Abstract: The extraction of RNA and lipids from a large number of biological samples is time-consuming and costly with steps required for both transcriptomic and lipidomic approaches. Most protocols rely on independent extraction of nucleic acids and lipids from a single sample, thereby increasing the need for biological material and inducing variability in data analysis. We investigated whether it is possible to use a standard RNA extraction procedure to analyze not only RNA levels, but also lipids in a single liver sample. We show that the organic phase obtained when using standard reagents for RNA extraction can be used to analyze lipids, including neutral lipids and fatty acids, by gas chromatography. We applied this technique to an analysis of lipids and the associated gene expression pattern in mice with hepatic steatosis induced by pharmacological activation of nuclear receptor LXR.

Abstract: Mollusca evolutionary success can be attributed partly to their efficiency to sustain and protect their soft body with an external biomineralized structure, the shell. Current knowledge of the protein set responsible for the formation of the shell microstructural polymorphism and unique properties remains largely patchy. In Pinctada margaritifera and Pinctada maxima , we identified 80 shell matrix proteins, among which 66 are entirely unique. This is the only description of the whole “biomineralization toolkit” of the matrices that, at least in part, is thought to regulate the formation of the prismatic and nacreous shell layers in the pearl oysters. We unambiguously demonstrate that prisms and nacre are assembled from very different protein repertoires. This suggests that these layers do not derive from each other.

Abstract: International audience; Metabolic diseases such as obesity, type II diabetes and hepatic steatosis are a public health concern in developed countries. The metabolic risk is gender-dependent. The constitutive androstane receptor (CAR), which is at the crossroads between energy metabolism and endocrinology, has recently emerged as a promising therapeutic agent for the treatment of obesity and type 2 diabetes. In this study we sought to determine its role in the dimorphic regulation of energy homeostasis. We tracked male and female WT and CAR deficient (CAR-/-) mice for over a year. During aging, CAR-/- male mice developed hypercortisism, obesity, glucose intolerance, insulin insensitivity, dyslipidemia and hepatic steatosis. Remarkably, the latter modifications were absent, or minor, in female CAR-/- mice. When ovariectomized, CAR-/- female mice developed identical patterns of metabolic disorders as observed in male mice. These results highlight the importance of steroid hormones in the regulation of energy metabolism by CAR. They unveil a sexually dimorphic role of CAR in the maintenance of endocrine and metabolic homeostasis underscoring the importance of considering sex in treatment of metabolic diseases.

Abstract: The pearl oyster Pinctada margaritifera has the specific ability to produce pearls with the widest range of colours among all pearl oyster species. This pearl colour diversity originates from the mantle biomineralising tissue (graft) of the donor oyster, which is originally responsible for the variety of colours of the inner shell surface. This study aimed to: 1) assess the geographic distribution and establish a first stocklist of the colourful oyster phenotypes used as donors in French Polynesia, and 2) investigate the phenotypic relation between inner shell colouration and the corresponding colour of harvested pearls. With the support of a pearl farmers' network, we investigated the different donor phenotype frequencies among five collection sites (Ahe, Apataki, Takaroa, TakumeandMangareva). This donor evaluation was made during grafting of pearl oysters (N=49,938) obtained from collector stations. Results showed that pearl production is mainly based on six common colourful donor phenotypes classified as monochromatic and polychromatic profiles, which shown different frequencies among the collection sites. Experimental grafts (N=4640)were then realised and subsequent culture conducted at a single site in order to avoid pearl colour variation due to environmental influences. Traceability between donors (N=232) and pearls (N=2776), revealed that: 1) yellow (gold) and aubergine (reddish) pearls could be mostly obtained by using the monochromatic yellowand red donor phenotypes, respectively, and 2) one third to one quarter of grey pearlswas inevitably harvested, whatever the polychromatic phenotype chosen as the donor, which leaves at least half the harvest composed of the attractive green and peacock colours. This preliminary stocklist of colour range together with analysis of the colour phenotype transmission between inner shell and pearl provide the basis for producing multiple pearl oyster “colour lines” through hatchery propagation and would be helpful for future selective breeding programs. Abstract: Genetic sex determination (GSD) can evolve from environmental sex determination (ESD) via an intermediate state in which both coexist in the same population. Such mixed populations are found in the crustacean Daphnia magna, where non-male producers (NMP, genetically determined females) coexist with male producers (MP), in which male production is environmentally inducible and can also artificially be triggered by exposure to juvenile hormone. This makes Daphnia magna a rare model species for the study of evolutionary transitions from ESD to GSD. Although the chromosomal location of the NMP-determining mutation has been mapped, the actual genes and pathways involved in the evolution of GSD from ESD remain unknown. Here, we present a transcriptomic analysis of MP and NMP females under control (female producing) and under hormone exposure conditions. We found similar to 100 differentially expressed genes between MP and NMP under control conditions. Genes in the NMP-determining chromosome region were especially likely to show such constitutive expression differences. Hormone exposure led to expression changes of an additional similar to 100 (MP) to similar to 600 (NMP) genes, with an almost systematic upregulation of those genes in NMP. These observations suggest that the NMP phenotype is not determined by a simple "loss-of-function" mutation. Rather, homeostasis of female offspring production under hormone exposure appears to be an active state, tightly regulated by complex mechanisms involving many genes. In a broader view, this illustrates that the evolution of GSD, while potentially initiated by a single mutation, likely leads to secondary integration involving many genes and pathways.

