Background The Leishmaniases are on the top of the global list of tropical neglected diseases. The number of infected dogs in South America is estimated in millions and correlated to disease cases in humans, especially in Brazil. Equines may get infected too and can play a role in the epidemiological chain. Thus, the aim of the present study was to evaluate risk and protective factors of leishmaniasis in rural areas of the western border region of Rio Grande do Sul state, Brazil by Leishmania spp. protozoa molecular detection and serological evaluation (ELISA) in equine and canine blood samples. This work included nine farms around the city of Uruguaiana. Epidemiologic information regarding farm characteristics and biologic material collection of canine (22) and equine (91), totalizing 113 samples was collected. The polymerase chain reaction (PCR) technique was used to detect Leishmania spp. in biological samples. Variables related to the farm were collected and evaluated through descriptive analysis followed by chi-square test and a logistic regression analysis. Results Nineteen positive samples (19/113 – 16,81%) were detected, being 18 equines and 1 canine, in six of the nine farms included in the study. No animal showed clinical signs of the disease. According to the variables analyzed, when compared each characteristic separately, the presence of abundant vegetation and poor hygiene demonstrated to be risk factors to Leishmania infection in rural areas. The logistic regression showed that excellent general hygiene, proximity to the weir and trimmed grass were protective factors (p=0.038, p=0.001 and p=0.014, respectively). Having excellent hygiene represents a 70% lower chance of getting infected, keeping the grass cut protects the animal by more than 90% and the proximity of the weir represents a protective factor of 96%. Conclusions The presence of Leishmania infection in the western border region of Rio Grande do Sul was 16,81% and it was influenced by farm characteristics. The role of the excellent general hygiene as a protective factor is extremely relevant in the leishmaniases prevention.

Visceral leishmaniasis (VL) is a zoonosis caused by the protozoan of the genus Leishmania. The disease is transmitted by the bite of a sand fly vector. Although the main reservoirs are dogs, other hosts can be infected and may play this role. Rio Grande do Sul western region, located on the triple border of Brazil-Uruguay-Argentina, represents a VL transmission area. The goal of the present study was to identify Leishmania spp. infection in animals from rural areas of Uruguaiana, Rio Grande do Sul. Nine farms in the Uruguaiana municipality, Rio Grande do Sul state, were included. Peripheral blood samples were collected from 113 animals (canine [n=22], equine [n=91]) for detection of Leishmania spp. DNA was isolated and polymerase chain reaction was performed. Eight (7%) animals with Leishmania spp. infection were detected on two farms in the same geographical area, seven of which were horses and one was canine, all of which were asymptomatic. To investigate the species of Leishmania, one of the positive equine samples was subjected to direct sequencing, which confirmed the presence of L. infantum genetic material. Results of this study confirm the presence of L. infantum-infected animals in rural areas of Uruguaiana, and provide evidence supporting further investigation of risk factors for dissemination in such areas.

Visceral leishmaniasis is an endemic zoonotic disease identified especially in developing territories. Brazil's northeast, southeast and midwest have been endemic for several years; currently, the infection is spreading to the south. Dogs are the main reservoirs; however, other mammal species have also been infected. Herein, we have identified the infecting Leishmania species in dogs and horses from the south of Brazil, a new outbreak of the infection. Blood samples were collected in the urban area of Uruguaiana city. DNA was extracted from peripheral blood, kinetoplast DNA (kDNA) and ribosomal DNA (rDNA) fragments were obtained by polymerase chain reaction (PCR) and sequenced. Out of 123 samples, 25 of them (14 dogs and 11 horses) were positive for Leishmania spp. Sequence alignment and phylogenetic analysis revealed that the kDNA in positive samples was similar to four species previously reported: L. infantum/L. chagasi, L. donovani, L. major. Despite kDNA minicircles regions are very useful due to high sensitivity to Leishmania spp. DNA detection, the sequence polymorphism among minicircles can be an obstacle to interspecific differentiation. Our results suggest that these strains are circulating in Brazil south region cross-border and indicate the susceptibility of new outbreak for visceral leishmaniasis infection in horses domiciled in endemic region for canine and human visceral leishmaniasis.

