The ion transport system responsible for intracellular pH (pHi) regulation in squid giant axons was examined in experiments with pH-sensitive microelectrodes and isotopic fluxes of Na+ and Cl-. In one study, axons were acid-loaded and the rate of the subsequent pHi recovery was used to calculate the acid extrusion rate. There was an absolute dependence of acid extrusion on external Na+, external HCO-3 (at constant pH), and internal Cl-. Furthermore, the dependence of the acid extrusion rate on each of these three parameters was described by Michaelis-Menten kinetics. Acid extrusion was stimulated by an acid pHi, required internal ATP, and was blocked by external 4-acetamido-4'-isothiocyanostilbene-2,2'-disulfonate (SITS). Under a standard set of conditions (i.e., [HCO-3]o = 12 mM, pHo = 8.00, [Na+]o = 425 mM, [Cl-]i = 150 mM, [ATP]i = 4 mM, pHi = 6.5, and 16 degrees C), the mean acid extrusion rate was 7.5 pmol X cm-2 X s-1. In a second study under the above standard conditions, the unidirectional Na+ efflux (measured with 22Na) mediated by the pHi-regulating system was found to be approximately 0, whereas the mean influx was about 3.4 pmol X cm-2 X s-1. This net influx required external HCO-3, internal Cl-, and acid pHi, internal ATP, and was blocked by SITS. In the final series of experiments under the above standard conditions, the unidirectional Cl- influx (measured with 36Cl) mediated by the pHi-regulating system was found to be approximately 0, whereas the mean efflux was approximately 3.9 pmol X cm-2 X s-1. This net efflux required external HCO-3, external Na+, an acid pHi, internal ATP, and was blocked by SITS. We conclude that the pHi-regulating system mediates the obligate net influx of HCO-3 (or equivalent species) and Na+ and the net efflux of Cl- in the stoichiometry of 2:1:1. The transport system is stimulated by intracellular acid loads, requires ATP, and is blocked by SITS.

Explant cultures were established from capsule/epithelium preparations from both normal and cataractous lenses to investigate properties of human lens epithelial cells. The cultured cells were found to have similar membrane potentials to whole human lenses and isolated epithelia, and similar free ionic concentrations of potassium, sodium, and calcium (131 mM, 17 mM and 0.8 microM respectively) to whole human lenses. The free ionic concentrations were measured in both cases using neutral resin-filled electrodes. Cellular communication was investigated using electrical (two internal microelectrodes) and dye injection techniques. The electrical resistance of a confluent cell monolayer was approximately 4 M omega when the voltage measuring and current passing microelectrodes were in neighbouring cells or several cell diameters apart. Additionally, Lucifer Yellow dye injected into one cell spread rapidly over a wide area of cells. The cells thus appear to be extremely well coupled. Electrical communication could be disrupted by internal acidification (following exposure to 100% CO2), exposure to 1 mM octanol and by membrane depolarization. In the latter case the blockade was only partial. All uncoupling methods proved to be reversible. The diffusion of Lucifer Yellow dye was also inhibited by internal acidification and exposure to octanol. The sensitivity of both dye and electrical coupling to internal acidification and exposure to octanol is similar to that observed in hepatocytes and other tissues, whereas the effect on cell communication induced by changing the resting potential appears to occur only in a few cell types such as those of embryonic origin.

We report here electrophysiological and ion distribution results obtained with mechanically skinned skeletal muscle fibres which indicate that a Donnan equilibrium, characterized by large ionic concentration and electrical potential differences, is established between the myofibrillar space and the rest of the sarcoplasm surrounding the myofibrils. The more negative electrical potential within the myofibrils (-15 to -29 mV for conditions similar to those in vivo) can strongly influence: (1) intracellular Ca2+ movements associated with contraction; (2) the location of sarcoplasmic enzymes, their substrates and products of reaction; (3) measurements of the electrical potential difference between the extra- and intracellular space and estimation of relative membrane permeabilities for certain ions.

Fluid and enzyme secretion from a number of mammalian exocrine glands is controlled by the action of neurotransmitters and hormones on acinar cell membranes. Sustained stimulation evoking sustained fluid and enzyme secretion also evokes sustained membrane depolarization and increase in conductance. Mouse and rat pancreatic fluid and enzyme secretion, as well as membrane depolarization and conductance increase evoked by sustained stimulation with acetylcholine or cholecystokinin-gastrin peptides, are acutely dependent on extracellular calcium. However, the initial stimulant-evoked conductance increase and secretion appear to be triggered by calcium released from inside the cells. Direct measurement of membrane current during sustained stimulation in voltage-clamp experiments with resolution of the total current into its Na, Cl and K components has allowed calculations of stimulant-evoked Na and Cl uptake into the acinar cells. The NaCl uptake is quantitatively sufficient to account for the stimulant-evoked fluid secretion. The role of the stimulant-evoked transmembrane ionic current appears to be the supply of salt for the fluid secretion. Calcium derived from intracellular sources in the initial phase of secretion, and from the extracellular fluid in the sustained phase, couples fluid and enzyme secretion to hormone-receptor interaction.

