The integrity of mitochondrial DNA (mtDNA) isolated from solid tissues is critical for analyses such as long-range PCR, but is typically assessed under conditions that fail to provide information on the individual mtDNA strands. Using denaturing gel electrophoresis, we show that commonly-used isolation procedures generate mtDNA containing several single-strand breaks per strand. Through systematic comparison of DNA isolation methods, we identify a procedure yielding the highest integrity of mtDNA that we demonstrate displays improved performance in downstream assays. Our results highlight the importance of isolation method choice, and serve as a resource to researchers requiring high-quality mtDNA from solid tissues.

Nitrogen containing heterocyclic compounds has acquired their remarkable and distinct place in the wide area of organic synthesis due to the broad range of applications. Among them, quinoline motifs have attracted researchers in the synthetic chemistry because of its presence in the large number of pharmacologically active compounds. Different methods for synthesis of quinoline derivatives are reported, among them the Friedlander synthesis have provided comparatively more efficient approach. Many of the reported conventional Friedlander methodologies have some problems such as difficult product isolation procedures, poor yields and use of expensive catalysts, etc. Recently, polymer or solid supported synthetic approaches have attracted the attention of researchers because of their easy execution, greater selectivity, increased product yields, simple work-up procedures, recoverability and reusability of the catalysts. In consideration with the advantages of polymer supported synthetic strategies, the proposed review covers the role of polymers in the Friedlander synthesis; which may use polymers of organic, inorganic or hybrid in nature and of nanolevel as well.

Background: The aim of the current study was to analyze the impact of single versus double transseptal puncture (TSP) for atrial fibrillation (AF) ablation. Methods: Consecutive patients undergoing AF ablation were prospectively included in the AF ablation registry and were analyzed according to single versus double TSP. Results: A total of 478 patients (female 35%, persistent AF 67%) undergoing AF ablation between 01/2014 and 09/2014 were included. Single TSP was performed in 202 (42%) patients, double TSP in 276 (58%) patients. Age, gender, body mass index, CHA2DS2-VASc score, left ventricular ejection fraction and operator experience (experienced operator defined as ≥ 5 years of experience in invasive electrophysiology) were equally distributed between the two groups. Repeat procedures (re-dos) were more frequently performed using single TSP access (p < 0.001). Left atrial (LA) diameter was larger in patients with double TSP (p = 0.001). Procedure duration in single TSP was identical to double TSP procedures (p = 0.823). Radiation duration was similar between the two groups (p = 0.217). There were 49 (10%) patients with complications after catheter ablation. There were no differences between complication rates and TSP type (p = 0.555). Similarly, recurrence rates were comparable between both TSP groups (p = 0.788). Conclusions: There was no clear benefit of single or double TSP in AF ablation.

Utilization of bone graft substitutes have increased due to the rising number of accidents, trauma, and aging population. Autograft is still considered as a gold standard for treating bone defects. However, limitations such as insufficient donor sites and secondary surgery, leads to the development of alternative grafts. Hydroxyapatite (HA) from natural resources gained much attention in recent years as a bone graft substitute due to its biocompatibility, excellent osteointegration, osteoconductive, and osteoinductive properties. In the current review, we have presented the isolation procedures of HA from marine fishbone and cuttlefish bone. Further, composite preparation using marine derived HA with other polymeric and ceramic materials were discussed, and cell-materials interaction were reviewed in detail towards bone tissue construction. Composite biomaterials with HA showed better cell proliferation, cell adhesion, increased gene expression (collagen, osteocalcin, osteopontin, bone sialoprotein, BMP-2 etc.), and in vivo studies demonstrated significant bone formation with HA composite materials. Hence, composite biomaterials with hydroxyapatite will be potential candidates for artificial synthetic bone graft substitute.

Extensive phytochemical work on the 1-BuOH-soluble fraction of a MeOH extract of the leaves of Symplocos cochinchinensis var. philippinensis resulted in the isolation of 14 new triterpenene saponins, along with four known ones. Their structures were elucidated by comparison of NMR spectroscopic data with related compounds reported in the literature. Three oleanane-type saponins, symplocosins K, M, and P, possessed glucuronic acid as a sugar component, and their carboxyl groups appeared as methyl esters. These are probably formed during extraction and isolation procedures. Symplocosin K (9) showed moderate cytotoxicity toward A549 cells. In addition, all isolated compounds did not show α-glucosidase inhibitory activity.

