ASCOT (an acronym derived from Ag-Silver, Copper Oxide, Titanium Oxide) is a user-friendly web tool for digital construction of electrically neutral, energy-minimized spherical nanoparticles (NPs) of Ag, CuO, and TiO2 (both Anatase and Rutile forms) in vacuum, integrated into the Enalos Cloud Platform (https://www.enaloscloud.novamechanics.com/sabydoma/ascot/). ASCOT calculates critical atomistic descriptors such as average potential energy per atom, average coordination number, common neighbour parameter (used for structural classification in simulations of crystalline phases), and hexatic order parameter (which measures how closely the local environment around a particle resembles perfect hexatic symmetry) for both core (over 4 Å from the surface) and shell (within 4 Å of the surface) regions of the NPs. These atomistic descriptors assist in predicting the most stable NP size based on lowest per atom energy and serve as inputs for developing machine learning models to predict the toxicity of these nanomaterials. ASCOT's automated backend requires minimal user input in order to construct the digital NPs: inputs needed are the material type (Ag, CuO, TiO2-Anatase, TiO2-Rutile), target diameter, a Force-Field from a pre-validated list, and the energy minimization parameters, with the tool providing a set of default values for novice users.

A robust predictive model was developed using 136 novel peroxisome proliferator-activated receptor delta (PPARδ) agonists, a distinct subtype of lipid-activated transcription factors of the nuclear receptor superfamily that regulate target genes by binding to characteristic sequences of DNA bases. The model employs various structural descriptors and docking calculations and provides predictions of the biological activity of PPARδ agonists, following the criteria of the Organization for Economic Co-operation and Development (OECD) for the development and validation of quantitative structure–activity relationship (QSAR) models. Specifically focused on small molecules, the model facilitates the identification of highly potent and selective PPARδ agonists and offers a read-across concept by providing the chemical neighbours of the compound under study. The model development process was conducted on Isalos Analytics Software (v. 0.1.17) which provides an intuitive environment for machine-learning applications. The final model was released as a user-friendly web tool and can be accessed through the Enalos Cloud platform’s graphical user interface (GUI).

In mediaeval Europe, the term “commons” described the way that communities managed land that was held “in common” and provided a clear set of rules for how this “common land” was used and developed by, and for, the community. Similarly, as we move towards an increasingly knowledge-based society where data is the new oil, new approaches to sharing and jointly owning publicly funded research data are needed to maximise its added value. Such common management approaches will extend the data’s useful life and facilitate its reuse for a range of additional purposes, from modelling, to meta-analysis to regulatory risk assessment as examples relevant to nanosafety data. This “commons” approach to nanosafety data and nanoinformatics infrastructure provision, co-development, and maintenance is at the heart of the “NanoCommons” project and underpins its post-funding transition to providing a basis on which other initiatives and projects can build. The present paper summarises part of the NanoCommons infrastructure called the NanoCommons Knowledge Base. It provides interoperability for nanosafety data sources and tools, on both semantic and technical levels. The NanoCommons Knowledge Base connects knowledge and provides both programmatic (via an Application Programming Interface) and a user-friendly graphical interface to enable (and democratise) access to state of the art tools for nanomaterials safety prediction, NMs design for safety and sustainability, and NMs risk assessment, as well. In addition, the standards and interfaces for interoperability, e.g., file templates to contribute data to the NanoCommons, are described, and a snapshot of the range and breadth of nanoinformatics tools and models that have already been integrated are presented Finally, we demonstrate how the NanoCommons Knowledge Base can support users in the FAIRification of their experimental workflows and how the NanoCommons Knowledge Base itself has progressed towards richer compliance with the FAIR principles.

Abstract Efficient and machine-readable representations are needed to accurately identify, validate and communicate information of chemical structures. Many such representations have been developed (as, for example, the Simplified Molecular-Input Line-Entry System and the IUPAC International Chemical Identifier), each offering advantages specific to various use-cases. Representation of the multi-component structures of nanomaterials (NMs), though, remains out of scope for all the currently available standards, as the nature of NMs sets new challenges on formalizing the encoding of their structure, interactions and environmental parameters. In this work we identify a set of principles that a NM representation should adhere to in order to provide “machine-friendly” encodings of NMs, i.e. encodings that facilitate machine processing and cooperation with nanoinformatics tools. We illustrate our principles by showing how the recently introduced InChI-based NM representation, might be augmented, in principle, to also encode morphology and mixture properties, distributions of properties, and also to capture auxiliary information and allow data reuse.

