User adaptation is important in conversational robots to enhance the user experience and engagement. One way of user adaptation is to change the dialogue behavior. A character (e.g. extrovert or agreeable) can be defined to achieve human-like behaviors and user adaptation, and the appropriate character differs depending on each user. In this study, we investigate user adaptation by character expression of conversational robots. Our character expression model adopts the Big Five traits for controlling four dialogue behaviors: amount of utterance, backchannel frequency, filler frequency, and switching pause length. We cluster the system character and user personality into four classes based on an analysis of a speed dating dialogue corpus, and we found the best combinations between the system character and user personality. We implemented the character expression model into a laboratory guide and a chit-chat robot and conducted subjective experiments, where the subjects talked with robots with four different characters and evaluated their impressions of each character. The results shows that the Role model character was preferred for the task-oriented dialogue of the laboratory guide, but a robot character complementary to the subject personality was preferred for the non-task-oriented dialogue of the speed dating and chit-chat. Empathy in human-robot conversations aims to endow the robot with the ability to comprehend user emotion and experience, and then respond to it appropriately. Generally, empathy is embodied in the aspects of both contextual understanding and affective expression, which occur when there exist content and emotion consistencies between context and response. However, previous studies only focus on either aspect. In this paper, we propose a dual variational generative model (DVG) for empathetic response generation to achieve both. Specifically, we integrate an emotion classifier and a variational autoencoder (VAE) into a dual response and context generative model to learn the emotion and content consistencies efficiently. DVG utilizes VAE to mimic the process of context/response understanding. In addition to the generative model, our model can effectively switch to another retrieval system as a fallback solution. Automatic and human evaluations on Japanese and English EmpatheticDialogue datasets demonstrate the effectiveness of our method for empathetic response generation. In this paper, we investigate the usefulness of an attentive-listening robot in psychiatric daycare, an outpatient treatment program for the rehabilitation of psychiatric disorders. The robot was developed based on counseling techniques, such as repeating words that the user has said. It can also generate backchannels as a listening behavior during user utterance. Conversation experiments have been conducted to evaluate whether the robot can provide effective activities in this setting. The robot attentively listened to 18 daycare attendees talking about their recent memorable events for up to 3 min. The results showed that the conversation increased self-rated arousal. The impressions of the robot showed that talking with the robot was more conversable than with strangers and more useful as a talking partner than a friend. The subjects also had a positive impression about whether they would keep the robot in their homes. A linear regression analysis indicates that the frequency of the robot's assessment responses and backchannels positively affect pleasure improvement. Mussels are a group of bivalves that includes the dominant species of shallow-sea, freshwater, and deep-sea chemosynthetic ecosystems. Mussels cling to various solid underwater surfaces using a proteinaceous thread, called the byssus, which is central to their ecology, physiology, and evolution. Mussels cluster using their byssi to form "mussel beds," thereby increasing their biomass per unit of habitat area, and also creating habitats for other organisms. Clustered mussels actively filter feed to obtain nutrients, but also ingest pollutants and suspended particles; thus, mussels are good subjects for pollution analyses, especially for microplastic pollution. The byssus also facilitates invasiveness, allowing mussels to hitchhike on ships, and to utilize other man-made structures, including quay walls and power plant inlets, which are less attractive to native species. Physiologically, mussels have adapted to environmental stressors associated with a sessile lifestyle. Osmotic adaptation is especially important for life in intertidal zones, and taurine is a major component of that adaptation. Taurine accumulation systems have also been modified to adapt to sulfide-rich environments near deep-sea hydrothermal vents. Atom probe tomography (APT) and transmission electron microscopy (TEM)/scanning transmission electron microscopy (STEM) have been used correlatively to explore atomic-scale local structure and chemistry of the exactly same area in the vicinity of growth front of a long-period stacking ordered (LPSO) phase in a ternary Mg–Al–Gd alloy. It is proved for the first time that enrichment of Gd atoms in four consecutive (0001) atomic layers precedes enrichment of Al atoms so that the formation of Al6Gd8 clusters occurs only after sufficient Al atoms to form Al6Gd8 clusters diffuse into the relevant portions. Lateral growth of the LPSO phase is found to occur by 'ledge' mechanism with the growth habit plane either {1 1 ¯ 00} or {11 2 ¯ 0} planes. The high concentration of vacancies induced by β″ phase formation in Al–Mg–Si alloys has been observed by positron annihilation lifetime spectroscopy. These vacancies differ from quenched-in vacancies. The induced vacancies are generated inside or at the interface of the β″ phase, i.e. the β″ phase contains a high concentration of vacancies. The induced vacancies survive during the β″-to-β′ phase transformation. The migration energy of the induced vacancies is determined to be 1.83 ± 0.11 eV, which is about three times higher than that of vacancies in pure Al. [ABSTRACT FROM AUTHOR]

