Breast cancer continues to be one of the main causes of morbidity and mortality globally and was the leading cause of cancer death in women in Spain in 2020. Early diagnosis is one of the most effective methods to lower the incidence and mortality rates of breast cancer. The human metalloproteinases (MMP) mainly function as proteolytic enzymes degrading the extracellular matrix and plays important roles in most steps of breast tumorigenesis. This retrospective cohort study shows the immunohistochemical expression levels of MMP-1, MMP-2, MMP-3, and MMP-9 in 154 women with breast cancer and 42 women without tumor disease. The samples of breast tissue are assessed using several tissue matrices (TMA). The percentages of staining (≤50%–&gt, 50%) and intensity levels of staining (weak, moderate, or intense) are considered. The immunohistochemical expression of the MMP-1-intensity (p = 0.043) and MMP-3 percentage (p = 0.018) and intensity, (p = 0.025) present statistically significant associations with the variable group (control–case), therefore, expression in the tumor tissue samples of these MMPs may be related to the development of breast cancer. The relationships between these MMPs and some clinicopathological factors in breast cancer are also evaluated but no correlation is found. These results suggest the use of MMP-1 and MMP-3 as potential biomarkers of breast cancer diagnosis.

Most of the snakebite envenomations in Central and South America are caused by species belonging to Bothrops genus. Their venom is composed mainly by zinc-dependent metalloproteinases, responsible of the hemorrhage characteristic of these envenomations. The aim of this study was to determine the inhibitory ability of ten flavonoids on the in-vitro proteolytic activity of Bothrops atrox venom and on the hemorrhagic, edema-forming and myonecrotic activities of Batx-I, the most abundant metalloproteinase isolated from this venom. Myricetin was the most active compound, exhibiting an IC 50 value of 150 μ M and 1021 μ M for the inhibition of proteolytic and hemorrhagic activity, respectively. Independent injection experiments, with a concentration of 1600 μ M of myricetin administered locally, immediately after toxin injection, demonstrated a reduction of 28 ± 6 % in the hemorrhagic lesion. Additionally, myricetin at concentrations 800, 1200 and 1600 μ M promoted a reduction in plasma creatine kinase activity induced by Batx-I of 21 ± 2 % , 60 ± 5 % and 63 ± 2 % , respectively. Molecular dynamics simulations coupled with the adaptive biasing method suggest that myricetin can bind to the metalloproteinase active site via formation of hydrogen bonds between the hydroxyl groups 3’, 4’ and 5’ of the benzyl moiety and amino acid Glu143 of the metalloproteinase. The hydroxyl substitution pattern of myricetin appears to be essential for its inhibitory activity. Based on this evidence, myricetin constitutes a candidate for the development of inhibitors to reduce local tissue damage in snakebite envenomations.

La capa híbrida es lograda mediante la desmineralización superficial de la dentina a través de la aplicación de acondicionadores acídicos y su posterior infiltración con resinas adhesivas, para conseguir una estructura compuesta por fibras de colágeno tipo I y proteoglicanos envueltos por cadenas de polímeros (Toledano et al, 2012). La matriz de colágeno parcialmente desmineralizada constituye un andamio adecuado para la remineralización en presencia de materiales bioactivos (Osorio et al, 2012). Sin embargo, en la base de la capa híbrida yace una capa de volumen reducido de colágeno desprotegido -dada la inadecuada infiltración resinosa así como también la ausencia de minerales intrafibrilares- y desmineralizado -tanto por acción del ácido fosfórico como por acción del ácido etilen diamino tetra acético (EDTA) - que puede sufrir degradación hidrolítica (Toledano et al, 2012; 2014 b) por acción de metaloproteinasas endógenas de la matriz (MMP) (Hebling et al, 2005). Esta actividad degradativa enzimática reduce el éxito clínico de las restauraciones adheridas debido a la pérdida de integridad en la interfaz de adhesión (Toledano et al, 2014 a). La barrera crítica al progreso en la adhesión a dentina, esto es el agua entrampada en los compartimentos intrafibrilares de las fibras de colágeno, no puede ser eliminada luego que éste es impregnado con resinas polimerizadas. Una matriz de colágeno llena de agua continúa siendo una matriz débil, independientemente de lo bien que sea preservada su integridad (Brackett et al, 20 11). La deshidratación progresiva de las fibras de colágeno a través del depósito de apatita intrafibrilar ocurre naturalmente en la mineralización de los tejidos duros y protege la matriz orgánica de la degradación durante un tiempo muy prolongado, mucho mayor al que se consigue cuando se utilizan inhibidores de las MMP o agentes reticuladores del colágeno (Brackett et al, 2011). La estabilidad e integridad de las fibrillas de colágeno dentro de la capa híbrida es crucial para el mantenimiento de la efectividad de la unión a lo largo del tiempo (Breschi et al, 2008). El sustrato dentinario puede ser modificado mediante el uso de agentes reticuladores cuyo fin es promover la formación de enlaces exógenos que permiten aumentar la resistencia de la red de colágeno (Bedran-Russo et al, 2007; 2008) y promover su remineralización (Osorio et al, 2005), de forma similar a como ocurre durante la mineralización de los tejidos duros. Este mecanismo protector debe ser recapitulado para la supervivencia a largo plazo de las uniones resinadentina (Brackett et al, 2011). En esta primera entrega se analizarán los inhibidores de las MMP.