espanolIntroduction. The aim of this study is to assess the value of systematic screening in asymptomatic women admitted for spontaneous delivery with a combination of reverse transcription polymerase chain reaction (RT-PCR) and cycle threshold (Ct) and serum antibodies. Material and methods. Since May 6 all women admitted for spontaneous delivery underwent RT-PCR in nasopharyngeal swabs and specific antibodies IgG of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) in serum that were performed as part of routine clinical care in our institution. Ct of the PCR was recorded. We analyzed the first 100 women consecutively admitted for spontaneous delivery at our institution. Results. Nine women were positive for SARS-CoV-2 in nasopharyngeal samples (9%) and 13 (13%) presented positive specific antibodies of the coronavirus. Overall, SAR-CoV-2 prior exposure was 15%. The Ct determination (RT-PCR test) of our 9 positive patients ranged from 36 to 41 cycles with a median of 40. Vaginal delivery occurred in 94% of the cases and only 6% underwent a cesarean section, always for obstetric reasons. No fetal transmission was observed and maternal and neonatal prognosis was excellent. Conclusions. During epidemic episodes in asymptomatic women in labor, universal testing with RT-PCR (considering Ct determination), and the detection of antibodies, permits a better interpretation of the results and avoid unnecessary isolation procedures. EnglishObjetivo. El objetivo de este estudio es evaluar, en mujeres asintomaticas que acuden a urgencias en trabajo de parto, el valor de la deteccion sistematica con una combinacion de reaccion en cadena de la polimerasa con transcriptasa inversa (RT-PCR) y umbral del ciclo (Ct) y anticuerpos sericos. Material y metodos. Desde el 6 de mayo, todas las mujeres ingresadas para parto espontaneo se sometieron a RT-PCR en hisopos nasofaringeos y anticuerpos especificos IgG del sindrome respiratorio agudo severo por coronavirus 2 (SARSCoV-2) en suero que se realizaron como parte de la atencion clinica de rutina en nuestra institucion. Se registro el Ct de la PCR. Analizamos las primeras 100 mujeres admitidas consecutivamente para parto espontaneo en nuestra institucion. Resultados. Nueve mujeres fueron positivas para SARSCoV-2 en muestras nasofaringeas (9%) y 13 (13%) presentaron anticuerpos especificos positivos del coronavirus. En general, la exposicion previa a SARS-CoV-2 fue del 15%. La determinacion de Ct (prueba de RT-PCR) de nuestros 9 pacientes positivos vario de 36 a 41 ciclos con una mediana de 40. El parto vaginal se produjo en el 94% de los casos y solo el 6% se sometio a una cesarea, siempre por razones obstetricas. No se observo transmision fetal y el pronostico materno y neonatal fue excelente. Conclusiones. Durante los episodios epidemicos en mujeres asintomaticas en trabajo de parto, las pruebas universales con RT-PCR (considerando la determinacion de Ct) y la deteccion de anticuerpos, permiten una mejor interpretacion de los resultados y evitan procedimientos de aislamiento innecesarios.

Background and aims Successful clinical human allo or auto-islet transplantation requires the recovery of a sufficient number of functional islets from either brain-dead or chronic pancreatitis pancreases respectively. Methods In the last two decades (2000–2019), significant progress has been made in improving the human islet isolation procedures and in standardizing the use of different tissue dissociation enzyme (TDE; a mixture of collagenase and protease enzymes) blends to recover higher islet yields. Results and Conclusions This review presents information focusing on properties and role of TDE blends during the islet isolation process, particularly emphasizing on the current developments, associated challenges and future perspectives within the field.

Current Aedes aegypti vector control strategies remain insufficient and efforts are needed to find new insecticides. Natural products represent a potential source of such larvicides. Modern analytical techniques and dereplication strategies can prove more efficient than classical bioguided approaches in the search for novel compounds. This method can detect the presence of known/unknown compounds/classes of compounds in a shorter time. Once the target is highlighted, knowledge of physicochemical properties and/or previous isolation procedures facilitates the isolation process. In the present study, dereplication of 34 Annonaceae extracts from Cardiopetalum calophyllum Schltdl., Duguetia furfuracea (A.St.-Hil.) Saff., Xylopia aromatica (Lam.) Mart. and Xylopia emarginata Mart. resulted in the isolation of three phenylpropanoids and one diterpene with larvicidal activity against A. aegypti.

COVID-19 is a new challenge in clinical medicine. Although typical presentations include fever and pneumonia, we describe a case of COVID-19 presenting with neurological symptoms of encephalitis and infectious-related syndrome of inappropriate anti-diuretic hormone secretion. Because of the epidemic health problems, it is crucial to identify these patients as early as possible to follow the isolation procedures. We suggest that unclear neurological clinical presentations of patients should be considered for COVID-19.

