5. Role of Royal Jelly Treated Adipose-Derived Stem Cell-Extracellular Vesicles on Fibroblast Proliferation, Migration, and Collagen Production

Abstract

Extracellular vesicles (EVs) secreted from adipose-derived stem cells (ADSCs) are known to exhibit collagen synthesis and migration activity in fibroblasts. These EVs can be used as a regenerative medicine and for cosmetic medicinal materials. Royal jelly (RJ) is produced from the hypopharyngeal and mandibular glands of honeybees (nurse bee), which contains apicin and 10-hydroxy-2-decenoic acid, compounds that promote fibroblast proliferation and collagen and elastin synthesis. Here, we investigated whether RJ could further enhance the physiological function of ADSC-EVs on fibroblasts. The findings confirmed that lyophilized RJ and enzyme-treated RJ enhanced the secretion of EVs by ADSCs, increasing the proliferation, migration, and collagen synthesis of fibroblasts compared to those induced by conventional ADSC-EVs. Bioinformatics analysis was conducted using various software programs, based on the data obtained from comprehensive gene expression and small RNA profiling analyses through next-generation sequencing; the results suggested that the RJ treatment of ADSC-EVs significantly enhanced the expression of genes related to extracellular matrix composition. Our findings suggest that the ADSC-EVs qualitatively altered using RJ could offer a more effective therapeutic material for regenerative and cosmetic medicine than conventional ADSCs-EVs.

Abstract

Acne vulgaris, typically caused by Cutibacterium acnes (C. acnes) involves chronic inflammation of the sebaceous follicles and is the most common skin disease, afflicting 85% of adolescents. We previously isolated 2 novel phenolic glycosides, 2-caffeoyl-3-hydroxy-3-methylbutyric 4′-β-D-glucopyranosyloxy-3′-hydroxybenzyl ester (citrulluside H [CH]) and 2-caffeoyl-3-hydroxy-3-methylbutyric 4′-β- d -glucopyranosyloxybenzyl ester (citrulluside T [CT]), from young fruits of watermelon (Citrullus lanatus). Both compounds suppressed UVB-induced photoaging in human fibroblasts by scavenging intracellular reactive oxygen species (ROS) and thus might be useful as natural skin care ingredients. In this study, we examined the inhibitory effects of these phenolic glycosides on C. acnes growth and C.acnes- induced inflammation. Neither phenolic glycoside inhibited the growth of C. acnes. However, they both significantly suppressed toll-like receptor (TLR) 1/2 or TLR2/6/nuclear factor κB (NF-κB) signaling in heat-killed C. acnes (hk- C. acnes) - stimulated RAW264.7 cells. Additionally, both phenolic glycosides decreased the expression of M1 macrophage biomarkers (cluster of differentiation [ CD ] 80, CD86, and inducible NO synthase [ iNOS ]), suggesting that they attenuate M1 macrophage activation. First published September 2, 2009; doi: 10.1152/ajpcell.00117.2009--Aquaporin 12 (AQPI2) is the most recently identified member of the mammalian AQP family and is specifically expressed in pancreatic acinar cells. In vitro expression studies have revealed that AQP12 is localized at intracellular sites. To determine the physiological roles of AQP12 in the pancreas, we generated knockout mice for this gene (AQP12-KO). No obvious differences were observed under normal conditions between wild-type (WT) and AQP12-KO mice in terms of growth, blood chemistry, pancreatic fluid content, or histology. However, when we induced pancreatitis through the administration of a cholecystokinin-8 (CCK-8) analog, the AQP12-KO mice showed more severe pathological damage to this organ than WT mice. Furthermore, when we analyzed exocytosis in the pancreatic acini using a two-photon excitation imaging method, the results revealed larger exocytotic vesicles (vacuoles) in the acini of AQP12-KO mice at a high CCK-8 dose (100 nM). Abstract

The operation of two-dimensional electron gas field-effect transistors at low temperatures isstudied as a function of stress biases and stress time. Experiments conducted on 2DEGFRTs with a 0.2-micrometer gate length demonstrate the severe current-voltage collapse properties of such devices when the stress drain voltage is 1.2 V. However, recovery from the collapse occurs when the applied drain voltage is more than 1.6 V.
