Abstract: Mid-infrared emission features probe the properties of ionized gas, and hot or warm molecular gas. The Orion Bar is a frequently studied photodissociation region (PDR) containing large amounts of gas under these conditions, and was observed with the MIRI IFU aboard JWST as part of the "PDRs4All" program. The resulting IR spectroscopic images of high angular resolution (0.2") reveal a rich observational inventory of mid-IR emission lines, and spatially resolve the substructure of the PDR, with a mosaic cutting perpendicularly across the ionization front and three dissociation fronts. We extracted five spectra that represent the ionized, atomic, and molecular gas layers, and measured the most prominent gas emission lines. An initial analysis summarizes the physical conditions of the gas and the potential of these data. We identified around 100 lines, report an additional 18 lines that remain unidentified, and measured the line intensities and central wavelengths. The H I recombination lines originating from the ionized gas layer bordering the PDR, have intensity ratios that are well matched by emissivity coefficients from H recombination theory, but deviate up to 10% due contamination by He I lines. We report the observed emission lines of various ionization stages of Ne, P, S, Cl, Ar, Fe, and Ni, and show how certain line ratios vary between the five regions. We observe the pure-rotational H$_2$ lines in the vibrational ground state from 0-0 S(1) to 0-0 S(8), and in the first vibrationally excited state from 1-1 S(5) to 1-1 S(9). We derive H$_2$ excitation diagrams, and approximate the excitation with one thermal (~700 K) component representative of an average gas temperature, and one non-thermal component (~2700 K) probing the effect of UV pumping. Abstract: Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photo-dissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, impacting planet formation within the disks. We report JWST and Atacama Large Millimetere Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modelling their kinematics and excitation allows us to constrain the physical conditions within the gas. We quantify the mass-loss rate induced by the FUV irradiation, finding it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.

Abstract: Forty years ago it was proposed that gas phase organic chemistry in the interstellar medium was initiated by the methyl cation CH3+, but hitherto it has not been observed outside the Solar System. Alternative routes involving processes on grain surfaces have been invoked. Here we report JWST observations of CH3+ in a protoplanetary disk in the Orion star forming region. Abstract: Water is a key ingredient for the emergence of life as we know it. Yet, its destruction and reformation in space remains unprobed in warm gas. Here, we detect the hydroxyl radical (OH) emission from a planet-forming disk exposed to external far-ultraviolet (FUV) radiation with the James Webb Space Telescope. The observations are confronted with the results of quantum dynamical calculations. The highly excited OH infrared rotational lines are the tell-tale signs of H2O destruction by FUV. The OH infrared ro-vibrational lines are attributed to chemical excitation via the key reaction O+H=OH+H which seeds the formation of water in the gas-phase. We infer that the equivalent of the Earth ocean's worth of water is destroyed per month and replenished. These results show that under warm and irradiated conditions water is destroyed and efficiently reformed via gas-phase reactions. This process, assisted by diffusive transport, could reduce the HDO/H2O ratio in the warm regions of planet-forming disks., Comment: Version submitted to Nature Astronomy