Background Leishmaniosis, zoonosis that produces significant public health impacts, is caused by Leishmania infantum. Canines are the main domestic reservoir and, besides humans, other species of mammals could be infected when living in endemic areas. In this study, we detected equine Leishmania infantum infections in a canine visceral leishmaniosis transmission area and evaluated the clinical, haematological, biochemical and oxidative stress disorders. This study was conducted in Uruguaiana, Rio Grande do Sul, south of Brazil. Peripheral blood samples were collected from 124 animals (98 horses and 26 dogs) of both genders and several breeds after they underwent general and dermatologic examinations. Results Twenty five Leishmania infantum infected animals (20.16%), 14 horses and 11 dogs were detected by PCR (Polymerase Chain Reaction) amplification of kinetoplast DNA regions with 96% homology to Leishmania infantum (GenBank Accession No. L 19877.1). The clinical and haematological alterations of infected equines were skin lesions, nodules, lymphadenopathy, decreased levels in red blood cells and haematocrit (p p p p p p > 0.05) in both species. Conclusions To our knowledge, this has been the first leishmaniosis equine survey performed in south of Brazil, caused by Leishmania infantum that were able to initially identify haematological and biochemical changes in the species, even in asymptomatic animals. We present evidence supporting those findings of haematological and biochemical changes could be related to infection. Surprisingly, the clinical manifestations of equine infection were similar to those found in canine visceral leishmaniosis. The equine population could be play an important role in the cycle of leishmaniosis in south Brazil and consequently indicates a great risk of public health. This evaluation of infected animals is important to establish the clinical and laboratory parameters involved in the disease progression.

O objetivo desse estudo foi realizar uma análise do manejo da ordenha realizada em propriedades rurais que utilizem o leite para subsistência, buscando evidenciar fatores de risco e, com isso, contribuir para a melhoria dessa prática e da qualidade do leite consumido em propriedades rurais localizadas no município de Uruguaiana, Rio Grande do Sul. Para a condução do estudo, foram selecionadas 10 fazendas em que são criados bovinos de corte e ovinos no sistema extensivo e que tenham vacas de raças leiteiras ou mestiças destinadas a ordenha para obtenção do leite para o consumo das pessoas da estância. Após a seleção foi entregue a cada proprietário, um questionário guia semiestruturado para serem obtidas informações referentes ao manejo durante a ordenha e ao leite obtido. Ao final da entrevista, cada resposta de cada propriedade foi considerada uma variável. Os dados obtidos foram tabulados em Excel, estudados descritivamente quanto aos percentuais de ocorrência e analisados com o auxílio do IBM SPSS Statistics 20.0 software. A partir das análises constatou-se que a ordenha manual é realizada na maior parte das propriedades e em local não apropriado para esta finalidade. Em relação aos cuidados com a higiene do leite e da sanidade das vacas verificou-se que estão aquém do desejado em todas as propriedades. Dessa forma, a partir desse conhecimento se torna possível a realização de ações objetivando melhorar a qualidade do leite, visando preservar a saúde das pessoas que consomem este produto, bem como seus subprodutos.

The present study aimed to evaluate factors associated with canine visceral leishmaniasis (CVL) in areas with higher seroprevalence of it in Uruguaiana city, Rio Grande do Sul state, Brazil, during the year of 2012. A semi-structured questionnaire with socioeconomic and environmental questions was applied in Leishmania endemic regions of the city. The survey data were analyzed by multivariate statistics and the associations between them were presented as odds ratio within a 95% confidence limit. The relationships between the studied variables showed no statistically significant difference between the dwellings with positive and negative dogs. However, when analyzing the odds ratio, the presence of green areas larger than 10m² turned the house into an area of potential risk for CVL (OR= 2.53). There was no difference among the socioeconomic variables education and income, though the groups with lower education and income showed a higher seroprevalence of CVL. The city of Uruguaiana is already located within the CVL broadcasting area and is taken as an endemic region of the disease, with seropositive dogs in all neighborhoods. Thus, the determinants for an increased Leishmania sp. infection could not be indicated, even so, the maintenance of green areas around the households represented a risk factor for being a potential vector shelter. O objetivo da presente pesquisa foi avaliar potenciais fatores de risco associados à ocorrência de leishmaniose visceral canina (LVC) em áreas de maior prevalência no município de Uruguaiana/RS, no ano de 2012. Foram aplicados questionários semiestruturados com questões socioeconômicas e ambientais em bairros endêmicos para LVC no município de Uruguaiana. Os dados foram analisados por meio de análise multivariada e as associações entre as variáveis foram expressas em odds ratio com intervalos de confiança de 95%. As relações entre as variáveis estudadas não apresentaram diferença estatística significativa quando se comparou residências com cães positivos àquelas com cães negativos. Entretanto, a presença de área verde maior que 10m² apresentou-se como um potencial fator de risco (OR = 2,53) para LVC. Não houve diferença significativa entre as variáveis socioeconômicas, escolaridade e renda embora, nos grupos de menor escolaridade e de menor renda familiar, tenha havido maior soro prevalência para LVC. O município de Uruguaiana-RS já pertence à área de transmissão de LVC tornando-se uma região endêmica da doença, com presença de cães soropositivos em todos os bairros. Desta forma, não foi possível apontar os determinantes para o aumento da infecção por Leishmania sp, embora a manutenção de áreas verdes peridomiciliares tenha representado um fator de risco devido à conservação do vetor nesses locais.