The equations that govern the behavior of ion-selective electrodes are evaluated for the composition ranges of Li+, Na+, K+, Mg2+, Ca2+, Cl-, and HCO-3 encountered in living cells. Various cause-effect relations are detailed, especially those pertaining to calibration procedures. The relation of the accuracy of measurement to the expected signal range, particularly in connection with the uncertainty of interpolation, is emphasized. Relevant numerical parameters are given for all ISE's discussed, the selectivity characteristics being those of the most advanced membrane systems. Figures are provided for the calibration curves in the vicinity of the typical cytosol composition.

At confluency, LLC-PK1 monolayers develop a transepithelial electrical resistance of 127 omega X cm2 and a spontaneous electrical potential that very seldom exceeds 1 mV. The monolayer shows a linear current-voltage relation in symmetrical solutions. The total conductance increases linearly with increases in the electrolyte concentration of the bathing solution. These characteristics indicate that the membrane that controls the permeability properties of the monolayer is a single membrane that it does not contain very weakly charged sites and it is sufficiently thick to obey the principle of microscopic electroneutrality. The sodium-to-chloride permeability determined from dilution potentials or from direct measurements of unidirectional Na+ and Cl- flux are 0.30 and 0.38, respectively, almost identical to the ratio obtained in the straight segment of the renal proximal tubule. The steady-state value of the electrical resistance depends on the Ca2+ concentration in the incubation medium with an apparent Km of 0.1 mM. A transitory opening of the occluding junctions results in a more uniform distribution of the Na+-dependent sugar transport system, which is normally confined to the apical membrane of the epithelial cell. This result indicates that the occluding junctions in LLC-PK1 monolayers act as a mechanical barrier, preventing the intermixing of extrinsic as well as intrinsic membrane proteins.

1. Body fluids contain ion pairs such as NaC10, CaCl+, CaHCO+3 and MgSO04. 2. Ion pairs must often be considered in the quantitation of acid-base, solubility, Donnan and other equilibria. 3. Fluxes of ion pairs, such as NaCO-3 and perhaps NaSO-4 and NaCl0, may sometimes contribute significantly to total ion fluxes across cell membranes. 4. Dissociation constants for ion pairs are discussed, but values appropriate to body fluids are often uncertain.

Spinal cord Na, K, Ca, and H2O changes were measured 6 h after graded contusion injuries in 40 Sprague-Dawley rats. A 10 g weight was dropped 1.25 cm (n = 6), 2.5 cm (n = 7), 5.0 cm (n = 6), or 7.5 cm (n = 7) onto the thoracic spinal cord of 26 rats. An additional 10 rats served as laminectomy controls and 4 rats were unoperated controls. At 6 h after surgery or injury, the spinal cords were rapidly cut into 4 mm segments, weighed to obtain tissue wet weights (W), dried for 14-16 h at 97 degrees C in a vacuum oven (30 mmHg), and reweighed for tissue dry weights (D). Water concentrations ([H2O]d) were estimated from (W-D)/D in units of ml/g D. Ionic concentrations ([Na]d, [K]d, and [Ca]d) of the tissue samples were measured by atomic absorption spectroscopy with units of mumol/g D. Ionic shifts (delta [Na]d, delta [K]d, delta [Ca]d) were calculated by subtracting laminectomy control values from those measured in injured cords. Laminectomy alone significantly increased [Na]d and [H2O]d compared to unoperated controls. Mean +/- standard deviations of [H2O]d, [Na]d, [K]d, and [Ca]d were, respectively, 1.95 +/- 0.07, 182.6 +/- 5.9, 277.2 +/- 11.8, and 12.1 +/- 1.4 in unoperated controls; 2.12 +/- 0.08, 238.6 +/- 9.2, 277.8 +/- 9.2, and 11.7 +/- 1.1 in laminectomy controls. At the impact site, [K]d fell by 14-37% and [H2O]d rose by 14-24%, [Na]d by 13-64%, and [Ca]d by 65-137% of laminectomy control values. delta [Na]d, delta [K]d, and delta [Ca]d correlated linearly with impact velocities; [Ca]d increased by 1.0% per cm/sec (r = 0.995, p less than 0.005), [Na]d increased 0.67% per cm/sec (r = 0.950, p less than 0.01), and [K]d decreased 0.34% per cm/sec (r = 0.964, p less than 0.01). Neither delta [H2O] nor delta [Na]d + delta [K]d consistently predicted impact velocity. [Na]d + [K]d correlated with [H2O]d with a slope of 177.4 mumol/ml (r = 0.697, p less than 0.005). Since Na and K constitute greater than 95% of tissue inorganic ions, the slope approximates net ionic shift per ml of water entry or the ionic osmolarity of edema fluid. These results indicate that increasing contusions produce graded ionic shifts and that edema does not predict contusion severity. These data support our hypothesis that net ionic shifts cause edema in injured spinal cords.

