Biofilm formation is a multifactorial process and often a multi-species endeavour that involves complex signalling networks, chemical gradients, bacterial adhesion, and production or acquisition of matrix components. Antibiotics remain the main choice when treating bacterial biofilm-associated infections despite their intrinsic tolerance to antimicrobials, and propensity for acquisition and rapid dissemination of antimicrobial resistance within the biofilm. Eliminating hard to treat biofilm-associated infections that are antibiotic resistant will demand a holistic and multi-faceted approach, targeting multiple stages of biofilm formation, many of which are already in development. This mini review will highlight the current approaches that are employed to treat bacterial biofilm infections and discuss new approaches in development that have promise to reach clinical practice.

Membrane vesicles (MVs) contribute to various biological processes in bacteria, including virulence factor delivery, host immune evasion, and cross-species communication. MVs are frequently being discharged from the surface of both Gram-negative and Gram-positive bacteria during growth. In some Gram-positive bacteria, genes affecting MV biogenesis have been identified, but the mechanism of MV formation is unknown. InEnterococcus faecalis, a causative agent of life-threatening bacteraemia and endocarditis, neither mechanisms of MV formation nor their role in virulence has been examined. Since MVs of many bacterial species are implicated in host-pathogen interactions, biofilm formation, horizontal gene transfer, and virulence factor secretion in other species, we sought to identify, describe, and functionally characterize MVs fromE. faecalis. Here we show thatE. faecalisreleases MVs that possess unique lipid and protein profiles, distinct from the intact cell membrane, and are enriched in lipoproteins. MVs ofE. faecalisare specifically enriched in unsaturated lipids that might provide membrane flexibility to enable MV formation, providing the first insights into the mechanism of MV formation in this Gram-positive organism.

Genetic tools to dissect complex interactions in mixed microbial communities are largely limited to transposon mutagenesis and traditional time- and labor-intensive allelic-exchange methods. Built upon streptococcal dCas9, we developed an easily modifiable, inducible CRISPRi system for E. faecalis that can efficiently silence single and multiple genes. This system can silence genes involved in biofilm formation and antibiotic resistance and can be used to interrogate gene essentiality. Uniquely, this tool is optimized to study genes important for biofilm initiation, maturation, and maintenance and can be used to perturb preformed biofilms. This system can be applied to study essential genes, genes involved in antimicrobial resistance, and genes involved in biofilm formation and persistence. The system is robust and can be scaled up for high-throughput screens or combinatorial targeting. This tool substantially enhances our ability to study enterococcal biology and pathogenesis, host-bacterium interactions, and interspecies communication.

Most bacteria express multiple adhesins that contribute to surface attachment and colonization. However, the network and relationships between the various adhesins of a single bacterial species are less well understood. Here, we examined two well-characterized adhesins in Enterococcus faecalis, aggregation substance and endocarditis- and biofilm-associated pili, and found that they exhibit distinct functional contributions depending on the growth stage of the bacterial community. Pili interfere with aggregation substance-mediated clumping and plasmid transfer under planktonic conditions, whereas the two adhesins structurally complement one another during biofilm development. This study advances our understanding of how E. faecalis, a ubiquitous member of the human gut microbiome and an opportunistic pathogen, uses multiple surface structures to evolve and thrive., Like many bacteria, Enterococcus faecalis encodes a number of adhesins involved in colonization or infection of different niches. Membrane proteins represent a significant proportion of total bacterial proteins and perform vital cellular functions ranging from exchanging metabolites and genetic material, secretion and sorting, sensing signal molecules, and cell division. Many of these functions are carried out at distinct foci on the bacterial membrane, and this subcellular localization can be coordinated by a number of factors, including lipid microdomains, protein-protein interactions, and membrane curvature. Elucidating the mechanisms behind focal protein localization in bacteria informs not only protein structure-function correlation, but also how to disrupt the protein function to limit virulence. Here we review recent advances describing a functional role for subcellular localization of membrane proteins involved in genetic transfer, secretion and sorting, cell division and growth, and signaling.

We demonstrate the development of organic semiconductor and passive material formulations optimized for photolithographic integration using existing photo-resist materials and technology. These materials can be coated and patterned to produce organic thin-film transistors suitable for integration into mass production processes with charge carrier mobility of greater than 2 cm2/Vs.

Dental caries is a multifactorial disease and the quality of oral hygiene has high influence on development of dental caries. Knowing that dental caries is a dynamic and reversible process led to the development of new technologies that can detect caries at its earliest stages (before the formation of the cavity), for its prompt treatment and prevention. 70 years of fluoride usage for prevention dental caries showed their reliability and ability to participate in the remineralization of hard tooth tissues. At the same time efficiency of fluoride is reduced under acidic conditions of dental plaque (in case of a poor oral hygiene). Daily oral hygiene with usage of a paste with 1.5% arginine, 1450 ppm of fluoride and calcium bicarbonate will help to stop the development of dental caries and reduce the risk of the new carious cavities appearance even in conditions of low level of oral hygiene.

