1. Modeling the decomposition signal and correcting bulk organic data from a peat deposit, a case study at low latitudes (Cameroon)

Abstract: International audience; Organic compounds are widely used for paleoclimatic and paleoenvironmental reconstructions. Bulk organic proxies, however, are more complicated to interpret due to the multiple causes of variation in climatic and environmental conditions and the degree of diagenetic alteration. As labile compounds, rich in easily degradable function and generally richer in heteroatoms such as oxygen and nitrogen, decompose, the remaining organic matter becomes progressively richer in refractory carbon and its carbon content increases. Thus, in a peat deposit composed almost entirely of organic matter, total organic carbon (TOC) is expected to increase with time and depth, which could mask the paleoclimatic signal. We propose a simple model for peat sediments to remove the decomposition signal based on a logarithmic function fitted with a partial dataset where decomposition is the main parameter. The subtraction of the obtained logarithmic function to the raw data (i.e., measured data) leads to “residual” data. We discuss the influence of different parameters (water table depth, vegetation, microbial community) on the “residual” data and their possible link to paleoclimatic and paleoenvironmental variations. This method is tested on bulk elemental and isotopic data obtained from a new peat core from the Adamawa Plateau (North-East Cameroon) covering nearly 10 ka cal BP. Comparison with Rock-Eval® parameters highlights similar variations between the Hydrogen Index and residual TOC variations, supporting the interpretation based on residual TOC. Our approach allows to extract paleoenvironmental information from decomposition-prone bulk organic proxies and can be generalized to peat deposits where decomposition plays a major role in controlling bulk data.

Abstract: The aim of this work is to characterize the geographical origin of cocoa beans and coffee beans. This study aims to contribute to the traceability of raw materials in order to fight against falsification. For this purpose, we based our work on the measurement of isotope ratios in 13C, 15N and 18O. The multi-element isotope ratios have been evaluated as a means to distinguish fermented cocoa beans of different geographic and varietal origins. The isotopic ratios of 13C, 15N and 18O were measured in theobromine obtained from samples of fermented cocoa beans. Twenty-two (22) samples of different geographical origins covering the four mainland cocoa producing areas were analyzed on the one hand and on the other hand, 16 caffeine samples from various origins were also analyzed. The treatment of the values resulting from these isotopic analyzes by statistical methods, namely the principal component analysis (PCA) makes it possible to visualize the discriminations between the different origins. The most discriminating variables identified as responsible for the geographic and varietal differences were the δ15N, δ13C and δ18O values of cocoa beans and certain extracts and tissues. We have shown that the isotope ratios are correlated with the altitude and precipitation conditions encountered in the different cocoa growing regions.

Abstract: International audience; Studies on microbial communities, and their associated organic biomarkers, that are found thriving in the aphotic euxinic waters in modern stratified ecosystems are scarce compared to those undertaken in euxinic photic zones. The Dziani Dzaha (Mayotte,Indian Ocean) is a tropical, saline, alkaline crater lake that has recently been presentedas a modern analog of Proterozoic Oceans due to its thalassohaline classification (having water of marine origin) and specific biogeochemical characteristics. Continuous intense photosynthetic production and microbial mineralization keep most of the water column permanently aphotic and anoxic preventing the development of a euxinic (sulfidic and anoxic) photic zone despite a high sulfide/sulfate ratio and the presence of permanent or seasonal haloclines. In this study, the molecular composition of the organic matter in Lake Dziani Dzaha was investigated and compared to the microbial diversity evaluated through 16S rRNA gene amplicon sequencing, over two contrasting seasons (rainy vs. dry) that influence water column stratification. Depth profiles of organic biomarker concentrations (chlorophyll-a and lipid biomarkers) and bacterial and archaeal OTU abundances appeared to be strongly dependent on the presence of aphotic haloclines and euxinia. OTU abundances revealed theimportance of specific haloalkaliphilic bacterial and archaeal assemblages in phytoplanktonic biomass recycling and the biogeochemical functioning of the lake, suggesting new haloalkaline non-phototrophic anaerobic microbial precursors for some of the lipid biomarkers. Uncultured Firmicutes from the family Syntrophomonadaceae (Clostridiales), and Bacteroidetes from the ML635J-40 aquatic group, emerged as abundant chemotrophic bacterial members in the anoxic or euxinic waters and were probably responsible for the production of short-chain n-alkenes, wax esters, diplopterol, and tetrahymanol. Halocline-dependent euxinia also had a strong impact on the archaeal community which was dominated by Woesearchaeota in the sulfide-free waters. In the euxinic waters, methanogenic Euryarchaeota from the Methanomicrobia, Thermoplasmata, and WSA2 classes dominated and were likely at the origin of common hydrocarbon biomarkers of methanogens (phytane, pentamethyl-eicosenes, and partially hydrogenated squalene).