The coronavirus disease 2019 (COVID-19) outbreak has reached pandemic status. Drastic measures of social distancing are enforced in society and healthcare systems are being pushed to and beyond their limits. To help in the fight against this threat on human health, a fully automated AI framework was developed to extract radiomics features from volumetric chest computed tomography (CT) exams. The detection model was developed on a dataset of 1381 patients (181 COVID-19 patients plus 1200 non COVID control patients). A second, independent dataset of 197 RT-PCR confirmed COVID-19 patients and 500 control patients was used to assess the performance of the model. Diagnostic performance was assessed by the area under the receiver operating characteristic curve (AUC). The model had an AUC of 0.882 (95% CI: 0.851&ndash, 0.913) in the independent test dataset (641 patients). The optimal decision threshold, considering the cost of false negatives twice as high as the cost of false positives, resulted in an accuracy of 85.18%, a sensitivity of 69.52%, a specificity of 91.63%, a negative predictive value (NPV) of 94.46% and a positive predictive value (PPV) of 59.44%. Benchmarked against RT-PCR confirmed cases of COVID-19, our AI framework can accurately differentiate COVID-19 from routine clinical conditions in a fully automated fashion. Thus, providing rapid accurate diagnosis in patients suspected of COVID-19 infection, facilitating the timely implementation of isolation procedures and early intervention.

Glycosaminoglycan (GAG) fine structures from the same animal cells and tissues are controlled not only by the biosynthetic and metabolic enzymes but also by other environmental factors, such as chemicals, growth factors, nutritional factors, and isolation procedures. To facilitate direct quantitative comparison of disaccharide compositions from different GAG preparations, several stable isotope labeling strategies have been developed. In this report, 1-phenyl-3-methyl-5-pyrazolone (PMP) and deuterated d5-PMP are used for differential disaccharide labeling and profiling of chondroitin sulfate GAG by high performance liquid chromatography (HPLC) coupled with mass spectrometry (MS).

Ergot Alkaloids are potent α-blockers that cause direct smooth muscle contraction. They are products of fungi in the genus Claviceps. The most prominent member of this group is Claviceps purpurea (rye ergot fungus). Their common sources include Rye, bread as well as other grains. These are nitrogen derived- natural substances which are grouped under indole alkaloids. These ergot alkaloids are a diverse category of secondary metabolites that have been classified into 3 groups as Clavines, Amides of lysergic acid, Ergopeptines and Lactum ergot alkaloids based on their bond arrangement. They contaminate a large variety of cereals such as rye triticale, wheat and barley. In this present review, basic information regarding the ergots along with their chemistry, extraction, purification and isolation procedures for ergot alkaloids were focused, that helps to produce a purified crude extract that is taken for chromatographic analysis. This review also provides detailed informations on the chromatographic approaches for the ergot alkaloids estimations such as TLC, HPLC, HPTLC, UPLC /UHPLC, LCMS, GCMS with different detection techniques like Mass spectrometry and Fluorescence detection which was done in the past decade and also provides information regarding the pharmacological activity of ergot alkaloids like its application in the treatment of severe, throbbing headaches such as migraine, cluster headaches and also other therapeutic applications.

Proteomics can map extracellular vesicles (EVs), including exosomes, across disease states between organisms and cell types. Due to the diverse origin and cargo of EVs, tailoring methodological and analytical techniques can support the reproducibility of results. Proteomics scans are sensitive to in-sample contaminants, which can be retained during EV isolation procedures. Contaminants can also arise from the biological origin of exosomes, such as the lipid-rich environment in human milk. Human milk (HM) EVs and exosomes are emerging as a research interest in health and disease, though the experimental characterization and functional assays remain varied. Past studies of HM EV proteomes have used data-dependent acquisition methods for protein detection, however, improvements in data independent acquisition could allow for previously undetected EV proteins to be identified by mass spectrometry. Depending on the research question, only a specific population of proteins can be compared and measured using isotope and other labelling techniques. In this review, we summarize published HM EV proteomics protocols and suggest a methodological workflow with the end-goal of effective and reproducible analysis of human milk EV proteomes.

Annona cherimola Mill., or the custard apple, is one of the species belonging to the Annonaceae family, is widely used in traditional medicine, and has been reported to be a valuable source of bioactive compounds. A unique class of secondary metabolites derived from this family are Annonaceous acetogenins, lipophilic polyketides considered to be amongst the most potent antitumor compounds. Due to the COVID-19 pandemic, higher education institutions were forced to make difficult decisions regarding the 2020-2021 academic year. Many institutions decided to have courses in an online remote format, others decided to attempt an in-person experience, while still others took a hybrid approach. Hope College (Holland, MI) decided that an in-person semester would be safer and more equitable for students. To achieve this at a residential college required broad collaboration across multiple stakeholders. Here, we share lessons learned and detail Hope College’s model, including wastewater surveillance, comprehensive testing, contact tracing and isolation procedures, that allowed us to deliver on our commitment of an in-person, residential college experience.