Traditionally, toxicology has strong links with other fields such as biology, chemistry, pharmacology and medicine. With the rise of synthetic and new engineered materials, alongside ongoing prioritisation needs in chemical risk assessment for existing chemicals, early predictive evaluations are becoming of utmost importance to both scientific and regulatory purposes. ELIXIR is an intergovernmental organisation that brings together life science resources from across Europe. To coordinate the linkage of various life science efforts around modern predictive toxicology, the establishment of a new ELIXIR Community is seen as instrumental. In the past few years, joint efforts, building on incidental overlap, have been piloted in the context of ELIXIR. For example, the EU-ToxRisk, diXa, HeCaToS, transQST, and the nanotoxicology community have worked with the ELIXIR TeSS, Bioschemas, and Compute Platforms and activities. In 2018, a core group of interested parties wrote a proposal, outlining a sketch of what this new ELIXIR Toxicology Community would look like. A recent workshop (held September 30th to October 1st, 2020) extended this into an ELIXIR Toxicology roadmap and a shortlist of limited investment-high gain collaborations to give body to this new community. This Whitepaper outlines the results of these efforts and defines our vision of the ELIXIR Toxicology Community and how it complements other ELIXIR activities.

(1) Background: Despite the encouraging indications regarding the suitability (biocompatibility) of iron carbide nanoparticles (ICNPs) in various biomedical applications, the published evidence of their biosafety is dispersed and relatively sparse. The present review synthesizes the existing nanotoxicological data from in vitro studies relevant to the diagnosis and treatment of cancer. (2) Methods: A systematic review was performed in electronic databases (PubMed, Scopus, and Wiley Online Library) on December 2023, searching for toxicity assessments of ICNPs of different sizes, coatings, and surface modifications investigated in immortalized human and murine cell lines. The risk of bias in the studies was assessed using the ToxRTool for in vitro studies. (3) Results: Among the selected studies (n = 22), cell viability emerged as the most frequently assessed cellular-level toxicity endpoint. The results of the meta-analysis showed that cell models treated with ICNPs had a reduced cell viability (SMD = −2.531; 95% CI: −2.959 to −2.109) compared to untreated samples. A subgroup analysis was performed due to the high magnitude of heterogeneity (I2 = 77.1%), revealing that ICNP concentration and conjugated ligands are the factors that largely influence toxicity (p < 0.001). (4) Conclusions: A dose-dependent cytotoxicity of ICNP exposure was observed, regardless of the health status of the cell, tested organism, and NP size. Inconsistent reporting of ICNP physicochemical properties was noted, which hinders comparability among the studies. A comprehensive exploration of the available in vivo studies is required in future research to assess the safety of ICNPs’ use in bioimaging and cancer treatment.

Introduction: Significant progress has been made in terms of best practice in research data management for nanosafety. Some of the underlying approaches to date are, however, overly focussed on the needs of specific research projects or aligned to a single data repository, and this “silo” approach is hampering their general adoption by the broader research community and individual labs.Methods: State-of-the-art data/knowledge collection, curation management FAIrification, and sharing solutions applied in the nanosafety field are reviewed focusing on unique features, which should be generalised and integrated into a functional FAIRification ecosystem that addresses the needs of both data generators and data (re)users.Results: The development of data capture templates has focussed on standardised single-endpoint Test Guidelines, which does not reflect the complexity of real laboratory processes, where multiple assays are interlinked into an overall study, and where non-standardised assays are developed to address novel research questions and probe mechanistic processes to generate the basis for read-across from one nanomaterial to another. By focussing on the needs of data providers and data users, we identify how existing tools and approaches can be re-framed to enable “on-the-fly” (meta) data definition, data capture, curation and FAIRification, that are sufficiently flexible to address the complexity in nanosafety research, yet harmonised enough to facilitate integration of datasets from different sources generated for different research purposes. By mapping the available tools for nanomaterials safety research (including nanomaterials characterisation, nonstandard (mechanistic-focussed) methods, measurement principles and experimental setup, environmental fate and requirements from new research foci such as safe and sustainable by design), a strategy for integration and bridging between silos is presented. The NanoCommons KnowledgeBase has shown how data from different sources can be integrated into a one-stop shop for searching, browsing and accessing data (without copying), and thus how to break the boundaries between data silos.Discussion: The next steps are to generalise the approach by defining a process to build consensus (meta)data standards, develop solutions to make (meta)data more machine actionable (on the fly ontology development) and establish a distributed FAIR data ecosystem maintained by the community beyond specific projects. Since other multidisciplinary domains might also struggle with data silofication, the learnings presented here may be transferrable to facilitate data sharing within other communities and support harmonization of approaches across disciplines to prepare the ground for cross-domain interoperability.

