Abstract Drainage for forestry has created ~ 1 million km of artificial waterways in Sweden, making it one of the largest human-induced environmental disturbances in the country. These extensive modifications of both peatland and mineral soil dominated landscapes still carry largely unknown, but potentially enormous environmental legacy effects. However, the consequences of contemporary ditch management strategies, such as hydrological restoration via ditch blocking or enhancing forest drainage to promote biomass production via ditch cleaning, on water resources and greenhouse gas (GHG) fluxes are unclear. To close the gap between science and management, we have developed a unique field research platform to experimentally evaluate key environmental strategies for drained northern landscapes with the aim to avoid further environmental degeneration. The Trollberget Experimental Area (TEA) includes replicated and controlled treatments applied at the catchment scale based on a BACI approach (before-after and control-impact). The treatments represent the dominant ecosystem types impacted by ditching in Sweden and the boreal zone: (1) rewetting of a drained peatland, (2) ditch cleaning in productive upland forests and (3) leaving these ditches unmanaged. Here we describe the TEA platform, report initial results, suggest ways forward for how to best manage this historical large-scale alteration of the boreal landscape, as well as warn against applying these treatments broadly before more long-term results are reported.

Boreal peatlands represent a biogeochemically unique and diverse environment in high-latitude landscape. They represent a long-term globally significant sink for carbon dioxide and a source of methane, hence playing an important role in regulating the global climate. There is an increasing interest in deciphering peatland biogeochemical processes to improve our understanding of how anthropogenic and climate change effects regulate the peatland biogeochemistry and greenhouse gas balances. At present, most studies investigating land-atmosphere exchanges of peatland ecosystems are commonly based on single-tower setups, which require the assumption of homogeneous conditions during upscaling to the landscape. However, the spatial organization of peatland complexes might feature large heterogeneity due to its varying underlying topography and vegetation composition. Little is known about how well single site studies represent the spatial variations of biogeochemical processes across entire peatland complexes. The recently established Kulbäcksliden Research Infrastructure (KRI) includes five peatland study sites located less than 3 km apart, thus providing a unique opportunity to explore the spatial variation in ecosystem-scale processes across a typical boreal peatland complex. All KRI sites are equipped with eddy covariance flux towers combined with installations for detailed monitoring of biotic and abiotic variables, as well as catchment-scale hydrology and hydrochemistry. Here, we review studies that were conducted in the Kulbäcksliden area and provide a description of the site characteristics as well as the instrumentation available at the KRI. We highlight the value of long-term infrastructures with ecosystem-scale and replicated experimental sites to advance our understanding of peatland biogeochemistry, hydrology, ecology, and its feedbacks on the environment and climate system.

Predicting the fate of carbon in peatlands relies on assumptions of behaviour in response to temperature. Here, the authors show that the temperature dependency of respiratory carbon losses shift strongly over day-night cycles, an overlooked facet causing bias in peatland carbon cycle simulations.

In a global field survey across a wide range of organic soils, the authors find that N2O flux can be predicted by models incorporating soil nitrate concentration (NO3 –), water content and temperature. N2O emission increases with NO3 – and temperature and follows a bell-shaped distribution with water content.

The triglyceride, cholesterol, and phospholipid contents of heart papillary muscle were measured in groups of obviously healthy and diseased females and males on whom either routine or forensic necropsies were performed. In healthy men the triglyceride content was 1·77 ± 1·30 mg/g of wet weight and in women 1·25 ± 0·48 mg/g wet weight. The corresponding values for cholesterol were 1·07 ± 0·24 mg/g and 1·21 ± 0·22 mg/g and those for phospholipids 17·70 ± 5·15 mg/g and 19·65 ± 10·21 mg/g. The differences between the sexes were not significant. The hypertensive or cardiac hypertrophy group had about the same or slightly lower means for lipid content. In the cholelithiasis group, women had significantly high triglyceride values (3·38 ± 2·36 mg/g). The cholesterol values were not significantly elevated in either men or women. In the diabetic group, triglycerides were significantly increased both in men (mean 8·12 ± 0·54 mg/g) and in women (6·85 ± 5·66 mg/g). The cholesterol mean values were also high in both sexes, but the rise was not significant because of the great variation. In the coronary atheroma group, both male and female hospital cases had high triglyceride contents (mean 4·48 ± 4·25 mg/g and 3·65 ± 3·94 mg/g) whereas the forensic cases had only slightly elevated or normal values. Cholesterol assays paralleled the triglyceride ones, but phospholipids showed an inverse trend. The results showed that the lipid content of papillary muscle was increased in diseases where disturbances of lipid metabolism are evident, as in diabetes and cholelithiasis. In coronary atheroma only those cases with advanced obstruction of the arteries were associated with abnormal values of papillary lipids. No increase of the lipid content with age alone was found, nor was there any correlation with obesity. [ABSTRACT FROM PUBLISHER]