Character of spoken dialogue systems is important not only for giving a positive impression of the system but also for gaining rapport from users. We have proposed a character expression model for spoken dialogue systems. The model expresses three character traits (extroversion, emotional instability, and politeness) of spoken dialogue systems by controlling spoken dialogue behaviors: utterance amount, backchannel, filler, and switching pause length. One major problem in training this model is that it is costly and time-consuming to collect many pair data of character traits and behaviors. To address this problem, semi-supervised learning is proposed based on a variational auto-encoder that exploits both the limited amount of labeled pair data and unlabeled corpus data. It was confirmed that the proposed model can express given characters more accurately than a baseline model with only supervised learning. We also implemented the character expression model in a spoken dialogue system for an autonomous android robot, and then conducted a subjective experiment with 75 university students to confirm the effectiveness of the character expression for specific dialogue scenarios. The results showed that expressing a character in accordance with the dialogue task by the proposed model improves the user's impression of the appropriateness in formal dialogue such as job interview. A suspension of nonpathogenic Xanthomonas campestris ( nXc) strain AZ98101 or AZ98106 was sprayed on peach trees in biological control tests against bacterial spot caused by Xanthomonas arboricola pv. pruni. Treatment with each strain reduced the number of leaves with spots. Meta-analysis of the results from seven field trials showed an integrated risk ratio (the ratio of leaf spot incidence on inoculated leaves to incidence on control leaves) of 0.46 by treatment with AZ98101 and 0.54 by AZ98106, indicating that leaf spot incidence was significantly reduced by each strain. Each also significantly reduced the incidence of fruit spot. The risk ratio with AZ98101 was significantly lower than that with AZ98106. The population of AZ98101R-1, a rifampicin-resistant mutant of AZ98101, was 6.3 × 10 CFU/g (fresh mass) on leaves and 8.0 × 10 CFU/g on fruit at 12 days after inoculation, but was undetectable (<10 CFU/g) by 24 days. Objectives: Concurrent chemoradiotherapy for regionally advanced stage III non-small cell lung cancer is the standard treatment method. However, the clinical implications of consolidation chemotherapy following chemoradiation have been unclear. Therefore, we conducted a phase II study of concurrent weekly carboplatin plus paclitaxel treatment in combination with radiotherapy followed by vinorelbine monotherapy. The primary endpoint was the 1-year survival rate. Patients and methods: Chemonaive PS 0–1 patients with stage IIIA/B NSCLC were enrolled. During the concurrent chemoradiation phase, patients were treated with weekly paclitaxel 40 mg/m 2 plus carboplatin AUC 2. The primary tumor and involved nodes received 60 Gy in 2-Gy fractions over 6 weeks. During the consolidation phase, vinorelbine 25 mg/m 2 on days 1 and 8 was repeated for three cycles. Results: A total of 40 eligible patients (72.5 % male; median age, 63 years; range 29–74 years) were analyzed for efficacy. Squamous cell carcinoma was the most common histology (47.5 %), and more patients had clinical stage IIIB (55 %) cancer. The average radiation dose was 56.5 Gy, and the average number of carboplatin plus paclitaxel cycles was 4.93. Seventeen patients proceeded to the consolidation chemotherapy phase, and 14 completed three cycles of vinorelbine monotherapy. The objective response rate was 75.0 %, including 1 patient who achieved a complete response. Progression-free survival and overall survival (OS) were 46 weeks [95 % confidence interval (CI) 31–64 weeks] and 110 weeks (95 % CI 90–184 weeks), respectively. The OS rate at 1 and 2 years was 85.0 % (95 % CI 69.6–93.0 %) and 53.9 % (95 % CI 37.1–68.0 %), respectively. Conclusion: Concurrent chemoradiation with weekly carboplatin and paclitaxel followed by vinorelbine consolidation is effective for stage III non-small cell lung cancer and shows a generally mild toxicity profile. Graft unions of nursery stock of grapevine ( Vitis vinifera L.) collected in Japan yielded non-pathogenic strains of Agrobacterium. On the basis of classic diagnostic tests, a sequence analysis and a previously reported multiplex PCR method, the non-pathogenic strains ARK-1, ARK-2 and ARK-3 were identified as Agrobacterium vitis. Stems of grapevine seedlings were inoculated with both a cell suspension of seven mixed strains of A. vitis (Ti) as a pathogen and one of a new strain or A. vitis strain VAR03-1, one of the biological control agents against crown gall previously reported, as competitors to assay the suppression of tumour formation caused by the pathogen. In a test with a 1:1 cell ratio of pathogen/nonpathogen, strains ARK-1, ARK-2 and ARK-3 reduced the tumour incidence.. In particular, strain ARK-1 was strongest at inhibiting tumour formation in this study. Strain ARK-1 established populations on roots of grapevine tree rootstock and persisted on roots for a year. ARK-1, ARK-2 and ARK-3 did not produce a halo of inhibition against A. vitis (Ti) strain on YMA medium. Moreover, strain ARK-1 did not reduce tumour incidence on the stems of grapevine when ARK-1 was dead or only culture filtrate was used. This result indicates the possibility that these new strains inhibit grapevine crown gall in planta by a different mechanism other than VAR03-1. In particular, one of the new strains, named ARK-1, was most effective in inhibiting tumour formation on grapevine and appears to be a promising new agent to control