entre o aumento da RCQ e o Escore de Framingham. Conclusões: Nossos achados descrevem correlações entre excesso de peso e presença de fatores de risco cardiovasculares modificáveis, podendo inferir que a redução do peso corporal contribua para a diminuição das doenças cardiovasculares.

Objetivo: delinear o perfil epidemiológico de pacientes vítimas de queimadura, que tenham sido submetidos à internação no Centro de tratamento de queimados de um hospital do Rio de Janeiro, no período de 2019 a 2021. Método: trata-se de uma pesquisa observacional, exploratória e descritiva, de abordagem quantitativa, documental e retrospectiva, realizada em um Centro de Tratamento de Queimados (CTQ), de um Hospital localizado no Rio de Janeiro. Abstract This study evaluated the peri-implant tissues under normal conditions and under the influence of experimental peri-implantitis (EPI) in osseointegrated implants installed in the maxillae of rats treated with oncologic dosage of zoledronate. Twenty-eight senescent female rats underwent the extraction of the upper incisor and placement of a titanium dental implant (DI). After eight weeks was installated a transmucosal healing screw on DI. After nine weeks, the following groups were formed: VEH, ZOL, VEH-EPI and ZOL-EPI. From the 9th until the 19th, VEH and VEH-EPI groups received vehicle and ZOL and ZOL-EPI groups received zoledronate. At the 14th week, a cotton ligature was installed around the DI in VEH-EPI and ZOL-EPI groups to induce the EPI. At the 19th week, euthanasia was performed, and the maxillae were processed so that at the implanted sites were analyzed: histological aspects and the percentage of total bone tissue (PTBT) and non-vital bone tissue (PNVBT), along with TNFα, IL-1β, VEGF, OCN and TRAP immunolabeling. ZOL group presented mild persistent peri-implant inflammation, higher PNVBT and TNFα and IL-1β immunolabeling, but lower for VEGF, OCN and TRAP in comparison with VEH group. ZOL-EPI group exhibited exuberant peri-implant inflammation, higher PNVBT and TNFα and IL-1β immunolabeling when compared with ZOL and VEH-EPI groups. Zoledronate disrupted peri-implant environment, causing mild persistent inflammation and increasing the quantity of non-vital bone tissue. Besides, associated with the EPI there were an exacerbated inflammation and even greater increase in the quantity of non-vital bone around the DI, which makes this condition a risk factor for medication-related osteonecrosis of the jaws.

Background: Hydroxyapatite (HA)-based ceramics are widely used as artificial bone substitutes due to their advantageous biological properties, which include biocompatibility, biological affinity, bioactivity, ability to drive bone formation, integration into bone tissue and induction of bone regeneration (in certain conditions). Phototherapy in bone regeneration is a therapeutic approach that involves the use of light to stimulate and accelerate the process of repair and regeneration of bone tissue. There are two common forms of phototherapy used for this purpose: Low-Level Laser Therapy (LLLT) and LED (Light Emitting Diode) Therapy. Understanding the mechanisms of laser therapy and its effects combined with hydroxyapatite has gaps. Therefore, this review was designed based on the PICO strategy (P: problem; I: intervention; C: control; O: result) to analyze the relationship between PBM therapy and hydroxyapatite. Methods: The bibliographic search, with the descriptors “hydroxyapatite AND low-level laser therapy” and “hydroxyapatite AND photobiomodulation” resulted in 43 articles in the PubMed/MEDLINE database, of which 1 was excluded for being a duplicate and another 33 due to inclusion/exclusion criteria, totaling 9 articles for qualitative analysis. In the Web of Science database, we obtained 40 articles, of which 7 were excluded for being duplicates, 1 for not having the full text available and another 17 due to inclusion/exclusion criteria, totaling 15 articles for qualitative analysis. Results: The most used biomaterial was composed of hydroxyapatite and β-tricalcium phosphate in a proportion of 70%–30%. In photobiomodulation, the gallium-aluminum-arsenide (GaAlAs) laser prevailed, with a wavelength of 780 nm, followed by 808 nm. Conclusions: The results indicated that the use of laser phototherapy improved the repair of bone defects grafted with the biomaterial, increasing the deposition of HA phosphate as indicated by biochemical estimators, spectroscopy and histological analyses.