Abstract: (Abridged) We investigate the impact of radiative feedback from massive stars on their natal cloud and focus on the transition from the HII region to the atomic PDR (crossing the ionisation front (IF)), and the subsequent transition to the molecular PDR (crossing the dissociation front (DF)). We use high-resolution near-IR integral field spectroscopic data from NIRSpec on JWST to observe the Orion Bar PDR as part of the PDRs4All JWST Early Release Science Program. The NIRSpec data reveal a forest of lines including, but not limited to, HeI, HI, and CI recombination lines, ionic lines, OI and NI fluorescence lines, Aromatic Infrared Bands (AIBs including aromatic CH, aliphatic CH, and their CD counterparts), CO2 ice, pure rotational and ro-vibrational lines from H2, and ro-vibrational lines HD, CO, and CH+, most of them detected for the first time towards a PDR. Their spatial distribution resolves the H and He ionisation structure in the Huygens region, gives insight into the geometry of the Bar, and confirms the large-scale stratification of PDRs. We observe numerous smaller scale structures whose typical size decreases with distance from Ori C and IR lines from CI, if solely arising from radiative recombination and cascade, reveal very high gas temperatures consistent with the hot irradiated surface of small-scale dense clumps deep inside the PDR. The H2 lines reveal multiple, prominent filaments which exhibit different characteristics. This leaves the impression of a "terraced" transition from the predominantly atomic surface region to the CO-rich molecular zone deeper in. Abstract: The JWST has captured the most detailed and sharpest infrared images ever taken of the inner region of the Orion Nebula, the nearest massive star formation region, and a prototypical highly irradiated dense photo-dissociation region (PDR). We investigate the fundamental interaction of far-ultraviolet photons with molecular clouds. The transitions across the ionization front (IF), dissociation front (DF), and the molecular cloud are studied at high-angular resolution. These transitions are relevant to understanding the effects of radiative feedback from massive stars and the dominant physical and chemical processes that lead to the IR emission that JWST will detect in many Galactic and extragalactic environments. Due to the proximity of the Orion Nebula and the unprecedented angular resolution of JWST, these data reveal that the molecular cloud borders are hyper structured at small angular scales of 0.1-1" (0.0002-0.002 pc or 40-400 au at 414 pc). A diverse set of features are observed such as ridges, waves, globules and photoevaporated protoplanetary disks. At the PDR atomic to molecular transition, several bright features are detected that are associated with the highly irradiated surroundings of the dense molecular condensations and embedded young star. Toward the Orion Bar PDR, a highly sculpted interface is detected with sharp edges and density increases near the IF and DF. This was predicted by previous modeling studies, but the fronts were unresolved in most tracers. A complex, structured, and folded DF surface was traced by the H2 lines. This dataset was used to revisit the commonly adopted 2D PDR structure of the Orion Bar. JWST provides us with a complete view of the PDR, all the way from the PDR edge to the substructured dense region, and this allowed us to determine, in detail, where the emission of the atomic and molecular lines, aromatic bands, and dust originate.

Abstract: (Abridged) Mid-infrared observations of photodissociation regions (PDRs) are dominated by strong emission features called aromatic infrared bands (AIBs). The most prominent AIBs are found at 3.3, 6.2, 7.7, 8.6, and 11.2 $\mu$m. The most sensitive, highest-resolution infrared spectral imaging data ever taken of the prototypical PDR, the Orion Bar, have been captured by JWST. We provide an inventory of the AIBs found in the Orion Bar, along with mid-IR template spectra from five distinct regions in the Bar: the molecular PDR, the atomic PDR, and the HII region. We use JWST NIRSpec IFU and MIRI MRS observations of the Orion Bar from the JWST Early Release Science Program, PDRs4All (ID: 1288). We extract five template spectra to represent the morphology and environment of the Orion Bar PDR. The superb sensitivity and the spectral and spatial resolution of these JWST observations reveal many details of the AIB emission and enable an improved characterization of their detailed profile shapes and sub-components. While the spectra are dominated by the well-known AIBs at 3.3, 6.2, 7.7, 8.6, 11.2, and 12.7 $\mu$m, a wealth of weaker features and sub-components are present. We report trends in the widths and relative strengths of AIBs across the five template spectra. These trends yield valuable insight into the photochemical evolution of PAHs, such as the evolution responsible for the shift of 11.2 $\mu$m AIB emission from class B$_{11.2}$ in the molecular PDR to class A$_{11.2}$ in the PDR surface layers. This photochemical evolution is driven by the increased importance of FUV processing in the PDR surface layers, resulting in a "weeding out" of the weakest links of the PAH family in these layers. Abstract: Kilonovae produced by the coalescence of compact binaries with at least one neutron star are promising standard sirens for an independent measurement of the Hubble constant ($H_0$). Through their detection via follow-up of gravitational-wave (GW), short gamma-ray bursts (sGRBs) or optical surveys, a large sample of kilonovae (even without GW data) can be used for $H_0$ contraints. Here, we show measurement of $H_0$ using light curves associated with four sGRBs, assuming these are attributable to kilonovae, combined with GW170817. Including a systematic uncertainty on the models that is as large as the statistical ones, we find $H_0 = 73.8^{+6.3}_{-5.8}$\,$\mathrm{km}$ $\mathrm{s}^{-1}$ $\mathrm{Mpc}^{-1}$ and $H_0 = 71.2^{+3.2}_{-3.1}$\,$\mathrm{km}$ $\mathrm{s}^{-1}$ $\mathrm{Mpc}^{-1}$ for two different kilonova models that are consistent with the local and inverse-distance ladder measurements. Abstract: The observation of a compact object with a mass of $2.50-2.67M_{\odot}$ on August 14, 2019, by the LIGO Scientific and Virgo collaborations (LVC) has the potential to improve our understanding of the supranuclear equation of state. While the gravitational-wave analysis of the LVC suggests that GW190814 likely was a binary black hole system, the secondary component could also have been the heaviest neutron star observed to date. We use our previously derived nuclear-physics-multimessenger astrophysics framework to address the nature of this object. Based on our findings, we determine GW190814 to be a binary black hole merger with a probability of $>99.9\%$. Even if we weaken previously employed constraints on the maximum mass of neutron stars, the probability of a binary black hole origin is still $\sim 81\%$. Furthermore, we study the impact that this observation has on our understanding of the nuclear equation of state by analyzing the allowed region in the mass-radius diagram of neutron stars for both a binary black hole or neutron star--black hole scenario. We find that the unlikely scenario in which the secondary object was a neutron star requires rather stiff equations of state with a maximum speed of sound $c_s\geq \sqrt{0.6}$ times the speed of light, while the binary black hole scenario does not offer any new insight., Comment: 9 pages, 3 figures, published version

