The effects of feeding an 80% plant protein diet, with and without fish protein hydrolysate (FPH) supplementation, on the growth and gut health of Atlantic salmon were investigated. Fish were fed either (A) a control diet containing 35% fishmeal, (B) an 80% plant protein diet with 15% fishmeal, (C) an 80% plant protein diet with 5% fishmeal and 10% partly hydrolysed protein, or (D) an 80% plant protein diet with 5% fishmeal and 10% soluble protein hydrolysate. Fish on the 80% plant- 15% fishmeal diet were significantly smaller than fish in the other dietary groups. However, partly-hydrolysed protein supplementation allowed fish to grow as well as fish fed the control 35% fishmeal diet. Fish fed the FPH diets (diets C and D) had significantly higher levels of amino acids in their blood, including 48% and 27% more branched chain amino acids compared to fish on the 35% fishmeal diet, respectively. Plant protein significantly altered gut microbial composition, significantly decreasing α-diversity. Spirochaetes and the families Moritellaceae, Psychromonadaceae, Helicobacteraceae and Bacteroidaceae were all found at significantly lower abundances in the groups fed 80% plant protein diets compared to the control fishmeal diet.

In this study, wheat and oat bran fibres were used to investigate the potential beneficial prebiotic-enhancing effects of superheated steam (SHS) treatment. Following SHS treatment of fibres, in-vitro simulated gastric and intestinal digestion followed by batch culture fermentation was performed. 16S rRNA gene sequencing and short chain fatty acid analysis were conducted, which demonstrated that SHS treatment was associated with significant differences in beta diversity and the relative abundances of several microbial taxa. Principal Coordinate Analysis (PCoA) revealed significant separation between SHS-treated oat and non-treated oat (p = 0.013), and between SHS-treated wheat and non-treated wheat following in vitro batch fermentation (p = 0.042). Abundances of Ruminococcus were found to be significantly higher by 1.09-log in SHS-treated oat when compared with non-treated oat while abundances of Escherichia_Shigella, and Alistipes were significantly lower by 1.47-log and 0.39-log, respectively in the SHS-treated oat samples than the non-treated oat samples (p = 6.85E-06, p = 0.002037, p = 0.002458; respectively). These data demonstrate that SHS treatment of dietary fibre enhanced its effectiveness to modulate the gut microbiome. Industrial Relevance: Superheated steam (SHS) treatment has emerged as an effective method to improve the shelf-life of grains by reducing lipid hydrolysis and oxidation. Our results revealed that SHS treatment of wheat and oat brans led to increased physiological and chemical properties including higher water-binding capacity, water-extractable arabinoxylan, and total phenolics as well as improvement in human gut microbiota. Our results suggest that this method could be implemented in the food industry in order to improve beneficial properties of oat and wheat brans.

Bacteriocins (ribosomally produced antimicrobial peptides) are potential alternatives to current antimicrobials given the global challenge of antimicrobial resistance. We identified a novel bacteriocin from Actinomyces ruminicola with no previously characterized antimicrobial activity. Using publicly available genomic data, we found a highly conserved yet divergent family of previously unidentified homologous peptide sequences within the genus Actinomyces with striking similarity to eukaryotic defensins. These actifensins may provide a potent line of antimicrobial defense/offense, and the machinery to produce them could be used for the design of new antimicrobials given the degeneracy that exists naturally in their structure., We identified a strain of Actinomyces ruminicola which produces a potent bacteriocin with activity against a broad range of Gram-positive bacteria, many of which are pathogenic to animals and humans. The bacteriocin was purified and found to have a mass of 4,091 ± 1 Da with a sequence of GFGCNLITSNPYQCSNHCKSVGYRGGYCKLRTVCTCY containing three disulfide bridges. Surprisingly, near relatives of actifensin were found to be a series of related eukaryotic defensins displaying greater than 50% identity to the bacteriocin. A pangenomic screen further revealed that production of actifensin-related bacteriocins is a common trait within the genus, with 47 being encoded in 161 genomes. Furthermore, these bacteriocins displayed a remarkable level of diversity with a mean amino acid identity of only 52% between strains/species. This level of redundancy suggests that this new class of bacteriocins may provide a very broad structural basis on which to deliver and design new broad-spectrum antimicrobials for treatment of animal and human infections. Milk consumption is estimated at billions of liters worldwide, the majority of which is consumed as pasteurized cow’s milk. In recent years there has been an increase in the consumption of raw milk due to perceived health benefits compared with heat-treated milk, including increased nutritional value and the potential presence of probiotic bacteria. However, raw milk and milk product consumption poses a significant health risk associated with ingestion of foodborne pathogens and consequent zoonotic illnesses. Contamination of raw milk generally occurs from environmental sources, or from sick animals. Good farming practices and effective post processing are generally sufficient for the eradication of milk-borne pathogens. Annually, a number of human illnesses are associated with consumption of raw milk worldwide, and the severity of these varies from mild symptoms to life threatening infections. In recent times, there have been calls for legislation preventing the sale of unpasteurized milk in many developed countries.

Liquid milk is a highly nutritious beverage which has seen a decline in consumption in recent years. Milk is an excellent source of dietary fats, proteins, and minerals such as calcium and magnesium, particularly for growing children. There is an ongoing debate on the potential benefits associated with the consumption of raw milk over its processed counterpart. Physiological and environmental factors can affect the composition and quality of milk and cows feeding system has been identified as an important factor which can alter milks nutritional status. Pasture-based feeding systems have been shown to produce milks with significantly higher conjugated linoleic acid and omega 3 fatty acid contents over more conventional indoor total mixed ration feeding systems. However, the consumption of raw milk poses a very real and serious health risk, through potential ingestion of pathogenic bacteria. The argument against treated milk has focused on the depletion of milks nutritional quality as a result of heat during processing, but these claims have been found to have no scientific basis. Nevertheless, it is widely agreed upon by experts that the risk of exposure to pathogenic bacteria in raw milk far outweighs such potential benefits.

Microbial fermentation has been used historically for the preservation of foods, the health benefits of which have since come to light. Early dairy fermentations depended on the spontaneous activity of the indigenous microbiota of the milk. Modern fermentations rely on defined starter cultures with desirable characteristics to ensure consistency and commercial viability. The selection of defined starters depends on specific phenotypes that benefit the product by guaranteeing shelf life and ensuring safety, texture, and flavour. Lactic acid bacteria can produce a number of bioactive metabolites during fermentation, such as bacteriocins, biogenic amines, exopolysaccharides, and proteolytically released peptides, among others. Prebiotics are added to food fermentations to improve the performance of probiotics. It has also been found that prebiotics fermented in the gut can have benefits that go beyond helping probiotic growth. Studies are now looking at how the fermentation of prebiotics such as fructo-oligosaccharides can help in the prevention of diseases such as osteoporosis, obesity, and colorectal cancer. The potential to prevent or even treat disease through the fermentation of food is a medically and commercially attractive goal and is showing increasing promise. However, the stringent regulation of probiotics is beginning to detrimentally affect the field and limit their application.