Abstract: Ventilation and drainage in termite nests are controlled by microscale morphological features of the outer walls., Termite nests have been widely studied as effective examples for ventilation and thermoregulation. However, the mechanisms by which these properties are controlled by the microstructure of the outer walls remain unclear. Here, we combine multiscale X-ray imaging with three-dimensional flow field simulations to investigate the impact of the architectural design of nest walls on CO2 exchange, heat transport and water drainage. We show that termites build outer walls that contain both small and percolating large pores at the microscale. The network of larger microscale pores enhances permeability by one to two orders of magnitude compared to the smaller pores alone, and it increases CO2 diffusivity up to eight times. In addition, the pore network offers enhanced thermal insulation and allows quick drainage of rainwater, thereby restoring the ventilation and providing structural stability to the wet nest.

Abstract: Background: In French Polynesia, thyroid cancer mortality and incidence is reported to be the highest in the world. Excessive levels of non-essential trace elements (nETE) in the body are associated with several types of cancer. Objective: The present study aims to provide quantitative information on food contamination by mercury (Hg), lead (Pb), arsenic (As) and cadmium (Cd) in French Polynesia and its potential correlation with measurements performed in fingernails of Polynesians, and then to investigate the potential association between these nETE and different thyroid cancer risks. Methods: The study population included 229 interviewed cases and 373 interviewed controls We performed a descriptive analysis of Polynesian food and examined the association between thyroid cancer risk and daily intake levels of nETE and with fingernail nETE levels. Results: Hg contamination was mainly present in sea products, Pb contamination was present in almost all samples, Cd was detectable in starchy food and As was detectable in all sea products. No patient exceeded dietary contamination WHO limits for Pb, 2 participants exceeded it for Hg and 3 individuals (0.5%) for cadmium. In fingernail clippings, the most detectable pollutant was Pb (553 participants), then Hg (543 participants) then Cd (only in 130 participants). Thyroid cancer risk was increased more than 4 times by Pb daily intake in patients with a history of cancer in first-degree relatives than in ones without (p for interaction =0.01), and 2 times more in women with more than 3 pregnancies than in those with none or less (p for interaction =0.005); it was also increased following As intake by more than 30% in patients with a history of cancer in first-degree relatives than in ones without (p for interaction =0.05). Conclusion: Locally produced foods are not a source of nETE exposure in French Polynesia. Dieatry nETE exposure and fingernail nETE concentration are not associated to differentiated thyroid cancer risk. No correlation found between nETE dietary exposure and fingernail nETE concentration.

We propose a new methodological framework, based on network theory and modularity concept, to determine which type of mechanisms (i.e. deterministic versus stochastic processes) has the strongest influence on structuring communities. This framework is based on the computation and comparison of two networks: the co-occurrence (based on species abundances) and the functional networks (based on the species traits values). In this way we can assess whether the species belonging to a given functional group also belong to the same co-occurrence group. We adapted the Dg index of Gauzens et al. (2015) to analyze congruence between both networks. This offers the opportunity to identify which assembly rule(s) play(s) the major role in structuring the community. We illustrate our framework with two datasets corresponding to different faunal groups and ecosystems, and characterized by different scales (spatial and temporal scales). By considering both species abundance and multiple functional traits, our framework improves significantly the ability to discriminate the main assembly rules structuring the communities. This point is critical not only to understand community structuring but also its response to global changes and other disturbances.

Abstract: Cyanobacterial blooms pose serious threats to aquatic organisms and strongly impact the functioning of aquatic ecosystems. Due to their ability to produce a wide range of potentially bioactive secondary metabolites, so called cyanotoxins, cyanobacteria have been extensively studied in the past decades. Proteomic and metabolomic analyses provide a unique opportunity to evaluate the global response of hundreds of proteins and metabolites at a glance. In this study, we provide the first combined utilization of these methods targeted to identify the response of fish to bloom-forming cyanobacteria. Medaka fish (Oryzias latipes) were exposed for 96 hours either to a MC-producing or to a non-MC-producing strain of Microcystis aeruginosa and cellular, proteome and metabolome changes following exposure to cyanobacteria were characterized in the fish livers. The results suggest that a short-term exposure to cyanobacteria, producing or not MCs, induces sex-dependent molecular changes in medaka fish, without causing any cellular alterations. Globally, molecular entities involved in stress response, lipid metabolism and developmental processes exhibit the most contrasted changes following a cyanobacterial exposure. Moreover, it appears that proteomic and metabolomic analyses are useful tools to verify previous information and to additionally bring new horizons concerning molecular effects of cyanobacteria on fish.