This study examined physiochemical conditions and prokaryotic community structure (the bacterial and archaeal 16S rRNA genes and mcrA gene abundances and proportions), and evaluated the effect of reed canary grass cultivation and mineral fertilisation on these factors, in the 60 cm thick residual peat layer of experimental plots located on an abandoned peat extraction area. The archaeal proportion was 0.67-39.56% in the prokaryotic community and the methanogens proportion was 0.01-1.77% in the archaeal community. When bacterial abundance was higher in the top 20 cm of peat, the archaea were more abundant in the 20-60 cm layer and methanogens in the 40-60 cm layer of the residual peat. The bacterial abundance was significantly increased, but archaeal abundance was not affected by cultivation. The fertiliser application had a slight effect on peat properties and on archaeal and methanogen abundances in the deeper layer of cultivated peat. The CH4 emission was positively related to mcrA abundance in the 20-60 cm of the bare peat, while in case of reed canary grass cultivation these two parameters were not correlated. Reed canary grass cultivation mitigated CH4 emission, although methanogen abundance remained approximately the same or even increased in different layers of residual peat under cultivated sites over time. This study supports the outlook of using abandoned peat extraction areas to produce reed canary grass for energy purposes as an advisable land-use practice from the perspective of atmospheric impact in peatland-rich Northern Europe.

Northern peatlands provide a globally important carbon (C) store. Since the beginning of the 20th century, however, large areas of natural peatlands have been drained for biomass production across Fennoscandia. Today, drained peatland forests constitute a common feature of the managed boreal landscape, yet their ecosystem C balance and associated climate impact are not well understood, particularly within the nutrient-poor boreal region. In this study, we estimated the net ecosystem carbon balance (NECB) from a nutrient-poor drained peatland forest and an adjacent natural mire in northern Sweden by integrating terrestrial carbon dioxide (CO 2 ) and methane (CH 4 ) fluxes with aquatic losses of dissolved organic C (DOC) and inorganic C based on eddy covariance and stream discharge measurements, respectively, over two hydrological years. Since the forest included a dense spruce-birch area and a sparse pine area, we were able to further evaluate the effect of contrasting forest structure on the NECB and component fluxes. We found that the drained peatland forest was a net C sink with a 2-year mean NECB of -115 ± 5 g C m -2  year -1 while the adjacent mire was close to C neutral with 14.6 ± 1.7 g C m -2  year -1 . The NECB of the drained peatland forest was dominated by the net CO 2 exchange (net ecosystem exchange [NEE]), whereas NEE and DOC export fluxes contributed equally to the mire NECB. We further found that the C sink strength in the sparse pine forest area (-153 ± 8 g C m -2  year -1 ) was about 1.5 times as high as in the dense spruce-birch forest area (-95 ± 8 g C m -2  year -1 ) due to enhanced C uptake by ground vegetation and lower DOC export. Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km 2 resolution for 0-5 and 5-15 cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1-km 2 pixels (summarized from 8519 unique temperature sensors) across all the world's major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10°C (mean = 3.0 ± 2.1°C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 ± 2.3°C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (-0.7 ± 2.3°C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Nevertheless, we highlight the need to fill remaining geographic gaps by collecting more in situ measurements of microclimate conditions to further enhance the spatiotemporal resolution of global soil temperature products for ecological applications., (© 2022 The Authors. Bottom-up synthetic biology aims to integrate artificial moieties with living cells and tissues. Here, two types of structural scaffolds for artificial organelles were compared in terms of their ability to interact with macrophage-like murine RAW 264.7 cells. The amphiphilic block copolymer poly(cholesteryl methacrylate)- block -poly(2-carboxyethyl acrylate) was used to assemble micelles and polymer-lipid hybrid vesicles together with 1,2-dioleoyl- sn -glycero-3-phosphocholine or 1,2-dioleoyl- sn -glycero-3-phosphoethanolamine (DOPE) lipids in the latter case. In addition, the pH-sensitive fusogenic peptide GALA was conjugated to the carriers to improve their lysosomal escape ability. All assemblies had low short-term toxicity toward macrophage-like murine RAW 264.7 cells, and the cells internalized both the micelles and hybrid vesicles within 24 h. Assemblies containing DOPE lipids or GALA in their building blocks could escape the lysosomes. However, the intracellular retention of the building blocks was only a few hours in all the cases. Taken together, the provided comparison between two types of potential scaffolds for artificial organelles lays out the fundamental understanding required to advance soft material-based assemblies as intracellular nanoreactors.