Abstract: A coupled pollen-brGDGT paleoclimate reconstruction approach has been tested to provide independent and robust estimates of Holocene climate and environment changes in the extremely arid environment of the mountainous areas ranging from northern Arid Central Asia (ACA) to the Mongolian Plateau. The two proxies were calibrated for both global and local modern data sets (NMSDB). This multi-proxy approach was then applied to a sediment core collected from Lake Ayrag, Arkhangai, covering the Late Holocene. In addition to brGDGTs and pollen, we also performed magnetic susceptibility, micro-XRF, elemental and isotopic bulk chemistry, and Non-Pollen Palynomorph (NPP) analyses on the Lake Ayrag sediments in order to better understand the lake system and human impact dynamics. While the globally calibrated record (both for pollen and brGDGTs) displayed a slight millennial-scale cooling, the locally calibrated results exhibit centennial-scale climate oscillations such as the 4.2 and 3.5 kyr events, the Roman Warm Period (RWP), Dark Ages Cold Period (DACP), Medieval Warm Period (MWP) and Little Ice Age (LIA). These climate oscillations and vegetation changes are discussed with regard to the main Mongolian human historical occupation events documented by pastoralism proxies, especially the Xiongnu, Mongol Empire, Mandchou and Soviet periods. The climate systems currently dominating the Mongolian Plateau are difficult to resolve because inter-annual climate variability is pronounced. However, precipitation mainly occurs in summer (easterly monsoon driven) when the winter Westerlies lead the air mass movement. In the past, both pollen and biomarkers exhibited anti-correlated trends with annual precipitation and temperature: over the last 4000 kcal yr BP, the warm periods (MWP, RWP) were dry and the cold periods (LIA, DACP, 3.5 kyrs) were humid. Thus, the East Asian Summer Monsoon (i.e., warm and wet conditions dominant during summer) seems not to have influenced central Mongolian climate during the Late Holocene, which could have remained dominated by the Westerlies/Siberian High cells conflict. A comparison between the Ayrag record and other paleoclimate records from the Baikal area (Dulikha), Mongolian Plateau (D3L6, D1L1, NRX, ATM), and continental China (Kesang, Baluk and Tonnel caves, XRD section) to the Loess Plateau (Huangye and Xianglong caves) suggests that the monsoon front has oscillated since the Early Holocene. A climate synthesis following strictly the same approach (locally calibrated brGDGTs vs. pollen-inferred climate) for all the ACA records available for the Late Holocene helps us to resolve the climate systems paced by centennial to millennial-scale oscillations and their consequences for human societies.