Abstract: International audience; To meet the dietary requirements of a burgeoning human population, the demand for animal-dependent crops continues to grow. To meet the demand, intensive farming practices are used. The gains in food production associated with agricultural intensification may be offset by its detrimental effects on pollinator populations through natural habitat fragmentation and pesticide use. Abundance and species richness of pollinators have been found to decrease with increasing distance to natural habitat in agroecosystems, reducing crop yields. A key aspect of crop pollination lies in the diversity of functional traits (functional diversity, FD) of flower-visitor communities within crop fields. Higher FD allows improved pollination success through complementarity between flower-visitors’ morphology, phenology and behaviour. Many studies reported negative effects of increasing distance to natural habitats on the abundance and richness of flower-visitor communities, but the link between FD and natural habitat isolation is less well understood. Also, a more complete understanding of the functional traits of flower-visitor communities within crops should consider potential variations through time. Differences in resources availability between seasons are important in tropical areas and could modify ecological responses of flower-visitor communities to isolation. In this study, we surveyed the Hymenoptera and Diptera communities within mango orchards of South Africa using pan traps at 100 m, 200 m and at the maximal distance possible from any natural habitat. We measured the response of insect abundance, wing span and body size as well as functional diversity to habitat isolation during mango flowering (dry season), and during the wet season (after mango fruit harvest). Flying insect abundance decreased with increasing distance to natural habitat during mango flowering, but no effect was detected during the wet season. FD of flying insects declined with increasing distance to natural habitat in both sampling periods. Insects captured during mango flowering were smaller but had higher wing length/body length ratios than those caught during the wet season. This study highlights that mango orchards are more inhospitable for flying insects during mango flowering. This effect might be due to low palatability of mango flowers, or pesticide use in mango fields. In order to maintain a high FD of flower-visiting species, and reduce the detrimental effects of habitat isolation to ultimately ensure better crop pollination, we propose establishment of patches of resource-rich habitats combined with judicious use of pesticides within orchards.

Abstract: The VAHINE mesocosm experiment was designed to trigger a diazotroph bloom and to follow the subsequent transfer of diazotroph-derived nitrogen (DDN) in the rest of the food web. Three mesocosms (50 m3) located inside the Nouméa lagoon (New Caledonia, southwestern Pacific) were enriched with dissolved inorganic phosphorus (DIP) in order to promote N2 fixation in these low-nutrient, low-chlorophyll (LNLC) waters. Initially, the diazotrophic community was dominated by diatom diazotroph associations (DDAs), mainly by Rhizosolenia/Richelia intracellularis, and by Trichodesmium, which fueled enough DDN to sustain the growth of other diverse diatom species and Synechococcus populations that were well adapted to limiting DIP levels. After DIP fertilization (1 µM) on day 4, an initial lag time of 10 days was necessary for the mesocosm ecosystems to start building up biomass. However, changes in community structure were already observed during this first period, with a significant drop of both Synechococcus and diatom populations, while Prochlorococcus benefited from DIP addition. At the end of this first period, corresponding to when most added DIP was consumed, the diazotroph community changed drastically and became dominated by Cyanothece-like (UCYN-C) populations, which were accompanied by a monospecific bloom of the diatom Cylindrotheca closterium. During the second period, biomass increased sharply together with primary production and N2-fixation fluxes near tripled. Diatom populations, as well as Synechococcus and nanophytoeukaryotes, showed a re-increase towards the end of the experiment, showing efficient transfer of DDN to non-diazotrophic phytoplankton.

Abstract: International audience; Key message Teak (Tectona grandis L.f.) is one of the most important tropical hardwood tree species, which is widely planted in Indonesia. Wood properties are strongly influenced by forest management conditioning further utilization of wood.Context In Indonesia, teak wood has been supplied from the state forests (Perhutani) for long rotation teak and from community teak plantations for short rotation teak. Short rotation teak has been harvested at 7-10 years and long rotation teak at 40-60 years.Aims This paper discusses the characterization of technical properties of short and long rotation teak wood based on the chemical, anatomical, physical, and mechanical properties.Methods The properties of short rotation and long rotation teak woods were characterized by measuring their density, extractive contents, chemical composition, swelling, wettability, water sorption isotherm, decay resistance, anatomical properties, bending strength (modulus of rupture (MOR), modulus of elasticity (MOE)), and hardness.Results The results indicate that short rotation teak was not particularly different in swelling, MOE and MOR, and Brinell hardness compared to long rotation teak, although it was less dense and less durable due to lower heartwood and extractive contents. Therefore, careful attention should be given to the use of short rotation teak in some wood-processing technologies.Conclusion Lower wood density and durability of the short rotation compared to the long rotation teak will restrict its utilization to some extent for both indoor and outdoor applications. Fast-growing teak from community cannot be used as usual heartwood teak from Perhutani because of the very low proportion of useful heartwood in the stem.