The effect of theophylline and a heat-stable enterotoxin of Escherichia coli (ST) on ion transport was examined using an in vitro, short-circuited preparation of the porcine colon. Theophylline abolished net Na absorption and elicited net Cl secretion, which quantitatively accounted for the increase in short-circuit current (Isc) observed. In contrast, a maximal dose of ST elicited an Isc response about one-half that of theophylline and only partially reduced the net absorption of Na and Cl. A significant residual ion flux, consistent with HCO3 secretion, was elicited by ST and was sustained after theophylline addition. Ion replacement experiments showed that the Isc and net ion transport response to ST was abolished when either Cl or HCO3 were removed from the bathing solutions. Voltage clamp experiments to evaluate the contribution of the paracellular and transcellular transepithelial pathways from serosa to mucosa showed that approximately one-half of the total serosa-to-mucosa flux (Jsm) of both Na and Cl was through the cells. Theophylline and ST both significantly reduced transcellular JNasm, but did not affect JClsm. Theophylline, but not ST, caused an increase in paracellular conductance of both ions. These results demonstrate significant differences in the effects of ST or theophylline on both transcellular and paracellular ion movement, and suggest that ST induces a Cl-dependent HCO3 secretion which is unobserved under control or theophylline-stimulated conditions. In addition, results are consistent with the operation of a neutral NaCl secretory process which is normally masked by the greater net rates of the neutral Na and Cl absorptive mechanisms. Thus, both ST and theophylline appear to reduce or abolished the neutral processes and convert the neutral secretory process into an electrogenic one. This latter effect could be explained simply by an increase in the anion conductance of the mucosal membranes.

The need to know the intracellular activity of an ion and how it changes under controlled conditions is as important today as it was 30 years ago. In 1956, one could fabricate only a H+-selective microelectrode and with a tip size not much smaller than 100 micron. Today, one can fabricate microelectrodes selective to H+, Na+, K+, Cl-, HCO3-, Ca2+, or Mg2+ (plus others) and with active tips less than 1 micron. The reduction of active tip size can be attributed mainly to the introduction of liquid ion exchanger (LIX) and neutral carrier ligands. Unfortunately, the LIX microelectrodes, as currently fabricated, do not yet function optimally as reliable and stable electrochemical measuring devices. A durable bond between the active membrane and its insulated container continues to remain the major design problem even after 30 years of development.

There are two groups of mechanisms through which ultrasound can affect biological systems, those of thermal origin and others of nonthermal origin. Since in almost every therapeutic application of ultrasound, movement of ions across cellular membranes is involved, it becomes important to study the effect of ultrasound on active and passive ionic conductance. In order to differentiate between thermal and nonthermal effects, a study was conducted on model systems in which the effect of temperature is known. The well-known sodium transporting epithelium, the epidermis of abdominal frog skin, was investigated and the effect of therapeutic ultrasound on its electrophysiological properties was determined. It was found that under open circuit conditions, irradiation of the skin with 1 MHz cw (60-480 mW/cm2) ultrasound caused a significant decrease (5-50%, depending on the applied power) in the transepithelial potential and resistance at room temperature (20-22 degrees C). Under short circuit conditions, also at room temperature, there was an increase in total ionic conductance (20-250%, depending on the applied power) and a decrease in the net actively transported current, measured as the short circuit current. These effects are reversible within the range of powers used. Furthermore, it was found that the magnitude of the observed changes was strongly dependent on the perfusion rate and the gas content of the bathing medium. The effect of ultrasound diminished in the presence of CO2 and was enhanced with faster perfusion rates. Pulsed ultrasound delivered at the same energy (Isata) as that of cw caused a significantly larger effect. At lower temperatures (12-14 degrees C) the effect of ultrasound was reduced. Analysis of the data reveals that the effects of ultrasound on ion transport reported here are not primarily of thermal origin but are probably due to cavitation and related effects, such as microsteaming.

The relationships among ion current, membrane potential difference, and resistance of an epithelium are studied. The short-circuit technique introduced by Ussing and Zerahn does not completely short circuit the epithelium if the series resistance parallel to the cell layer between the voltage electrodes is not properly compensated. The residual potential difference across the epithelial cell layer in the "short-circuit state" is proportional to both the measured short-circuited small intestinal mucosa the villus and crypt areas are hypo-polarized to different degrees rather than simultaneously hyper- and hypo-polarized. Short-circuiting the whole tissue reduces but does not abolish the passive net ion movement across the tissue. Measurements of the electrical properties of the whole and denuded rat distal small intestine in HCO3-Ringer solution containing 10 mM glucose reveal that the measured short-circuit current has under-estimated approximately 33% of the true short-circuit current and that the passive net Na flux from serosa to mucosa and Cl flux from mucosa to serosa are not negligible in the "short circuit state."

This paper reviews a theory of pattern establishment and pattern restoration by endogenous ionic currents. These currents are supposed to be generated by a certain separation of ion leaks and ion pumps in cell membranes. In so far as these currents act back to further this separation, they would be part of a regenerative process that initially establishes positional values. Later in development, particularly in epimorphic regeneration, when positional values are restored or extended, these currents are supposed to leak through sites of discontinuity in such values and thus trigger growth. This paper also reviews the factual evidence for this view: evidence that developmental currents are, indeed, very widespread; evidence in a few cases, particularly in Cecropia follicles and in wounded cavy skin, that they can generate substantial voltage differences or gradients; evidence that comparable artificial fields can move charged macromolecules along cell membranes and polarize cell growth; and direct evidence in a few case, particularly fucoid eggs, Cecropia follicles and regenerating amphibian limbs, that ion currents do, in fact, act back to direct or further development. The paper also presents a particular theory, based upon ionic currents, of the reversal of thyroid cell polarity by serum.