grapevine crown gall. The meat quality of farmed yellowtail Seriola quinqueradiata fed on extruded pellets (EP) containing 0.5% (v/v) red pepper (experimental group) was compared with yellowtail of the same age fed on EP (control group). In 1-year-old yellowtail, the crude lipid content of the dorsal muscle of the experimental group tended to be lower than that of the control group. In contrast, there was no difference in the lipid content of the dorsal muscle between the control group and the experimental group in 2-year-old yellowtail. The muscle texture of the experimental group was significantly firmer than that of the control group, with the effect of red pepper unrelated to fish age and lipid content. Color change of red muscle of the experimental group was significantly lower than that of the control group, and the content of thiobarbituric acid-reactive substances in the red muscle was significantly lower in the experimental group than in the control group. These results are the first to demonstrate that the inclusion of red pepper in the diet is able to reduce the loss of muscle texture firmness and to slow down color change in red muscle of yellowtail. Abstract: The natriuretic peptide (NP) family consists of at least seven members; cardiac ANP, BNP and VNP and brain CNPs (CNP1–4). Phylogenetic and comparative genomic analyses showed that CNP4 is the ancestral molecule of the family, from which CNP3 and CNP1/2 were duplicated in this order, and that the three cardiac NPs were generated from CNP3 by tandem duplication. Seven members existed at the divergence of ray-finned fishes and lobe-finned fishes (tetrapods), but some of the NP genes have disappeared during the course of evolution. In ray-finned fishes, all three cardiac NPs exist in chondrostei and some migratory teleost species, but VNP is generally absent and ANP is absent in a group of teleosts (Beloniformes). In tetrapods, ANP and BNP are present in mammals and amphibians, but ANP is usually absent in reptiles and birds. Thus, BNP is a ubiquitous cardiac NP in bony fishes and tetrapods though elasmobranchs and cyclostomes have only CNP3/4 as a cardiac NP. Functional studies indicate that cardiac NPs are essential Na+-extruding hormones throughout vertebrates; they play critical roles in seawater (SW) adaptation in teleosts, while they are important volume-depleting hormones in mammals as water and Na+ are regulated in parallel in terrestrial animals. In mammals, cardiac NPs become prominent in pathological conditions such as heart failure where they are used in diagnosis and treatment. Although the functional role of BNP has not yet been fully elucidated compared with ANP in non-mammalian vertebrates, it appears that BNP plays pivotal roles in the cardiovascular and body fluid regulation as shown in mammals. ANP has previously been recognized as the principal cardiac NP in mammals and teleosts, but comparative studies have revealed that BNP is the only cardiac NP that exists in all tetrapods and teleosts. A new decomposition system for volatile organic compounds (VOCs), based on dielectric barrier discharges combined with zeolite honeycomb sheets, has been developed. A discharge element was made of layered flat metal sheets covered with mica sheets separated by 2 mm, in which corrugated (honeycomb-shaped) ceramic sheets embedded with zeolites were inserted. This element was shown to satisfy the requirements of durability and flexibility to meet various sizes. Barrier discharges were investigated using this discharge element over wide ranges of operating conditions, in terms of the power consumption and decompositions of various types of VOCs. By selecting suitable operating scenarios, it was found that decomposition of more than 90% of most VOCs was possible, at reasonable power consumption and without electrode contamination by decomposition products. In 2005, characteristic symptoms of crown gall on grapevines ( Vitis vinifera L. cv. Muscat of Alexandria, and cv. Seto Giants) were observed in a commercial greenhouse-orchard in Okayama Prefecture, Japan. Isolations from diseased tissues consistently yielded bacterial colonies that were white, glistening, and produced abundant polysaccharide on potato semi-synthetic agar (PSA) medium. Ten representative isolates were chosen for further characterization. A multiplex polymerase chain reaction (PCR) assay showed these strains were not Rhizobium vitis but did possess a Ti plasmid. The bacteriological characteristics of the isolates corresponded well with R. radiobacter. The almost complete 16S ribosomal DNA sequences of isolates AT06-1 and AT06-2, selected from 10 grapevine isolates, were determined and corresponded to sequences of R. radiobacter. The pathogenicity of the isolates was tested on young grapevine and tomato ( Lycopersicon esculentum Mill.) plants. Gall symptoms developed on both plant species after inoculation, and bacteria with the same colony morphology as those inoculated were reisolated. Based on these results, the isolates were identified as R. radiobacter (Ti). This report is the first of the occurrence of R. radiobacter (Ti) on grapevine in Japan. Phylogenetic analyses using the partial nucleotide sequences of virC operon located on a Ti plasmid showed that the isolate of R. radiobacter (Ti) isolated from grapevine and some strains of R. vitis (Ti) belonged to the same monophyletic group, which differed from the groups of R. radiobacter (Ti) isolated from plants other than grapevine and of the majority of R. vitis (Ti) strains isolated from grapevine. Abstract: The renin–angiotensin system is an enzyme-linked hormonal cascade that plays an important role in body fluid and cardiovascular regulation. The system is initiated by the action of renin on the precursor protein, angiotensinogen (AGT), whose sequence information is scarce because of its high variability among species. In the present study, we