12. Effect of serpentinization on carbon speciation: an experiment with formic acid

Abstract: One of the principal theories about the origin of life is based on the abiotic reduction of carbon oxides to various organic molecules in hydrothermal systems. This synthesis is most favored in ultramafic environments undergoing hydrothermal alteration where the serpentinization reaction efficiently produces H2. Nevertheless, decades of hydrothermal experiments have hardly succeeded in producing abundant organic volatiles such as CH4 and short-chain hydrocarbons. On another hand, natural observations have shown the occurrence of other abiotic compounds such as organic acids in fluids and carbonaceous matter (CM) within serpentinized rocks. But organic acids as carbon source and CM as product have not been investigated so far in experiments reproducing hydrothermal peridotite alteration. Here, we explored the effect of formic acid (HCOOH) on the serpentinization reaction and possible feedback effects on carbon speciation in both fluid and solid. We performed reactions at 300°C and 250 bar using peridotite powder (2, CO, and CO2. After 4 months, H2, CO, CO2, CH4 and short-chain alkanes (mainly ethane) were measured in the fluid, and the powder was completely indurated. The solidified powder displayed a black and white layering perpendicular to fluid diffusion. Its analysis showed the advancement of the serpentinization reaction, and the incorporation of carbon compounds into the solid phase. XRD analysis indicated 70% of serpentinization. SEM-EDX observations showed peculiar texture with large and localized euhedral magnetite grains alternating with larger magnetite grains mixed with C-enriched areas of long chrysotile fibers. FT-IR measurement attested of the widespread formation of carbonaceous material in the solid. Liquid analyses are under progress. Those first results suggest that serpentine formation not only provides additional H2 to the system, but also mineral surfaces that could play a role in the precipitation of carbonaceous material and carbon speciation in natural systems. The nature and formation mechanisms of this latter remain to be addressed but this opens new paths for abiotic organic synthesis under hydrothermal conditions. In addition to their implications as an abiotic carbon source for deep hydrocarbon degraders ecosystems, it could have important implications for the total carbon cycle.

Abstract: International audience; Specific n-alkanes and n-alkanoic acids are commonly used as biomarkers in paleoenvironmental reconstruction, yet any individual homologue may originate from multiple biological sources. Here we improve source and age controls for these compounds in meromictic systems by measuring the radiocarbon (C-14) ages of specific homologues preserved in twentieth century Lake Pavin (France) sediments. In contrast to many studies, C-14 ages generally decreased with increasing carbon chain length, from 7.3 to 2.6 ka for the C-14-C-30 n-alkanoic acids and from 9.2 to 0.3 ka for the C-21-C-33 n-alkanes. Given a known hard water effect, these values suggest that aquatic microbial sources predominate and contributed to most of the homologues measured. Only the longest chain n-alkanes exclusively represent inputs of higher plant waxes, which were previously sequestered in soils over centennial to millennial timescales prior to transport and deposition. These findings suggest that biomarker source and age should be carefully established for lacustrine settings.