The regional variability in tundra and boreal carbon dioxide (CO 2 ) fluxes can be high, complicating efforts to quantify sink-source patterns across the entire region. Statistical models are increasingly used to predict (i.e., upscale) CO 2 fluxes across large spatial domains, but the reliability of different modeling techniques, each with different specifications and assumptions, has not been assessed in detail. Here, we compile eddy covariance and chamber measurements of annual and growing season CO 2 fluxes of gross primary productivity (GPP), ecosystem respiration (ER), and net ecosystem exchange (NEE) during 1990-2015 from 148 terrestrial high-latitude (i.e., tundra and boreal) sites to analyze the spatial patterns and drivers of CO 2 fluxes and test the accuracy and uncertainty of different statistical models. CO 2 fluxes were upscaled at relatively high spatial resolution (1 km 2 ) across the high-latitude region using five commonly used statistical models and their ensemble, that is, the median of all five models, using climatic, vegetation, and soil predictors. We found the performance of machine learning and ensemble predictions to outperform traditional regression methods. We also found the predictive performance of NEE-focused models to be low, relative to models predicting GPP and ER. Our data compilation and ensemble predictions showed that CO 2 sink strength was larger in the boreal biome (observed and predicted average annual NEE -46 and -29 g C m -2  yr -1 , respectively) compared to tundra (average annual NEE +10 and -2 g C m -2  yr -1 ). This pattern was associated with large spatial variability, reflecting local heterogeneity in soil organic carbon stocks, climate, and vegetation productivity. Globally, soils store two to three times as much carbon as currently resides in the atmosphere, and it is critical to understand how soil greenhouse gas (GHG) emissions and uptake will respond to ongoing climate change. In particular, the soil-to-atmosphere CO 2 flux, commonly though imprecisely termed soil respiration (R S ), is one of the largest carbon fluxes in the Earth system. An increasing number of high-frequency R S measurements (typically, from an automated system with hourly sampling) have been made over the last two decades; an increasing number of methane measurements are being made with such systems as well. Such high frequency data are an invaluable resource for understanding GHG fluxes, but lack a central database or repository. Here we describe the lightweight, open-source COSORE (COntinuous SOil REspiration) database and software, that focuses on automated, continuous and long-term GHG flux datasets, and is intended to serve as a community resource for earth sciences, climate change syntheses and model evaluation. Contributed datasets are mapped to a single, consistent standard, with metadata on contributors, geographic location, measurement conditions and ancillary data. The design emphasizes the importance of reproducibility, scientific transparency and open access to data. While being oriented towards continuously measured R S , the database design accommodates other soil-atmosphere measurements (e.g. ecosystem respiration, chamber-measured net ecosystem exchange, methane fluxes) as well as experimental treatments (heterotrophic only, etc.). Identifying the atomic structure of organic-inorganic interfaces is challenging with current research tools. Interpreting the structure of complex molecular adsorbates from microscopy images can be difficult, and using atomistic simulations to find the most stable structures is limited to partial exploration of the potential energy surface due to the high-dimensional phase space. In this study, we present the recently developed Bayesian Optimization Structure Search (BOSS) method as an efficient solution for identifying the structure of non-planar adsorbates. We apply BOSS with density-functional theory simulations to detect the stable adsorbate structures of (1 S )-camphor on the Cu(111) surface. We identify the optimal structure among eight unique types of stable adsorbates, in which camphor chemisorbs via oxygen (global minimum) or physisorbs via hydrocarbons to the Cu(111) surface. This study demonstrates that new cross-disciplinary tools, such as BOSS, facilitate the description of complex surface structures and their properties, and ultimately allow us to tune the functionality of advanced materials., (Copyright © 2020, Järvi et al.; licensee Beilstein-Institut.)