cloned AGT in chondrichthyans (elasmobranchs: Triakis scyllium, Dasyatis akajei, Leucoraja erinacea and a holocephalan: Callorhinchus milii). Homology was low among AGTs thus far identified; 25–28% between elasmobranchs and tetrapods and 33–61% even within chondrichthyans. All chondrichthyan angiotensin (ANG) II’s have a unique Pro3 instead of Val3 as seen in all other species. In addition, holocephalan ANG II has an unusual His4 instead of Tyr4. In addition, and the N-terminal amino acid, which is usually Asp1 in tetrapods and Asn1 in fishes, was highly variable (Asp, Asn or Tyr) in chondrichthyans. Molecular phylogenetic analysis showed that chondrichthyan AGT precursors are clustered into a group separated from those of tetrapods and teleosts. The AGT gene was most abundantly expressed in the liver, followed by the kidney, interrenal tissue and rectal gland of Triakis where biological actions of ANG II have been demonstrated. Collectively, we identified diversified AGT genes for the first time in chondrichthyes and showed that their ANG II’s have unique amino acid residues at positions 1, 3 and 4. High variability of ANG II sequences in chondrichthyans is discussed in relation to their unique regulatory mechanisms such as urea-based osmoregulation. [Copyright &y& Elsevier]

A virus that caused a distinct yellow mosaic was isolated in Okayama, Japan from Chinese cabbage ( Brassica rapa L., Pekinensis group). The virus, with spherical particles ca. 28 nm in diameter, was mechanically transmissible only to cruciferous species. From the host range, characteristic morphology of virus particles, serology and sequence analysis of coat protein gene, the causal virus was identified as Turnip yellow mosaic virus (TYMV). Seed transmission of TYMV at 0–2.2% in Chinese cabbage was confirmed. This report is the first of TYMV from Chinese cabbage and in Japan. [ABSTRACT FROM AUTHOR]

Abstract: Various invertebrates inhabiting hydrothermal vents possess sulfur-oxidizing bacteria in their tissues; however, the mechanisms by which toxic sulfides are delivered to these endosymbionts remain unknown. Recently, detoxification of sulfides using thiotaurine, a sulfur-containing amino acid, has been suggested. In this study, we propose the involvement of a taurine transporter in sulfide detoxification in the deep-sea mussel Bathymodiolus septemdierum by demonstrating: (i) the abundance of its mRNA in the gill; (ii) its activity under a wide range of salinities; (iii) its low Michaelis constant value in taurine transportation; and (iv) its affinity for thiotaurine and the thiotaurine precursor, hypotaurine. In Vitis labruscana ‘Pione’ and ‘Kyoho’, 57 grapevines with rugose wood (RW) symptoms and 31 asymptomatic grapevines were obtained from vineyards in Hiroshima and Okayama prefectures. We surveyed vines for RW-associated viruses and other viruses by reverse transcription-polymerase chain reaction with virus-specific primers and degenerate primers for simultaneous detection of viruses associated with RW or leafroll disease. Rupestris stem pitting-associated virus (RSPaV) was detected from all grapevines with RW symptoms and from eight asymptomatic grapevines. Grapevine leafroll-associated virus-1, -2, -3, Grapevine virus B, Grapevine fleck virus, and Grapevine asteroid mosaic-associated virus were detected from some of the grapevines tested. RSPaV isolates detected were clustered in four groups by phylogenetic analysis using the nucleotide sequences of coat protein gene. Abstract: The natriuretic peptide (NP) family consists of multiple subtypes in teleosts, including atrial, B-type, ventricular, and C-type NPs (ANP, BNP, VNP, CNP-1–4, respectively), but only ANP, BNP, CNP-3, and CNP-4 have been identified in tetrapods. As part of understanding the molecular evolution of NPs in the tetrapod lineage, we identified NP genes in the chicken genome. Previously, only BNP and CNP-3 have been identified in birds, but we characterized two new chicken NP genes by cDNA cloning, synteny and phylogenetic analyses. One gene is an orthologue of CNP-1, which has only ever been reported in teleostei and bichir. The second gene could not be assigned to a particular NP subtype because of high sequence divergence and was named renal NP (RNP) due to its predominant expression in the kidney. CNP-1 mRNA was only detected in brain, while CNP-3 mRNA was expressed in kidney, heart, and brain. In the developing embryo, BNP and RNP transcripts were most abundant 24h post-fertilization, while CNP mRNA increased in a stage-dependant manner. Synthetic chicken RNP stimulated an increase in cGMP production above basal level in chicken kidney membrane preparations and caused a potent dose-dependant vasodilation of pre-constricted dorsal aortic rings. From conserved chromosomal synteny, we propose that the CNP-4 and ANP genes have been lost in chicken, and that RNP may have evolved from a VNP-like gene. A nonpathogenic strain of Agrobacterium vitis VAR03-1 was tested as a biological control agent against crown gall of grapevine ( Vitis vinifera L.). A mixture of the nonpathogenic strain VAR03-1 and a tumorigenic strain G-Ag-27 of A. vitis at cell ratios of 1 : 1, 3 : 1, 9 : 1, and 99 : 1 significantly inhibited gall formation and size on stems of tomato ( Lycopersicon esculentum Mill.). Strain VAR03-1 also inhibited gall formation on stems of both tomato and grapevine at a 1 : 1 cell ratio with several tumorigenic A. vitis strains isolated from different fields of grapevine in Japan. In biological control tests, when roots of grapevine and tomato seedlings were soaked in a cell suspension of strain VAR03-1 for 24 h before a 1-h soaking in a cell suspension of the pathogen and subsequent planting in pots of infested soil, strain VAR03-1 significantly reduced the incidence of gall formation on both plants. Abstract: The calcitonin gene-related peptide (CGRP) family is composed of CGRP, amylin