Due to a shortage of donation after brain death (DBD) organs, donation after circulatory death (DCD) is increasingly performed. In the field of islet transplantation, there is uncertainty regarding the suitability of DCD pancreas in terms of islet yield and function after islet isolation. The aim of this study was to investigate the potential use of DCD pancreas for islet transplantation. Islet isolation procedures from 126 category 3 DCD and 258 DBD pancreas were performed in a 9-year period. Islet yield after isolation was significantly lower for DCD compared to DBD pancreas (395 515 islet equivalents [IEQ] and 480 017 IEQ, respectively; p = .003). The decrease in IEQ during 2 days of culture was not different between the two groups. Warm ischemia time was not related to DCD islet yield. In vitro insulin secretion after a glucose challenge was similar between DCD and DBD islets. After islet transplantation, DCD islet graft recipients had similar graft function (AUC C-peptide) during mixed meal tolerance tests and Igls score compared to DBD graft recipients. In conclusion, DCD islets can be considered for clinical islet transplantation.

ObjectivesTo understand the experiences of those who underwent supported isolation as part of the response to the COVID-19 pandemic, after returning to the UK from Wuhan, China.DesignWe used semi-structured interviews to capture participants’ experiences and perceptions of supported isolation.SettingTelephone interviews carried out within approximately one month of an individual leaving supported isolation.Participants26 people who underwent supported isolation at either Arrowe Park Hospital (n = 18) or Kents Hill Park Conference Centre (n = 8) after being repatriated from Wuhan in January – February 2020.ResultsParticipants were willing to undergo supported isolation because they understood that it would protect themselves and others. Positive treatment by staff was fundamental to participants’ willingness to comply with isolation procedures. Despite the high level of compliance, participants expressed some uncertainty about what the process would involve.ConclusionsAs hotel quarantine is introduced across the UK for international arrivals, our findings suggest that those in charge should: communicate effectively before, during and after quarantine, emphasising why quarantine is important and how it will protect others; avoid enforcement and focus on supporting and promoting voluntary compliance; facilitate shared social experiences for those in quarantine; and ensure all necessary supplies are provided. Doing so will increase adherence and reduce any negative effects on wellbeing.

As the powerhouse of the cell, mitochondria, plays a crucial role in many aspects of life, whereby mitochondrial dysfunctions are associated with pathogenesis of many diseases, like neurodegenerative diseases, obesity, cancer, and metabolic as well as cardiovascular disorders. Mitochondria analysis frequently starts with isolation and enrichment procedures, which have become increasingly important in biomedical research. Unfortunately, isolation procedures can easily cause changes in the structural integrity of mitochondria during in vitro handling having impact on their function. This carries the risk that conclusions about isolated mitochondria may be drawn on the basis of experimental artifacts. Here we critically review a commonly used isolation procedure for mitochondria utilizing differential (gradient) centrifugation and depict major challenges to achieve "functional" mitochondria as basis for comprehensive physiological studies.

Bacterial magnetosomes represent magnetic core-shell nanoparticles biomineralized by magnetotactic bacteria like Magnetospirillum gryphiswaldense. The establishment of fermentation regimes for high-yield particle production, standardized isolation procedures as well as the development of a genetic toolkit for the generation of “tailored” particles might soon pave the way for the application of engineered magnetosomes in the biomedical and biotechnological field.

Lung isolation is used for many thoracic surgeries to provide exclusive ventilation of the nonoperative lung. This allows for the operative lung to be desufflated, improving surgical conditions and exposure. Additionally, in severe cases of unilateral lung infection or bleeding, ventilation of only the healthy lung can decrease the risk for contamination with blood and infected material from the diseased lung. Techniques used for achieving lung isolation include bronchial blocker placement, mainstem intubation, and use of a double-lumen tube. Although provider comfort and surgical preference often influence choice of technique, patient size and quality of lung isolation required are also guiding factors.

Hyptis monticola Mart. ex Benth. (Lamiaceae) is an endemic species of altitude regions of Brazil. From the leaves of this plant, two 5,6-dihydro-α-pyrones, named monticolides A and B, have been reported as cytotoxic agents against different tumor cell lines. The isolation by high-speed countercurrent chromatography in combination with recycling preparative high-performance liquid chromatography of the undescribed monticolides C-F is presented. These compounds corresponded to a series of related monticolide derivatives differing from each other by the number of acyl substituents. Their characterization by mass spectrometry and nuclear magnetic resonance is also presented, in conjunction with an evidence by a simple chemical correlation for their absolute stereochemistry. The distribution of these chemical markers in extracts of flowers, leaves and branches collected in different seasons by electrospray ionization ion trap mass spectrometry in positive mode was analyzed. Multivariate data analyses indicated that seasonality affects monticolide concentrations in different organs of the aerial parts. Monticolides A-F seem to be present as the original markers of the analyzed plant. However, mono-, di- and triacetylated monticolides can undergo acid-catalyzed transesterifications and their natural yields estimated were affected during the isolation procedures.