Abstract: Observations of neutron-star mergers based on distinct messengers, including gravitational waves and electromagnetic signals, can be used to study the behavior of matter denser than an atomic nucleus, and to measure the expansion rate of the Universe described by the Hubble constant. We perform a joint analysis of the gravitational-wave signal GW170817 with its electromagnetic counterparts AT2017gfo and GRB170817A, and the gravitational-wave signal GW190425, both originating from neutron-star mergers. We combine these with previous measurements of pulsars using X-ray and radio observations, and nuclear-theory computations using chiral effective field theory to constrain the neutron-star equation of state. We find that the radius of a $1.4$ solar mass neutron star is $11.75^{+0.86}_{-0.81}\ \rm km$ at $90\%$ confidence and the Hubble constant is $66.2^{+4.4}_{-4.2}\ \rm km \,Mpc^{-1}\, s^{-1}$ at $1\sigma$ uncertainty.

Abstract: Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photodissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, which affects planet formation within the disks. We report James Webb Space Telescope and Atacama Large Millimeter Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modeling their kinematics and excitation allowed us to constrain the physical conditions within the gas. We quantified the mass-loss rate induced by the FUV irradiation and found that it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.

Abstract: Forty years ago, it was proposed that gas-phase organic chemistry in the interstellar medium can be initiated by the methyl cation CH3+ (refs. 1–3), but so far it has not been observed outside the Solar System 4,5. Alternative routes involving processes on grain surfaces have been invoked 6,7. Here we report James Webb Space Telescope observations of CH3+ in a protoplanetary disk in the Orion star-forming region. We find that gas-phase organic chemistry is activated by ultraviolet irradiation.

21. Flerspråkiga barns språkutveckling i förskolan med stöd av lek : En kvalitativ studie om förskollärarnas arbete med lek

Abstract: Syftet med studien är att undersöka förskollärares arbete med flerspråkiga barns språkutveckling med stöd av lek. Jag valde att genomföra semistruktuerade intervjuer med åtta förskollärare i fyra olika förskolor i två medelstora städer i Sverige. Jag valde fyra mångkulturella förskolor och åtta förskollärare med god erfarenhet av arbetet med flerspråkiga barn. Resultatet visade att leken i förskolan ses som centrala för lärandet och att barn lär sig i samspel med andra. Det som blir betydelsefullt är att det finns en närvarande pedagog under leken, som stöttar barnen i leken samt upprepar och benämner ord och begrepp. Slutsatsen är att oavsett om det är en mångkulturell förskola eller svensktalande förskola blir det pedagogernas ansvar att se till att barn utvecklas samt stödja deras språkutveckling.