Many neurones are extremely invaginated and possess branching processes, axons and dendrites. In general, they are surrounded by a restricted diffusion space. Many of these cells exhibit large, slow potential changes during the passage of current across their membranes. Whenever currents cross membranes separating aqueous solutions, differences in transport numbers of the major permeant ions give rise to local concentration changes of these ions adjacent to the membranes, which will result in various electrical and osmotic effects. These transport number effects are expected to be enhanced by the presence of membrane invaginations. Dendrites are equivalent to reversed invaginations and there should be significant changes in concentrations of permeant ions within them. In general, the effects of such changes on the electrical response of a cell will be greater when the concentration of a major permeant ion is low. The effects have been modelled in terms of two nondimensional parameters: the invagination transport number parameter beta and the relative area occupied by the invaginations delta A. If these two parameters are known, the magnitudes and time course of the slow potential changes can immediately be estimated and the time course converted to real time, if the length of the invaginations (l) and ionic diffusion coefficient (D) within them are alos known. Both analytical and numerical solutions have been given and predictions compared. It is shown that in the case of large currents and potentials the analytical solution predictions will underestimate the magnitudes and rates of onset of the voltage responses. The relative magnitude of the transport number effect within the invaginations (or dendrites) and other transport number contributions to slow potential changes have also been assessed and order-of-magnitude values of these are estimated for some biological data.

Proteins labeled with [35S]methionine or [3H]leucine were generated in vitro in bullfrog dorsal root ganglia and their fast axonal transport in the spinal nerves was followed during a subsequent incubation period. Incubation of the ganglia in a medium where sucrose, choline chloride, or sodium isethionate replaced NaCl caused respectively an 88, a 37, or a 76% reduction in the quantity of proteins carried by the fast axonal transport system; no decrease in synthesis of labeled proteins was observed and protein transport followed the usual time course. Incubation of desheathed spinal nerves in a medium where sucrose replaced NaCl reduced by 67% the quantity of labeled proteins which were transported past the desheathed region. Although both the axons and the dorsal root ganglia exhibit the requirement for monovalent ions to maintain fast axonal transport, the possibility that the ionic requirements of the ganglia pertain to the somal portion of the nerve cell is discussed.

A series of studies on the signal responsible for initiating the tubuloglomerular feedback (TGF)-mediated changes in SNGFR have utilized a retrograde perfusion technique that permits close control of macula densa segment tubular fluid composition. The feedback response has been demonstrated to be an approximately linear function of luminal NaCl concentration between 15 and 60 mM, with increasing NaCl concentration resulting in lower nephron filtration rate. Feedback responses could be initiated by NaCl and NaBr solutions but not by NaI, NaNO3, NaF, NaHCO3, NaSCN, or Na acetate. All chloride salts tested, in which chloride was accompanied by a univalent cation, elicited a response. The feedback mechanism thus shows a special requirement for chloride or bromide and not for sodium. With the chloride salts the response varied in magnitude depending on the accompanying cation with the following order of decreasing response, Rb+, Na+, Cs+, NH4+, K+, choline+, and Li+, CaCl2 and MgCl2 did not elicit a feedback response. The feedback response was not influenced by large changes in luminal osmolarity by addition of urea, suggesting either that the feedback sensing site is water impermeable or that water movement does not influence the signal. When electrolyte-free solutions are perfused through the loop of Henle in an orthograde direction, sufficient electrolyte may enter the luminal fluid to transiently influence the feedback mechanism.

Iodinated radiologic contrast agents should, ideally, passively provide radiographic contrast in blood vessels and soft tissues but have no effects on physiology, hematology or biochemistry. In practice, no such agent exists although the nonionics most closely approach the ideal. All agents have a bewildering variety of effects on the cardiovascular system, which, in turn, engender a variety of neurogenic and humoral responses. The mechanisms underpinning these many phenomena appear to involve hyperosmolality, molecular structure, ionic content and balance. This review presents the current state of knowledge of these effects and their mechanisms, with emphasis on the differences between conventional ionic and more inert nonionic agents.

The ionic currents flowing in developing organisms produce weak magnetic fields that can be detected using SQUID magnetometers. The method is non-invasive and dc recording is possible. To date SQUID magnetometers have mainly been used in human studies. The features of the technique are described and the prospects of extending its use to developmental studies are discussed. Feasible instrumental specifications are indicated. A recent SQUID magnetometer investigation of ionic current flow in the developing chick in ovo is summarised as an illustration of the magnetometer method. The paper as a whole argues that magnetometry is a useful alternative or adjunct to electrode-based experiments on the electrophysiology of developing organisms.