Nanoplastic contamination is a serious environmental concern and could have implications on plant life depending upon interactions. Here, the authors study the effect of size and charge on the accumulation and uptake of model polymer nanoparticles by plant roots which has implications for environmental exposure and nanoparticle delivery to plants.

Abstract Management of nanomaterials and nanosafety data needs to operate under the FAIR (findability, accessibility, interoperability, and reusability) principles and this requires a unique, global identifier for each nanomaterial. Existing identifiers may not always be applicable or sufficient to definitively identify the specific nanomaterial used in a particular study, resulting in the use of textual descriptions in research project communications and reporting. To ensure that internal project documentation can later be linked to publicly released data and knowledge for the specific nanomaterials, or even to specific batches and variants of nanomaterials utilised in that project, a new identifier is proposed: the European Registry of Materials Identifier. We here describe the background to this new identifier, including FAIR interoperability as defined by FAIRSharing, identifiers.org, Bioregistry, and the CHEMINF ontology, and show how it complements other identifiers such as CAS numbers and the ongoing efforts to extend the InChI identifier to cover nanomaterials. We provide examples of its use in various H2020-funded nanosafety projects. Graphical Abstract

The importance of the cladoceran Daphnia as a model organism for ecotoxicity testing has been well-established since the 1980s. Daphnia have been increasingly used in standardised testing of chemicals as they are well characterised and show sensitivity to pollutants, making them an essential indicator species for environmental stress. The mapping of the genomes of D. pulex in 2012 and D. magna in 2017 further consolidated their utility for ecotoxicity testing, including demonstrating the responsiveness of the Daphnia genome to environmental stressors. The short lifecycle and parthenogenetic reproduction make Daphnia useful for assessment of developmental toxicity and adaption to stress. The emergence of nanomaterials (NMs) and their safety assessment has introduced some challenges to the use of standard toxicity tests which were developed for soluble chemicals. NMs have enormous reactive surface areas resulting in dynamic interactions with dissolved organic carbon, proteins and other biomolecules in their surroundings leading to a myriad of physical, chemical, biological, and macromolecular transformations of the NMs and thus changes in their bioavailability to, and impacts on, daphnids. However, NM safety assessments are also driving innovations in our approaches to toxicity testing, for both chemicals and other emerging contaminants such as microplastics (MPs). These advances include establishing more realistic environmental exposures via medium composition tuning including pre-conditioning by the organisms to provide relevant biomolecules as background, development of microfluidics approaches to mimic environmental flow conditions typical in streams, utilisation of field daphnids cultured in the lab to assess adaption and impacts of pre-exposure to pollution gradients, and of course development of mechanistic insights to connect the first encounter with NMs or MPs to an adverse outcome, via the key events in an adverse outcome pathway. Insights into these developments are presented below to inspire further advances and utilisation of these important organisms as part of an overall environmental risk assessment of NMs and MPs impacts, including in mixture exposure scenarios.

Microplastics (MPs) have been widely detected in the world's water, which may pose a significant threat to the ecosystem as a whole and have been a subject of much attention because their presence impacts seas, lakes, rivers, and even the Polar Regions. There have been numerous studies that report direct adverse effects on marine organisms, but only a few have explored their ecological effects on freshwater organisms. In this field, there is still a lack of a systematic overview of the toxic effects and mechanisms of MPs on aquatic organisms, as well as a consistent understanding of the potential ecological consequences. This review describes the fate and impact on marine and freshwater aquatic organisms. Further, we examine the toxicology of MPs in order to uncover the relationship between aquatic organism responses to MPs and ecological disorders. In addition, an overview of the factors that may affect the toxicity effects of MPs on aquatic organisms was presented along with a brief examination of their identification and characterization. MPs were discussed in terms of their physicochemical properties in relation to their toxicological concerns regarding their bioavailability and environmental impact. This paper focuses on the progress of the toxicological studies of MPs on aquatic organisms (bacteria, algae, Daphnia, and fish, etc.) of different trophic levels, and explores its toxic mechanism, such as behavioral alternations, metabolism disorders, immune response, and poses a threat to the composition and stability of the ecosystem. We also review the main factors affecting the toxicity of MPs to aquatic organisms, including direct factors (polymer types, sizes, shapes, surface chemistry, etc.) and indirect factors (persistent organic pollutants, heavy metal ions, additives, and monomer, etc.), and the future research trends of MPs ecotoxicology are also pointed out. The findings of this study will be helpful in guiding future marine and freshwater rubbish studies and management strategies.