22. Infection prevention and aseptic practice in dentistry during the Covid-19 pandemic : A systematic review

Abstract: Aim: To assess and compile current knowledge and recommendations forinfection prevention and control (IPC) in dentistry during the COVID-19pandemic. Furthermore, to assess whether previous knowledge about IPC hascontributed to constructing evidence-based measures for IPC that can beuniversally applicable. Also, identifying knowledge gaps that need to beaddressed regarding IPC to be used effectively in future pandemics.Method: Searches were made in PubMed, Web of Science, Scopus, Cochrane,and Embase, according to PRISMA guidelines. There were no time or publicationtype restrictions. Studies that met the inclusion criteria were read in full text andresulted in 44 included articles. Quality assessment and risk of bias (RoB) wereperformed using checklists by AMSTAR-2 and ROBIS.Results: The four main recommended measures for IPC in dentistry were: triage,personal protective equipment, antimicrobial mouthwash before the procedure,and surface disinfection. Most of the included studies were literature reviews withlow quality and a high RoB.Conclusion: Measures taken during this pandemic were based on evidence,previous knowledge, and subjective opinions. However, there were limitations inthe included studies as their evidence was of lower quality and had a high RoB.Future research is needed to minimize the risk of infection spreading in dentistryduring the pandemic. The lack of research on SARS-COV-2 was considered aknowledge gap that should be addressed to be used effectively in futurepandemics. This present study shows that IPC policies were to some extent basedon evidence but not regarding the current virus., Syfte: Syftet med studien var att bedöma och sammanställa nuvarandekunskapsläget samt rekommendationerna för infektionspreventionen och kontroll(IPC) inom tandvården under COVID-19 pandemin. Vidare var syftet attgenomföra en bedömning av huruvida tidigare kunskap om infektionspreventionhar bidragit till att konstruera evidensbaserade åtgärder för infektionspreventionsom kan bli universellt applicerbara. Dessutom, att identifiera kunskapsluckorsom behöver åtgärdas gällande infektionsprevention och kontroll för att effektivtkunna användas för framtida pandemier.Material och metod: Sökningar gjordes i PubMed, Web of Science, Scopus,Cochrane och Embase, enligt PRISMA riktlinjer. Det fanns inga tids- ellerpublikationstypsbegränsningar. Studier som uppfyllde inklusionskriterierna lästesi fulltext och resulterade i 44 inkluderade artiklar. Kvalitetsbedömning och riskenför bias av de inkluderade studierna utfördes med hjälp av checklistor avAMSTAR-2 och ROBIS.Resultat: De fyra främsta rekommenderade åtgärderna för IPC inom tandvårdenvar: triage, personlig skyddsutrustning, antimikrobiell munsköljning föreproceduren samt ytdesinfektion. De flesta av de inkluderade studierna varlitteraturöversikter med låg kvalitet och hög risk för bias.Slutsats: Åtgärder som vidtogs under denna pandemi baserades på både bevis ochtidigare kunskap men också subjektiva åsikter. Det fanns dock begränsningar i deinkluderade studierna på grund av att deras evidens inte var av god kvalitet samthade en hög RoB. Framtida forskning behöver göras för att minimera risken förinfektionsspridning inom tandvården under COVID-19-pandemin. Brist påforskning om SARS-COV-2 ansågs vara en kunskapslucka som måste åtgärdasför att effektivt kunna användas för framtida pandemier.