Abstract: Glutamine synthetase (GS, EC 6.3.1.2) catalyzes the production of glutamine from,glutamate, ammonium and ATP. Although being essential in plants for N assimilation and recycling, kinetic commitments and transition states of the reaction have not been clearly established yet. Here, we examined C-12/C-13,N-14/N-15 and H2O/D2O isotope effects in Arabidopsis GS1 catalysis and compared to the prokaryotic (Escherichia coli) enzyme. A(14)N/N-15 isotope effect (V-15/K approximate to 1.015, with respect to substrate NH4+) was observed in the prokaryotic enzyme, indicating that ammonium utilization (deprotonation and/or amidation) was partially rate-limiting. In the plant enzyme, the isotope effect was inverse (V-15/K = 0.965), suggesting that the reaction intermediate is involved in an amidation-deamidation equilibrium favoring N-15. There was no C-12/C-13 kinetic isotope effect (V-13/K = 1.000), suggesting that the amidation step of the catalytic cycle involves a transition state with minimal alteration of overall force constants at the C-5 carbon. Surprisingly, the solvent isotope effect was found to be inverse, that is, with a higher turn-over rate in heavy water (V-D approximate to 0.5), showing that restructuration of the active site due to displacement of H2O by D2O facilitates the processing of intermediates. Abstract: International audience; Deciphering the role of sulfur in biogeochemical cycles strongly relies on its stable isotope composition, which ultimately depends on the ability to quantitatively recover different sulfur species from geological samples. For decades most studies have been restricted to the 34S/32S composition of bulk samples, using simple methods based on the analysis of SO2 released by sample combustion combined to mass spectrometry. The wet chemistry procedures required to selectively extract specific sulfur species were generally avoided due to their tediousness and inefficiency for some complex matrices, especially when targeting organic sulfur. With the recent advent of multi-isotope studies (investigating the minor sulfur stable isotopes 33S and 36S) which rely either on the analysis of sulfur as SF6, or on the use of secondary ion or multi-collector inductively coupled plasma mass spectrometry, wet chemistry-based preparation procedures were brought back to the stage with a renewed interest in developing procedures better adapted to the investigation of specific sulfur species. Here we propose a new stepwise chemical procedure for the quantitative recovery and multi-isotope analysis of organic sulfur from both solvent soluble (total lipid extract) and insoluble (kerogen) fractions, based on a wet oxidation by sodium hypochlorite. This procedure also allows the multi-isotope analysis of inorganic sulfur species (elemental sulfur, sulfates and sulfides) in the same sample. Its application to different well-known petroleum source rocks and to an oil demonstrates its relevance for disentangling the interactions between the different sulfur pools and for shedding new light on the sulfur biogeochemical cycle.

Abstract: Archaea and Bacteria that inhabit the deep subsurface (known as the deep biosphere) play a prevalent role in the recycling of sedimentary organic carbon. In such environments, this process can occur over millions of years and requires microbial communities to cope with extremely limited sources of energy. Because of this scarcity, metabolic processes come at a high energetic cost, but the ways heterotrophic microbial communities develop to minimize energy expenses for a maximized yield remain unclear. Here, we report molecular biomarker evidence for the recycling of archaeal cell wall constituents in extreme evaporitic facies of Dead Sea deep sediments. Wax esters derived from the recombination of hydrolyzed products of archaeal membrane lipids were observed in gypsum and/or halite sedimentary deposits down to 243 m below the lake floor, implying the reutilization of archaeal necromass possibly by deep subsurface bacteria. By recycling the building blocks of putatively better-adapted archaea, heterotrophic bacteria may build up intracellular carbon stocks and mitigate osmotic stress in this energy-deprived environment. This mechanism illustrates a new pathway of carbon transformation in the subsurface and demonstrates how life can be maintained in extreme environments characterized by long-term isolation and minimal energetic resources.

Abstract: International audience; Breast cancer is the most common cancer in women worldwide. Despite the information provided by anatomopathological assessment and molecular markers (such as receptor expression ER, PR, HER2), breast cancer therapies and prognostics depend on the metabolic properties of tumor cells. However, metabolomics have not provided a robust and congruent biomarker yet, likely because individual metabolite contents are insufficient to encapsulate all of the alterations in metabolic fluxes. Here, we took advantage of natural 13 C and 15 N isotope abundance to show there are isotopic differences between healthy and cancer biopsy tissues or between healthy and malignant cultured cell lines. Isotope mass balance further suggests that these differences are mostly related to lipid metabolism, anaplerosis and urea cycle, three pathways known to be impacted in malignant cells. Our results demonstrate that the isotope signature is a good descriptor of metabolism since it integrates modifications in C partitioning and N excretion altogether. Our present study is thus a starting point to possible clinical applications such as patient screening and biopsy characterization in every cancer that is associated with metabolic changes. Medical applications of stable isotopes are now widespread, like the well-known 13 C-urea breath assay for ulcer detection 1. This takes advantage of 13 C-labelling and thus usually neglects differences in reaction rates between isotopic forms, because the isotopic signal used for diagnosing is far above small natural variations in 13 C. By contrast, the use of isotopes at natural abundance exploits such subtle differences (referred to as isotope effects) to identify bottlenecks in metabolic pathways (rate-limiting steps) or the contribution of multiple elemental sources (mass balance), without the need to introduce expensive isotope tracers into the patient. Isotope effects in metabolism are mostly caused by enzymatic reactions that preferentially consume substrates containing either the light or the heavy isotope (isotopologues) and therefore, the natural isotope abundance in metabolites depends on metabolic fluxes and source substrates 2. For example, the natural 13 C abundance in respired CO 2 has been used to trace diet composition and substrate changes during exercise 3,4. In cancer biology, the use of natural variations in Cu and S stable isotopes in hepatocellular carcinoma has been attempted recently 5. But to our knowledge, no study has looked at alterations of natural isotope abundance in breast cancer. Due to changes in primary C and N metabolism such as increased glycolysis, glutaminolysis and nucleotide synthesis 6 , important changes in 13 C and 15 N natural abundance can be anticipated. To address this question, we examined the isotopic signature of intact breast cancer biopsies (mostly from invasive ductal carcinoma, IDC) and cultured breast cancer cell lines (Supplementary Tables S1 and S2) using elemental analysis coupled to isotope ratio mass spectrometry (EA-IRMS). This technique has been recently shown to be applicable to the biochemical analysis of cancerous cell lines 7 .