Renal epithelial cells were isolated from the outermost superficial cortex of adult and young rats. The cells, likely of proximal origin, were plated on silicon pieces, and cultured during 1-3 days. Intracellular content and concentrations of K, Na, Cl, and P, and the kinetics of change in intracellular content, after inhibition of Na-K ATPase by incubation with ouabain or in K-free medium, were measured in individual cells in small populations using electron probe analysis. In control medium, concentrations in mM were approximately: K, 130; Na, 15; Cl, 28; P, 140. After 6 h inhibition of Na-K ATPase, cells exchanged all K for Na, and the intracellular Na concentration increased to 139 mM in K-free medium. The Cl concentration increased at most to 46 mM. The sum of intracellular K + Na + Cl did not increase more than 25% after 24 h incubation in K-free medium. There were no differences in intracellular K, Na, and Cl for adult and young rat cells in similar conditions. The half-times of K efflux and Na influx after inhibition of Na-K ATPase measured in adult rat were approximately 16-20 min. In the absence of serum, in K-free medium, the half-times of K efflux and Na influx in young rat cells were approximately 30 min, significantly higher than the half-time in the presence of serum, and with ouabain, being approximately 13 min. Histograms of distributions of K and Na content showed that the cells behaved as a single functional population. Ouabain Ki was estimated to be 10(-4) M. After 24 h preincubation in K-free medium, when returned in 5 mM K-containing medium, adult rat cells recovered rapidly normal intracellular K and Na concentrations. Using this approach, expression of the kinetics ionic transport properties of renal epithelial cells during development, and the hormonal influences on terminal differentiation may be studied.

Astrocytes comprise about 25% of the cellular volume of the brain, and their main function is to maintain homeostasis of the neuronal environment. These cells are commonly identified on the basis of their membrane electrical properties and the presence of specific proteins. We have characterized the human astrocytoma cell line designated UC-11MG and have shown these cells have many of the traits of differentiated astrocytes. Many of the UC-11MG cells have a large resting membrane potential, averaging -74 mV. The slope of the Em vs log [K]o cuve was 58.5 mV per decade [K]o. The cells were inexcitable when exposed to brief depolarizing current pulses. The astrocytoma traits are virtually identical to those previously reported for normal astrocytes. The astrocytoma cells also express glutamine synthetase activity which is considered specific to astrocytes among brain cells. Previous work had also demonstrated the presence of other astrocyte markers glial fibrillary acidic protein and S-100 protein in the UC-11MG cells. The steady-state ion transport properties of Na+, Cl-, and K+ were also characterized in these cells, and the rates of efflux were found to be similar to those in other astrocytes, with the major difference being the presence of a second kinetic compartment in the UC-11MG cells. From this work, we conclude that the UC-11MG cell line displays prominent features associated with differentiated astrocytes, and may provide an excellent model system for the study of human astrocytes.

Striking convergent evolution is found in the properties of the organic osmotic solute (osmolyte) systems observed in bacteria, plants, and animals. Polyhydric alcohols, free amino acids and their derivatives, and combinations of urea and methylamines are the three types of osmolyte systems found in all water-stressed organisms except the halobacteria. The selective advantages of the organic osmolyte systems are, first, a compatibility with macromolecular structure and function at high or variable (or both) osmolyte concentrations, and, second, greatly reduced needs for modifying proteins to function in concentrated intracellular solutions. Osmolyte compatibility is proposed to result from the absence of osmolyte interactions with substrates and cofactors, and the nonperturbing or favorable effects of osmolytes on macromolecular-solvent interactions.

Patterns of ionic currents accompanying shape and surface changes during growth of cell clusters from carrot suspension cultures were examined using a vibrating probe. Electrical polarity was established in clusters undergoing apparently disorganized proliferation in the presence of 2,4-dichlorophenoxyacetic acid (2,4-D). This electrical polarity is similar to that found in organized somatic embryos which form upon removal of 2,4-D, and implies that the proliferating clusters are actually suppressed embryos. This implication is further supported by observations using scanning electron microscopy. We conclude that the potential to undergo embryogenesis is present even before the 2,4-D is removed, but cannot be realized in the presence of the auxin.

Requirements for a reliable use of liquid membrane microelectrodes are discussed in terms of stability, response time, and lifetime on the basis of membrane technological considerations. The selectivity of H+, Li+, Na+, K+, Mg2+, Ca2+, and Cl- microelectrodes is critically evaluated using the Nikolskii-Eisenman formalism. Recent progress in the design of new ionophores is presented. A novel neutral carrier-based Ca2+-selective microelectrode with a detection limit of about 5 X 10(-10) M Ca2+ at a background of 125 mM K+ has been realized. An neutral carrier-based microelectrode for H+ with extended pH range of the sample solution is now available. Promising developments in the field of Li+-, Mg2+-, and Cl--selective ionophores are discussed.