Cell-derived Drug Delivery Systems (DDSs), particularly exosomes, have grown in popularity and have been increasingly explored as novel DDSs, due to their intrinsic targeting capabilities. However, clinical translation of exosomes is impeded by the tedious isolation procedures and poor yield. Cell-derived nanovesicles (CDNs) have recently been produced and proposed as exosome-mimetics. Various methods for producing exosome-mimetics have been developed. In this chapter, we present a simple, efficient, and cost-effective CDNs production method that uses common laboratory equipment (microcentrifuge) and spin cups. Through a series of extrusion and size exclusion steps, CDNs are produced from in vitro cell culture and are found to highly resemble the endogenous exosomes. Thus, we envision that this strategy holds great potential as a viable alternative to exosomes in the development of ideal DDS.

Introduction Ablation Index (AI) is a novel catheter-based parameter developed to improve and increase efficacy and safety of Pulmonary Vein Isolations (PVI) in the treatment of Atrial Fibrillation (AF). This method involves the incorporation of contact force, time and power in deliverance of ablation lines. The aim of this study is to evaluate the impact of AI on the AF free burden over one-year post redo ablation for persistent AF. The study evaluates a secondary endpoint in medical management escalations post procedure. Methods A retrospective single centre study of 39 patients (Mater Private Hospital, Dublin) who had redo ablations for persistent AF between the period of Jan 2016 till December 2019. We evaluated and analysed the efficacy of AI on redo PVIs in comparison to conventional established ablation techniques. Both groups were matched for age, gender, and duration of PeAF. Group 1 (17 patients) was the AI group with ablation index intervention and group 2 was the contact force (CF) -guided group with 22 patients. Each cardiologist carried out a minimum of 100 ablations per year to confirm the uniformity of performance. Patient follow-up data was analysed at staged intervals for one-year post procedure. Inclusion criteria are: Patients must have had one prior PVI procedure for PeAF, complete follow up data available and all procedures completed using the Carto 3D Mapping System. Statistical analysis was carried out using SPSS (IBM SPSS Statistic Version 26). A Kaplan Meier graph was generated to evaluate the AF free interval. All continuous variables were expressed as the mean ± SD and Students T Test to was applied to give the significant differences for continuous variables. Results Patient characteristics are demonstrated in Table 1. There were no significant differences in age, sex, weight, height, CHADVASC or anti-arrythmia agents, which indicated similar patient profile in each cohort. Pulmonary vein reconnections at redo PVI procedure were comparable at 100% in the AI group and 86% in the CF group (p = 0.1). Freedom from AF burden was (mean 8.72 ± 4.33 months) in CF group Versus (mean 9.35 ± 4.1 months) in AI guided ablation (p=0.71) (figure 1). The AI group demonstrated greater numbers of patients in whom antiarrhythmic therapy could be deescalated over one year (AI, n=11/17, 65% Vs. CF, n=2/22, 9% with p=0.01) while fewer patients underwent escalation of their antiarrhythmic therapy (AI n=2/17, 12% vs CF n=6/22, 27% p= 0.03) (table 1). Conclusion This is the first study to analyse the outcomes of ablation index on repeat PVI procedures. Despite no significant difference in AF recurrence outcomes demonstrated in this study, there was a significant difference in the medical de-escalation in favour of the use of the AI over the short term follow up period. This may reflect effective ablation lesions. A longer-term analysis would be recommended to determine the efficacy of AI use in PeAF redo procedures.

COVID-19 associated lung diseases can mimic radiological characteristics of other viral lung diseases such as influenza which may lead to misdiagnosis. In this study, we proposed an Artificial Intelligence framework based on a combination of a Convolutional Neural network architecture and a Recurrent Neural Network architecture to classify CT volumes with COVID-19, Influenza, and no-infection. The model was trained on a dataset of 300 patients (100 patients in each class). Each set of 15 consecutive axial slices with the associated label of the corresponding CT volume was input as a 3 channel input at 5 time points to the CNN-RNN network. Benchmarked against RT-PCR confirmed cases of COVID-19 and Influenza, our model, when evaluated on an independent validation set of 400 CT patients, can accurately classify CT volumes of patients with COVID-19, Influenza, or no-infection with a sensitivity of 96% (COVID-19) and 95% (Influenza) (Table1). Figure1 shows the percentage of correctly classified and misclassified cases in each class. Our model provides rapid accurate diagnosis in patients suspected of COVID-19 infection, facilitating the timely implementation of isolation procedures and early intervention.