26. Key determinants of corruption in the Middle East and North Africa.

Abstract: This study examines the effect of some economic and non-economic factors on corruption. It tests a number of hypotheses formulated around the impact of specific dimensions on increasing or reducing corruption in the Middle East and North Africa Region over a time series from 2008 to 2018. The objective of this study is to illuminate the impact of several independent factors (average income, economic freedom, education, income distribution and globalization) on corruption increase or decrease in the MENA region taking a few countries as a sample case. To reach this end, secondary data is employed and retrieved from secondary sources using published reports and indexes and official websites and databases maintained by various research institutions. The results are tested using panel data regression analysis as a statistical tool for data analysis and hypotheses testing. Findings indicate a significant effect of the presented economic and non-economic determinants on corruption in these sample countries except income distribution which shows an insignificance effect. Abstract: Neutron-star mergers are associated with violent phenomena that probe physical principles under extreme conditions that are not reproducible in any terrestrial laboratory. Thus, their multi-messenger analysis combining gravitational-wave and electromagnetic signatures from multiple wavelengths is not only of astrophysical interest, but it also allows studying the behavior of supranuclear dense matter, testing the fabric of spacetime, probing the principles of general relativity, and measuring the expansion rate of the Universe. In this work, we perform a multi-messenger analysis of the gravitational wave signal GW170817 and its electromagnetic counterparts AT2017gfo and GRB170817A. By incorporating information from the NICER observation of PSR J0030+0451, the radio observations of PSR J0740+6620, and nuclear theory computations using chiral effective field theory, we provide new constraints on the neutron-star equation of state and the Hubble constant. For our analysis, we make use of the new waveform approximant IMRPhenomPv2_NRTidalv2 for the interpretation of the gravitational wave signal, we employ a newly developed kilonova model, and we derive a new prediction for the debris disk mass surrounding the binary neutron star merger remnant. We determine the radius of a 1.4 solar mass neutron star to be $R_{1.4M_\odot}=10.98^{+0.37}_{-0.38} \rm km$ at $1\sigma$ uncertainty and $R_{1.4M_\odot}=10.98^{+1.00}_{-0.69} \rm km$ at $2\sigma$ uncertainty. In addition, we estimate the Hubble constant to be $H_0=68.4^{+5.2}_{-4.7}\ \rm km /Mpc/s$ at $1\sigma$ uncertainty. We test the consistency of our results and the robustness of our methods by also analyzing the second binary neutron-star merger detection GW190425 and the fact that no electromagnetic counterpart was observed for this event.

Abstract: The observation of a compact object with a mass of 2.50-2.67Me on 2019 August 14, by the LIGO Scientific and Virgo collaborations (LVC) has the potential to improve our understanding of the supranuclear equation of state. While the gravitational-wave analysis of the LVC suggests that GW190814 likely was a binary black hole system, the secondary component could also have been the heaviest neutron star observed to date. We use our previously derived nuclear-physics-multimessenger astrophysics framework to address the nature of this object. Based on our findings, we determine GW190814 to be a binary black hole merger with a probability of >99.9%. Even if we weaken previously employed constraints on the maximum mass of neutron stars, the probability of a binary black hole origin is still ∼81%. Furthermore, we study the impact that this observation has on our understanding of the nuclear equation of state by analyzing the allowed region in the mass-radius diagram of neutron stars for both a binary black hole or neutron star-black hole scenario. We find that the unlikely scenario in which the secondary object was a neutron star requires rather stiff equations of state with a maximum speed of sound cs ≥0.6 times the speed of light, while the binary black hole scenario does not offer any new insight.