Abstract: The quality of the determination of compound-specific isotopic content at natural abundance by gas chromatography-isotope ratio measurement-mass spectrometry (GC-irm-MS) relies on the stability of the voltage generated by the ion detector Faraday cages. The application of GC-irm-MS to the determination of δ(13)C (‰) and δ(15)N (‰) is now routine. However, for numerous applications, it is necessary to determine both the isotope content (δ(15)N) and the quantity (in micromoles) of analyte present. We now show that it is possible for nitrogen-containing compounds to measure how much analyte is present with an irm mass spectrometer linked to a GC by exploiting the integrated N(2) total ion current intensity (Vs) generated by measuring the (15)N/(14)N isotope ratio. The method is validated over a range of concentration (2-70 mmol/L) and δ(15)N (-70 to +50‰) values for six molecules of diverse chemical nature and functionality (nortropine, norpseudotropine, nortropinone, cysteine, taurine, glutathione). It is shown that once the ion current is calibrated, the quantitative values are of a comparable quality to those obtained from GC with flame ionization detection (GC-FID). In addition, it is demonstrated that over a definable range, the δ(15)N (‰) value is independent of the quantity of analyte introduced, confirming the validity of this method.

Abstract: The measurement of the nitrogen isotope ratio of taurine (2-aminoethanesulphonic acid) in biological samples has a large number of potential applications. Taurine is a small water-soluble molecule which is notoriously difficult to analyze due to its polarity and functionality. A method is described which allows the determination of the natural abundance delta N-15 values of taurine and structural analogues, such as 3-amino-1-propanesulphonic acid (APSA), by isotope ratio mass spectrometry interfaced to gas chromatography (GC-irm-MS). The one-step protocol exploits the simultaneous derivatization of both functionalities of these aminosulphonic acids by reaction with triethylortho-acetate (TEOA). Conditions have been established which ensure quantitative reaction thus avoiding any nitrogen isotope fractionation during derivatization and workup. The differences in the delta N-15 values of derivatized and non-derivatized taurine and APSA all fall within the working range of 0.4 parts per thousand (-0.02 to 0.39 parts per thousand). When applied to four sources of taurine with various delta N-15 values, the method achieved excellent reproducibility and accuracy. The optimized method enables the determination of the natural abundance delta N-15 values of taurine over the concentration range 1.5-7.84 mu mol.mL(-1) in samples of biological origin. Isotope ratios and percentage composition for C and N were measured in different tissues (cotyledons, shells) and extracts (pure theobromine, defatted cocoa solids, protein, lipids) obtained from fermented cocoa bean samples. Sixty-one samples from 24 different geographical origins covering all four continental areas producing cocoa were analyzed. Treatment of the data with unsupervised (Principal Component Analysis) and supervised (Partial Least Squares Discriminant Analysis) multiparametric statistical methods allowed the cocoa beans from different origins to be distinguished. The most discriminant variables identified as responsible for geographical and varietal differences were the δ15N and δ13C values of cocoa beans and some extracts and tissues. It can be shown that the isotope ratios are correlated with the altitude and precipitation conditions found in the different cocoa-growing regions.