In the New World, especially in Brazil, visceral leishmaniasis (VL) is caused by Leishmania infantum. The pathogen can infect several animal species including dogs, foxes, rodents, primates, felines, equines and humans. Dogs act as the primary domestic reservoirs. This study aimed to use polymerase chain reaction (PCR) for detecting Leishmania infection in horses living in a canine visceral leishmaniasis (CVL) endemic region. DNA samples from horse peripheral blood were used to perform PCR. Templates were amplified using primers for the kinetoplast DNA (kDNA) minicircles, which were able to detect different species of Leishmania. In addition, primers for internal transcribed spacer (ITS) of ribosomal DNA were used for detection of Trypanosomatidae sp. Amongst the 75 (39%) positive PCR samples from total 192 samples, 21 samples were positive for kDNA and 63 samples were positive for either ITS, ITS1, or ITS2 gene markers. The kDNA PCR and sequencing allowed the detection of L. infantum in horse blood samples. To our knowledge, this is the first report of equine infection with L. infantum in Southern Brazil. These results proved that L. infantum could also infect horses in addition to humans and dogs, as well as in European countries. This conclusion emphasizes the urgent need to follow up investigation of the infection in these animals. The aim of this study was to verify the feasibility of synovial fluid (SF) analysis for the parasitological diagnosis of leishmaniasis in naturally infected dogs. Aseptic arthrocentesis of the tarsal and carpal joints of 40 infected dogs (158 Leishmania sp samples) was conducted after euthanasia. The SF smears were evaluated under optic microscopy and amastigotes were detected in the SF of 35 dogs (87.5%). Of the 93 positive samples, 53% were obtained from the tarsal joints, and 47% were obtained from the carpal joints. Amastigote forms were confirmed in one (17%), two (26%), three (31%) or all four investigated joints (26%). This research highlights the feasibility and high sensitivity of SF evaluation for the parasitological diagnosis of leishmaniasis in symptomatic dogs.

The negative aspects of traditional pest control have led to the investigation of alternative methods such as biological control. Metarhizium anisopliae , well known as an entomopathogenic fungus capable of actively invading and killing its hosts and thus a candidate biopesticide, is here tested against two agricultural pests of economic/social importance and also evaluated for its chitinolytic secretion and capacity to grow and sporulate at different temperatures. None of the isolates was able to grow below 4 °C or above 37 °C. Chitinolytic activity under artificial growth conditions revealed that Rhipicephalus ( Boophilus ) microplus cuticle induces N -acetyl-β- d -glucosaminidase and endochitinase activities more efficiently than Dysdercus peruvianus cuticle and that glucose did not repress those activities. Bioassays were carried out with R. microplus females and fourth instar D. peruvianus. Six isolates of M. anisopliae were pathogenic to the engorged female cattle ticks. E6, GC47 and CG97 were the most virulent isolates for both arthropod models although differences were seen among them. M. anisopliae strains caused 90–100 % mortality on the fourth post-infection day in R. microplus. D. peruvianus females were more sensitive to fungal infection than males, and the most virulent strains caused 50 % mortality on the third to fourth day post-infection. Our studies suggest that M. anisopliae strain CG47 is a candidate for commercial pesticide formulations due to its capacity to kill both hosts and its ability to sporulate at higher temperatures.