and adrenomedullin (AM) in mammals. In teleost fish, AM forms an independent subfamily of five members (AM1–5), which inspired us to trace the evolutionary history of the CGRP family throughout vertebrates by comparative genomic approach. Linkage mapping and synteny analyses of the CGRP family genes in medaka, Oryzias latipes, revealed that AM1/CGRP, AM2/amylin, and AM5 genes were located on respective proto-chromosomes before the divergence of teleost lineage. In teleost fish, additional whole genome duplication generated AM1/4, CGRP1/2, AM2/3, but one of the duplicated amylin and AM5 genes was silenced. The following report is a record of the results from the reconstruction of tire blow-out accidents on highway roads. PC-Crash traffic accident simulation software was used to reconstruct the accidents based on the tire marks left on the road surface and the damages on the vehicles at the scene of the accidents. The results of the simulations are quite comparable to trails at the accident scenes. Graft unions of nursery stock of grapevine ( Vitis vinifera L.) collected in Japan yielded pathogenic and nonpathogenic strains of Agrobacterium. On the basis of classical diagnostic tests, a sequence analysis, and a multiplex polymerase chain reaction method previously reported, the pathogenic strain was identified as Agrobacterium tumefaciens biovar 3, whereas the nonpathogenic strains were assigned to Agrobacterium radiobacter biovar 3. Stems of tomato ( Lycopersicon esculentum Mill.) seedlings were inoculated with both A. tumefaciens biovar 3 strain G-Ag-27 as a pathogen and one of the control strains isolated from grapevine or A. radiobacter biovar 2 strain K84 as competitors to assay the suppression of gall formation caused by the pathogen. In a test with a 1 : 1 pathogen/nonpathogen cell ratio, all A. radiobacter biovar 3 strains reduced gall incidence and size compared to that of the positive control inoculated only with the pathogen. Strain VAR03-1 was especially effective in reducing the incidence of gall formation on grapevine and reduced gall size by 84%–100% of those on the positive control. Many tested nonpathogenic biovar 3 strains were bacteriocinogenic, causing an inhibition zone against A. tumefaciens biovar 3 strains on YMA medium. To study the mechanism of faster growth of the sculpin Cottus kazika in seawater (SW) than in fresh water (FW), we transferred 120-day-old juveniles from FW to SW, 1/3 SW or FW, and survival, growth and levels of transcripts of growth hormone (GH) gene in the pituitary and insulin-like growth factor I (IGF-I) gene in the liver were examined 20 and 40 days after transfer. The survival rates of fish reared in SW and 1/3 SW were higher than the FW controls. The growth was fastest in SW and slowest in the controls. IGF-I gene transcripts increased in SW-reared fish 40 days after transfer compared with the controls, while GH gene transcripts did not. Juveniles of 1 year of age were transferred from FW to SW or 1/3 SW, and GH and IGF-I gene transcripts were compared after 48 h. In this short-term experiment, the level of GH gene transcripts was higher in fish reared in SW and 1/3 SW, while no difference was found in the IGF-I gene transcripts. Thus, the GH gene transcripts increased only transiently after SW transfer but the IGF-I gene transcripts increased gradually for 40 days, suggesting that the long-term increase in IGF-I gene expression is involved in the enhanced growth in SW and GH may play an initiation role for growth enhancement. Losses of neutral beam (NB) injected fast ions from the confinement region of a field-reversed configuration (FRC) with a strong magnetic mirror are numerically analyzed for parameters relevant to NB injection experiments on the FIX (FRC injection experiment) device [T. Asai et al., Phys. Plasmas 7, 2294 (2000)]. Ionization processes of beam particles are calculated by the Monte Carlo method. The confinement of beam ions is discussed with the concept of accessible regions that restrict the ion excursion and are determined from two constants of motion, the kinetic energy and canonical angular momentum, in the case of an axisymmetric and a steady state FRC without an electrostatic field. From the calculation of the accessible regions, it is found that all the fast ions suffer from the orbit loss on the wall surface and/or the end loss. Single particle orbits are also calculated to find a difference of confinement properties from the results by employing the accessible regions. The magnetic moment is observed to show nonadiabatic motions of the beam ions, which cause a gradual orbit loss on the wall even in a case that a strong magnetic mirror is applied. The results show that the correlation of the magnetic moment disappears as the fast ions experience the density gradient around the separatrix surface and the field-null points. © 2004 American Institute of Physics. We have identified cDNA encoding a new member of the adrenomedullin (AM) family, AM2, for the first time in mammals (mouse, rat and human). The predicted precursor carried mature AM2 in the C-terminus, which had an intramolecular ring formed by an S–S bond and a possibly amidated C-terminus. Phylogenetic analyses clustered AM2 and AM into two distinct but closely related groups. Similarity of exon–intron structure and synteny of neighboring genes showed that mammalian AM2 is an ortholog of pufferfish AM2 and a paralog of mammalian AM. AM2 mRNA was expressed in submaxillary gland, kidney, stomach, ovary, lymphoid tissues and pancreas of mice, but not in adrenal and testis. Intravenous injection of synthetic mature AM2 decreased arterial pressure more potently than AM, and induced antidiuresis and antinatriuresis in mice. These results show that at least two peptides, AM and AM2, comprise an adrenomedullin family in mammals, and that AM2 may play pivotal roles in cardiovascular and body fluid regulation. Japanese