Abstract

This study aimed to examine how regular aerobic training can affect the muscle hypertrophy induced by overloading. Male C57BL/6J mice were randomly divided into three groups: rest group, low-intensity aerobic exercise group, and high-intensity aerobic exercise group. Mice in the exercise groups were assigned to run at a speed of 10 m/min (low-intensity) or 25 m/min (high-intensity) for 30 min/day, five days/week, for four weeks. Then, the right hind leg gastrocnemius muscles were surgically removed to overload the plantaris and soleus muscles, while the left hind leg was subjected to a sham-operation. Both the plantaris and soleus muscles grew larger in the overloaded legs than those in the sham-operated legs. Muscle growth increased in the plantaris muscles in the low-intensity exercise group compared to that in the rest or high-intensity exercise groups at one and two weeks after overloading. This enhancement was not observed in the soleus muscles. Consistently, we observed changes in the expression of proteins involved in anabolic intracellular signaling, including Akt, mechanistic target of rapamycin (mTOR), and p70S6K, in the plantaris muscles. 18. Epitaxial growth and critical currents in (013)/(103)- and (110)-orientedYBa2Cu3Oxfilms

Abstract

Thin films of YBa{sub 2}Cu{sub 3}O{sub {ital x}} were grown epitaxially on (110)-oriented SrTiO{sub 3} crystals using {ital in} {ital situ}, off-axis, rf magnetron sputter deposition. The films were investigated by x-ray diffraction, reflection high-energy electron diffraction, scanning electron microscopy, resistance-temperature, and critical current measurements. Depending on growth conditions, the films formed platelets oriented with YBa{sub 2}Cu{sub 3}O{sub {ital x}} (110) or (013)/(103) parallel to the (110) substrates. Direct comparison of simultaneous growth on (110)- and (100)-oriented substrates showed that the critical temperature was significantly less for growth on the (100) surface for the same growth conditions. This is ascribed to the more open structure of the (110) and (013)/(103) planes which allow more complete oxidation of the film during growth and cool down. For growth on (110) substrates and substrate temperatures below about 650 {degree}C the (110) orientation dominated; above 650 {degree}C the films were mainly (013) or (103) oriented. The critical current density for the (013)/(103)-oriented films was 0.25 MA cm{sup {minus}2} at 77 K. This compares with about 5 MA cm{sup {minus}2} for {ital J}{sub {ital c}} parallel to the {ital ab} plane and 10 kA cm{sup {minus}1} for {ital J}{sub {ital c}} parallel to the {italmore » c} axis. The temperature dependence of the (013)/(103) films was {ital J}{sub {ital c}}{proportional_to}(1{minus}{ital T}/{ital T}{sub {ital c}}){sup 3/2}, 0.9{ital T}{sub {ital c}}{lt}{ital T}{lt}{ital T}{sub {ital c}}. This suggests that the measured critical currents correspond to the depairing current, although intergranular twinning gives a similar temperature dependence. The overall results are discussed in relation to the nucleation and growth kinetics of the different epitaxial modes and also the effects of energetic particles on sputter deposition.« less

Abstract

AbstractObjective: Childhood onset Graves’ disease (GD) has been documented to be clinically distinct from adult onset GD, and an association with the genes encoding HLA and CTLA-4 (cytotoxic T lymphocyte antigen-4) has been reported in both Caucasian and Japanese adult GD patients. The aim of this study was to determine whether HLA-DR, -DQ and CTLA-4 are associated with childhood onset GD in Japanese individuals. Methods: We investigated the genotype of HLA class II (DRB1, DQB1) and the A/G transition polymorphism of CTLA-4 exon 1 position 49 in 43 GD patients and in healthy controls for comparison. The CTLA-4 alleles were identified by the polymerase chain reaction (PCR) of genomic DNA and restriction fragment-length polymorphism analysis (PCR-RFLP) with Ita1. Results: The frequency of both HLA-DRB1*0405 and DQB1*0401 was increased in the patient group (DRB1*0405: 26.7%, p < 0.001; DQB1*0401: 25.6%, p < 0.005) compared with the controls. Patients with GD had a significantly lower frequency of the AA genotype of CTLA-4 than the controls, but there was no difference in allele frequency between the G and A allele. Conclusions: the association of HLA-DRB1 and DQB1 genotype with susceptibility to childhood onset GD differs from that in adult onset GD, whereas the association between CTLA-4 gene polymorphism and childhood onset GD is similar to that in adult onset GD in Japanese individuals, but the association is weak.Copyright © 2005 S. 45. Novel Ligands Lacking a Positive Charge for the δ- and μ-Opioid Receptors