Abstract: International audience; In the article (Romek et al. 2013) we reported the values of δ15N (‰) and δ13C (‰) obtained bySince exclusively breast-suckled infants obtain their nutrient only from their mother's milk, it might be anticipated that a correlation will exist between the (15)N/(14)N isotope ratios of amino acids of protein of young infants and those supplied by their mother. The work presented here aimed to determine whether amino nitrogen transfer from human milk to infant hair protein synthesized within the first month of life conserves the maternal isotopic signature or whether post-ingestion fractionation dominates the nitrogen isotope spectrum. The study was conducted at 1 month post-birth on 100 mother-infant pairs. Isotope ratios (15)N/(14)N and (13)C/(12)C were measured using isotope ratio measurement by Mass Spectrometry (irm-MS) for whole maternal milk, and infant hair and (15)N/(14)N ratios were also measured by GC-irm-MS for the N-pivaloyl-O-isopropyl esters of amino acids obtained from the hydrolysis of milk and hair proteins. The δ(15)N and δ(13)C (‰) were found to be significantly higher in infant hair than in breast milk (δ(15)N, P < 0.001; δ(13)C, P < 0.001). Furthermore, the δ(15)N (‰) of individual amino acids in infant hair was also significantly higher than that in maternal milk (P < 0.001). By calculation, the observed shift in isotope ratio was shown not to be accounted for by the amino acid composition of hair and milk proteins, indicating that it is not simply due to differences in the composition in the proteins present. Rather, it would appear that each pool-mother and infant-turns over independently, and that fractionation in infant N-metabolism even in the first month of life dominates over the nutrient N-content.

Abstract: International audience; RATIONALE In isotope tracer experiments used in nutritional studies, it is frequently desirable both to determine the 15N/14N ratios of target compounds and to quantify these compounds. This report shows how this can be achieved in a single chromatographic run for protein amino acids using an isotope ratio mass spectrometer. METHODS Protein hydrolysis by acidic digestion was used to release amino acids, which were then derivatized as their N-pivaloyl-O-isopropyl esters. Suitable conditions for sample preparation were established for both hair and milk proteins. The N-pivaloyl-O-isopropyl esters of amino acids were separated by gas chromatography (GC) on a 60m ZB-WAX column linked via a combustion interface to an isotope ratio mass spectrometer. The 15N/14N ratios were obtained from the m/z 28, 29 and 30 peak intensities and the quantities from the Area All (Vs) integrated peak areas. RESULTS It is shown from a five-point calibration curve that both parameters can be measured reliably within the range of 1.0 to 2.0mg/mL for the major amino acids derived from the hydrolysis of human maternal milk or hair samples. The method was validated in terms of inter-day and inter-user repeatability for both parameters for 14 amino acids. The amino acid percentage composition showed a good correlation with literature values. The method was applied to determine the variability in a population of lactating mothers and their infants. CONCLUSIONS It has been established that 15N values can be simultaneously determined for a complex mixture of amino acids at variable concentrations. It is shown that the percentage composition obtained correlates well with that obtained by calculation from the protein composition or from literature values. This procedure should provide a significant saving in analysis time, especially in those cases where the GC run-time is necessarily long. It allows the satisfactory determination of the variation within a sample population. Copyright (c) 2013 John Wiley & Sons, Ltd.