medaka (Oryzias latipes) is a freshwater (FW) teleost that is popular throughout the world for laboratory use. In this paper, we discuss the utility of Japanese medaka and related species for studying mechanisms of seawater (SW) adaptation. In addition to general advantages as an experimental animal such as their daily spawning activity, transparency of embryos, short generation time and established transgenic techniques, Japanese medaka have some adaptability to SW unlike the strictly stenohaline zebrafish (Danio rerio). Since other species in the genus Oryzias exhibit different degrees of adaptability to SW, comparative studies between Japanese medaka, where molecular-biological and genetic information is abundant, and other Oryzias species are expected to present varying approaches to solving the problems of SW adaptation. We introduce some examples of interspecies comparison for SW adaptabilities both in adult fish and in embryos. Oryzias species are good models for evolutionary, ecological and zoogeographical studies and a relationship between SW adaptability and geographic distribution has been suggested. Medaka fishes may thus deliver new insights into our understanding of how fish have expanded their distribution to a wide variety of osmotic environments. [Copyright &y& Elsevier]

Adrenomedullin (AM) is a multifunctional peptide known to form a hormone family with calcitonin gene-related peptide (CGRP) and amylin. We have cloned five distinct AM cDNAs from the pufferfish, Takifugu rubripes, and named them TrAM-1, -2, -3, -4, and -5. Judging from the deduced precursor sequences and processing pattern of the C-terminal mature peptides, TrAMs may be divided into at least two groups; AM-2 and -3, and AM-1, -4, and possibly -5. Phylogenetic analysis of the mature peptides, exon–intron structure of their genes, and tissue distribution of their mRNA also support this classification. TrAM-1 and -4 were ubiquitously expressed in various tissues including the kidney and interrenal (adrenal homolog) as in the case of mammalian AM, while TrAM-2 and -3 were expressed most abundantly in the brain followed by the vascular tissues. Synteny of the genes around AM gene showed that TrAM-1 is the ortholog of mammalian AM. The presence of a PAMP-like sequence in the prosegment of TrAM-1 also supports this notion. Multiple AMs were also detected in another pufferfish, Tetraodon nigroviridis, and in zebrafish, Danio rerio. The present study shows for the first time the presence of a novel AM family in teleost fish that is independent from CGRP and amylin, which further suggests the possible existence of multiple AMs in mammals. Natriuretic peptides (NPs) are major cardiovascular and osmoregulatory hormones in vertebrates. Although tetrapods generally have three subtypes, atrial NP (ANP), B-type NP (BNP), and C-type NP (CNP), some teleosts lack BNP, and sharks and hagfish have only one NP. Thus, NPs have diverged during fish evolution, possibly reflecting changes in osmoregulatory systems. In this study, we found, by cDNA cloning, four distinct CNPs (1 through 4) in the medaka (Oryzias latipes) and puffer fish (Takifugu rubripes), although to our knowledge no more than two CNPs have been isolated from a vertebrate species. Predicted mature CNP-1 was most similar, and CNP-4 was most dissimilar, to mammalian CNPs. However, synthetic CNP-4 most potently activated OIGC1, a medaka CNP-specific receptor (NPR-B) expressed in cultured cells, whereas CNP-1 and CNP-3 most activated OIGC7 and OIGC2, two medaka homologues of the ANP/BNP receptor (NPR-A), respectively. Linkage mapping in medaka followed by comparative genomic analyses among fishes and humans located four CNP genes in separate medaka chromosomes corresponding to human chromosomes 1, 2, 12, and 17. From conserved synteny, the following evolutionary history of NPs was evoked: (i) four CNP genes were generated by chromosomal duplications before the divergence of elasmobranchs; (ii) the CNP-3 gene generated ANP and BNP genes through tandem duplication before the divergence of tetrapods and teleosts; (iii) CNP-1 and -2 genes were retained in the teleost lineage but not in the tetrapod lineage; (iv) the CNP-3 gene disappeared from the tetrapod lineage after divergence of amphibians; and (v) the CNP-4 gene is retained in humans as CNP. The four-spine sculpin Cottus kazika is a euryhaline teleost, in which faster growth in seawater (SW) and freshwater (FW) has been reported. In this study, cDNA clones encoding growth hormone (GH) and insulin-like growth factor I (IGF-I) were isolated from this species to examine the involvement of the GH/IGF-I axis in osmotic adaptation. The amino acid sequence of GH predicted from cDNA was highly similar to those of other fish species, 92% to Sparus aurata, 67% to Paralichthys olivaceus, and 63% to Oncorhynchus keta. The predicted sequence of IGF-I was also exhibited high similarity to those of other fishes, 97% to Myxocephalus scorpius, 95% to P. olivaceus, and 81% to O. keta. Tissue distribution of GH and IGF-I mRNA in fish reared in FW and SW was examined by reverse transcription-polymerase chain reaction (RT-PCR). The GH mRNA was detected only in the pituitary gland. The major site of IGF-I mRNA expression was the liver while minor signals were detected in various tissues including the pituitary, gill, fin, heart, spleen, intestine, and kidney. The expression level of GH mRNA in the pituitary was not different between FW- and SW-reared fish. However, the level of IGF-I mRNA in the liver of SW-reared fish was significantly higher than that of FW-reared fish. Natriuretic peptides (NPs) play important roles in osmoregulatory and cardiovascular systems of vertebrates. For functional studies of NPs, rainbow trout (Oncorhynchus mykiss), a euryhaline fish, is an interesting model. The information on homologous NPs of salmonid fish is, however, still incomplete with