Abstract: The detection of the optical transient AT2017gfo proved that binary neutron star mergers are progenitors of kilonovae (KNe). Using a combination of numerical-relativity and radiative-transfer simulations, the community has developed sophisticated models for these transients for a wide portion of the expected parameter space. Using these simulations and surrogate models made from them, it has been possible to perform Bayesian inference of the observed signals to infer properties of the ejected matter. It has been pointed out that combining inclination constraints derived from the KN with gravitational-wave measurements increases the accuracy with which binary parameters can be estimated, in particular breaking the distance-inclination degeneracy from gravitational wave inference. To avoid bias from the unknown ejecta geometry, constraints on the inclination angle for AT2017gfo should be insensitive to the employed models. In this work, we compare different assumptions about the ejecta and radiative reprocesses used by the community and we investigate their impact on the parameter inference. While most inferred parameters agree, we find disagreement between posteriors for the inclination angle for different geometries that have been used in the current literature. According to our study, the inclusion of reprocessing of the photons between different ejecta types improves the modeling fits to AT2017gfo and, in some cases, affects the inferred constraints. Our study motivates the inclusion of large similar to 1-mag uncertainties in the KN models employed for Bayesian analysis to capture yet unknown systematics, especially when inferring inclination angles, although smaller uncertainties seem appropriate to capture model systematics for other intrinsic parameters. Abstract: Temaet for denne bacheloroppgaven er plutselig idiopatisk sensorinevralt hørselstap. Ved gjennomgang av litteratur har vi sett nærmere på ulike faktorer som er avgjørende for en god prognose ved diagnosen, og hvor avgjørende de er for gjenvinningsgraden av hørselen. For å få svar på dette ble det gjennomført en systematisk litteraturgjennomgang ved hjelp av ulike databaser. Vi bestemte oss for å inkludere artikler fra 1980-2014 fordi mange eldre artikler inneholder gode, relevante undersøkelser, samt at det er færre nyere artikler som undersøker det som er relevant for vår problemstilling. Studiene som skrev om en spesifikk årsak til hørselstapet, som var forårsaket av for eksempel slag, sykdom, traume, ble forkastet da vi var ute etter plutselig idiopatisk hørselstap. Det resulterte til slutt i 12 relevante artikler som utgjør hoveddelen av datamaterialet vårt. Resultatet viser blant annet at alle artiklene konkluderte med at en hadde størst gjenvinningsgrad av hørselen dersom en oppsøkte hjelp og mottok behandling innen det hadde gått sju dager etter opplevelsen av plutselig idiopatisk hørselstap. Alder er en avgjørende faktor for gjenvinningsgraden av hørselen. Høyere alder resulterte i en dårligere forbedring av hørselen i dB og sjansen for permanent hørselstap er høyere. The theme of this bachelor thesis is sudden idiopathic sensorineural hearing loss. We did a systematic review of literature to find which factors that are important for a good prognosis, and how crucial they are for the recovery rate of hearing. In order to achieve this, a systematic review was conducted with help of different databases. We decided to include articles from 1980-2014. The oldest articles contain good and relevant research, and there are fewer new articles that examine what is relevant to our issue. We discarded studies that wrote about specific causes to the hearing loss, caused by, for example stroke, other disease, trauma because we were searching for articles that included patients who experienced sudden idiopathic hearing loss. This resulted in 12 relevant articles that made up the main part of our data material. The result from all the articles concluded that the greatest recovery rate of hearing if the patient received treatment and help within seven days after the first experience of the hearing loss. Age is a crucial factor in the recovery rate of hearing. Higher age resulted in a poorer improvement in hearing in dB and the chance of permanent hearing loss is higher. 37. The role of CDK5RAP2 in autosomal recessive primary microcephaly (MCPH)