Abstract: Two-dimensional nuclear magnetic resonance (2D NMR) forms a powerful tool for the quantitative analysis of complex mixtures such as samples of metabolic relevance. However, its use for quantitative purposes is far from being trivial, not only because of the associated experiment time, but also due to its subsequent high sensitivity to hardware instabilities affecting its precision. In this paper, an alternative approach is considered to measure absolute metabolite concentrations in complex mixtures with a high precision in a reasonable time. It is based on a ``multi-scan single shot'' (M3S) strategy, which is derived from the ultrafast 2D NMR methodology. First, the analytical performance of this methodology is compared to the one of conventional 2D NMR. 2D correlation spectroscopy (COSY) spectra are obtained in 10 min on model metabolic mixtures, with a precision in the 1-4% range (versus 5-18% for the conventional approach). The M3S approach also shows a better linearity than its conventional counterpart. It ensures that accurate quantitative results can be obtained provided that a calibration procedure is carried out. The M3S COSY approach is then applied to measure the absolute metabolite concentration in three breast cancer cell line extracts, relying on a standard addition protocol. M3S COSY spectra of such extracts are recorded in 20 min and give access to the absolute concentration of 14 major metabolites, showing significant differences between cell lines.

28. A test to identify cyanide origin by isotope ratio mass spectrometry for forensic investigation

Abstract: Cyanide is one of the common poisons in murders. When cyanide has been used, to identify the origin of cyanide may be necessary in the forensic investigation. We have examined the possibility of distinguishing different commercial cyanide samples through the δ 13 C and δ 15 N values and developed a protocol for the isotope analysis of cyanide extracted from several matrices as food and medicine. Several cyanide precipitates were tested for the isotope analysis. The results show that cupric ferrocyanide Cu 2 [Fe(CN) 6 ] is the most appropriate precipitate for the analysis. Thirteen batches of KCN and nine batches of NaCN chemicals were randomly chosen from different suppliers. The cyanides were converted to cupric ferrocyanide and then analysed by isotope ratio mass spectrometry coupled to elemental analysis (EA-IRMS). The isotopic signature of the commercial samples varied from −51.96 to −25.77‰ for δ 13 C and from −4.51 to +3.81‰ for δ 15 N, highlighting the potential of applying EA-IRMS technique to identify cyanide from different batches and sources. The influence of the cyanide extraction and isolation from spiked matrix on the isotopic analysis was also studied. Three matrices: orange juice, yogurt drink and a medicine were tested. In many cases, the isotopic analysis results obtained from the original cyanides precipitates and those isolated from the matrices showed a good accordance, especially for δ 15 N. In some matrices, the 13 C analysis was interfered by co-precipitates. With carefully elaborated working protocol, determining the isotope ratio of N and C in cyanide by EA-IRMS is a promising method for forensic investigations.

Abstract: The measurement of the nitrogen isotope ratio of taurine (2-aminoethanesulphonic acid) in biological samples has a large number of potential applications. Taurine is a small water-soluble molecule which is notoriously difficult to analyze due to its polarity and functionality. A method is described which allows the determination of the natural abundance δ(15)N values of taurine and structural analogues, such as 3-amino-1-propanesulphonic acid (APSA), by isotope ratio mass spectrometry interfaced to gas chromatography (GC-irm-MS). The one-step protocol exploits the simultaneous derivatization of both functionalities of these aminosulphonic acids by reaction with triethylorthoacetate (TEOA). Conditions have been established which ensure quantitative reaction thus avoiding any nitrogen isotope fractionation during derivatization and workup. The differences in the δ(15)N values of derivatized and non-derivatized taurine and APSA all fall within the working range of 0.4‰ (-0.02 to 0.39‰). When applied to four sources of taurine with various δ(15)N values, the method achieved excellent reproducibility and accuracy. The optimized method enables the determination of the natural abundance δ(15)N values of taurine over the concentration range 1.5-7.84 µmol.mL(-1) in samples of biological origin.