respect to C-type NP (CNP). In this study, we isolated cDNAs encoding the precursor of CNP from the brain of trout. Predicted mature CNP (CNP-22) sequence was identical to that of killifish Fundulus heteroclitus, and only one amino acid was different from that of the eel Anguilla japonica, demonstrating a greater conservation among different teleost species than is found with atrial NP (ANP) and ventricular NP (VNP). While the preprosegment of trout CNP retained 57% similarity to the eel sequence, similarities were low to those of sharks and tetrapods. The major site of expression identified by RT-PCR was the brain with minor expression in the atrium. The putative mature CNP-22 was synthesized and its biological activity was compared with other trout NPs (ANP and VNP) using trout ventral aorta, efferent branchial and celiacomesenteric arteries and anterior cardinal vein in vitro. A new position-sensitive γ-ray detector to be used in a two-dimensional angular correlation of positron annihilation radiation (2D-ACAR) apparatus has been developed. It consists of 36 compact position-sensitive photomultiplier tubes (PS-PMT: HAMAMATSU R5900-00-C8), a light guide, and 2676 Bi4Ge3O12 (BGO) scintillator pieces of size 2.6 mm×2.6 mm×18 mm. A high detection efficiency for 511 keV γ-ray is achieved with the length of BGO scintillators used. The detection area is about 160 mm×160 mm. The 288 anode outputs of the PS-PMTs are wired and connected to resistor chains from which 16 outputs (8 outputs each along the X and Y directions) are taken to identify the incident position of the γ-ray. The spatial resolution is about 3 mm (FWHM). The timing signal taken from the last dynodes of the PS-PMTs gives a timing resolution of 7.7 ns (FWHM) for 511 keV positron annihilation γ-rays. Nematodes belonging to the family Anisakidae including Anisakis simplex and Pseudoterranova decipiens are known to cause anisakiasis when their live larvae are ingested by humans. We estimated the possibility of anisakid infection to salmonids, farmed in sea net-pens at Onagawa Bay, Miyagi, Japan, in 1992, 1998 and 1999, by direct examination of the edible muscle and examination of the contents of the alimentary canal. From direct examination of the muscle, no nema-tode was found in the 249 farmed coho salmon Oncorhynchus kisutch and 40 farmed rainbow trout Oncorhynchus mykiss. In contrast, third-stage larvae of Anisakis simplex were found in seven of 14 wild coho salmon caught in Russia and all the 40 wild chum salmon Oncorhynchus keta caught at Nemuro and Kesennuma in 1998. The stomach and intestines of 521 farmed coho salmon and 40 farmed rainbow trout were examined carefully for the existence of possible carrier organisms such as crustaceans, fish or squid. Such carrier organisms were not found in the stomach and intestines of farmed fish. Thus, we conclude that the possibility of anisakid infection is very low in farmed salmonids. We have investigated a post-annealing effect on microscopic structural changes of kink boundaries (KBs), which have been initially introduced by a hot-extrusion of an Mg-Zn-Y alloy containing long period stacking/order (LPSO) phase, based on scanning transmission electron microscopy (STEM) and atom-probe tomography (APT). STEM observations show that most of the KBs appear to be segmented into several submicron-scale, each of which is composed of arrays of extended basal (a -) dislocations that are geometrically necessary to account for the LPSO crystalline rotations. Atomic-scale STEM/APT observations reveal significant reconstructions of these extended dislocation-core structures, which occur at the solute-enriched face-centered cubic layers in the LPSO structures and forms local hexagonal close-packed Mg region with distinct solute depletions. These recovery-like features indeed enhance the thermal stability of the KBs, and the corresponding driving force can be reasonably described according to disclinations, whose elastic energy is defined by crystal rotations and the relevant dipole distances. • Microscopic structures of kink boundaries in unique long-period structured Mg-Zn-Y alloys have been investigated. • Microscopic segmentations of kink boundaries have been promoted during the post-annealing of the hot-extruded specimen. • Dislocation-core structures at kink boundaries have been significantly reconstructed with distinct solute redistributions. • Driving force to promote the observed recovery features can be reasonably described according to disclinations. Objective Patients and Methods Results Conclusion To report the outcomes of repeat biopsies, metastasis and survival in the Prostate Cancer Research International: Active Surveillance (PRIAS)‐JAPAN study, a prospective observational study for Japanese patients, initiated in 2010.At the beginning, inclusion criteria were initially low‐risk patients, prostate‐specific antigen (PSA) density (PSAD) <0.2, and ≤2 positive biopsy cores. As from 2014, GS3+4 has also been allowed for patients aged 70 years and over. Since January 2021, the age limit for Gleason score (GS) 3 + 4 cases was removed, and eligibility criteria were expanded to PSA ≤20 ng/mL, PSAD <0.25 nd/mL/cc, unlimited number of positive GS 3 + 3 cores, and positive results for fewer than half of the total number of cores for GS 3 + 4 cases if magnetic resonance imaging fusion biopsy was performed at study enrolment or subsequent follow‐up. For patients eligible for active surveillance, PSA tests were performed every 3 months, rectal examination every 6 months, and biopsies at 1, 4, 7 and 10 years, followed by every 5 years thereafter. Patients with confirmed pathological reclassification were recommended for secondary treatments.As of February 2024, 1302 patients were enrolled in AS; 1274 (98%) met the eligibility criteria. The median (interquartile range) age, PSA level, PSAD, and number of positive cores were 69 (64–73) years, 