O objetivo do presente estudo foi avaliar a influência do parasitismo intestinal crônico sobre parâmetros hematológicos e de líquido peritoneal por meio da comparação dessas características em equinos naturalmente parasitados e após administração de anti-helmíntico. Utilizaram-se 21 cavalos de tração urbana, entre dois e 19 anos, sem raça definida e com resultado de exame parasitológico superior a 300 ovos por grama de fezes. Foi realizada avaliação física e coleta de fezes, de líquido peritoneal e de sangue em dois momentos do experimento (D0 e D15), sendo efetuado tratamento antiparasitário no D0. No fluido peritoneal foram avaliadas características físicas, bioquímicas, bem como contagem de células nucleadas (CTCN) e diferenciação celular. No sangue foram determinados valores eritrocitários, leucocitários, proteínas plasmáticas totais, glicose e fibrinogênio plasmáticos, além de fosfatase alcalina (FA) sérica. A análise dos parâmetros avaliados não demonstrou diferença significativa entre animais parasitados e após administração de anti-helmíntico, exceto para valores de CTCN, contagem de neutrófilos segmentados e grau de turbidez do líquido peritoneal. As médias se mantiveram dentro dos intervalos de referência, com exceção da CTCN do líquido peritoneal no D0. No líquido peritoneal, houve predomínio de neutrófilos segmentados, seguidos por macrófagos, linfócitos e eosinófilos em ambos os momentos de avaliação. Observaram-se tendência do quadro eritrocitário em manter-se próximo aos limites inferiores e leve leucocitose no D0. A infecção parasitária nos animais estudados foi predominantemente moderada, o que oferece poucos riscos clínicos. Nessas condições, pode-se afirmar que a CTCN, a contagem absoluta de neutrófilos segmentados e o grau de turbidez do líquido peritoneal são influenciados e podem ser considerados ferramentas diagnósticas e prognósticas úteis nas parasitoses intestinais crônicas. The aim of this study was to evaluate the influence of chronic intestinal parasitism on hematological parameters and peritoneal fluid. It was done by comparing these features in horses used for traction naturally parasitized and after the administration of anthelmintic. Twenty-one horses, between two and nineteen years of age, of mixed breed and with results of parasitological examination of more than 300 eggs per gram of feces were studied. Physical assessment and samples of feces were conducted, as well as blood and peritoneal fluid in the two phases of the experiment (D0 and D15). Antiparasitic treatment in D0 has also been done. The peritoneal fluid was evaluated for physical and biochemical features, and also total count of nucleated cells (TCNC) and cell differentiation. The blood was determined for erythrocyte, leukocyte, plasma total protein, glucose and plasma fibrinogen, and alkaline phosphatase (ALP) levels. The analysis of these parameters showed no significant difference between parasitized animals and after administration of anthelmintic except for TCNC values, segmented neutrophil count and degree of turbidity in peritoneal fluid. The averages remained within the reference ranges, except the TCNC in the peritoneal fluid in D0. In the peritoneal fluid there was a predominance of segmented neutrophils, followed by macrophages, lymphocytes and eosinophils in both time points. A trend was observed in erythrocyte frame to keep close to the lower limits and mild leukocytosis in D0. Parasitic infection of the animals studied was predominantly moderate, which offered minimal clinical risks. After that, it can be affirmed that the TCNC, absolute segmented neutrophil count and targeted degree of turbidity in peritoneal fluid are influenced and can be considered useful diagnostic and prognostic tools in chronic intestinal parasitism.