The effect of molecular charge on protein permeability of the dually perfused guinea-pig placenta has been investigated by measurement of the permeability surface area products (PS) and observation of the ultrastructural localization of cationic horseradish peroxidase (cHRP) and anionic horseradish peroxidase (aHRP) molecules. Steady-state PS calculated from the experimental data was 0.032 +/- 0.0045 and 0.0045 +/- 0.0008 ml min-1 (mean +/- s.e.m.) for cHRP and aHRP respectively (P less than 0.01). The PS for a diffusional marker, 51Cr-ethylenediaminetetraacetic acid, showed no significant difference between the two groups. Ultrastructurally, placentae perfused with cHRP showed fewer microvilli and a dilated interstitial space compared with placentae perfused with aHRP; large vacuoles were also found in the syncytiotrophoblast in the former but not in the latter tissue. Reaction product in placentae perfused with cHRP was localized to the maternal-facing plasma membrane of the syncytiotrophoblast, in vacuoles and vesicles of the syncytiotrophoblast, and also in the basement membrane of the interstitial space, whereas placentae perfused with aHRP only had reaction product in vesicles in the syncytiotrophoblast. These results suggest that anionic sites in the guinea-pig placenta affect its permeability to charged proteins, cationic molecules inducing structural changes associated with increased permeability.

The ionic mechanisms of adaptation of the receptor potential, especially the early adaptation, was studied in an isolated mechanoreceptor, the slowly adapting crustacean stretch receptor, with electrophysiological methods including potential clamp technique. This receptor has frequently been used as a model to demonstrate general principles for the generation of signals in sensory receptors. The receptor potential is depolarizing and consists of an initial peak followed by a static phase in response to a ramp and hold stretch with a high rate of rise. The receptor current, defined as the stretch induced current, is inward and has a similar appearance. The adaptive fall of the receptor potential and receptor current occurs in two phases, an initial rapid fall from the peak (early adaptation) followed by a slow decline during the static phase (late adaptation). The two phases could be characterized by different time constants. By polarizing the cell to various steady potentials before stretch it was shown that the time constant of the early adaptive fall of the receptor potential varied with potential and had a minimum close to resting membrane potential. The time course of the late adaptive fall appeared to be independent of holding potential. These results suggest that ionic mechanisms play an important role during early adaptation, whereas late adaptation probably is governed by mechanical factors. Prolonged exposure to K+-free solutions or injection of TEA caused nearly complete abolition of the early adaptive fall of the receptor potential, making the response almost rectangular in shape. The outward current associated with a positive potential step was considerably reduced after injection of TEA. Intracellular injection of Ca2+ or exposure to isotonic Ca2+-saline decreased the amplitude of the static phase of the receptor potential, whereas injection of EGTA caused an increase in amplitude. Ion substitution experiments indicated that Ca2+ might enter the neuron through the transducer channels opened by stretch. It is concluded that the major cause of the early adaptive fall of the receptor potential is an outward K+ current activated by the stretch-induced depolarization. The effects of varied intra- and extracellular Ca2+ concentration suggest that a part of the K+ current might be activated by Ca2+ influx. Another possibility is that intracellular Ca2+ contributes to adaptation by inactivation of the transducer channels. The present results demonstrate a possible mechanism for adaptation in sensory receptors. A potential activated outward K+ current enhances the fall of the receptor potential from the initial peak.(ABSTRACT TRUNCATED AT 400 WORDS)

The axial elastic modulus as a function of relative fiber width was measured in relaxed skinned fiber segments from the semitendinosus muscle of the frog. Fiber width was reduced by adding the non- penetrating long-chain polymer Dextran T500 (Mm = 181,800 D) to the fiber bathing solution. The axial elastic modulus increased steeply as relative fiber width was reduced. This relationship is independent of both ionic strength and of the presence of low molecular weight fractions of dextran within the interfilament space. The observed increase in axial elastic modulus with compression may reflect an interaction between crossbridges and thin filaments, an hypothesis which is corroborated by the similarity of responses obtained from compressed fibers and from width fibers in rigor.

Isolated segments of hamster small intestine were perfused with oxygenated salt-fluorocarbon emulsions with or without 10-25 mM glucose, alanine or leucine. Resistances of intercellular occluding junctions and of lateral spaces and the distributed capacitance of epithelial plasma membranes were estimated from steady-state transepithelial impedances at frequencies from 0.01-10 kHz. The segments were then fixed in situ with isorheic 2.5% glutaraldehyde while continuing to measure impedance. This method of fixation increased the resistance of lateral spaces but had little effect on the resistance of occluding junctions or on membrane capacitance. The large decreases of impedance induced by glucose or amino acids were preserved in fixed tissue and could therefore be correlated with changes in structure. The observed changes of impedance were interpreted as decreased resistance of occluding junctions and lateral spaces together with increased exposed surface of lateral membranes (capacitance). Glucose, alanine or leucine induced expansion of lateral intercellular spaces as seen by light and electron microscopy. Large dilatations within absorptive cell occluding junctions were revealed by electron microscopy. Freeze-fracture analysis revealed that these dilatations consisted of expansions of compartments bounded by strands/grooves. These solute-induced structural alterations were also associated with condensation of microfilaments in the zone of the perijunctional actomyosin ring, typical of enhanced ring tension. Similar anatomical changes were found in epithelia fixed in situ at 38 degrees C during luminal perfusion with glucose in blood-circulated intestinal segments of anesthetized animals. These structural changes support the hypothesis that Na-coupled solute transport triggers contraction of perijunctional actomyosin, thereby increasing junctional permeability and enhancing absorption of nutrients by solvent drag as described in the two accompanying papers.