Biomaterials and biopharmaceuticals for correcting large bone defects are a potential area of translational science. A new bioproduct, purified from snake venom and fibrinogen from buffalo blood, aroused interest in the repair of venous ulcers. Expanding potential uses, it has also been used to form biocomplexes in combination with bone grafts, associated with physical therapies or used alone. The aim of this preclinical study was to evaluate low-level laser photobiomodulation (PBM) in critical defects in the calvaria of rats filled with nanohydroxyapatite (NH) associated with the heterologous fibrin biopolymer (HFB). Sixty animals were used, divided into six groups (n = 10 each): G1 (NH); G2 (HFB); G3 (NH + HFB); G4 (NH + PBM); G5 (HFB + PBM); G6 (NH + HFB + PBM). PBM simultaneously used red (R) and infrared (IR) light emission, applied intraoperatively and twice a week, until the end of the experiment at 42 days. Microtomography, bone formation can be seen initially at the margins of the defect, more evident in G5. Microscopically, bone formation demonstrated immature and disorganized trabeculation at 14 days, with remnants of grafting materials. At 42 days, the percentage of new bone formed was higher in all groups, especially in G5 (HFB, 45.4 ± 3.82), with collagen fibers at a higher degree of maturation and yellowish-green color in the birefringence analysis with Picrosirius-red. Therefore, it is concluded that the HFB + PBM combination showed greater effectiveness in the repair process and presents potential for future clinical studies.

(1) Background: Dysregulated serum amino acids (AA) are known to be associated with obesity and risk of Type 2 Diabetes (T2D) in adults, and recent studies support the same notion in the pubertal age. It is, however, unknown whether childhood overweight may already display alterations of circulating AA. (2) Methods: We used liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS)—targeted metabolomics to determine plasma concentrations of AA and AA-related molecules in 36 children aged 7–12 years with normal weight or overweight. Clinical and anthropometric parameters were measured. (3) Results: Overweight in children is associated with an altered AA profile, with increased branched-chain amino acids (BCAA) and decreased glycine levels, with no clinically manifested metabolic conditions. Moreover, z-BMI was positively and negatively correlated with BCAA and glycine levels, respectively, even after adjustment for age and gender. We also found a correlation between the AA profile and clinical parameters such as lipids profile and glycemia. (4) Conclusions: A pattern of low glycine, and increased BCAA is correlated to z-BMI, total cholesterol, and triglycerides in overweight but otherwise healthy children. Our data suggest that, in childhood overweight, AA disturbances may precede other clinical parameters, thus providing an early indicator for the later development of metabolic disease.

The Hereditary Pyropoikilocytosis is a rare and severe form of Elliptocytosis, characterized by an alteration in the skeleton of the red blood cell membrane. It is an autosomal dominant condition with variable penetration in elliptocytosis, which may be in homozygosity or double heterozygosity in Pyropoikilocytosis, starting from the neonatal period, with clinical manifestations of persistent jaundice. We report here a case of a newborn with persistent jaundice, associated with anemia requiring blood transfusion. The clinical evolution and laboratory findings were conclusive for the diagnosis of Hereditary Pyropoikilocytosis.

Objetivo: avaliar o debriefing virtual, realizado com estudantes de enfermagem que participaram como observadores em um cenário de simulação, gravado, sobre rebaixamento do nível de consciência por hipoglicemia. Método: estudo com delineamento transversal descritivo na fase quantitativa, e exploratório-descritivo na fase qualitativa entre 2020 e 2021, com 60 estudantes de graduação em enfermagem que assistiram a um cenário gravado e vivenciaram um debriefing virtual. Foram aplicados dois instrumentos de avaliação do debriefing, e na abordagem qualitativa utilizou-se análise pelo Iramuteq. Resultados: na Escala de Avaliação do Debriefing Associada à Simulação, a média geral de avaliação foi 4,25, mostrando que os participantes tiveram uma percepção positiva. Na Escala de Experiência com o Debriefing, a avaliação geral foi 4,38 sugerindo que foi uma estratégia útil. Conclusão: os estudantes apontaram o debriefing virtual como uma estratégia positiva para o aprendizado.

