Abstract

Minute examination of local lattice structures in matter affected by impurity doping is of special importance for the development of functional materials. In order to obtain microscopic information on spinel ferrites, in the present work, we introduced nonmagnetic In3+ ions in Fe3O4 and probed their site selectivity and the doping effect on the local lattice structures and bulk magnetism by means of 57Fe Mössbauer spectroscopy and positron annihilation spectroscopies. The Mössbauer parameters of the area intensity and isomer shift (IS) show that In3+ ions predominantly reside in the tetrahedral A site, especially at low doping level. With increasing concentration of In ions, however, they gradually occupy the octahedral B site replacing Fe3+ ions. Along with the site information, the IS values confirmed that the introduced In ions squeeze the B-site Fe ions at their nearest neighbors. Supporting results were obtained from positron annihilation lifetime spectroscopy; positron lifetimes become shorter with increasing In concentration, signifying that the oxygen ions are pressed by the introduced In ions resulting in lowering the volume of the adjacent lattice vacancies. The results of Doppler broadening spectroscopy also support the squeezing effect; the positrons in the vacancies adjacent to In ions are more likely to annihilate with the inner shell electrons of the surrounding oxygen ions as a result of a reduction in the vacancy volume. Abstract

Objective To investigate the immunology of host-tumour interaction, critical for the development of immunotherapy against cancers, by assessing the major histocompatibility complex (MHC) class I expression in both benign and malignant prostate disease, and the relationship between their expression and degree of tumour-infiltrating lymphocytes. Materials and methods Direct serial analysis of gene expression in tumours is an extremely sensitive and powerful tool for monitoring immunological changes in the immunotherapy of solid tumours. Most previous monitoring protocols rely mainly on the analysis of patient's peripheral blood but in the present study the direct molecular analysis of small tissue samples was used, and its accuracy compared with that of conventional immunohistochemical analysis. Twenty-four formalin-fixed, paraffin-embedded prostate samples (11 benign and 13 carcinoma) were used for the immunohistochemical analysis of CD8[SUP+] T lymphocytes and MHC class I expression. CD8[SUP+] T lymphocytes were counted using an ocular grid and MHC class I measured using digital image-analysis software. Twenty-seven frozen prostate tissue samples (12 benign and 15 carcinoma) were used for direct gene measurements of CD8 and interferon-&b.gamma; using a quantitative real-time polymerase chain reaction. Results There were significantly fewer CD8[SUP+] T lymphocytes in prostate carcinoma nests than in benign prostate. There was a significant correlation between the number of CD8[SUP+] T lymphocytes and MHC class I expression in the prostate. There was a strong correlation between the immunohistochemical estimates of CD8[SUP+] T lymphocytes and CD8 gene by polymerase chain reaction, but no significant difference between benign prostate and prostate carcinoma tissue in gene measurements. RNA-DNA covalent hybrids containing viral RNA have been isolated from nuclear fractions of Rous sarcoma virus-infected chicken embryo fibroblast cells shortly after virus infection. The formation of covalent hybrid structures depends upon a functional reverse transcriptase in vivo, since its appearance in cells is temperature dependent when infected with Rous sarcoma virus mutant LA335, which contains a temperature-sensitive reverse transcriptase.

50. Characterization of Y73, an avian sarcoma virus: a unique transforming gene and its product, a phosphopolyprotein with protein kinase activity.

Abstract

The Y73 strain of avian sarcoma virus recently isolated in Japan is defective in replication and is associated with subgroup A leukosis virus (YAV). The virus caused sarcoma but not acute leukosis when inoculated into chickens. Studies on the viral RNA showed that a 26S RNA, etimated to be 4.8 kilobases long, was Y73 viral RNA carrying a transforming gene. The 26S RNA has sequences in common with the RNA of an avian leukosis virus but no homology with the src gene sequence of avian sarcoma virus (ASV). Thus, Y73 has a unique sarcoma-inducing gene. A phosphorylated polyprotein of 90,000 daltons (p90) was immunoprecipitated from extracts of Y73-transformed chicken embryo cells by a variety of antisera reacting with gag gene products. When a bacteria-bound immunocomplex containing the p90 protein was incubated with [gamma-32P]ATP, the Y73-specific p90 and the IgG heavy chain were phosphorylated by a p90-associated protein kinase. The amino acid phosphorylated in vitro was exclusively tyrosine in both cases, whereas p90 phosphorylated in vivo contained phosphoserine as a major phospho amino acid with traces of phosphotyrosine and phosphothreoine.
