Introdução: A Doença de Alzheimer (DA) é a causa mais frequente de demência e caracteriza-se por uma deterioração gradual e irreversível das funções cognitivas. Seu diagnóstico é realizado quando há indícios de mutação genética causadora dessa doença, dados por história familiar ou teste genético, além de sinais claros de decréscimo da memória e da aprendizagem. No que concerne ao tratamento, os principais fármacos baseiam-se nas hipóteses etiológicas da doença, de modo que são utilizados inibidores da acetilcolinesterase. Apesar dos efeitos benéficos da intervenção farmacológica, atualmente, não existe tratamento que impeça o avanço da doença, sendo questionável sua eficácia para alguns pacientes. Essa situação impulsionou o aparecimento de estudos baseados em terapias não-farmacológicas, as quais possuem como princípio intervenções psicossociais, que melhoram os sintomas cognitivos e comportamentais, atuando como otimizadoras dos tratamentos medicamentosos tradicionais. Portanto, este estudo visa descrever a importância e a eficiência da utilização da musicoterapia na vida de pacientes com Alzheimer. Metodologia: Artigo desenvolvido com base na literatura consultada nos bancos de dados Pubmed, Scielo e Lilacs. Foram incluídos artigos entre 2012 e 2022, que tratavam diretamente do assunto. Os descritores utilizadas foram “Alzheimer”, “Musicoterapia”, “Demência na doença de Alzheimer”, “Music therapy and Alzheimer”. Após avaliação dos critérios de inclusão, o embasamento teórico da discussão foi desenvolvido por meio de 19 artigos selecionados dentre todos os encontrados. Discussão: O avanço das Neurociências possibilitou uma maior compreensão sobre a relação entre música e sistema nervoso. A percepção do som envolve uma série de estruturas cerebrais, tais como áreas do córtex e áreas do sistema límbico. Essas áreas são envolvidas na percepção musical desde a percepção auditiva do som. Tanto a percepção primária do som quanto seu entendimento são modulados pela experiência emocional de ouvir música. A percepção pode estar preservada na demência e a presença da música pode recrutar atividade motora ou recuperação da memória. Isso ocorre tanto pela capacidade da música com ritmo regular, que ativa os sistemas motor e linguagem, como por meio da associação das palavras de uma canção e o efeito emocional evocado pela música poder minimizar a apraxia verbal decorrente do estresse. Conclusão: A musicoterapia frente aos fármacos tradicionais é uma alternativa para o tratamento de Alzheimer. É notória a sua importância, pois melhora o humor dos pacientes, atua na cognição e minimiza os efeitos da demência, o que torna essa terapia uma alternativa eficaz, devido ao baixo custo e a inexistência de efeitos colaterais. Contudo, mesmo com as pesquisas realizadas, ainda existem aspectos a serem aprofundados.

Leishmaniasis is a neglected disease caused by protozoa of the genus Leishmania, which causes different clinical manifestations. Drugs currently used in the treatment such as pentavalent antimonial and amphotericin B cause severe side effects in patients, and parasite resistance has been reported. Thus, it is necessary and urgent to characterize new and effective alternative drugs to replace the current chemotherapy of leishmaniasis. In this regard, it has been experimentally demonstrated that quinoline derivatives present significative pharmacological and parasitic properties. Thus, the aim of this work was to demonstrate the leishmanicidal activity of 8-hydroxyquinoline (8-HQ) in vitro and in vivo. The leishmanicidal activity (in vitro) of 8-HQ was assayed on promastigote and intracellular amastigote forms of L. (L.) amazonensis, L. (L.) infantum chagasi, L. (V.) guyanensis L. (V.) naiffi, L. (V.) lainsoni, and L. (V.) shawi. Additionally, the levels of nitric oxide and hydrogen peroxide were analyzed. The therapeutic potential of 8-HQ was analyzed in BALB/c mice infected with a strain of L. (L.) amazonensis that causes anergic cutaneous diffuse leishmaniasis. In vitro data showed that at 24 and 72 h, 8-HQ eliminated promastigote and intracellular amastigote forms of all studied species and this effect may be potentialized by nitric oxide. Furthermore, 8-HQ was more selective than miltefosine. Infected animals treated with 8-HQ by the intralesional route dramatically reduced the number of tissue parasites in the skin, and it was associated with an increase in IFN-γ and decrease in IL-4, which correlated with a reduction in inflammatory reaction in the skin. These results strongly support the idea that 8-HQ is an alternative molecule that can be employed in the treatment of leishmaniasis, given its selectivity and multispectral action in parasites from the Leishmania genus.