Abstract: 9 pages; International audience; French Polynesia is renowned for the production of Tahitian black pearl. These gems are obtained by grafting a nucleus into the gonad of a receiving oyster together with a graft, i.e. a small section of mantle tissue of a donor oyster. This procedure initiates the formation of a pearl sack around the nucleus, and subsequently, the deposition of concentric layers of nacre. The nucleus plays a key-role in pearl formation and its characteristics influence markedly the quality of the final product. As it is manufactured from mollusc shells, it contains a small percentage of organics. In the present paper, we used a set of biochemical techniques to characterize and compare the organic matrices from two types of nuclei that are currently used in French Polynesia: that from the freshwater mussel Amblema sp., and that from the pearl oyster Pinctada sp. To this end, we extracted the matrices from nuclei and performed FT-IR, monodimensional electrophoresis, and enzyme-linked immuno-sorbent assay (ELISA). Our data show that the matrix associated with Amblema nuclei has a very different biochemical signature from that of Pinctada nuclei, a fact that may explain the improved tolerance of grafted oysters to nuclei of Pinctada origin. In the absence of complex physical methods of investigation, simple immunological techniques and FT-IR performed on the extracted organic matrix are extremely reliable and effective for discriminating nuclei from these two sources. We assert that such techniques can be used as a diagnostic test to track unambiguously the biological origin of nuclei to avoid fraud.

Abstract: International audience; AimTwo frameworks (BASVIL and PODCAR), based on two different functional representations (ordination and dendrogram), have been proposed for partitioning overall functional beta diversity into two analogous components: turnover and nestedness‐resultant dissimilarity, or replacement and difference of functional richness, respectively. We compared the two frameworks by testing the influence of functional representations and partitioning methods on the measurement of overall functional beta diversity and its components.InnovationWe computed beta‐diversity indices from the two frameworks on a set of communities simulated according to five scenarios of assembly: random, richness gradient, pure nestedness, pure turnover and mixed turnover/loss scenarios. To disentangle the effects of the partitioning approach and those of the functional representation on measurement of functional beta diversity, we also computed PODCAR indices in multidimensional space.Main conclusionsBASVIL and PODCAR frameworks led to different results for overall functional beta diversity and their analogous partitioning components. Most of the difference between the two frameworks was due to the functional representation used. The goodness‐of‐fit measure (mean squared deviation, mSD) to assess the quality of functional spaces showed that the one computed on the basis of the dendrogram used in PODCAR remained lower than that of the functional ordination considered in BASVIL. In addition, only functional turnover derived from the BASVIL framework is independent of difference in functional richness. Finally, BASVIL measured functional variations derived from nested phenomena while PODCAR did not allow separation of this variation derived from richness difference. However, the sensitivity of BASVIL to functionally extreme species may make it difficult to know whether variations of the nestedness‐resultant dissimilarity components are due to a turnover with few extreme species or a loss in functional richness. Particular attention with regard to the properties of the two frameworks is required before drawing conclusions regarding processes that structure communities.

Abstract: International audience; Estimates of trophic position are used to validate ecosystem models and understand food web structure. A consumer's trophic position can be estimated by the stable nitrogen isotope values (delta N-15) of its tissue, once the baseline isotopic variability has been accounted for. Our study established the first data-driven baseline delta N-15 isoscape for the Western and Central Pacific Ocean using particulate organic matter. Bulk delta N-15 analysis on 1039 muscle tissue of bigeye and yellowfin tuna were conducted together with amino acid compound-specific delta N-15 analysis (AA-CSIA) on a subset of 21 samples. Both particulate organic matter and tuna bulk delta N-15 values varied by more than 10 parts per thousand across the study area. Fine-scaled trophic position maps were constructed and revealed higher tuna trophic position (by similar to 1) in the southern latitudes compared to the equator. AA-CSIA confirmed these spatial patterns for bigeye and, to a lesser extent, yellowfin tuna. Using generalized additive models, spatial variations of tuna trophic positions were mainly related to the depth of the 20 degrees C isotherm, a proxy for the thermocline behavior, with higher tuna trophic position estimates at greater thermocline depths. We hypothesized that a deeper thermocline would increase tuna vertical habitat and access to mesopelagic prey of higher trophic position. Archival tagging data further suggested that the vertical habitat of bigeye tuna was deeper in the southern latitudes than at the equator. These results suggest the importance of thermocline depth in influencing tropical tuna diet, which affects their vulnerability to fisheries, and may be altered by climate change.