We present here a technique for setting up detection limits on any fluorescent microscope in conjunction with the fluorophore Nile Red for microplastic identification. Our method also describes a rigorous morphology-specific identification key for microplastics to reduce subjectivity between researchers. The detection limits were established for nine common polymer types and five natural substrates which could result in false-positive signals when using Nile Red for microplastic identification. This method was then applied to real freshwater samples and identified particles were validated with micro-FTIR or Raman spectroscopy. The aquatic system is a major sink for engineered nanomaterials released into the environment. Here, we assessed the toxicity of graphene oxide (GO) using the freshwater planarian Dugesia japonica, an invertebrate model that has been widely used for studying the effects of toxins on tissue regeneration and neuronal development. GO not only impaired the growth of normal (homeostatic) worms, but also inhibited the regeneration processes of regenerating (amputated) worms, with LC10 values of 9.86 mg/L and 9.32 mg/L for the 48-h acute toxicity test, respectively. High concentration (200 mg/L) of GO killed all the worms after 3 (regenerating) or 4 (homeostasis) days of exposure. Whole-mount in situ hybridization (WISH) and immunofluorescence analyses suggest GO impaired stem cell proliferation and differentiation, and subsequently caused cell apoptosis and oxidative DNA damage during planarian regeneration. Mechanistic analysis suggests that GO disturbed the antioxidative system (enzymatic and non-enzymatic) and energy metabolism in the planarian at both molecular and genetic levels, thus causing reactive oxygen species (ROS) over accumulation and oxidative damage, including oxidative DNA damage, loss of mitochondrial membrane integrity, lack of energy supply for cell differentiation and proliferation leading to retardance of neuron regeneration. The intrinsic oxidative potential of GO contributes to the GO-induced toxicity in planarians. These data suggest that GO in aquatic systems can cause oxidative stress and neurotoxicity in planarians. Overall, regenerated tissues are more sensitive to GO toxicity than homeostatic ones, suggesting that careful handling and appropriate decisions are needed in the application of GO to achieve healing and tissue regeneration.

The call for adaptive governance approaches to guide the sustainable transformation of urban water management systems is growing amongst scholars and policy professionals. Responding to this call, the Global North (GN) has focused significant evidence-based research on issues of scale, capacity, and institutional arrangements to support such transformations, whereas evidence from the Global South remains nascent. This paper contributes to the growing body of knowledge from the Global South, discussing how adaptive governance operates under different local contexts and conditions. Following empirical investigations in two cities in Bangladesh, which involved 58 semi-structured interviews, 17 oral histories, and secondary data analysis, and drawing on the adaptive capacity and attributes framework, we examined how scale, capacity, and institutional hybridization might deliver the conditions necessary for guiding a sustainable transformation in water governance. The research revealed that a large-scale urban system such as Dhaka is currently experiencing “lock-in” due to ongoing investments in large-scale infrastructure, inappropriate transfer of technology from GN contexts, bureaucratic complexity, and general resistance to change. In contrast, the relatively smaller urban system represented by the secondary city Mymensingh was found to be more open, flexible, showcasing key enabling factors that might support sustainable growth. Overall, this study sheds light on the role of adaptive governance in the context of system scales and capacity (i.e., institutional / organizational / individual) and reveals how capacity development is linked to key enabling attributes including multi-level and polycentric institutions, participatory approaches, networking, bridging organizations, and leadership. Collectively these findings offer insights into how adaptive attributes can inform sustainable transformation processes