A Lei dos Crimes Contra o Sistema Financeiro - Lei nº 7.492/1986 – vulgarmente chamada de “Lei do Colarinho Branco”, regula parte do grupo dos Crimes contra o Sistema Econômico Nacional. Dentre estes, destaca-se o crime de evasão de divisas, tipificado ao teor do artigo 22 da referida norma. Ocorre que, passados cerca de 30 (trinta) anos desde a promulgação da lei nº 7.492/1986, é indubitável que houve certo amadurecimento legislativo, bem como a consequente criação de novas figuras delitivas em razão das mudanças sociais, políticas e jurídicas ocorridas ao longo dos anos. Diante disso, o cenário jurídico econômico-financeiro foi afetado pelo fenômeno do conflito aparente de normais penais, causado em razão da proximidade das figuras típicas regulamentadoras da matéria. Frente a esta problemática, o presente estudo objetivou analisar o crime de evasão de divisas à luz dos princípios da consunção e da especialidade, como meio de resolução de eventual conflito normativo. O levantamento de informações foi realizado mediante pesquisas bibliográficas e documentais, através do emprego do método dedutivo, com caráter qualitativo, e à luz de ditames principiológicos, realizando-se, ainda, abordagem jurisprudencial como forma de demonstrar a aplicação prática da temática em voga.

Background: The fractionation of the n-hexane phase of the EtOH extract from the leaves of Duguetia lanceolata (Annonaceae) led to the identification of the sesquiterpene (–)-cyclocolorenone. Objective: Chemical characterization, including determination of the absolute stereochemistry, and in vitro evaluation of antileishmanial activity of the sesquiterpene (–)-cyclocolorenone, isolated from D. lanceolata, were carried out. Methods: (–)-Cyclocolorenone was isolated from D. lanceolata leaves using different chromato-graphic steps and its structure was defined by analysis of NMR and ESI-HRMS data. Additionally, the absolute configuration of (–)-cyclocolorenone was ambiguously assigned by means of vibra-tional circular dichroism (VCD). Antileishmanial activity of (–)-cyclocolorenone was evaluated on promastigote and amastigote forms of Leishmania (Leishmania) amazonensis. The integrity of the cell membrane of L. (L.) amazonensis was analyzed using the SYTOX green probe. Results: (–)-(1R,6S,7R,10R)-Cyclocolorenone displayed activity against promastigotes and amastigotes forms of L. (L.) amazonensis with IC50 of 4.54 and 28.44 , respectively. Further-more, this compound was non-toxic in J774 macrophage cells (CC50 > 458.71 ) with a selectivi-ty index > 100 (promastigotes) and > 32.2 (amastigotes). Additionally, (–)-cyclocolorenone was ob-served to target the parasite cell membrane. Conclusion: Obtained data suggested that (–)-cyclocolorenone, in which absolute configuration was determined, can be considered as a scaffold for the development of new drugs for the treatment of leishmaniasis.

Visceral leishmaniasis (VL) is one of the most severe clinical manifestations of leishmaniasis as it leads to death in 90% of untreated cases. The liver, spleen and bone marrows are the organs most affected, however, Leishmania parasites are able to reach the intestines where the gut-associated lymphoid tissue (GALT) is located. Under physiological conditions, the gastrointestinal tract and GALT interact with the enteric nervous system (SNE), however, there are no studies exploring the modulatory role of Leishmania (Leishmania) infantum in the intestines. Thus, this work aimed to investigate the parasitism, stratigraphy, and morphological changes in the myenteric plexus of golden hamsters infected with L. (L.) infantum. The animals were infected intraperitoneally, and the parasite load was evaluated in the spleen, the liver, and the jejunum. The stratigraphic evaluation and the quantitative and morphometric analyses of NADH-dp and NADPH-dp myenteric neurons were studied at 30-, 60-, and 90-days post-infection (DPI). Parasites in the spleen, the liver, and the jejunum increased during the progression of the infection. Stratigraphy studies showed a significant hypertrophy of the villi and the crypts associated with the increased intraepithelial lymphocytes that were observed in the jejunum of the infected animals. In addition, mucosal atrophy associated with a reduction in AB+ and PAS+ goblet cells was observed at 60 DPI and 90 DPI. These morphological changes were associated with an atrophy of the cell profile from NADPH-dp myenteric neurons. Furthermore, a significant decrease in the densities of this neuron population was observed in the chronic phase of the infection. This study suggests that L. (L.) infantum parasites are able to alter the morphology and innervation of the jejunum in golden hamsters.