A fully automated AI framework was developed to extract radiomics features from chest CT scans to detect COVID-19 patients. We curated and analysed the data from a total of 1381 patients. A cohort of 181 RT-PCR confirmed COVID-19 patients and 1200 control patients was included for model development. An independent dataset of 697 patients was used to validate the model. The datasets were collected from CHU Liege, Belgium. Model performance was assessed by the area under the receiver operating characteristic curve (AUC). Assuming 15% disease prevalence, a comprehensive analysis of classification performance in terms of accuracy, sensitivity, specificity, negative predictive value (NPV) and positive predictive value (PPV) was performed for all possible decision thresholds. The final curated dataset used for model development and testing consisted of chest CT scans of 1224 patients and 641 patients, respectively. The model had an AUC of 0.882 (95% CI: 0.851-0.913) in the independent test dataset. Assuming the cost of false negatives is twice as high as the cost of false positives, the optimal decision threshold resulted in an accuracy of 85.18%, a sensitivity of 69.52, a specificity of 91.63%, an NPV of 94.46% and a PPV of 59.44%. Our AI framework can accurately detect COVID-19. Thus, providing rapid accurate diagnosis in patients suspected of COVID-19 infection, facilitating the implementation of isolation procedures and early intervention.

Tuber and root crops are matured as fundamental food crops universally especially in tropical and subtropical regions. Among them, Taro (Colocasia esculenta) considered as 5th chief root crop due to its medicinal, ornamental and food formulation facets. Competitively it holds a considerable amount of starch even more than that to potato, sweet potato, cassava and so on. Taro corms starch (70-80%) contemplate as a cheapest abode for food industries due to its multifarious potentiality in food like stabilizer, emulsifier, fat substitute and as filler agent too. Baby foods, packaging material novelties, geriatric foods with resistant starch incorporation are the recently explored deliverables. Moreover, quality attributes of taro starch increases its behavioral versatility after modification competitively in a much better manner than native starch. This review aims to outline the current awareness about taro starch's molecular pattern, isolation procedures, properties, modifications and novel hit approaches for commercial viability. Gluten-free trait, hypoallergenic features and high digestibility are the additional benefits that widens its application scope to adapt better among pharma and textile in along with food sector.

Since the 1990s, improvements in ventilation techniques and isolation procedures have been widely credited with the decline in nosocomial transmission of tuberculosis and other airborne diseases. Little effort, however, has been made to study the risk of isolation patients acquiring secondary infections from contaminated air migrating into negatively pressurized isolation rooms from adjacent spaces. As a result, an actual hospital was used to observe the transport of aerosol from a nursing station and general patient room to a nearby airborne infectious isolation room (AIIR). Aerosols ≤3.0μm (viruses and most airborne bacteria) were found to be capable of migrating 14.5m from a general patient room to an AIIR anteroom entrance in 3.0μm to the entrance of the general patient room (4.5m).

Microbiological surveillance of the food chain plays a critical role in improving our understanding of the distribution and circulation of food-borne pathogens along the farm to fork continuum toward the development of interventions to reduce the burden of illness. The application of molecular subtyping to bacterial isolates collected through surveillance has led to the identification of strains posing the greatest risk to public health. Past evidence suggests that enrichment methods for Campylobacter jejuni, a leading bacterial foodborne pathogen worldwide, may lead to the differential recovery of subtypes, obscuring our ability to infer the composition of a mixed-strain sample and potentially biasing prevalence estimates in surveillance data. To assess the extent of potential selection bias resulting from enrichment-based isolation methods, we compared enrichment and non-enrichment isolation of mixed subtype cultures of C. jejuni, followed by subtype-specific enumeration using both colony plate-counts and digital droplet PCR. Results differed from the null hypothesis that similar proportions of C. jejuni subtypes are recovered from both methods. Our results also indicated a significant effect of subtype prevalence on isolation frequency post-recovery, with the recovery of more common subtypes being consistently favored. This bias was exacerbated when an enrichment step was included in the isolation procedure. Taken together, our results emphasize the importance of selecting multiple colonies per sample, and where possible, the use of both enrichment and non-enrichment isolation procedures to maximize the likelihood of recovering multiple subtypes present in a sample. Moreover, the effects of subtype-specific recovery bias should be considered in the interpretation of strain prevalence data toward improved risk assessment from microbiological surveillance data.