Introdução/Objetivos: Sepse é a disfunção múltipla de órgãos causada pela resposta inflamatória irregular do corpo a uma infecção, incluída como prioridade em Saúde Pública pela Organização Mundial de Saúde. É a maior causa de mortes entre pacientes admitidos UTIs, o que é associado à falta de diagnóstico/tratamento eficiente em tempo adequado. O diagnóstico por hemocultura considerado padrão ouro na diagnose, demanda de 3 a 7 dias para o resultado final e baixa sensibilidade. Avanços têm sido realizados na identificação de patógenos a partir de hemocultura positiva, como automação dos testes fenotípicos e bioquímicos, testes moleculares e espectrometria de massas (MS) pela técnica MALDI-TOF, porém a dependência do cultivo prévio ocasiona importantes limitações, com aproximadamente 70% de resultados falso negativos e longo tempo necessário para o crescimento (1 a 5 dias ou mais). Neste estudo desenvolvemos uma prova de conceito para metodologia baseada em LC-MS/MS com o objetivo de monitorar íons de alta intensidade específicos para micro-organismos relacionados à sepse, diretamente de amostras de sangue total, sem a necessidade de cultivo microbiológico. Metodologia: O método que desenvolvemos tem como etapas metodológicas partindo de amostra de sangue total: lise diferencial de pH básico e lise celular ácida, extração/digestão rápida das proteínas do microrganismo e o uso da LC/ESI-MS/MS na análise/identificação dos peptídeos únicos discriminatórios de cada patógeno estudado. Resultados: Demonstramos a eficácia de nossa metodologia ao diagnosticar amostras infectadas com um ou mais dos seguintes patógenos: S. aureus, P. aeruginosa e C. albicans. Nosso método selecionou peptídeos discriminantes a partir dos dados gerados por LC-MS/MS que forneceram identificações corretas para todos os microrganismos mencionados acima com sensibilidade de 87,5% em sete horas e sem necessidade de enriquecimento em microcultura. Conclusão: Apresentamos um procedimento simples e rápido para a pré-seleção de um painel de peptídeos a ser usado para diagnóstico. A vantagem do nosso método é que podemos diagnosticar patógenos diretamente do sangue total, ao invés de passar pelo processo de cultura, configurando uma alternativa diagnóstica para sangue infecção. Prevemos também que nosso método será útil na identificação de fungos filamentosos e no diagnóstico de resistência antimicrobiana, em última análise, contribuindo para dados epidemiológicos.

A nocardiose é uma doença infecciosa que pode ocorrer de forma aguda ou crônica com características piogranulomatosas, podendo acometer o animal de forma sistêmica, sendo considerada rara em felinos. A infecção ocorre através do contato de feridas com o agente Nocardia spp. que se encontra em solo, água, madeira e outros locais favoráveis ao seu crescimento e contaminação. A doença pode gerar formação de fístulas, abscessos de forma localizada ou disseminada e apresentar secreção serossanguinolenta. Assim, o objetivo desse artigo é relatar um caso de nocardiose em um felino. O animal foi atendido em uma clínica veterinária privada de Lagarto/SE, tinha 3 anos de idade, sem raça definida (SRD), apresentando abscessos na região rostral, no abdômen, nos membros torácicos e membros pélvicos. O exame histopatológico diagnosticou nocardiose e o animal passou por tratamento farmacológico juntamente com cirurgia reconstrutiva. No entanto, o paciente foi a óbito apresentando ascite, anorexia e do baixo estado nutricional quando retornou à clínica. O tutor não autorizou a necropsia para elucidar a causa do óbito.

In equine ophthalmology, ulcerative keratitis is among the most common conditions and, in general, arises as a consequence of some trauma suffered. Secondarily, subsequent contamination by pathogenic or resident bacteria of the horse’s ocular microbiota may have undesirable consequences. Under physiological conditions, the normal microbiota coexists with the immune status of the host, serving as a barrier, ensuring the health of the ocular surface, and inhibiting the proliferation of pathogens. However, in the imbalance of immune barriers, the normal microbiota can become pathogenic and lead to infection, acting as an opportunistic agent. The present study aims to demonstrate the antimicrobial effect of platelet-rich plasma (PRP), its time of action, and its correlation with the concentration of its same components in vitro on Staphylococcus sciuri, a bacterium with high prevalence in the normal ocular microbiota of horses in the municipality of Minas Gerais. For the preparation of the PRP, eight adult Quarter Horse (QH) horses were used. The individual PRP was prepared by the double centrifugation protocol, and then, the PRPs were added to a pool, followed by testing their interaction in culture with Brain Heart Infusion (BHI) broth at different dilutions against five strains collected from different animals. After 3, 6, 12, and 18 hours, the colony formation units (CFU) count on a 5% horse blood agar plate was evaluated for each time point. Our study showed that Staphylococcus sciuri, the resident microorganism of the ocular conjunctival microbiota of horses, is more susceptible when compared to the standard strain “American Type Culture Collection” (ATCC-29213) Staphylococcus aureus, a pathogenic microorganism, which was used for the validation of our study. The antibacterial effect shown in this study was bacteriostatic for up to 6 hours. The most concentrated PRP dilutions, 1 : 1 and 1 : 2, were also most effective, suggesting that the antibacterial effect is volume dependent.