To overcome such limitations, it becomes urgent to characterize new bioactive molecules. Plants produce and accumulate different classes of bioactive compounds, and these molecules can be studied as a strategy to combat leishmaniasis. The study presented herein evaluated the leishmanicidal effect of maslinic acid isolated from the leaves of Hyptidendron canum (Lamiaceae) and investigated the morphological that occurred on Leishmania (Leishmania) infantum upon treatment. Maslinic acid was active and selective against promastigote and amastigote forms in a dose-dependent manner. Additionally, it was not toxic to peritoneal macrophages isolated from golden hamsters, while miltefosine and amphotericin B showed mild toxicity for macrophages. Morphological changes in promastigotes of L. (L.) infantum treated with maslinic acid were related to cytoplasmic degeneration, intense exocytic activity, and blebbing in the kDNA; disruption of mitochondrial cristae was observed in some parasites. The nucleus of promastigote forms seems to be degraded and the chromatin fragmented, suggesting that maslinic acid triggers programmed cell death. These results indicate that maslinic acid may be an interesting molecule to develop new classes of drugs against leishmaniasis. O presente artigo propõe-se a investigar, de forma clara e objetiva, as contribuições metodológicas desenvolvidas pelo professor a favor da construção da autonomia do aluno. Nesse sentido, deseja-se criar um espaço de reflexão e crítica sobre a prática docente, cuja premissa é ampliar possibilidades didático-pedagógicas para o aprimoramento de saberes que favoreçam o objetivo ora abordado, no decorrer do processo ensino-aprendizagem. É relevante apontar que neste trabalho o diálogo foi apresentado como uma das ferramentas possíveis para elaboração de propostas educativas autônomas, levantadas neste estudo. A metodologia utilizada para a condução dessa pesquisa foi a revisão bibliográfica, também conhecida como revisão de literatura, cujo objetivo é aprofundar-se na questão apresentada, tendo como base autores que investigam o tema, o que garante a confiabilidade e a qualidade técnica e científica do trabalho. Esta pesquisa ressalta conceitos relevantes para a relação do professor e aluno na busca de um ensino significativo, autônomo e transformador.

A multipara pregnant woman, 38 years old, presented with an initial suspicion of antibiotic-refractory mastitis. A breast biopsy revealed luminal type B invasive carcinoma and the image showed bone and liver metastases. She developed oligosymptomatic hypercalcemia requiring hydration and underwent cesarean at 35 weeks and 6 days; 2 days later, she received an osteolysis inhibitor, and a week later started chemotherapy. A baby boy was born with Apgar 9/10; hypercalcemia was observed after 12 h; then, he developed lethargy, hypotonia, and bradypnea after 21 h, requiring continuous positive airway pressure and monitoring. He received volume expansion with an improvement in his condition. Signs of breast cancer are confused with physiological breast changes in pregnancy such as engorgement, hypertrophy, and nipple discharge, implying a delay in diagnosis. Hypercalcemia during pregnancy occurs in about 0.03% of women; maternal complications include acute kidney injury, severe hypertension, and pre-eclampsia. Fetal adverse outcomes include intrauterine growth restriction, fetal demise in utero, neonatal hypocalcemia, tetany, and permanent hypoparathyroidism. Primary hyperparathyroidism is the main cause; hypercalcemia from metastatic breast cancer can be due to osteolytic metastases and humoral hypercalcemia of malignancy caused by peptide release related to parathyroid hormone, which promotes bone resorption and renal calcium reabsorption. In cases of malignant hypercalcemia, pamidronate is effective in inhibiting osteolysis, but it reduces uterine contractions and presents a teratogenic risk. Calcitonin is well tolerated during pregnancy but has a limited effect; the use of glucocorticoids can also be considered. In case of maternal hypercalcemia, the newborn’s calcium levels must be monitored. Hypercalcemia in pregnancy is a rare condition when cancer-related hypercalcemia may cause great maternal morbidity and fetal and neonatal morbidity and mortality. The manifestations presented by the neonate were compatible with the suspicion of neonatal hypercalcemia. Diagnostic confirmation was made based on the metabolic condition; the basis of emergency treatment is intravenous hydration.

Introdução: O uso de plantas medicinais como alternativa terapêutica no tratamento de inúmeras doenças é um fato que acontece a milénios antes de Cristo, e a inovação do Sistema Único de Saúde em liberar fitoterápicos como adjuvante ou tratamento, abriu caminho para o conhecimento da população sobre as plantas medicinais e seu uso correto para tratamento. Plantas como espinheira-santa, romã e unha-de-gato são frequentemente empregados para tratamento, e o objetivo desse trabalho é identificar os principais metabólitos secundários que são vinculados às principais características farmacológicas das plantas.Metodologia: Trabalho realizado no laboratório de pesquisa do CENTRO UNIVERSITÁRIO – FIMCA, Porto Velho - RO. As análises prospectivas foram realizadas no mês de agosto de 2021. No desenvolvimento da pesquisa foram utilizados: laboratório, artigos científicos, teses, sites, livros e revistas especializadas no tema.Resultado: Nota-se que tanto a romã, unha-de-gato e espinheira-santa apresentaram uma quantidade expressiva de metabólitos secundários. Através da prospecção fitoquímica para a romã os testes foram positivos para alcaloides, glicosídeos cardiotônicos, flavonoides, taninos e esteroides. Para a unha-de-gato os testes foram positivos para alcaloides, taninos, saponinas e triterpenos. E para a Espinheira-Santa os testes foram positivos para alcaloides, glicosídeos, cumarinas, flavonoides, taninos, saponinas e triterpenos. Sendo assim, entende-se que as espécies possuem possíveis ações terapêuticas que são direcionadas por estes metabólitos secundários.Conclusão: Durante a elaboração desta pesquisa foi possível determinar que tanto a romã, unha-de-gato e a espinheira santa tiveram resultados que sugerem que as espécies possuem ações farmacológicas devido as presenças dos metabólitos secundários e por isso podem ser utilizadas para tratamento de algumas patologias.