The distribution of pyruvate, phosphate, malate, citrate, K+, aspartate, glutamate, ADP, and ATP between the mitochondrial and cytosolic compartments was studied in isolated rat hepatocytes exposed to 30 min anoxia. The results show that pyruvate and citrate gradients are comparable to aerobic values, indicating that the pH gradient across the membrane under anaerobic conditions is comparable to that under normal aerobic conditions. In contrast, the distribution of phosphate, malate, ATP, ADP, aspartate, and glutamate suggests that transport of these species may be inhibited during anoxia. The results are discussed in terms of potential regulation of mitochondrial function to provide a quiescent anoxic state that is capable of recovering normal function on reoxygenation.

Intra-and extracellular ion activities were measured with ion sensitive microelectrodes in motoneurones and glia cells of the spinal cord of the frog. These data were corrected for cross sensitivities of the ion exchangers to intracellular interfering ions, and equilibrium potentials for K +, Na +, Ca2 + and C1- (EK, ENa, ECa and EC1) were calculated. In motoneurones with membrane potentials exceeding -60 mV the following mean equilibrium potentials were determined. ENa = + 29.4mV, EK = -87.9 mV, ECa = + 52.6 mV, EC1 = -34.1 mV. The corresponding values for glia cells were: ENa = + 40.5 mV, EK = -84.0 mV, ECa = + 35.7 mV, EC1 = -59.7 mV. The intracellular ionic milieu is probably disturbed by the impalement of the cells. This transiently decreases the intracellular K + and increases intracellular Na +. These effects were estimated and their origin is discussed. The results of the experiments suggest a non-passive transmembrane distribution of K +, Na + and Ca2 + in motoneurones and glia cells, a non-passive transmembrane distribution of C1- in motoneurones, and a passive transmembrane distribution of C1- in glia cells.

Morphogenetic fields must be generated by mechanisms based on known physical forces which include gravitational forces, mechanical forces, electrical forces, or some combination of these. While it is unrealistic to expect a single force, such as a voltage gradient, to be the sole cause of a morphogenetic event, spatial and temporal information about the electrical fields and ion concentration gradients in and around a cell or embryo undergoing morphogenesis can take us one step further toward understanding the entire morphogenetic mechanism. This is especially true because one of the handful of identified morphogens is Ca2+, an ion that will not only generate a current as it moves, but which is known to directly influence the plasma membrane's permeability to other ions, leading to other transcellular currents. It would be expected that movements of this morphogen across the plasma membrane might generate ionic currents and gradients of both electrical potential and intracellular concentration. Such ionic currents have been found to be integral components of the morphogenetic mechanism in some cases and only secondary components in other cases. My goal in this review is to discuss examples of both of these levels of involvement that have resulted from investigations conducted during the past several years, and to point to areas that are ripe for future investigation. This will include the history and theory of ionic current measurements, and a discussion of examples in both plant and animal systems in which ionic currents and intracellular concentration gradients are integral components of morphogenesis as well as cases in which they play only a secondary role. By far the strongest cases for a direct role of ionic currents in morphogenesis is the polarizing fucoid egg where the current is carried in part by Ca2+ and generates an intracellular concentration gradient of this ion that orients the outgrowth, and the insect follicle in which an intracellular voltage gradient is responsible for the polarized transport from nurse cell to oocyte. However, in most of the systems studied, the experiments to determine if the observed ionic currents are directly involved in the morphogenetic mechanism are yet to be done. Our experience with the fucoid egg and the fungal hypha of Achlya suggest that it is the change in the intracellular ion concentration resulting from the ionic current that is critical for morphogenesis.

Simultaneous intra- and extracellular recordings with K+, Na+, Ca2+, and Cl- sensitive microelectrodes were performed in motoneurones of the spinal cord of the frog during depolarizations mediated by glutamate (GLUT) and by experimentally increased extracellular K+. Depolarization resulting from increased K+ activity (alpha K+) in the bathing solution evoked a decrease of intracellular Na+ activity (alpha Na+i); a transient increase of alpha Na+i accompanied by a decrease of alpha Na+e was observed during the depolarization induced by GLUT. Both modes of depolarization led to an increase of alpha Cl-i and a concomitant decrease of alpha Cl-e. An experimental increase of alpha K+e led to a threshold dependent increase of alpha Ca2+i by at least one order of magnitude and to an equally threshold dependent strong decrease of alpha Ca2+e. The threshold of these changes of alpha Ca2+ was at a membrane potential of -25 mV. During a depolarization of half the amplitude induced by GLUT a comparable increase of alpha Ca2+i and a smaller decrease of alpha Ca2+e were observed. The GLUT mediated changes of alpha Ca2+ were not threshold dependent and occurred synchronously with the onset of depolarization. A transient decrease of alpha K+i and a parallel strong increase of alpha K+e occurred during the GLUT induced depolarization. Depolarization evoked by an experimental increase of alpha K+e led to an increase of alpha K+i. The observed changes in the ionic composition of the intra- and extracellular fluids indicate that GLUT evokes an increase in membrane permeability to Na+ and Ca2+ and a subsequent influx of these ions into motoneurones, while the inward shift of Cl- and the outward shift of K+ are presumably passive. A voltage dependent Ca2+ influx is triggered at -25 mV membrane potential.

