In the laste years several articles focalized on human microbioma - the microorganisms from skin, mucosae, bowel - and on its role in chronic inflammatory diseases of the skin as acne, rosacea, atopic dermatitis, seborrheic dermatitis. In this article the authors present an update on particular acne skin’s microbioma, on innate immunity in acne and new physiopathology mecanisms described in inflammatory process in acne, and at the end we present in vitro, ex vivo and in vivo studies on the microbioma modulation and microbiofilm of pathogenic ribotypes of P. acnes leading to a significant improvement of acne in a series of 74 acne patients.

International audience; We have previously shown that the C-type Natriuretic Peptide (CNP), a peptide produced by lungs, is able to impact Pseudomonasaeruginosa physiology. In the present work, the effect of CNP at different concentrations on P. aeruginosa biofilm formation was studied and the mechanisms of action of this human hormone on P. aeruginosa were deciphered. CNP was shown to inhibit dynamic biofilm formation in a dose-dependent manner without affecting the bacterial growth at any tested concentrations. The most effective concentrations were 1 and 0.1 µM. At 0.1 µM, the biofilm formation inhibition was fully dependent on the CNP sensor protein AmiC, whereas it was only partially AmiC-dependent at 1 µM, revealing the existence of a second AmiC-independent mode of action of CNP on P. aeruginosa. At 1 µM, CNP reduced both P. aeruginosa adhesion on glass and di-rhamnolipid production and also increased the bacterial membrane fluidity. The various effects of CNP at 1 µM and 0.1 µM on P. aeruginosa shown here should have major consequences to design drugs for biofilm treatment or prevention.

International audience; Neurohormones diffuse in sweat and epidermis leading skin bacterial microflora to be largely exposed to these host factors. Bacteria can sense a multitude of neurohormones, but their role in skin homeostasis was only investigated recently. The first study focused on substance P (SP), a neuropeptide produced in abundance by skin nerve terminals. SP is without effect on the growth of Gram-positive (Bacillus cereus, Staphylococcus aureus, and Staphylococcus epidermidis) and Gram-negative (Pseudomonas fluorescens) bacteria. However, SP is stimulating the virulence of Bacillus and Staphylococci. The action of SP is highly specific with a threshold below the nanomolar level. Mechanisms involved in the response to SP are different between bacteria although they are all leading to increased adhesion and/or virulence. The moonlighting protein EfTu was identified as the SP-binding site in B. cereus and Staphylococci. In skin nerve terminals, SP is co-secreted with the calcitonin gene-related peptide (CGRP), which was shown to modulate the virulence of S. epidermidis. This effect is antagonized by SP. Identification of the CGRP sensor, DnaK, allowed understanding this phenomenon as EfTu and DnaK are apparently exported from the bacterium through a common system before acting as SP and CGRP sensors. Many other neuropeptides are expressed in skin, and their potential effects on skin bacteria remain to be investigated. Integration of these host signals by the cutaneous microbiota now appears as a key parameter in skin homeostasis.

Introduction P . acnes presente 2 ribotypes (RT) RT 4 et 5 particulierement adherents et virulents ; leur microbiofilm est un element important dans l’induction de l’inflammation acneique. Objectifs de l’etude L’observation in vitro de la formation du microbiofilm de P. acnes issus de peaux acneiques, puis l’evaluation in vitro de l’effet d’un extrait vegetal (MPA-regul) sur la virulence et la croissance de 2 souches P. acnes RT4 et RT5. Dans une deuxieme etape, ont ete analyses l’effet et la tolerance d’une emulsion H/E contenant l’actif MPA-regul chez les patients adultes presentant une acne polymorphe legere a moderee. Materiel et methodes In vitro A. Le microbiofilm des souches de P. acnes a ete analyse sur des lamelles en verre traitees par MPA-regul (a 0,5 % et 1 %) a 24, 48 et 72 heures par le Confocal Laser Scanning Microscopy. B. La croissance des 2 souches de P. acnes –HL045PA1 (RT4) et HL043PA2 (RT5) a ete etudiee sur culture en microplaques. C. La cytotoxicite (virulence) des P. acnes RT4 et RT5 a ete etudiee sur la lignee keratinocytaire HaCaT par dosage de la liberation de LDH dans le milieu de culture. La cytotoxicite a ete egalement evaluee en presence de l’actif MPA-regul a 2 concentrations (etude en triplicate). International audience; BACKGROUND: The cephalopod mollusk Sepia officinalis can be considered as a relevant model for studying reproduction strategies associated to seasonal migrations. Using transcriptomic and peptidomic approaches, we aim to identify peptide sex pheromones that are thought to induce the aggregation of mature cuttlefish in their egg-laying areas. RESULTS: To facilitate the identification of sex pheromones, 576 5'-expressed sequence tags (ESTs) were sequenced from a single cDNA library generated from accessory sex glands of female cuttlefish. Our analysis yielded 223 unique sequences composed of 186 singletons and 37 contigs. Three major redundant ESTs called SPα, SPα' and SPβ were identified as good candidates for putative sex pheromone transcripts and are part of the 87 unique sequences classified as unknown. The alignment of translated SPα and SPα' revealed a high level of conservation, with 98.4% identity. Translation led to a 248-amino acid precursor containing six peptides with multiple putative disulfide bonds. The alignment of SPα-α' with SPβ revealed a partial structural conservation, with 37.3% identity. Translation of SPβ led to a 252-amino acid precursor containing five peptides. The occurrence of a signal peptide on SPα, SPα' and SPβ showed that the peptides were secreted. RT-PCR and mass spectrometry analyses revealed a co-localization of transcripts and expression products in the oviduct gland. Preliminary in vitro experiments performed on gills and penises revealed target organs involved in mating and ventilation. CONCLUSIONS: The analysis of the accessory sex gland transcriptome of Sepia officinalis led to the identification of peptidic sex pheromones. Although preliminary functional tests suggested the involvement of the α3 and β2 peptides in ventilation and mating stimulation, further functional investigations will make it possible to identify the complete set of biological activities expected from waterborne pheromones.
