Abstract

Research has been conducted on the Mn atoms which function as catalysts in the water-oxidizing complex in the PS II oxidizing site. The effect of the PS II proteins' amine groups on the photoligation of the Mn (super)2+ to the apo water-oxidizing complex has been investigated via photoactivation and chemical modification techniques and the results are discussed.

Abstract

The function of the extrinsic 33 kilodalton protein (OEC33) in the turnover of the reaction center-binding protein D1 during photoinhibition was studied by measuring D1 degradation following strong illumination of samples with and without OEC33, PSII-enriched membrane and spinach manganese-depleted samples. The results demonstrated that the presence of the OEC33 greatly affects the light-induced degradation and aggregation of D1. The effects were attributed to the role of the OEC33 in maintaining the stability of PS II required for the efficient turnover of the D1 protein.

30. Characterization of the stromal protease(s) degrading the cross-linked products of the D1 protein generated by photoinhibition of photosystem II

Abstract

When photosystem (PS) II-enriched membranes are exposed to strong light, cross-linking of the intrinsic D1 protein with the surrounding polypeptides and degradation of the D1 protein take place. The cross-linking of the D1 protein with the α-subunit of cytochrome b559is suggested to be an early event of photoinduced damage to the D1 protein (Barbato et al., FEBS Lett. 309 (1992) 165–169). The relationship between the cross-linking and the degradation of the D1 protein, however, is not yet clear. In the present study, we show that the addition of stromal extract from chloroplasts degrades the 41 kDa cross-linked product of D1/cytochrome b559α-subunit and enhances the degradation of the D1 protein. Incubation of the preilluminated PS II-enriched membranes with the stromal extract at 25°C causes the degradation of the cross-linked product by more than 70%. The activity of the stromal extract showed a pH optimum at 8.0, and was enhanced by the addition of ATP or GTP. Consistent with the nucleotide effect, this stromal activity was eliminated by the preincubation of the stromal extract with apyrase, which hydrolyzes nucleotides. Also, the stromal activity was nearly fully inhibited by a serine-type protease inhibitor, 3,4-dichloroisocoumarin, which suggests participation of a serine-type protease(s).

Abstract

Precise measurement of blood flow is important because blood flow closely correlates formation of thrombus and atherosclerotic plaque. Among clinically applied modalities for blood flow measurement, color Doppler ultrasound shows two-dimensional (2D) distribution of one-dimensional blood flow component along the ultrasound beam. In the present study, 2D blood flow vector is obtained with high temporal and bidirectional Doppler ultrasound technique. Linear array probe with the central frequency of 7.5 MHz and an ultrasound data acquisition system with 128 transmit and 128 receive channels were equipped. Frame rate of 5 kHz was achieved by parallel receive beam forming with a wide transmitted wave. The flow velocity was measured from two different angles by beam steering. The interval of two measurements was 0.8 msec and it was considered as almost one moment to obtain 2D blood flow vector. B-mode image and 2D blood flow vector of the pulsatile flow in a carotid artery model showed small vortex at the bifurcation area. The method was also applied for visualization of in vivo blood flow vector in human carotid arteries. 2D blood flow measurement may predict the risk area of thrombus and plaque formation induced by abnormal blood flow.