Abstract: International audience; The semi-enclosed Solomon Sea in the southwestern tropical Pacific is on the pathway of a major oceanic circuit connecting the subtropics to the equator via energetic western boundary currents. Waters transiting through this area replenish the Pacific Warm Pool and ultimately feed the equatorial current system, in particular the equatorial undercurrent. In addition to dynamical transformations, water masses undergo nutrient and micronutrient enrichment when coming in contact with the coasts, impacting the productivity of the downstream equatorial region. Broadscale observing systems are not well suited for describing the fine-scale currents and water masses properties in the Solomon Sea, leaving it relatively unexplored. Two multidisciplinary oceanographic cruises were conducted in the Solomon Sea region, the first in July–August 2012 and the second in March 2014, by investigators from France and the United States. The experimental approach combined physical, chemical, geochemical and biogeochemical analyses, providing access to a wide range of space and time scales of the circulation. This collection of data allows describing the fine-scale structure of the currents and the water properties, transformations and mixing from the surface to the sill depth in the Solomon Sea and in the straits connecting it to the equator. Ocean-margin exchanges were documented through a comprehensive sampling of trace elements and isotopes as efficient tracers of natural fertilization processes. As air chemistry is largely impacted by the regional volcanic plumes, rainwater pH was also sampled. Dinitrogen fixation rates were measured and found to be among the highest in the global ocean, highlighting this region as a hot spot of nitrogen fixation. This study provides an overview of the climatic context during both cruises and the physical circulation and water masses properties. It provides a comprehensive description of all measurements made onboard, and presents preliminary results, aiming to serve as a reference for further physical, geochemical and biogeochemical studies.

Abstract: International audience; Size is the most important and valuable quality of the cultured black-lip pearl, Pinctada margaritifera. As this pearl aquaculture is carried out at numerous grow-out sites, this study analyzes the environmental influence on pearl size parameters (nacre weight and thickness) in relation to the recipient oyster biometric parameters (shell thickness, height, width, and oyster weight) at harvest time. Toward this end, an experimental graft was designed by using a homogeneous donor oyster phenotype. The recipient oysters were randomly and equally transferred and reared in five commercial and contrasting grow-out locations. Overall inter-site comparisons revealed that the cultured pearl size (N = 2168) and the biometric parameters of the recipient oysters were highest for sites with warmer temperatures with low seasonal variation in comparison to the southern latitude sites. These results were supported by positive correlations between pearl nacre thickness and recipient oyster shell thickness, height, and width. In parallel, the biomineralization potential of the mantle graft was screened through four genes encoding aragonite (Pif 177, MSI60) and calcite (shematrin 9, aspein). As the gene expression levels were the same among all the donor oysters, this finding demonstrates that: 1) the pearl sac that originated from the mantle graft was not isolated from environmental variations during the culture period and 2) the phenotypic expressions of the two biomineralizing tissues in the recipient oyster were consistent (shell and pearl). In the near future, this knowledge will be helpful at the production sites of genetically selected donor oyster lines for growth produced in hatchery systems. Abstract: International audience; This article reports the results of a survey of the pearl oyster industry in French Polynesia territory. Its purpose is to examine the perceptions of the priorities for the development of this industry towards sustainable development. These perceptions were apprehended by a survey of pearl oyster farmers and other stakeholders of the sector (management authorities, scientists). After describing the methodological protocol of these investigations, it comes to confront the priorities chosen by professionals (i.e. pearl farmers) concerning sustainable development, with the perceptions of others stakeholders in the sector. Secondly it comes to build a typology of the priorities of pearl farmers concerning sustainable development. This analysis enables the assessment of the degree of convergence within the sector, which is the base material for defining a shared action plan at the territory scale. This is the first study compiling data of surveys of various professionals and stakeholders of the pearl farming industry in such a large area in French Polynesia.