Abstract: Autosomal recessive primary microcephaly (MCPH) is a rare, genetically heterogeneous disease characterized by a reduced head circumference at birth and intellectual deficit. Biallelic mutations in the Cyclin-dependent kinase 5, regulatory subunit-associated protein 2 gene CDK5RAP2 cause MCPH type 3 (MCPH3). CDK5RAP2 function contributes to cellular processes such as centrosome function, cell cycle checkpoint control, DNA repair, chromosome condensation, and kinetochore attachment to spindles. One current model for the microcephaly phenotype in MCPH3 invokes a premature shift from symmetric to asymmetric cell divisions and thus premature neurogenesis with a subsequent depletion of the progenitor pool. In addition, other mechanisms may also play a role. The aim of my PhD thesis work was to better understand the role of CDK5RAP2 in physiological brain development and in disease. Specific aims were (i) to characterize the spatiotemporal CDK5RAP2 expression during normal murine and human brain development, (ii) to characterize the phenotype of a Cdk5rap2 mutant mouse generated in our research group, and (iii) to identify further MCPH3 patients with novel CDK5RAP2 mutations and thereby better characterize the phenotype spectrum of MCPH3. We found that in the murine brain Cdk5rap2 expression boosts during early embryonic stages, when proliferation rates are high and neocortical development is initiated. In human and murine brain, CDK5RAP2 is present in brain structures with high proliferative rates and, colocalizes with progenitor cells, glial cells, and early neurons. As the brain matures, CDK5RAP2 is refined to specific substructures within regions, which correspond to preserved proliferation zones. Moreover, we found concordance between regions of high CDK5RAP2 expression in the mouse and sites of pathology suggested by neuroimaging studies in humans and mouse. Our findings in human and mouse tissue confirm the function of CDK5RAP2 in cell proliferation. In addition, we described for the first time in detail the clinical, radiological, and also the cellular phenotype of MCPH3 patients with a novel homozygous nonsense mutation in the CDK5RAP2 gene. Cells from the patients showed mitotic spindle defects and disrupted γ-tubulin localization to the centrosome, which underlines the importance of CDK5RAP2 in mitotic spindle pole organization and centrosome integrity. These findings suggest that MCPH3 pathomechanism is partially due to mitotic spindle and centrosomal defects. To further mimic the human mutation and study the function of CDK5RAP2 in vivo, we generated a transgenic Cdk5rap2 mouse. These mice did not display the expected microcephaly phenotype. Our further analysis indicated that a previously unknown splice variant of the Cdk5rap2 gene exists, which allows translation of Cdk5rap2, even in the mutant mice. With our studies, we provide for the first time a systematic description of the spatiotemporal expression of CDK5RAP2 in murine and human developing brain. Moreover, the cellular phenotype findings we collected from the new MCPH3 patients provide a glimpse into processes that could lead to the microcephaly phenotype in MCPH., Die Autosomal-rezessive primäre Mikrozephalie (MCPH) ist eine seltene und genetisch heterogene Erkrankung, die sich durch einen kongenital reduzierten Kopfumfang und eine mentale Retardierung auszeichnet. Biallelische Mutationen im Cyclin-dependent-kinase-5-regulatory-subunit-associated-Protein 2 Gen CDK5RAP2 verursachen die MCPH vom Typ 3 (MCPH3). Das CDK5RAP2 spielt eine Rolle in zellulären Prozessen wie der Funktion von Zentrosomen, der Kontrolle der Zellzyklus-Checkpoints, DNA-Reparatur, Chromosomenkondensation und Kinetochor Befestigung an Spindeln. Eine der Haupthypothesen für die Entstehung des Mikrozephalie-Phänotyps bei MCPH3 beinhaltet einen vorzeitigen Wechsel von symmetrischer zu asymmetrischer Zellteilungen und damit eine vorzeitige Neurogenese sowie eine Depletion des Vorläuferpools. Das Ziel der vorliegenden Doktorarbeit war es, die Rolle des CDK5RAP2-Proteins in der physiologischen Entwicklung des Gehirns und in der Entstehung von MCPH besser zu verstehen. Spezifische Ziele waren (i) die zeitliche und örtliche Expression von CDK5RAP2 während der normalen Entwicklung des Gehirns von Menschen und Mäusen zu veranschaulichen, (ii) den Phänotyp einer in unserer Forschungsgruppe erzeugten Cdk5rap2-Mausmutanten zu charakterisieren, und (iii) weitere MCPH3 Patienten mit bisher nicht beschriebenen Mutationen im CDK5RAP2-Gen zu identifizieren und damit das Phänotypspektrum der MCPH3 besser zu erforschen. Wir fanden, dass Cdk5rap2 während der frühen embryonalen Stadien im murinen Gehirn besonders hoch exprimiert wird, d.h. während einer Zeit in der hohe Proliferationsraten auftreten und die Neokortex-Entwicklung bei Mensch und Maus beginnt. In humanen und murinen Gehirnen, ist CDK5RAP2 in Hirnstrukturen mit hoher Proliferationsrate vorhanden und kolokalisiert mit Vorläuferzellen, Gliazellen und unreifen Neuronen. Mit der Entwicklung des Gehirns reduziert sich das Vorhandensein von CDK5RAP2 auf spezifische Unterstrukturen, in denen die Proliferation anhält. Darüber hinaus fanden wir Übereinstimmungen zwischen Regionen mit hoher CDK5RAP2 Expression in der Maus und der durch bildgebene Verfahren beschriebenen Pathologie bei Mensch und Maus. Unsere Ergebnisse in Human- und Mausgewebe bestätigen die Rolle von CDK5RAP2 in der Zellproliferation. Zusätzlich beschrieben wir zum ersten Mal detailliert den klinischen, radiologischen und auch den zellulären Phänotyp von Patienten mit MCPH3 mit einer bisher nicht beschriebenen homozygoten Nonsense-Mutation im CDK5RAP2-Gen. Wir fanden, dass Zellen von Patienten eine abnorme Morphe ihrer mitotischen Spindelapparate sowie eine gestörte Lokalisation des γ-Tubulins am Zentrosom aufwiesen. Diese Ergebnisse unterstreichen die Bedeutung von CDK5RAP2 in der Organisation der mitotischen Spindel und der Zentrosomintegrität. Diese Ergebnisse legen nahe, dass der MCPH3-Pathomechanismus teilweise auf Defekte der mitotischen Spindel und der Zentrosomen zurückzuführen ist. Um die Auswirkungen einer CDK5RAP2-Mutation im Menschen weiter zu imitieren und die Funktion des CDK5RAP2-Proteins in vivo zu untersuchen, generierten wir eine transgene Cdk5rap2-Mausmutante. Diese Mäuse bildeten aber nicht den erwarteten Mikrozephalie-Phänotyp aus. Weitere Analysen zeigten, dass es eine bisher unbekannte Spleißvariante des Cdk5rap2-Gens gibt, die selbst in den Mausmutanten die Translation von Cdk5rap2 ermöglicht. Diesen Studien bieten zum ersten Mal eine systematische Beschreibung der CDK5RAP2-Expression während der Entwicklung muriner und humaner Gehirne. Darüber hinaus geben unsere am Patienten erhobenen Ergebnisse des zellulären Phänotyps einen Einblick in Prozesse, die zum Mikrozephalie- Phänotyp bei MCPH führen könnten.