A pitiose, doença granulomatosa de eqüinos causada pelo oomiceto Pythium insidiosum, tem como característica a evolução rápida seguida de morte dos animais. Estas mortes muitas vezes são causadas por diagnósticos errôneos ou demorados quando os doentes já não respondem ao tratamento. Este trabalho teve por objetivo a padronização do ensaio imunoenzimático indireto (ELISA) para diagnóstico sorológico de pitiose em eqüinos e coelhos, visando a diminuição de erros e de tempo necessário para o diagnóstico. Para o desenvolvimento e validação do teste foram utilizadas 72 amostras de soro de eqüinos saudáveis e 44 soros de eqüinos com pitiose confirmada. Os resultados da validação do ELISA para eqüinos foram: sensibilidade 97,72%, especificidade 90,27%, valor preditivo positivo 86%, valor preditivo negativo 98,4% e eficiência de 93,1%. Para coelhos, o teste foi padronizado com 48 amostras de soro de animais saudáveis e 24 amostras de coelhos imunizados com antígenos de P. insidiosum. Os resultados foram: sensibilidade 91,66%, especificidade 95,83%, valor preditivo positivo 91,66%, valor preditivo negativo 95,83% e eficiência de 94,44%. Os resultados deste trabalho demonstram que o ensaio imunoenzimático indireto é um método seguro e eficaz para o diagnóstico sorológico da pitiose. Pythiosis is a granulomatous disease caused by the oomycete Pythium insidiosum that affects humans and animals, especially horses. Deaths are very often the consequence of incorrect or late diagnosis when animals no longer respond to treatment. This study aimed standardization of the ELISA assay for the serodiagnostic of pythiosis in horses and rabbits, in order to minimize errors and delays in the diagnosis of the disease. Sera of 72 healthy and 44 of by pythiosis affected horses were used for development and evaluation of the test. The ELISA for equine diagnostic showed 97.72% sensitivity, 90.27% specificity, 86% positive predictive value, 98.4% negative predictive value, and 93.1% efficiency. The rabbit test was standardized with 48 sera of healthy rabbits and 24 sera of rabbits immunized with P. insidiosum antigens. The results were 91.66 % sensitivity, 95.83% specificity, 91.66% positive predictive value, 95.83% negative predictive value, and 94.44% efficiency. It can be concluded that ELISA is a reliable test for diagnostic and serological monitoring of pythiosis.

Pythium insidiosum is the etiological agent of pythiosis, a chronic granulomatous disease affecting people and several animal species, mainly horses. Immunotherapy has recently arisen as a promising therapeutic alternative for the disease. This article reports a preliminary characterization of the specificity of the humoral response to P. insidiosum antigens. The immunodominant P. insidiosum proteins were identified by western immunoblot using sera from naturally infected horses and cattle, and sera from rabbits immunized with mycelial mass antigen (MMA). Sera from naturally infected horses and from immunized rabbits reacted with five main polypeptides of 33.5-, 38-, 39-, 40-, and 80-kDa, respectively. Sera from horses submitted to immunotherapy with MMA showed the same pattern of reactivity. Sera from two naturally infected calves reacted with four main polypeptides: two of them presented a molecular mass of 39- and 40-kDa and were probably the same as those recognized by horse and rabbit sera, whereas the other two (35- and 70-kDa) were exclusively recognized by cattle serum. The initial characterization of P. insidiosum immunodominant antigens may help in understanding the immune response behind the clinical cure obtained through immunotherapy and in designing more effective immunotherapeutic strategies.

Pythiosis is a granulomatous disease of horses, cattle, dogs, cats and humans identified in tropical and subtropical areas and caused by Pythium insidiosum, a zoosporic fungus. Experimental models of pythiosis in naturally infected species have not yet been reported but, rabbits may be inoculated with zoospores as an experimental model for studying the disease. The present study evaluates the efficacy of three different of immunotherapics in the rabbit model. Approximately 17,500 zoospores of oomycete P. insidiosum(CBS 101555 strain) were inoculated in each animal to generate the disease. Immunotherapics were produced from vortexed or sonicated cultures of the same strain. Four groups of five animals were employed: group 1, placebo; group 2, sonicated immunotherapic; group 3, mixed immunotherapic; and group 4, vortexed immunotherapic. All rabbits were inoculated with viable zoospores one month before administration of the immunotherapics. Eight doses of immunotherapic or placebo were used in each animal with a 14 day interval between injections. Rabbits receiving the vortexed immunotherapic were most effectively protected ( P< 0.05), showing a decrease in the area of coastal nodules due to Pythiosis insidiosum by 71.8% after 26 weeks of evaluation. Moreover, two animals in this group showed complete remission of the infection at the end of the 26 weeks. In contrast to these findings, rabbits given the sonicated immunotherapic did not show any protection and had an increase of 211.8% in the size of lesions. This failure of sonicated immunotherapic may reflect denaturation of protective antigens due to the sonication method. © 2003 Elsevier Science Ltd. All rights reserved.