Objetivo: compreender como enfermeiros supervisores da Atenção Primária à Saúde realizam a gestão dos recursos materiais. Método: abordagem qualitativa, tipo estudo de caso, com 13 enfermeiros supervisores e/ou assistenciais, que responderam a entrevista semiestruturada. Resultados: a gestão dos recursos materiais foi entendida de maneira diversificada pelos enfermeiros mencionando etapas isoladas de previsão e provisão; controle, organização e gestão dos recursos materiais; sendo realizadas de forma incompleta. É parametrizada pelo nível central sem prever o estoque de segurança e dividida em duas etapas: previsão e controle de estoque. Fatores desafiadores: estrutura física das unidades, interrupções frequentes no intervalo entre a solicitação e a entrega dos materiais. Potencialidades: integração da equipe multidisciplinar, comunicação entre os supervisores por meio de redes sociais. Conclusão: a racionalização e a gestão dos recursos materiais eficiente e eficaz são necessárias, pois contribuirão para manutenção da continuidade da assistência, garantindo a qualidade.

Introdução: O saber popular e o conhecimento empírico há muito vêm demonstrando os efeitos benéficos proporcionados pelo uso de plantas como terapia complementar ou alternativa. Objetivo: Este estudo teve como objetivo avaliar o conhecimento e o uso de plantas medicinais e fitoterápicos como prática terapêutica por acadêmicos de enfermagem. Metodologia: Trata-se de um estudo transversal, de caráter quantitativo, composto por uma mostra de 90 acadêmicos de enfermagem, utilizando um questionário semiestruturado adaptado para a coleta de dados. Resultados: Nenhum entrevistado conhecia a diferença de conceito entre fitoterápico e planta medicinal e 67% destes, afirmaram saber pouco sobre o assunto, mas a totalidade dos entrevistados afirmou que já utilizaram plantas medicinais pelo menos uma vez, principalmente na forma de chá. Conclusão: Conclui-se que há uma lacuna no aprendizado dos acadêmicos de enfermagem sobre o conhecimento da fitoterapia e das plantas medicinais. A inserção de uma disciplina com esta temática poderá contribuir para a solução deste problema, melhorando o conhecimento na área.

Visceral leishmaniasis is a neglected tropical disease caused by parasites belonging to the Leishmania genus that infect macrophages in different tissues such as the spleen, liver, lymph nodes, bone marrow, and intestine. Therefore, this study aimed to investigate the integrity of the intestinal tract and the nitrergic (NADPH-dp) and metabolically active (NADH-dp) myenteric neurons of the duodenum of golden hamsters infected with L. (L.) infantum. Therefore, thirty golden hamsters were divided into six groups (n = 5); three of them were infected with 2 × 10

Ursolic acid, a triterpene produced by plants, displayed leishmanicidal activity in vitro and in vivo, however, the low solubility of this triterpene limits its efficacy. To increase the activity of ursolic acid (UA), this triterpene was entrapped in nanostructured lipid carriers (UA-NLC), physical-chemical parameters were estimated, the toxicity was assayed in healthy golden hamsters, and the efficacy of UA-NLC was studied in experimental visceral leishmanisis. UA-NLC exhibited a spherical shape with a smooth surface with a size of 266 nm. UA-NLC displayed low polydispersity (PDI = 0.18) and good colloidal stability (−29.26 mV). Hamsters treated with UA-NLC did not present morphological changes in visceral organs, and the levels of AST, ALT, urea and creatinine were normal. Animals infected with Leishmania (Leishmania) infantum and treated with UA-NLC showed lower parasitism than the infected controls, animals treated with UA or Amphotericin B (AmB). The therapeutic activity of UA-NLC was associated with the increase in a protective immune response, and it was associated with a high degree of spleen and liver preservation, and the normalization of hepatic and renal functions. These data indicate that the use of lipid nanoparticles as UA carriers can be an interesting strategy for the treatment of leishmaniasis.