5.3 (4.5–6.6) ng/mL, 0.15 (0.12–0.17) ng/mL, and 1 (1–2), respectively. The clinical stage was T1c in 1089 patients (86%) and T2 in 185 (15%). The rates of acceptance by patients for the first, second, third and fourth re‐biopsies were 83%, 64%, 41% and 22%, respectively. The pathological reclassification rates for the first, second, third and fourth re‐biopsies were 29%, 30%, 35% and 25%, respectively. The 1‐, 5‐ and 10‐year persistence rates were 77%, 45% and 23%, respectively. Six patients developed metastasis, and one patient died from prostate cancer.Pathological reclassification was observed in approximately 30% of the patients during biopsy; however, biopsy acceptance rates decreased over time. Transurethral procedures such as direct vision internal urethrotomy and urethral dilation have been the traditional treatments for urethral strictures. However, transurethral procedures are associated with high recurrence rates, resulting in many uncured cases and prompting major international urological societies to recommend urethroplasty as the standard treatment owing to its high success rate. In contrast, many Japanese general urologists have little doubts about treating urethral strictures with transurethral treatment. Therefore, urethral stricture treatments in Japan are not in line with those used in other countries. To address this, the Trauma, Emergency Medicine, and Reconstruction Subcommittee of the Japanese Urological Association has developed guidelines to offer standardized treatment protocols for urethral stricture, based on international evidence and tailored to Japan's medical landscape. These guidelines target patients with a clinically suspected urethral stricture and are intended for urologists and general practitioners involved in its diagnosis and treatment. Following the Minds Clinical Practice Guideline Development Manual 2020, the committee identified eight critical clinical issues and formulated eight clinical questions using the “patient, intervention, comparison, and outcome” format. A comprehensive literature search was conducted. For six clinical questions addressed by the existing guidelines or systematic reviews, the level of evidence was determined by qualitative systematic reviews. Quantitative systematic reviews and meta‐analyses were performed for the two unique clinical questions. The recommendation grades were determined using the Delphi method and consensus by the committee. Objectives: Immune checkpoint inhibitors and enfortumab vedotin have opened new avenues for sequential treatment strategies for locally advanced/metastatic urothelial carcinoma (la/mUC). In the pre‐enfortumab vedotin era, many patients could not receive third‐line treatment owing to rapid disease progression and poor general status. This study aimed to analyze real‐world sequential treatment practices for la/mUC in Japan, with a focus on patients who do not receive third‐line treatment. Methods: We analyzed data for 1023 la/mUC patients diagnosed between January 2020 and December 2021 at 54 institutions from a Japanese nationwide cohort. Results: At the median follow‐up of 28.5 months, the median overall survival from first‐line initiation for 905 patients who received systemic anticancer treatment was 19.1 months. Among them, 81% and 32% received second‐ and third‐line treatment. Notably, 52% had their treatment terminated before the opportunity for third‐line treatment. Multivariate logistic regression analysis revealed that low performance status (≥1), elevated neutrophil‐to‐lymphocyte ratio (≥3), and low body mass index (<21 kg/m2) at the start of first‐line treatment were independent risk factors for not proceeding to third‐line treatment (p = 0.0024, 0.0069, and 0.0058, respectively). In this cohort, 33% had one of these factors, 36% had two, and 15% had all three. Conclusions: This study highlights the high frequency of factors associated with poor tolerance to anticancer treatment in la/mUC patients. The findings suggest the need to establish optimal sequential treatment strategies, maximizing efficacy within time and tolerance constraints, while concurrently providing strong supportive care, considering immunological and nutritional aspects. A segment of Mytilus galloprovincialis foot protein 3 (Mgfp-3) cDNA was amplified by means of reverse-transcription (RT)/PCR with degenerate primers. The 5' and 3' regions of the cloned segment were amplified by means of rapid amplification of eDNA ends. The 5'-region clones had almost identical nucleotide sequences, but two sequences were found among 3'-region clones. The Mgfp-3 coding region was amplified between a 5' untranslated sequence and one of two 3' untranslated sequences. Two cDNA clones which encoded variants Mgfp-3A and Mgfp-3B. were isolated. These two clones encoded proteins with 70 and 77 amino acid residues, of which the first 24 residues are predicted to be signal peptides. The existence of additional variants was suggested by the sequences of other clones. Thus, it was suggested that Mgfp-3 genes constitute a gene family. RT/PCR of RNA from developing larvae indicates that Mgfp-3 genes are transcribed after settlement. RT/PCR of RNA from major organs and in situ hybridization of the foot indicate that Mgfp-3 genes are transcribed in a limited part of the foot, i.e., the phenol gland and a distal part of the accessory gland. Atom-probe technique was applied to analyze three-dimensional dopant distribution in Si substrate of metal-oxide-semiconductor field effect transistor (MOSFET) structure. As a result, three-dimensional As atom distribution implanted in Si was obtained. The quantification of the As atom distribution in a depth direction was confirmed as compared with the one-dimensional distribution measured by secondary ion mass spectroscopy. Moreover, monolayer segregation of As atoms at the interface between gate oxide and Si substrate was clearly observed.