Singlet oxygen (1O2) generation quantum yields from chromophoric dissolved organic matter (CDOM) have been reported for many samples over the past 4 decades. Yet even for standardized isolates such as those from the International Humic Substance Society (IHSS), wide-ranging values exist in the literature. In this manuscript, time-resolved 1O2 phosphorescence was used to determine the 1O2 quantum yields (ΦΔ) of a variety of dissolved organic matter (DOM) isolates and natural waters. In general, the 1O2 quantum yield values in this study are in the middle, although below the median of the range of past reported values (e.g., for Suwannee River Natural Organic Matter IHSS isolate: 1.8% vs 0.23–2.89%). Notably, hydrophobic neutral fractions of DOM isolates were found to possess the highest 1O2 quantum yields, an interesting result given that these fractions are not retained in typical humic and fulvic acid isolation procedures that use XAD resins. The excitation wavelength dependence of 1O2 generation from CDOM was also examined, and an approximate linear decrease with longer excitation wavelength was observed. Transmission of varicella occurs frequently in schools and households. We investigated the characteristics of varicella cases derived from within-household transmission and the modes of varicella transmission between school and household settings in Shanghai, China, from 2009 to 2018. Within-household transmission occurred in 278 households, of which 134 transmission events were between children. Sixty-one household varicella transmission events may be attributed to isolation procedures for infected students during school outbreaks, and 7.6% of school outbreaks were caused by schoolchildren cases derived from within-household transmission. The frequency of ‘school-household-school’ transmission adds an additional layer of complexity to the control of school varicella outbreaks. Administration of varicella vaccine as post-exposure prophylaxis after exposure is considered to be an effective measure to control varicella spread within households and schools.

Extracellular vesicles (EVs) are small vesicles ensuring transport of molecules between cells and throughout the body. Their small size and biological and physical functions make them optimal candidates as therapeutic agents in several medical frameworks. The elucidation of biophysical and biochemical characteristics of EVs is crucial for the understanding of their interaction with recipient cells and their functional activity. In particular, for therapeutic applications understanding how the process of manufacturing affects the biological function of EVs is mandatory before going to the clinical testing. The absence of standardized methodologies and technologies to establish reliable criteria has been the main hurdle for real therapeutic applications of EVs. Here we exploited a combination of different biophysical techniques (Fourier Transform Infrared Spectroscopy, Ultraviolet Resonant Raman Spectroscopy, Atomic Force Microscopy and Small Angle X-Ray Scattering) to screen EVs derived from umbilical cord-mesenchymal stem cells with different isolation procedures, evidencing subtle differences in the biophysical properties of EVs. The results underline the need of a multiparametric analysis to address size, stability and purity of different preparations and to correlate phenotypic parameters of EVs with their functional activity.

Two previously undescribed chimonanthine-type [sinodamines A and B] and five related known dimeric tryptamine-derived alkaloids were isolated and characterized from the leaves of the endangered ornamental plant Sinocalycanthus chinensis under the guidance of LC-MS detection and dereplication analyses, along with conventional isolation procedures. Their structures were established on the basis of spectroscopic methods and chemical transformations. Sinodamine A can be regarded as the naturally occurring N-oxide derivative of its pseudo-mesomer sinodamine B. An acid-catalyzed Meisenheimer rearrangement from sinodamine A to its oxazine-form with a final equilibrium of 1:2 was observed by monitoring their NMR spectra. (−)-Folicanthine showed significant cytotoxicity against human lung carcinoma A549 and colorectal carcinoma HT29 cells, with IC50 values of 7.76 and 6.16 μM, respectively.

Extracellular vesicles (EVs) are particles naturally released from cells, delimited by a lipid bilayer, carrying functionally active biological molecules. In addition to their physiological role in cellular communication, the interest of the scientific community has recently turned to the use of EVs as vehicles for delivering therapeutic molecules. Several attempts are being made to ameliorate drug encapsulation and targeting, but these efforts are thwarted if the starting material does not meet stringent quality criteria. Here, we take a step back to the sources and isolation procedures that could guarantee significant improvements in the purification of EVs to be used as drug carriers, highlighting the advantages and shortcomings of each approach.

Finding the conduction gaps in redo PV isolation procedures is challenging, and several maneuvers have been described. In the present study, we analyze the pace and map (PM) maneuver [atrial mapping during pulmonary vein (PV) pacing] to locate the gaps in redo PV isolation procedures.Consecutive patients undergoing a second PV isolation procedure at a single institution over a 4-year period were included. For the last 2 years, all the patients (n = 38) studied underwent PV isolation based on the PM maneuver and were compared to the previous patients (n = 45). The atrial side of the ablation line was mapped with the ablation catheter during PV pacing, and the earliest site was considered a gap site.Shorter radiofrequency time was required to obtain PV isolation in the PM group (485 ± 374 vs. 864 ± 544 s; p 0.001), which remained significant after adjusting for the number of reconnected PVs (p = 0.01). We did not find significant differences in the procedure duration (106 ± 46 vs. 112 ± 53 min; p = 0.57) and arrhythmia recurrence during 1-year follow-up (26.6 vs. 28.9%; p = 0.82) after adjusting for several confounding factors (HR 1.32; 95% CI 0.5-3.4; p = 0.57).PM is a simple maneuver to identify the gaps in ablation lines around the PV. It remains efficacious in redo procedures despite the difficulties in localizing the ablation lines performed in the first procedure. The PM maneuver reduced the radiofrequency time required to isolate the PV without compromising the efficacy.