Leishmaniasis is a neglected tropical disease that affects people living in tropical and subtropical areas of the world. There are few therapeutic options for treating this infectious disease, and available drugs induce severe side effects in patients. Different communities have limited access to hospital facilities, as well as classical treatment of leishmaniasis; therefore, they use local natural products as alternative medicines to treat this infectious disease. The present work performed a bibliographic survey worldwide to record plants used by traditional communities to treat leishmaniasis, as well as the uses and peculiarities associated with each plant, which can guide future studies regarding the characterization of new drugs to treat leishmaniasis. A bibliographic survey performed in the PubMed and Scopus databases retrieved 294 articles related to traditional knowledge, medicinal plants and leishmaniasis; however, only 20 were selected based on the traditional use of plants to treat leishmaniasis. Considering such studies, 378 quotes referring to 292 plants (216 species and 76 genera) that have been used to treat leishmaniasis were recorded, which could be grouped into 89 different families. A broad discussion has been presented regarding the most frequent families, including Fabaceae (27 quotes), Araceae (23), Solanaceae and Asteraceae (22 each). Among the available data in the 378 quotes, it was observed that the parts of the plants most frequently used in local medicine were leaves (42.3% of recipes), applied topically (74.6%) and fresh poultices (17.2%). The contribution of Latin America to studies enrolling ethnopharmacological indications to treat leishmaniasis was evident. Of the 292 plants registered, 79 were tested against Leishmania sp. Future studies on leishmanicidal activity could be guided by the 292 plants presented in this study, mainly the five species Carica papaya L. (Caricaceae), Cedrela odorata L. (Meliaceae), Copaifera paupera (Herzog) Dwyer (Fabaceae), Musa × paradisiaca L. (Musaceae), and Nicotiana tabacum L. (Solanaceae), since they are the most frequently cited in articles and by traditional communities.

Drugs employed in the treatment of leishmaniasis have limited efficacy and induce local and systemic side effects to the patients. Natural products are an interesting alternative to treat leishmaniasis, because some purified molecules are selective toward parasites and not to the host cells. Thus, the aim of the present study was to compare the in vitro antileishmanial activity of the triterpenes betulin (Be), lupeol (Lu), and ursolic acid (UA); analyze the physiology and morphology of affected organelles; analyze the toxicity of selected triterpenes in golden hamsters; and study the therapeutic activity of triterpenes in hamsters infected with L. (L.) infantum as well as the cellular immunity induced by studied molecules. The triterpenes Lu and UA were active on promastigote (IC50=4.0±0.3 and 8.0±0.2 μM, respectively) and amastigote forms (IC50=17.5±0.4 and 3.0±0.2 μM, respectively) of L. (L.) infantum, and their selectivity indexes (SI) toward amastigote forms were higher (≥13.4 and 14, respectively) than SI of miltefosine (2.7). L. (L.) infantum promastigotes treated with Lu and UA showed cytoplasmic degradation, and in some of these areas, cell debris were identified, resembling autophagic vacuoles, and parasite mitochondria were swelled, fragmented, and displayed membrane potential altered over time. Parasite cell membrane was not affected by studied triterpenes. Studies of toxicity in golden hamster showed that Lu did not alter blood biochemical parameters associated with liver and kidney functions; however, a slight increase of aspartate aminotransferase level in animals treated with 2.5 mg/kg of UA was detected. Lu and UA triterpenes eliminated amastigote forms in the spleen (87.5 and 95.9% of reduction, respectively) and liver of infected hamster (95.9 and 99.7% of reduction, respectively); and UA showed similar activity at eliminating amastigote forms in the spleen and liver than amphotericin B (99.2 and 99.8% of reduction). The therapeutic activity of both triterpenes was associated with the elevation of IFN-γ and/or iNOS expression in infected treated animals. This is the first comparative work showing the in vitro activity, toxicity, and therapeutic activity of Lu and UA in the chronic model of visceral leishmaniasis caused by L. (L.) infantum; additionally, both triterpenes activated cellular immune response in the hamster model of visceral leishmaniasis. Such enzymes can be blocked by azoles and allylamines drugs, such as the antifungal butenafine chloride. This drug was active on parasites that cause cutaneous and visceral leishmaniasis. Based on the leishmanicidal activity of butenafine chloride and considering the absence of reports about the therapeutic potential of this drug in cutaneous leishmaniasis, the present work is aimed at analyzing the efficacy of butenafine formulated in two different topical delivery systems, the self-nanoemulsifying drug delivery systems (BUT-SNEDDS) and in a SNEDDS-based nanogel (BUT-SNEDDS gel) as well as in the free form in experimental cutaneous leishmaniasis. Cathelicidins form a class of such molecules, which are produced by a variety of organisms. Batroxicidin (BatxC) is a cathelicidin found in the venom of the common lancehead (Bothrops atrox). In the present work, BatxC and two synthetic analogues, BatxC(C-2.15Phe) and BatxC(C-2.14Phe)des-Phe1, were assessed for their microbicidal activity. All three peptides showed a broad-spectrum activity on Gram-positive and -negative bacteria, as well as promastigote and amastigote forms of Leishmania (Leishmania) amazonensis. Circular dichroism (CD) and nuclear magnetic resonance (NMR) data indicated that the three peptides changed their structure upon interaction with membranes. Biomimetic membrane model studies demonstrated that the peptides exert a permeabilization effect in prokaryotic membranes, leading to cell morphology distortion, which was confirmed by atomic force microscopy (AFM). The molecules considered in this work exhibited bactericidal and leishmanicidal activity at low concentrations, with the AFM data suggesting membrane pore formation as their mechanism of action. Nesse sentido, a atual pesquisa se propôs a construir indicadores de qualidade da educação a partir dos dados sobre a trajetórias escolares, buscando sintetizar as informações de permanência e conclusão da educação básica de uma coorte de estudantes que cursaram o 6° ano do ensino fundamental em Curitiba em 2010. A metodologia de pesquisa adotada foi a pesquisa quantitativa de caráter longitudinal com base no Banco de Dados do INEP, na qual foram coletados dados de matrícula de estudantes do 6° ano do ensino fundamental de Curitiba de 2010 e esses momentos foram acompanhados ao longo de 7 anos. Utilizar a trajetória escolar como um indicador de qualidade significa discutir política educacional e olhar para a garantia do direito à educação a partir das dimensões de acesso, permanência e condições de conclusão. Assim como outros indicadores propostos também possuem limitações, mas abrem a possibilidade de ter outros pontos de vista sobre a qualidade da educação e permitem análises cotejadas com outros indicadores.