We demonstrate the co-development of organic semiconductors, passive materials and formulations with process optimization to enable the manufacture of high performance Organic Thin Film Transistor (OTFT) arrays suitable for mass production of printable display backplanes. These materials can be printed or coated to fabricate top and bottom gate OTFT's with charge carrier mobility greater than amorphous silicon.

Three new diindenodithienothiophene (DITT) based materials were synthesised and their electrochemical properties investigated. The HOMO–LUMO gaps were observed to be 3.33, 3.48 and 2.81 eV, respectively. Cyclic voltammetry results indicate increased stability for the alkylated derivatives. The dioxide exhibits strong photoluminescence, giving a photoluminescence quantum yield of 0.72 in solution and 0.14 in the solid state. Hole mobility measurements were carried out on the non-alkylated derivative and the corresponding values were ∼10−4 cm2 V−1 s−1.

A substantial body of evidence has confirmed human papillomavirus (HPV) infection as an etiologic agent in human cervical cancer. To evaluate the association between HPV and cervical cancer in Chinese women, we examined tumor specimens from women who lived in Shanghai, People's Republic of China. Biopsies from 40 women, diagnosed with either squamous-cell carcinoma (n = 35) or adenocarcinoma (n = 5) were tested for HPV DNA by PCR. The HPV types present in tumors were determined either by hybridization of PCR products with HPV type-specific probes or by PCR-based sequencing. A total of 35 of the 40 cervical cancer specimens (87.5%) contained HPV DNA. The following distribution and types were detected: 7.5% HPV 16, 10% HPV 18, 20% HPVs 16 and 18, 15% HPV 52, 15% HPV 58, 12.5% HPVs 52 and 58 and 7.5% unclassified HPVs. In this population of Chinese women with cervical cancer, HPV 52 and 58 were as prevalent as the "high-risk" (for cervical cancer) viruses HPVs 16 and 18.

To enable the integration of organic transistors into flexible displays we have developed passive materials for a robust, layer-to-layer compatible device stack to be manufactured using a variety of industrially applicable coating and printing processes. The solution processable materials for the OSC protection and the passivation material are designed for good interlayer adhesion and OTFT stability. The 365 nm UV-curable protection layers for the OSC are compatible with fabrication processes commonly used in the display industry, e.g ., sputtering of metals. Fabrication of ink-jet printed devices is demonstrated with usage of a passive material suitable for further pixel integration. The bias stress and ambient stability was shown for the integrated stacks of transistors with the carrier mobility greater than that of amorphous silicon. We further demonstrate the development of formulations, suitable for high throughput roll-to-roll processing of soluble small molecule semiconductors allowing a high degree of structural order in the final solid film. Transistor mobility 2 /V·s is achieved with a range of viscosity values compatible with flexographic printing. Fine pattern of OSC is demonstrated with flexographic printing by adjusting the viscosity of the formulation. Uniform layers are achieved with coating and printing techniques and uniformity values are sufficient for display application.

To determine the malaria prevalence by microscopy, antigen detection and nucleic acid detection in a defined subpopulation in a Plasmodium falciparum-endemic region during the peak transmission season.Blood specimens were collected in a cross-sectional study involving children aged 5-10 years (n = 195) presenting with acute fever to two clinics in Western Kenya. All specimens underwent microscopy, HRP2 and aldolase antigen detection by enzyme immunoassay (EIA), parasite-specific DNA and total nucleic acid (RNA and DNA) by real-time PCR (qPCR) and reverse-transcriptase PCR (qRT-PCR).Microscopy detected 65/195 cases of malaria infection [95% confidence interval (CI) 52-78]. HRP2 and aldolase EIA had similar sensitivity levels detecting antigen in 65/195 (95% CI, 52-78) and 57/195 (95% CI, 45-70) cases. Discordants in antigen detection vs. microscopy occurred at470 parasites/μl and4900 parasites/μl for HRP2 and aldolase, respectively. Detection of total nucleic acid allowed a 3 log lower limit of detection than just DNA detection by real-time PCR in vitro. In clinical specimens, 114/195 (95% CI, 100-127) were qPCR positive (DNA), and 187/195 (95% CI, 179-191) were qRT-PCR positive (DNA plus RNA).The prevalence of submicroscopic malaria infection was significantly higher when detecting total nucleic acid than just DNA in this outpatient population during the high transmission season. Defining standards for submicroscopic infection will be important for control programmes, diagnostics development efforts and molecular epidemiology studies.

The title compound, C(18)H(12)S, comprises five fused rings forming an essentially planar mol-ecule, with a total puckering amplitude (Q) of 0.032 Å and a maximum deviation from the mean plane of 0.014 (4) Å for the C atoms of the methyl-ene groups. A crystallographic mirror plane orthogonal to the mol-ecular plane passes through the S atom and the midpoint of the opposite C-C bond within the central five-membered ring. The mol-ecules lie in layers, forming edge-to-face C-H⋯π inter-actions, with a separation of 2.66 Å between one H atom of the methyl-ene group and the centroid of an adjacent indene ring.