Amaranth, chia and quinoa seeds contain proteins which have biological and functional properties that provide nutritional benefits due to their reasonably well-balanced aminoacid content. This review analyses these vegetable proteins and focuses on recent research on protein classification and isolation as well as structural characterization by means of fluorescence spectroscopy, surface hydrophobicity and differential scanning calorimetry. Isolation procedures have a profound influence on the structural properties of the proteins and, therefore, on their in vitro digestibility. The present article provides a comprehensive overview of the properties and characterization of these proteins.

Soil protists are major biotic forces driving microbial dynamics within aggregates. Interpretation of protistan spatial distribution and ecological processes in aggregates primarily relies on aggregate isolation methods. However, comparative studies illustrating how isolation methods influence protistan communities are lacking. Here, two isolation procedures, dry- and wet-sieving, were evaluated. The wet-sieving led to separation of dry mega-aggregates, which consequently altered aggregate size distribution and associated protistan communities. The variable influences of sieving methods on aggregate protists were dependent on soil type. However, the aggregate isolation method did not affect the prominent aggregate-effects on protistan communities and detection of dominant taxa. Collectively, our findings highlight the importance of isolation procedures for exploring the spatial heterogeneity of protists at the micro-level.

Natural polymer is a frequently used polymer in various food applications and pharmaceutical formulations due to its benefits and its biocompatibility compared to synthetic polymers. One of the natural polymer groups (i.e., polysaccharide) does not only function as an additive in pharmaceutical preparations, but also as an active ingredient with pharmacological effects. In addition, several natural polymers offer potential distinct applications in gene delivery and genetic engineering. However, some of these polymers have drawbacks, such as their lack of water retention and elasticity. Sacran, one of the high-molecular-weight natural polysaccharides (megamolecular polysaccharides) derived from Aphanothece sacrum (A. sacrum), has good water retention and elasticity. Historically, sacran has been used as a dietary food. Moreover, sacran can be applied in biomedical fields as an active material, excipient, and genetic engineering material. This article discusses the characteristics, extraction, isolation procedures, and the use of sacran in food and biomedical applications.

Diversity analyses of bioaerosol samples from highly loaded workplaces as found in agricultural production or waste management help to improve the knowledge of exposure levels of workers. However, different used methods resulting in the detection of different bacterial species at the same work places. The present study obviously supports the deviation of received results using cultivation and further isolation approaches. Within the present study, the bacterial community at workplaces was estimated using the powerful tool of 16S rRNA gene sequence analyses after cultivation procedure. To avoid complex isolation procedures, the suitability of cultivation and subsequent cloning procedures was determined in bioaerosols from a duck hatchery. Diversity analysis of one bioaerosol sample, which was prepared independently three times in parallel, resulted in similarity values of 38.5%-57.1%. Further, similarity analysis calculated from three independent bioaerosol samplings on 1 day resulted in 31%-40% similarity. Although similar concentration between 2.22 × 106 and 4.46 × 106 CFU per m3 hatchery air were measured, in a ring-like trail, diversity analyses from six labs differ widely, resulting in 38.9%-78.6% divergence. The present method seems to be very useful for diversity analysis of bioaerosol samples, although heterogeneity in monitoring of airborne bacteria via cultivation was pointed out.

Intraoperative application of the stromal vascular fraction (SVF) of adipose tissue requires a fast and efficient isolation procedure of adipose tissue. This review was performed to systematically assess and compare procedures currently used for the intraoperative isolation of cellular SVF (cSVF) and tissue SVF (tSVF) that still contain the extracellular matrix. Pubmed, EMBASE and the Cochrane central register of controlled trials databases were searched for studies that compare procedures for intraoperative isolation of SVF (searched 28 September 2016). Outcomes of interest were cell yield, viability of cells, composition of SVF, duration, cost and procedure characteristics. Procedures were subdivided into procedures resulting in a cSVF or tSVF. Thirteen out of 3038 studies, evaluating 18 intraoperative isolation procedures, were considered eligible. In general, cSVF and tSVF intraoperative isolation procedures had similar cell yield, cell viability and SVF composition compared to a nonintraoperative (i.e. culture laboratory-based collagenase protocol) control group within the same studies. The majority of intraoperative isolation procedures are less time consuming than nonintraoperative control groups, however. Intraoperative isolation procedures are less time-consuming than nonintraoperative control groups with similar cell yield, viability of cells and composition of SVF, and therefore more suitable for use in the clinic. Nevertheless, none of the intraoperative isolation procedures could be designated as the preferred procedure to isolate SVF. Copyright © 2017 John Wiley & Sons, Ltd.
