Abstract

It is well known that the main parts of Solid Oxide Fuel Cells (SOFCs) are build from ceramic materials. Namely the ceramic materials and composites, used for SOFCs manufacturing, are objects of the overview in the present work. The analysis carried out covers the last current publications in the field discussed. Special attention and examination in details have been done on patents state-of-the-art. After a background and short classification of the ceramic SOFCs materials the attention is focused on cathode, electrolyte, anode, interconnection and sealing materials. Their requirements, structure, thermal stability, composition control and behavior, processing and performance are the object of overview. A correlation has been made between the phase diagrams oxygen incorporation and transport, and SOFC advantages, generally for materials of lanthanum- base perovskite family. In order to analyze the innovative investigations regarding the patent branch of the SOFCs development and application, an object of review was patents from Japan, USA, Germany and European Union. Some examples of the inventions with accent on the ceramic materials are shown. In addition the tendency regarding R & D activities of SOFCs development materials from the leading companies in the world is analyzed. On the base of the most important technological and economical parameters of cell cathode/electrolyte/anode materials an attempt for evaluation and correlation has been made and innovative conceptions are shown.Es bien sabido que los componentes principales de las celdas de combustible de óxido sólido (SOFCs) estan constituidos por materiales cerámicos. Dichos materiales cerámicos y materiales compuestos que se utilizan en la fabricación de SOFCs son objeto de estudio en el presente trabajo. El análisis llevado a cabo incluye la revisión de las últimas publicaciones en la materia, con una especial atención y examen minucioso sobre las patentes más relevantes. Después de una revisión y breve clasificación de los materiales SOFCs cerámicos, el estudio se centra en la descripción del cátodo, electrolito y ánodo, así como de la unión y materiales de sellado. También se han estudiado sus requisitos, estructura, estabilidad térmica, control composicional y de comportamiento, procesado y rendimiento. Se ha establecido una correlación entre los diagramas de fase de incorporación y transporte de oxígeno y las ventajas de los SOFC, generalmente para materiales de la familia de las perovsquitas basadas en lantano. Con el fin de analizar las investigaciones innovadoras que conciernen al campo del desarrollo y aplicación de SOFCs, se han revisado patentes de Japón, Estados Unidos, Alemania y la Unión Europea. Se dan algunos ejemplos de dichas patentes en las que destaca el papel de los materiales cerámicos. Además, se ha analizado la tendencia en actividades de I + D sobre los SOFCs en compañías líderes mundiales. Se ha intentado realizar una evaluación y correlación en base a los parámetros tecnológicos y económicos más importantes de los materiales de celda cátodo/electrolito/ánodo y se han mostrado conceptos innovadores.

Abstract

The Human Major Histocompatibility Complex (HLA) is the most polymorphic region of the human genome and encodes molecules with a central role in antigen-specific immune responses. Class I and class II HLA genes have an important role in bone marrow and solid organ transplantations. HLA typing by Next Generation Sequencing (NGS) provides unambiguous second field or allelic level resolution and, therefore, better matching of donor and recipient and a higher chance of survival for transplanted patients. The aim of this study was to validate GenDx NGS HLA typing protocol for 6 HLA loci (HLA-A, -B, -C, DRB1, -DQB1 and DPB1).

Abstract

The Adolescent Brain Cognitive Development (ABCD) Study® is a 10-year longitudinal study of children recruited at ages 9 and 10. A battery of neuroimaging tasks are administered biennially to track neurodevelopment and identify individual differences in brain function. This study reports activation patterns from functional MRI (fMRI) tasks completed at baseline, which were designed to measure cognitive impulse control with a stop signal task (SST; N = 5,547), reward anticipation and receipt with a monetary incentive delay (MID) task (N = 6,657) and working memory and emotion reactivity with an emotional N-back (EN-back) task (N = 6,009). Further, we report the spatial reproducibility of activation patterns by assessing between-group vertex/voxelwise correlations of blood oxygen level-dependent (BOLD) activation. Analyses reveal robust brain activations that are consistent with the published literature, vary across fMRI tasks/contrasts and slightly correlate with individual behavioral performance on the tasks. These results establish the preadolescent brain function baseline, guide interpretation of cross-sectional analyses and will enable the investigation of longitudinal changes during adolescent development.