Entomopathogenic fungi are able to produce several chitinases, which serve a variety of biological functions, such as fungal cell wall organization and the degradation of exogenous chitin for nutrition or insect infection processes. In this study, we analyzed the contribution of the CHIT30 chitinase from Metarhizium anisopliae in morphogenetic development and virulence as a model of chitinase function. The analysis of chi3 gene expression revealed transcript accumulation in response to heat-shock stress conditions as well as cultivation in medium supplemented with chitin. Null chi3 mutants were constructed to determine the biological role of CHIT30. No substantial differences in the secreted chitinase activity could be detected between the wild type and the Δchi3 mutant. However, both endochitinase and exochitinase activities were diminished in the mutant strain following heat-shock treatment, suggesting that CHIT30 is involved in heat-shock adaptation. Mutants lacking CHIT30 chitinase showed reduced virulence against the cotton stainer bug Dysdercus peruvianus, indicating that the CHIT30 chitinase plays a role in the infection process of M. anisopliae.

Current analysis investigated the tickcide effects of the aqueous extract and chloroform fractions of Ruta graveolens L. (rue) on engorged females of Rhipicephalus microplus, as well as their genotoxic and mutagenic effects on human leukocytes. The best tickcide activity (non-dependent dose) and genotoxic / mutagenic effects (dependent-dose) were observed on exposure to chloroform fractions. Results suggest that extract fractions of R. graveolens L are efficient against R. microplus, although the fraction and the tested concentrations show genotoxic and mutagenic potential for human leukocytes.

Chronological histological alterations of Metarhizium anisopliae during interaction with the cattle tick Boophilus microplus were investigated by light and scanning electron microscopy. M. anisopliae invades B. microplus by a process which involves adhesion of conidia to the cuticle, conidia germination, formation of appressoria and penetration through the cuticle. Twenty-four hours post-infection conidia are adhered and germination starts on the surface of the tick. At this time, the conidia differentiate to form appressoria exerting mechanical pressure and trigger hydrolytic enzyme secretion leading to penetration. Massive penetration is observed 72 h post-inoculation, and after 96 h, the hyphae start to emerge from the cuticle surface to form conidia. The intense invasion of adjacent tissues by hyphae was observed by light microscopy, confirming the ability of M. anisopliae to produce significant morphological alterations in the cuticle, and its infective effectiveness in B. microplus.

The purpose of the present study was to compare the susceptibility to four antifungal agents of 69 Cryptococcus neoformans strains isolated from AIDS patients with that of 13 C. neoformans strains isolated from the environment. Based on the NCCLS M27-A methodology the Minimal Inhibitory Concentrations (MICs) obtained for amphotericin B, itraconazole and ketoconazole were very similar for clinical and environmental isolates. Clinical isolates were less susceptible to fluconazole than environmental isolates. The significance of these findings and aspects concerning the importance, role and difficulties of C. neoformans susceptibility testing are also discussed. Comparou-se a suscetibilidade de 69 amostras de C. neoformans isoladas de pacientes com SIDA com 13 amostras de C. neoformans isoladas do meio ambiente, frente a quatro agentes antifúngicos. Com base na metodologia preconizada pelo NCCLS (M27-A) as concentrações inibitórias mínimas (CIMs) obtidas para a anfotericina B, itraconazol e cetoconazol foram muito semelhantes nos dois grupos estudados. This article reports an outbreak of intoxication of female horses with Claviceps purpurea in southern Brazil. The outbreak affected twelve pregnant mares which were fed with black oat (Avena strigosa) during the pre-delivery period. Underdevelopment of the mammary gland in the pre-delivery period resulting in post-delivery agalactia was the most pronounced finding. These mares delivered weak and unviable foals, which showed no suckling reflex and died within a few hours of birth. Laboratory analysis of oat samples fed to the animals resulted in the identification of Claviceps purpurea sclerotia. The fungus was identified in 0.22% of the examined seeds.