Thus, treatment options rely on systemic therapy, which fail to produce high drug local concentrations but can lead to significant systemic toxicity. Amphotericin B (AmB) is highly efficacious in the treatment of both fungal and parasitic diseases such as cutaneous leishmaniasis, but is only reserved for parenteral administration in patients with severe pathophysiology. Here, we have designed and optimised AmB-transfersomes [93.5 % encapsulation efficiency, size of 150 nm, and good colloidal stability (-35.02 mV)] that can remain physicochemically stable (>90 % drug content) at room temperature and 4 °C over 6 months when lyophilised and stored under desiccated conditions. AmBtransfersomes possessed good permeability across mouse skin (4.91 ± 0.41 μg/cm2/h) and 10-fold higher permeability across synthetic Strat-M® membranes. In vivo studies after a single topical application in mice showed permeability and accumulation within the dermis (>25 μg AmB /g skin at 6 h post-administration) indicating the delivery of therapeutic amounts of AmB for mycoses and cutaneous leishmaniasis, while a single daily administration in Leishmania (Leishmania) amazonensis infected mice over 10 days resulted in excellent efficacy (98 % reduction in Leishmania parasites). Combining the application of AmB-transfersomes with metallic microneedles in vivo increased levels in the SC and dermis but is unlikely to elicit transdermal levels. In conclusion, AmB-transfersomes are promising and stable topical nanomedicines that can be readily translated for parasitic and fungal infectious diseases.