Accurate projections of climate change impacts on the vast carbon stores of northern peatlands require detailed knowledge of ecosystem respiration (ER) and its heterotrophic (Rh) and autotrophic (Ra) components. Currently, however, standard flux measurement techniques, i.e. eddy covariance and manual chambers, generate empirical ER data during only night- or daytime, respectively, which are extrapolated to the daily scale based on the paradigm that assumes a uniform diel temperature response. Here, using continuous autochamber measurements, we demonstrate a distinct bimodal pattern in diel peatland ER which contrasts the unimodal pattern inherent to the classical assumption. This feature results from divergent temperature dependencies of day- and nighttime ER due to varying contributions from Rh and Ra. We further find that disregarding these bimodal dynamics causes significant bias in ER estimates across multiple temporal scales. This calls for improved process-based understanding of ER to advance our ability to simulate peatland carbon cycle-climate feedbacks.

The role of plant phenology as a regulator for gross ecosystem productivity (GEP) in peatlands is empirically not well constrained. This is because proxies to track vegetation development with daily coverage at the ecosystem scale have only recently become available and the lack of such data has hampered the disentangling of biotic and abiotic effects. This study aimed at unraveling the mechanisms that regulate the seasonal variation in GEP across a network of eight European peatlands. Therefore, we described phenology with canopy greenness derived from digital repeat photography and disentangled the effects of radiation, temperature and phenology on GEP with commonality analysis and structural equation modeling. The resulting relational network could not only delineate direct effects but also accounted for possible effect combinations such as interdependencies (mediation) and interactions (moderation). We found that peatland GEP was controlled by the same mechanisms across all sites: phenology constituted a key predictor for the seasonal variation in GEP and further acted as a distinct mediator for temperature and radiation effects on GEP. In particular, the effect of air temperature on GEP was fully mediated through phenology, implying that direct temperature effects representing the thermoregulation of photosynthesis were negligible. The tight coupling between temperature, phenology and GEP applied especially to high latitude and high altitude peatlands and during phenological transition phases. Our study highlights the importance of phenological effects when evaluating the future response of peatland GEP to climate change. The net ecosystem CO 2 exchange (NEE) drives the carbon (C) sink-source strength of northern peatlands. Since NEE represents a balance between various production and respiration fluxes, accurate predictions of its response to global changes require an in depth understanding of these underlying processes. Currently, however, detailed information of the temporal dynamics as well as the separate biotic and abiotic controls of the NEE component fluxes is lacking in peatland ecosystems. In this study, we address this knowledge gap by using an automated chamber system established across natural and trenching/vegetation removal plots to partition NEE into its production (i.e., gross and net primary production; GPP and NPP) and respiration (i.e., ecosystem, heterotrophic and autotrophic respiration; ER, Rh and Ra) fluxes in a boreal peatland in northern Sweden. Our results showed that daily NEE patterns were driven by GPP while variations in ER were governed by Ra rather than Rh. Moreover, we observed pronounced seasonal shifts in the Ra/Rh and above/belowground NPP ratios throughout the main phenological phases. Generalized linear model analysis revealed that the greenness index derived from digital images (as a proxy for plant phenology) was the strongest control of NEE, GPP and NPP while explaining considerable fractions also in the variations of ER and Ra. In addition, our data exposed greater temperature sensitivity of NPP compared to Rh resulting in enhanced C sequestration with increasing temperature. Overall, our study suggests that the temporal patterns in NEE and its component fluxes are tightly coupled to vegetation dynamics in boreal peatlands and thus challenges previous studies that commonly identify abiotic factors as key drivers. Nitrous oxide (N 2 O) is a powerful greenhouse gas and the main driver of stratospheric ozone depletion. Since soils are the largest source of N 2 O, predicting soil response to changes in climate or land use is central to understanding and managing N 2 O. Here we find that N 2 O flux can be predicted by models incorporating soil nitrate concentration (NO 3 - ), water content and temperature using a global field survey of N 2 O emissions and potential driving factors across a wide range of organic soils. N 2 O emissions increase with NO 3 - and follow a bell-shaped distribution with water content. Combining the two functions explains 72% of N 2 O emission from all organic soils. Above 5 mg NO 3 - -N kg -1 , either draining wet soils or irrigating well-drained soils increases N 2 O emission by orders of magnitude. As soil temperature together with NO 3 - explains 69% of N 2 O emission, tropical wetlands should be a priority for N 2 O management.