The water governance crisis has critical implications for the transformation of cities through establishment of sustainable water management practices. Adaptive governance approaches, with their potential to address global water governance challenges, are emerging in the context of the global South (GS). A key feature of adaptive governance is its enabling context, or hybridized governance approach which bridges organizational and network activities across multiple implementation scales. Transforming urban water management toward sustainable water governance practices requires flexibility and agility, and a willingness to enable new ideas, features often associated with smaller and newer cities (secondary cities) which are less constrained than the major cities. However, unpacking their potentiality and the scope of such secondary cities to lead the way in transitioning to sustainable water governance remains under-researched. To address this gap, a qualitative study in a representative GS secondary city (Mymensingh in Bangladesh) was undertaken to investigate whether national and local strategies directed toward improving governance and management capacity of the local municipality are yielding sustainable transformations. We identified a significant shift within the governance regime that influences existing power dynamics and decision-making for the delivery of urban water services. Within the traditional state-led governance structure, a hybridized governance is emerging that builds both institutional and actor capacity. However, these hybridized governance activities are strongly dependent on (international) donor investment and guidance, therefore the presence or absence of donor support will likely determine the impact of these activities in the future.

Nanotoxicology is a relatively new field of research concerning the study and application of nanomaterials to evaluate the potential for harmful effects in parallel with the development of applications. Nanotoxicology as a field spans materials synthesis and characterisation, assessment of fate and behaviour, exposure science, toxicology / ecotoxicology, molecular biology and toxicogenomics, epidemiology, safe and sustainable by design approaches, and chemoinformatics and nanoinformatics, thus requiring scientists to work collaboratively, often outside their core expertise area. This interdisciplinarity can lead to challenges in terms of interpretation and reporting, and calls for a platform for sharing of best-practice in nanotoxicology research. The F1000Research Nanotoxicology collection, introduced via this editorial, will provide a place to share accumulated best practice, via original research reports including no-effects studies, protocols and methods papers, software reports and living systematic reviews, which can be updated as new knowledge emerges or as the domain of applicability of the method, model or software is expanded. This editorial introduces the Nanotoxicology Collection in F1000Research. The aim of the collection is to provide an open access platform for nanotoxicology researchers, to support an improved culture of data sharing and documentation of evolving protocols, biological and computational models, software tools and datasets, that can be applied and built upon to develop predictive models and move towards in silico nanotoxicology and nanoinformatics. Submissions will be assessed for fit to the collection and subjected to the F1000Research open peer review process.

Nanotechnology has enabled the discovery of a multitude of novel materials exhibiting unique physicochemical (PChem) properties compared to their bulk analogues. These properties have led to a rapidly increasing range of commercial applications; this, however, may come at a cost, if an association to long-term health and environmental risks is discovered or even just perceived. Many nanomaterials (NMs) have not yet had their potential adverse biological effects fully assessed, due to costs and time constraints associated with the experimental assessment, frequently involving animals. Here, the available NM libraries are analyzed for their suitability for integration with novel nanoinformatics approaches and for the development of NM specific Integrated Approaches to Testing and Assessment (IATA) for human and environmental risk assessment, all within the NanoSolveIT cloud-platform. These established and well-characterized NM libraries (e.g. NanoMILE, NanoSolutions, NANoREG, NanoFASE, caLIBRAte, NanoTEST and the Nanomaterial Registry (>2000 NMs)) contain physicochemical characterization data as well as data for several relevant biological endpoints, assessed in part using harmonized Organisation for Economic Co-operation and Development (OECD) methods and test guidelines. Integration of such extensive NM information sources with the latest nanoinformatics methods will allow NanoSolveIT to model the relationships between NM structure (morphology), properties and their adverse effects and to predict the effects of other NMs for which less data is available. The project specifically addresses the needs of regulatory agencies and industry to effectively and rapidly evaluate the exposure, NM hazard and risk from nanomaterials and nano-enabled products, enabling implementation of computational ‘safe-by-design’ approaches to facilitate NM commercialization.