The treatment of all forms of leishmaniasis relies on first-line drug, pentavalent antimonial, and in cases of drug failure, the second-line drug amphotericin B has been used. Besides the high toxicity of drugs, parasites can be resistant to antimonial in some areas of the World, making it necessary to perform further studies for the characterization of new antileishmanial agents. Thus, the aim of the present work was to evaluate the leishmanicidal activity of tolnaftate, a selective reversible and non-competitive inhibitor of the fungal enzyme squalene epoxidase, which is involved in the biosynthesis of ergosterol, essential to maintain membrane physiology in fungi as well as trypanosomatids. Tolnaftate eliminated promastigote forms of L. (L.) amazonensis, L. (V.) braziliensis and L. (L.) infantum (EC50 ~ 10 μg/mL and SI ~ 20 for all leishmanial species), and intracellular amastigote forms of all studied species (EC50 ~ 23 μg/mL in infections caused by dermatotropic species; and 11.7 μg/mL in infection caused by viscerotropic species) with high selectivity toward parasites [SI ~ 8 in infections caused by dermatotropic species and 17.4 for viscerotropic specie]. Promastigote forms of L. (L.) amazonensis treated with the EC50 of tolnaftate displayed morphological and physiological changes in the mitochondria and cell membrane. Additionally, promastigote forms treated with tolnaftate EC50 reduced the level of ergosterol by 5.6 times in comparison to the control parasites. Altogether, these results suggest that tolnaftate has leishmanicidal activity towards Leishmania sp., is selective, affects the cell membrane and mitochondria of parasites and, moreover, inhibits ergosterol production in L. The available treatments are not well tolerated and present diverse side effects in patients, justifying the search for new therapeutic compounds. In the present study, the therapeutic potential and toxicity of ursolic acid (UA), isolated from the leaves of Baccharis uncinella C. DC. (Asteraceae), were evaluated in experimental visceral leishmaniasis. To evaluate the therapeutic potential of UA, hamsters infected with L. (L.) infantum were treated daily during 15 days with 1.0 or 2.0 mg UA/kg body weight, or with 5.0 mg amphotericin B/kg body weight by intraperitoneal route. Fifteen days after the last dose, the parasitism of the spleen and liver was stimated and the main histopathological alterations were recorded. The proliferation of splenic mononuclear cells was evaluated and IFN-γ, IL-4, and IL-10 gene expressions were analyzed in spleen fragments. The toxicity of UA and amphotericin B were evaluated in healthy golden hamsters by histological analysis and biochemical parameters. Animals treated with UA had less parasites in the spleen and liver when compared with the infected control group, and they also showed preservation of white and red pulps, which correlate with a high rate of proliferation of splenic mononuclear cells, IFN-γ mRNA and iNOS production. Moreover, animals treated with UA did not present alterations in the levels of AST, ALT, creatinine and urea. Taken together, these findings indicate that UA is an interesting natural compound that should be considered for the development of prototype drugs against visceral leishmaniasis. No. of bitstreams: 0 Previous issue date: 2020-10-15 Leishmaniasis is a neglected disease presenting cutaneous, mucosal and visceral forms and affecting an estimated 12 million mostly low-income people. Treatment of cutaneous leishmaniasis (CL) is recommended to expedite healing, reduce risk of scarring, prevent parasite dissemination to other mucocutaneous (common with New World species) or visceral forms and reduce the chance of relapse, but remains an unmet need. Available treatments are painful, prolonged (>20 days) and require hospitalisation, which increases the cost of therapy. Here we present the development of optimised topical self-nanoemulsifying drug delivery systems (SNEDDS) loaded with buparvaquone (BPQ, a hydroxynapthoquinone from the open Malaria Box) for the treatment of CL from New World species. The administration of topical BPQ-SNEDDS gels for 7 days resulted in a reduction of parasite load of 99.989 ± 0.019% similar to the decrease achieved with intralesionally administered Glucantime® (99.873 ± 0.204%) in a L. amazonensis BALB/c model. In vivo efficacy was supported by ex vivo permeability and in vivo tape stripping studies. BPQ-SNEDDS and their hydrogels demonstrated linear flux across non-infected CD-1 mouse skin ex vivo of 182.4 ± 63.0 μg cm−2 h−1 and 57.6 ± 10.8 μg cm−2 h−1 respectively localising BPQ within the skin in clinically effective concentrations (227.0 ± 45.9 μg and 103.8 ± 33.8 μg) respectively. These levels are therapeutic as BPQ-SNEDDS and their gels showed nanomolar in vitro efficacy against L. amazonensis and L. braziliensis amastigotes with excellent selectivity index toward parasites versus murine macrophages. In vivo tape stripping experiments indicated localisation of BPQ within the stratum corneum and dermis. Histology studies confirmed the reduction of parasitism and indicated healing in animals treated with BPQ-SNEDDS hydrogels. These results highlight the potential clinical capability of nano-enabled BPQ hydrogels towards a non-invasive treatment for CL. Understanding the dynamics of soil organisms in natural environments can be very useful for determining the quality of forest and agricultural ecosystems. This study aimed to assess arbuscular mycorrhizal fungal (AMF) communities in pasture and regenerating riparian forest sites in Guajara-Mirim, Rondonia, Brazil. Ten soil samples were randomly collected from each ecosystem in three seasons (2018 dry and rainy seasons and 2019 dry season). AMF species richness, frequency, spore density, Shannon–Wiener diversity index, Simpson’s dominance index, and ideal sample size were determined. Species richness varied among sites and seasons, with a tendency toward higher richness in pasture soil in the 2018 dry season. The dominant AMF species in both ecosystems were Glomus macrocarpum and Acaulospora mellea . The pasture site in the 2018 dry season had higher AMF diversity than the riparian forest in the 2018 rainy season and the 2019 dry season. The complete linkage distance between the pasture site in the 2019 dry season and all other sites/seasons was 100%. More than 10 soil samples are needed to accurately determine the richness of AMF species in pasture and riparian forest sites in Guajara-Mirim, Brazil.

Objetivo: Avaliar o uso da Aroeira no tratamento da mucosite oral induzida pela radioterapia exclusiva ou associada à quimioterapia em pacientes com câncer de cabeça e pescoço. Método: Foi realizado um ensaio clínico controlado, randomizado e duplo-cego, onde doze indivíduos foram alocados em dois grupos: intervenção e controle e orientados a aplicar a pomada três vezes por dia. A avaliação diária do grau da mucosite e da nota da dor referida pelo paciente permitiram observar a regressão ou não da sua manifestação. Foram também aplicados formulários referentes à situação clínica, sociodemográfica e de higiene bucal dos participantes. Uma análise descritiva dos dados foi realizada. Resultados: No grupo controle, 50% (3) dos participantes apresentaram regressão do grau de mucosite; já no grupo de intervenção 17% (1). Em relação a nota da dor, 83% do grupo placebo relatou regressão e 50% do grupo intervenção. Conclusões: Neste estudo o uso da aroeira não mostrou-se eficaz no tratamento da mucosite, em relação ao uso do placebo. por outro lado apresentou uma porcentagem significativa de redução da nota da dor. Devido às limitações do estudo não se pode comprovar o efeito fitoterápico da aroeira neste tipo de abordagem.