Study Objectives: To investigate the body movement-associated heart rate responses during sleep in postmenopausal women and to evaluate the possible effect of transdermal oestrogen replacement therapy and metabolic factors on these responses., Design: A prospective double-blind cross-over placebo-controlled trial., Setting: A university sleep research unit., Patients: Seventy-one healthy postmenopausal women., Methods: The subjects underwent a whole-night polysomnography including an electrocardiogram (ECG) and a sensitive whole-body movement sensor channel. Body movements of 2-15 s duration with an artefact-free ECG were identified and heart rate was automatically analysed for 30 s before and 60 s after movement. The movement-induced reflex tachycardia and the following rebound bradycardia were measured for the awake state, non-rapid eye movement (REM) sleep and REM sleep. Indices of the heart rate response correlated with age, body mass index (BMI) and high-density lipoprotein (HDL)/low-density lipoprotein (LDL) cholesterol ratio., Results: The tachycardia response to a body movement was stronger during sleep than in the awake state, being strongest in REM sleep. The rebound bradycardia following a tachycardia response was relatively similar in REM and in the awake states, but attenuated in non-REM sleep. Oestrogen did not influence the heart rate response. Increasing age and BMI attenuated the response, while an increase in the HDL/LDL cholesterol ratio enhanced the rebound bradycardia response., Conclusions: Sleep powerfully modulates the rapid cardiac autonomic responses to internal stressors. Postmenopausal hormone replacement therapy does not influence these responses, but they are strongly modulated by lipid metabolism and closely correlated with age and BMI.

The study examined three methodological aspects of expressed emotion (EE) as assessed in the course of PACE (Psychosocial Assessment of Childhood Experiences) interviews with a parent. In a sample of 87 children, aged 6 13 years, enrolled in a prospective study examining the role of stress on the course of asthma, EE was assessed at three time points, 9 months apart. A high degree of agreement was found among the three concurrent measures of negative and positive EE (kappas from 0.74 to 0.97, and from 0.45 to 0.88, respectively; p < or = 0.0001 in all instances). The temporal stability of all measures was lower, although statistically significant in all but 2 instances (kappas from 0.19 to 0.59, and from 0.11 to 0.39, respectively). The temporal stability across measures, as well as across interviewers and over time, was broadly similar (kappas from 0.21 to 0.56 for negative EE, and from 0.09 to 0.38 for positive EE, with all but three of the 36 statistically significant). The findings provide support for the underlying assumptions of the PACE-EE and show the utility of measures based on just very brief periods of non-directive interviewing, making them practical in a wide range of studies with EE just one of a larger set of measures.

31P nuclear magnetic resonance (NMR) spectroscopy enables us to study intracellular energy metabolism noninvasively. The present work concerns the analysis of a series of 31P NMR spectra on human muscle during exercise. The spectra contain signals corresponding to certain metabolites in the sample and the aim is to identify and quantify these signals. We have written a PC program to perform this task automatically. With the program the results can be achieved substantially faster compared to operating with the spectrometer software. The methods implemented in the program and the functions of the program itself are described. Although we have focused on the 31P NMR spectrum series, the program can also be applied to other liquid state NMR spectra.

To study the effects of uremia and hemodialysis on the production rates of antiaggregatory prostacyclin (PGI2) and proaggregatory thromboxane A2 (TxA2), we collected serial plasma samples from eight patients with chronic uremia before, during and after hemodialysis and assayed them for 6-keto-PGF1 alpha and TxB2, the stable metabolites of PGI2 and TxA2, respectively. In addition, the capacity of the platelets to produce TxB2 during spontaneous clotting was studied by measuring the TxB2 levels in serum incubated at +37 degrees C for 60 minutes. The PGI2 production of the uremia patients before hemodialysis was less (P less than 0.001) than that of healthy volunteers. It rose significantly following heparinization and remained elevated during hemodialysis. TxB2 generation by platelets during clotting was diminished in uremia. Plasma TxB2 levels were normal before, but increased during hemodialysis. Thus, profound changes in the PGI2/TxA2-system seem to be associated with uremia and hemodialysis.

We report epidemiologic, clinical, laboratory, and biopsy findings in 14 cases of nephropathia epidemica. The patients were between 19 and 49 years of age. The onset of the disease was characterized by high fever, nausea, headache, backache, abdominal pain, proteinuria, oliguria, hematuria, and uremia. The symptoms subsided rapidly during the polyuria phase, which followed the oliguria stage. Because of renal failure, hemodialysis was required in eight cases. Edema of eyelids, conjunctival injection and hemorrhages, transitory myopia, and acute glaucoma were the most common eye abnormalities. Renal biopsy specimens showed glomerular changes, with mild swelling of the epithelial cells of Bowman's capsule, thickening of the basement membrane of glomerular capillaries, glomerular adhesions, inflammatory cell infiltration, leukocytoclasis and hemorrhages in the interstitium, and eosinophilic hyaline degeneration and vacuolization of the epithelial cells of the proximal tubuli.