Nanoparticle incorporated membranes are potential tools to remove hazardous organic pollutants from drinking water sources. Cellulose acetate membranes were impregnated by incorporating zinc nanoparticles, produced using a green synthesis with Mentha plant extract utilizing microwave energy. The synthesized nanoparticles were extensively characterized using ultraviolet–visible spectroscopy, X-ray diffraction, Scanning electron microscopy, energy dispersive X-ray spectroscopy and Nanoparticle tracking analysis. The spectroscopy results showed an absorbance peak at 300 nm, confirming the successful synthesis, while X-ray diffraction confirmed that the particles are crystalline with a mean particle size of 36 nm. The surface roughness and topography of the zinc nanoparticle impregnated membranes was studied using atomic force microscopy to check the homogeneity of dispersion of the nanoparticles in the membrane. The efficiency of the zinc nanoparticles and the zinc nanoparticle loaded cellulose acetate membranes for adsorption of organic herbicides such as metolachlor and acetochlor was studied. The particles themselves and the particle-embedded membranes proved to be very adsorbent due to their unique structural properties, providing large surface area and high adsorption capacity. The incorporation of zinc nanoparticles into the membrane decreased the time needed for the removal of the selected herbicides relative to that of the nanoparticles alone, and provides a promising method for water purification.

The application of nanomaterials (NMs) in agriculture has become a global concern in recent years. However, studies on their effects on plants are still limited. Here, we conducted a seed germination experiment for 5 days and a hydroponics experiment for 14 days to study the effects of silicon dioxide NMs(nSiO2) and silicon carbide NMs(nSiC) (0,10, 50, 200 mg/L) on rice (Oryza sativa L.). Bulk SiO2 (bSiO2) and sodium silicate (Na2SiO3) were used as controls. The results showed that nSiO2 and nSiC increased the shoot length (11–37%, 6–25%) and root length (17–87%, 59–207%) of germinating seeds, respectively, compared with the control. Similarly, inter-root exposure to nSiO2, bSiO2, and nSiC improved the activity of aboveground catalase (10–55%, 31–34%, and 13–51%) and increased the content of trace elements magnesium, copper, and zinc, thus promoting the photosynthesis of rice. However, Na2SiO3 at a concentration of 200 mg/L reduced the aboveground and root biomass of rice by 27–51% and 4–17%, respectively. This may be because excess silicon not only inhibited the activity of root antioxidant enzymes but also disrupted the balance of mineral elements. This finding provides a new basis for the effect of silica-based NMs promotion on seed germination and rice growth.

A freely available “in vitro dosimetry” web application is presented enabling users to predict the concentration of nanomaterials reaching the cell surface, and therefore available for attachment and internalization, from initial dispersion concentrations. The web application is based on the distorted grid (DG) model for the dispersion of engineered nanoparticles (NPs) in culture medium used for in vitro cellular experiments, in accordance with previously published protocols for cellular dosimetry determination. A series of in vitro experiments for six different NPs, with Ag and Au cores, are performed to demonstrate the convenience of the web application for calculation of exposure concentrations of NPs. Our results show that the exposure concentrations at the cell surface can be more than 30 times higher compared to the nominal or dispersed concentrations, depending on the NPs’ properties and their behavior in the cell culture medium. Therefore, the importance of calculating the exposure concentration at the bottom of the cell culture wells used for in vitro arrays, i.e., the particle concentration at the cell surface, is clearly presented, and the tool introduced here allows users easy access to such calculations. Widespread application of this web tool will increase the reliability of subsequent toxicity data, allowing improved correlation of the real exposure concentration with the observed toxicity, enabling the hazard potentials of different NPs to be compared on a more robust basis.

The removal of pesticide pollution is imperative, because of their high environmental load and persistence, and their potential for bioaccumulation in, and toxicity to the environment. Most pesticides are found to be toxic even at trace levels. AgNPs can be effectively used for the adsorption of pesticides, and the incorporation of the AgNPs onto a support polymeric membrane enhances their effectiveness and reduces the potential unwanted consequences of intentionally adding free nanoparticles to the environment. Here, silver nanoparticles (AgNPs) were synthesized using a reliable, eco-friendly, and one-step “green” method, by reacting Mentha Piperita (mint) extract with AgNO3 aqueous solution at 60 °C in a microwave. The resulting high surface area nanoparticles are both economic and effective environmental remediation agents, playing a promising role in the elimination of aquatic pesticide pollution. Embedding the nanoparticles into a cellulose membrane at a low concentration (0.1 g) of AgNPs was shown to result in effectively adsorption of representative pesticides (Cypermethrin, Paraquat, and Cartap) within 60 min, while increasing the concentration of nanoparticles incorporated into the membrane further enhanced the removal of the exemplar pesticides from water. The high adsorption capacity makes the cellulose-AgNPs membrane an excellent substrate for the remediation of pesticide-polluted water.