Abstract: ObjectiveTo measure the impact of a short-term variable-workload endurance-training programme (PEP’C) on cardiorespiratory parameters and endurance, and also on quality of life for seniors. The respective roles of the perception of physical activity and the financial factor involved in joining such a program were also investigated.PopulationTwo hundred and thirty-two volunteers aged 50 or over voluntarily participated in the “Physical Fitness for Health” assessment (CAPS). Subjects presenting the common contra-indications to physical activity were previously excluded.MethodAfter an initial assessment of aerobic capacity using a maximum cardiorespiratory test, a special variable-workload endurance-training programme on ergometric bicycles was run, comprising 18 sessions of 30 minutes each for 9 weeks. A final assessment of aerobic capacity completed the program. Quality of life was measured before and after the PEP’C using the MOS-SF-36.ResultsThe volunteers’ mean age was 66.0 ± 6.5 years ; 77.5% were aged 60 to 74 years and 11.6% were 75 years or older. Among the volunteers, 15.9% had never practiced any sport or physical activity. One out of two said that maintaining their physical fitness was their main motivation. The programme adherence rate was 100%. Over 18 sessions, maximum cardiorespiratory parameters (MTP VO2peak and MMV) and aerobic endurance parameters (VT1, HR, VT1, and lactate at MTP) improved (P > 0.05). An effect of gender was observed whereby improvement in performance was significantly greater in women. For aerobic endurance abilities, a significant gain was measured for 92.2% of the participants. Before starting PEP’C, scores on the MOS-SF-36 were high in all dimensions. At the end, all quality of life dimensions were significantly improved and the increase in global score was 10.0% ± 20.0%. The mean scores in the dimensions of “perceived health” and “vitality” were those that increased most. In men, physical activity particularly improved the “vitality” dimension, while for women it was rather the “mental health” dimension (P < 0.01). On average, 134 € was the amount considered affordable for a program of this sort, this amount being strongly dependent on the level of satisfaction.ConclusionThis study confirms the feasibility and benefits of this “individually tailored” PEP’C on maximum cardiorespiratory and endurance parameters, and on quality of life, among seniors. It also shows that adherence to a programme of this type is more influenced by the satisfaction derived from practicing physical activity and sport than by affordability., ObjectifsMesurer l’impact d’un programme d’entraînement à l’effort personnalisé par créneaux (PEP’C) sur les paramètres cardiorespiratoires et d’endurance, mais également la qualité de vie des seniors. Parmi les volontaires, 15,9 % n’avaient jamais pratiqué d’activité physique et sportive. Un sur deux déclarait que le maintien ou la remise en forme physique était leur motivation pour faire du sport. Le taux d’adhésion était de 100 %. En 18 séances, une amélioration des paramètres cardiorespiratoires maximaux (PMT, VO2pic, VMM) et des paramètres d’endurance aérobie (SV1, FC au SV1, taux de lactate à la PMT) était observée (p < 0,05). Il a été observé un effet du sexe avec une augmentation des performances significativement supérieure chez les femmes. Pour les aptitudes d’endurance aérobie, il était mesuré un gain significatif pour 92,2 % des participants. Avant d’initier le PEP’C, les scores au MOS-SF-36 étaient élevés dans tous les domaines. À la fin du PEP’C, tous les domaines de qualité de vie étaient significativement augmentés et l’augmentation du score global était de 10,0 ± 20,0 %. Les scores moyens dans les dimensions de « santé perçue » et de « vitalité » étaient les plus augmentés. Chez les hommes, la pratique d’une activité physique améliorait particulièrement la dimension « vitalité » alors que pour les femmes c’était plutôt la « santé psychique » (p < 0,01). En moyenne, 134 € était la somme qui pourrait être dédiée à un tel programme, le montant étant fortement dépendant du niveau de satisfaction.ConclusionCette étude confirme la faisabilité et les bénéfices du programme personnalisé PEP’C sur les principaux paramètres cardiorespiratoires maximaux, l’endurance et la qualité de vie des seniors. Elle montre aussi que l’adhésion à un tel dispositif est peu influencée par la satisfaction induite par la pratique d’une activité physique et sportive que par les coûts engendrés.

Abstract: International audience; The nests of social insects are not only impressive because of their sheer complexity but also because they are built from individuals whose work is not centrally coordinated. A key question is how groups of insects coordinate their building actions. Here, we use a combination of experimental and modeling approaches to investigate nest construction in the ant Lasius niger. We quantify the construction dynamics and the 3D structures built by ants. Then, we characterize individual behaviors and the interactions of ants with the structures they build. We show that two main interactions are involved in the coordination of building actions: (i) a stigmergic-based interaction that controls the amplification of de-positions at some locations and is attributable to a pheromone added by ants to the building material; and (ii) a template-based interaction in which ants use their body size as a cue to control the height at which they start to build a roof from existing pillars. We then develop a 3D stochastic model based on these individual behaviors to analyze the effect of pheromone presence and strength on construction dynamics. We show that the model can quantitatively reproduce key features of construction dynamics, including a large-scale pattern of regularly spaced pillars, the formation and merging of caps over the pillars, and the remodeling of built structures. Finally, our model suggests that the lifetime of the pheromone is a highly influential parameter that controls the growth and form of nest architecture.