Abstract: Recent data have found that aging-related hearing loss (ARHL) is associated with the development of Alzheimer's Disease (AD). However, the nature of the relationship between these two disorders is not clear. There are multiple potential factors that link ARHL and AD, and previous investigators have speculated that shared metabolic dysregulation may underlie the propensity to develop both disorders. Here, we investigate the distribution of serum lipidomic biomarkers in AD subjects with or without hearing loss in a publicly available dataset. Serum levels of 349 known lipids from 16 lipid classes were measured in 185 AD patients. Using previously defined co-regulated sets of lipids, both age- and sex-adjusted, we found that lipid sets enriched in phosphatidylcholine and phosphatidylethanolamine showed a strong inverse association with hearing loss. Examination of biochemical classes confirmed these relationships and revealed that serum phosphatidylcholine levels were significantly lower in AD subjects with hearing loss. A similar relationship was not found in normal subjects. These data suggest that a synergistic relationship may exist between AD, hearing loss and metabolic biomarkers, such that in the context of a pathological state such as AD, alterations in serum metabolic profiles are associated with hearing loss. These data also point to a potential role for phosphatidylcholine, a molecule with antioxidant properties, in the underlying pathophysiology of ARHL in the context of AD, which has implications for our understanding and potential treatment of both disorders. 45. Toxic Metals Impact Gut Microbiota and Metabolic Risk in Five African-Origin Populations.

Abstract: Exposure to toxic metals impacts obesity and type 2 diabetes (T2DM) risk. Yet, the underlying mechanisms remain largely unknown. Gut microbiota has been strongly associated with progression of cardiometabolic risk. To determine whether high metal exposures and gut dysbiosis interact to promote metabolic dysregulation and cardiometabolic risk, we assessed relationships between these factors. We analyzed cross-sectional associations between arsenic, lead, mercury, cadmium, and cardiometabolic health markers in 178 randomly selected African-origin adults (52% female, 51% obese, mean age=43.0±6.4 years) from Ghana, South Africa, Seychelles, Jamaica, and USA. Metal levels were dichotomized to high or low at the median level of each metal. We analyzed associations between gut microbiome taxa, metal levels, clinical measures (BMI, fasting blood glucose, and blood pressure) and diagnoses (hypertension, obesity, and diabetes status). High vs. low lead and arsenic exposures had a significant effect on beta diversity (p <0.05). 71 taxa were associated with high lead levels: 30 with elevated BMI, 22 with T2DM, and 23 with elevated fasting blood glucose (p<0.05). 115 taxa were associated with high arsenic levels: 32 with elevated BMI, 33 with T2DM, and 26 with elevated blood glucose (p<0.05). Of the taxa associated with high lead and arsenic exposure and either elevated BMI or fasting blood glucose, porphyrin metabolism was the most enriched metabolic pathway. These data collectively provide the first findings in a human study that the gut microbiome may drive the association between lead and arsenic exposure and obesity and T2DM risk., Competing Interests: Competing Interests. Abstract: Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photodissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, which affects planet formation within the disks. We report James Webb Space Telescope and Atacama Large Millimeter Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modeling their kinematics and excitation allowed us to constrain the physical conditions within the gas. We quantified the mass-loss rate induced by the FUV irradiation and found that it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.