The aim of the present study is to measure the solubility of urea in aqueous solutions of prop-2-enoic acid and propane-1,2,3-triol, as these compounds are used as co-monomers to produce urea base co-polymers. Experimental values have been obtained at several concentrations and temperatures. Solubility results were modelled with the help of an exponential empirical correlation, ideal solid-liquid equilibrium correlation and the Universal Functional Activity Coefficient (UNIFAC) method, used to describe the activity coefficient in the liquid phase. The empirical correlation requires two empirical parameters for each solvent and leads to the best fit for the available data. The UNIFAC method correlation also has a good numerical performance and is completely predictive, and it does not require the estimation of additional parameters.

Studies on the situation of mortality from traffic accidents among workers are still insufficient. This study aimed to describe the mortality rates for traffic accidents / path between state workers of Bahia and Brazil. This is a quantitative, descriptive study of fatal work accidents related to traffic accidents / path with the Mortality Information System data in the period from 2009 to 2011. The results show 317 reported deaths from traffic accidents in Bahia and 5118 in Brazil, with a predominance of male workers, married, white, between 15 to 35 years, with an average level of education, employed in the industrial sector, victims of accidents vessel, water transport and air; The mortality rate ranged from 1.2 to 1.9 / 100 000 workers in Bahia and Brazil 1.6 to 2.0 / 100 000 workers in the investigation period; proportional mortality in Bahia ranged from 55.5% to 61.3% and Brazil 47.6% to 51.1%. It is concluded that the findings relating to sociodemographic and occupational exposure showed important aspects that should be considered in the planning of preventive measures to improve worker health and working conditions.

Ursolic acid (UA) is a triterpene with a broad array of pharmacological activities. In leishmaniasis, UA killed different species of parasites, and it was active in the experimental model of cutaneous and visceral leishmaniasis. Thus, the objective of this work was to study the therapeutic efficacy of the conventional drugs amphotericin B (AmB) or glucantime (Glu) combined with UA in experimental visceral and cutaneous leishmaniasis, respectively. L. (L.) infantum-infected hamsters were treated with AmB alone or combined with UA. L. (L.) amazonensis-infected BALB/c mice were treated with Glu alone or combined with UA. Animals were treated for 15 consecutive days by intraperitoneal or intralesional routes. Following one week after the last dose, the tissue parasitism and cellular immune responses were analyzed. Hamsters treated with 0.2 and 1.0 mg/kg of AmB plus 1.0 mg/kg of UA showed low hepatic and splenic parasitisms, however, AmB given as monotherapy did not reduce the number of viable parasites in the spleen of treated animals. In cutaneous leishmaniasis, Glu given as monotherapy was inactive at 2.0 mg/kg, showed mild activity at 10.0 mg/kg, and at 50.0 mg/kg was highly active at eliminating parasites in the skin. When animals were treated with Glu plus UA, higher leishmanicidal activity was observed in comparison to all groups treated with monotherapy schemes, and such activity was related to lesion improvement and upregulation of IFN-&gamma, production. Altogether, data suggest that the association of drugs for the treatment of leishmaniasis can increase the efficiency of the treatment and decrease the toxicity associated to the conventional drugs.

No estágio final em unidade de cuidados na comunidade os objetivos do projeto de intervenção foram a avaliação do risco e o desenvolvimento do plano de intervenção de enfermagem de reabilitação para prevenção de quedas em idosos no domicílio. Estudo de caso descritivo, participaram quatro utentes idosos, portadores de doenças neurológicas, com alteração do equilíbrio e com potencial de recuperação. Os resultados foram a diminuição de fatores de risco intrínsecos relacionados com o equilíbrio, extrínsecos relacionados com o domicílio e a diminuição da dependência nas atividades de transferência e mobilidade. Conclui-se que os cuidados de enfermagem de reabilitação produzem ganhos na diminuição do risco de queda, diminuição da dependência e aumentam a segurança do idoso e dos seus cuidadores. A enfermagem de reabilitação no domicílio nas fases subagudas da doença neurológica pode potencializar estes resultados. In the aging process are associated physiologic changes that contribute to increased risk of fall. The goals of the intervention project in the final training in the community care unit were the risk assessment and the development of the plan of rehabilitation nursing intervention for the falls prevention in the elderly at home. Descriptive case study included four elderly patients with neurological diseases with change of balance and with recovery potential. The results were the decrease of intrinsic risk factors related to balance, decrease of extrinsic risk factors related with home and the reduction of dependence on transfer and mobility activities. It is concluded that the rehabilitation nursing care produce gains in reducing the risk fall, decreased dependency and increase the safety of the patients and informal caregivers. The rehabilitation nursing at home in subacute phases of neurological disease can enhance these results.