Abstract: International audience; BackgroundCalophyllum inophyllum L. (Calophyllaceae) is an evergreen tree ethno-medically used along the seashores and islands of the Indian and Pacific Oceans, especially in Polynesia. Oil extracted from the seeds is traditionally used topically to treat a wide range of skin injuries from burn, scar and infected wounds to skin diseases such as dermatosis, urticaria and eczema. However, very few scientific studies reported and quantified the therapeutic properties of Calophyllum inophyllum oil (CIO). In this work, five CIO from Indonesia (CIO1), Tahiti (CIO2, 3), Fiji islands (CIO4) and New Caledonia (CIO5) were studied and their cytotoxic, wound healing, and antibacterial properties were presented in order to provide a scientific support to their traditional use and verify their safety.MethodsThe safety of the five CIO was ascertained using the Alamar blue assay on human keratinocyte cells. CIO wound healing properties were determined using the scratch test assay on human keratinocyte cells. CIO-stimulated antibacterial innate immune response was evaluated using ELISA by measuring beta defensin-2 release in human derivative macrophage cells. CIO antibacterial activity was tested using oilogramme against twenty aerobic Gram-bacteria species, twenty aerobic Gram+ bacteria species, including a multi-drug resistant Staphylococcus aureus strain and two anaerobic Gram+ bacteria species e.g. Propionibacterium acnes and Propionibacterium granulosum. To detect polarity profile of the components responsible of the antibacterial activity, we performed bioautography against a Staphylococcus aureus strain.ResultsBased on Alamar Blue assay, we showed that CIO can be safely used on keratinocyte cells between 2.7% and 11.2% depending on CIO origin. Concerning the healing activity, all the CIO tested accelerated in vitro wound closure, the healing factor being 1.3 to 2.1 higher compared to control when keratinocytes were incubated after scratch with CIO at 0.1%. Furthermore, our results showed that CIO exhibit two distinct antibacterial effects: one against Gram + bacteria by direct inhibition of mitotic growth and another potent effect against Gram-bacteria due to increased release of beta-defensin 2 peptide by macrophages. Interestingly, the needed concentrations of CIO to inhibit bacteria growth and to promote wound healing are lower than concentrations exhibiting cytotoxic effects on keratinocyte cells. Finally, we performed bioautography assay against Staphylococcus aureus to determine polarity profile of the components responsible for CIO antibacterial activity. Our results showed for the five tested CIO that components responsible of the bacterial growth inhibition are the more polar one on the TLC chromatographic profile and are contained in the resinous fraction of the oil.ConclusionsThis study was conducted to evaluate cytotoxicity, wound healing and antibacterial properties of five CIO traditionally used to treat infected wounds. Using cell and bacteria cultures, we confirmed the pharmacological effects of CIO as wound healing and antimicrobial agent. Moreover, we showed that concentration of CIO needed to exhibit therapeutic effects are lower than concentrations exhibiting cytotoxic effects in vitro. For the first time, this study provides support for traditional uses of CIO. These wound healing and antibiotic properties make CIO a valuable candidate to treat infected wounds especially in tropical areas.

Abstract: Cultured pearls are human creations formed by inserting a nucleus and a small piece of mantle tissue into a living shelled mollusc, usually a pearl oyster. Although many pearl observations intuitively suggest a possible rotation of the nucleated pearl inside the oyster, no experimental demonstration of such a movement has ever been done. This can be explained by the difficulty of observation of such a phenomenon in the tissues of a living animal. To investigate this question of pearl rotation, a magnetometer system was specifically engineered to register magnetic field variations with magnetic sensors from movements of a magnetic nucleus inserted in the pearl oyster. We demonstrated that a continuous movement of the nucleus inside the oyster starts after a minimum of 40 days post-grafting and continues until the pearl harvest. We measured a mean angular speed of 1.27° min −1 calculated for four different oysters. Rotation variability was observed among oysters and may be correlated to pearl shape and defects. Nature's ability to generate so amazingly complex structures like a pearl has delivered one of its secrets.

Abstract: International audience; Many animals in heterogeneous environments bias their trajectories displaying a preference for the vicinity of boundaries. Here we propose a criterion, relying on recent invariance properties of residence times for microreversible Boltzmann's walks, that permits detecting and quantifying boundary-following behaviors. On this basis we introduce a boundary-following model that is a nonmicroreversible Boltzmann's walk and that can represent all kinds of boundary-following distributions. This allows us to perform a theoretical analysis of field-resolved boundary following in animals. Two consequences are pointed out and are illustrated: A systematic procedure can now be used for extraction of individual properties from experimental field measurements, and boundary-curvature influence can be recovered as an emerging property without the need for individuals perceiving the curvature via complex physiological mechanisms. The presented results apply to any memoryless correlated random walk, such as the run-and-tumble models that are widely used in cell motility studies.