Current understanding of nano- and microplastic movement, propagation and potential effects on biota in freshwater environments is developing rapidly. Still, there are significant disconnects in the integration of knowledge derived from laboratory and field studies. This review synthesises the current understanding of nano- and microplastic impacts on freshwater biota from field studies and combines it with the more mechanistic insights derived from laboratory studies. Several discrepancies between the field and laboratory studies, impacting progress in process understanding, were identified including that the most prevalent plastic morphologies found in the field (fibres) are not those used in most of the laboratory studies (particles). Solutions to overcome these disparities are proposed to aid comparability of future studies. For example, environmental sampling and separation of biota into its constituents is encouraged when conducting field studies to map microplastic uptake preferences. In laboratory studies, recommendations include performing toxicity studies to systematically test possible factors affecting toxicity of nano- and microplastics, including morphology, chemical makeup (e.g., additives) and effects of plastic size. Consideration should be given to environmentally relevant exposure factors in laboratory studies, such as realistic exposure medium and effects of plastic ageing. Furthermore, based on this comprehensive review recommendations of principal toxicity endpoints for each of the main trophic levels (microbes, primary producers, primary consumers and secondary consumers) that should be reported to make toxicity studies more comparable in the future are given.

Despite their ubiquity in personal care products, the health implications of titanium dioxide (TiO2) nanomaterials (NMs) are under strenuous investigation for their potential as a carcinogen, whereas other evidence has shown links with premature ageing. Both potential hazards are manifested after chronic exposure. To explore the chronic effects of TiO2 NMs in the environment, a multigenerational study using the model test species Daphnia magna is conducted. Phenotypical characteristics associated with ageing are observed (loss or shortening of tails and lipid accumulation) with increased expression of highly conserved key stress response genes involved in inflammatory responses and oxidative stress. These responses are visible in continuously exposed daphnids over four generations and in daphnids removed from maternal exposure even three generations later. However, exposure to the “aged” variants of these NMs at the same concentrations significantly reduced these effects, and exposure in medium containing natural organic matter is less severe than in salt‐only medium.

Abstract Background Particles and fibres affect human health as a function of their properties such as chemical composition, size and shape but also depending on complex interactions in an organism that occur at various levels between particle uptake and target organ responses. While particulate pollution is one of the leading contributors to the global burden of disease, particles are also increasingly used for medical purposes. Over the past decades we have gained considerable experience in how particle properties and particle-bio interactions are linked to human health. This insight is useful for improved risk management in the case of unwanted health effects but also for developing novel medical therapies. The concepts that help us better understand particles’ and fibres’ risks include the fate of particles in the body; exposure, dosimetry and dose-metrics and the 5 Bs: bioavailability, biopersistence, bioprocessing, biomodification and bioclearance of (nano)particles. This includes the role of the biomolecule corona, immunity and systemic responses, non-specific effects in the lungs and other body parts, particle effects and the developing body, and the link from the natural environment to human health. The importance of these different concepts for the human health risk depends not only on the properties of the particles and fibres, but is also strongly influenced by production, use and disposal scenarios. Conclusions Lessons learned from the past can prove helpful for the future of the field, notably for understanding novel particles and fibres and for defining appropriate risk management and governance approaches.