43. Structural and biochemical characterisation of PKCι assemblies and substrate targeting mechanisms

Abstract

The PKCι (protein kinase C isoform ι) plays crucial roles in the formation and maintenance of cell polarity through the formation of the Par complex (PKCι, Par6, Cdc42, Par3), a master regulator of epithelial apical-basal polarity. The Par complex is an apical polarity marker and is directly opposed by the baso-lateral markers Lgl, Dlg and Scribble by mutual antagonism. The molecular basis for the mutual antagonism between the complexes is unclear but may involve direct inhibition of PKCι activity. Conversely, several proteins able to inhibit PKCι are also functionally validated substrates, for example Lgl, Par3 and Kibra. The focus of this thesis has been to explore the molecular details of PKCι regulation and RIPR-dependent substrate recruitment. Different forms of PKCι have been characterised using biophysical, biochemical and structural biology methods. An active mature PKCι kinase domain structure was defined at high resolution. Comparison with a Par3-inhibited conformer reveals how inhibition perturbs the active conformer. To probe the molecular interaction between PKCι and RIPR-dependent substrates, Llgl1 and Periphillin1, peptides from these proteins were characterised and crystallised with the PKCι kinase domain. Amino-terminal sequences flanking their PKC consensus motif make contacts exclusively with the PKCι C-lobe. Surprisingly, both Llgl1 and Periphillin1 peptides structures lacked a bound nucleotide within of PKCι. In contrast, an ATP-bound PKCι lacked the Periphillin substrate peptide. These data suggest that RIPR-dependent substrate peptides may influence nucleotide loading of PKCι. Finally, the thesis explored a second mechanism for Par6-PKCι recruitment to the apical domain through Crb-Pals1 interaction. The basis for Crb intracellular domain interaction with Pals1 and the Par complex was defined indicating contacts from the Pals-PDZ-SH3 unit are required to engage Crb. The higher Pals1 affinity for Crb than the Par complex, suggests a hierarchy of substrate interactions to recruit Par6-PKCι through the Crb complex.

Abstract

Secondary neoplastic lesions in lymph nodes are predominantly metastases from solid tumors, whereas primary lymph node hemangiomas are exceptionally uncommon, with only 24 well-documented cases in the literature. Histologically, they are characterized by endothelial cells that may appear flattened or enlarged, with variable vascular density, and the presence of stromal elements. Notably, the concurrent presence of a primary hemangioma and a metastasis from breast cancer – the latter being the most prevalent secondary lesion in axillary lymph nodes – represents an unprecedented observation. The unique case presented herein underscores the exceptional rarity of primary lymph node hemangiomas and demonstrates for the first time their possible coexistence with breast cancer metastasis within the same axillary lymph node. In sharing and discussing this case study, we pay homage to Professor Juan Rosai, whose work in redefining rare and complex diagnoses continues to enlighten our understanding of lymph node vascular lesions.

Abstract

The diagnostic assessment of thyroid nodules is hampered by the persistence of uncertainty in borderline cases, and further complicated by the inclusion of non-invasive follicular tumor with papillary-like nuclear features (NIFTP) as a less aggressive alternative to papillary thyroid carcinoma (PTC). In this setting, computational methods might facilitate the diagnostic process by unmasking key nuclear characteristics of NIFTPs. The main aims of this work were to (1) identify morphometric features of NIFTP and PTC that are interpretable for the human eye, and (2) develop a deep learning model for multi-class segmentation as a support tool to reduce diagnostic variability. Our findings confirmed that nuclei in NIFTP and PTC share multiple characteristics, setting them apart from hyperplastic nodules (HP). The morphometric analysis identified 15 features that can be translated into nuclear alterations readily understandable by pathologists, such as a remarkable inter-nuclear homogeneity for HP in contrast to a major complexity in the chromatin texture of NIFTP, and to the peculiar pattern of nuclear texture variability of PTC. A few NIFTP cases with available NGS data were also analyzed to initially explore the impact of RAS-related mutations on nuclear morphometry. Finally, a pixel-based deep learning model was trained and tested on whole slide images (WSIs) of NIFTP, PTC, and HP cases. The model, named NUTSHELL (NUclei from Thyroid tumors Segmentation to Highlight Encapsulated Low-malignant Lesions), successfully detected and classified the majority of nuclei in all WSIs' tiles, showing comparable results with already well-established pathology nuclear scores. NUTSHELL provides an immediate overview of NIFTP areas and can be used to detect microfoci of PTC within extensive glandular samples or identify lymph node metastases. NUTSHELL can be run inside WSInfer with an easy rendering in QuPath, thus facilitating the democratization of digital pathology.