Abstract: International audience; Ceramides are known to promote insulin resistance in a number of metabolically important tissues including skeletal muscle, the predominant site of insulin-stimulated glucose disposal. Depending on cell type, these lipid intermediates have been shown to inhibit protein kinase B (PKB/Akt), a key mediator of the metabolic actions of insulin, via two distinct pathways: one involving the action of atypical protein kinase C (aPKC) isoforms, and the second dependent on protein phosphatase-2A (PP2A). The main aim of this study was to explore the mechanisms by which ceramide inhibits PKB/Akt in three different skeletal muscle-derived cell culture models; rat L6 myotubes, mouse C2C12 myotubes and primary human skeletal muscle cells. Our findings indicate that the mechanism by which ceramide acts to repress PKB/Akt is related to the myocellular abundance of caveolin-enriched domains (CEM) present at the plasma membrane. Here, we show that ceramide-enriched-CEMs are markedly more abundant in L6 myotubes compared to C2C12 myotubes, consistent with their previously reported role in coordinating aPKC-directed repression of PKB/Akt in L6 muscle cells. In contrast, a PP2A-dependent pathway predominantly mediates ceramide-induced inhibition of PKB/Akt in C2C12 myotubes. In addition, we demonstrate for the first time that ceramide engages an aPKC-dependent pathway to suppress insulin-induced PKB/Akt activation in palmitate-treated cultured human muscle cells as well as in muscle cells from diabetic patients. Collectively, this work identifies key mechanistic differences, which may be linked to variations in plasma membrane composition, underlying the insulin-desensitising effects of ceramide in different skeletal muscle cell models that are extensively used in signal transduction and metabolic studies.

Abstract: Interactions between individuals and the structure of their environment play a crucial role in shaping self-organized collective behaviors. Recent studies have shown that ants crossing asymmetrical bifurcations in a network of galleries tend to follow the branch that deviates the least from their incoming direction. At the collective level, the combination of this tendency and the pheromone-based recruitment results in a greater likelihood of selecting the shortest path between the colony's nest and a food source in a network containing asymmetrical bifurcations. It was not clear however what the origin of this behavioral bias is. Here we propose that it results from a simple interaction between the behavior of the ants and the geometry of the network, and that it does not require the ability to measure the angle of the bifurcation. We tested this hypothesis using groups of ant-like robots whose perceptual and cognitive abilities can be fully specified. We programmed them only to lay down and follow light trails, avoid obstacles and move according to a correlated random walk, but not to use more sophisticated orientation methods. We recorded the behavior of the robots in networks of galleries presenting either only symmetrical bifurcations or a combination of symmetrical and asymmetrical bifurcations. Individual robots displayed the same pattern of branch choice as individual ants when crossing a bifurcation, suggesting that ants do not actually measure the geometry of the bifurcations when travelling along a pheromone trail. Finally at the collective level, the group of robots was more likely to select one of the possible shorter paths between two designated areas when moving in an asymmetrical network, as observed in ants. This study reveals the importance of the shape of trail networks for foraging in ants and emphasizes the underestimated role of the geometrical properties of transportation networks in general., Author Summary Most ant species form transportation networks, be they foraging trails linking food sources to the main colony or underground galleries connecting the different parts of the nest. As for human transportation networks (roads, airlines, etc.), the design and the placement of the connecting points (or nodes) dramatically affects the movement of individuals and hence the exchanges of material and information. In a previous study, we have shown that the geometrical configuration of these nodes (i.e., the angles between the different exiting branches) can affect the route followed by an ant in a network of galleries and, as a consequence, the efficiency of the pheromone-based recruitment toward a food source. Here we show that we can reproduce these results using ant-like robots with minimal perceptual and cognitive capabilities. We demonstrate that the simple interaction between the displacement of an ant and the geometrical configuration of the gallery network can greatly affect the foraging performances of the colony. This result increases our understanding of how workers move through structures built by ant colonies and more generally points toward possible improvements for the design of man-made transportation networks.

Abstract: International audience; The last decades have seen an increasing interest in modeling collective animal behavior. Some studies try to reproduce as accurately as possible the collective dynamics and patterns observed in several animal groups with biologically plausible, individual behavioral rules. The objective is then essentially to demonstrate that the observed collective features may be the result of self-organizing processes involving quite simple individual behaviors. Other studies concentrate on the objective of establishing or enriching links between collective behavior researches and cognitive or physiological ones, which then requires that each individual rule be carefully validated. Here we discuss the methodological consequences of this additional requirement. Using the example of corpse clustering in ants, we first illustrate that it may be impossible to discriminate among alternative individual rules by considering only observational data collected at the group level. Six individual behavioral models are described: They are clearly distinct in terms of individual behaviors, they all reproduce satisfactorily the collective dynamics and distribution patterns observed in experiments, and we show theoretically that it is strictly impossible to discriminate two of these models even in the limit of an infinite amount of data whatever the accuracy level. A set of methodological steps are then listed and discussed as practical ways to partially overcome this problem. They involve complementary experimental protocols specifically designed to address the behavioral rules successively, conserving group-level data for the overall model validation. In this context, we highlight the importance of maintaining a sharp distinction between model enunciation, with explicit references to validated biological concepts, and formal translation of these concepts in terms of quantitative state variables and fittable functional dependences. Illustrative examples are provided of the benefits expected during the often long and difficult process of refining a behavioral model, designing adapted experimental protocols and inversing model parameters.