Synchronous beating between chick embryonic heart cell aggregates and heart cell layers was used to study the relationship between intercellular adhesion and ionic coupling. Adhesion was measured by counting the proportion of aggregates which were not to be removed from cell layers by gentle washing after a 30 min incubation. Synchrony between bound aggregates and contiguous layers was assessed by phase microscopy. The first evidence of synchrony was seen 1.5 h after addition of aggregates to layers, following which there was an increase in the percentage of aggregates beating synchronously, reaching over 50% at 7 h and slowly increasing to a maximum of 65% by 24 h. Scanning electron microscopy and autoradiography of thymidine-labeled cells suggest that synchrony does not depend on cell movement at the interface between aggregate and layer. Acquisition of synchrony can be prevented completely by inhibiting protein synthesis, although pulsation of aggregates and layers continues in proportions unchanged from controls. After reversal of protein synthesis inhibition, synchrony is acquired at a rate and to an extent closely resembling that of newly adherent controls. These data indicate that ionic coupling is neither an inevitable nor an immediate consequence of adhesion. Since ionic coupling has been shown to correlate with the presence of gap junctions, the findings suggest that gap junctions are not involved in the initial events responsible for intercellular adhesion in vitro and that their formation following adhesion in this system may depend upon protein synthesis.

Carotid body chemoreceptors are activated by low PO2, high PCO2, acidity, increased temperature, and tonicity. These receptors are important in homeostasis and mediate their reflex effects on the CNS through sensory discharges of the carotid (sinus) nerve. The receptor complex is formed by glomus (type I) cells and carotid nerve endings, which, morphologically, appear to form a sensory synapse. The junction between glomus cells and nerve endings is enveloped by processes of sustentacular (type II) cells. The mechanisms of chemoreceptor transduction are complex; there is no agreement about the identity of the primary receptor element (glomus cell or nerve terminal) or what mechanisms are responsible for the onset of the sensory discharge in the carotid nerve. There is increasing evidence that integrity of the glomus cell is essential for normal transduction and that the receptor synapse described by morphologists may be functionally active. There is no conclusive evidence, however, that the glomus cell is the primary site of sensory transduction. Stimuli act on the glomus cell to release "transmitter" and/or "modulator" substances; but it is unknown if the released chemicals are directly responsible for the accompanying change in sensory impulse frequency or merely modify an already ongoing discharge. Interactions between glomus cells and nerves may be complicated enough to make it very difficult to resolve this question.

Three related hypotheses are proposed. (1) Many simple viruses disassemble in the act of crossing a membrane, drawing energy from ionic disequilibria across the membrane. (2) Many of the metabolic changes early in virus infection are due to altered ion fluxes across membranes and the consequently altered ion concentrations in cytoplasm. (3) The crucial determinant of the transformed phenotype is also a change in ion concentrations. Ca2+ ions may play a major role in these effects.

Currents flowing through the postsynaptic membrane of an active synapse will tend to change the concentrations of ions in the synaptic cleft. Published experimental data are used to predict (a) the sodium and potassium concentration changes in the cleft at the frog neuromuscular junction, and (b) the sodium depletion in the cleft under a Ia synaptic bouton on a cat motoneuron. Significant concentration changes are predicted at both synapses. These changes will contribute to the time dependence of the observed current and will cause the reversal potential of the current to be time dependent. At the frog neuromuscular junction, the time course of the endplate current has been shown previously to depend on the magnitude of the current flowing (at a given potential). We attribute this to changes of the cleft ion concentration. The time dependent changes of the endplate current reversal potential that we predict for the neuromuscular junction are probably too small to be detected. This is because the effects of sodium depletion and potassium accumulation on the reversal potential almost cancel. We predict that near the reversal potential small currents of complex time course will remain, i.e. no true reversl potential exists. Such currents have previously been experimentally. At the cat Ia synapse, the synaptic current is predicted to deplete a significant fraction of the available extracellular sodium ions. Consequently, the magnitude of the synaptic current should be relatively independent of the number of postsynaptic channels activated, and of the membrane potental, as has previously been found experimentally.

Changes in ion concentration can be expected to accompany natural activity in any preparation with infolded membranes or narrow spaces between cells. Such preparations have complex electrical properties because some of the membranes are isolated by the resistance of the solution in narrow extracellular spaces. Analyses of the diffusional and electrical consequences of these structural complexities are intimately related: both require morphometric measurement and theoretical analysis of the the structure. The measured properties of complex tissues, either diffusional or electrical, depend on the properties of many membranes and intra- and extracellular compartments. Fitting a structural theory to one type of experimental data separates some of the measured properties of the tissue into the properties of its components. Fitting electrical data alone give many electrical parameters of the tissue components. Fitting diffusional data alone may give some of the permeability and diffusional parameters of the tissue components. A complete understanding of ion accumulation probably requires a concomitant analysis of elctrical properties.