Effective microplastic extraction from sediment and soil samples requires a density separation step, with the ability to remove >80 % of plastic particles without introducing substantial contamination. Additional benefits such as affordability and simplicity allow microplastic campaigns on limited budgets the ability to achieve high extraction efficacies. Coppock et al. (2017) designed the Sediment Microplastic Isolation (SMI) unit with these criteria in mind, warning that long-term use may lead to polyvinyl chloride (PVC) contamination. As part of the method validation work for a large-scale international project, collecting samples from more than 100 rivers globally, a pilot study of extraction efficiency and contamination potential of an SMI unit was performed. PVC contamination occurred during the extraction of 20 samples, with indicative grey shavings found in both negative controls and field samples. The original protocol was modified and artificially spiked sediments (positive blanks) were run to test extraction efficacy. The modification, requiring the PVC ball valve to remain open throughout the extraction. This modification eliminated contamination caused by wear and tear of the ball valve, while still maintaining recovery rates >80 %.Three points describing the change not the original: • The PVC ball valve is open while sample is agitated with a magnetic stirrer. • The PVC ball valve remains open while the solution is decanted. • The upper chamber is unscrewed and rinsed; recovering particles attached to the inner walls that would be lost using other filtration approaches. Toxicity of ZnO nanoparticles (NPs) are often related to the release of Zn2+ ions due to their dissolution. Studies also suggest that the toxicity of ZnO NPs cannot be solely explained by the release of Zn2+ ions; however, there is a lack of direct evidence of ZnO particulate effects. This study compared the acute toxicity of ZnO NPs and ZnSO4 following intranasal exposure using a combination of metallomics and metabolomics approaches. Significant accumulation of Zn in the liver was only found in the ZnO NP treatment, with 29% of the newly accumulated Zn in the form of ZnO as revealed by X-ray fine structure spectroscopy (XAFS). This is the first direct evidence suggesting the persistence of ZnO NPs in liver upon intranasal exposure. Although both ZnO NPs and ZnSO4 altered the metabolite profiles, with some overlaps and considerable specificity, of both liver and plasma samples, more and distinct metabolites in the liver and opposite effects in the plasma were altered by ZnO NPs compared with ZnSO4, consistent with no accumulation of Zn detected in liver from ZnSO4. Specifically, a large number of antioxidant-related compounds and energetic substrates were exclusively elevated in the liver of ZnO NP-treated animals. These findings provided direct evidence that persistence of ZnO NPs induced particle-specific effects on the antioxidant systems and energy metabolism pathways. Evaluation of the uptake pathways in cells during exposure to nanoparticles (NPs) is key for risk assessment and the development of safer nanomaterials, as the internalisation and fate of NPs is linked to their toxicity and mode of action. Here, we determined the uptake mechanisms activated during the internalisation of 10, 30, and 100 nm AgNPs by embryonic zebrafish cells (ZF4). The uptake results demonstrated an NP size- and time-dependent uptake, showing the highest total silver uptake for the smallest AgNP (10 nm) at the lowest exposure concentration (2.5 μg/mL) after 2 h, while after 24 h, the highest exposure concentration (10 μg/mL) of the 10 nm AgNPs revealed the highest cellular load at 8 pg/cell. Inhibition of the caveolae, clathrin, and macropinocytosis endocytic pathways by pharmaceutical inhibitors (genistein, chlorpromazine, and wortmannin respectively) revealed that uptake was mainly via macropinocytosis for the 10 nm AgNPs and via the caveolae-mediated pathway for the 30 and 100 nm AgNPs. The induction of autophagy was also strongly related to the NP size, showing the highest percentage of induction for the 10 nm (around 3%) compared to naive cells, suggesting that autophagy can be activated along with endocytosis to deal with exposure to NPs. TEM imaging revealed the distribution of NPs across the cytoplasm inside intracellular vesicles. An increase in Early Endosome formation (EE) was observed for the 30 and 100 nm sizes, whereas the 10 nm AgNPs disrupted the activity of EE. The data supports the establishment of adverse outcome pathways by increasing knowledge on the link between a molecular initiating event such as receptor-mediated endocytosis and an adverse outcome, as well as supporting the reduction of animal testing by using alternative testing models, such as fish cell lines.

ACEnano is an EU-funded project which aims at developing, optimising and validating methods for the detection and characterisation of nanomaterials (NMs) in increasingly complex matrices to improve confidence in the results and support their use in regulation. Within this project, several interlaboratory comparisons (ILCs) for the determination of particle size and concentration have been organised to benchmark existing analytical methods. In this paper the results of a number of these ILCs for the characterisation of NMs are presented and discussed. The results of the analyses of pristine well-defined particles such as 60 nm Au NMs in a simple aqueous suspension showed that laboratories are well capable of determining the sizes of these particles. The analysis of particles in complex matrices or formulations such as consumer products resulted in larger variations in particle sizes within technologies and clear differences in capability between techniques. Sunscreen lotion sample analysis by laboratories using spICP-MS and TEM/SEM identified and confirmed the TiO2 particles as being nanoscale and compliant with the EU definition of an NM for regulatory purposes. In a toothpaste sample orthogonal results by PTA, spICP-MS and TEM/SEM agreed and stated the TiO2 particles as not fitting the EU definition of an NM. In general, from the results of these ILCs we conclude that laboratories are well capable of determining particle sizes of NM, even in fairly complex formulations.