Objective: To analyze the self-reported need of patients compared to professional indications for tooth whitening. Material and Methods: Initially, 58 undergraduate students responded to a form that highlighted the question: "Do you think you need to have your teeth whitened?" Among those who answered positively to the previous question, ten individuals were photographed with their smiles. In addition, they were asked to point out, on the Vita 3D-Master scale, which color they believed their teeth had, a value that was compared to the actual color obtained by a spectrophotometer. Finally, the photographs were presented to dentists, who were asked about the indication or not of the whitening treatment. Results: Most interviewees (63.8%) self-reported the need for whitening, as well as there was a greater incidence of a positive indication among professionals (53.9%). Pearson's Chi-square test revealed a relationship between patient gender and the training course on the desire to have teeth whitened. Among the professionals, the specialty, as well as time since graduation, interfered in the indication for whitening. Conclusion: Professionals and patients share the aesthetic ideal directly related to light teeth; most patients self-perceive the color of their teeth darker than it actually is; the opinion about the color of the teeth has an extremely subjective character and varies greatly from one professional to another.

Objetivo: apresentar a concepção e implantação do Modelo de Gestão de Altas do Complexo Hospital de Clínicas da Universidade Federal do Paraná. Método: relato de experiência ocorrido entre 2017 e 2020, sobre a concepção e implantação do modelo de Gestão de Altas com apoio do modelo lógico. Resultados: continuidade do cuidado e gerenciamento de caso são operacionalizados por uma equipe exclusiva de enfermeiras de ligação que gerenciam e contrarreferenciam os cuidados para a atenção primária e outros pontos da rede de assistência à saúde. Conclusão: a gestão de altas se consolidou como uma estratégia gerencial que integra a rede de atenção à saúde, evita descontinuidades da assistência, promove segurança aos pacientes e familiares, além de otimização de leitos, e ainda insere na agenda da atenção primária os pacientes complexos. Contribui para a continuidade do cuidado e representa evidência de um novo campo de atuação do Enfermeiro.

Este trabalho objetivou avaliar os efeitos da aplicação de cama de aviário com diferentes tempos de compostagem sobre a ocorrência da hérnia das crucíferas e o desenvolvimento de plantas de couve-flor. Utilizaram-se como tratamentos parcelas inoculadas com Plasmodiophora brassicae (Pb) e adubadas com cama de aviário fresca (CA-0 + Pb) e com cama de aviário compostada por 15 (CA-15 + Pb) e por 30 dias (CA-30 + Pb). Como testemunhas utilizaram-se parcelas inoculadas e sem cama de aviário (Testemunha + Pb), parcelas não inoculadas (NI) e tratadas com cama de aviário compostada por 30 dias (CA-30 - NI), e sem cama de aviário (Testemunha - NI). O grau de estabilização e maturação da cama de aviário interferiu na severidade da hérnia das crucíferas, volume e massa de raízes sadias e das galhas e no crescimento da parte aérea. Os tratamentos CA-15 + Pb e CA-30 + Pb foram equivalentes a Testemunha + Pb quanto à intensidade da doença. Na presença do patógeno (Pb), a aplicação de CA-15 resultou em menor volume e massa de raízes sadias e menor massa de parte aérea. O tratamento CA-0 foi equivalente aos tratamentos não inoculados. Novos estudos são necessários para se elucidar os processos e interações relacionados ao efeito da maturidade de compostos de cama de aviário na infecção das raízes e ocorrência da hérnia das crucíferas.