Abstract

Recently we reported using minilibraries to replace Lys9 [somatostatin (SRIF) numbering] of the potent somatostatin agonist L-363,301 (c[-Pro-Phe-d-Trp-Lys-Thr-Phe-]) to generate the potent neurokinin receptor (NK-1) antagonist c[-Pro-Phe-d-Trp-p-F-Phe-Thr-Phe-]. This novel cyclic hexapeptide did not bind the SRIF receptor. Thus, a single mutation converted L-363,301, a SRIF agonist with potency ca. 2−8 times the potency of SRIF in laboratory animals,24 into a selective NK-1 receptor antagonist with an IC50 of 2 nM in vitro. During the screening of the same libraries for ligands of the δ-opioid receptor, we identified four compounds (1−4) which represent a new class of δ-opioid antagonists, some of which were also NK-1 receptor antagonists. The most potent δ-opioid antagonist, c[-Pro-1-Nal-d-Trp-Tyr-Thr-Phe-] (2), showed a Ke value of 128 nM in the mouse vas deferens assay and a δ-receptor binding affinity constant of 152 nM in the rat brain membrane binding assay. These results are of interest because they represent a novel class of δ-opioid antagonists and, like two previously reported δ-opioid antagonists, they lack a positive charge. To examine further the requirement for a positive charge in the δ-opioid ligands, we prepared two analogues of the β-casomorphin-derived mixed μ-agonist/δ-antagonist, H-Dmt-c[-d-Orn-2-Nal-d-Pro-Gly-] (7), in which we eliminated the positive charge either through formylation of the primary amino group (5) or by the deletion of this N-terminal amino group (6). These latter compounds proved to be δ-opioid antagonists with Ke values in the 16−120 nM range, as well as fairly potent μ-opioid antagonists (Ke ≈ 200 nM). These six compounds provide the most convincing evidence to date that there is no requirement for a positive charge in μ- and δ-opioid receptor antagonists. In addition, cyclic hexapeptide 4 lacks a phenolic hydroxyl group. Taken together, these data suggest that the prevailing assumptions about δ- and μ-opioid receptor binding need revision and that the receptors for these opioid ligands have much in common with the NK-1 and somatostatin receptors.

Abstract

Chemoembolization by selective intra-arterial infusion of mitomycin C in microcapsule form exerts potential therapeutic effects through infarction and prolonged activity. Of 20 patients with inoperable hepatocellular carcinoma (HCC) treated with one or more courses via a hepatic arterial catheter, a change in tumor size could be measured in 16. Measurable tumor regression amounting to greater than 50% was shown in 6 patients (38%), 25-50% in 3 (19%), and less than or equal to 25% in 6 (38%), while tumor increased in 1. Serum alpha-fetoprotein, which was greater than 300 ng/ml before treatment in 13 patients, clearly fell after treatment in 10 (77%). The survival rate at 3, 6, and 12 months after treatment was 75%, 65%, and 24%, respectively, for all patients, and 92%, 85%, and 38% in 11 patients without a tumor cast in the portal vein or its major branches. Systemic toxicity was mild, and all patients tolerated treatment well. These results indicate that chemoembolization with mitomycin C microcapsules is a useful palliative measure for treatment of inoperable HCC.
